Due to the accumulating evidence that suggests that numerous unhealthy conditions in the indoor environment are the result
of abnormal growth of the filamentous fungi (mold) in and on building surfaces it is necessary to accurately determine the
organisms responsible for these maladies and to identify them in an accurate and timely manner. Historically, identification
of filamentous fungal (mold) species has been based on morphological characteristics, both macroscopic and microscopic. These
methods may often be time consuming and inaccurate, necessitating the development of identification protocols that are rapid,
sensitive, and precise. To this end, we have devised a simple PAN-PCR approach which when coupled to cloning and sequencing
of the clones allows for the unambiguous identification of multiple fungal organisms. Universal primers are used to amplify
ribosomal DNA sequences which are then cloned and transformed into Escherichia coli. Individual clones are then sequenced and individual sequences analyzed and organisms identified. Using this method we were
capable of identifying Stachybotrys chartarum, Penicillium purpurogenum, Aspergillus sydowii, and Cladosporium cladosporioides from a mixed culture. This method was found to be rapid, highly specific, easy to perform, and cost effective. 相似文献

The respiration kinetics of Aspergillus oryzae on different varieties of whole wheat kernels were studied. Six wheat varieties were pretreated in two different ways. Five of the six substrates fermented similarly and independently of the pretreatment method. However, pretreatment affected fermentation of one variety of soft wheat (Apollo). T21H-NMR imaging of the water inside the kernels showed a change in water binding inside the kernels when a different pretreatment strategy was used. Differences in free sugar or amino acid content or in kernel stiffness were not significant. 相似文献

Buildings with poor indoor air quality (IAQ) frequently have many areas with surface fungal contamination. Studies have demonstrated that certain fungal genera (e.g., Cladosporium, Penicillium, and Stachybotrys) are able to grow on building materials such as wallpaper, drywall, and ceiling tiles, particularly after water damage has occurred. Due to the increasing awareness of sick building syndrome (SBS), it has become essential to identify building materials that prevent the interior growth of fungi. The objective of this study was to identify building materials that would not support the growth of certain fungal genera, regardless of whether an external food source was made available. The growth of three fungal genera (Cladosporium, Penicillium, and Stachybotrys) was evaluated on cellulose-containing ceiling tile (CCT) and inorganic ceiling tile (ICT). Both types of ceiling tile were exposed to environmental conditions which can occur inside a building. Our results show that ICT did not support the growth of these three fungal genera while CCT did. Our data demonstrate that ICT could serve as an ideal replacement for CCT. This revised version was published online in June 2006 with corrections to the Cover Date. 相似文献

Carbon catabolite repression and pH regulation are regulatory circuits with a wide domain of action in the Plectomycetes. Penicillin biosynthesis is one of the pathways which are under their control. The conclusions obtained so far, which are based on studies of the genetic and molecular regulation of the penicillin pathway of Aspergillus nidulans, would have been much harder to produce using an organism such as Penicillium chrysogenum (the industrial penicillin producer). However, A. nidulans and P. chrysogenum are close in terms of their phylogeny and one can reasonably predict that the conclusions about A. nidulans, which are summarized in this review and which are of unquestionable biotechnological relevance, will be extrapolable to the industrial organism. 相似文献

