The analysis regarded 304 strains of Acinetobacter genus isolated from various diagnostic materials, objects from hospital environment and from non-hospital sources (soil, water, various animals). Applying API ZONE system, five species were isolated: Acinetobacter juni, (18.42%), Acinetobacter baumanii (70.39%), Acinetobacter haemolyticus (5.59%), Acinetobacter lwoffii (4.6%) and Acinetobacter johnsonii (0.99%). Most frequently isolated species were present in purulent materials and in samples from respiratory tract infections and urinary tract infections. Over 47% Acinetobacter species strains were present in clinical material as single aerobic bacteria.  相似文献

Contribution of Acinetobacter genus in the degradation of atrazine and its analogs is reported here. An interesting bacterial isolate capable of degrading atrazine as high as 250 ppm was isolated from a soil heavily contaminated with atrazine. The permissible level of atrazine in drinking water is 3 ppb and hence use of a strain capable of atrazine degradation as high as 250 ppm would be of immense help for rapid environmental cleanup. This isolate was found to be capable of best growth at 37 degrees C and at pH inclined towards the alkaline side. It was found that atrazine was utilized as a carbon and not as a nitrogen source. Acinetobacter species was also active on other triazine pesticides, viz., simazine, terbutryn, cyanazine, and prometon. There are very few reports on the degradation of atrazine by any member of this genus and hence this could lead to new degradation pathways and new metabolites.  相似文献

通过选择性富集培养,从沈抚灌区石油污染土壤中分离到1株菲降解细菌.试验证明该菌株能以菲为唯一碳源和能源生长.经形态学、生理生化鉴定和16S rRNA基因序列比对分析,确定该菌株属于不动杆菌属,命名为Acinetobacter sp. L2. 系统发育进化分析发现,L2菌株与Acinetobacter sp. DG880[AY258108]亲源关系最近.L2菌株培养7 d后对菲的降解率达96.3%.邻苯二酚2,3-双加氧酶活力测定表明,L2菌株可能含有菲降解基因.  相似文献

Acinetobacter sp. strain 4CB1 was isolated from a polychlorobiphenyl-contaminated soil sample by using 4-chlorobenzoate as a sole source of carbon and energy. Resting cells of Acinetobacter sp. strain 4CB1 hydrolytically dehalogenated 4-chlorobenzoate under aerobic and anaerobic conditions, but 4-hydroxybenzoate accumulated only under anaerobic conditions. Cell extracts of Acinetobacter sp. strain 4CB1 oxidized 4-hydroxybenzoate by an NADH-dependent monooxygenase to form protocatechuate, which was subsequently oxidized by both ortho- and meta-protocatechuate dioxygenase reactions. When grown on biphenyl, Acinetobacter sp. strain P6 cometabolized 4,4'-dichlorobiphenyl primarily to 4-chlorobenzoate; however, when this strain was grown in a coculture with Acinetobacter sp. strain 4CB1, 4-chlorobenzoate did not accumulate but was converted to inorganic chloride. When resting cells of Acinetobacter sp. strain 4CB1 were incubated anaerobically with 3,4-dichlorobenzoate, they accumulated 4-carboxy-1,2-benzoquinone as a final product. Since 3,4-dichlorobenzoate is a product that is formed from the cometabolism of 3,4-dichloro-substituted tetrachlorobiphenyls by Acinetobacter sp. strain P6, the coculture has a potential application for dehalogenation and mineralization of specific polychlorobiphenyl congeners.  相似文献

Bacterial flora in bottled uncarbonated mineral drinking water   总被引:16,自引:0,他引:16  
A quantitative study of bacterial populations in mineral water was carried out. Samples were stored at 6 and 20 degrees C, and the colony counts were determined on tryptone agar plates incubated at 22 and 37 degrees C. Samples were collected from the spring source in sterile glass flasks and from the bottling factory in conventional plastic and glass containers. In both cases, the initial population (10(1)-10(2) cfu/mL water) increased to 10(5)-10(6) cfu/mL after 3 days storage as determined from plate counts incubated at 22 degrees C. The levels reached by this population were similar to those of samples of mineral water obtained at the market stage. Results from plate counts incubated at 37 degrees C showed that populations in samples collected at the bottling factory reached 10(2)-10(3) cfu/mL. No growth was observed in water collected from spring source. Bacterial multiplication was not stopped even when water was stored at 6 degrees C. Caulobacter was the genus found most frequently in both types of samples, followed by Sphaerotilus-Leptothrix. Acinetobacter calcoaceticus and Pseudomonas fluorescens were frequently found in only two springs, and Pseudomonas putida, Arthrobacter, Aeromonas hydrophilia, and Corynebacterium were isolated less frequently. Janthinobacterium was recovered only once from a single spring. A giant bacterium closely resembling Hyphomicrobium and a budding one similar to Pasteuria were recovered from all samples of a single spring and from some of the commercial samples.  相似文献

