Sulfate-reducing bacteria in littoral sediment of Lake Constance   总被引:6,自引:0,他引:6  
Abstract The viable population of sulfate-reducing bacteria (SRB) in littoral sediments of Lake Constance was investigated using enrichment and enumeration techniques. Enrichment studies established that most types of SRB grew best in media with low salt concentrations (max. 0.4 g Cl/1), consistent with the low salinity of the freshwater habitat. Enumerations were based on an adequate medium with the following electron donors: H2, lactate, acetate, propionate, butyrate, caprylate, succinate, benzoate, or S2O32− for thiosulfate-disproportionating bacteria. Cultures were incubated for 6 weeks to obtain maximum counts. A maximum cell density of 6.3 × 106 cells per ml sediment was estimated, which is the highest number of SRB ever reported for anoxic sediments. A comparison with measured sulfate reduction rates showed that the enumeration techniques were about 10–100-fold more efficient than those previously used. The population of SRB had a characteristic structure consisting of 87.7% H2-utilizing SRB (physiologically resembling the classical Desulfovibrio species); 12.0% propionate utilizers (tentatively identified as Desulfobulbus species); 0.3% long chain fatty acid-oxidizing Desulfovibrio sapovorans species. Acetate-utilizing SRB ( Desulfotomaculum acetoxidans ) constituted ≤ 0.05% of the total estimated population. Moreover, the latter species was only present as inactive spores. Benzoate-degrading SRB were not detected.  相似文献

Abstract In the profundal sediment ot Lake Constance (143 m depth) the temperature is constant at 4 °C. Despite the constant temperature, CH4 concentrations changed with season between about 120 μM in winter and about 750 μM in summer, measured down to 30 cm depth. The acetate concentration profiles also varied between seasons. In summer, acetate concentration reached a maximum at about 100 μM in 2 or 4 cm depth. In winter, maximal concentrations of about 5 μM were observed over the entire depth. Input of organic material in late spring may be the reason for the seasonal change of both compounds. To simulate such a sedimentation event, intact sediment cores were covered with suspensions of Porphyridium aerugenium or Synechococcus sp. The addition of the phytoplankton material resulted in a drastic increase of acetate concentrations with a maximum at 2 cm depth, similar to in situ acetate concentrations measured in summer. The same applies for CH4 for which increased concentrations were observed down to 6 cm depth. H2 concentrations, on the other hand, showed no distinct increase. Treatment of intact sediment cores with 14C-labeled Synechococcus cells resulted in the formation of 14C-acetate, 14CH4 and 14CO2. Maximum concentrations of 14CH4 were found at 4 cm depth, i.e. just above the depth to which 14C-acetate penetrated. The results show that phytoplankton blooms may cause a seasonal variation of acetate and CH4 in profundal sediments of deep lakes despite the constant low temperature. They also indicate that acetate is the dominant substrate for methanogenic bacteria in the profundal sediments of Lake Constance.  相似文献

Maximum rates of CH4 production in the littoral sediment were observed in 2–5 cm depth. The CH4 production rates increased during the year from about 5 mmol m−2d−1 in December to a maximum of about 95 mmol m−2d−1 in September. CH4 production rates showed a temperature optimum at 30°C and an apparent activation energy of 76 kJ mol−1. A large part of the seasonality of CH4 production could be ascribed to the change of the sediment temperature. Most of the produced CH4 was lost by ebullition. Gas bubbles contained about 60–70% CH4 with an average δ13C of −56.2% and δD of −354%, and 2% CO2 with an average δ13C of −14.1% indicating that CH4 was produced from methyl carbon, i.e. mainly using acetate as methanogenic substrate. This result was confirmed by inhibition of methanogenesis with chloroform which resulted in an accumulation rate of acetate equivalent to 81% of the rate of CH4 production. Most probable numbers of methanogenic bacteria were in the order of 104 bacteria g−1d.w. sediment for acetate-, methanol- or formate-utilizing, and of 105 for H2-utilizing methanogens. The turnover times of acetate were in the order of 2.3–4.8 h which, with in situ acetate concentrations of about 25–50 μM, resulted in rates of acetate turnover which were comparable to the rates of CH4 production. The respiratory index (RI) showed that [2−14C]acetate was mainly used by methanogenesis rather than by respiratory processes, although the zone of CH4 production in the sediment overlapped with the zone of sulfate reduction.  相似文献

We have tested the hypothesis that spheroidal carbonaceous particles (SCP), emitted from oil and coal combustion, can be used as a marker for recent sediment distribution in lake basins. Sediment traps have been used to study when and how the particles are introduced into a lake, a laboratory experiment has been made to study the depositional behaviour of the particles in relation to other sedimentary constituents, and spatial SCP distributions have been surveyed in three lakes. The results support the hypothesis that SCP can be used as a marker, and also indicate that acidification can change sediment distribution in lakes.  相似文献

