研究了从土壤中筛选产脂肪酶的菌株,利用紫外线、快中子、快中子和磁场复合、γ射线、γ射线和磁场复合诱变,以酶活为筛子进行诱变育种。结果,出发菌酶活较低的一株得到了一株酶活为396.22U/mL的诱变株,此酶活比出发菌株高92倍,并发现此菌对紫外线和快中子比较敏感;而出发菌酶活较高的一株得到了酶活为424.60U/mL发酵液的诱变株,此酶活为出发菌株3.0倍。在此基础上,初步探讨了快中子、γ射线及磁场复合处理在产脂肪酸菌种诱变中的作用,并认为,在产脂肪酸菌株的诱变中快中子诱变更为有效。  相似文献

本实验以妥布霉素产生菌黑暗链霉菌(S.tenebrarius)ATCC17920为出发菌株,经紫外线处理,获得一株产量较高且稳定的菌株UV-59,其抗生素效价比原株提高92.3%对UV_59菌株进行高温处理,得到一株T-541菌株,所产抗生素只有两个组分,比原出发株减少一个组份,不再含有出发株产生的氨甲酸卡那霉素;再对T-541菌株进行原生质体制备,分别用紫外线和紫外线加氯化锂以及亚硝基胍诱变处理原生质体,获得四株高产菌株,效价比原株提高115~150%,且稳定。此外还研究了变异菌株的形态与产量的关系。  相似文献

曲酸发酵的研究I.紫外线照射对黄曲霉产生曲酸的影响   总被引:2,自引:1,他引:1  
用曲酸生产菌株黄曲霉 Aspergtllus flavus Link AS 3.2789为出发菌株进行紫外线诱变,从73个突变株中选出UI 1202,产曲酸达34.7毫克/毫升,比原始菌株提高50.8%。再以UI 1202进行第二次紫外线照射,从183个突变株中选出UII 1223,产曲酸达46.5毫克/毫升,比原始菌株提高78.8%。说明以紫外线为诱变剂,可以提高曲酸产量,并有积累诱变效果。将UII 1223在5000升发酵罐进行工业水平生产[实际装置4000升,搅拌速度160转/分,通风量每分钟1:0.5(体积:体积),温度31--33℃,发酵120小时左右],说明经二次紫外线诱变后的突变株性能比原始菌株有不少优越性,不仅提高了曲酸产量,而且性能稳定,不产毒素,降低了原料消耗,节约生产时间,已在生产中正式使用。  相似文献

紫外线与亚硝酸钠复合诱变选育L-组氨酸产生菌   总被引:1,自引:0,他引:1  
以1株谷氨酸棒杆菌(Corynebacterium glutamicum)S_6作为出发菌株,利用亚硝酸钠(NaNO_2)、紫外线(UV)进行诱变,通过实验证明亚硝酸钠诱变时间在180 s后致死率达到80%,紫外线在照射30 s后致死率达到80%。诱变后的突变菌株经6-巯基嘌呤结构类似物的抗性平板筛选,最终筛得3株菌,Y_1产L-组氨酸量达到331 mg/L,比出发菌株S_6高了5.08%,Z_2产L-组氨酸量达到325 mg/L,比出发菌株S_6高了1.9%,F_6产L-组氨酸量达到330 mg/L,比出发菌株S_6高了7.14%。结果显示,经紫外线与亚硝酸钠复合诱变后的菌株F_6产L-组氨酸的产量最高,比亚硝酸钠诱变后的菌株产L-组氨酸量提高2.06%,比紫外线诱变后的菌株产L-组氨酸量提高5.24%。  相似文献

以树状黄杆菌(Flavobacteriumaraborescens)NRRL11022为出发菌株,用紫外线对其进行诱变,经筛选得到一株葡萄糖异构酶的高产菌株U-616,其酶活力提高31%。经保存三年和多次传代复测,其产酶能力保持稳定。其生长和产酶需较高的溶氧水平,最适产酶温度为30℃,最适产酶pH为7.0-7.5,铁离子对其生长和产酶无明显的影响。所产葡萄糖异构酶的最适温度为60-80℃,最适pH为7.5-8.5,Co2+和Mg2+对酶有激活作用,对金属离子耐受性较强,对Ca2+不敏感,热稳定性较好。树状黄杆菌变株U-616是一株产胞内葡萄糖异构酶的优良菌株。  相似文献

以黄曲霉(Aspergillus flavus.)为出发菌株,经3次紫外线、1次^60Co、3次亚硝基胍多重复合诱变处理,选育获得曲酸生产菌UCN7—17,配以最佳培养条件,发酵7d,曲酸产量由原来的0.926%,提高到6.3%。实验证明采用多因子复合诱变,能有效改变菌株对诱变因素敏感性,提高变异率,逐步提高突变株的产酸水平。  相似文献

