青海可可西里嗜碱芽胞杆菌资源调查   总被引:3,自引:0,他引:3  
【目的】了解可可西里嗜碱芽胞杆菌资源多样性及产酶多样性,为芽胞杆菌功能资源挖掘和菌剂开发提供基础。【方法】采用Horikoshi I培养基,通过可培养法分离青海可可西里土壤中的嗜碱芽胞杆菌,利用16S r RNA基因序列初步鉴定分离获得的芽胞杆菌。采用透明圈法分析分离菌株的产蛋白酶、纤维素酶及木聚糖酶活性。【结果】从青海可可西里土壤中共分离获得66株嗜碱芽胞杆菌,根据16S r RNA基因序列相似性分析,发现它们隶属于6个属22个种,分别为芽胞杆菌属(Bacillus)、纤细芽胞杆菌属(Gracilibacillus)、喜盐芽胞杆菌属(Halobacillus)、咸海鲜芽胞杆菌属(Jeotgalibacillus)、类芽胞杆菌属(Paenibacillus)和嗜冷芽胞杆菌属(Psychrobacillus),其中以芽胞杆菌属(Bacillus)为优势属。2株嗜碱芽胞杆菌与它们最近匹配模式菌株的16S r RNA基因序列相似性为97.00%和98.65%,为潜在新种。三种酶活检测结果表明产酶菌株约占总分离菌株的95.00%,其中55株具有产蛋白酶活性,27株具有产纤维素酶活性,8株能够产木聚糖酶。【结论】青海可可西里蕴藏着较丰富的嗜碱芽胞杆菌资源及丰富的产酶资源,为后续嗜碱芽胞杆菌的挖掘提供理论基础。  相似文献

利用纯培养和筛选培养,从思茅松毛虫幼虫肠道中分离得到7株产脂肪酶的菌株.通过提取基因组DNA并进行16S rDNA序列测定,构建产酶菌株的系统发育树,初步鉴定结果显示:菌株D2、D12、D19属于假单胞菌属(Pseudomonas sp.),菌株D7、D17属于芽胞杆菌属(Bacillus sp.),菌株D9、D16属于克雷伯氏菌(Klebsiella sp.).初步研究所产脂肪酶的酶学性质,确定这些酶的最适作用温度30~40℃、最适作用pH值8.0~9.0,为中温碱性脂肪酶.  相似文献

为了解淡水湖渔场底泥中产几丁质酶菌株的产酶量和分布情况,对环洞庭湖的4个淡水湖渔场的表层底泥样品进行了无菌采集。利用稀释涂布平板法、点种法和摇瓶发酵法从底泥样品中筛选分离到26株产几丁质酶菌株,几丁质酶活在0.07~0.69 U/mL之间。对26株产几丁质酶菌株进行16S rRNA基因鉴定和系统发育分析。结果表明,26株菌株都分布于变形菌门(Proteobacteria)和厚壁菌门(Firmicutes)的芽胞杆菌属(Bacillus)、假单胞菌属(Pseudomonas)、不动杆菌属(Acinetobacter)和微小杆菌属(Exiguobacterium)。且产几丁质酶细菌在4个淡水湖渔场表层底泥中的分布情况为安乐湖>东湖>北民湖>西湖。对产几丁质酶菌株的降解活力、种类组成及数量分布的研究可为淡水湖渔场底泥中产几丁质酶微生物资源的开发及应用提供参考。  相似文献

摘要:目的 分离小菌虫肠道可培养细菌,并研究其产消化酶活性,探讨肠道细菌对小菌虫消化食物的影响。方法 采用传统细菌分离培养方法分离小菌虫肠道细菌,利用16S rDNA序列进行细菌分子鉴定;利用筛选培养基鉴别各细菌的产蛋白酶、脂肪酶、淀粉酶和纤维素酶活性。结果 在小菌虫肠道中分离到4种可培养细菌,分别是枯草芽胞杆菌(Bacillus subtilis)、肺炎克雷伯菌(Klebsiella pneumoniae)、Pseudocitrobacter faecalis和芽胞杆菌(Bacillus sp.)。其中,2种芽胞杆菌属细菌有产消化酶活性。枯草芽胞杆菌有产纤维素酶、淀粉酶和蛋白酶活性;芽胞杆菌仅有产蛋白酶活性,但产酶能力低于枯草芽胞杆菌。结论 小菌虫肠道细菌中可培养细菌结构简单,但其中的芽胞杆菌属细菌有产纤维素酶、淀粉酶和蛋白酶能力,说明小菌虫肠道中的2种芽胞杆菌属细菌可能有协助小菌虫进行食物消化的功能。  相似文献

