The aim of this brief analytical review is to highlight and disentangle research dilemmas in the field of exercise addiction. Research examining exercise addiction is primarily based on self-reports, obtained by questionnaires (incorporating psychometrically validated instruments), and interviews, which provide a range of risk scores rather than diagnosis. Survey methodology indicates that the prevalence of risk for exercise addiction is approximately 3 percent among the exercising population. Several studies have reported a substantially greater prevalence of risk for exercise addiction in elite athletes compared to those who exercise for leisure. However, elite athletes may assign a different interpretation to the assessment tools than leisure exercisers. The present paper examines the: 1) discrepancies in the classification of exercise addiction; 2) inconsistent reporting of exercise addiction prevalence; and 3) varied interpretation of exercise addiction diagnostic tools. It is concluded that there is the need for consistent terminology, to follow-up results derived from exercise addiction instruments with interviews, and to follow a theory-driven rationale in this area of research. 相似文献

The study of intrinsic phosphorylation dynamics and kinetics in the context of complex protein architecture in vivo has been challenging: Method limitations have prevented significant advances in the understanding of the highly variable turnover of phosphate groups, synergy, and cooperativity between P-sites. However, over the last decade, powerful analytical technologies have been developed to determine the full catalog of the phosphoproteome for many species. The curated databases of phospho sites found by mass spectrometry analysis and the computationally predicted sites based on the linear sequence of kinase motifs are valuable tools. They allow investigation of the complexity of phosphorylation in vivo, albeit with strong discrepancies between different methods. A series of hypothetical scenarios on combinatorial processive phosphorylation is proposed that are likely unverifiable with current methodologies. These proposed a priori postulates could be considered as possible extensions of the known schemes of the activation/inhibition signaling process in vivo. 相似文献

Neuroimaging studies involving human subjects raise a range of ethics issues. Many of these issues are heightened in the context of neuroimaging research involving persons with mental health disorders. There has been growing interest in these issues among legal scholars, philosophers, social scientists, and as well as neuroimagers over the last decade. Less clear, however, is the extent to which members of the neuroimaging community are engaged with these issues when they undertake their research and report results. In this study, we analyze the peer-reviewed review literature involving fMRI as applied to the study of mental health disorders. Our hypothesis is that, due to the critical orientation of reviews, and the vulnerability of mental health population, the penetrance of neuroethics will be higher in the review literature in this area than it is in the primary fMRI research literature more generally. We find that while authors of reviews do focus a great deal of attention on the methodological limitations of the studies they discussed, contrary to our hypothesis, they do not frame concerns in ethical terms despite their ethical significance. We argue that an ethics lens on such discussion would increase the knowledge-value of this scholarly work. 相似文献

The partial amino acid sequences of 121 rice proteins separated by two-dimensional gel electrophoresis (2D-PAGE), were determined for a protein sequence data file. In the Rice Genome Research Program (RGP), more than 20,000 cDNA clones randomly selected from rice cDNA libraries have been sequenced to construct a cDNA catalog. Complimentary DNAs encoding about 30% of proteins in the protein sequence data file could be identified in the catalog by computer search. It was deduced that 20,000–40,000 genes are present in the rice genome. Only half of about 20,000 cDNAs sequenced in the RGP, corresponding to 1/4–1/2 of genes present in the entire rice genome, should have unique sequences after considering gene redundancy. This is consistent with the fact that the cDNAs encoding about 30% of the sequenced proteins could be identified in the catalog. If the size of the cDNA catalog is enlarged further, cDNAs encoding all proteins separated by 2D-PAGE could be easily identified from the catalog by using the protein sequence data. 相似文献