通过紫外线—高温复合诱变处理衣康酸生产菌株土曲霉构Aspergillus terreus As3.2811,用以琥珀酸为唯一碳源的选择性乎板定向筛选高产菌株,获得产酸率较其亲株提高了5倍以上的突变株。用正交试验的方法对突变株的适宜产酸条件进行了研究,通过分批补糖发酵可提高其产酸率高达39.92%。  相似文献

以耐高渗酵母为出发菌株,采用紫外线诱变处理,得到了一株变异株C1。其摇瓶发酵产赤藓糖醇24.9g/L左右。通过摇瓶发酵试验,研究了菌株C1的遗传稳定性。研究结果表明,经多次传代实验,菌株C1的发酵产糖醇能力没有改变,表明其是稳定的变异株。赤藓糖醇的产量由诱变前的12、594g/L提高到24.943g/L,提高了98.1%。关键词:  相似文献

本文采用原生质紫外诱变方法对生香酵母进行诱变处理,并利用重氮染色法检测突变株的酯酶活性,成功地选育出两株高产酯生香酵母FU4和FU7,实验室发酵实验结果表明,FU4和FU7菌最大产酯量分别为出发菌株的1.6倍和1.5倍,且产物风味好,具有一定的应用价值。  相似文献

Coenzyme Q10 (CoQ10) is an industrially important molecule having nutraceutical and cosmeceutical applications. CoQ10 is mainly produced by microbial fermentation and the process demands the use of strains with high productivity and yields of CoQ10. During strain improvement program consisting of sequential induced mutagenesis, rational selection and screening process, a mutant strain UF16 was generated from Sporidiobolus johnsonii ATCC 20490 with 2.3-fold improvements in CoQ10 content. EMS and UV rays were used as mutagenic agents for generating UF16 and it was rationally selected based on atorvastatin resistance as well as survival at free radicals exposure. We investigated the genotypic and phenotypic changes in UF16 in order to differentiate it from wild type strain. Morphologically it was distinct due to reduced pigmentation of colony, reduced cell size and significant reduction in mycelial growth forms with abundance of yeast forms. At molecular level, UF16 was differentiated based on PCR fingerprinting method of RAPD as well as large and small-subunit rRNA gene sequences. Rapid molecular technique of RAPD analysis using six primers showed 34 % polymorphic fragments with mean genetic distance of 0.235. The partial sequences of rRNA-gene revealed few mutation sites on nucleotide base pairs. However, the mutations detected on rRNA gene of UF16 were less than 1 % of total base pairs and its sequence showed 99 % homology with the wild type strain. These mutations in UF16 could not be linked to phenotypic or genotypic changes on CoQ10 biosynthetic pathway that resulted in improved yield. Hence, investigating the mutations responsible for deregulation of CoQ10 pathway is essential to understand the cause of overproduction in UF16. Phylogenetic analysis based on RAPD bands and rRNA gene sequences coupled with morphological variations, exhibited the novelty of mutant UF16 having potential for improved CoQ10 production.  相似文献

Improved production of citric acid by a diploid strain of Aspergillus niger   总被引:1,自引:0,他引:1  
Aspergillus niger strain CGU 87 was treated with UV radiation and some auxotrophic mutants were obtained. These mutants were less productive than CGU 87, which produced an average of 7.4% citric acid. All possible crosses in pair wise combinations were carried out between these auxotrophs, and three heterokaryons were synthesised. Finally, one heterozygous diploid was isolated from each of them. These heterokaryons and diploids showed improved productivity when compared with their component parents, but except in one diploid D5, all others produced less citric acid than CGU 87. The yield of D5 exceeded that of CGU 87 by 1.2 times and it produced 9% citric acid. This is a significant improvement and the increased productivity seems to be the result of successful adaptation of D5 to its fermentation environment.  相似文献

AIMS: To isolate a bacterium that produces high yield of melanin and to examine the effect of this bacterial pigment on the efficacy of a bioinsecticide. METHODS AND RESULTS: A novel melanin-producing bacterium, designated as strain WS, was isolated from the East Lake, Wuhan, China. Taxonomic studies of this strain indicate that it belongs to Aeromonas media. Physicochemical analysis of the pigment produced by strain WS (melanin WS) suggests that it is the authentic 3,4-dihydroxyphenylalanine (DOPA)-melanin. This melanin and that produced by Pseudomonas maltophilia P28 (melanin P28) share many biophysical properties, but the yield of the melanin WS is significantly higher than that of the melanin P28. In addition, the melanin WS appears to be more effective in the protection of a bioinsecticide against ultraviolet (UV) or solar radiation. At the concentration of 10 ppm, the melanin P28 exhibited no photoprotective effect on the bioinsecticide against UV radiation; in contrast, 5 ppm of melanin WS displayed an obvious protective effect. Similarly, the melanin WS displayed more protective effect on the bioinsecticide against solar radiation than the melanin P28 did over a 4-day period, with the effect being more dramatic for the last 2 days. CONCLUSIONS: We have isolated a novel bacterial strain of A. media that produces high levels of melanin. The melanin produced by this strain offers effective photoprotection of a commercial bioinsecticide BTI against UV and solar radiation. SIGNIFICANCE AND IMPACT OF THE STUDY: Our study suggests that the melanin produced by our newly isolated A. media strain has the potential to be used as a general photoprotective agent for bioinsecticides.  相似文献

