The preference of paddy rice for NH4+ rather than NO3- is associated with its tolerance to low pH since a rhizosphere acidification occurs during NH4+ absorption. However, the adaptation of rice root to low pH has not been fully elucidated. This study investigated the acclimation of plasma membrane H+-ATPase of rice root to low pH. Rice seedlings were grown either with NH4+ or NO3-. For both nitrogen forms, the pH value of nutrient solutions was gradually adjusted to pH 6.5 or 3.0. After 4 d cultivation, hydrolytic H+-ATPase activity, V max, K m, H+-pumping activity, H+ permeability and pH gradient across the plasma membrane were significantly higher in rice roots grown at pH 3.0 than at 6.5, irrespective of the nitrogen forms supplied. The higher activity of plasma membrane H+-ATPase of adapted rice roots was attributed to the increase in expression of OSA1, OSA3, OSA7, OSA8 and OSA9 genes, which resulted in an increase of H+-ATPase protein concentration. In conclusion, a high regulation of various plasma membrane H+-ATPase genes is responsible for the adaptation of rice roots to low pH. This mechanism may be partly responsible for the preference of rice plants to NH4+ nutrition.  相似文献

White lupin ( Lupinus albus L.) is able to grow on soils with sparingly available phosphate (P) by producing specialized structures called cluster roots. To mobilize sparingly soluble P forms in soils, cluster roots release substantial amounts of carboxylates and concomitantly acidify the rhizosphere. The relationship between acidification and carboxylate exudation is still largely unknown. In the present work, we studied the linkage between organic acids (malate and citrate) and proton exudations in cluster roots of P-deficient white lupin. After the illumination started, citrate exudation increased transiently and reached a maximum after 5 h. This effect was accompanied by a strong acidification of the external medium and alkalinization of the cytosol, as evidenced by in vivo nuclear magnetic resonance (NMR) analysis. Fusicoccin, an activator of the plasma membrane (PM) H+-ATPase, stimulated citrate exudation, whereas vanadate, an inhibitor of the H+-ATPase, reduced citrate exudation. The burst of citrate exudation was associated with an increase in expression of the LHA1 PM H+-ATPase gene, an increased amount of H+-ATPase protein, a shift in pH optimum of the enzyme and post-translational modification of an H+-ATPase protein involving binding of activating 14-3-3 protein. Taken together, our results indicate a close link in cluster roots of P-deficient white lupin between the burst of citrate exudation and PM H+-ATPase-catalysed proton efflux.  相似文献

Chilling leads to cytoplasmic acidification in chill-sensitive plants. A possible explanation for this observation is that a ΔpH-stat between the cytosol and vacuole (ΔpHvac-stat) is perturbed by chilling. To understand the nature of this ΔpHvac-stat, the effect of temperature, between 20 and 0 °C, on pyrophosphate (PPi)- or ATP-dependent acidification of vacuolar vesicles, isolated from mung bean hypocotyls, was determined. Over the temperature range investigated, the H+-influx mediated by PPase was balanced with the H+-efflux, which was PPi-dependently suppressed, and consequently a constant pH in vesicles (pHin) of ca. 5 was maintained against temperature changes. However, the ΔpHin driven by ATP decreased as the temperature dropped. Thus, the PPi-dependent H+-accumulation may function as an essential factor to form a ΔpHvac-stat against temperature changes. Next, to study the chilling sensitivity of PPi-dependent H+-accumulation, vacuolar vesicles were isolated from control seedlings or from seedlings chilled at 0 °C for 1 d. Chilling treatment resulted in a decrease in the H+-accumulation rate and in the steady-state ΔpHin formed by PPi, the causes of which were enhanced by PPi-dependent H+-efflux and reduced by H+-influx driven by PPase. Together, the results suggest that the decrease of PPi-dependent H+-accumulation associated with the ΔpHvac-stat could result in cytoplasmic acidification.  相似文献