Stachybotrys chartarum is a fungal species that can produce mycotoxins, specifically trichothecenes. Exposures in the indoor environment have reportedly induced neurogenic symptoms in adults and hemosiderosis in infants. However, little evidence has linked measured exposures to any fungal agent with any health outcome. We present here a study that focuses on quantitatively assessing the health risks from fungal toxin exposure. Male, 10 week old Charles River-Dawley rats were intratracheally instilled with approximately 9.6 million Stachybotrys chartarum spores in a saline suspension. The lungs were lavaged 0 h (i.e., immediately post-instillation), 6, 24 or 72 h after instillation. Biochemical indicators (albumin, myeloperoxidase, lactic dehydrogenase, hemoglobin) and leukocyte differentials in the bronchoalveolar lavage fluid and weight change were measured. We have demonstrated that a single, acute pulmonary exposure to a large quantity of Stachybotrys chartarum spores by intratracheal instillation causes severe injury detectable by bronchoalveolar lavage. The primary effect appears to be cytotoxicity and inflammation with hemorrhage. There is a measurable effect as early as 6 h after instillation, which may be attributable to mycotoxins in the fungal spores. The time course of responses supports early release of some toxins, with the most severe effects occurring between 6 and 24 h following exposure. In the past 15 years, many class I and class II transposons were identified in filamentous fungi. However, little is known about the influence of transposons during industrial strain development. The availability of the complete genome sequences of the industrially relevant fungi Aspergillus niger and Penicillium chrysogenum has enabled an analysis of transposons present in these two fungi. Here, a compilation of the transposon-like sequences identified is provided. We investigated a yet undescribed A. niger retrotransposon, ANiTa1, as well as two P. chrysogenum transposons (PeTra1 and PeTra2), which are the first P. chrysogenum transposons ever described, in more detail. Analysis of the genomic distribution of selected transposable elements in five strains of A. niger and seven strains of P. chrysogenum revealed the transposon distribution to be virtually identical. However, one element, Vader-previously published-from A. niger, showed strain-specific differences in transposon distribution, suggesting transposition activity during classical strain improvement programs. 相似文献

We are interested in isolating and identifying antigenic fungal proteins from species that grow on damp building materials. Fungal spores contain a modest number of proteins present in high concentrations and mycelia can contain large numbers of such proteins. Their distribution in mycelium when grown on laboratory media often does not reflect their occurrence in nature. Stachybotrys chartarum occurs on wet cellulose based building materials including paper-faced gypsum board and wood. To find potentially characteristic antigenic proteins from this fungus, we screened mycelium and spores from geographically representative strains using sera from a large number of patients who had been identified as having antibodies to fungi that grow in the built environment. Proteins were sought that were common across strains and for which antibodies were present in many sera. The use of human sera as the source of antibodies permitted the conclusion that any proteins of interest were produced in nature. The target proteins were shown to be present in spores/mycelium produced on natural substrata including straw and paper. This report concerns the isolation and partial characterization of a 34-kDa exoantigen. It is a glycoprotein that appears to comprise two subunits with molecular weights of 34 and 21 kDa which are not bound together through disulfide bonds. Both proteins have pIs in the acidic range. The sequences indicate that the protein is related to an extracellular DNase from Nectria haematococca. The partial sequences of the larger subunit are similar to that of a hypothetical protein from Gibberella zeae. 相似文献

EPg222 protease is a novel extracellular enzyme produced by Penicillium chrysogenum (Pg222) isolated from dry-cured hams that has the potential for use over a broad range of applications in industries that produce dry-cured meat products. The gene encoding EPg222 protease has been identified. Peptide sequences of EPg222 were obtained by de novo sequencing of tryptic peptides using mass spectrometry. The corresponding gene was amplified by PCR using degenerated primers based on a combination of conserved serine protease-encoding sequences and reverse translation of the peptide sequences. EPg222 is encoded as a gene of 1,361 bp interrupted by two introns. The deduced amino acid sequence indicated that the enzyme is synthesized as a preproenzyme with a putative signal sequence of 19 amino acids (aa), a prosequence of 96 aa and a mature protein of 283 aa. A cDNA encoding EPg222 has been cloned and expressed as a functionally active enzyme in Pichia pastoris. The recombinant enzyme exhibits similar activities to the native enzyme against a wide range of protein substrates including muscle myofibrillar protein. The mature sequence contains conserved aa residues characteristic of those forming the catalytic triad of serine proteases (Asp42, His76 and Ser228) but notably the food enzyme exhibits specific aa substitutions in the immunoglobulin-E recognition regions that have been identified in protein homologues that are allergenic. 相似文献

The virulence of two isolates of the hyphomycete fungi, Beauveria bassianaand B. brongniartii, and additional fungal species isolated from diseased Bactrocera oleae pupae and Sesamia nonagrioideslarvae were assessed against adults of the olive fruit fly B. oleae and the Mediterranean fruit fly Ceratitis capitata (Diptera: Tephritidae). Contact and oral bioassays revealed that moderate to high mortality rates for the olive fruit fly occurred when the adults were exposed to conidia of Mucor hiemalis, Penicillium aurantiogriseum, P. chrysogenum and B. bassianaisolates. A strain of M. hiemalis isolated from S. nonagrioides larvae was the most toxic resulting in 85.2% mortality to the olive fruit fly adults. B. brongniartiiand B. bassiana were the most pathogenic to the C. capitataadults causing 97.4 and 85.6% mortality. Metabolites collected from the M. hiemalis and P. chrysogenum isolates were toxic to adults of both species. 相似文献

