Bog peat soils have been accumulating at Wellington Plain peatland, Victoria, Australia for the last 3300 years. Now, dried peat soils are common adjacent to bog peats. The 14C basal age of dried peat is not different from the 14C basal age of bog peat, which supports the theory that dried peat formed from bog peat. A novel application of 210Pb dating links the timing of this change with the introduction of livestock to Wellington Plain in the mid‐1800s. Physical loss of material appears to have been the dominant process removing material as bog peats drained to form dried peats, as indicated by the mass balances of carbon and lead. This research has implications for the post‐fire and post‐grazing restoration of bogs in Victoria's Alpine National Park, and the contribution of peat soils to Australia's carbon emissions.  相似文献

Use of molecular techniques to study plant systematics has been restricted to either largely temperate groups or groups from which seed is readily obtained. Dried material has advantages over other methods of preservation for molecular analyses; it is cheap, easily undertaken, overcomes the difficulties associated with seed recalcitrance and the absence of seeding plants in the field, and most taxonomists are familiar with the method. Three drying techniques were assessed using material collected in the tropics. No differences in either the quantity or the quality of DNA extracted from material dried by these methods was detected. The implications of using dried material in molecular analyses are discussed.  相似文献

Although not laying claim to being the inventor of the light microscope, Antonj van Leeuwenhoek (1632–1723) was arguably the first person to bring this new technological wonder of the age properly to the attention of natural scientists interested in the study of living things (people we might now term ‘biologists’). He was a Dutch draper with no formal scientific training. From using magnifying glasses to observe threads in cloth, he went on to develop over 500 simple single lens microscopes (Baker & Leeuwenhoek 1739 Phil. Trans. 41, 503–519. (doi:10.1098/rstl.1739.0085)) which he used to observe many different biological samples. He communicated his finding to the Royal Society in a series of letters (Leeuwenhoek 1800 The select works of Antony Van Leeuwenhoek, containing his microscopical discoveries in many of the works of nature, vol. 1) including the one republished in this edition of Open Biology. Our review here begins with the work of van Leeuwenhoek before summarizing the key developments over the last ca 300 years, which has seen the light microscope evolve from a simple single lens device of van Leeuwenhoek''s day into an instrument capable of observing the dynamics of single biological molecules inside living cells, and to tracking every cell nucleus in the development of whole embryos and plants.  相似文献

A saturated solution of SbCl3 in 60% HClO4 was used to detect steroids in plant tissue, using (a) cut sections of fresh material (b) dried and powdered material and (c) dried aqueous ethanolic extracts of microquantities of tissues. The utility of this reagent as a microchemical test for diosgenin and yamogenin has been established by application to 55 plant specimens from 35 species in 12 families, including plant parts which hold steroidal sapogenin (spirostans) as well as those that do not. The presence or absence of spirostans in the plant parts was confirmed by infrared analysis after processing the material on a macro scale. Colours obtained with the reagent and pure sapogenins, some of their common derivatives and their glycosides are given for reference.  相似文献

The photoelectric response of a detector using dried bacteriorhodopsin (bR) film as the light sensing material is mathematically modeled and experimentally verified in this paper. The photocycle and proton transfer kinetics of dried bR film differ dramatically from the more commonly studied aqueous bR material because of the dehydration process. The photoelectric response of the dried film is generated by charge displacement and recombination instead of transferring a proton from the cytoplasmic side to the extracellular side of the cell membrane. In this work, the wild-type bR samples are electrophoretically deposited onto an indium tin oxide (ITO) electrode to construct a simple multiple layered photo-detector with high sensitivity to small changes in incident illumination. The light absorption characteristics of the thin bR film are mathematically represented using the kinetics of the bR photocycle and the charge displacement theorem. An electrically equivalent RC circuit is used to describe the intrinsic photoelectric properties of the film and external measurement circuitry to analyze the detector's response characteristics. Simulated studies and experimental results show that the resistance of the dried bR film is in the order of 10(11) Omega. When the input impedance of the measurement circuitry is one order of magnitude smaller than the dried film, the detector exhibits a strong differential response to the original time-varying light signal. An analytical solution of the equivalent circuit also reveals that the resistance and capacitance values exhibited by the dried bR film, in the absence of incident light, are almost twice as large as the values obtained while the material is under direct illumination. Experimental observations and a predictive model both support the notion that dried bR film can be used in simple highly sensitive photo-detector designs.  相似文献