Acinetobacter sp. strain ADP1 is a nutritionally versatile soil bacterium closely related to representatives of the well-characterized Pseudomonas aeruginosa and Pseudomonas putida. Unlike these bacteria, the Acinetobacter ADP1 is highly competent for natural transformation which affords extraordinary convenience for genetic manipulation. The circular chromosome of the Acinetobacter ADP1, presented here, encodes 3325 predicted coding sequences, of which 60% have been classified based on sequence similarity to other documented proteins. The close evolutionary proximity of Acinetobacter and Pseudomonas species, as judged by the sequences of their 16S RNA genes and by the highest level of bidirectional best hits, contrasts with the extensive divergence in the GC content of their DNA (40 versus 62%). The chromosomes also differ significantly in size, with the Acinetobacter ADP1 chromosome <60% of the length of the Pseudomonas counterparts. Genome analysis of the Acinetobacter ADP1 revealed genes for metabolic pathways involved in utilization of a large variety of compounds. Almost all of these genes, with orthologs that are scattered in other species, are located in five major 'islands of catabolic diversity', now an apparent 'archipelago of catabolic diversity', within one-quarter of the overall genome. Acinetobacter ADP1 displays many features of other aerobic soil bacteria with metabolism oriented toward the degradation of organic compounds found in their natural habitat. A distinguishing feature of this genome is the absence of a gene corresponding to pyruvate kinase, the enzyme that generally catalyzes the terminal step in conversion of carbohydrates to pyruvate for respiration by the citric acid cycle. This finding supports the view that the cycle itself is centrally geared to the catabolic capabilities of this exceptionally versatile organism.  相似文献

Total number of bacteria, viable counts of aerobic and anaerobic heterotrophic bacteria and 16S rRNA gene diversity were investigated during drilling of three boreholes in the walls of the Äspö hard rock laboratory tunnel, at depths ranging from 380 to 446 m below sea level. Water samples were taken from the drill water source, the drilling equipment and from the drilled boreholes. The drill water was kept under nitrogen atmosphere and all equipment was steam cleaned before the start of a new drilling. Total and viable counts of bacteria in the drilled boreholes were several orders of magnitude lower than in the samples from the drilling equipment, except for sulphate reducing bacteria. A total of 158 16S rRNA genes that were cloned from the drill water source, the drilling equipment and the drilled boreholes were partially sequenced. The drilled boreholes generally had a 16S rRNA diversity that differed from what was found in samples from the drilling equipment. Several of the sequences obtained could be identified on genus level as one of the genera Acinetobacter, Methylophilus, Pseudomonas and Shewanella. In conclusion, the tubing used for drill water supply constituted a source of bacterial contamination to the rest of the drilling equipment and the boreholes. The results show, using molecular and culturing methods, that although large numbers of contaminating bacteria were introduced to the boreholes during drilling, they did not establish in the borehole groundwater at detectable levels.  相似文献

Chloromethane gas is produced naturally in the phyllosphere, the compartment defined as the aboveground parts of vegetation, which hosts a rich bacterial flora. Chloromethane may serve as a growth substrate for specialized aerobic methylotrophic bacteria, which have been isolated from soil and water environments, and use cmu genes for chloromethane utilization. Evidence for the presence of chloromethane-degrading bacteria on the leaf surfaces of Arabidopsis thaliana was obtained by specific quantitative PCR of the cmuA gene encoding the two-domain methyltransferase corrinoid protein of chloromethane dehalogenase. Bacterial strains were isolated on a solid mineral medium with chloromethane as the sole carbon source from liquid mineral medium enrichment cultures inoculated with leaves of A. thaliana. Restriction analysis-based genotyping of cmuA PCR products was used to evaluate the diversity of chloromethane-degrading bacteria during enrichment and after strain isolation. The isolates obtained, affiliated to the genus Hyphomicrobium based on their 16S rRNA gene sequence and the presence of characteristic hyphae, dehalogenate chloromethane, and grow in a liquid culture with chloromethane as the sole carbon and energy source. The cmu genes of these isolates were analysed using new PCR primers, and their sequences were compared with those of previously reported aerobic chloromethane-degrading strains. The three isolates featured a colinear cmuBCA gene arrangement similar to that of all previously characterized strains, except Methylobacterium extorquens CM4 of known genome sequence.  相似文献