通过组织研磨法对南方红豆杉(Taxus wallichiana var. mairei)新鲜组织进行内生细菌的分离,利用16S rRNA测序技术对分离的内生细菌进行鉴定分析,使用高效液相色谱-高分辨飞行质谱测定菌液提取物IAA含量,利用HPLC-TOF-MS对提取物脂肽类化合物进行分离、鉴定,结合数据库筛选菌株产脂肽类化合物种类及含量差异。结果表明,从南方红豆杉根茎叶中共分离鉴定出内生细菌31株,筛选出产IAA菌株20株,其中编号为KLBMPTC10的芽胞杆菌属菌株产IAA含量高达210.3955 μg·L-1。在对产IAA菌株所产脂肽类化合物的研究中发现,菌株KLBMPTC10是所有产IAA内生细菌中脂肽类化合物含量最丰富的,其产Iturin A2相对含量为26.06%,明显优于其他产IAA菌株。芽胞杆菌属菌株KLBMPTC10同时具备高产IAA和脂肽类化合物的能力,可作为研究菌株进一步开发。  相似文献

目的 对云南兰坪铅锌尾矿区样品中的可培养细菌进行分离及初步鉴定,同时挖掘具有酶活性功能的菌株。方法 从兰坪铅锌尾矿区及周边农田采集了20份样品,运用10种培养基进行细菌分离,对分离菌株的16S rRNA基因测序以鉴定其分类地位,再以平板透明圈法检测分离菌株的淀粉酶、纤维素酶、蛋白酶和脂肪酶活性。结果 从20份样品中分离获得了320株细菌,隶属于6个纲、14个目、26个科、39个属,有5个潜在新分类单元。其中链霉菌属的菌株数最多,高达102株,占菌株总数的31.88%,为优势种群;其次为芽胞杆菌属菌株40株,肠杆菌属菌株17株。去重后对165株菌进行酶活检测,有41株菌具有淀粉酶活性,占筛选菌株总数的24.70%,主要为链霉菌属和芽胞杆菌属;有40株菌对纤维素酶具有活性,占筛选菌株总数的24.10%,主要为链霉菌属和芽胞杆菌属;有14株菌对蛋白酶具有活性,占筛选菌株总数的8.43%,主要为链霉菌属和芽胞杆菌属;有12株菌对脂肪酶具有活性,占筛选菌株总数的7.23%,主要为链假单胞菌属。结论 兰坪铅锌尾矿区可培养细菌的种类丰富,且蕴藏着大量具有酶活性的菌株。研究结果为了解兰坪铅锌尾矿细菌多...  相似文献

目的筛选并鉴定一种产纤维素酶能力较高的菌株,为纤维素的高效利用贮备菌源。方法用羧甲基纤维素钠(CMC-Na)平板筛选产纤维素酶菌株,通过LB培养基对其进行纯化,16SrDNA基因序列分析其分类地位,3,5-二硝基水杨酸法(DNS)测定其产酶能力。结果分离纯化得到的产纤维素酶菌株(S1)为芽胞杆菌属(Bacillus genus)的短小芽胞菌,在最佳产酶条件下产酶含量达到1 204U/mL,产纤维素酶能力与里氏木霉(Trichoderma reesei)相当,但其产酶速率较里氏木霉低。结论 S1是一株产纤维素酶能力较高的菌株,产酶条件温和,初步鉴定为一种新种,具有较高研究及应用价值。  相似文献

