目的 :观察滋肾方对亚急性衰老大鼠学习记忆及海马组织胆碱酯酶 (CHE)和单胺氧化酶 (MAO)的影响 ,以探讨滋肾方抗衰老的作用。方法 :持续注射D 半乳糖法致亚急性衰老大鼠模型 ,“Y”型电迷宫法进行学习记忆观察 ,并对海马组织进行酶学测定。结果 :滋肾方可以减少大鼠达标所需的电击次数 (P <0 .0 1) ,提高大鼠海马组织中胆碱酯酶活性 (P <0 .0 5 ) ,降低大鼠海马组织中单胺氧化酶活性 (P <0 .0 1)。结论 :滋肾方可以提高衰老大鼠学习记忆能力 ,其作用机制可能与提高大鼠海马组织中胆碱酯酶活性、降低单胺氧化酶活性有关  相似文献

单侧切断大鼠海马缴和部分穹窿可使海马部分去神经。损伤后7d,海马内胆碱能系统中乙酰胆碱(ACh)含量下降72.5%,胆碱乙酰基转移酶(ChAT)活力下降45.7%,胆碱酯酶活力下降52.2%,在单胺能系统中,去甲肾上腺素(NA)含量下降16.3%,多巴胺(DA)含量下降31.3%,5-羟色胺含量下降30.3%。在损伤过程中,海马内氨基酸含量没有改变。实验结果表明,海马缴和穹窿是脑内胆碱能和单胺能传入神经到达海马靶区的部分共同通路。  相似文献

本文旨在研究α2肾上腺素能受体激动剂右美托咪定对酒精依赖大鼠戒断症状的影响及作用机制,以期为酒精戒断综合征(alcohol withdrawal syndrome, AWS)的治疗提供科学依据。成年Sprague-Dawley (SD)雄性大鼠连续28 d饮用6%酒精水溶液建立饮酒模型,然后停止饮酒以诱导AWS。用酶联免疫吸附法测定大鼠脑组织蓝斑核及海马去甲肾上腺素的含量。在大鼠AWS显著时分别腹腔注射5、10、20μg/kg右美托咪定,并记录戒断症状变化,部分大鼠预先在海马注射α2肾上腺素能受体拮抗剂育亨宾。结果显示,与对照组相比,酒精戒断6 h组大鼠AWS评分和再次饮酒量显著提高,末次饮酒组和戒断6 h组海马及蓝斑核去甲肾上腺素含量显著升高。右美托咪定干预后,戒断6 h组AWS评分及海马去甲肾上腺素含量均显著降低,而育亨宾可以逆转右美托咪定的上述作用。以上结果提示,右美托咪定可能通过激活海马α2肾上腺素能受体改善酒精依赖大鼠戒断症状。  相似文献

中药天年饮对衰老大鼠脑单胺类神经递质含量的影响   总被引:2,自引:0,他引:2  
目的:观察中药天年饮(Tiannianyin,TNY-traditional chinese medicine)对D-半乳糖衰老大鼠脑单胺类神经递质去甲肾上腺素(NE)、多巴胺(DA)、5-羟色胺(5-HT)含量的影响。方法:选用成年雄性SD大鼠40只.随机分为4组.每组均为10只:正常组、衰老模型组、TNY用药组、阴性对照组。Ⅱ半乳糖连续腹腔注射制作亚急性衰老的大鼠模型.采用高效液相色谱-电化学法检测各组大鼠下丘脑、海马NE、DA、5-HT的含量。结果:D-半乳糖衰老大鼠下丘脑、海马NE、DA、5-HT的含量明显降低(与正常大鼠相比P〈0.01):TNY可明显提高脑单胺类神经递质的含量(用药组与模型组相比P〈0.05)。结论:TNY可有效调整中枢神经递质的合成,具有良好延缓衰老的作用。  相似文献

