Carvacrol and thymol in combination at 6.7 mM each completely inhibited the production of short-chain volatile fatty acids and lactate from cattle waste in anoxic flasks over 23 days. Fecal coliforms were reduced from 4.6 × 106 to 2.0 × 103 cells per ml 2 days after treatment and were nondetectable within 4 days. Total anaerobic bacteria were reduced from 8.4 × 1010 to 1.5 × 107 cells per ml after 2 days and continued to be suppressed to that level after 14 days. If the concentration of carvacrol or thymol were doubled (13.3 mM), either could be used to obtain the same inhibitory fermentation effect. We conclude that carvacrol or thymol may be useful as an antimicrobial chemical to control pathogens and odor in stored livestock waste.  相似文献

Various antimicrobial agents were evaluated with the purpose of reducing the microbial fermentation in stored cattle waste and the resulting odor emissions. Duplicate sealed 2-L flasks with 500 ml waste slurry, with and without antimicrobial inhibitors, were used to measure the production of short-chain volatile fatty acids, lactate, and total fermentation gas over 27–30 days. A combination of chlorhexidine diacetate (2 mM), iodoacetate (2 mM), and α-pinene (3.8 mM) reduced gas production 80% (1000 ml to 200 ml) and total volatile fatty acid production 50% (145 mM to 72 mM). Pinene had little antimicrobial effect; rather, it served as an effective masking agent, giving the waste a less offensive odor. A combination of chlorhexidine diacetate and the deaminase inhibitor, diphenyliodonium chloride (1.3 mM) had a similar effect in reducing short-chain volatile fatty acid production (145 mM to 80 mM). It is concluded that a combination of antimicrobial agents may be useful in controlling odor emissions and conserving organic matter in livestock wastes, therefore providing a potentially more useful byproduct waste when used as plant fertilizer. AIM: To determine how eugenol affects fermentation parameters and faecal coliforms in cattle and swine waste slurries stored anaerobically. METHODS AND RESULTS: Waste slurries (faeces:urine:water, 50:35:15) were blended with and without additives and aliquoted to triplicate 1-l flasks. Faecal coliforms were eliminated in cattle and swine waste slurries within 1 or 2 days with additions of eugenol at 10.05 mm (0.15%) and 16.75 mm (0.25%). At these concentrations volatile fatty acids (VFA) were reduced ca 70 and 50% in cattle and swine waste, respectively, over 6-8 weeks. Additionally, in cattle waste, eugenol stimulated the accumulation of lactate (>180 mm) when compared with thymol treatment (20 mm lactate). In swine waste, lactate accumulation did not occur without additives; eugenol and thymol stimulated lactate accumulation to concentrations of 22 and 32 mm, respectively. CONCLUSIONS: Eugenol added to cattle waste may be more beneficial than thymol because not only does it effectively control faecal coliforms and odour (VFA production), it also stimulates lactate accumulation. This in turn, causes the pH to drop more rapidly, further inhibiting microbial activity and nutrient emissions. SIGNIFICANCE AND IMPACT OF THE STUDY: Plant essential oils have the potential to solve some of the environmental problems associated with consolidated animal feeding operations. Thymol and eugenol reduce fermentative activity, thus, have the potential to reduce emissions of greenhouse gases and odour, and curtail transmission of pathogens in cattle and swine wastes.  相似文献

Growth performance, carcass quality, survival and hematological responses of Oncorhynchus mykiss juveniles (initial weight 8.4 ± 0.1 g) fed diets containing thymol‐carvacrol powder at the levels of 0, 1.0, 2.0, 3.0 g kg?1 were tested. Thymol‐carvacrol powder originated from Origanum vulgare, a Mediterranean plant, added to diets. Each diet was fed to triplicate groups of fish for 45 days. Fish fed diets containing thymol‐carvacrol had significantly higher final weight and growth than the control group. Food conversion ratio in fish fed diets containing 2.0 and 3.0 g kg?1 thymol‐carvacrol was statistically better than in other treatments. Survival was not different among all treatments. The number of lymphocytes increased when thymol‐carvacrol was used at higher levels. Furthermore, whole body lipid content was higher in fish fed 1.0 and 2.0 g kg?1 thymol‐carvacrol than the other groups, but body protein in the group fed 3.0 g kg?1 was higher than in other groups. Also, body ash in control and 1.0 g kg?1 was higher than in other groups. Whole body dry matter was not affected by dietary treatments. These results indicated that dietary administration of thymol‐carvacrol can influence some growth, hematological parameters and tissue composition in rainbow trout juveniles.  相似文献

Biofilms can cause severe problems in industrial paper mills, particularly of economic and technological types (clogging of filters, sheet breaks or holes in the paper, machine breakdowns, etc.). We present here some promising results on the use of essential oil compounds to control these biofilms. Biofilms were grown on stainless-steel coupons with a microbial white water consortium sampled from an industrial paper mill. Five essential oil compounds were screened initially in the laboratory in terms of their antimicrobial activity against planktonic cells and biofilms. The three most active compounds were selected and then tested in different combinations. The combination finally selected was tested at the pilot scale to confirm its efficiency under realistic conditions. All the compounds tested were as active against biofilms as they were against planktonic cells. The most active compounds were thymol, carvacrol, and eugenol, and the most efficient combination was thymol–carvacrol. At a pilot scale, with six injections a day, 10 mM carvacrol alone prevented biocontamination for at least 10 days, and a 1 mM thymol–carvacrol combination enabled a 67 % reduction in biofilm dry matter after 11 days. The use of green antimicrobials could constitute a very promising alternative or supplement to the treatments currently applied to limit biofilm formation in the environment of paper mill machines.  相似文献

