目的:研究通辽地区女性患者下生殖道人乳头瘤病毒(HPV)感染率和基因型分布。方法:采用凯普导流杂交法,对364例妇科就诊患者宫颈标本进行HPV基因分型检测。结果:HPV基因检测总阳性率为17.03%(62/364),阳性患者中单一感染率为70.97%(44/62),多重感染率为29.03%(18/62);共检出16种HPV亚型,包括高危型12型,低危型4型,感染以HPV31、HPV18、HPV16三种亚型最多见,所占比例分别为13.09%、11.90%、10.71%。结论:通辽地区妇科门诊女性患者HPV感染以HPV31、HPV18、HPV16三型较为多见,常见感染亚型总体符合亚洲人群分布规律,但又具有独特的地区分布特点。  相似文献

目的 了解性病门诊患者人乳头瘤病毒(HPV)的感染情况及其基因型别。方法 用导流杂交分型检测法对1 516例性病门诊患者进行HPV检测与基因分型,评价HPV及其基因型别与疾病类型的关系。结果 1 516例受检者中,HPV总阳性率为55.61%。基因型分析发现,26例鲍温病样丘疹病患者均有高危型HPV感染,而762例尖锐湿疣患者中低危型HPV感染占70.86%。21种HPV基因型均被检出,以HPV6(28.86%)、11(17.23%)、16(10.41%)型感染最为常见。检出的843例阳性患者中单一型别感染占61.68%,以HPV6和HPV11感染为主;HPV多重感染以高低危型HPV混合感染为主。结论 HPV基因型别分布与疾病类型密切相关,HPV亚型的鉴别对相关疾病的预防和治疗具有临床应用价值。  相似文献

目的 对广州市花都区789例疑似人乳头瘤病毒( HPV)感染的妇女进行调查,研究HPV感染状况和基因亚型分布情况.方法 采用荧光定量聚合酶链式反应(Fluorescence quantitative PCR,FQ-PCR)对789例疑似HPV感染患者的宫颈分泌物进行21种人乳头瘤病毒基因亚型的检测.结果 (1)789例标本中,HPV阳性占48.42%(382/789),共检出10种基因型,其中检出单一低危型(HPV6、11、43和44型)279例,占感染者73.04%;单一高危型( HPV16、18、33、52、53、58)68例,占感染者17.80%;多重感染35例,占感染者9.16%.低危型以HPV6、11为主,高危型以HPV16、18为主.HPV6为主要致病基因型,检出率为34.0% (P <0.01);(2)各年龄段间感染率差异无统计学意义(P>0.05).结论 花都区HPV感染率较高,且多为单一轻危型感染,以HPV6为主要感染亚型,HPV基因型分布有助于确定宫颈癌发生的高危因素及筛查高危人群,对本地区HPV疫苗开发、应用具有重大意义.  相似文献

目的 探讨广东韶关地区女性人乳头瘤病毒(HPV)感染现状及病毒基因型分布情况。方法 采用PCR-反向点杂交技术对54 597例女性宫颈脱落细胞标本进行23种HPV基因分型检测。结果 54 597例女性中共检出HPV阳性13 824例,阳性率为25.32%。全部标本中高危型感染占19.91%(10870/54597),主要包含HPV52(2187/54597)、HPV16(1852/54597)、HPV58(1226/54597)、HPV53(801/54597)和HPV68(729/54597)。低危型感染占5.41%(2954/54597),主要包含HPV43(896/54597)和HPV81(833/54597)。感染类型中单一感染占79.22%(8545/10787),二重感染占15.62%(1685/10787),三重及以上感染占5.16%(557/10787)。结论 韶关地区女性HPV感染率较高,且以高危型和单一型感染为主。病毒主要基因型为HPV52、HPV16和HPV58。进行HPV基因分型检测对宫颈癌的防治和疫苗研发接种具有重要意义。  相似文献

应用第二代基因杂交捕获技术(HC-Ⅱ)对7068例21-58岁妇女官颈细胞进行13种高危型人乳头瘤病毒(HPV)DNA的检测。该人群中13种高危型HPVDNA的总检出率为23.45%。在检出的1440例HPV阳性者中,宫颈癌、官颈上皮内高度病变(CINⅡ/Ⅲ)及宫颈上皮内低度病变(CINⅠ)的高危型HPVDNA检出率分别为100%、100%、65.06%。女性生殖道高危型HPV感染是宫颈癌及CIN流行的主要危险因素,提示言颈癌的防治应重点预防HPV感染,而对HPV感染的筛查和密切监测应是已感染高危型HPV的对象;  相似文献