目的:通过观察慢性温和应激对大鼠行为及5-羟色胺、色氨酸、应激激素的影响,进一步阐明应激致抑郁发生的机制。方法:利用慢性不可预计温和应激(CUMS)结合行为学指标建立大鼠抑郁动物模型;高效液相检测血浆和脑5-HT、儿荼酚胺;放免方法检测血浆皮质醇换算成皮质酮含量;氨基酸分析仪检测血清色氨酸的含量。结果:①CUMS诱导8周大鼠糖水偏爱性与对照组相比明显减低(P〈0.05);体重和旷场实验评分下降(P〈0.05)。②CUMS诱导8周大鼠大脑和血浆5-HT含量显著下降(P〈0.05);而血清色氨酸含量与对照组相比则明显升高(P〈0.05)。③CUMS诱导8周大鼠血浆去甲肾上腺素和肾上腺素含量与对照组相比明显升高(P〈0.05):血浆皮质酮含量与对照组相比统计学上无显著性差异(P〉0.05)。结论:利用CUMS建立的抑郁模型大鼠存在着神经内分泌紊乱,CUMS大鼠抑郁样行为的发生可能与血浆去甲肾上腺素和肾上腺素升高对色氨酸代谢的影响有关。  相似文献

目的:建立大鼠抑郁模型,研究和探讨灵孢多糖注射液辅助治疗抑郁症的作用及机制。方法:将48只SD大鼠随机分为正常组、抑郁组、生理盐水组及灵孢多糖(低剂量、中剂量、高剂量)治疗组各8只。应用慢性应激刺激的方法诱导抑郁组、生理盐水组、灵孢多糖治疗组大鼠抑郁,通过行为学指标评估建模是否成功。建模成功后,在生理盐水组中注射生理盐水;在灵孢多糖治疗组中分别注射低、中、高剂量灵孢多糖注射液,比较各组间的疗效差异。采用免疫组化的方法测定大鼠脑内多巴胺、去甲肾上腺素、5-羟色胺、酪氨酸羟化酶等单胺类神经递质含量的表达情况。结果:成功的建立了大鼠抑郁模型,抑郁组中大鼠各项行为学指标与正常组相比均明显减少,差异有统计学意义(P0.05);灵孢多糖治疗组大鼠脑内的多巴胺、去甲肾上腺素、5-羟色胺、酪氨酸羟化酶与抑郁组、生理盐水组大鼠脑内的含量均明显升高,差异有统计学意义(P0.05)。结论:灵孢多糖注射液对改善抑郁症状有良好的疗效,具有辅助治疗抑郁症的作用。  相似文献

为探讨合欢花对慢性应激大鼠生长和脑单胺类神经递质的影响,采用15只大鼠,设置了对照组、应激组和合欢花组3组实验。应激组和合欢花组均接受7天的应激刺激,之后合欢花组再灌胃合欢花10天。实验结束后,取3组大鼠的脑组织,用高效液相色谱法测定高香草酸(HVA)、去甲肾上腺素(NE)、多巴胺(DA)和5-羟色胺(5-HT)的含量。结果表明,应激组大鼠日增重显著低于对照组(P=0.011);而合欢花组大鼠的日增重极显著高于应激组(P=0.002)。应激组大鼠海马、纹状体和前额叶中的HVA含量与对照组相比,虽有升高的趋势,但无显著差异;两组间的NE、DA和5-HT也无显著差异。合欢花组大鼠海马中的HVA、DA含量明显高于应激组,而前额叶中的多巴胺和5-羟色胺,以及纹状体中的5-羟色胺均明显低于应激组。这表明合欢花对慢性应激引起的大鼠生长受抑有缓解作用,对其脑内单胺类神经递质有调节作用。  相似文献

下丘脑弓状核区是脑内合成β-内啡肽等物质的主要部位,并与脑内中缝背核、蓝斑等结构有密切的交互纤维支配。本实验用新生期大鼠注射谷氨酸—钠(MSG)损毁弓状核区的方法,观察对脑内β-内啡肽、5-羟色胺、去甲肾上腺素含量及针刺镇痛的影响。MSG 处理组大鼠下丘脑弓状核神经元减少72%左右,脑β-内啡肽含量降低67%,针刺镇痛效应明显下降,电针后脑去甲肾上腺素含量明显高于电针对照组;将 MSG 处理大鼠的垂体摘除后,针刺镇痛效应几乎消失,同时电针后脑去甲肾上腺素含量则显著高于单纯 MSG处理组。本文对可能的机理进行了讨论。  相似文献