从浙江省金华市花生地土样中筛选、分离纯化得到1株产聚-β-羟基丁酸编号为PX-95的芽胞杆菌,PHB产量为135.81 mg/L。根据形态特征、生理生化特征初步鉴定为芽胞杆菌属蜡样芽胞杆菌群,16S rDNA序列分析显示PX-95菌株与苏云金芽胞杆菌以及蜡样芽胞杆菌均具有高度同源性,最后采用扩增gyrB(DNA促旋酶B亚单位)基因的方法将其鉴定为苏云金芽胞杆菌(Bacillus thuringiensis)。  相似文献

目的获得具有良好性状的产细菌素菌株。方法测定菌株形态学和生理生化学特性以进行菌种鉴定;采用牛津杯法测定抑菌谱及细菌素的理化特性。结果经鉴定实验菌株为凝结芽胞杆菌,所产细菌素在100℃加热60min后,残余活力为90%;在pH5~9范围内,抗菌活力稳定。结论凝结芽胞杆菌G1可产性能良好的抑制革兰阳性菌生长的细菌素。  相似文献

从南方酸性水稻土和腐烂秸秆中采集样品,经初筛、复筛获得4株产生明显水解圈且D/d值较大同时传代效果稳定的酸性半纤维素降解细菌。DNS法测定4株细菌半纤维素酶活力,最终结合其产酶速度及酶活大小得到菌株NB8,液体摇瓶发酵72 h其半纤维素酶活达85.12 U/mL。经形态学观察和生理生化指标测定,初步确定该菌为芽胞杆菌属。结合16S rDNA基因序列分析结果,可以初步鉴定NB8为短小芽胞杆菌(Ba-cillus pumilus)。  相似文献

Aims:  Isolation and characterization of an agarase-producing bacterium Agarivorans sp. HZ105.
Methods and Results:  An agarase-producing bacterium strain HZ105 had been isolated from marine sediment sample. Based on phylogenetic analysis of the 16S rRNA gene sequence and phenotypic analysis, as well as biochemical analyses, this strain was named Agarivorans sp. HZ105. Effect of pH, NaCl on the growth and agarase production of strain HZ105 was studied. Strain HZ105 produced three extracellular agarases which were purified to homogeneity from bands in the PAGE gel. Two agarases of these three had a molecular mass of 54, 58 kDa, respectively. And the MS and MS/MS spectra were used to identify the agarases.
Conclusions:  The MS spectra result showed that the agarases of strain HZ105 should be beta-agarase and belong to the family 50 of glycosyl hydrolases. The agarases could keep stable activity at room temperature.
Significance and Impact of the Study:  The strain HZ105 was useful to produce stable agarases. The solution produced by agar's degradation in the agar plates was first reported to be used for purification of agarase. Agarases were purified to homogeneity directly from the PAGE gel without stained by Coomassie brilliant blue.  相似文献

A strain of bacterium producing antifungal antibiotic was isolated and identification of the strain was attempted. We could identify the bacterium as being a Bacillus sp., based on morphological observation, physiological characteristics, and 16S rDNA sequence analysis, thus leading us to designate the strain as Bacillus sp. AH-E-1. The strain showed potent antibiotic activity against phytopathogenic and human pathogenic fungi by inducing mycelial distortion and swelling and inhibiting spore germination. The antibiotic metabolite produced by the strain demonstrated excellent thermal and pH (2-11) stability, but was labile to autoclaving. From these results, we could find a broader antifungal activity of Bacillus genus. Isolation and characterization of the active agent produced by the strain are under progress.  相似文献

