The centromere position on the chromosomes of three European stocks of Nicotiana plumbaginifolia (2n=20) was determined by investigating in root tip mitoses the shape of the chromosomes during congression and polar movement and during C-metaphase. One pair of chromosomes is subtelocentric and 9 pairs are acrocentric. Some chromosomes mimic (sub-)metacentrics by having noncentric constrictions. A recently published (A. Villa; Genetica 64: 145–148, 1984) idiogram of one of the stocks is commented on.  相似文献

Patterns of secondary association between the acrocentric autosomes of man   总被引:5,自引:0,他引:5  
The pattern of association between acrocentric chromosomes at C-metaphase has been studied in 1140 unlabelled and 473 labelled normal human cells. Models are proposed which examine alternative hypotheses relating to those factors which may determine within and between individual variation in this character. It is concluded that the pattern of association is determined by the nucleolar organising ability of the chromosomes, their capacity for association either by proximity or by attraction to a dominant nucleolar organiser or fusion site and to a limitation on the number and type of chromosome which may be accepted into a single association.  相似文献

Duplicate staining of human metaphase chromosomes, first with Giemsa followed by silver, revealed the presence of two small silver-stained bodies not seen in the Giemsa stained metaphases. Similar bodies were subsequently found in the metaphases of several animal groups. The size, structure, spatial relationships to the nucleus, behavior throughout the cell cycle, and apparent universal presence of these bodies suggest that they are either centrioles or associated centriolar structures. These centriole-bodies can be seen throughout the cell cycle, even in hypotonically spread C-metaphase chromosome plates. The silver stain procedure allows enough resolution to distinguish parent and daughter centriole-bodies at interphase, thus permitting visualization of the replication, maturation and separation stages of these bodies with the light microscope.  相似文献

A drastic reduction was observed in the mitotic index in Allium cepa roots treated for long periods with various concentrations of pantopon hydrochloride.Pantopon hydrochloride induced various types of mitotic abnormality in root tips of Allium cepa. The most common abnormality occurring at the metaphase stage was C-metaphase. The C-metaphase was maintained even after a few hours of recovery. In addition, C-anaphase was frequently noticed which sometimes led, after restitution, to the production of polyploid cells. The most prominent anaphase abnormality induced by pantopon hydrochloride was the chromosome bridge.  相似文献

Treatment of HeLa cells with Colcemid at concentrations of 0.06-0.10 mug/ml leads to irreversible arrest in mitosis. Colcemid-arrested cells contained few microtubules, and many kinetochores and centrioles were free of microtubule association. When these cells were exposed to microtubule reassembly buffer containing Triton X-100 and bovine brain tubulin at 37 degrees C, numerous microtubules were reassembled at all kinetochores of metaphase chromosomes and in association with centriole pairs. When bovine brain tubulin was eliminated from the reassembly system, microtubules failed to assemble at these sites. Similarly, when EGTA was eliminated from the reassembly system, microtubules failed to polymerize. These results are consistent with other investigations of in vitro microtubule assembly and indicate that HeLa chromosomes and centrioles can serve as nucleating sites for the assembly of microtubules from brain tubulin. Both chromosomes and centrioles became displaced from their C-metaphase configurations during tubulin reassembly, indicating that their movements were a direct result of microtubule formation. Although both kinetochore- and centriole- associated microtubules were assembled and movement occurred, we did not observe direct extension of microtubules from kinetochores to centrioles. This system should prove useful for experimental studies of spindle microtubule formation and chromosome movement in mammalian cells.  相似文献

Data are presented on the effect of chlorahydrate on microtubule organization in the root meristem of Allium cepa. Our studies show that an incomplete preprophase band commonly appears during G2-prophase transition, yet the major effect is the lack of perinuclear microtubules, leading to inhibition of the prophase spindle formation and transition to C-mitosis. Upon chloralhydrate treatment of metaphase cells, we found cells with chromosomes regularly aligned within the metaphase plate and differently disorganized mitotic spindles. Concurrently, C-metaphase cells with remnants of kinetochore fibers were present. In addition, normal bipolar and abnormal irregular types of chromosome segregation were detected, this representing multipolar and diffuse anaphases. The major difference between them is the presence of polar microtubules during multipolar anaphase, and their lacking during diffuse anaphase. Alternatively, microtubule clusters between segregated groups of chromosomes are typical for cells with diffuse anaphase. During bipolar anaphase, excessive aster-like microtubules emanate from the spindle poles, and in telophase accessory phragmoplasts are observed at the cell periphery. The formation of incomplete phragmoplasts was observed after normal bipolar and abnormal chromosome segregation. We conclude that chloralhydrate may affect the nuclear surface capability to initiate the growth of perinuclear microtubules, thus blocking the prophase spindle formation. It also disturbs the spatial interaction between microtubules, which is crucial for the formation and functioning of various microtubular systems (preprophase band, spindle and phragmoplast).  相似文献

