The insect antifeedant activities of pterocarpans and a sesquiterpene alcohol from the dichloromethane extract of Pterocarpus macrocarpus Kruz. (Leguminosae) were evaluated against the common cutworm, Spodoptera litura F. (Noctuidae), and the subterranean termite, Reticulitermes speratus (Kolbe)(Rhinotermitidae). Three pterocarpans, (?)-homopterocarpin (1), (?)-pterocarpin (2), and (?)-hydroxyhomopterocarpin (3) and the sesquiterpene alcohol, (+)-pterocarpol (5), were isolated from the dichloromethane extract of the heartwood of P. macrocarpus under guidance by a biological assay. Among these natural products, the most active insect antifeedant against both S. litura and R. speratus was 1. On the other hand, sesquiterpene alcohol 5 showed less insect antifeedant activity than the other pterocarpans against both insect species. While its methylated derivative, (?)-methoxyhomopterocarpin (4), showed high biological activity, 3 showed less insect antifeedant activity in this study. Interestingly, racemic 1 did not show insect antifeedant activity against S. litura. However, all of the test pterocarpans and isoflavones showed antifeedant activity against the test termites. Additionally, since these compounds were major constituents of P. macrocarpus, these antifeedant phenolics may act as chemical defense factors in this tree. In Thailand, lumber made from this tree is used to make furniture and in building construction due to its resistance to termite attack.  相似文献

The agricultural sector, and particularly the horticultural production, has a singular importance in agriculture, considering that it ranks second on agricultural products, nationally and worldwide. Fungal diseases are one of the major causes of vegetable loss during storage, reducing their nutritional value, quality and sale price. Vegetables are usually exposed to diverse treatments with chemical products before storage; as a result, fungal populations develop an increased resistance over time becoming more difficult to control. Because of this, research efforts toward finding more suitable chemicals to control fungal diseases are needed. Natural extracts may be an alternative solve this problem. In the present investigation the fungicidal activity of aqueous and ethanol extracts of Agave scabra was evaluated on the growth of Botrytis cinerea, Mucor sp., Aspergillus niger, Fusarium sp. and Penicillium sp., whose strains were isolated from potato and tomato. To assess their effects, the agar-dilution and agar-well techniques were performed. The ethanol extract was more effective against Botrytis cinerea and Mucor sp. when the agar-well method was used. However, when using the agar-dilution method the ethanol extract of Agave scabra inhibited the growth of Botrytis cinerea, Mucor sp. and Penicillium sp.  相似文献

  • The research conducted including its rationale: Spodoptera litura is the major pest of tomato causing significant reduction in tomato yield. Application of Plant growth promoting rhizobacteria(PGPR) prevent use of chemical fertilizer and synthetic pesticides through enhancement of plant growth and yield and induction of systemic resistance. Present investigation is an attempt to evaluate the role of PGPR, Pseudomonas putida and Rothia sp. on the physiology and yield of tomato fruit infested with the S. litura.
  • Central methods applied : The surface sterilized seeds of tomato were inoculated with 48 h culture of P. putida and Rothia sp. At 6–7 branching stage of the plant, the larvae of S. litura at 2nd in star was used to infect the tomato plant leaves.
  • Key results: The S. litura infestation decreased dry weight of shoots and roots by 46% and 22%, and significant reduction was recorded in tomato fruit yield. The P. putida and Rothia sp. inoculations alleviated the adverse effects of insect infestation and resulted in 60% increase in plant biomass and 40% increase in yield over infested plants.
  • Main conclusions including key points of discussion: PGPR: Defense appears to be mediated via increase in proline production, enhanced activities of antioxidant enzymes, stimulation in the activities of protease and polyphenol oxidases, increased contents of phenolics, protein and chlorophyll. The formulation of biopesticide involving PGPR comprise an environment friendly and sustainable approach to overcome insect infestation.

