Heterogenous chitinases have been introduced in many plant species with the aim to increase the resistance of plants to fungal diseases. We studied the effects of the heterologous expression of sugar beet chitinase IV on the intensity of ectomycorrhizal (ECM) colonization and the structure of fungal communities in the field trial of 15 transgenic and 8 wild-type silver birch (Betula pendula Roth) genotypes. Fungal sequences were separated in denaturing gradient gel electrophoresis and identified by sequencing the ITS1 region to reveal the operational taxonomic units. ECM colonization was less intense in 7 out of 15 transgenic lines than in the corresponding non-transgenic control plants, but the slight decrease in overall ECM colonization in transgenic lines could not be related to sugar beet chitinase IV expression or total endochitinase activity. One transgenic line showing fairly weak sugar beet chitinase IV expression without significantly increased total endochitinase activity differed significantly from the non-transgenic controls in the structure of fungal community. Five sequences belonging to three different fungal genera (Hebeloma, Inocybe, Laccaria) were indicative of wild-type genotypes, and one sequence (Lactarius) indicated one transgenic line. In cluster analysis, the non-transgenic control grouped together with the transgenic lines indicating that genotype was a more important factor determining the structure of fungal communities than the transgenic status of the plants. With the tested birch lines, no clear evidence for the effect of the heterologous expression of sugar beet chitinase IV on ECM colonization or the structure of fungal community was found.  相似文献

A field trial of 15 transgenic birch lines expressing a sugar beet chitinase IV gene and the corresponding controls was established in southern Finland to study the effects of the level of sugar beet chitinase IV expression on birch resistance to fungal diseases. The symptoms caused by natural infections of two fungal pathogens, Pyrenopeziza betulicola (leaf spot disease) and Melampsoridium betulinum (birch rust), were analysed in the field during a period of 3 years. The lines that had shown a high level of sugar beet chitinase IV mRNA accumulation in the greenhouse also showed high sugar beet chitinase IV expression after 3 years in the field. The level of sugar beet chitinase IV expression did not significantly improve the resistance of transgenic birches to leaf spot disease. Instead, some transgenic lines were significantly more susceptible to leaf spot than the controls. The level of sugar beet chitinase IV expression did have an improving effect on most parameters of birch rust; the groups of lines showing high or intermediate transgene expression were more resistant to birch rust than those showing low expression. This result indicates that the tested transformation may provide a tool for increasing the resistance of silver birch to birch rust.  相似文献

The effect of two stilbene compounds, pinosylvin and resveratrol, on the growth of several fungi was evaluated in plate tests. Wood decay tests were carried out with birch and aspen samples impregnated with the two stilbenes. In plate experiments, resveratrol had an enhancing effect on growth at concentrations where pinosylvin was already enough to prevent the growth of most fungi studied. Pinosylvin impregnated at 0.2% (w/w) concentration significantly reduced the decay caused by all fungi except Phellinus tremulae. In contrast, a resveratrol content of 0.8%, did not protect the wood from decay. A pinosylvin-synthase-encoding gene from Pinus sylvestris was transferred into aspen (Populus tremula) and two hybrid aspen clones (Populus tremula×tremuloides) by Agrobacterium tumefaciens-mediated transformation. Transgenic plants accumulated pinosylvin synthase-specific mRNA and showed stilbene synthase enzyme activity in vitro. Transgenic aspen line H4 showed increased resistance to Phellinus tremulae, while two hybrid aspen transformants decayed faster than the control trees. The diet and population dynamics of the Eurasian lynxLynx lynx Linnaeus, 1758 as well as an index of its main prey abundance were studied in transitional mixed forests of northern Belarus in 1985-2004. Monitoring of the lynx population and its main potential prey (the mountain hareLepus timidus, and the roe deerCapreolus capreolus) was done by snow-tracking. Also, abundance of tetraonids (Tetraonidae) was monitored by sight count. Hare numbers were fairly stable during the study period, whereas density of the roe deer population markedly increased, and tetraonids decreased. Composition of the lynx diet was stable seasonally. Lynx fed mostly on hares, roe deer and birds (usually tetraonids) year-round. However, the share of roe deer in lynx diet increased significantly during the period of its higher abundance and the share of tetraonids decreased with their decreasing numbers. There was also a remarkable increase of lynx population, which followed that of the roe deer, despite the pronounced decline of tetraonids. The results of the study emphasised the importance of roe deer as a prey of the Eurasian lynx.  相似文献