Summary Exposure of germlings of Allomyces neo-moniliformis to colchicine for 0 to 5 min after zoospore encystment was found to block 30% of germlings derived from flagellated zoospores and 55% of germlings derived from deflagellated zoospores in C-metaphase configurations at the first mitotic division. The zoospore lacks a pool of colchicine binding protein, and protein synthesis is absent during the time when colchicine first becomes effective in inducing C-metaphase. From these observations it is concluded that the microtubule subunit protein of the spindle apparatus of the first mitotic division to a large extent is derived from the depolymerization of the cytoplasmic microtubules of the zoospore. GTP, Mg2+, and ATP were observed to be antagonistic to the action of colchicine in vivo. It is suggested that these compounds may compete with colchicine for binding to the subunit protein in vivo. Germlings derived from flagellated zoospores are appreciably less subject to the action of colchicine in the presence of the antagonistic compounds than are germlings derived from deflagellated zoospores. This differential sensitivity to colchicine is interpreted as reflecting a difference in the quantity of microtubule subunit protein present at the time of exposure to colchicine.  相似文献

The aim of this study was to compare the chromosomal aneuploidy rate between transgenic and non-transgenic rabbits derived from the F4 generation. Chromosomal analysis was carried out on bone marrow samples of New Zealand White transgenic (carrying human factor VIII gene) and non-transgenic rabbits (F4 generation) each having a different genetic background (female no. 1-3-5 line I and female no. 1-9-7 line II). C-metaphase plates were obtained from the bone marrow lymphocytes synchronized by the addition of 0.25 microg/ml colcemide. No significant difference in chromosomal aneuploidy between transgenic (61%) and non-transgenic (51.27%) rabbits of line I was observed. A higher but non-significant aneuploidy rate between transgenic and non-transgenic rabbits was found in line II, on the other hand a significant difference (P<0.05) was observed in diploidy rate. In conclusion, chromosomal aneuploidy rates in this experiment were higher than published previously in other reports.  相似文献

Interduplex crosslinks by a bifunctional anthramycin DNA crosslinker produced triradial and quadriradial chromosomes. The crosslinker alkylates guanine at N-2. Bovine chromosomes contain GC-rich density satellite DNAs at the centromeric heterochromatin and is the basis for the formation of triradial and quadriradial chromosomes at the centromeres. The in situ crosslinking of interphase chromosomes indicates that the distance between centromeres is 17.5 A. We conclude that the nuclear matrix associated DNA in the centromeric heterochromatin of interphase chromosomes are positioned close enough for crosslinking to occur. We propose a model for the generation of triradial and quadriradial chromosomes based upon the number of interduplex crosslinks between two chromosomes.  相似文献

Study on meiotic chromosomes of grasshopper, Gesonula punctifrons and interphase polytene chromosomes from Dipteran larvae as of Chironomus striatipennis and Drosophila melanogaster following staining by periodic acid-Schiff technique revealed that chromosomes contained polysaccharides as an integral part of their organization. PAS +ve nature of the chromosomes both at highly condensed state as available during meiotic cell division and at extended state as in polytene chromosomes supports the idea that chromosomes contain polysaccharides as one of the constituent biological macromolecules. PAS +ve chromosomes appeared to be fluorescent under fluorescence microscope and fluorescence was found to be more or less uniform along the whole length of the meiotic chromosomes, while in case of polytene chromosomes intense fluorescence could be noticed along the band regions of the chromosomes.  相似文献

Methylation pattern peculiarities revealed by immunocytochemical analysis of metaphase chromosomes from preimplanted human embryos with monoclonal antibodies against 5-methylcytosine are described. Chromosomes of 2-8-cell triploid human embryos are undermethylated, if compared to those from PHA-stimulated fetal cord blood lymphocytes. Hemimethylation (asymmetric labeling of sister chromatids) is typical for the most of embryonic chromosomes at 2-cell--blastocyst stages due most probably to a passive loss of methylation during initial cleavages. Diffuse labeling and sister chromatid exchanges are two other cytogenetic peculiarities revealed by immunofluorescent staining of early human embryos. Hypomethylation of pericentromeric heterochromatin of chromosomes 1, 9, 16 and different methylation status of some homologous chromosomes may distinguish them from metaphase chromosomes of lymphocytes. M-banding pattern typical for chromosomes from adult and cord blood lymphocytes initially appears in embryonic metaphase chromosomes as early as at a 8-cell stage to be established for most part of chromosomes of the karyotype at the morula-blastocyst stage only.  相似文献