以紫杉醇产生菌NCEU_1为出发菌株,分别采用紫外线和紫外线与氯化锂复合诱变方法对其原生质体进行诱变,获得了两株高产紫杉醇的突变株———UV4 0 - 1 9和UL50 - 6 ,其紫杉醇产量从出发菌株的314 0 7μg L分别提高至376 38μg L和392 6 3μg L ;同时,又采用RAPD和同工酶技术对出发菌株NCEU_1与两高产株UV4 0 - 1 9和UL50 - 6 间的遗传差异进行了研究。结果表明,出发菌株与诱变菌株之间以及两诱变菌株之间都存在明显差异,为进一步研究与紫杉醇合成相关基因及诱变株产量提高的分子机制奠定了基础  相似文献

The effect of low doses of ionizing and nonionizing radiation on the radiation response of yeast Saccharomyces cerevisiae toward ionizing and nonionizing radiation was studied. The wild-type strain D273-10B on exposure to 54 Gy gamma radiation (resulting in about 10% cell killing) showed enhanced resistance to subsequent exposure to UV radiation. This induced UV resistance increased with the incubation time between the initial gamma radiation stress and the UV irradiation. Exposure to low doses of UV light on the other hand showed no change in gamma or UV radiation response of this strain. The strains carrying a mutation at rad52 behaved in a way similar to the wild type, but with slightly reduced induced response. In contrast to this, the rad3 mutants, defective in excision repair, showed no induced UV resistance. Removal of UV-induced pyrimidine dimers in wild-type yeast DNA after UV irradiation was examined by analyzing the sites recognized by UV endonuclease from Micrococcus luteus. The samples that were exposed to low doses of gamma radiation before UV irradiation were able to repair the pyrimidine dimers more efficiently than the samples in which low gamma irradiation was omitted. The nature of enhanced repair was studied by scoring the frequency of induced gene conversion and reverse mutation at trp and ilv loci respectively in strain D7, which showed similar enhanced UV resistance induced by low-dose gamma irradiation. The induced repair was found to be essentially error-free. These results suggest that irradiation of strain D273-10B with low doses of gamma radiation enhances its capability for excision repair of UV-induced pyrimidine dimers.  相似文献

发酵法生产曲酸还未实现大规模工业化生产的原因之一是曲酸菌种产酸率较低,本文以平展米曲霉(Aspergillus oryzae effusus)AS32为出现菌株,经UV和^60Co诱变处理,筛选获得一株高产曲酸变异株AUR163,以葡萄糖为碳源,酵母膏为氮源,32℃摇瓶和30L罐发酵培养4天,产酸达6.8g/100mL。平均生产效率为17.0g/L.d最高可达30.5g/L.d比出发菌AS32提高190%以上,这表明UV和^60Co作诱剂,可以大幅度提高米曲霉的曲酸生产效率。  相似文献

Aspergillus niger ORS-4.410, a mutant of Aspergillus niger ORS-4 was produced by repeated irradiation with UV rays. Treatments with chemical mutagnes also resulted into mutant strains. The mutants differed from the parent strain morphologically and in gluconic acid production. The relationship between UV treatment dosage, conidial survival and frequency of mutation showed the maximum frequency of positive mutants (25%) was obtained along with a conidial survival of 59% after second stage of UV irradiation. Comparison of gluconic acid production of the parent and mutant ORS-4.410 strain showed a significant increase in gluconic acid production that was 87% higher than the wild type strain. ORS-4.410 strain when transferred every 15 days and monitored for gluconic acid levels for a total period of ten months appeared stable. Mutant ORS-4.410 at 12% substrate concentration resulted into significantly higher i.e. 85-87 and 94-97% yields of gluconic acid under submerged and solid state surface conditions respectively. Further increase in substrate concentration appeared inhibitory. Maximum yield of gluconic acid was obtained after 6 days under submerged condition and decreased on further cultivation. Solid state surface culture condition on the other hand resulted into higher yield after 12 days of cultivation and similar levels of yields continued thereafter.  相似文献

以阿维链霉菌(Streptomyces avermitilis)76-12为出发菌株,采用亚硝基胍、吖啶橙、紫外线和氯化锂分别对其孢子和原生质体进行诱变,经抗代谢物理性筛选,获得一系列高产突变株,其中N-1-2高产突变株的发酵单位是出发菌株的2.47倍。实验中同时获得了只产阿维菌素a组分的突变株G-32、Bla组分含量高的Ave8菌株和产蓝绿色孢子的突变株UA-G等。  相似文献