Abstract. White lupin ( Lupinus albus L.) was grown for 13 weeks in a phosphorus (P) deficient calcareous soil (20% CaCO3, pH(H2O)7.5) which had been sterilized prior to planting and fertilized with nitrate as source of nitrogen. In response to P deficiency, proteoid roots developed which accounted for about 50% of the root dry weight. In the rhizosphere soil of the proteoid root zones, the pH dropped to 4.8 and abundant white precipitates became visible. X-ray spectroscopy and chemical analysis showed that these precipitates consisted of calcium citrate. The amount of citrate released as root exudate by 13-week-old plants was about 1 g plant−1, representing about 23% of the total plant dry weight at harvest. In the rhizosphere soil of the proteoid root zones the concentrations of available P decreased and of available Fe, Mn and Zn increased. The strong acidification of the rhizosphere and the cation/anion uptake ratio of the plants strongly suggests that proteoid roots of white lupin excrete citric acid, rather than citrate, into the rhizosphere leading to intensive chemical extraction of a limited soil volume. In a calcareous soil, citric acid excretion leads to dissolution of CaCO3 and precipitation of calcium citrate in the zone of proteoid roots.  相似文献

In embryos of the sea urchin, Hemicentrotus pulcherrimus , as well as in cultured cells derived from isolated micromeres, spicule formation was inhibited by allylisothiocyanate, an inhibitor of H+, K+-ATPase, at above 0.5 μM and was almost completely blocked at above 10 μM. Amiloride, an inhibitor of Na+, H+ antiporter, at above 100 μM exerted only slight inhibitory effect, if any, on spicule formation. Intravesicular acidification, determined using [ dimethylamine -14C]-aminopyrine as a pH probe, was observed in the presence of ATP and 200 mM KCl in microsome fraction obtained from embryos at the post gastrula stage, at which embryos underwent spicule calcification. Intravesicular acidification and K+-dependent ATPase activity were almost completely inhibited by allylisothiocyanate at 10 μM. Allylisothiocyanate-sensitive ATPase activity was found mainly in the mesenchyme cells with spicules isolated from prisms. H+, K+-ATPase, an H+ pump, probably mediates H+ release to accelerate CaCO3 deposition from Ca2+, CO2 and H2O in the primary mesenchyme cells. Intravesicular acidification was stimulated by valinomycin at the late gastrula and the prism stages but not at the pluteus stage. K+ permeability probably increases after the prism stage to activate H+ release.  相似文献

The figleaf gourd ( Cucurbita ficifolia Bouché) root system has the ability to take up water and nutrients at low soil temperatures, and in the present paper, we attempt to reveal some of the molecular mechanisms behind this low-temperature tolerance. Exposure of figleaf gourd root system to low temperature induced accumulation of H2O2 along the plasma membrane but not in the cytoplasm. H+-ATPase (EC 3.6.1.35) activity of isolated root plasma membranes and root hydraulic conductivity ( Lpr ) were largely insensitive to externally applied H2O2. However, using bromocresol purple, it was shown that the acidification of the medium surrounding the root was strongly inhibited with low temperature- and H2O2-treated roots. Addition of catalase (EC 1.11.1.6) to the root medium during low-temperature exposure led to a recovery of H+-efflux along the root surface and increased Lpr , demonstrating the importance of an H2O2 detoxification system in the root cells. Additional evidence for an increased Lpr was obtained by the Fenton reaction wherein a warming of the solution increased the activity of the detoxification system. All available evidence suggests that the ability of figleaf gourd root system to maintain a low level of H2O2 in the cytoplasm and to detoxify reactive oxygen species is related to the maintenance of water transport activity at low temperatures.  相似文献