Cultivated rye (Secale cereale) and its weedy relative (S. segetale) carry B chromosomes. The B chromosomes are known to be morphologically alike at somatic metaphase and they are of the standard type in natural populations. To clarify the cytogenetic relationship between the standard B chromosomes of S. cereale and those of S. segetale, we made four crosses between Afghan S. segetale with two standard B chromosomes as a pistillate parent and Turkish, Iranian, Korean, and Japanese S. cereale, all with two standard B chromosomes as pollen parents. We observed the pairing of B chromosomes at diakinesis in pollen mother cells in all F1 hybrids with four standard B chromosomes, two from each of the pistillate and the pollen parents. The degree of pairing of B chromosomes in all F1 hybrids with four standard B chromosomes was similar to or somewhat lower than, that in parental strains with four standard B chromosomes. These results showed that the standard B chromosomes in S. segetale from Afghanistan are homologous with those in S. cereale from Turkey, Iran, Korea, and Japan. We therefore propose monophyletic origin of the standard B chromosomes in S. segetale and S. cereale.  相似文献

CENP-E is a kinesin-like protein that when depleted from mammalian kinetochores leads to mitotic arrest with a mixture of aligned and unaligned chromosomes. In the present study, we used immunofluorescence, video, and electron microscopy to demonstrate that depletion of CENP-E from kinetochores via antibody microinjection reduces kinetochore microtubule binding by 23% at aligned chromosomes, and severely reduces microtubule binding at unaligned chromosomes. Disruption of CENP-E function also reduces tension across the centromere, increases the incidence of spindle pole fragmentation, and results in monooriented chromosomes approaching abnormally close to the spindle pole. Nevertheless, chromosomes show typical patterns of congression, fast poleward motion, and oscillatory motions. Furthermore, kinetochores of aligned and unaligned chromosomes exhibit normal patterns of checkpoint protein localization. These data are explained by a model in which redundant mechanisms enable kinetochore microtubule binding and checkpoint monitoring in the absence of CENP-E at kinetochores, but where reduced microtubule-binding efficiency, exacerbated by poor positioning at the spindle poles, results in chronically monooriented chromosomes and mitotic arrest. Chromosome position within the spindle appears to be a critical determinant of CENP-E function at kinetochores.  相似文献

A small population of complex translocation heterozygote plants of Allium roylei from the Bani region of Jammu Province was studied for meiosis in the female track. This study resulted in identification of two variants, having embryo-sac mother cells (EMCs) with more than 16 chromosomes. EMCs of the remaining plants invariably had diploid (2n = 16) chromosome complement. Female meiosis, in general, was found to be abnormal, with nearly 23% and 11% chromosomes associating as quadrivalents or trivalents at prophase I and at metaphase I, respectively. This was followed by irregular segregation of chromosomes at anaphase I. Amongst the variants; one had 38% EMCs with eight bivalents plus two small sized chromosomes. Their small size, dispensable nature and tendency to affect the pairing behaviour of normal complement are some of the features that latter chromosomes share with the B chromosomes. Seventeen to nineteen chromosomes were observed in 35% EMCs of other variant; the remaining cells had 16 chromosomes. Chromosomal behaviour in both kind of cells (euploid and aneuploid) was more or less similar. Unlike female meiocytes, male meiocytes analysed earlier of this strain always had 16 chromosomes which paired to form extremely complex associations involving 3-16 chromosomes. The most likely cause of this asynchrony with regards to number of chromosomes involved in multivalent formation seems to be interaction of genes controlling chiasma formation with the different physiological conditions of male and female meiocytes.  相似文献

A method for the separation of large quantities of human metaphase chromosomes at neutral pH is described. The separation was performed in a specially designed sedimentation chamber which was placed in a bucket of a speed-controlled centrifuge. Flow deflectors in the chamber allowed both the undisturbed introduction of gradient and chromosomes, as well as the undisturbed fractionation of the content of the chamber. The centrifuge was run at 20 g for 1 h taking care of slow acceleration and deceleration. The slow morphological changes of the chromosomes due to ageing at neutral pH do not affect the resolving power of the sedimentation technique within this hour. This in contrast to separation of chromosomes at neutral pH by velocity sedimentation at unit gravity during 20 h, as described before. The main advantage of chromosome sorting at neutral pH is that the fractionated chromosomes are more suitable for gene transfer and gene mapping experiments.  相似文献

