The effects of a series of sodium alkyl sulfates (C(4) to C(16)) on flagellation, motility, swarming, and growth of Proteus were examined. The concentrations of the various sodium alkyl sulfates completely inhibiting the swarming phenomenon (on solid medium) and motility (in liquid medium) were in the same order of magnitude. The inhibiting effect of the detergents examined increased from sodium hexyl sulfate (inhibitory concentration, 20 to 30 mmoles per liter) to sodium tetradecyl sulfate (inhibitory concentration, 0.1 to 0.5 mmoles per liter). Flagella were produced neither in liquid nor on solid medium at these concentrations as could be observed by electron microscopy. At concentrations where motility was not impaired, intact flagellation could be observed. At a concentration of 0.1 mmole per liter, sodium tetradecyl sulfate completely inhibited the motility of Proteus in the liquid medium employed without impairing growth.  相似文献

The ruminal selenomonad strain H18 grew rapidly (mu = 0.50 h-1) in a defined medium containing glucose, ammonia, purified amino acids, and sodium (95 mM); little if any ammonia was utilized as a nitrogen source. When the sodium salts were replaced by potassium salts (0.13 mM sodium), there was a small reduction in growth rate (mu = 0.34 h-1), and under these conditions greater than 95% of the cell nitrogen was derived from ammonia. No growth was observed when the medium lacked sodium (less than 0.35 mM) and amino acids were the only nitrogen source. At least six amino acid transport systems (aspartate, glutamine, lysine, phenylalanine, serine, and valine) were sodium dependent, and these systems could be driven by an electrical potential (delta psi) or a chemical gradient of sodium. H18 utilized lactate as an energy source for growth, but only when sodium and aspartate were added to the medium. Malate or fumarate was able to replace aspartate, and when these acids were added, sodium was no longer required. Glucose-grown cells accumulated large amounts of polysaccharide (64% of dry weight), and when the exogenous glucose was depleted, this material was converted to acetate and propionate as long as sodium was present. When the cells were incubated in buffers lacking sodium, succinate accumulated and exogenous succinate could not be decarboxylated. Because sodium had little effect on the transmembrane pH gradient at pH 6.7 to 4.5, it did not appear that sodium was required for intracellular pH regulation.  相似文献

The study of the microalgal growth kinetics is an indispensable tool in all fields of phycology. Knowing the optimal nutrient concentration is an important issue that will help to develop efficient growth systems for these microorganisms. Although nitrogen and phosphorus are well studied for this purpose, sulfur seems to be less investigated. Sulfate is a primary sulfur source used by microalgae; moreover, the concentration of this compound is increasing in freshwater systems due to pollution. The aim of this study was to investigate the effects of different sodium sulfate concentrations in the culture medium on growth and growth kinetics of the freshwater microalga Chlamydomonas moewusii. Production of biomass, chl content, kinetic equations, and a mathematical model that describe the microalgal growth in relation with the concentration of sodium sulfate were obtained. The lowest concentration of sodium sulfate allowing optimal growth was 0.1 mM. Concentrations higher than 3 mM generated a toxic effect. This work demonstrates that this toxic effect was not directly due to the excess of sulfate ion but by the elevation of the ionic strength. An inhibition model was successfully used to simulate the relationship between specific growth rate and sodium sulfate in this microalga.  相似文献

Calcium chloride (0.3 or 10 mM) was applied to the growth medium before, together with, or after sodium chloride application, and the effect of the timing of Ca application on the alleviation of salt stress was investigated. Seedlings of maize, tall fescue, and reed canarygrass were grown in medium with 0 and 200 mM NaCl for 5 d. Regardless of the plant species the maximum alleviation of NaCl stress was achieved when CaCl2 and NaCl coexisted in the growth medium. The effects of Ca application were connected with the decrease in the Na content in the plant roots and shoots and increased ATPase activity in the roots.  相似文献

The effect of fluctuating temperatures on microbial growth is important in the passage of foods from production to consumption. Suspensions of Salmonella typhimurium have been subjected to sinusoidally time-varying temperatures of periods from 60 to 240 min between 4°C and 22°C, that is within and below the growth temperature range. The suspensions were prepared with two concentrations of sodium chloride and adjusted to two different values of pH. The change in the numbers of viable bacteria was measured with time and the experimental growth curves and average generation times compared with predictions based on isothermal growth data. Generally, the experimental average generation times exceeded the predictions by not more than 10% In enumerating viable bacteria in the suspensions containing 3.5% (w/v) sodium chloride it was necessary to use sodium chloride in the diluent and recovery medium in order to recover the bacteria quantitatively.  相似文献