The secondary metabolites that play a defensive role in plants not only affect the growth and development of herbivores, but they can also influence their natural enemies. In the present study, the effect of coumarin was evaluated on a braconid parasitoid, Bracon hebetor (Say) (Hymenoptera: Braconidae) using polyphagous pest, Spodoptera litura (Fabricius) (Lepidoptera: Noctuidae). Reproductive potential of the parasitoid declined significantly due to parasitization of host larvae feeding on coumarin supplemented diet. Total development period of B. hebetor extended significantly (2.17–2.38 days) when the host larvae were reared on diet amended with higher concentrations (625–3,125 parts per million) of coumarin as compared to unamended diet. Ingestion of coumarin supplemented diet by S. litura adversely affected the emergence and parasitization potential of B. hebetor. As compared to the control a significant decline in hatching of eggs was recorded when parasitoid wasps developed on host larvae fed on diet containing higher concentrations of coumarin (625–3,125 ppm). Negative effects of coumarin were also recorded from the next generation of the parasitoid.  相似文献

A member of the potato proteinase inhibitor II (PPI II) gene family that encodes for a chymotrypsin iso-inhibitor has been introduced into tobacco (Nicotiana tabacum) usingAgrobacterium tumefaciens-mediated T-DNA transfer. Analysis of the primary transgenic plants (designated R0) confirmed that the introduced gene is being expressed and the inhibitor accumulates as an intact and fully functional protein. For insect feeding trials, progeny from the self-fertilization of R0 plants (designated R1) were used. Leaf tissue, either from transgenic or from control (non-transgenic) plants, was fed to larvae ofChrysodeixis eriosoma (Lepidoptera: Noctuidae, green looper),Spodoptera litura (F.) (Lepidoptera: Noctuidae) andThysanoplusia orichalcea (F.) (Lepidoptera: Noctuidae) and insect weight gain (increase in fresh weight) measured. Consistently,C. eriosoma larvae fed leaf tissue from transgenic plants expressing thePPI II gene grew slower than insects fed leaf tissue from non-transgenic plants or transgenic plants with no detectablePPI II protein accumulation. However, larvae of bothS. litura andT. orichalcea consistently demonstrated similar or faster growth when fed leaf tissue from transgenic plants compared with those fed non-transgenic plants. In agreement with the feeding trials, the chymotrypsin iso-inhibitor extracted from transgenic tobacco effectively retarded chymotrypsin-like activity measured inC. eriosoma digestive tract extracts, but not in extracts fromS. litura. We conclude, therefore, that for certain insects the use of chymotrypsin inhibitors should now be evaluated as an effective strategy to provide field resistance against insect pests in transgenic plants, but further, that a single proteinase inhibitor gene may not be universally effective against a range of insect pests. The significance of these observations is discussed with respect to the inclusion of chymotrypsin inhibitors in the composite of insect pest resistance factors that have been proposed for introduction into crop plants.  相似文献

Abstract

Millettia pachycarpa (Bentham), a perennial climbing tree, found endemic in north-east India used by tribes for treating various ailments. To ascertain the potential of M. pachycarpa for pest management, the present investigation was formulated against a polyphagous pest, Spodoptera litura (Fabricius). Growth inhibitory activity of various extracts and extract fractions was evaluated against third instar larvae of S. litura through topical as well as diet incorporation method. It was observed that dichloromethane extract showed very good growth inhibition in both topical (GI50 7.77?ppm) and diet incorporation methods (GI50< 5?ppm). The growth inhibitory potential of this plant is also demonstrated by the presence of various abnormalities and reduction in larval and pupal weight.  相似文献

Differential accumulation of plant defence metabolites has been suggested to have important ecological consequence in the context of plant–insect interactions. Feeding of generalist pests on Brassica juncea showed a distinct pattern with selective exclusion of leaf margins which are high in glucosinolates. Molecular basis of this differential accumulation of glucosinolates could be explained based on differential expression profile of BjuMYB28 homologues, the major biosynthetic regulators of aliphatic glucosinolates, as evident from quantitative real‐time PCR and promoter:GUS fusion studies in allotetraploid B. juncea. Constitutive overexpression of selected BjuMYB28 homologues enhanced accumulation of aliphatic glucosinolates in B. juncea. Performance of two generalist pests, Helicoverpa armigera and Spodoptera litura larvae, on transgenic B. juncea plants were poor compared to wild‐type plants in a no‐choice experiment. Correlation coefficient analysis suggested that weight gain of H. armigera larvae was negatively correlated with gluconapin (GNA) and glucobrassicanapin (GBN), whereas that of S. litura larvae was negatively correlated with GNA, GBN and sinigrin (SIN). Our study explains the significance and possible molecular basis of differential distribution of glucosinolates in B. juncea leaves and shows the potential of overexpressing BjuMYB28 for enhanced resistance of Brassica crops against the tested generalist pests.  相似文献