A Giemsa C-banding technique was used to study the amount and location of constitutive heterochromatin in diploid (2n = 2x = 16) Medicago sativa ssp. falcata (L.) Arcangeli. Most accessions had the standard C-banding pattern with centromeric bands on all the chromosomes and a prominent heterochromatic band at the nucleolar organizer regions (NOR) of the satellited (SAT) chromosomes. However, we observed in various accessions that constitutive heterochromatic C-bands can exist at the telomeric ends of all the chromosomes. Interstitial bands occurred on the short arms of all chromosomes except for chromosome 3 and on the long arms of chromosomes 1, 2, 3, and 6, only. Rearranged chromosomes such as isochromosomes have been observed for the short arms of chromosomes 2 and 6. This is the first report on the existence of C-banding polymorphisms and the detection of putative isochromsomes in the chromosomes of diploid ssp. falcata which could have contributed to the variation observed in cultivated alfalfa.  相似文献

The interpretation of the significance of marker chromosomes, which can be encountered at prenatal diagnosis, is extremely problematic. Various factors contribute to the difficulty of clarifying the phenotypic risks of supernumerary marker chromosomes, including differences in the size, structure, and origin of marker chromosomes, as well as the occurrence of multiple marker chromosomes of different origin in the same proband. Research on marker chromosomes is currently in a data-accumulation phase. We report the presence of two marker chromosomes, originating from chromosomes 6 and 11, in a child with developmental delay and craniofacial dysmorphism and discuss the related literature.  相似文献

Morphology and behaviour of the X and Y chromosomes of four species of genus Microtus were studied at pachytene, metaphase I and meiotic metaphase. The X chromosomes of the species varied with respect to their size and location of heterochromatic blocks. The axes of X and Y chromosomes of these species as well as Microtus agrestis never formed true synaptonemal complexes at any sub-stage of the pachytene. They approached each other at the start of the pachytene throughout to metaphase I, getting situated closely. At the end of the pachytene, they formed sex vesicle. The X and Y chromosomes kept their proximity during metaphase I, but never formed true bivalents. It is suggested that lack of synapsis of the X and Y chromosomes in the genus Microtus is the final step of evolutionary trend to reduction of the size of the pseudo-autosomal region. The abolition of restrictions on homology between the X and Y chromosomes is supposed to be a cause for the fast divergence in morphology of sex chromosomes in the genus.  相似文献

Application of the fluorescent DNA-intercalator propidium iodide for stabilization of the mitotic chromosome structure during isolation of chromosomes from V79 Chinese hamster cells and subsequent staining with the fluorochromes 33258 Hoechst or DAPI allowed bivariate flow karyotyping of isolated chromosomes. Fluorescence of 33258 Hoechst bound to isolated chromosomes containing 5-bromodeoxyuridine (BrdUrd) was quenched in comparison with the fluorescence of control chromosomes. Despite structural relationship and similarity of both absorption and fluorescence spectra of DAPI and 33258 Hoechst, reduction of fluorescence of DAPI-stained isolated chromosomes was not observed, by contrast with findings in conventional cytological metaphase preparations. It could be obtained, however, by preirradiation of the chromosomes with near-UV in the presence of DAPI. This led to a progressive destruction of the chromosomes. Destruction also occurred without BrdUrd, though at a slower rate. Preirradiation of chromosomes in the presence of 33258 Hoechst hardly affected the integrity of the chromosomes. Preirradiation of a 33258 Hoechst solution and its subsequent use as a stain resulted in a considerably decreased fluorescence of chromosomes. For DAPI this effect was small. Thus, whereas 33258 Hoechst itself is much more sensitive to near-UV irradiation than DAPI, DAPI bound to DNA in chromosomes renders the DNA much more sensitive to irradiation than 33258 Hoechst bound to DNA. Presumably, these differences can at least partly be reduced to the different molecular sizes of the dyes.  相似文献

In this paper,Psammosilene tunicoides,an endemic species to China,was cytologically studied for the first time.The morphology of the nuclei at resting stage was categorized to be simple chromocentre type.The morphology of mitotic prophase chromosomes was categorized to be the interstitial type.28 chromosomes were observed at the mitotic metaphase,and 14 bivalent chromosomes were observed at diakinesis.So,the basic chromosome number was confirmed to be x=14.Psammosilene tunicoides is different from Silene rubicunda in the basic chromosome number and the morphology of the nuclei at resting stage and mitotic prophase chromosomes,because Silene rubicund has the basic chromosome number of x=10 and 12,and its nuclei at resting stage and mitotic prophase chromosomes is sparsely diffuse type and continuous type respectivrly.  相似文献