A small amount (0.5 mg) of isolated membrane fragments of rabbit cardiac muscle was dried on Corning cover glasses at 4°C under reduced pressure. The membrane fragments so dried did not come off the cover glasses during incubation in reaction mixtures and subsequent washing. The Mg2+- and Na+-K+-activated adenosine triphosphatase (ATPase) activities of dried membrane fragments were similar and comparable to those of original membrane fragments before drying. Furthermore, the specific binding of ouabain to phosphorylated intermediate forms of Na+-K+-activated ATPase and the ATP-dependent 24Na binding to the membrane were found to occur in dried membrane fragments. Retention of these vital characteristics of cell membrane, the requirement of small quantities of membrane material, and an advantage of instantaneous removal of membrane fragments from reaction mixtures make this preparation uniquely suited for certain kinds of investigations on the cellular membrane.  相似文献

Summary The production of mineralized carbon and nitrogen by a slightly acid sandy loam soil, in the presence and absence of finely chopped fresh plant material or powdered dry plant material, was followed by determination of the amounts of carbon and nitrogen mineralized at intervals during continuous incubation over a period of twelve weeks. Mineralization of carbon and nitrogen was also followed in parallel soil samples and soil plant material mixtures which were dried at 35°C or 105°C and then rewetted every two weeks during the incubation period.The amounts of carbon and nitrogen mineralized were determined at intervals during the incubation period.Mineralization of the carbon and nitrogen of the humus of the soil was stimulated by periodic drying of the soil and particularly when the soil was dried at 105°C.It was found that more mineral nitrogen was produced from fresh plant material than from dried plant material in all the treatments. Periodic drying of the soil-plant material mixtures did not stimulate the production of mineral nitrogen from the added plant material and reduced it considerably when the drying was carried out at 105°C.Periodic drying at 35°C did not stimulate the mineralization of the carbon of fresh or dried plant material. It is clear therefore that, at temperatures occurring in nature, it is unlikely that the decomposition of plant material added to the soil will be stimulated as a consequence of drying of the soil. Periodic drying of the soil-plant material mixture at 105°C increased the mineralization of the carbon of the dried plant material. The amounts of carbon mineralized in 12 weeks from the dried plant material did not, however, exceed the amounts from fresh plant material incubated continuously in fresh soil or in soil periodically dried at 35°C.  相似文献

The increasing importance of natural beta-carotene in fighting xerophthalmia and cancer has given special importance to algal sources of beta-carotene. The susceptibility to quick degradation of this valuable nutrient in oxygen atmosphere, light or heat calls for specific attention to processing and storage practices. In the case of Spirulina it was found that initial losses of beta-carotene on spray drying were between 7 and 10%. On storage in coloured bottles containing air, more than 50% was lost in less than 45 days. The particle size of the dried material seems to have an influence. Flakes (about 20 mesh+) retained 52% of the original beta-carotene level while the spray-dried fine powder (100 mesh-), retained only 34% of the original level. This is explainable in terms of surface area available for active reaction which is higher in the powder than in flakes. This questions the suitability of using spray drying for Spirulina drying. In this paper, data will be presented to substantiate the behaviour of beta-carotene on drying and storage by various methods.  相似文献

Environmental DNA analysis (eDNA) has revolutionized the field of biomonitoring in the past years. Various sources have been shown to contain eDNA of diverse organisms, for example, water, soil, gut content and plant surfaces. Here we show that dried plant material is a highly promising source for arthropod community eDNA. We designed a metabarcoding assay to enrich diverse arthropod communities while preventing amplification of plant DNA. Using this assay, we analysed various commercially produced teas and herbs. These samples recovered ecologically and taxonomically diverse arthropod communities, a total of over a thousand species in more than 20 orders, many of them specific to their host plant and its geographical origin. Atypically for eDNA, arthropod DNA in dried plants shows very high temporal stability, opening up plant archives as a source for historical arthropod eDNA. Considering these results, dried plant material appears excellently suited as a novel tool to monitor arthropods and arthropod–plant interactions, detect agricultural pests and identify the geographical origin of imported plant material. The simplicity of our approach and the ability to detect highly diverse arthropod communities from all over the world in tea bags also highlights its utility for outreach purposes and to raise awareness about biodiversity.  相似文献

Summary Lipases were investigated with respect to their ability to catalyse the incorporation of fatty acids into phosphatidylcholine (PC) by interesterification reactions. The enzymes were dried onto solid support materials and the conversions were carried out in water-saturated toluene. Three lipases (two fungal and one plant enzyme) had the desired activity; immobilized lipase from Mucor miehei (Lipozyme) was the most active enzyme. The Lipozyme-catalysed interesterification was selective for the sn-1 position of PC and during 48 h of reaction around 50% of the fatty acids in this position were replaced with heptadecanoic acid, a fatty acid which was practically absent in the original phospholipid. Due to adsorption on the support material and the competing hydrolysis reaction the total amount of PC in the reaction solution decreased to about 40% of the original amount. Gomori's original aldehyde-fuchsin method has been modified by the combination of Halmi's counter stain with Gabe's preparation, consisting of basic fuchsin, 1 gm; boiling water, 200 ml; with HC1, 2 ml and paraldehyde, 2 ml added after cooling and filtering. The solution so made was allowed to ripen 3-4 days at room temperature, and the precipitate which formed was filtered off and dried at 55-60°C. The staining solution consisted of 0.5 gm of the dry precipitate dissolved in 100 ml of 70% alcohol. The staining follows original procedures except that it is very important to bring slides from water to 70% alcohol before placing them in the aldehyde-fuchsin solution and also to remove all excess staining solution by rinsing in 95% alcohol after staining. The staining solution is stable for at least 6 mo.  相似文献