The research aim was to assess the effects of the plant hormone abscisic acid (ABA) and the growth regulator paclobutrazol (PBZ) on root system development during the in vitro culture of different birch and aspen genotypes. The studied genotypes involved two aspen (Populus tremula and Populus tremuloides × P. tremula) and two silver birch (Betula pendula) trees, with one of the birches characterized by its inability to root in vitro. For experiments, apical shoot segments were cultured on nutrient medium enriched with either ABA or PBZ. Additionally, the analysis of the endogenous hormones in shoots developed on hormone‐free medium was conducted by high‐performance liquid chromatography. The endogenous concentration of auxin indole‐3‐acetic acid was much higher in the aspens than that in the birches, while the highest concentration of ABA was found in the root‐forming birch. The culturing of this birch genotype on medium enriched with ABA resulted in an increased root length and a higher number of lateral roots without any negative effect on either shoot growth or adventitious root (AR) formation, although these two processes were largely inhibited by ABA in the aspens. Meanwhile, PBZ promoted AR formation in both aspen and birch cultures but impaired secondary root formation and shoot growth in birches. These results suggest the use of ABA for the in vitro rooting of birches and PBZ for the rooting of aspens.  相似文献

Direct and interactive effects of CO2 and light on tree phytochemistry and insect fitness parameters were examined through experimental manipulations of plant growth conditions and performance of insect bioassays. Three species of deciduous trees (quaking aspen, Populus tremuloides; paper birch, Betula papyrifera; sugar maple, Acer saccharum) were grown under ambient (387±8 μL/L) and elevated (696±2 μL/L) levels of atmospheric CO2, with low and high light availability (375 and 855 μmol×m?2×s?1 at solar noon). Effects on the population and individual performance of a generalist phytophagous insect, the white‐marked tussock moth (Orgyia leucostigma) were evaluated. Caterpillars were reared on experimental trees for the duration of the larval stage, and complementary short‐term (fourth instar) feeding trials were conducted with insects fed detached leaves.
Phytochemical analyses demonstrated strong effects of both CO2 and light on all foliar nutritional variables (water, starch and nitrogen). For all species, enriched CO2 decreased water content and increased starch content, especially under high light conditions. High CO2 availability reduced levels of foliar nitrogen, but effects were species specific and most pronounced for high light aspen and birch. Analyses of secondary plant compounds revealed that levels of phenolic glycosides (salicortin and tremulacin) in aspen and condensed tannins in birch and maple were positively influenced by levels of both CO2 and light. In contrast, levels of condensed tannins in aspen were primarily affected by light, whereas levels of ellagitannins and gallotannins in maple responded to light and CO2, respectively.
The long‐term bioassays showed strong treatment effects on survival, development time, and pupal mass. In general, CO2 effects were pronounced in high light and decreased along the gradient aspen birch maple. For larvae reared on high light aspen, enriched CO2 resulted in 62% fewer survivors, with increased development time, and reduced pupal mass. For maple‐fed insects, elevated CO2 levels had negative effects on survival and pupal mass in low light. For birch, the only negative CO2 effects were observed in high light, where female larvae showed prolonged development. Fourth instar feeding trials demonstrated that low food conversion efficiency reduced insect performance. Elevated levels of CO2 significantly reduced total consumption, especially by insects on high light aspen and low light maple.
This research demonstrates that effects of CO2 on phytochemistry and insect performance can be strongly light‐dependent, and that plant responses to these two environmental variables differ among species. Overall, increased CO2 availability appeared to increase the defensive capacity of early‐successional species primarily under high light conditions, and of late‐successional species under low light conditions. Due to the interactive effects of tree species, light, CO2, and herbivory, community composition of forests may change in the future.  相似文献