The microbial community structure of an anoxic profundal lake sediment, i.e., subtropical Lake Kinneret, was analysed with respect to its composition by culture-independent molecular methods including terminal restriction fragment length polymorphism (T-RFLP) analysis, comparative sequence analysis, and quantitative real-time PCR. In particular we were interested in the structure, species composition, and relative abundance of the overall microbial community in the methanogenic sediment layer (0-10 cm depth). Pairwise comparison of archaeal and bacterial 16S rRNA gene T-RFLP profiles obtained from three independent samplings indicated stability of the microbial community. The numbers of Archaea and Bacteria, quantified by real-time PCR, amounted to about 10(8) and 10(10) 16S rRNA gene copies cm(-3) sediment, respectively, suggesting that Archaea may account for only a minor fraction (approximately 1%) of the total prokaryotic community. Hydrogenotrophic Methanomicrobiales and acetoclastic Methanosaeta spp. dominated T-RFLP profiles of the archaeal community. T-RFLP profiles of the bacterial community were dominated by Deltaproteobacteria, sulphate reducers and syntrophs in particular. The second most abundant group was assigned to the Bacteroidetes-Chlorobi-group. Only one bacterial group, which was affiliated with halorespiring bacteria of subphylum II of the Chloroflexi, showed variation in abundance within the sediment samples investigated. Our study gives a comprehensive insight into the structure of the bacterial and archaeal community of a profundal lake sediment, indicating that sulphate reducers, syntrophs, bacteroidetes, halorespirers and methanogens are of particular importance in Lake Kinneret sediment.  相似文献

The effects of natural nematode communities on bacterial activity and abundance were investigated in a microcosm study. Nematodes were added at different densities to a freshwater sediment and bacterial parameters were measured after 1, 5, 9, and 17 days. Significant effects of nematode density on bacterial activity were noted on day 5. No long-term changes in bacterial activity were recorded. Bacterial abundance displayed an overall decrease in both treatments and controls. In a second experiment, the effect of nematode feeding-type on bacterial activity was studied. Microcosms were incubated with 100 individuals of a fungus-feeding (Aphelenchus avenae) or a bacteria-feeding nematode species (Caenorhabditis elegans) respectively, and bacterial activity was determined after 0, 1, 2, 4, and 7 days. Significant time and feeding-type effects were found, with consistently higher bacterial activity estimates in treatments with bacteria-feeding nematodes. These results suggest that grazing affects bacterial activity, and indicate that grazing by nematodes may be more important in stimulating bacterial activity than bioturbation or excretion. Combining these results, we conclude that natural nematode communities may have an impact on bacterial activity, and that the magnitude of this impact depends on the proportion of actively feeding bactivores within the community. Lake Varese (northern Italy) has shown deterioration in water quality since the 1960s and, as a result of the long duration of direct discharge of untreated sewage into the lake, it was classified as being hypertrophic. To recover the lake water quality, a series of externally and internally remedial actions were implemented in subsequent years. The applied sewage collecting system induced a reduction of the external P loads from 50 t P year−1 to 16t P year−1 and the weighted mean annual TP concentration decreased from 352 μg P l−1 to 85 μg P l−1, typical of eutrophic conditions. The hypolimnetic water withdrawals, adopted in the years 2000–2003, allowed a reduction of the internal P loads of about 3–5 t P. In the same years, 500t O2 were injected at depths of 4.5–8 m during the summer months. In spite of these internal remedial actions, no significant reduction of the weighted mean annual concentration of the TP could be observed, and during the summer stratification period no significant reduction of the volumes of anoxic water and of the duration of the anoxia were detected. The anoxic conditions are still the prevailing force driving the lake P-budget, maintaining the lake in eutrophic status.  相似文献

The Lake Taihu drainage basin is an economically developed area with some of the highest population densities in China. The lake has deteriorated due to ecological destruction and eutrophication. Three short sediment cores from eastern, northeastern and southwestern Lake Taihu were collected. Total organic carbon (TOC), total nitrogen (TN), pigments, elements and particle size were analyzed for the purpose of understanding past trophic status and pollution levels. Sedimentation rates were based on 137Cs or 210Pb methods. Results indicated that sediment particle size became coarser since the 1920s, and the lake was contaminated by heavy metals, such as Cu and Zn, since the 1970s. A remarkable increase in eutrophication since the 1980s due to increased loading of untreated effluents from industry, agriculture and urbanization is reflected by total organic carbon, total nitrogen and pigments in the studied cores. However the onset times of eutrophication in different parts of Lake Taihu were not synchronous.  相似文献