高危型人乳头瘤病毒(humanpa pilloma virus,HPV)的持续感染是导致妇女宫颈癌发生的一个关键因素。HPV感染宫颈上皮细胞后,可以通过抑制免疫应答,在部分个体中建立潜伏感染。高危HPV编码的蛋白在持续感染的过程中,操控了细胞多种重要功能,如凋亡、增殖、细胞周期调控等,使得宫颈上皮细胞的形态学、遗传物质、表观遗传学等都发生重要改变。部分感染人群的宫颈上皮细胞因此会被转化,并在协同因子相互作用下,逐渐转化为宫颈癌。HPV在宫颈癌发生过程中起着重要的作用,本文将对HPV感染致宫颈癌机制最近的研究进展进行综述。  相似文献

目的:调查安康地区女性人乳头瘤病毒(HPV)感染的基因型别及年龄分布特征,分析其与宫颈癌的关系,为宫颈癌防治及HPV疫苗研发提供可靠的依据。方法:收集2010年6月-2012年8月间在本院及安康市部分县级医院妇产科就诊的2736名女性的液基细胞学和组织学标本,分为8个年龄组:16-24岁119例、25-29岁230例、30-34岁343例、35-39岁472例、40-44岁574例、45-49岁512例、50-54岁206例、55-86岁280例,进行病理学分类及HPV分型检测,分析不同年龄组及不同类型宫颈组织中的HPV感染率。结果:2736例女性中发生HPV感染720例(26.32%),共检出21种型别,感染率最高的基因型别是HPV16(25.05%),其他常见型别依次为HPV58、HPV52、HPV6、HPV11。单一感染占76.25%,多重感染占23.75%。HPV感染率在16-24岁、35-39岁和55-86岁三个年龄段出现高峰;而高危型HPV的感染率在35-39岁和55-86岁两个年龄段分别出现高峰。HPV的检出率随着宫颈病变的严重程度而增加,其中正常或炎症人群的HPV感染率显著低于宫颈病变及宫颈鳞状细胞癌患者(均P0.05),且意义未明的不典型鳞状细胞(ASCUS)、CIN1-3及宫颈鳞状细胞癌患者的HPV感染率对比结果存在显著差异(P0.05)。CIN1组、CIN2-CIN3组及宫颈鳞状细胞癌组单一感染率逐渐增加(P0.05),且其二重、三重感染率比较差异均有统计学意义(P0.05)。结论:安康地区HPV16型别感染较广,临床需加强对HPV16型单一感染宫颈病变患者的癌症预防工作。  相似文献

Emergence of methicillin-resistant Staphylococcus aureus (MRSA) and methicillin-resistant coagulase-negative staphylococci (MR-CoNS) in bovine milk is a major public health concern. The primary purpose of this research was to determine molecular genetic characteristics and antibiotic resistance of staphylococcal isolates recovered from milk of mastitic cows in the Shaanxi Province in Northwestern China. One hundred and thirteen methicillin-susceptible Staphylococcus aureus (MSSA), one mecA-positive and phenotype-positive MRSA, seven mecA- and mecC- negative but phenotype-positive MRSA and two MR-CoNS including one oxacillin-susceptible mecA-positive Staphylococcus haemolyticus (OS-MRSH) and one mecA-positive and methicillin-resistant Staphylococcus epidermidis (MRSE) isolates were recovered from 214 quarter milk samples on 4 dairy farms. All above 123 isolates were subjected to antibiotic resistance profiling. S. aureus isolates were also genotyped using the spa typing and the multilocus sequence typing (MLST). Eight MRSA and 2 MR-CoNS isolates were additionally tested for SCCmec types. Resistance was common among isolates against ampicillin or penicillin (80.5%), kanamycin (68.3%), gentamicin (67.5%), tetracycline (43.9%) and chloramphenicol (30.1%). However, no isolate was resistant to vancomycin or teicoplanin. Twenty, 29 and 58 isolates showed resistance to 1, 2 or more than 2 antibiotics, respectively. The predominant multidrug resistance profile was penicillin/ampicillin/kanamycin/gentamicin/tetracycline (46 isolates). Most S. aureus isolates belonged to spa types t524 (n = 63), t11772 (a new type, n = 31) and t4207 (n = 15). At the same time, MLST types ST71 (n = 67) and ST2738 (a new type, n = 45) were identified as dominant sequence types. The mecA-positive and phenotype-positive MRSA isolate had a composite genotype t524-ST71-SCCmecIVa, while 7 mecA-negative but phenotype-positive MRSA isolates were all t524-ST71. The OS-MRSH isolate contained a type V SCCmec cassette, while the MRSE isolate possessed a non-typeable SCCmec. The spa-MLST types t11772-ST2738 (n = 27), t11807-ST2683 (n = 4) and t11771-ST2738 (n = 3) were newly identified genotypes of S. aureus. These new genotypes and multidrug-resistant staphylococci could pose additional threat to animal and human health.  相似文献