The aim with this study was to, under controlled conditions, determine the food preference of mountain haresLepus timidus Linnaeus, 1758 and bank volesClethrionomys glareolus (Schreber, 1780) for a substantial part of the woody plants potentially available for these herbivores during winter. In addition, we compared hare and vole preference patterns. Thirteen woody plant species were simultaneously presented to 9 captive voles and 9 captive hares in preference tests during winter. Consumption by hares from 50 g bundles (one per species) was measured after 3 h, whereas shoot consumption by voles was measured after 12 h. Both hares and voles preferred deciduous species to conifers,Populus tremula, andVaccinium myrtillus being the most preferred species. However, there was considerable variation in palatability among deciduous plant species and only a marginally significant correlation was found between hare and vole preference. One striking differences between hares and voles was that Sorbusaucuparia was the most utilised by voles but the least preferred by hares. In conclusion, deciduous plant species were generally considerably more palatable to hares and voles than conifers, which is consistent with current theories. Nevertheless, the high variation in palatability among deciduous trees and the difference in preference between hares and voles indicate more complex and species-specific patterns with regard to plant— animal interactions. The latter also suggests that hares and voles differ in their dietary adaptations and have different dietary constrains.  相似文献

A sugar beet chitinase gene driven by the (42) CaMV 35S promoter was introduced into silver birch (Betula pendula) through Agrobacterium-mediated transformation. Transgenic shoots were regenerated and grown on WPM medium supplemented with 150 mg/ml kanamycin. From a total of 220 explants, 52 transgenics were obtained and 13 transgenic lines were randomly taken for molecular analysis to confirm the presence of the introduced sugar beet chitinase 4 cDNA by polymerase chain reaction and Southern hybridisation. All 13 transgenic lines were confirmed to contain the gene and further characterised. Northern blot analysis of total RNA indicated that the transgenic lines differed with respect to the steady-state levels of chitinase mRNA. Transgenic lines with high levels of mRNA of chitinase 4 cDNA consistently showed higher levels of resistance to Pyrenopeziza betulicola than transgenics with intermediate or low mRNA levels or a non-transgenic control plant. This report demonstrates that the constitutive expression of this gene in transgenic birch lines increased the resistance of birch against the leaf spot fungus P. betulicola.  相似文献

Juvenile trees of temperate and boreal regions cease growth and set buds in autumn in response to short day-lengths (SD) detected by phytochrome. Growth cessation and bud set are prerequisites for the development of winter dormancy and full cold hardiness. In this study we show that the SD-requirement for bud set and cold hardening can be overcome in hybrid aspen (Populus tremula L. × tremuloides Michx.) by low night temperature and inhibition of gibberellin (GA) biosynthesis. Bud set and increased cold hardiness were observed under normally non-inductive long day-length (LD) in wild-type plants, when exposed to low night temperature and paclobutrazol. In addition, the effect of PHYA overexpression could be overcome in transgenic plants, producing bud set and cold acclimation by treatment with: SD, low night temperature and paclobutrazol. After cold acclimation, the degree of bud dormancy was lower for wild-type plants prior treated with LD and transgenic plants (overexpressing PHYA), than SD-treated, wild-type plants. Thus, low night temperature in combination with reduced GA content induced bud set and promoted cold hardiness under normally non-inductive photoperiods in hybrid aspen, but was unable to affect development of dormancy. This might suggest separate signalling pathways from phytochrome regulating the induction of cold/cold hardiness and bud dormancy in hybrid aspen or alternatively, there might be one pathway that fails to complete its action in the transgenic and paclobutrazol treated plants.  相似文献

Summary To evaluate the general extent to which sex-related differences in palatability occur in boreal dioecious woody plants, males and females of five dioecious woody plant species were presented to free-ranging mountain hares (Lepus timidus) during winter. Hares strongly preferred branches from male plants when feeding on Populus tremula and Salix caprea and weakly preferred male S. pentandra. However, they did not show any sex-related preference when feeding on the other two species studied (Myrica gale and Juniperus communis). Nitrogen concentration and, to some degree, digestibility showed strong relationships with hare food preferences. By contrast, the concentration of phenolics was only weakly related to feeding preference. Phenolics could, nevertheless, still be important if only one or a few specific compounds deter hare feeding. These results indicate that sex-related differences in plant palatability in the boreal forest might be more widespread than previously believed, particularly for species of the family Salicaceae. Thus, herbivores might be responsible for the female-biased sex ratios found in willow populations in northern Scandinavia (e.g. Elmqvist et al. 1988).  相似文献