A novel bacterium, strain BMP-1(T), was isolated from a continuous wastewater treatment culture system operating with a bacterial consortium. Cells of the isolate were Gram-variable, aerobic, moderately halotolerant, motile and endospore-forming rods. Strain BMP-1(T) grew chemolithoautotrophically by oxidation of thiosulfate to sulfate with a growth yield of 1.07 g protein mol(-1) of thiosulfate consumed. DNA G+C content was 43.8 mol%. Its cell wall had peptidoglycan based on m-diaminopimelic acid, and the major component of fatty acid was C(15 : 0). The 16S rRNA gene analysis showed that strain belongs to the genus Bacillus, sharing a 99.5% of sequence similarity with Bacillus jeotgali CCM 7133(T). DNA-DNA hybridization between the isolate of this study and this strain was 44%. Thus, the inclusion of strain BMP-1(T) in the genus Bacillus is suggested as a novel species and the name Bacillus thioparus sp. nov. (Type strain BMP-1(T)=BM-B-436(T)=CECT 7196(T)) is proposed. The sequence of the 16S rRNA gene has been deposited in GenBank with accession number DQ371431.  相似文献

松材线虫生防细菌的筛选、鉴定及其毒性因子的初步研究   总被引:4,自引:0,他引:4  
从河南南阳不同农田的植物根部采取土壤样本,共分离获得了198株细菌。通过毒性测试和平板生测从中筛选出松材线虫生防细菌6株,其中NS-3菌株对松材线虫的杀灭活性最高。结合该菌株的形态学、生理学特征及16S rDNA序列分析等结果将该菌株归为芽孢杆菌属,命名为Bacillus sp.strain NS-3。将该细菌液体培养的上清液和上清蛋白粗提物分别处理松材线虫48h后线虫的死亡率分别达到50%和100%;线虫死亡后虫体消解。然而,细菌的上清蛋白粗提物经煮沸变性后,基本丧失了对松材线虫的侵染活性,结果显示细菌Bacillus sp.strain NS-3的杀线虫活性物质主要要存在于细菌培养上清液中,并且为热不稳定性物质。  相似文献

【目的】从运城盐湖中分离获得一株耐盐细菌LG,对其进行分类鉴定及抗菌特性研究。【方法】利用16S rRNA基因序列分析对菌株进行分类鉴定。以金黄色葡萄球菌为指示菌,采用杯碟法对菌株LG发酵上清液进行抗菌活性检测,利用扫描电镜和透射电镜观察其抗菌效果。研究不同因素对上清液抗菌活性的影响,并采用PCR技术对菌株基因组进行功能基因筛查。【结果】系统发育分析表明该菌为Bacillus属成员,在0–25%的NaCl浓度范围内生长良好,为耐盐细菌。电镜观察发现,菌株LG发酵上清液处理金黄色葡萄球菌可导致细胞结构明显出现异常,细胞质泄漏。抗菌稳定性研究表明,菌株LG发酵上清液活性稳定,表现出了良好的对紫外光、温度、pH和NaCl的耐受性。采用特异性引物,通过PCR筛查发现菌株LG基因组中含有聚酮合酶(PKS I)基因和非核糖体肽合成酶(NRPS)基因,表明该菌具有产多种代谢产物的潜力。【结论】极端环境中的微生物资源可作为抗菌活性物质的潜在新来源。  相似文献

An endospore-forming bacterium, designated strain B-16T, was isolated from a forest soil sample in Yunnan, China. The isolate presented remarkable nematotoxic activity against nematode Panagrellus redivivus. The organism was strictly aerobic, motile, spore forming and rod shaped, catalase- and oxidase-positive. The predominant isoprenoid quinone was menaquinone 7 (MK-7). The major cellular fatty acid profiles were anteiso-C15:0 (48.67%), iso-C15:0 (13.45%), C16:0 (9.06%) and anteiso-Cl7:0 (8.29%). The DNA G+C content was 46%. Phylogenetic analyses based on 16S rDNA sequence revealed that isolate belongs to the genus Bacillus. Strain B-16T exhibited high 16S rDNA similarity with its closest neighbors Bacillus vallismortis (99.79%), B. subtilis (99.43%), B. atrophaeus (99.43%), B. amyloliquefaciens (99.36%), B. licheniformis (98.0%) and less than 97.0% with all the other relative type strains in the genus Bacillus. The phenotypic and genotypic characteristics and DNA-DNA relatedness data indicate that strain B-16T should be distinguished from all the relative species of genus Bacillus. Therefore, on the basis of the polyphasic taxonomic data presented, a new species of the genus Bacillus, B. nematocida, with the type strain B-16T ( = CGMCC 1128T) is proposed. The taxonomic position of a novel halophilic endospore-forming bacterium previously isolated from a desert iguana was investigated by 16S rRNA gene sequencing. Comparative sequence analyses showed the unidentified bacterium to be phylogenetically loosely associated with some other spore-forming ( Bacillus pantothenticus, Sporosarcina halophila ) and non-spore-forming ( Marinococcus albus ) halotolerant bacteria. Based on the phenotypic and phylogenetic distinctiveness of the unidentified bacterium, it is proposed that it is classified in the genus Bacillus as a new species, Bacillus dipsosauri.  相似文献

