Genetic and biochemical evidence has established that a SNARE complex consisting of syntaxin 5 (Sed5)-mYkt6 (Ykt6)-GOS28 (Gos1)-GS15 (Sft1) is required for transport of proteins across the Golgi stack in animals (yeast). We have utilized quantitative immunogold labeling to establish the cis-trans distribution of the v-SNARE GS15 and the t-SNARE subunits GOS28 and syntaxin 5. Whereas the distribution of the t-SNARE is nearly even across the Golgi stack from the cis to the trans side, the v-SNARE GS15 is present in a gradient of increasing concentration toward the trans face of the stack. This contrasts with a second distinct SNARE complex, also required for intra-Golgi transport, consisting of syntaxin 5 (Sed5)-membrin (Bos1)-ERS24 (Sec22)-rBet1 (Bet1), whose v-(rBet1) and t-SNARE subunits (membrin and ERS24), progressively decrease in concentration toward the trans face. Transport within the stack therefore appears to utilize countercurrent gradients of two Golgi SNAREpins and may involve a mechanism akin to homotypic fusion.  相似文献

A combination of cell-free biochemical and morphological studies has revealed that a coated bud-coated vesicle transport system shuttles newly synthesized proteins through the successive processing compartments of the Golgi apparatus. These Golgi-coated vesicles operate in a manner formally analogous to the clathrin-coated, pit-coated vesicle system responsible for receptor-mediated endocytosis; however Golgi-coated vesicles do not contain clathrin.  相似文献

We have used an in vitro Golgi protein transport assay dependent on high molecular weight (greater than 100 kD) cytosolic and/or peripheral membrane proteins to study the requirements for transport from the cis- to the medial-compartment. Fractionation of this system indicates that, besides the NEM-sensitive fusion protein (NSF) and the soluble NSF attachment protein (SNAP), at least three high molecular weight protein fractions from bovine liver cytosol are required. The activity from one of these fractions was purified using an assay that included the second and third fractions in a crude state. The result is a protein of 115-kD subunit molecular mass, which we term p115. Immunodepletion of the 115-kD protein from a purified preparation with mAbs removes activity. Peptide sequence analysis of tryptic peptides indicates that p115 is a "novel" protein that has not been described previously. Gel filtration and sedimentation analysis indicate that, in its native state, p115 is a nonglobular homo-oligomer. p115 is present on purified Golgi membranes and can be extracted with high salt concentration or alkaline pH, indicating that it is peripherally associated with the membrane. Indirect immunofluorescence indicates that p115 is associated with the Golgi apparatus in situ.  相似文献

An N-ethylmaleimide-sensitive transport component (NSF) has been purified on the basis of its ability to support transport between Golgi cisternae. We now report that NSF is needed for membrane fusion. Thus, when NSF is withheld from incubations of Golgi stacks with cytosol and ATP, uncoated transport vesicles accumulate. Biochemical experiments confirm this conclusion and reveal that NSF is needed to form the first of two previously described prefusion complexes. NSF, therefore, acts within a cascade in which a vesicle-cisterna complex is matured until it is competent for fusion. We suggest that this reflects the stepwise assembly of a multisubunit "fusion machine" following vesicle attachment.  相似文献

Involvement of GTP-binding "G" proteins in transport through the Golgi stack   总被引:101,自引:0,他引:101  
GTP gamma S irreversibly inhibits protein transport between successive compartments of the Golgi stack in a cell-free system. Fluoride, potentiated by the addition of aluminum ion, also causes a strong inhibition. These are hallmarks of the involvement of a guanine nucleotide-binding or regulatory "G" protein. Inhibition by GTP gamma S requires a cytosolic inhibitory factor that binds to Golgi membranes during inhibition. Preincubation experiments reveal that GTP gamma S blocks the function of acceptor Golgi but not donor Golgi membranes. More specifically, a processing step in between vesicle attachment and the actual fusion event seems to be affected. Electron microscopy demonstrates a corresponding 5-fold accumulation of non-clathrin-coated buds and vesicles associated with the Golgi cisternae during inhibition by GTP gamma S.  相似文献