Summary Plant carbon/nutrient balance has been implicated as an important factor in plant defensive chemistry and palatability to herbivores. We tested this hypothesis by fertilizing juvenile growth form Alaska paper birch and green alder with N, P and N-plus-P in a balanced 2x2 factorial experiment. Additionally, we shaded unfertilized plants of both species. Fertilization with N and N-plus-P increased growth of Alaska paper birch, reduced the concentration of papyriferic acid in internodes and increased the palatability of birch twigs to snowshoe hares. Shading decreased birch growth, decreased the concentration of papyriferic acid in internodes and increased twig palatability. These results indicate that the defensive chemistry and palatability of winter-dormant juvenile Alaska paper birch are sensitive to soil fertility and shade. Conversely the defensive chemistry and palatability of green alder twigs to snowshoe hares were not significantly affected by soil fertility or shade. The greater sensitivity of Alaska paper birch defensive chemistry and palatability to snowshoe hares in comparison to green alder is in agreement with the hypothesis that early successional woody plants that are adapted to high resource availability are more plastic in their chemical responses to the physical environment than are species from less favorable environments.  相似文献

Among the prey of wolfCanis lupus Linnaeus, 1758, in many European regions, roe deerCapreolus capreolus, red deer Cervuselaphus and wild boarSus scrofa are of primary significance. Up to now, these species were not represented in models which allow an estimation of the consumed biomass on the basis of wolf scats. To develop such formulae for roe deer, red deer and wild boar, we fed these species to 5 captive adult wolves of European origin. In 10 feeding trials, the wolves were fed 10 prey, whose body mass ranged from 15.3 to 118.5 kg. We related the body mass of the prey in accordance with Weaver (1993) to one collectable scat (Model l:y=0.00554+0.00457x) and, as it is difficult to define one unit scat, we alternatively related it to one meter length of scats (Model 2:y=0.141+0.0487x). The Model 1 equation differed from both of Weaver’s (1993) regressions, which he had calculated on the basis of his own feeding trials and on the basis of the summarised data of Floydet al. (1978), Traves (1983) and Weaver (1993). Applying an equation gained through the European ungulates resulted in lower estimates of prey mass. Model 1 and Model 2 estimates were comparable in size.  相似文献

We studied the three‐way interaction of elevated CO2, nitrogen (N), and temperature (T), and the two‐way interaction of elevated CO2 and early‐season defoliation on the secondary chemistry and resistance of Eurasian silver birch (Betula pendula) and North American paper birch (B. papyrifera) against the Eurasian hare (Lepus timidus) and the North American eastern cottontail rabbit (Sylvilagus floridanus), respectively. Elevated CO2 decreased the palatability of winter‐dormant silver and paper birch stems to both hares and rabbits, respectively. But the effect on hares was only apparent at intermediate levels of N fertilization. Elevated T had no effect on palatability. The effects of elevated CO2, N, and T on levels of silver birch bark phenolics and terpenoids were dominated by two‐way interactions between N and CO2, and N and T. Generally, however, N amendments elicited a parabolic response in carbon partitioning to most biosynthetic classes of silver birch phenolics (i.e. highest concentrations occurring at intermediate N). CO2 elevation was most enhancing at highest levels of N. On the other hand, T increases, more often than not, elicited reductions in phenolics, but especially so at the highest N level. In the case of B. papyrifera, elevated CO2 increased carbon partitioning to Folin‐Denis stem and branch phenolics and condensed tannins. Early‐season defoliation, on the other hand, had no effect on phenolics and tannins but lowered both N and energy levels of branches. Elevated CO2 substantially ameliorated the negative effects of severe defoliation on tree growth. These results support the hypothesis that continuing anthropogenic alterations of the atmosphere may trigger significant changes in plant phenotypic resistance to mammalian herbivores owing to an increasing net carbon balance between the highly vagile supply and demand capacities of plant carbon sources and sinks.  相似文献