In recent yearsAspergillus flavus and aflatoxin production have been noted on several occasions in grain preserved with formic acid. Samples of mouldy barley treated with formic acid and stored in an open bin were investigated for the presence of fungi. In the lower part of the bin there was a clear dominance ofFusarium sporotrichioides, and deoxynivalenol and neosolaniol were detected.A. flavus andA. fumigatus were also present.Paecilomyces variotii occurred, almost as a pure culture, in the upper part of the bin, but no patulin was found. Cultivation of four fungal isolates from these genera on laboratory substrates containing formic acid showedP. variotii to be the most tolerant to formic acid, withstanding 150 mM, but still without patulin production.F. sporotrichioides andA. fumigatus tolerated only 6 mM formic acid. The growth, biochemical composition and fatty acid profiles of six Antarctic microalgae cultured at different temperatures, ranging from 4, 6, 9, 14, 20 to 30 ∘C, were compared. The algae were isolated from seawater, freshwater, soil and snow samples collected during our recent expeditions to Casey, Antarctica, and are currently deposited in the University of Malaya Algae Culture Collection (UMACC). The algae chosen for the study were Chlamydomonas UMACC 229, Chlorella UMACC 234, Chlorella UMACC 237, Klebsormidium UMACC 227, Navicula UMACC 231 and Stichococcus UMACC 238. All the isolates could grow at temperatures up to 20 ∘C; three isolates, namely Navicula UMACC 231 and the two Chlorella isolates (UMACC 234 and UMACC 237) grew even at 30 ∘C. Both Chlorella UMACC 234 and Stichococcus UMACC 238 had broad optimal temperatures for growth, ranging from 6 to 20 ∘C (μ = 0.19 – 0.22 day–1) and 4 to 14 ∘C (μ = 0.13 – 0.16 day–1), respectively. In contrast, optimal growth temperatures for NaviculaUMACC 231 and Chlamydomonas UMACC 229 were 4 ∘C (μ = 0.34 day–1) and 6–9 ∘C (μ = 0.39 – 0.40 day–1), respectively. The protein content of the Antarctic algae was markedly affected by culture temperature. All except Navicula UMACC 231 and Stichococcus UMACC 238 contained higher amount of proteins when grown at low temperatures (6–9 ∘C). The percentage of PUFA, especially 20:5 in Navicula UMACC 231 decreased with increasing culture temperature. However, the percentages of unsaturated fatty acids did not show consistent trend with culture temperature for the other algae studied. 相似文献

One-year-old tree seedlings were incubated in a greenhouse from April to July, under natural daylight conditions, with their root systems at constant temperatures of 5, 10, 15, 20, 25, 30 and 35 °C and with the above ground parts kept at a constant air temperature of 18–20 °C. The course of height growth, total mass increment, root, shoot and leaf weight as well as leaf areas were measured. The results indicate that clear differences exist in the optimal root zone temperatures for various growth parameters in different tree species. Pinus sylvestris had a maximal height increment at about 5–10 °C and maximal total mass increment at 15 °C root temperature. In contrast, the optimum for Quercus robur was at 25 °C. Tilia cordata and Fagus sylvatica had their optima for most growth parameters at 20 °C. The root temperature apparently indirectly influenced photosynthesis (dry weight accumulation) and respiration loss. From the observed symptoms and indications in the literature it seems probable that a change in hormone levels is involved as the main factor in the described effects. Variation of root temperature had only an insignificant effect on bud burst and the time at which the shoots sprouted. Apparently species of northern origin seem to have lower root temperature optima than those of more southern origin. This is to be verified by investigation of other tree species. 相似文献