The effect of different levels of salt, sodium nitrite, polyphosphate and various sugars on growth, pigment production, protease activity and culture pH caused by Monascus purpureus was studied in broth medium and ground meat. The addition of sodium chloride (> 50.0 g l(-1)) and polyphosphate (> 3.0g l(-1)) to broth medium decreased mycelial growth, pigment production and protease activity of M. purpureus, whereas low concentrations of sodium nitrite (< 0.2 g l(-1)) promoted mycelial growth and pigment production. When the basal medium and ground meat contained salt, 150.0 g l(-1), the mould growth was stopped. The medium with fructose as carbon source proved to be the most suitable for mycelium growth and pigment production, with maltose and glucose being the second most productive. When sucrose and lactose were used as carbon sources, mycelium growth and pigment production were inhibited but the protease activity increased significantly. The mould showed more tolerance to salt and polyphosphate in ground meat than in broth medium and used sucrose as a carbon source as well as glucose for growth and pigment production in the meat mixture.  相似文献

Summary The importance of serum-free medium components on the growth of Chinese hamster ovary (CHO) cells and production of recombinant human interferon(IFN)-gamma was investigated. The complexity of the medium led to the adoption of a statistical optimization approach based on a Plackett-Burman design. From this analysis a set of nutritional components was identified as important for cell growth and recombinant protein production. Glycine was identified as an important determinant of specific growth rate, whereas for cell production bovine serum albumin (BSA), phenylalanine and tyrosine were also identified as important. BSA, sodium pyruvate, glutamate, methionine, proline, histidine, hydroxyproline, tyrosine and phenylalanine were shown to be important for IFN-gamma production. Other medium components, such as insulin, arginine, aspartate and serine produced an inhibitory effect on both cell growth and IFN-gamma production. The effect of the stimulatory nutrients as a whole group was tested by increasing their concentration in the medium. A significant improvement in specific cell growth rate, cell production and IFN-gamma production (up to 45%) was achieved on both shake-flask and fermentor cultures. An increase in the medium concentration of the negative variables had only a small inhibitory effect (approximately 10%) on the same parameters. Analysis of the effects of the group of stimulatory amino acids and BSA on CHO cell growth showed that the effect of the former was independent of BSA.Correspondence to: A. T. Bull  相似文献

The toxicity of potassium arsenate, as measured by retardation or inhibition of growth on solid nutrient media, showed wide variation among different fungi but was consistently reduced by the addition of large amounts of potassium phosphate, with both arsenic-sensitive and arsenic-tolerant fungi. Poria monticola was completely inhibited by 0.0025 M arsenate but was progressively less inhibited as the phosphate content of the medium increased and grew slowly at 0.04 M arsenate when 0.16 M KH(2)PO(4) was added. Cladosporium herbarum showed 36% reduction in growth at 0.08 M arsenate in a low-phosphate medium, but when 0.01 M KH(2)PO(4) was added, arsenate concentrations up to 0.64 M (at which the medium contains 4.8% As) caused no reduction in growth rate. Addition of phosphate also reduced the toxicity of potassium arsenite but not that of dimethyl sodium arsonate (sodium cacodylate). The counteracting effect of phosphate on arsenate toxicity was found to occur with every one of a wide variety of microorganisms tested. The author interprets the results as supporting the thesis that the fungitoxicity of arsenate is due to its competitive interference with phosphorus in oxidative phosphorylation and not to a reaction with the -SH groups of essential proteins. The latter mechanism is, however, probably operative with dimethyl sodium arsonate. The practical implications of the counter-inhibition phenomenon in laboratory investigations and standard tests of arsenical fungicides, in biochemical research, and in the commercial use of arsenical biocides are set out.  相似文献

目的:考察培养基中葡萄糖、谷氨酰胺、血清、碳酸氢钠含量对CHO细胞生长繁殖的影响。方法:在CHO细胞培养基中添加不同成分的葡萄糖、谷氨酰胺、血清、碳酸氢钠,通过单因素实验结果结合Box-Behnken效应面法,根据二次回归模型的分析结果,以细胞表达蛋白体外活性为指标进行实验,考察培养基中葡萄糖、谷氨酰胺、血清、碳酸氢钠含量对细胞生长繁殖的影响。结果:根据回归方程分析结果,作出相应的曲面图和等高线图,优选出培养基中各组分的最佳配比为:葡萄糖2.54 g/L、谷氨酰胺0.59 g/L、血清8.3%,碳酸氢钠2.96 g/L。结论:Box-Behnken实验设计法用于细胞培养过程中考察培养基中各组分的优选是可行的,数学模型的预测值与实验观察值相符。通过对CHO细胞培养基成分的优化,使CHO细胞蛋白表达量高,有利于提高产品质量和降低生产成本。  相似文献