Three tame, hand-reared adult roe deer (Capreolus capreolus) were fed a low-quality diet (blueberry stems) or high-quality concentrate either restricted or ad libitum in summer and winter. Total mean retention time (TMRT) and rumen retention time (RRT) were significantly longer, indigestible fill was significantly higher and apparent digestibility and intake were significantly lower when fed blueberry stems compared to concentrate. The feeding regimes of concentrate did not affect the digesta kinetic estimates, except that total fill and apprent digestibility were significantly higher when deer were fed ad libitum than when they were fed a restricted ration. TMRT was significantly longer when they were fed a restricted ration of blueberry stems than for ad libitum intake, whereas all other estimates were equal. Intake was significantly higher and TMRT and RRT were significantly shorter in summer compared to winter when they were fed concentrate. These findings support the theory that roe deer, as small-bodied concentrate selectors, are adapted to high intake, rapid turnover, and rapid digestion when fed high-quality concentrate. When fed a low-quality diet the roe deer may be limited by rumen bulk. A strategy based on increased rumen retention accompanied by increased cell-wall digestion has therefore severe limitations in roe deer. This indicates that roe deer are dependent on a selective feeding strategy to enhance winter survival.  相似文献

Application experiments have suggested that short‐day‐induced cessation of elongation growth in trees is caused by photoperiodic regulation of the conversion of gibberellin GA19 to GA20. In the present study we examined further the photoperiodic control of GA metabolism in trees with focus on the conversion of GA19 in Salix pentandra, hybrid aspen (Populus tremula × tremuloides) and silver birch (Betula pendula) using [17,17‐2H2]‐GA19 or unlabelled GAs in application studies. GA20 and GA1 were able to restore growth also in hybrid aspen and silver birch under short days (SD), whereas GA19 had no or only a very small activity. Contrary to hybrid aspen and S. pentandra, the activity of GA20 in silver birch was significantly lower than that of GA1. Gas chromatography‐mass spectrometry (GC‐MS) analysis revealed a smaller turnover of [2H2]‐GA19 in SD than in long days (LD) in hybrid aspen. No such difference in turnover of [2H2]‐GA19 was observed in photoperiod‐insensitive hybrid aspen overexpressing PHYA. Application of unlabelled GAs to seedlings of S. pentandra, hybrid aspen and silver birch under SD followed by quantification of metabolites by GC‐MS analysis, showed that applied GA19 was not readily converted to GA20 and GA1. Although the sensitivity to GAs is also known to decrease under SD, the present data are in favour of a photoperiodic regulation of the metabolism of GA19in vivo in the woody species S. pentandra, hybrid aspen and silver birch. The data might also suggest that silver birch differs from S. pentandra and hybrid aspen by exhibiting a possible photoperiodic control also of the conversion of GA20 to GA1.  相似文献

Aim We aimed to describe the large‐scale patterns in population density of roe deer Caprelous capreolus in Europe and to determine the factors shaping variation in their abundance. Location Europe. Methods We collated data on roe deer population density from 72 localities spanning 25° latitude and 48° longitude and analysed them in relation to a range of environmental factors: vegetation productivity (approximated by the fraction of photosynthetically active radiation) and forest cover as proxies for food supply, winter severity, summer drought and presence or absence of large predators (wolf, Canis lupus, and Eurasian lynx, Lynx lynx), hunter harvest and a competitor (red deer, Cervus elaphus). Results Roe deer abundance increased with the overall productivity of vegetation cover and with lower forest cover (sparser forest cover means that a higher proportion of overall plant productivity is allocated to ground vegetation and thus is available to roe deer). The effect of large predators was relatively weak in highly productive environments and in regions with mild climate, but increased markedly in regions with low vegetation productivity and harsh winters. Other potentially limiting factors (hunting, summer drought and competition with red deer) had no significant impact on roe deer abundance. Main conclusions The analyses revealed the combined effect of bottom‐up and top‐down control on roe deer: on a biogeographical scale, population abundance of roe deer has been shaped by food‐related factors and large predators, with additive effects of the two species of predators. The results have implications for management of roe deer populations in Europe. First, an increase in roe deer abundance can be expected as environmental productivity increases due to climate change. Secondly, recovery plans for large carnivores should take environmental productivity and winter severity into account when predicting their impact on prey.  相似文献