At extremely low values of moisture pressure (?96.4 MPa; aw 0.50), the spores of xerotolerant streptomycetes (Streptomyces odorifer and S. rubiginosohelvolus) germinated, their germ lengths increased, and lateral branching of the mycelium was observed after 5 days of incubation in a thin layer of agarized nutrient medium. At ?22.6 MPa (aw 0.86), the mycelium begins to branch after a 2-day incubation; over a 5-day incubation at ?2.8 MPa (aw 0.98), it goes through a reproduction cycle, which culminates in spore formation. The mathematical model approach enabled us to elucidate the behavioral patterns of Streptomyces spores in a thin layer of agarized nutrient medium at low humidity levels. The dynamics of spore germination is governed by the exponential law, which allows calculation of the average duration of the period a before spore germination, as well as the time needed for 50% of viable spores to germinate. 相似文献

The fungus Fusarium bulbigenum var. blasticola distinguished by secondary tumor-like growth (TLG) in aging was taken as a novel test system for antitumor drugs. EPR spectroscopy was used to assess the effect of various drugs introduced into the growth medium on the type and content of free radicals in the primary mycelium and TLGs. The EPR spectra in all cases featured a single slightly asymmetrical line of ΔH ~ 0.4–0.6 mT and g = 2.0036 ± 0.0006, whereby the paramagnetic centers could be assigned to melanin radicals. The content of free radicals was always higher in TLGs than in the primary mycelium, which could naturally be due to more intense metabolism. A number of antitumor drugs (fluorouracil, hydroxyurea, methotrexate, etoposide) completely prevented TLG, with varying effect on free radicals in mycelia. Another group (cyclophosphane, dacarbazine, adriablastine, vinblastine), on the contrary, promoted TLG and increased the free radical content in all fungal cells. The third group (mercaptopurine, thioguanine, pharmorubicin) were weakly suppressive for TLG despite the elevated free radical content therein. Vitamins B2, B12, C, and PP were stimulatory for TLG, whereas in combination with antitumor drugs they could enhance or attenuate their effects. 相似文献

Summary Methods were developed to study the effects of absorbent materials from diapers on microbial survival, growth and toxic shock syndrome toxin-1 (TSST-1) production under specified in vitro conditions. Growth of representative skin and fecal flora organisms was equivalent in cultures in which materials from cotton cloth diapers, disposable diapers or disposable diapers containing absorbent gelling material were added as the sole carbon source. In urine used as an enrichment medium, growth of the test organisms in media containing material from the three diaper types was equivalent and no contribution to growth from the diaper material was detected. TSST-1 was not produced byStaphylococcus aureus under conditions in which urine was added to the diaper materials. Pathogenic strains of organisms purposefully introduced onto diapers failed to survive and the few microbial cells normally found in diaper material did not multiply when stored under conditions favorable to microbial growth. The data indicate that all three diaper types tested were the same with respect to growth and survival of representative skin and fecal organisms. 相似文献

Biomass and toxic metabolite production by Altemaria hemicota, A. solani and A. tenuis in modified Czapek Dox broth (mCDB) as affected by incubation time (up to 4 wk) temperature (6, 15 and 25 °C), pH (3.0, 5.0 and 8.0) and sugars (glucose, fructose and sucrose) was studied. Biomass production was greatest at 25 ° C and pH 8.0 but toxin production was enhanced at 15 °C. In general, biomass production was promoted by up to 9% glucose and sucrose and 6% fructose; however, toxin production was simultaneously reduced by elevated sugars in mCDB over a 4-wk incubation period. 相似文献

Chromosome-substituted haploid segregants of anA. nidulans × A. tetrazonus somatic hybrid were used to allocate several random amplified polymorphic DNA and isoenzyme markers to parental chromosomes. Twenty-six amplified DNA fragments, and nine isoenzyme activities, including lactate dehydrogenase, superoxide dismutase, and arylesterase isoenzymes were assigned to chromosomes. Chromosome-specific markers were found for eachA. nidulans andA. tetrazonus chromosome. These markers could be used to saturate the genetic map ofA. nidulans. The formation of two secondary metabolites was also assigned to chromosomes III and VIII. Because little is known about filamentous fungal tensile strength, it is assumed to be constant in many models. A method involving off-line fragmentation and image analysis is used now to measure relative tensile strength of Aspergillus oryzae hyphae. During the course of shake-flask and fed-batch cultures, hyphal tensile strength increases during growth then decreases. Hyphal tensile strength can change up to 3-fold over time. 相似文献