The aim of this study was to assess the occurrence of microorganisms in 39 intraoral abscesses. The samples were place in transport medium. The materials were inoculated on adequate enrichment and selective media and cultivated under anaerobic and aerobic conditions. The microorganisms were found in all samples (100%), Anaerobic bacteria most frequently were recovered. The predominant anaerobes were from genus Prevotella, Bacteroides, Fusobacterium and Peptostreptococcus. Among aerobic bacteria, the most frequent were gram-positive cocci. The microaerophilic bacteria and fungi most rarely were isolated from pus samples.  相似文献

研究宜春富硒温泉水体与泉底沉积物的细菌群落多样性。利用高通量测序技术分析泉水与沉积物中细菌群落结构与多样性。温泉水中主要的细菌类群为变形菌门和拟杆菌门,而在沉积物样品中的主要优势菌群为OP1、蓝细菌、浮霉菌门和绿弯菌门。细菌在属分类水平上,温泉水中优势菌群为不动杆菌属、假单胞菌属、水栖菌属、Thermosynechococcus、鞘脂杆菌属和金黄杆菌属等。沉积物样品细菌中优势菌群属于未知物种,在数据库中并没有相关的注释信息;其中已知的优势菌属为Candidatus acetothermum、Thermosynechococcus、亚热栖菌属、不动杆菌属。宜春温汤富硒温泉水体与沉积物中存在着丰富的微生物群落且组成差异性很大,该研究为了解与发掘温泉微生物菌种资源具有重要价值。  相似文献

A novel psychrotolerant, alkalitolerant bacterium, strain Ths, was isolated from a soil sample immersed in hot spring water containing hydrocarbons and grown on a chemically defined medium containing n-tetradecane as the sole carbon source. The isolate grew at 0 degrees C but not at temperatures higher than 45 degrees C; its optimum growth temperature was 27 degrees C. It grew in the pH range of 7-9. The strain utilized C(13)-C(30) n-alkane and fluorene at pH 9 and 4 degrees C. To our knowledge, this is the first report on the bacterium that utilizes a wide range of hydrocarbons at a high pH and a low temperature. Phylogenetic analysis based on 16S rRNA gene sequences showed that strain Ths is closely related to genomic species 6 ATCC 17979 (99.1% similarity), genomic species BJ13/TU14 ATCC 17905 (97.8% similarity), genomic species 9 ATCC 9957 (97.6% similarity) belonging to the genus Acinetobacter and to Acinetobacter calcoaceticus JCM 6842(T) (97.5% similarity). DNA-DNA hybridization revealed that the isolate has 62, 25, 18 and 19% relatedness, respectively, to genomic species 6 ATCC 17979, genomic species BJ13/TU14 ATCC 17905, genomic species 9 ATCC 9957 and A. calcoaceticus, respectively.  相似文献

Several new microorganisms have been isolated from soil samples with high epoxide hydrolase activity toward ethyl 3,4-epoxybutyrate. Screening was performed by enrichment culture on alkenes as sole carbon source, followed by chiral gas chromatography. Eight strains were discovered with enantioselectivity from moderate to high level and identified as bacterial and yeast species. Cells were cultivated under aerobic condition at 30°C using glucose as carbon source and resting cells were used as biocatalysts for kinetic resolution of ethyl 3,4-epoxybutyrate. Among isolated microorganisms, Acinetobacter baumannii showed highest enantioselectivity for (S)-enantiomer, resulting in (R)-ethyl-3,4-epoxybutyrates (>99%ee, 46% yield). It is the first report on the fact that epoxide hydrolases originating from bacterial species of A. baumannii was applied to kinetic resolution of ethyl 3,4-epoxybutyrate in order to obtain enantiopure high-value-added (R)-ethyl-3,4-epoxybutyrate.  相似文献

Abstract Isolates of the genus Streptomyces were readily obtained from the intestines of two African species of soil-feeding termites by an aerobic explant technique using starch casein medium, and from their parent soil and mound materials by dilution plating. Discriminant analysis of the isolates, based on 44 representative characters, showed that the population derived directly from the termites was significantly different from that of the feed soil or the mound. The termite gut was considered to be a good source of unusual actinomycetes, but strains isolated under aerobic conditions are likely to be allochthons selected by the intestinal environment, which is highly alkaline and anaerobic. An anaerobic, filamentous isolate was obtained which may be a component of the prokaryotic symbiont population mediating termite digestion.  相似文献