In agro-ecosystems,plants are important mediators of interactions between their associated herbivorous insects and microbes,and any change in plants induced by one species may lead to cascading effects on interactions with other species.Often,such effects are regulated by phytohormones such as jasmonic acid(JA)and salicylic acid(SA).Here,we investigated the tripartite interactions among rice plants,three insect herbivores(Chilo suppressalis,Cnaphalocrocis medinalis or Nilapai-vata lugens),and the causal agent of rice blast disease,the fungus Magnaporthe oryzae.We found that pre-infestation of rice by C.suppressalis or N.lugens but not by C.medinalis conferred resistance to M.oryzae.For C.suppressalis and N.lugens,insect infestation without fungal inoculation induced the accumulation of both JA and SA in rice leaves.In contrast,infestation by C.medinalis increased JA levels but reduced SA levels.The exogenous application of SA but not of JA conferred resistance against M.oryzae.These results suggest that preinfestation by C suppressalis or N.lugens conferred resistance against M.oryzae by increasing SA accumulation.These findings enhance our understanding of the interactions among rice plant,insects and pathogens,and provide valuable information for developing an ecologically sound strategy for controlling rice blast.  相似文献

Dothediomycetes sp., Alternaria tenuissima, Thielavia subthermophila, Alternaria sp., Nigrospora oryzae, Colletotrichum truncatum, and Chaetomium sp. were isolated as endophytic fungi from leaves and stems of the medicinal plant, Tylophora indica, based on rDNA sequencing of ITS region and microscopic examination. Alternaria tenuissima, Colletotrichum truncatum, and Alternaria sp. were found to be active against both Sclerotinia sclerotiorum and Fusarium oxysporum. Chaetomium sp. showed very mild activity against Sclerotinia sclerotiorum and Fusarium oxysporum. Whereas Dothideomycete sp. and Thielavia subthermophila showed high activity against Sclerotinia sclerotiorum. Methanol extract of Dothediomycetes sp. showed 66.5% growth inhibition (GI) at 500 μg/ml.  相似文献

We recently characterized a highly dynamic fungal disease outbreak in native populations of Nicotiana attenuata in the southwestern United States. Here, we explore how phytohormone signalling contributes to the observed disease dynamics. Single inoculation with three native Fusarium and Alternaria fungal pathogens, isolated from diseased plants growing in native populations, resulted in disease symptoms characteristic for each pathogen species. While Alternaria sp.‐infected plants displayed fewer symptoms and recovered, Fusarium spp.‐infected plants became chlorotic and frequently spontaneously wilted. Jasmonic acid (JA) and salicylic acid (SA) levels were differentially induced after Fusarium or Alternaria infection. Transgenic N. attenuata lines silenced in JA production or JA conjugation to isoleucine (JA‐Ile), but not in JA perception, were highly susceptible to infection by F. brachygibbosum Utah 4, indicating that products derived from the JA‐Ile biosynthetic pathway, but not their perception, is associated with increased Fusarium resistance. Infection assays using ov‐nahG plants which were silenced in pathogen‐induced SA accumulations revealed that SA may increase N. attenuata's resistance to Fusarium infection but not to Alternaria. Taken together, we propose that the dynamics of fungal disease symptoms among plants in native populations may be explained by a complex interplay of phytohormone responses to attack by multiple pathogens.  相似文献

Plant diseases and insect pests are serious threat to the growth and yield of oilseed rape. In this study, a binary vector carrying sporamin and chitinase PjChi-1 genes in tandem was introduced into Brassica napus cv. ZS 758 via Agrobacterium tumefaciens for dual resistance against disease and insect attack. Thirty-two regenerated plantlets exhibiting hygromycin resistance were selected following Agrobacterium-mediated transformation of 600 leaf petiole explants. Of these, 27 transformants were confirmed to carry the two transgenes as detected by polymerase chain reaction (PCR) with 4.5% transformation efficiency. Eight plantlets were randomly selected for further confirmation by Southern and northern blot hybridization analyses. Four plants carried single copy of the transgenes, while the remaining four plants carried either two or three copies of the transgenes. Moreover, expression of the sporamin transgene was detected by northern blot hybridization in transgenic lines, but not in wild-type plants. These eight T0 plants were grown in vitro, and inoculated with the Lepidoptera larvae of Plutella xylostella and with spores of the fungal pathogen of Sclerotinia sclerotiorum. Transgenic plants exhibited high levels of resistance to P. xylostella and S. sclerotiorum when compared to untransformed wild-type plants. Genetic analysis of T1 progeny confirmed Mendelian segregation of the introduced genes. Therefore, these transgenic lines demonstrate a promising potential for variety development of oilseed rape lines with enhanced resistance against both P. xylostella and S. sclerotiorum.  相似文献