以3d龄Sprague-Dawley大鼠腹腔注射苯丙氨酸(Phe)诱导高苯丙氨酸血症,荧光法测定大脑皮层及其突触体中去甲肾上腺素(NE)、多巴胺(DA)和5-羟色胺(5-HT)含量;Y型电迷宫法测其学习记忆能力.结果显示:高苯丙氨酸血症大鼠大脑皮层NE、DA及5-HT含量降低38.6%~67.4%,突触体中NE、DA和5-HT含量降低51.9%~70.2%,学习记忆能力明显低于对照组.结果提示,苯酮尿症智力障碍可能与大脑皮层及其突触体中某些单胺类递质含量降低相关.  相似文献

目的:观察甘麦大枣汤对抑郁模型大鼠行为学及单胺递质的影响,并从突触结构及结构蛋白MAP-2与GAP-43表达改变探讨其潜在作用机制。方法:60只SD大鼠随机分为5组:正常对照组、模型组、氟西汀组(10.8mg/kg)、甘麦大枣汤高、低剂量组(9.72、4.86 g/kg),每组12只。除对照组外,其余各组大鼠均采用慢性不可预见性温和应激(CUMS)建立抑郁模型,并于造模同时给药组灌胃给药,连续21 d。采用糖水消耗实验和旷场测试评价大鼠抑郁样行为,ELISA法检测海马单胺递质5-HT、NE含量,Golgi染色观察神经元突触损伤情况,免疫组化法和Western blot法检测海马突触结构蛋白MAP-2和GAP-43的表达。结果:与对照组比较,抑郁模型大鼠糖水偏好度及自主活动评分均显著下降(P<0.01),海马5-HT、NE含量显著下降(P<0.01),树突棘缺失明显,同时MAP-2和GAP-43表达均显著下调(P<0.01);甘麦大枣汤干预后,模型大鼠抑郁样行为明显缓解(P<0.01),5-HT、NE含量显著升高(P<0.05),树突棘密度、长度及分枝增加,...  相似文献

In our recent studies on nicotine-induced changes in neurotransmitters in brain areas associated with cognitive function using a nicotine dose of 0.5 mg/kg administered subcutaneously to conscious freely moving rats, we found changes in dopamine, norepinephrine, and serotonin, and their metabolites, in the areas examined. For the present report we examined changes in these neurotransmitters following administration of lower nicotine doses, to test regional differences in nicotine response and possible threshold levels for some effects of nicotine. The doses used were 0.15 mg/kg and 0.03 mg/kg nicotine administered subcutaneously. Nicotine levels in the brain reached peak values in less than 10 min and decreased with a half-life of about 60 min (0.15 mg/kg) or 30 min (0.03 mg/kg) to values below detection limits (1 ng/g), by the later time points of the 0.03 mg/kg experiments. Nicotine-induced dopamine (DA) increase (and increase in DA metabolites) and decrease in 5-HT levels at 0.15 mg/kg were significant in the cortex, less so in the hippocampus. Norepinephrine (NE) increase at 0.15 mg/kg was much less significant than found previously at 0.5 mg/kg. At a low nicotine dose (0.03 mg/kg), the significant changes observed were a decrease in 5-HT in the hippocampus and small increases of DA and NE in the prefrontal cortex and of NE in the medial temporal cortex. In the nucleus accumbens DA, NE, and 5-HT and their metabolites in the ventral tegmental area, mostly DA and metabolites were increased. We conclude that in areas of cognitive function nicotine-induced DA changes are more concentration dependent than changes in NE or 5-HT, and that there are regional differences in neurotransmitter changes induced by nicotine, with NE changes detectable only in the cortex and 5-HT changes only in the hippocampus at a low nicotine dose, indicating significant regional variation in sensitivity to nicotine-induced neurotransmitter changes in brain areas associated with cognitive function. The decrease in 5-HT shows that nicotine also has indirect effects caused by neurotransmitters released by nicotine. The effects of low nicotine dose are more significant in areas of reward function, indicating differences in sensitivity between cognitive and reward functions.  相似文献