Abstract A new obligately anaerobic, extremely thermophilic, cellulolytic bacterium is described. The strain designated Tp8T 6331 is differentiated from thermophilic cellulolytic clostridia on the basis of physiological characteristics and phylogenetic position within the Bacillus/Clostridium subphylum of the Gram-positive bacteria. Strain Tp8T 6331 is assigned to a new genus Caldicellulosiruptor , as Caldicellulosiruptor saccharolyticus gen., nov., sp. nov.  相似文献

A slightly yellowish, Gram-positive, filamentous with 'cross-like' branching, aerobic, spore-forming bacterium was isolated from enrichment culture during degradation of the thallus of the brown alga Fucus evanescens. The bacterium studied was chemoorganotrophic, tolerant to 3% NaCl, alkalitolerant, and alginolytic. The predominant cellular fatty acid was ai15:0 which accounted more than 65% of total fatty acids, while i14:0, il5:0 i16:0, and ai17:0 made up 25%. DNA base composition was 37 mol% GC. Phylogenetic analysis of 16S rDNA gene revealed that this isolate was a member of the genus Bacillus, with no close relatives at the species level (16S rRNA gene sequence similarity less 97%). On the basis of the significant differences demonstrated in the phenotypic and chemotaxonomic characteristics, it is suggested that the bacterium be classified as a novel species; the name Bacillus algicola sp. nov. is proposed. The taxonomic position of a Gram-positive, endospore-forming bacterium isolated from soil sample collected from an industrial site was analyzed by a polyphasic approach. The strain designated as IITR-54T matched most of the phenotypic and chemical characteristics of the genus Bacillus and represents a novel species. It was found to biodegrade 4-chlorobiphenyl through dechlorination and was isolated through enrichment procedure from an aged polychlorinated biphenyl-contaminated soil. Both resting cell assay and growth under aerobic liquid conditions using 4-chlorobiphenyl as sole source of carbon along with 0.01 % yeast extract, formation of chloride ions was measured. 16S rRNA (1,489 bases) nucleotide sequence of isolated strain was compared with those of closely related Bacillus type strains and confirmed that the strain belongs to the genus Bacillus. Strain IITR-54T differs from all other species of Bacillus by at least 2.1 % at the 16S rRNA level, and the moderately related species are Bacillus oceanisediminis (97.9 %) followed by Bacillus infantis (97.7 %), Bacillus firmus (97.4 %), Bacillus drentensis (97.3 %), Bacillus circulans (97.2 %), Bacillus soli (97.1 %), Bacillus horneckiae (97.1 %), Bacillus pocheonensis (97.1 %) and Bacillus bataviensis (97.1 %), respectively. The cell wall peptidoglycan contained meso-diaminopimelic acid and the major isoprenoid quinone was MK-7. Major fatty acids are iso-C15:0 (32.4 %) and anteiso-C15:0 (27.4 %). Predominant polar lipids are diphosphatidylglycerol, phosphatidylglycerol and phosphatidylethanolamine. The results of physiological and biochemical tests allowed the genotypic and phenotypic distinctiveness of strain IITR-54T with its phylogenetic relatives and suggest that the strain IITR-54T should be recognized as a novel species, for which the name Bacillus mesophilum sp. nov. is proposed. The type strain is IITR-54T (= MTCC 11060T = JCM 19208T).  相似文献