Despite the involvement of many members of the chitinase family in the plant immune system, the exact functions of most chitinases remain poorly understood, especially in plant defense responses to phytophagous insects. Here, the gene GhChi6, which encodes a chitinase protein in Gossypium hirsutum, was shown to be induced by cotton aphid feeding and mechanical wounding. Overexpression of GhChi6 in Arabidopsis plants improved their defense response to aphids. The activities of chitinase and PPO in GhChi6 transgenic Arabidopsis plants were higher than those in wild-type plants. Callose deposition in leaves from GhChi6 transgenic Arabidopsis plants was clearly increased compared with wild-type plants. The levels of AtEDS1, AtPAD4, and AtEDS5 in the SA signaling pathway were higher in GhChi6 transgenic Arabidopsis Line4 than those in wild-type plants, while the expression levels of AtLOX2 in the JA signaling pathway and AtEIN2 in the ethylene signaling pathway were lower in GhChi6 transgenic Arabidopsis Line4 than those in wild-type plants. These results collectively showed that the cotton chitinase gene GhChi6 modulated the plant defense response to aphid attack, which may help guide strategies for improving cotton aphid prevention.

In this study, we demonstrate that the mountain hare and roe deer compete with each other. This was determined using "natural experiments" of populations found in sympatry and allopatry on the islands along the west coast of Norway. We demonstrate that both species occupy the same habitats, share the same food resources and that food availability is limited. Two browsing species as different in size as roe deer and mountain hare might be expected to partition the available vegetation (e.g. woody scrub) in terms of height above ground level. However, from the evidence collected, the feeding-height-separation hypothesis must be rejected as an explanation for ecological separation between roe deer and mountain hares because there was extensive height overlap in resource utilisation by both species and neither species changed its feeding height in response to the presence of the other. Total browse utilisation did not increase when both species were together; rather, species-specific browse utilisation declined when the other species was present. However, the foraging behaviour of each herbivore varied significantly between the allopatric and sympatric sites. When both herbivores were present, the clip diameter of shoots browsed by mountain hares declined to match those selected by roe deer, while roe deer switched from a browse-dominated diet to a diet dominated by winter-green gramineae. The change to smaller-diameter shoots likely resulted in the hare increasing its intake of digestion-inhibiting or toxic secondary metabolites, while the alternative choice of digging through the snow like roe deer to reach the winter-green gramineae is a practice considered energetically too costly for hares. On this basis, we conclude that the enforced switch to a nutritionally inferior diet by mountain hares at the sympatric sites may result in changes to growth rate and body size which consequently impact on mortality and may explain the competitive superiority of the roe deer.  相似文献

1 The feeding preference of the adult pine weevil Hylobius abietis (L.) (Coleoptera: Curculionidae) for Betula pendula Roth was studied in no‐choice and paired‐choice feeding experiments. 2 In the first no‐choice test, large quantities of silver birch bark in Petri dishes were consumed; on average, the daily consumption of each weevil was 67 mm2. 3 In the second no‐choice test, the weevils were offered 1‐year‐old silver birch seedlings for 6 days. Initially, the weevils fed mostly on the stem bases; later, they moved upward to feed on other parts of the stems. In addition to the main shoots, scars caused by gnawing were also found on leaf bases, blades, veins and petioles. Feeding resulted in the death of the main stems in 15% of the seedlings. 4 In the paired‐choice tests, the conifers were preferred to silver birch, even though a large amount of silver birch was also consumed in the presence of conifers. 5 In the paired‐choice tests, equal amounts of Scots pine and Norway spruce were always consumed. When hybrid aspen was offered, only small amounts were gnawed.  相似文献