Six brain areas of rats and guinea-pigs, killed by microwave irradiation, were used for the concomitant measurement of the levels and regional distribution of cholinergic, biogenic amine, and amino acid neurotransmitters and metabolites. Acetylcholine (ACh) and choline (Ch) were quantified by chemiluminescence; noradrenaline (NA), dopamine (DA), 5-hydroxytryptamine (5-HT), and their metabolites by HPLC with electrochemical detection (HPLC-EC); and six putative amino acid neurotransmitters by HPLC-EC following derivatisation. The levels and regional distribution of these transmitters and their metabolites in the rat were similar to those reported in previous studies, except that biogenic amine transmitter levels were higher and metabolite concentrations were lower. The guinea-pig showed a similar regional distribution, but the absolute levels of ACh were lower in striatum and higher in hippocampus, midbrain-hypothalamus, and medulla-pons. In all areas, the levels of Ch were higher and those of NA, 5-HT, and taurine were lower than in the rat. The most marked differences between the rat and guinea-pig were in the relative proportion of DA metabolites and 5-HT turnover, as estimated by metabolite/transmitter ratios. This study can be used as a basis for a comprehensive understanding of the central effects of drugs on the major neurotransmitter systems.  相似文献

探讨电磁脉冲(EMP)对大鼠神经系统的效应。实验采用Wistar大鼠,EMP辐照后不同时间用Y-型迷宫测其学习能力,高效液相色谱法检测脑不同部位的神经递质含量。与假照射组(对照组)相比,照后三天内各测定组大鼠学习能力降低(P<0.05),其中照射后第1天组的海马内5-羟色胺(5-HT)和多巴酸(DOPAC)含量升高(P<0.05),下丘脑多巴胺(Dopamine)含量升高(P<0.05),肾上腺素(Adr)含量降低;照后2天组海马Adr含量降低(P<0.05),海马5-HT含量升高(P<0.05);照后3天组海马内Adr含量降低(P<0.05)。EMP能够改变大鼠不同脑区神经递质的含量,降低大鼠学习能力  相似文献

The present study was undertaken to determine cerebrospinal fluid (CSF) and brain levels of norepinephrine (NE), serotonin (5-HT) and their metabolites--3,4-dihydroxyphenylacetic acid (DOPAC), 4-hydroxy-3-methoxyphenylacetic acid (HVA) and 5-hydroxyindole-3-acetic acid (5-HIAA)--in rats pretreated with 6-hydroxydopamine (6-OHDA) or 5,7-dihydroxytryptamine (5,7-DHT). In the 6-OHDA pretreated rats, both CSF and brain concentrations of NE, DOPAC and HVA sustained significant decreases as compared with those in non-treated rats. Positive and significant correlations between CSF and brain levels were observed in respect to NE, DOPAC and HVA. In 5,7-DHT pretreated rats, both CSF and brain concentrations of 5-HT and 5-HIAA were significantly decreased. A positive and significant correlation between CSF and brain levels in respect to 5-HT and 5-HIAA was observed. Further studies were carried out to determine ACh levels of both the CSF and the brain in microspheres (MS)-treated rats, which are used as a model of microembolization. The CSF ACh concentrations in MS-treated groups were significantly decreased as compared with those in non-treated rats. The brain ACh contents also tended to decrease in this group. A positive and significant correlation was observed between CSF and brain levels of ACh. These findings suggest that NE, 5-HT and ACh concentrations in the CSF are direct indications of central noradrenergic, serotonergic and cholinergic nerve activity, respectively.  相似文献

为探讨贯叶连翘对慢性应激大鼠生长和脑单胺类神经递质的影响,用15只大鼠设置对照组、应激组和贯叶连翘组3组实验。应激组和贯叶连翘组均进行7天的应激刺激后,贯叶连翘组灌胃贯叶连翘10d。实验结束后,取3组大鼠的脑组织,用高效液相色谱法测定高香草酸(HVA)、去甲肾上腺素(NE)、多巴胺(DA)和5-羟色胺(5-HT)的含量。结果表明,应激组大鼠日增重明显低于对照组;而贯叶连翘组大鼠的日增重明显高于应激组。应激组大鼠海马、纹状体和前额叶中的HVA、NE、DA和5-HT与对照组间均无显著差异。贯叶连翘组大鼠纹状体中的DA含量明显高于应激组;而前额叶中的DA则明显低于应激组。因此,贯叶连翘对慢性应激引起的大鼠生长受抑有缓解作用,对其脑内单胺类神经递质有部分调节作用。  相似文献