The electrical conductivity (EC) of milk is mainly a function of the electrolyte concentration in the milk and therefore raised in mastitis. The present investigation was aimed at elaborating, if possible, a diagnostic model for screening purposes based on EC determinations and consistent with the diagnostic procedures and interpretations commonly used in laboratory milk diagnosis in the Nordic countries (Klastrup 1975). According to this diagnosis (here called reference diagnosis) cell numbers above 300,000/ml (cell count or the corresponding CMT-score) in foremilk quarter samples during the main part of the lactation period and significantly above the lowest value on within-udder comparison during late lactation are considered indicative of mastitis and bacteriological examinations are made when called for.  相似文献

In this study the benefit of metabolome level analysis for the prediction of genetic value of three traditional milk traits was investigated. Our proposed approach consists of three steps: First, milk metabolite profiles are used to predict three traditional milk traits of 1,305 Holstein cows. Two regression methods, both enabling variable selection, are applied to identify important milk metabolites in this step. Second, the prediction of these important milk metabolite from single nucleotide polymorphisms (SNPs) enables the detection of SNPs with significant genetic effects. Finally, these SNPs are used to predict milk traits. The observed precision of predicted genetic values was compared to the results observed for the classical genotype-phenotype prediction using all SNPs or a reduced SNP subset (reduced classical approach). To enable a comparison between SNP subsets, a special invariable evaluation design was implemented. SNPs close to or within known quantitative trait loci (QTL) were determined. This enabled us to determine if detected important SNP subsets were enriched in these regions. The results show that our approach can lead to genetic value prediction, but requires less than 1% of the total amount of (40,317) SNPs., significantly more important SNPs in known QTL regions were detected using our approach compared to the reduced classical approach. Concluding, our approach allows a deeper insight into the associations between the different levels of the genotype-phenotype map (genotype-metabolome, metabolome-phenotype, genotype-phenotype).  相似文献

The maternal effect has been widely proposed to affect the production traits in domestic animals. However, the sequence polymorphisms of mitochondrial DNA (mtDNA) and association with milk production traits in Holstein cows have remained unclear. In this study, we investigated the single nucleotide polymorphisms (SNPs) of mtDNA ATPase 8/6 genes and association with four milk production traits of interest in 303 Holstein cows. A total of 18 SNPs were detected among the 842 bp fragment of ATPase 8/6 genes, which determined six haplotypes of B. taurus (H1-H4) and B. indicus (H5-H6). The mixed model analysis revealed that there was significant association between haplotype and 305-day milk yield (MY). The highest MY was observed in haplotype H4. However, we did not detect statistically significant differences among haplotypes for the traits of milk fat (MF), milk protein (MP), and somatic cell count (SC). The overall haplotype diversity and nucleotide diversity of ATPase 8/6 genes were 0.563 ± 0.030 and 0.00609 ± 0.00043, respectively. The results suggested that mitochondrial ATPase 8/6 genes could be potentially used as molecular marker to genetically improve milk production in Holstein cows.  相似文献

徐家城遗址是甘肃省新发现的含丰富旧石器文化遗物的遗址,主要埋藏于水洛河二级阶地上覆的马兰黄土中,年代在距今4.3~2.3万年间。徐家城遗址的拼合分析显示,4B、4C层剥片与石器加工行为是石制品空间分布形成的主要原因,但可能存在遗址使用时古人类对石制品位置改变的行为;4A层石制品零散分布可能反映了此时遗址处于古人类活动的边缘地带。遗址剥片方法以硬锤锤击法为主,石核剥片序列主要以转向为主,古人类对石料进行单层或多层开发;同时部分石器破碎后古人类可能对其进行再利用。平面分布显示发掘区内可能存在两个相对集中的剥片区域,并且古人类在同一区域剥片和加工石器。然因遗址石制品原料性质、发掘面积等局限性,拼合分析的优势并未在此项研究中尽现。今后此类研究应结合其他研究方法,综合多方面信息解释遗址形成过程、石器技术及遗址空间利用方式。  相似文献
