Summary A new variant band in the Bf system has been found in the serum of three individuals belonging to a tribe of Brazilian Indians (Karaja—Bananal—Goias) and in the serum of a Caucasian individual from the area of Strasbourg. Summary A variant band in the Bf polymorphism has been found in the serum of a healthy women, her mother, her sister, and two brothers. By direct comparison the band was found to migrate faster than F but slower than F1. It is highly probable that the band represents a new allele at the Bf locus, Bf 0.55. The authors have studied the genetic polymorphism of the properdin factor B (Bf) by the isoelectrofocusing technique. The SS phenotypes, all similar on agarose gel electrophoresis, were shown to be heterogeneous after isoelectrofocusing; this heterogeneity corresponds to the expression of two new suballeles SA and SB, inherited in a codominant manner. Gene frequencies for 121 individuals with SS phenotype are 0.57 for SA, and 0.43 for SB.  相似文献

Three different population samples have been tested for properdin factor B markers: 395 individuals from Schleswig-Holstein (Germany), 343 individuals (Europids) from Southern Brazil, and 309 individuals (Negroids) from Guinea-Bissau (Western Africa). These samples are showing marked differences in the distribution of Bf gene frequencies. As for the sample from Southern Brazil the Bf data are confirming the assumption that the Caucasoid population in Southern Brazil is somewhat mixed with Negroids.  相似文献

The green fluorescent protein (GFP) from the jellyfish Aequorea victoria has provided a myriad of applications for biological systems. Over the last several years, mutagenesis studies have improved folding properties of GFP (refs 1,2). However, slow maturation is still a big obstacle to the use of GFP variants for visualization. These problems are exacerbated when GFP variants are expressed at 37 degrees C and/or targeted to certain organelles. Thus, obtaining GFP variants that mature more efficiently is crucial for the development of expanded research applications. Among Aequorea GFP variants, yellow fluorescent proteins (YFPs) are relatively acid-sensitive, and uniquely quenched by chloride ion (Cl-). For YFP to be fully and stably fluorescent, mutations that decrease the sensitivity to both pH and Cl- are desired. Here we describe the development of an improved version of YFP named "Venus". Venus contains a novel mutation, F46L, which at 37 degrees C greatly accelerates oxidation of the chromophore, the rate-limiting step of maturation. As a result of other mutations, F64L/M153T/V163A/S175G, Venus folds well and is relatively tolerant of exposure to acidosis and Cl-. We succeeded in efficiently targeting a neuropeptide Y-Venus fusion protein to the dense-core granules of PC12 cells. Its secretion was readily monitored by measuring release of fluorescence into the medium. The use of Venus as an acceptor allowed early detection of reliable signals of fluorescence resonance energy transfer (FRET) for Ca2+ measurements in brain slices. With the improved speed and efficiency of maturation and the increased resistance to environment, Venus will enable fluorescent labelings that were not possible before.  相似文献

Summary A healthy, 10-year-old male born in Milano to a family from Southern Italy (Puglia) was found to be a homozygote for an albumin variant of the fast type. His parents are half-first cousins. Their common grandfather, the parents, one sister, and three brothers of the child were all heterozygotes for the same albumin variant. This seems to be the first case of a homozygote for a fast albumin variant described in Europe.  相似文献

The distribution of properdin factor B (Bf) phenotypes and the gene frequencies were investigated in 918 Saudi Arabs. A high frequency of the 'rare' allele BFS0.7 was observed BfS0.7 = 0.1514). The frequencies of the common Bf alleles (BfS = 0.5174, BfF = 0.3213) are outside the corresponding ranges of BfS, BfF gene frequencies found in European Caucasoids.  相似文献

Bovine beta-lactoglobulin (BLG) is a globular protein of uncertain physiological function and a member of the lipocalin superfamily of proteins. Here, we present the X-ray structure at 3.0 angstroms of BLG (variant A) from an orthorhombic (P2(1)2(1)2(1)) pseudo-tetragonal crystal form that suffers from pseudo-merohedral twinning (final R(working) = 0.224, R(free) = 0.265). Crystals were grown by dialysis against ultra-purified water (i.e., at very low ionic strength), at pH approximately 5.2 (approximately pI), conditions vastly different from all other BLG structures determined previously. This allows critical assessment of the BLG structure and of the influence that pH, ionic strength, and crystal packing may have on the molecular structure of BLG. The pH-sensitive EF loop is found in the closed conformation characteristic of BLG at pH less than 7 and moderate to high ionic strength. Although the hydrophobic pocket appears to be empty, the presence of highly disordered water molecules cannot be excluded. The dimer interface and the hydrophobic pocket (calyx) are preserved. However, the orientation of the subunits in the dimer varies considerably with crystal form. Structure is deposited with PDB ID 2akq.  相似文献