The effects of changes in the inorganic nitrogen source of themedium on the growth and development of Marsilea in asepticculture are described. A change in nitrate concentration to0·2 or 5 x that of the normal medium had initially noeffect on growth or heteroblastic development, but in the 0·2nitrogen medium there was finally a reversion to juvenile stagesfollowed by early cessation of growth. Growth and heteroblastic development were stimulated by thesubstitution of the nitrate of the medium by ammonium saltsor urea. Nitrite was little inferior to nitrate as a nitrogensource, but hydroxylamine was toxic even at low concentrations.Addition of sodium chloride led to improved growth and developmentin nitrate media, an effect referred to beneficial effects ofthe chloride ion. Symptoms of toxicity and eventual death of the cultures whichoccurred after prolonged growth on ammonium media were due toan acid drift of the media rather than to a specific effectof the ammonium ion.  相似文献

The present paper deals with the effect of osmotic value of the medium on the growth of the young sunflower embryos grown in vitro. Based upon the results of a series of experiments carried out during 1963 the following conclusions are reached: 1. The sunflower embryos grown in vitro require different concentrations of sucrose, varying with the age at which the embryos are excised. Generally speaking, the optimum concentration of sucrose for the embryos 1,000–1,100μ in length is about 17.5%, for 2,000–2,500 μ, about 16% and for 5,000–5,500 μ, about 12.5%. 2. The sucrose may be partially replaced by sodium chloride (0.2%–0.4%) or mannitol (1.125%–5.5%) as an osmotically active substance, but 0.8% sodium chloride is apparently toxic to the young embryos. 3. Embryos cultured in hypotonic medium usually germinate precociously, those in approximately isotonic medium continue their embryonic growth, while those in hypertonic medium become more or less checked in their growth.  相似文献

为了避免玫瑰红钠培养基平皿法计数验证时细菌在培养基上生长而干扰真菌计数的准确性这一问题,采用在该培养基中选加了0.08g/L、0.10g/L和0.12g/L三种不同浓度的氯霉素的方法。结果证实,在玫瑰红钠培养基中加入浓度为0.10g/L的氯霉素可完全抑制供试大肠杆菌CMCC(B)44102株的生长,但又不影响供试真菌如白色念珠菌CMCC(F)98001、黑曲霉CMCC(F)98003的培养生长。因此,在微生物限度检查时的玫瑰红钠培养基中加入氯霉素最适宜的有效浓度为0.10g/L。  相似文献

Persistent bacteria were separated fromS. parasitica by means of the oligodynamic effect of a silver ring in a modification ofRaper's technique. Inoculation of fungal cultures was by means of mycelial macerate. Growth was measured by mycelial dry weight. A chemically defined medium (standard medium) was developed which consisted of a mineral base (chlorides of magnesium, manganese, zinc, calcium, and iron) chelated with EDTA, supplemented with glucose, sodium glutamate, and methionine, and buffered at pH 7.0 with 0.01 M. KH2PO4. Shaking culture methods supported increased growth rates and higher dry weight yields compared to stationary methods. Excellent growth occurred between 15 to 30°C. in the standard medium and between pH 4.0 and 8.0 in the standard medium plus 0.01 M. sodium succinate and 0.01 M. TRIS used as additional buffers. Significant phosphate toxicity was demonstrated at concentrations exceeding 0.05 M. Sodium succinate and TRIS, used as buffers at 0.01 M. each, were compatible withS. parasitica, whereas boric acid, sodium barbital, and sodium citrate inhibited growth under similar conditions. Substitution of other carbon sources for glucose in the standard medium (on an equal carbon basis where possible) indicated that cellobiose, dextrin, fructose, glycerine, glycogen, sodium lactate, and soluble starch supported significantly heavier growth than did the standard medium minus glucose; glycogen had a greater yield than did the standard medium minus glucose; glycogen had a greater yield than the standard medium. Arabinose, dulcitol, galactose, inulin, lactose, mannitol, mannose, raffinose, rhamnose, sorbitol, sucrose, and xylose neither stimulated nor inhibited growth; however, growth inhibition was produced by α-ketoglutaric acid, sodium citrate, and sodium succinate. When fatty acids and lipids were substituted for glucose (on an equal carbon basis where possible), only butter, lard, oleo, and palmitic acid supported heavier growth ofS. parasitica than the standard medium minus glucose. Stearicacid neither stimulated nor inhibited growth; acetic acid, butyric acid, formic acid, octanoic acid, and propionic acid significantly inhibited the growth of the fungus. Various nitrogen sources were substituted for sodium glutamate in the standard medium (on an equal nitrogen basis where possible). Casein hydrolysate and gelatin produced yields higher than that developed in the standard medium; other nitrogen sources produced lesser yields but still greater than those from the standard medium minus sodium glutamate:
  1. Alanine, arginine, aspartic acid, and histidine (good nitrogen sources).
  2. Ammonium chloride, cysteine, leucine, serine, and urea (fair nitrogen sources).
  3. Glycine, isoleucine, lysine, methionine, phenylalanine, potassium nitrate, sodium nitrate, threonine, tryptophan, and valine (poor nitrogen sources).
When various sulfur sources were substituted for methionine in the standard medium (on an equal sulfur basis), only cysteine and cystine produced dry weights comparable to that which developed in the standard medium. The following were very poor sulfur sources yet supported more growth than did the standard medium minus methionine: sodium sulfide, sodium thiosulfate, and thiourea. The ability of the other sulfur sources to support growth was questionable: potassium persulfate, sodium bisulfite, sodium dithionate, sodium hydrosulfite, sodium sulfate, sodium sulfite, and sodium thiocyanate. The standard medium contained only two nitrogen sources: sodium glutamate and methionine. Sodium glutamate served as a carbon source as well as a nitrogen source, but methionine could serve only as a source of sulfur.  相似文献