Shading of maize plants ( Zea mays L. cv. Blizzard) reduced net H+ extrusion by roots and increased K+ release, whereas there was no significant effect on anion efflux in deionized water. With lower light intensity the concentrations of carbohydrates in the roots decreased, but ATP levels and energy charge remained unchanged. Also, shading raised the tissue pH of roots and made the cytoplasmic pH of root cells drop. There was a significant influence of light intensity on H+ uptake by roots from an acidified test solution and CCCP (carbonylcyanide-3-chlorophenylhydrazone)-in-duced H+ uptake was modified by shading.
It is concluded that low light intensity does not limit active H+ release by plasmalemma ATPase activity in the root cells, but that a reduced carbohydrate supply brings about a change in biochemical reactions which alter the membrane permeability for protons. An increased passive reflux of H+ into the cells rather than a reduced H+ ATPase activity explains the decrease of net H+ release by roots of intact maize plants under low light intensity.  相似文献

Transport across the plasma membrane is driven by an electrochemical gradient of H+ ions generated by the plasma membrane proton pump (H+-ATPase). Random mutants of Arabidopsis H+-ATPase AHA1 were isolated by phenotypic selection of growth of transformed yeast cells in the absence of endogenous yeast H+-ATPase (PMA1). A Trp-874-Leu substitution as well as a Trp-874 to Lys-935 deletion in the hydrophilic C-terminal domain of AHA1 conferred growth of yeast cells devoid of PMA1. A Trp-874-Phe substitution in AHA1 was produced by site-directed mutagenesis. The modified enzymes hydrolyzed ATP at 200–500% of wild-type level, had a sixfold increase in affinity for ATP (from 1.2 to 0.2 mM; pH 7.0), and had the acidic pH optimum shifted towards neutral pH. AHA1 did not contribute significantly to H+ extrusion by transformed yeast cells. The different species of aha1, however, displayed marked differences in initial rates of net H+ extrusion and in their ability to sustain an electrochemical H+ gradient. These results provide evidence that Trp-874 plays an important role in auto-inhibition of the plant H+-ATPase and may be involved in controlling the degree of coupling between ATP hydrolysis and H+ pumping. Finally, these results demonstrate the usefulness of yeast as a generalized screening tool for isolating regulatory mutants of plants transporters.  相似文献

NaCl-induced changes in the accumulation of message for the 70 kDa subunit of the tonoplast H+-ATPase and plasma membrane H+-ATPase were studied in hydroponically grown plants of Lycopersicon esculentum Mill. cv. Large Cherry Red. There was increased accumulation of message for the 70 kDa (catalytic) subunit of the tonoplast H+-ATPase in expanded leaves of tomato plants 24 h after final NaCl concentrations were attained. This was a tissue-specific response; levels of this message were not elevated in roots or in young, unexpanded leaves. The NaCl-induced accumulation of this message was transient in the expanded leaves and returned to control levels within 7 days. The temporal and spatial patterns of NaCl-induced accumulation of message for the plasma membrane H+-ATPase differed from the patterns associated with the 70 kDa subunit of the tonoplast H+-ATPase. NaCl-induced accumulation of the plasma membrane H+-ATPase message occurred in both roots and expanded leaves. Initially accumulation of the plasma membrane H+-ATPase message was greater in root tissue than in expanded leaves, but increased to higher levels in expanded leaves after 7 days. These results suggest that increased expression of the tonoplast H+-ATPase is an early response to salinity stress and may be associated with survival mechanisms, rather than with long-term adaptive processes.  相似文献

The generally rhizotoxic ion Al3+ often enhances root growth at low concentrations. The hypothesis that Al3+ enhances growth by relieving H+ toxicity was tested with wheat seedlings ( Triticum aestivum L.). Growth enhancement by Al3+ only occurred under acidic conditions that reduced root elongation. Al3+ increased cell membrane electrical polarity and stimulated H+ extrusion. Previous investigations have shown that Al3+ decreases solute leakage at low pH and that the alleviation of H+ toxicity by cations appears to be a general phenomenon with effectiveness dependent upon charge (C3+>C2+>Cl+). Alleviation of one cation toxicity by another toxic cation appears to be reciprocal so that Al3+ toxicity is relieved by H+. It has been argued previously that this latter phenomenon accounts for the apparent toxicity of ALOH2+ and Al(OH)+2. Reduction of cell-surface electrical potential by the ameliorative cation may reduce the cell-surface activity of the toxic cation.  相似文献