Fungal endophytes were isolated from surface sterilised leaf segments of five medicinal plants collected from Mandi district, Himachal Pradesh, India. A total of 373 fungal strains belonging to 15 fungal genera and 18 species, Aspergillus niger, A. flavus, A. clavatus, A. variecolor, Penicillium chrsogenum, Alternaria alternata, Curvularia lunata, Haplosporium sp., Phoma sp., Nigrospora sp., Colletotrichum sp., Cladosporium sp., Stemphylium sp., Fusarium sp., Geotrichum sp., Phomopsis sp., Trichoderma sp., Rhizopus sp. and some sterile mycelium were isolated from all the plants. The relative frequency, isolation rate and colonisation rates of endophytes were used to study the endophytic diversity. The results showed that the highest colonisation rate (93.05%) was observed in Adhathoda vasica, while it was 91.66% in Ocimum sanctum, 85% in Viola odorata, 82.81% Cannabis sativa and lowest (61.11%) in Withania somnifera. Moreover, reading the richness and diversity of the endophytic fungi, the highest was obtained for O. sanctum, W. somnifera and C. sativa having eight species each, while lowest (6 and 4) was obtained from A. vasica and V. odorata, respectively. As the role of endophytic organisms in defensive mechanisms of plants is now well established, the present study is an important step to find new and interesting endophytes among the medicinal plants.  相似文献

Plant structural traits often act as defenses against herbivorous insects, causing them to avoid feeding on a given plant or tissue. Mineral crystals of calcium oxalate in Medicago truncatula Gaertn. (Fabaceae) leaves have previously been shown to be effective deterrents of lepidopteran insect feeding. They are also inhibitors of conversion of plant material into insect body mass during or after consumption. Growth of beet armyworm, Spodoptera exigua Hübner (Lepidoptera: Noctuidae), larvae was correspondingly greater on calcium oxalate‐defective (cod) mutants of M. truncatula with lower levels of crystal accumulation. Data presented here show that insects feeding on M. truncatula leaves with calcium oxalate crystals experience greater negative effects on growth and mandible wear than those feeding on artificial diet amended with smaller amorphous crystals from commercial preparations. Commercial calcium oxalate can be added to insect artificial diet at levels up to 7.5‐fold higher than levels found in wild‐type M. truncatula leaves with minimal effect on insect growth or lepidopteran mandibles. These data suggest that negative impacts of calcium oxalate in the diet of larvae are due to physical factors, and not toxicity of the compound, as high levels of the commercial crystals are readily tolerated. In contrast to the dramatic physical effects that M. truncatula‐derived crystals have on insect mandibles, we could detect no damage to insect peritrophic gut membranes due to consumption of these crystals. Taken together, the data indicate that the size and shape of prismatic M. truncatula oxalate crystals are important factors in determining effects on insect growth. If manipulation of calcium oxalate is to be used in developing improved insect resistance in plants, then our findings suggest that controlling not only the overall amount, but also the size and shape of crystals, could be valuable traits in selecting desirable plant lines.  相似文献

Spodoptera litura, a polyphagus insect pest of economic importance, having the ability to develop resistance to various classes of insecticides was selected for the study. Leaf dip bioassay studies were done after ten generations of selection pressure reported the development of resistance up to 80 folds by S. litura against chlorantraniliprole compared to lab susceptible strain. Bioassay studies conducted using enzyme inhibitors such as triphenyl phosphate (TPP) followed by diethyl-maleate (DEM) and piperonyl butoxide (PBO) showed good amount of synergism with chlorantraniliprole and improved efficacy against resistant strain. Findings of bioassay studies were supported by in-vitro enzyme inhibition assays. Esterase activity in gut homogenate of resistant strain was significantly inhibited by TPP suggesting esterase mediated biochemical resistance development in S. litura against chlorantraniliprole.  相似文献