The lysosomotropic amine primaquine has previously been shown to inhibit both secretory and recycling processes of cells in culture. We have used a cell-free assay that reconstitutes glycoprotein transport through the Golgi apparatus to investigate the mechanism of action of primaquine. In this assay, primaquine inhibits protein transport at a half-maximal concentration of 50 microM, similar to the concentration previously reported to disrupt protein secretion in cultured cells. Kinetic analysis of primaquine inhibition indicates that its point of action is at an early step in the vesicular transport mechanism. Primaquine does not inhibit the fusion of vesicles already attached to their target membranes. Primaquine irreversibly inactivates the membranes that form transport vesicles (donor), but not the membranes that are the destination of those vesicles (acceptor). Morphological data indicate that primaquine inhibits the budding of vesicles from the donor membranes. Once formed, the vesicles are refractile to primaquine action, and their attachment to and fusion with acceptor membranes proceeds unimpeded. In addition to illuminating the mechanism of action of primaquine, this study suggests that the selective action of this agent will make it a useful tool in the study of the formation of transport vesicles.  相似文献

A number of protein toxins from plants and bacteria take advantage of transport through the Golgi apparatus to gain entry into the cytosol where they exert their action. These toxins include the plant toxin ricin, the bacterial Shiga toxins, and cholera toxin. Such toxins bind to lipids or proteins at the cell surface, and they are endocytosed both by clathrin-dependent and clathrin-independent mechanisms. Sorting to the Golgi and retrograde transport to the endoplasmic reticulum (ER) are common to these toxins, but the exact mechanisms turn out to be toxin and cell-type dependent. In the ER, the enzymatically active part is released and then transported into the cytosol, exploiting components of the ER-associated degradation system. In this review, we will discuss transport of different protein toxins, but we will focus on factors involved in entry and sorting of ricin and Shiga toxin into and through the Golgi apparatus.  相似文献

A new composite membrane was designed and studied for permselectivity of various molecular weight proteins. The membrane is composed of a porous substrate membrane [Durapore; poly(vinylidene fluoride)] coated with a thin dense layer of regenerated cellulose. This composite membrane was fabricated by spin coating a cellulose acetate solution onto the membrane, followed by alkaline hydrolysis of the cellulose acetate coating to regenerate cellulose. The coated layer was physically characterized by scanning electron microscopy (SEM) and infrared (IR) spectroscopy. In addition, the water uptake into and permeation properties of macromolecules across the coated and uncoated membranes were studied. A typical composite membrane coating was 0.8 +/- 0.2 mum thick, resulting in a molecular weight cutoff of approximately 40,000 daltons. Isolated Golgi membranes incubated in the presence of ATP and a cytosolic protein fraction form a population of coated buds or vesicles from the Golgi cisternae. The coats do not have the characteristic hexagonal-pentagonal basketwork of clathrin, and do not react with anti-clathrin polyclonal antibody. The conditions that produce these apparently nonclathrin-coated buds also reconstitute protein transport between compartments of the Golgi stack. The membrane of the buds contains the glycoprotein in transit through these Golgi stacks (VSV-encoded G protein). This suggests that protein transport through the Golgi stack is mediated by a new type of coated vesicle that does not contain clathrin. The concentration of G protein in the coated buds reflects the local concentration of G protein in the cisternae, raising the possibility that the Golgi coated vesicles may be "bulk" membrane carriers.  相似文献

We have applied agarose gel electrophoresis as a novel step in the purification of clathrin-coated vesicles. Preparations of coated vesicles obtained by sedimentation velocity and isopycnic centrifugation are resolved into two distinct fractions upon electrophoresis. The slower migrating fraction contains smooth vesicles, whereas the faster contains only coated vesicles and empty clathrin coats. The faster mobility of the coated vesicles is primarily caused by the acidic nature of clathrin. Coated vesicles from three different cell types have different mobilities. In each case, however, all of the major polypeptides previously attributed to coated vesicles comigrate with the now homogeneous particles, even though a powerful ATPase activity is completely removed.  相似文献

Members of the rab/YPT1/SEC4 gene family of small molecular weight GTPases play key roles in the regulation of vesicular traffic between compartments of the exocytic pathway. Using immunoelectron microscopy, we demonstrate that a dominant negative rab1a mutant, rab1a(N124I), defective for guanine nucleotide binding in vitro, leads to the accumulation of vesicular stomatitis virus glycoprotein (VSV-G) in numerous pre-cis-Golgi vesicles and vesicular-tubular clusters containing rab1 and beta-COP, a subunit of the coatomer complex. Similar to previous observations (Balch et al. 1994. Cell. 76:841-852), VSV-G was concentrated nearly 5-10-fold in vesicular carriers that accumulate in the presence of the rab1a(N124I) mutant. VSV-G containing vesicles and vesicular-tubular clusters were also found to accumulate in the presence of a rab1a effector domain peptide mimetic that inhibits endoplasmic reticulum to Golgi transport, as well as in the absence of Ca2+. These results suggest that the combined action of a Ca(2+)-dependent protein and conformational changes associated with the GTPase cycle of rab1 are essential for a late targeting/fusion step controlling the delivery of vesicles to Golgi compartments.  相似文献