Summary Factors affecting the release of sulphate from a number of eastern Australian soils were studied.All of the soils released sulphate when dried. The amounts released were influenced by the manner in which the soil was dried. Air-drying in the laboratory at 20°C released least sulphate.Sulphate was mineralized in all soils by incubation at 30°C but the amounts mineralized could not be related to soil type or any single soil property. The ratio of nitrogen mineralized: sulphur mineralized varied widely between soils and was generally appreciably greater than the ratio of total nitrogen: organic sulphur in the soils.A rapid flush of mineralization of both sulphur and nitrogen took place when some of the soils were rewetted and incubated after they had been dried in the laboratory and stored for 4 to 5 months. Following this, the rate of mineralization was similar to that in the original undried soil. During this flush, the enhancement of sulphur mineralization was relatively greater than that of nitrogen so that the ratio of nitrogen mineralized: sulphur mineralized was considerably smaller than that during later phases of the incubation or that of the original moist soil. Soils collected after they had remained dry in the field for a similar period of time did not show this type of mineralization although they had initially done so when collected moist and air-dried in the laboratory.The effects of temperature, soil moisture, toluene and formaldehyde, and the addition of calcium carbonate to soils on the mineralization of sulphur were similar to their effects on the mineralization of nitrogen.  相似文献

A method for the dry-preservation of fixed plant material, root tips and buds, is described. The method seems to be advantageous on long expeditions and when material has to be sent away.

The material is transferred from the fixative to 70% alcohol (3 changes, 1/2 hour in the last). It is dried on blotting-paper. The dried material may be preserved a long time. Material kept dry for 4 1/2 years has proved to be quite satisfactory. Drying has been tried after fixation with CRAF-solutions (Webber and Randolph modifications) and fixatives containing osmic acid (Fleming-Benda and 2BD).

The dry material is swollen by keeping for 2 days in 10% alcohol. It is embedded in paraffin according to the usual method. A satisfactory staining has been obtained after these fixatives using iodine-gentian-violet and Feulgen stainings. In addition to chromosome counts dry material may be used for chromosome morphology studies.

Dried material fixed in aceto-alcohol (1:3) has not turned out to be specially suitable for squash preparations owing to the fragility of the chromosomes. If strong pressure is not applied, satisfactory results may, however, be obtained.  相似文献

The PVC superphylum consists of the core phyla Planctomycetes, Verrucomicrobia and Chlamydiae, together with additional ones. Historically, the cell plan of PVC bacteria has been interpreted as an ‘exception’ to the classical Gram-negative (Gneg) one (Fuerst Antonie van Leeuwenhoek 104:451–466, 2013). However recent genomic and electron-microscopy data have argued against this exceptional status and suggested the need for a reinterpretation of the data in a more classical framework. In this perspective, I evaluate the arguments that have recently been presented by Fuerst as supporting the PVC cell plan as an ‘exception’ and present an alternative interpretation that is based on proposed evolutionary events that may have shaped the PVC genomes and proteomes. This interpretation supports the alternative proposal that the PVC cell plan is derived from a Gneg one.  相似文献

Two human ring chromosomes 18 were isolated intact in a Langmuir trough, critical-point dried, and studied by quantitative electron microscopy. One of the fundamental discoveries of W. Arnold was the detection of thermally stimulated light emission from preilluminated photosynthetic material (Arnold and Sherwood (1957) Proc Natl Acad Sci USA 43: 105–114). This phenomenon, called thermoluminescence (TL), is characteristic of a wide range of materials (semiconductors, minerals, inorganic and organic crystals, and complex biological systems such as the photosynthetic apparatus) which share the common ability of storing radiant energy in thermally stabilized trap states.The original discovery of TL in dried chloroplasts later proved to be a phenomenon common to all photosynthetic organisms: photosynthetic bacteria, cyanobacteria, algae and higher plants. Following the pioneering work of Arnold, considerable effort has been devoted to identification and characterization of photosynthetic TL components. This work has firmly established the participation of various redox states of the water-oxidizing complex and the quinone electron acceptors of Photosystem II in the generation of photosynthetic glow curves. Since TL characteristics are very sensitive to subtle changes in redox properties of the involved electron transport components, the TL method has become a powerful tool in probing a wide range of PS II redox reactions.