Hippocampal cholinergic neuronal activity is reported to be regulated, at least partly, through serotonin1A (5-HT1A) receptors. Chronic lithium treatment has been shown to alter both behavioral and neurochemical responses mediated by postsynaptic 5-HT1A receptors. We investigated whether long-term lithium treatment affects central cholinergic neurotransmission through 5-HT1A receptor-mediated pathways. Changes in acetylcholine (ACh) release induced by 8-hydroxy-2-(di-n-propylamino)tetralin (8-OH-DPAT), a 5-HT1A receptor agonist, in the rat hippocampus were measured using a microdialysis technique and a radioimmunoassay for ACh. Administration of lithium for 21 days resulted in a serum lithium concentration of 1.03 mM and caused little change in density or affinity of [3H]8-OH-DPAT binding sites in the hippocampus. The local application of 8-OH-DPAT into the hippocampus of lithium treated rats increased the ACh efflux in both the absence and the presence of physostigmine, a cholinesterase (ChE) inhibitor, in the perfusion fluid. The basal ACh efflux of lithium treated rats was not different from that of the control rats under normal conditions, but was significantly higher than that of the controls when ChE was inhibited. These results demonstrate that chronic lithium treatment increases spontaneous ACh release in the hippocampus under conditions of ChE inhibition, but not under normal conditions, and enhances cholinergic neurotransmission through 5-HT1A receptor-mediated pathways, and suggest that activation of 5-HT1A receptor function by lithium is related to the enhancement of hippocampal cholinergic neurotransmission.  相似文献

目的通过对吗啡诱导的躯体依赖与精神依赖两种大鼠模型脑内单胺类递质水平的比较,探讨其在吗啡依赖形成中的作用。方法采用剂量递增法复制吗啡依赖大鼠模型,然后用纳洛酮催促,引起躯体戒断症状。连续给予吗啡(5mg/kg,ip)6d,引起大鼠产生显著的条件性位置偏爱效应。脑组织去甲肾上腺素(NE)、5-羟色胺(5-HT)和多巴胺(DA)含量采用荧光分光光度法测定。结果吗啡依赖大鼠催促戒断后脑内NE和5-HT水平明显升高,DA水平下降。吗啡在引起大鼠明显位置偏爱的同时,使大鼠脑内DA和5-HT水平显著升高,NE无明显改变。结论吗啡依赖的形成和戒断与脑内单胺神经递质有密切关系,吗啡依赖的躯体戒断症状与NE升高有关,而吗啡诱导的精神依赖则与脑内DA水平升高有关。  相似文献

Ageing is associated with structural and functional alterations of the vasculature. The nature of age-related vascular disorders is not completely understood. Oxidative stress is hypothesized to play a crucial role in the pathophysiology of vascular complications. We investigated the effects of chronic treatment with the superoxide dismutase mimetic tempol (4-hydroxy-2,2,6,6-tetramethyl piperidinoxyl) on vascular function in the mesenteric vasculature of aged rats. Young (3 weeks) and old (40 weeks) Sprague-Dawley rats were treated with tempol (1 mM in drinking water) or vehicle for 3 weeks. Arterial blood pressure was slightly, but significantly, higher in old than in young rats. Tempol had no effect on arterial blood pressure. The vasoconstrictor responses to norepinephrine (NE) and serotonin (5-HT) were exaggerated in the mesenteric vascular bed (MVB) removed from old rats. Vasodilator responses to acetylcholine (ACh), papaverine (PPV), and isoprenaline (ISO) were reduced in the MVB of old rats in comparison with young rats. Chronic treatment of old rats with tempol normalized their responses to NE and 5-HT. The dilator responses to ACh, PPV, and ISO were similar between old rats receiving tempol and young rats. The present findings suggest that oxidative stress contributes to vascular dysfunction in the mesentery of old rats. The vasculoprotective effects of tempol remain to be elucidated.  相似文献