Inappropriate land use practices, pollutants, exploitation, and overpopulation have simplified stream habitats and degraded water quality worldwide. Management agencies are now being tasked to ameliorate impacts and restore stream “health,” yet there is a dearth of rigorous scientific methods and theory on which to base sound restoration design and monitoring. Despite this, many localized restoration projects are being constructed to stabilize erosion and enhance habitat heterogeneity in streams. Many restoration attempts adopt the paradigm that increasing habitat heterogeneity will lead to restoration of biotic diversity, yet there have been few studies that have manipulated variation of a physical parameter independent of the mean to isolate the effects of heterogeneity per se. We conducted a field experiment to mimic restoration of habitat heterogeneity in a shallow. stony stream. By using an experimental approach rather than a detailed assessment of existing restoration work, we were able to control the starting conditions of replicate riffles so that organism responses could be unambiguously attributed to the heterogeneity treatments. We successfully manipulated the variability of streambed particle sizes and consequently near‐bed flow characteristics of entire riffles. These factors define axes of habitat heterogeneity at scales relevant to the resident macroinvertebrate fauna. Despite this, we were unable to distinguish differences in community structure between high and low habitat heterogeneity treatments. Power analysis indicated that macroinvertebrate populations were more sensitive to individual site conditions at each riffle than to the heterogeneity treatments, suggesting that increasing habitat heterogeneity may be an ineffective technique if the restoration goals are to promote macroinvertebrate recovery in denuded streams. With extremely high variability between replicate riffles, monitoring programs for localized restoration projects or point source impacts are unlikely to detect gradual shifts in community structure until the differences between the reference and treatment sites are extreme. Innovative measurement of other parameters, such as ecosystem function variables (e.g., production, respiration, decomposition), may be more appropriate indicators of change at local scales. 相似文献

This paper presents an overview of the key ethical questions of performing gene editing research on military service members. The recent technological advance in gene editing capabilities provided by CRISPR/Cas9 and their path towards first-in-human trials has reinvigorated the debate on human enhancement for non-medical purposes. Human performance optimization has long been a priority of military research in order to close the gap between the advancement of warfare and the limitations of human actors. In spite of this focus on temporary performance improvement, biomedical enhancement is an extension of these endeavours and the ethical issues of such research should be considered. In this paper, we explore possible applications of CRISPR to military human gene editing research and how it could be specifically applied towards protection of service members against biological or chemical weapons. We analyse three normative areas including risk–benefit analysis, informed consent, and inequality of access as it relates to CRISPR applications for military research to help inform and provide considerations for military institutional review boards and policymakers. 相似文献

The ramifications of a current debate about informant accuracy are shown to extend well beyond informant accuracy per se, since they invite scrutiny of fundamental epistemological assumptions. Accuracy is analyzed as a complex concept that must be evaluated along several dimensions, suggesting that multiple standards of "accuracy" require multiple methods of data collection and interpretation known generally as triangulation. Rapid physiological profiling of heterotrophic microbial communities enables intensive analysis of the factors affecting activity
in aerobic habitats, such as soil. Previous methods for performing such profiling were severely limited due to enrichment
bias and inflexibility in incubation conditions. We tested a new physiological profiling approach based on a microtiter plate
oxygen sensor system (Becton Dickinson Oxygen Biosensor System (BDOBS)), which allows for testing of lower substrate addition
(i.e., lower enrichment potential) and manipulation of physiochemical assay conditions, such as pH and nutrients. Soil microbial
communities associated with a scrub-oak forest ecosystem on Merritt Island Wildlife Refuge in central Florida, USA, were studied
in order to evaluate microbial activity in a nutrient poor soil and to provide baseline data on the site for subsequent evaluation
of the effects of elevated CO2 on ecosystem function. The spatial variation in physiological activity amongst different habitats (litter, bulk soil, and
rhizosphere) was examined as a function of adaptation to local resources (i.e., water soluble extracts of roots and leaf litter)
and the degree of N and P limitation. All the communities were primarily N-limited, with a secondary P limitation, which was
greater in the rhizosphere and bulk soil. The litter community showed greater overall oxygen consumption when exposed to litter
extracts relative to the rhizosphere or soil, suggesting acclimation toward greater use of the mixed substrates in the extract.
Root extracts were readily used by communities from all the habitats with no habitat specific acclimation observed. A priming
effect was detected in all habitats; addition of glucose caused a significant increase in the use of soil organic carbon.
Response to added glucose was only observed with N and P addition, suggesting that C may be lost to the groundwater from these
porous soils because nutrient limitation prevents C immobilization. 相似文献