Planktonic microbial community structure and classical food web were investigated in the large shallow eutrophic Lake Taihu (2338 km2, mean depth 1.9 m) located in subtropical Southeast China. The water column of the lake was sampled biweekly at two sites located 22 km apart over a period of twelve month. Site 1 is under the regime of heavy eutrophication while Site 2 is governed by wind-driven sediment resuspension. Within-lake comparison indicates that phosphorus enrichment resulted in increased abundance of microbial components. However, the coupling between total phosphorus and abundance of microbial components was different between the two sites. Much stronger coupling was observed at Site 1 than at Site 2. The weak coupling at Site 2 was mainly caused by strong sediment resuspension, which limited growth of phytoplankton and, consequently, growth of bacterioplankton and other microbial components. High percentages of attached bacteria, which were strongly correlated with the biomass of phytoplankton, especially Microcystis spp., were found at Site 1 during summer and early autumn, but no such correlation was observed at Site 2. This potentially leads to differences in carbon flow through microbial food web at different locations. Overall, significant heterogeneity of microbial food web structure between the two sites was observed. Site-specific differences in nutrient enrichment (i.e. nitrogen and phosphorus) and sediment resuspension were identified as driving forces of the observed intra-habitat differences in food web structure.  相似文献

巢湖渔业资源现状及其对水体富营养化的响应研究   总被引:15,自引:0,他引:15  
2002年至2004年期间,对巢湖鱼类资源进行调查,共发现鱼类54种,隶属16科、9目,主要以鲤科鱼类为主(35种),占64.8%,与20世纪80年代相比,鱼类种类数减少了40种,主要表现在洄游性种类急剧减少,甚至消失。渔业资源结构(渔获物)表现为以湖鲚、太湖新银鱼等小型鱼类为优势种类,在渔产量的比例不断上升,而大型鱼类(如翘嘴、鲤等)的种群结构趋于低龄化,产量呈下降趋势。结合已有的历史资料,分析巢湖渔业资源变化的影响因素,结果表明造成巢湖渔业资源结构变化受人为活动的影响主要表现在水利工程修建、过度捕捞和水体富营养化等方面。同时探讨近20年巢湖渔业捕捞产量与水体氮磷含量变化的相互关系发现,水体磷的含量变化显著影响巢湖渔业的捕捞产量。因此,合理利用巢湖渔业资源,需要减轻水体富营养化,控制捕捞强度和人为调整渔业结构使得巢湖渔业可持续发展。  相似文献

Phosphorus release rates from profundal sediments of Lake Constance (Obersee) have been determined in D.O., pH regulated sediment-water systems. Above 10% O2 saturation (> 1.2 ppm D. O.) and with pH as in situ, no net release could be found. Sedimentations of diatom sludge (Asterionella formosa) and carbonate-phosphate coprecipitate (CaCO3.CaHPO4) increased the release to 0.5 mg × m–2 × d–1 which, however, will not be relevant to the P balance in Obersee. 为揭示水生植物对富营养化河涌底泥的生理生态适应性及其净化能力,选取11种水生挺水植物(包含6种本土植物和5种外来植物)结合河涌底泥进行试验。通过测定试验一年后植物叶片的光饱和光合速率(P_(sat),μmol m~(-2)s~(-1))、比叶面积(SLA,m~2/kg)、总氮含量(TN,mg/g)和光合作用氮利用效率(PNUE,μmol mol~(-1)s~(-1)),比较分析物种间生理与结构特性及其相互关系。结果表明:种间的SLA层次比较分明,最高的大叶皇冠草(20.31±0.30)与最低的鸢尾草(7.22±0.31)相差近3倍。种间的P_(sat)在(3.76±0.57)(鸢尾草)—(21.53±1.20)(水罂粟)之间,水罂粟比鸢尾草高81.79%。种间的PNUE从42.53±8.42(鸢尾草)至655.8±100.93(天使花),美人蕉、水罂粟、风车草和香蒲的PNUE值均较高,且差异不明显(P0.05),这些植物的PNUE显著高于较低PNUE的种类(包括菖蒲、蓝花草和鸢尾草)(P0.05)。种间SLA分别与PNUE和P_(sat)(μmol kg~(-1)s~(-1))呈显著的正相关,SLA和P_(sat)(μmol m~(-2)s~(-1))分别与TN(mmol/m~2)呈显著负相关(P0.05)。外来植物类群的PNUE、SLA、P_(sat)和TN均显著高于本地植物类群(T-test,P0.05),说明外来水生植物在养分富集化环境下能更有效地利用资源,具有潜在的高生长速率和种间竞争优势。  相似文献