Abstract Washed cells of Rhodopseudomonas sphaeroides forma sp. denitrificans , grown under photodenitrifying conditions, exhibited K+ uptake dependent on the transmembrane proton gradient (Δ pH). These cells also acidified the suspension medium in response to K+ pulses both aerobically and anaerobically in light and in the dark. The results indicate that the photodenitrifier has a reversible K+/H+ exchange activity which reflects its role in regulating the intracellular K+ concentration, as well as intracellular pH. The acidification of the external medium resulting from K+ pulses was inhibited by carbonyl cyanide- m -chlorophenylhydrazone (CCCP) indicating that the antiporter is energy-dependent. Addition of KCl to washed cells depolarized the membrane potential (Δψ) with a concomitant increase in ΔpH, indicating that the K+/H+ antiporter was electrogenic.  相似文献

The involvement of Ca2+ and H+ flux oscillations in root nutation was studied for decapped roots of corn ( Zea mays L. cv. Aussie Gold) placed horizontally. Net ion fluxes were measured around the elongation and meristematic regions using a microelectrode ion flux measuring system. High correlation between H+ flux oscillations and root nutations was found in the elongation region. Two oscillatory components of H+ flux, with periods of about 90 min and 7 min, correlated with root circumnutations and micronutations, respectively. The periods of H+ flux oscillations and rhythmical root movements in this region could be modified similarly by external factors including pH. In the meristematic region no association between ion flux behaviour and nutation was apparent. Ion flux oscillations and nutations both decreased in amplitude as the growth rate at the measured location decreased. Possible involvement of ion flux oscillations in root circumnutation is discussed. It is concluded that a model involving an internal oscillator must be developed to explain the H+ flux involvement in root nutations.  相似文献

The phytohormone abscisic acid (ABA) inhibits blue light‐induced apoplastic acidification of guard cells. The signal transduction pathway of ABA, mediating this response, was studied using ABA‐insensitive ( abi ) mutants of Arabidopsis thaliana . Apoplastic acidification was monitored with a flat tipped pH‐electrode placed on epidermal strips, in which only guard cells were viable. Blue light‐induced apoplastic acidification was reduced by vanadate and diethylstilbestrol (DES), indicating involvement of plasma membrane‐bound H+‐ATPases. In wild type epidermal strips, ABA reduced blue light‐induced acidification to 63%. The inhibition did not result from an increased cytoplasmic free Ca2+ concentration in guard cells, since factors that increase the Ca2+ concentration stimulated apoplastic acidification. Apoplastic acidification was not inhibited by ABA in abi1 and abi2 mutants. In abi1 epidermal strips ABA had no effect on the acidification rate, while it stimulated apoplastic acidification in abi2 . The ABA response in both mutants could be partially restored with protein kinase and phosphatase inhibitors. The abi1 guard cells became ABA responsive in the presence of okadaic acid, a protein phosphatase inhibitor. In abi2 guard cells the wild type ABA response was partially restored by K‐252a, a protein kinase inhibitor. Apoplastic inhibition is thus mediated through the protein phosphatases encoded by ABI1 and ABI2 . The results with protein kinase and protein phosphatase inhibitors indicate that ABI1 and ABI2 are involved in separate signal transduction pathways.  相似文献

