Growth of the yeast Pichia pastoris IFP 206 in methanol- and glucose-containing media led respectively to very and weakly flocculent cells. Mannans from both kinds of cells were extracted and compared. Chemical analysis and molecular mass estimation showed some differences between the mannans from very and weakly flocculent cells, especially in quantitative amino acid content. 1H NMR analysis showed that both kinds of mannan contained alpha-1,2 and beta-1,2 linkages. Two acetolysis conditions, combined with 1H NMR analysis, revealed that mannans from both kinds of cells were composed of mannose, mannobiose, mannotriose, mannotetraose and mannopentaose side-chains with the following respective structures: Man; Man alpha 1---2Man; Man alpha 1----2Man alpha 1----2Man; Man beta 1----2Man alpha 1----2Man; Man beta 1----2Man beta 1----2Man alpha 1----2Man; Man alpha 1----2Man beta 1----2Man beta 1----2Man alpha 1----2Man. Additionally the beta-1,2 linkages of the non-reducing terminal residues of the mannotetraose were shown to be acetolysis-labile. The mannans from very flocculent cells were richer in mannopentaose than the mannans from weakly flocculent cells. According to these results, the extended conformations in the branching moieties of the mannan could be the basis of the higher degree of flocculation of the methanol-grown cells.  相似文献

A high molecular weight glycoprotein antigen was isolated by size exclusion chromatography on Sepharose 4B from an extract of the yeast Saccharomyces cerevisiae. The glycoprotein antigen Sc 500 was shown to be identical to the antigen termed gp200 previously isolated (Heelan et al., 1991). The MW of Se 500 was determined to be about 500 kDa by size exclusion chromatography on Superose 6 and 460 kDa ± 20k Da by size-exclusion chromatography/multi-angle laser light scattering (SEC/MALLS). Sc 500 contained 90% mannose and traces of N-acetylglucosamine. The amino acid composition revealed that serine and threonine were the most abundant amino acids of the protein part. By alkaline borohydride treatment some, but not all bonds between protein and carbohydrate were broken. This indicates that the main type of linkage between protein and carbohydrate is O-glycosidic and that a minor type is of N-glycosidic nature. Methylation analysis revealed that the mannose residues were connected by 1 → 2 and 1 → 3 linkages with 1 → 2, 1→ 6 linked branch points.

Purified Sc 500 was subjected to a series of chemical and enzymatic modifications followed by studies of antibody binding activity. Treatments with both periodate and alkaline sodium borohydride reduced the human serum IgA, IgG and monoclonal IgM antibody binding activity of Sc 500 whereas trypsin and pronase did not affect its ability to bind these antibodies. The mannosidase Man1 → 2,3,6Man reduced the IgM binding to Sc 500 while the other enzymes included in this experiment (Man1→2 Man, Manβ1 →4GlcNAc and PNGase F) had no effect on the antibody binding.  相似文献

Co-flocculation between cells of beer yeast IFO 2018, a flocculent strain, and non-flocculent strains was investigated by means of a chemical modification method. Treatment with periodate deprived non-flocculent cells, but not flocculent cells, of the ability to co-flocculate. Treatment with mercaptoethanol or photo-irradiation in the presence of methylene blue deprived flocculent cells, but not non-flocculent cells, of the co-flocculating ability. Mercaptoethanol-treated or photoirradiated flocculent cells (beer yeast IFO 2018) co-flocculated with periodate-treated flocculent cells, but periodate-treated cells subsequently subjected to mercaptoethanol treatment or photoirradiation neither flocculated by themselves nor co-flocculated with other cells. Thus, it is likely that both protein and carbohydrate components of the yeast cell surface play important roles in the mutual recognition and intercellular interaction involved in flocculation. It is strongly suggested that the essential carbohydrate which is widely distributed among Saccharomyces species is the mannan fraction on the cell wall, and that a flocculent yeast strain produces surface protein component(s) which recognize and bind the mannan component of adjacent cells.  相似文献