Engineered protein aggregates ranging up to 400 nm in diameter were selectively deposited within the cis-most cisternae of the Golgi stack following a 15 degrees C block. These aggregates are much larger than the standard volume of Golgi vesicles, yet they are transported across the stack within 10 min after warming the cells to 20 degrees C. Serial sectioning reveals that during the peak of anterograde transport, about 20% of the aggregates were enclosed in topologically free "megavesicles" which appear to pinch off from the rims of the cisternae. These megavesicles can explain the rapid transport of aggregates without cisternal progression on this time scale.  相似文献

A protein designated as a 100-kDa protein on the basis of sodium dodecyl sulfate gel electrophoresis was purified from coated vesicles obtained from bovine brain, with uncoated vesicles as starting material. Two gel filtration steps, one involving 0.5 M tris(hydroxymethyl)aminomethane, pH 8.0, buffer, and the other 0.01 M tris(hydroxymethyl)aminomethane, pH 8.0, and 3 M urea buffer, were employed. The purified protein has a native molecular weight of 114,000 as determined by sedimentation equilibrium analysis. Circular dichroism data showed that the protein has 28% helical structure, 29% beta-structure, and 15% beta-turns, and the rest is random coil. Addition of the purified protein to clathrin results in the polymerization of clathrin to homogeneous size baskets of sedimentation velocity 150 S. A scan of the Coomassie Blue stained electrophoresis gels of the polymerized baskets shows that, for every clathrin trimer, there is approximately one 100-kDa protein molecule.  相似文献

Two different methods, stimulation of transport by fatty acyl-coenzyme A (CoA) and inhibition of transport by a nonhydrolyzable analogue of palmitoyl-CoA, reveal that fatty acylation is required to promote fusion of transport vesicles with Golgi cisternae. Specifically, fatty acyl-CoA is needed after the attachment of coated vesicles and subsequent uncoating of the vesicles, and after the binding of the NEM-sensitive fusion protein (NSF) to the membranes, but before the actual fusion event. We therefore suggest that an acylated transport component participates, directly or indirectly, in membrane fusion.  相似文献

Examination of a cell-free reconstitution of intercompartmental transport through the Golgi apparatus has enabled detection of two intermediates in the pathway (Balch, W. E., Glick, B. S., and Rothman, J. E. (1984) Cell 39, 525-536). These intermediates are thought to represent stages in the budding and fusion reactions of transport vesicles mediating such a transport process. Here we describe a new transport intermediate that is interposed between the previously established primed donor formation and the N-ethylmaleimide (NEM)-resistant acceptor intermediates. Consumption of this intermediate requires much less cytosol than its formation, and thus it has been termed the "dilution-resistant" intermediate. The dilution-resistant intermediate only forms in the presence of donor and acceptor membranes, and its consumption is sensitive to NEM. The transition from this state to the later, NEM-resistant form of the prefusion complex requires ATP as well as cytosol and may represent a processing of transport vesicles to permit their fusion.  相似文献

Membrane proteins were isolated from purified Trypanosoma brucei coated endocytotic vesicles by phase separation with Triton X-114. The largest abundant membrane protein was a doublet band with a molecular mass of about 77 kDa. A specific antiserum was prepared against this protein by immunization with antigen bands excised from sodium dodecyl sulfate-polyacrylamide gels. Immunoblot analyses with this antiserum showed that the 77-kDa protein was present in other T. brucei, in T. congolense, and in T. vivax bloodstream-stage parasites but absent from procyclic (tsetse fly midgut)-stage trypanosomes. Antigenically related molecules of 58, 300, and 15.5 kDa were also detected. The 300- and 15.5-kDa molecules were not in purified coated vesicles; they were detected in whole bloodstream- and procyclic-form T. brucei organisms. Immunofluorescent studies localized the antigen to the region between the flagellar pocket and the nucleus of bloodstream-form parasites. Ultrastructurally, the antigen was detected on membranes of endosomes and lysosome-like structures that contained endocytosed markers.  相似文献