Hypoxic pretreatment is known to induce anoxia tolerance in plant species sensitive to oxygen deprivation. However, we still do not have detailed information on changes in cytoplasmic and vacuolar pH (pHcyt and pHvac) in plants under low-oxygen availability (hypoxia) and under anoxia. To investigate this, we have studied the influence of hypoxia and anoxia on pHcyt and pHvac, glucose-6-phosphate (Glc-6-P) and nucleotide triphosphate (NTP) contents in rice ( Oryza sativa L.) root tips in comparison with those of wheat ( Triticum aestivum L.) with in vivo 31P-nuclear magnetic resonance. Both cereals responded to hypoxia similarly, by rapid cytoplasmic acidification (from pH 7.6–7.7 to 7.1), which was followed by slow partial recovery (0.3 units after 6 h). Anoxia led to a dramatic pHcyt drop in tissues of both species (from pH 7.6–7.7 to less than 7.0) and partial recovery took place in rice only. In wheat, the acidification continued to pH 6.8 after 6 h of exposure. In both plants, NTP content followed the dynamics of pHcyt. There was a strong correlation between NTP content and cytoplasmic H+ activity ([H+]cyt= 10−pHcyt) for both hypoxic and anoxic conditions. Glc-6-P content increased in rice under anoxia and hypoxia. In wheat, Glc-6-P was not detectable under anoxia but increased under hypoxia. In this study, rice root tips were shown to behave as anoxia tolerant tissues. Our results suggest that the initial cytoplasmic acidification and subsequent pHcyt are differently regulated in anoxia tolerant and intolerant plants and depend on the external oxygen concentration.  相似文献

The regulation of the H+-ATPase of plasma membrane is a crucial point in the integration of transport processes at this membrane. In this work the regulation of H+-ATPase activity induced by changes in turgor pressure was investigated and compared with the stimulating effect of fusicoccin (FC). The exposure of cultured cells of Arabidopsis thaliana L. (ecotype Landsberg 310–14-2) to media containing mannitol (0. 15 or 0. 3 M ) or polyethylene glycol 6000 (PEG) (15. 6% or 22% w/v) resulted in a decrease in the turgor pressure of the cells and in a strong stimulation of H+ extrusion in the incubation medium. The osmotica-induced H+ extrusion was (1) inhibited by the inhibitor of plasma membrane H+-ATPase, erythrosin B (EB), (2) dependent on the external K+ concentration, (3) associated with a net K+ influx, and (4) lead to an increase of cellular malate content. These results show that the reduction of external osmotic potential stimulates the activity of plasma membrane H+-ATPase
The effect of mannitol was only partially inhibited by treatments with cycloheximide (CH) and cordycepin, which block protein and mRNA synthesis, respectively. All the effects of osmotica were qualitatively and quantitatively similar to those induced by 5 μ M FC. However, when FC and mannitol (or PEG) were fed together, their effects on H+ extrusion appeared synergistic, irrespective of whether FC was present at suboptimal or optimal concentrations. This behaviour suggests that the modes of action of FC and of the osmotica on H+-ATPase activity differ at least in some step(s)  相似文献

Nutrient acquisition in the mature root zone is under systemic control by the shoot and the root tip. In maize, exposure of the shoot to light induces short-term (within 1–2 min) effects on net K+ and H+ transport at the root surface. H+ efflux decreased (from −18 to −12 nmol m−2 s−1) and K+ uptake (∼2 nmol m−2 s−1) reverted to efflux (∼−3 nmol m−2 s−1). Xylem probing revealed that the trans-root (electrical) potential drop between xylem vessels and an external electrode responded within seconds to a stepwise increase in light intensity; xylem pressure started to decrease after a ∼3 min delay, favouring electrical as opposed to hydraulic signalling. Cutting of maize and barley roots at the base reduced H+ efflux and stopped K+ influx in low-salt medium; xylem pressure rapidly increased to atmospheric levels. With 100 m m NaCl added to the bath, the pressure jump upon cutting was more dramatic, but fluxes remained unaffected, providing further evidence against hydraulic regulation of ion uptake. Following excision of the apical part of barley roots, influx changed to large efflux (−50 nmol m−2 s−1). Kinetin (2–4  µ m ), a synthetic cytokinin, reversed this effect. Regulation of ion transport by root-tip-synthesized cytokinins is discussed.  相似文献