We studied phosphopeptidomannans (PPMs) of two Saccharomyces cerevisiae NCYC 625 strains (S. diastaticus): a wild type strain grown aerobically, anaerobically, and in the presence of antimycin and a [rho(0)] mutant grown aerobically and anaerobically. The aerobic wild-type cultures were highly flocculent, but all others were weakly flocculent. Ligands implicated in flocculation of mutants or antimycin-treated cells were not aggregated as much by concanavalin A as were those of the wild type. The [rho(0)] mutants and antimycin-treated cells differ from the wild type in PPM composition and invertase, acid phosphatase, and glucoamylase activities. PPMs extracted from different cells differ in the protein but not in the glycosidic moiety. The PPMs were less stable in mitochondrion-deficient cells than in wild-type cells grown aerobically, and this difference may be attributable to defective mitochondrial function during cell wall synthesis. The reduced flocculation of cells grown in the presence of antimycin, under anaerobiosis, or carrying a [rho(0)] mutation may be the consequence of alterations of PPM structures which are the ligands of lectins, both involved in this cell-cell recognition phenomenon. These respiratory chain alterations also affect peripheral, biologically active glycoproteins such as extracellular enzymes and peripheral PPMs.  相似文献

Summary Cell walls from aKluyveromyces marxianus either non flocculent or flocculent strain were isolated and analysed for protein, carbohydrates and phosphate content. Alkaline extract of proteins were analysed by SDS-PAGE. The results revealed a higher protein content in the cell walls from the flocculent strain. Electrophoresis of the cell wall proteins of the flocculent strain showed an extra peptide with an apparent molecular weight of 37 KDa which is absent fom non-flocculent cells. The involvement of this protein in cell adhesion during flocculation is discussed.  相似文献

Abstract The flocculation character in strain IM1-8b of Saccharomyces cerevisiae is controlled by a single and dominant gene shown to be allelic to FLO1 . Such a gene has been both mitotically and meiotically mapped on the right arm of chromosome I at 4.7 cM from PHO11 . The phenotype was suppressed by a single gene of wide distribution among non-flocculent strains (proposed as fsu3 ) that, however, was unable to suppress other FLO1 genes in other flocculent strains.  相似文献

This study deals with the chemical characterization of a capsular polysaccharide (CPS) produced by a thermal biomass largely comprising the cyanobacterium Mastigocladus laminosus. The sugar moiety of this polymer is composed of seven neutral monosaccharides (Rha, Fuc, Ara, Xyl, Man, GIc, Gal) and two uronic acids (GalA, GIcA). Proteins represent 18% of the dry weight of the CPS. Organic acid substituents (acetate, pyruvate, succinate) were also detected and estimated by high-performance liquid chromatography. The presence of sulfate groups (5% w/w) was observed, which represents a relatively rare feature for cyanobacteria. Acidic hydrolysis of the purified polysaccharide led to the isolation of four oligosaccharidic fractions. Flocculation is an eco-friendly process of cell separation, which has been traditionally exploited by the brewing industry. Cell surface charge (CSC), cell surface hydrophobicity (CSH) and the presence of active flocculins, during the growth of two (NCYC 1195 and NCYC 1214) ale brewing flocculent strains, belonging to the NewFlo phenotype, were examined. Ale strains, in exponential phase of growth, were not flocculent and did not present active flocculent lectins on the cell surface; in contrast, the same strains, in stationary phase of growth, were highly flocculent (>98%) and presented a hydrophobicity of approximately three to seven times higher than in exponential phase. No relationship between growth phase, flocculation and CSC was observed. For comparative purposes, a constitutively flocculent strain (S646-1B) and its isogenic non-flocculent strain (S646-8D) were also used. The treatment of ale brewing and S646-1B strains with pronase E originated a loss of flocculation and a strong reduction of CSH; S646-1B pronase E-treated cells displayed a similar CSH as the non-treated S646-8D cells. The treatment of the S646-8D strain with protease did not reduce CSH. In conclusion, the increase of CSH observed at the onset of flocculation of ale strains is a consequence of the presence of flocculins on the yeast cell surface and not the cause of yeast flocculation. CSH and CSC play a minor role in the auto-aggregation of the ale strains since the degree of flocculation is defined, primarily, by the presence of active flocculins on the yeast cell wall.  相似文献