Conjugative plasmids can mediate gene transfer between bacterial taxa in diverse environments. The ability to donate the F-type conjugative plasmid R1 greatly varies among enteric bacteria due to the interaction of the system that represses sex-pili formations (products of finOP) of plasmids already harbored by a bacterial strain with those of the R1 plasmid. The presence of efficient donors in heterogeneous bacterial populations can accelerate plasmid transfer and can spread by several orders of magnitude. Such donors allow millions of other bacteria to acquire the plasmid in a matter of days whereas, in the absence of such strains, plasmid dissemination would take years. This "amplification effect" could have an impact on the evolution of bacterial pathogens that exist in heterogeneous bacterial communities because conjugative plasmids can carry virulence or antibiotic-resistance genes.  相似文献

Summary: Zerg is a library of sub-routines that parses the output from all NCBI BLAST programs (Blastn, Blastp, Blastx, Tblastn and Tblastx) and returns the attributes of a BLAST report to the user. It is optimized for speed, being especially useful for large-scale genomic analysis. Benchmark tests show that Zerg is over two orders of magnitude faster than some widely used BLAST parsers. Sulphate induces very fast cell rounding and detachment   总被引:1,自引:0,他引:1  
Within a few minutes of incubation with SO4(2-), cultured monolayer cells retract into round shapes with drastically reduced surface area. Concomitant elevation of phosphoinositide second messenger levels, viz, 1,2-diacylglycerol (DAG) and inositol 1,4,5-trisphosphate (IP3), is observed. A causal relationship with sulphation seems to be suggested by finding (a) sulphation of an added acceptor, 4-methylumbelliferone, (b) sulphation of endogenous glycosaminoglycan (GAG) polymers, (c) inhibition by phenol sulphotransferase inhibitor, DCNP (2,6-dichloro-4-nitrophenol). DCNP also inhibits the second messenger production and cell rounding. Reduced surface area appears to be caused by massive plasma membrane internalization in a distinctive endocytosis which is also seen in cell rounding from directly imposed ionic gradients. Reducing the surface area would reduce the adhesive or attachment sites. Besides demonstrating a highly efficient cell detachment potential, huge macromolecules appear to be readily internalized. The association of sulphation, signal transduction and cell detachment is novel.  相似文献

Two moulds — Aspergillus flavus and Alternaria tenuissima — were isolated from wheat-based fast foods in Nigeria and their cultural conditions determined. A warm ascorbic acid environment is preferred by A. flavus but aeration enhances the growth of both organisms. At 45°C, Al. tenuissima is unaffected by vitamins. The minimum inhibitory concentrations of sodium chloride for A. flavus and Al. tenuissima are 5% and 10% w/v respectively. Lysine, casein hydrolysate, phenylalanine, raffinose, rhamnose and stachyose enhance growth in both species.  相似文献

The glyoxalase I polymorphism was studied in 1,490 unrelated subjects from three different areas of Italy (Milan, Rome and Naples). The following frequencies of GLO2 allele were observed: Milan 0.57, Rome 0.59, Naples 0.61, and a significant difference between Milan and Naples was found (p less than 0.02). An anomalous phenotype is also described which permits us to postulate a new allele producing an isozyme with low activity and electrophoretically slower than those corresponding to GLO1 and GLO2 alleles.  相似文献

A new, simple, and very sensitive assay for phospholipase A and C is described. The assay is based on the bioluminescence developed by the mutant of the bacterium Beneckea harveyi as a response to myristic acid released from dimyristoyl phosphatidylcholine by either phospholipase A or by a phospholipase C-lipase coupled system. It is possible to assay these enzymes at a rate corresponding to a release of as little as 1 to 2 pmol of myristic acid per minute.  相似文献

The genetic polymorphism of human plasminogen (PLG) was investigated in Libyans using wide-scale ultrathin-layer polyacrylamide isoelectric focusing with subsequent immunoblotting. The 2 common alleles, PLG*A and PLG*B, and 4 previously reported rare ones, PLG*A3, PLG*M4, PLG*B1 and PLG*B2, were observed. In addition, a new intermediate rare allele designated PLG*MTripoli (PLG*MT) was encountered. The estimated allele frequencies for the genes PLG*A, PLG*B, PLG*A3, PLG*MT, PLG*M4, PLG*B1 and PLG*B2 were 0.6409, 0.3091, 0.0182, 0.0045, 0.0091, 0.0045 and 0.0136, respectively. The isolated probability of exclusion in cases of disputed paternity among Libyans is 23.3%.  相似文献