Free-living amoebae (FLA) of the genus Acanthamoeba are widely distributed in the environment, in the air, soil, and water, and have also been isolated from air-conditioning units. The objective of this work was to investigate the presence of this genus of FLA in the air-conditioning equipment at the Institute of Public Health of Chile in Santiago, Chile. Water and air samples were collected from air-conditioning systems and were checked for the presence of Acanthamoeba spp. Positive samples were further classified at the genotype level after sequencing the highly variable diagnostic fragment 3 (DF3) region of the 18S rRNA gene. This is the first report of the T3, T4, and T11 genotypes of Acanthamoeba in air-conditioning units from Chile. Overall, the widespread distribution of potentially pathogenic Acanthamoeba strains in the studied source demands more awareness within the public and health professionals in Chile as this pathogen is emerging as a risk for human health worldwide.  相似文献

Time-series DNA-stable isotope probing (SIP) was used to identify the microbes assimilating carbon from [(13)C]toluene under nitrate- or sulfate-amended conditions in a range of inoculum sources, including uncontaminated and contaminated soil and wastewater treatment samples. In all, five different phylotypes were found to be responsible for toluene degradation, and these included previously identified toluene degraders as well as novel toluene-degrading microorganisms. In microcosms constructed from granular sludge and amended with nitrate, the putative toluene degraders were classified in the genus Thauera, whereas in nitrate-amended microcosms constructed from a different source (agricultural soil), microorganisms in the family Comamonadaceae (genus unclassified) were the key putative degraders. In one set of sulfate-amended microcosms (agricultural soil), the putative toluene degraders were identified as belonging to the class Clostridia (genus Desulfosporosinus), while in other sulfate-amended microcosms, the putative degraders were in the class Deltaproteobacteria, within the family Syntrophobacteraceae (digester sludge) or Desulfobulbaceae (contaminated soil) (genus unclassified for both). Partial benzylsuccinate synthase gene (bssA, the functional gene for anaerobic toluene degradation) sequences were obtained for some samples, and quantitative PCR targeting this gene, along with SIP, was further used to confirm anaerobic toluene degradation by the identified species. The study illustrates the diversity of toluene degraders across different environments and highlights the utility of ribosomal and functional gene-based SIP for linking function with identity in microbial communities.  相似文献

从3种不同土壤中分离和纯化得到10个石油降解细菌菌株,分离菌株均为好氧菌、革兰氏阴性菌、不形成芽孢的杆菌,10个菌株均能利用中等链长的烷烃、柴油和原油作为碳源,而不能以单环芳烃和多环芳烃为碳源。根据其生理生化性状和16SrDNA序列分析结果表明,分离菌株EVA5,EVA6,EVA7,EVA8和EVA9为假单胞菌(Pseudomonas spp.),EVA10、EVA11、EVA12、EVA13和EVA14为不动杆菌(Acinetobacter spp.),均属于Proteobac-teria的γ亚群。  相似文献

目的:研究塔里木河天然胡杨林部分地区可培养细菌的生态分布。方法:通过塔里木河胡杨林采样,可培养菌分离及16S rDNA序列鉴定。结果:从3种不同样品(水样、土样和胡杨树杆分泌物)中分离筛选了22株细菌,其中17株菌为革兰氏阳性菌,5株为革兰氏阴性菌。根据生理生化特征与16S rDNA序列分析结果表明,其中15株菌属于芽孢杆菌属,4株属于不动杆菌属、假单胞菌属、动性球菌属和Agrococcus属各含有1个分离株。结论:塔里木河胡杨林可培养微生物中芽孢杆菌比较丰富,其中有3个可能的新种。  相似文献

不动杆菌属细菌分布广泛,作为重要的石油烃降解者,在乳化和降解石油烃、降低石油烃生物毒性等方面有重要作用。本文概述了不动杆菌属细菌对烷烃、芳香烃等石油烃组分的降解,总结了该属细菌中已发现的烷烃氧化酶和芳香烃氧化酶,综述了该属细菌所分泌的表面活性剂的类型和乳化机理,讨论了固定化对该属细菌降解石油烃的影响,展望了该属细菌降解石油烃的应用前景。基于此,作者认为探索不动杆菌属细菌降解石油烃的详细机理和途径、发现关键酶、寻找遗传工具、构建基因工程菌、发掘环境友好的固定化材料,应是未来的研究重点及热点。  相似文献