Inonotus levis biomass was extracted with 5% NaOH containing NaBH4, the insoluble material was discarded and the solution dialyzed. It was further treated with proteinase and the polymeric fraction isolated by gel chromatography. It contained mostly a polysaccharide of the following structure:
β-GlcA-(1→2)--Gal-(1→6)--Gal3OMe-(1→[→6)--Gal-(1→6)--Gal3OMe-(1→]5–10
where a non-reducing terminal glucuronic acid residue was present in about half of the molecules, making thus some of the chains acidic and others neutral. Methyl groups were present at about half of the galactose residues, however, no direct proof of the presence of defined repeating units was obtained due to NMR signals overlap. We believe that these short polymeric chains might be originally attached to a protein via serine or threonine residues and were cleaved off due to the alkaline conditions of extraction. Another polymer, co-extracted with this galactan, was a branched phosphorylated mannan with a structure similar to that of the mannan from Saccharomyces cerevisiae yeast.  相似文献

A flocculent strain of Saccharomyces cerevisiae S646-1B accumulated more Cu2+ (81 nmol mg–1 dry wt) than the isogenic (except for the marker genes ade1 and trp1 and the gene FLO1) non-flocculent strain S646-8D (30 nmol mg–1 dry wt), in the first 10 min of contact of the cells with Cu2+. Additionally, this strain flocculated in solutions of 0.2 mM Cu2+, Ni2+, Zn2+ and Cd2+. The potential of using flocculent strains in the bioremediation of heavy metals contaminated waste waters is discussed.  相似文献

乙酸是生物质乙醇发酵过程中酵母细胞面临的重要抑制剂之一,对细胞生长及发酵性能有强烈的抑制作用。增强酵母菌对乙酸胁迫的耐受性对提高乙醇产率具有重要意义。用分别带有完整絮凝基因FLO1及其重复序列单元C发生缺失的衍生基因FLO1c的重组表达质粒分别转化非絮凝型工业酿酒酵母CE6,获得絮凝型重组酵母菌株6-AF1和6-AF1c。同时以空载体p YCPGA1转化CE6的菌株CE6-V为对照菌株。与CE6-V相比,絮凝酵母明显提高了对乙酸胁迫的耐受性。在0.6%(V/V)乙酸胁迫下,6-AF1和6-AF1c的乙醇产率分别为对照菌株CE6-V的1.56倍和1.62倍;在1.0%(V/V)乙酸胁迫下,6-AF1和6-AF1c的乙醇产率分别为对照菌株CE6-V的1.21倍和1.78倍。可见絮凝能力改造能明显提高工业酿酒酵母的乙酸胁迫耐受性及发酵性能,而且FLO1内重复序列单元C缺失具有更加明显的效果。  相似文献

Abstract A non-flocculent strain of Saccharomyces cerevisiae was selected after EMS mutation of a flocculent and heterozygous FLO1 locus diploid. The analysis of 25 asci from this diploid showed in all cases segregation 0F:4NF, thus confirming that it was probably affected in the desired gene. After sporulation and dissection of asci, three haploid strains were chosen, which were altered in the locus FLO1 . Crossing these three strains with two other ones having markers for ADE1 and pho11::LEU2 , we could map the mutation at ca. 4.3 cM and ca. 37.7 cM from the PHO11 and ADE1 loci respectively.  相似文献

During cultivation of a flocculent yeast, Saccharomyces cerevisiae 1001, two cell fractions, flocs and free cells, appeared in the medium. Free cells contained cells with a normal ability to flocculate, less flocculent cells and not-flocculent cells. When the non-flocculent cells and not-flocculent cells. When the non-flocculent cell fraction from the postexponential phase of growth was collected and used as an inoculum, the culture showed synchronous growth. The floc forming ability of the yeast cells from this culture increased gradually with the number of divisions.  相似文献