An endophytic fungus was isolated from the root of the medicinal plant Moringa oleifera Lam. Based on analyzing the rDNA sequence, the fungus was identified as Nigrospora sp. This is the first report of the isolation of endophytic Nigrospora from M. oleifera. By bioassay-guided fractionation, four antifungal secondary metabolites were isolated from liquid cultures of the fungus Nigrospora sp. LLGLM003, and their chemical structures were determined to be griseofulvin (1), dechlorogriseofulvin (2), 8-dihydroramulosin (3) and mellein (4) on the basis of spectroscopic analyses. Compound 2, 3 and 4 were isolated from Nigrospora sp. for the first time. In vitro antifungal assay showed that griseofulvin displayed clear inhibition of the growth of 8 plant pathogenic fungi. Dechlorogriseofulvin and mellein exhibited only weak antifungal activities, whereas 8-dihydroramulosin displayed no antifungal activities.  相似文献

茉莉酸甲酯(Methyl jasmonate, MeJA)与植物间的防御通讯密切相关,其挥发性强,可以在空气中传播而诱导邻近植物产生防御反应,为探明MeJA诱导邻近植株抗虫性的浓度和距离效应,本研究在大棚(长4 m×宽1 m×高1 m)进口处喷以不同浓度MeJA,检测离气味源60 cm、120 cm、180 cm、240 cm和300 cm远的番茄植株叶片多酚氧化酶(Polyphenol oxidase, PPO),过氧化物酶(Peroxidase, POD)和脂氧合酶(Lipoxygenase, LOX)的活性以及斜纹夜蛾幼虫取食不同处理番茄植株叶片后的体重增长率,研究结果表明1 mmol/L和10 mmol/L MeJA挥发物可以显著诱导增强邻近番茄植株叶片PPO,POD和LOX酶的活性,以离10 mmol/L MeJA气味源60 cm处植株叶片的酶活性最高,并随着距离的增加而呈降低的趋势;取食60 cm和300 cm对照组植株叶片的斜纹夜蛾幼虫其体重增长率没有显著差异,然而1 mmol/L和10 mmol/L MeJA处理下,取食300 cm处植株叶片的斜纹夜蛾幼虫其体重增长率显著高于取食60 cm处的,相同时间下以取食10 mmol/L MeJA处理组60 cm处植株叶片的幼虫体重增长率最低,以上研究结果表明MeJA对邻近番茄植株抗虫性的诱导有明显的浓度和距离效应。  相似文献

Abstract

Citral and geraniol are dominating constituents of lemon and rose scented essential oils of Cymbopogon spp. Here, we report their antifungal activity against some pathogenic fungi Drechslera oryzae, Fusarium oxysporum, Rhizoctonia solani and Sclerotium rolfsii and larvicidal activity against two insects Helicoverpa armigera and Spodoptera litura. Results revealed that both citral and geraniol exhibited strong fungicidal effects at concentrations > 0.2 µL mL?1. Citral was found most active against F. oxysporum and S. rolfsii while geraniol against S. rolfsii at 0.2–2 µL mL?1. Nanoemulsions of citral and geraniol caused 91 and 94% larval mortality of H. armigera and S. litura, respectively, at 1% concentration. Hence, the study revealed potential antifungal and larvicidal activities of citral and geraniol, which can be implicated in formulation of biocides for effective control of phytopathogenic fungal and insects.  相似文献

The biological activity of a crude methanolic extract of Trichilia americana (Sesse and Mocino) Pennington (Meliaceae) was assessed using the Asian armyworm, Spodoptera litura (Fabr.) (Lepidoptera: Noctuidae). The extract exhibited strong antifeedant activity in a choice leaf disc bioassay with 0.18 g cm–2 extract deterring feeding by 50%. In nutritional assays, the crude extract reduced growth, consumption and the utilisation of ingested and digested food in a dose-dependent manner when fed to larvae, suggesting both antifeedant and toxic activities. When relative growth rates were plotted against relative consumption rates, the growth efficiency of the S. litura fed on diet containing T. americana crude extract was significantly less than that of control larvae. This result further indicates that the extract acts as both an antifeedant and chronic toxin. Toxicity is only seen following ingestion and was not observed following topical application or injection into the hemocoel. Larvae reared initially on extract-containing diet then transferred to control diet showed nutritional indices comparable to those of larvae fed continuously on control diet. This suggests that the extract is not permanently damaging the insect's digestive tract. The mode-of-action of the extract as a chronic toxin remains unknown.  相似文献