To determine recent sediment movement, we measured the levels of 137Cs (an artificial radionuclide produced during nuclear weapons testing) of 118 southern Lake Michigan samples and 27 in Green Bay. These samples, taken from 286 grab samples of the upper 3 cm of sediment, were collected in 1975 as part of a systematic study of Lake Michigan sediment. 137Cs levels correlated well with concentrations of organic carbon, lead, and other anthropogenic trace metals in the sediment. 137Cs had a higher correlation with silt-sized than with clay-sized sediment (0.55 and 0.46, respectively). Atmospherically derived 137Cs and trace metals are being redistributed by sedimentary processes in Lake Michigan after being incorporated in suspended sediment. We determined a distribution pattern of 137Cs that represents areas of southern Lake Michigan where sediment deposition is occurring.  相似文献

Short-term variations in phosphorus (P) concentrations must be considered while assessing the long-term changes in trophic status and estimating the P load and export. Furthermore, given the challenges of conventional monitoring of river systems, a sediment-specific biomonitoring tool may be more successful inferring P related human controls. In this study, conducted along a 37 km river channel representing up-and downstream urban control, and through a trajectory from a major point source (Assi drain), we tested the patterns of concordance between alkaline phosphatase (AP) activity and soluble reactive-P (SRP) and between AP activity and trophic status in the Ganga River. To validate data comparison, we selected a reference site at Dev Prayag, situated ∼1130 km upstream to the main study stretch. Samples were collected for three consecutive year (March 2013 to February 2016) with respect to atmospheric deposition, surface runoff, point source loading, river water and sediment analysis. For trajectory analysis, samples were collected from 15 locations starting from the drain outlet (zero distance) upto 1.5 km downstream with sampling location 100 m away from the preceding one. We found marked spatial and temporal variations in P concentrations which could be traced by quantifying the AP activity. The AP activity, recorded highest at reference site, declined with increases in P; and at drain mouth it was close to zero reflecting strong influence of P level on alkaline phosphatase activity in the river. We used canonical correlation analysis (CCorA) to test the degrees of concordance and similarity in different variables. Most of the environmental variables and indicators of eutrophy appear largely clustered at one side of the coordinate separating AP activity and dissolved oxygen towards opposite side of the axis. The dynamic fit function relating AP activity with different variables showed significant positive correlation with DO (R2 = 0.67; p < 0.001) and negative correlations with BOD (R2 = 0.82; p < 0.001), Chl a biomass (R2 = 0.52; p < 0.001) and trophic status index (R2 = 0.54 (Chl a), 0.96 (DRP); p < 0.001). Furthermore, the enzyme activity did not show significant negative correlation with heavy metals in sediment. Because anthropogenic activities continue to enhance P loads; AP is inhibited directly by P availability; and eutrophy feedbacks sediment P release, our observations on P-AP activity relationship provide a valuable alternative means for detecting P related controls on water quality, trophic status and biogeochemical feedbacks in human impacted rivers.  相似文献

Benthic macroinvertebrate distribution was studied on April 9, 1994, and March 2, 2003, in Lake Yamanakako. The average density of the benthic community for the entire lake was 3168ind.m–2, comprising principally oligochaetes (41.0%) and chironomids (59.0%) in 1994. In 2003, the benthic community for the entire lake was 1847ind.m–2, principally consisting of oligochaetes (69.9%) and chironomids (30.1%). In 1994, the larval density of Propsilocerus akamusi was 3.5 times that of Chironomus nipponensis and in 2003 the figure was 5.7 times. However, the larval biomass of P. akamusi was 2.1 times greater than that of C. nipponensis in 1994 and 2.8 times greater in 2003. The larval density of Tanypodinae decreased drastically, by about 12-fold, from 1994 to 2003. P. akamusi larvae were particularly abundant at the lake center in 1994, but they inhabited the entire lake bottom in 2003. P. akamusi density was closely related to water depth and ignition loss. C. nipponensis larvae also showed the widest distribution pattern in 2003, whereas their larvae had inhabited the northeastern parts and the lake center in 1994. Recently, the number of C. nipponensis larvae in Lake Yamanakako is tending to decrease, whereas that of P. akamusi larvae is increasing, suggesting ongoing eutrophication.  相似文献

揭示湖泊的富营养化发生机制、定量了解关键生源要素与藻类爆发的因果关联对有效改善湖泊水质和富营养化状况具有重要的科学与决策意义。本研究以云南省程海为例,建立了基于神经网络的响应模型,对富营养化机理进行了研究,并从富营养化核心驱动因子识别、神经网络模型构建与架构分析以及叶绿素a(Chl a)与TN、TP浓度降低的响应模拟几个方面对面临的科学问题进行探索。模拟结果表明,神经网络模型必须在适当的架构下才能产生科学合理的结果;程海的富营养化机制由一个氮(N)、磷(P)共限制的营养盐-藻类动力结构主导,但在此主导结构下拥有氮型限制的次级结构。基于神经网络模型模拟,推导出一系列基于湖体水质控制的Chl a响应的非线性函数,为程海的富营养化控制提供了快速决策支持。  相似文献