The lipopolysaccharide (LPS) of a galU mutant of Pseudomonas aeruginosa PA103, a serogroup O11 strain, was sequentially extracted with phenol–chloroform–petroleum ether (PCP) followed by hot phenol–water extraction of the bacterial pellet remaining after PCP extraction. LPS was found in both the PCP extract as well as in the water phase of the hot phenol–water extract. Analysis of the carbohydrate portion released by mild acid hydrolysis of both LPS preparations, both before and after removal of all phosphate groups by treatment with aqueous HF, was performed by glycosyl composition and linkage analyses as well as by NMR and mass spectrometric analyses. The results showed that the carbohydrate portion of these two LPS extracts contained the same structure: namely, -GalN(Ala)-(1→3)--(7-Cm)HepII-(1→3)--HepI-(1→5)--Kdo-(2→. The oligosaccharide preparation from PCP-extracted LPS consisted of a variety of structures containing up to six phosphate groups present as mono-, pyro-, and possibly triphosphate, primarily located on the HepI residue with some molecules having a monophosphate on HepII. The oligosaccharide preparation from the hot phenol–water-extracted LPS contained a similar variety of structures, but with an additional structure in which HepI contained a PPEA group at O-2. In addition, PAGE immunoblot analysis of the crude cellular extract with anti-A-antibodies revealed the presence of A-band material in both PA103 and the galU mutant. The A-band material was purified and characterized by glycosyl composition and linkage analyses, as well as by NMR spectroscopy, which confirmed that the A-band rhamnan polysaccharide was present but not as typical LPS since lipid-A or LPS core oligosaccharide components were not detected.  相似文献

Structural data are presented on six oligosaccharide-fractions (hexa- to octa-saccharides) released from sheep -gastric-glycoproteins having blood-group I and i activity by degradation with alkaline borohydride. Previous data on two of the oligosaccharides are included for comparison. The fractions were analysed, before and after treatment with exo-β- -glycosidases and an endo-β- -galactosidase, on Bio-Gel P4 and by p.c., by direct-insertion m.s. (after methylation), and by g.l.c.—m.s. of the derived, partially O-methylated alditol acetates. Each fraction contained 1–3 oligosaccharides, each of which had 2-acetamido-2-deoxy- -galactitol (GalNAc-ol) at the reduced end and involved one of the structures
The majority of the oligosaccharides contained the unsubstituted “type 2” blood group precuisor-chain sequences, β- -Gal-(1→4)-β- -GlcNAc-(1→6) and single or repeating β- -Gal-(1→4)-β- -GlcNAc-(1→3), which are recognised by various anti-blood-group I and i cold agglutinins. The “type 1” sequence, β- -Gal-(1→3)-β-blood-group Ii activities, the following structural model can be proposed, which consists of (a) a core region; (b) a backbone region having (1→3)- and (1→6)-linked N-acetyl-lactosamine [β-Gal-(1→4)-GlcNAc] branches with I activities, and linear, repeating, (1→3)-linked N-acetyl-lactosamine units with i activities; and (c) a peripheral region with blood-group isotype activities.

Pectins were extracted either by water from extruded lemon fibre or by hot acid from the raw lemon fibre. The amount of water-soluble polysaccharides from lemon plant cell walls was greatly increased after extrusion-cooking. The pectins obtained by extraction with water from the extruded fibre and the pectins extracted from the raw material by hot acid were studied. The water-soluble pectins obtained after extrusion-cooking had the distinctive feature of being very highly (92%) methylated; they were also particularly rich in arabinose side-chains. High molecular weight material coming from the “hairy” regions was isolated after digestion by an endo-polygalacturonase. Methylation analysis revealed the presence in both pectins of fairly branched (1 → 5)-linked arabinans and arabinogalactans of type I and II side-chains.  相似文献

In higher plants, mannans are found as dominant reserve material in the endosperm of Arecaceae seeds and also in some species from Apiaceae, Rubiaceae and Asteraceae. A linear β(1 → 4)-d-mannan was now isolated from the endosperm of Schizolobium parahybae, family Caesalpiniaceae, a native of Southern Brazil. Its seeds were germinated and the consumption of polysaccharides from the endosperm, namely galactomannans and β(1 → 4)-d-mannan, was analysed at differents stages of germination. At the 6th day after germination no residual 3:1 Man:Gal galactomannan was found, indicating that complete degradation of galactomannan had been reached. However, after 12 days of germination, the mannan was recovered from the remaining endosperm. Its presence in the endosperm after germination demonstrated that it is not a reserve material as described for seeds of other species.  相似文献

Oligomeric procyanidins were isolated from the leaves and flowers of hawthorn (Crataegus laevigata). A trimer, epicatechin-(4β→8)-epicatechin-(4β→6)-epicatechin, and a pentamer consisting of (−)-epicatechin units linked through C-4β/C-8 bonds have been isolated from hawthorn for the first time, in addition to known procyanidins including dimers B-2, B-4 and B-5, trimers C-1 and epicatechin-(4β→6)-epicatechin-(4β→8)-epicatechin, and tetramer D-1.