The biosynthesis of extracellular alkaline phosphatase in the streptomycin-resistant strains Bacillus intermedius S3-19 and S7 in the presence in the medium of 5'-nucleoside monophosphates and different sources of carbon--glucose, sodium pyruvate, sodium lactate, or glycerol--was studied. It was established that, in the presence of mononucleotides, the content of extracellular alkaline phosphatase in both strains increased; the maximal effect was caused by 5'-AMP at a concentration of 20 micrograms/ml. In medium with a low orthophosphate content, where active biosynthesis of alkaline phosphatase occurred, 1% glucose and 0.5% pyruvate stimulated this process 2.5-4 times, and 2% sodium lactate and sodium pyruvate, on the contrary, inhibited it by 20-40%. Analysis of the dynamics of growth and accumulation of extracellular phosphatase in the presence of different sources of carbon in the medium gives evidence of an interrelationship between the biosynthesis of alkaline phosphatase and carbon metabolism in Bacillus intermedius.  相似文献

目的:探讨当归口服对硫化钠致小鼠斑秃模型的影响及其可能机制。方法:将50只雄性KM小鼠随机均分为空白对照组、模型组和当归高、中、低剂量组,每组10只。模型组和当归高、中、低剂量组均以8%硫化钠酒精溶液局部外涂建立斑秃模型,当归高、中、低剂量组予以不同剂量当归煎剂灌胃,空白对照组和模型组予以等体积生理盐水灌胃,每天1次,连续7 d。采用酶联免疫吸附(ELISA)和苏木-伊红素(HE)染色法分别检测小鼠血清白细胞介素-10(IL-10)水平和单位视野毛囊数,同时对小鼠背部脱毛区毛发生长情况进行评分。结果:模型组毛发生长评分、单位视野毛囊数及血清IL-10水平均显著低于空白对照组(P0.05)。与模型组比较,高、中、低剂量组毛发生长评分、单位视野毛囊数及血清IL-10水平均显著高于模型组(P0.05)。中剂量组组毛发生长评分明显高于高、低剂量组(P0.05),低剂量组评分与高剂量组毛发生长评分、单位视野毛囊数及血清IL-10水平比较无明显差异(P0.05)。结论:口服当归对硫化钠致斑秃小鼠模型有生发作用,且以中剂量效果最佳,其生发作用可能与提高血清IL-10水平有关。  相似文献

Two representative strains of Gram-negative rumen bacteria from the genus Prevotella were used as model organisms in order to evaluate the effect of cinnamaldehyde (the secondary metabolite found in extracts of the Cinnamomum family) vs. sodium monensin on growth, cell size and cell protein production. Prevotella bryantii B(1)4 was found to be remarkably more resistant to the action of both compounds than Prevotella ruminicola 23. The approximate IC(50) concentrations of sodium monensin influenced the increase in cell size of both strains during growth, which was much more pronounced in the case of the B(1)4 strain. A similar effect was observed in strain B(1)4 when 1.438 mmol/L cinnamaldehyde was added to the growth medium, indicating a possible interference with cell division. The action of cinnamaldehyde on P. bryantii B(1)4 was concentration-dependent, in contrast to the effect observed on P. ruminicola 23.  相似文献

Interacting effects of phosphorus, potassium and sodium on the growth and nutrient uptake in Stylosanthes fruticosa were studied in its native sandy soil and solution culture. High phosphorus in the medium had adverse effects on growth. High potassium level completely masked or lessened adverse effects of high phosphorus. High sodium level adversely affected growth only in combination with high phosphorus and low potassium. Shoot nutrient content data indicated that phosphorus per se may not inhibit growth. Selective uptake of potassium against sodium was eliminated by high phosphorus. It is suggested that high concentration of phosphorus in the medium effects membrane permeability and the effect of phosphorus on plant growth is primarily through its influence on uptake of other ions.  相似文献

The effects of sodium and potassium (from 0.005 M to 0.2 M) and the two surfactants, Tween 80 and Triton X were tested.