The Simon effect shows that choice reactions are faster if the location of the stimulus and the response correspond, even when stimulus location is task-irrelevant. The Simon effect raises the question of what factors influence spatial coding. Until now, the effects of handedness, responding hand, and visual field were addressed in separate studies that used bimanual and unimanual tasks, providing inconclusive results. Here we aimed to close this empirical gap by looking at the effects of these variables in the same study. We used a unimanual version of a Simon task with four groups of participants: left-handed and right-handed, responding with the dominant or nondominant hand. Our results show that the Simon effect is substantially reduced in the field of the responding hand for all groups of participants, except for left-handed individuals responding with the left-hand. These findings highlight the importance of attention mechanisms in stimulus-response coding. They reflect that stimulus-response interference is influenced by hierarchical activation of response units. At a practical level, these findings call for a number of methodological considerations (e.g., handedness, responding hand, and visual field) when using stimulus-response conflict to address spatial coding and cognitive control functions in neurological populations. 相似文献

The diversity of online resources storing biological data in different formats provides a challenge for bioinformaticians to integrate and analyse their biological data. The semantic web provides a standard to facilitate knowledge integration using statements built as triples describing a relation between two objects. WikiPathways, an online collaborative pathway resource, is now available in the semantic web through a SPARQL endpoint at http://sparql.wikipathways.org. Having biological pathways in the semantic web allows rapid integration with data from other resources that contain information about elements present in pathways using SPARQL queries. In order to convert WikiPathways content into meaningful triples we developed two new vocabularies that capture the graphical representation and the pathway logic, respectively. Each gene, protein, and metabolite in a given pathway is defined with a standard set of identifiers to support linking to several other biological resources in the semantic web. WikiPathways triples were loaded into the Open PHACTS discovery platform and are available through its Web API (https://dev.openphacts.org/docs) to be used in various tools for drug development. We combined various semantic web resources with the newly converted WikiPathways content using a variety of SPARQL query types and third-party resources, such as the Open PHACTS API. The ability to use pathway information to form new links across diverse biological data highlights the utility of integrating WikiPathways in the semantic web. 相似文献

We document the phylogenetic behavior of the 18S rRNA molecule in 67 taxa from 28 metazoan phyla and assess the effects of
among-site rate variation on reconstructing phylogenies of the animal kingdom. This empirical assessment was undertaken to
clarify further the limits of resolution of the 18S rRNA gene as a phylogenetic marker and to address the question of whether
18S rRNA phylogenies can be used as a source of evidence to infer the reality of a Cambrian explosion. A notable degree of
among-site rate variation exists between different regions of the 18S rRNA molecule, as well as within all classes of secondary
structure. There is a significant negative correlation between inferred number of nucleotide substitutions and phylogenetic
information, as well as with the degree of substitutional saturation within the molecule. Base compositional differences both
within and between taxa exist and, in certain lineages, may be associated with long branches and phylogenetic position. Importantly,
excluding sites with different degrees of nucleotide substitution significantly influences the topology and degree of resolution
of maximum-parsimony phylogenies as well as neighbor-joining phylogenies (corrected and uncorrected for among-site rate variation)
reconstructed at the metazoan scale. Together, these data indicate that the 18S rRNA molecule is an unsuitable candidate for
reconstructing the evolutionary history of all metazoan phyla, and that the polytomies, i.e., unresolved nodes within 18S
rRNA phylogenies, cannot be used as a single or reliable source of evidence to support the hypothesis of a Cambrian explosion.