Two trials were conducted to study the effects of dose and formulation of carvacrol and thymol on bacterial counts, metabolites and functional traits of the gut in weaned piglets. In the first experiment (Exp. I), 25 piglets (28 d, 6.59 ± 0.48 kg BW) were allocated to five dietary treatments: a control diet, or the same diet supplemented with either carvacrol or thymol at doses of 500 and 2000 mg kg?1. In the second experiment (Exp. II), 35 piglets (28 d, 7.99 ± 0.73 kg BW) were assigned to seven dietary treatments: the same control diet as in Exp. I, or this diet supplemented with thymol in one of three formulations (on celite, on alphacel or microencapsulated) at doses of 500 and 2000 mg kg?1. At 11/12 days post-weaning piglets were euthanised, and digesta from stomach, proximal and distal small intestine were sampled for bacteriological and biochemical analysis. Small intestinal tissue was sampled for histo-morphological determinations. In none of the experiments or sections of the gut was the number of bacteria lowered by the carvacrol or thymol supplementation. In Exp. I, the villus/crypt ratio at the distal small intestine for the experimental diets (1.30–1.32) was higher than for the control diet (1.24) (p < 0.05). Thymol fed animals in Exp. II had a lower number of intra-epithelial lymphocytes at the proximal (p < 0.05) and at the distal (p < 0.1) small intestine as compared to control animals. Mean concentration of the active ingredient in the stomach and proximal small intestine for the 2000 mg kg?1 carvacrol diet was 521 and 5 mg kg?1 fresh digesta, respectively, and for the 2000 mg kg?1 thymol diets it ranged between 475 and 647 and between 13 and 24 mg kg?1 fresh digesta, respectively. Cumulative absorption in the proximal small intestine was higher than 90% for all treatments and was not affected by formulation type. These data suggest that carvacrol and thymol can improve gut health, but evidence for clear antimicrobial effects towards the major culturable bacteria of the pig foregut is limited.  相似文献

Five experiments were conducted to either ferment fresh shrimp or crab waste with molasses, molasses and bacterial inoculant, or to preserve it with salt. Experiment 1 was a 4 × 2 factorial arrangement. Crab waste was combined with 0, 5, 10, or 15% liquid molasses, and stored in mini-silos (15 l) with or without lids for 14 days. The addition of molasses slightly decreased pH and offensive odors; mini-silo temperatures without lids were higher than those with lids. Experiment 2 was a 5 × 2 factorial arrangement designed to enhance fermentation. Fresh shrimp waste was combined with 0, 10, 15, 20, or 25% dry molasses and 0 or 1.0 × 108 colony forming bacteria/g inoculant and ensiled for six days. As the level of molasses increased, dry matter and lactic acid increased but, the pH, crude protein, ammonia acetic, butyric, and propionic acid concentrations decreased. Significant molasses by inoculant interactions occurred which were highly variable for each acid. Evidence of fermentation was supported by production of lactic acid at all levels of molasses. The pH decreased from 7.7 in the untreated waste to an average of 7.4 for the 10, 15 and 20% molasses treated wastes to 6.8 in the 25% molasses treated waste. The high pH was an indication that the waste may be unstable with longer storage (> 6 days). Therefore, in Experiment 3, designed as a 2 × 2 factorial arrangement, shrimp waste treated with 15 and 20% molasses, with or without inoculant was ensiled for 21 days to test stability. By day 21, shrimp waste had deteriorated as indicated by a mean pH of 7.5, low lactic acid, and high butyric acid concentration, an unacceptable odor, and the presence of mold on the surface of the samples.In Experiments 4 and 5, shrimp or crab waste was combined with salt at 0, 2.5, 5.0, 7.5, 10.0, and 12.5%. Increasing levels of salt decreased crude protein percent, ammonia concentration, and lactic and volatile fatty acids while increasing the pH and improving the acceptability of the odors in both the shrimp and crab wastes. Treatment of crustacean waste with 7.5% or greater salt was more effective at preserving crude protein and minimizing odor than either dry or liquid molasses.  相似文献

Bakhtiarian savory (Satureja bachtiarica Bunge) is an endemic herb growing wild in Iran with interesting pharmacological and biological properties. In this research, the variability of essential oil content and phytochemical compositions among seven populations of this species was studied in both natural and field conditions. There was a wide interpopulation variability in phytochemical variation of studied populations under both natural and field conditions, indicating the existence of different chemotypes. Six chemical compositions showed high value in studied plants so that carvacrol and thymol were known as major compounds and confirmed by principal component analysis. Ilam population had the highest thymol in both field (77.10 %) and natural (42.32 %) conditions and was known as a thymol chemotype. Beside, Yazd and Shahrekord populations, respectively, showed the highest carvacrol percentages in both field (84.83 and 63.81 %) and natural (72.50 and 58.05 %) conditions and were known as carvacrol chemotypes. In other populations, carvacrol and thymol were the main components and showed partly equal percentage and these populations were known as carvacrol/thymol chemotypes. According to Pearson correlation analysis, carvacrol was negatively correlated with p-cymene, γ-terpinene and thymol (r = ?0.61, ?0.70 and ?0.98, respectively), indicating very high reverse correlation between carvacrol and thymol. In conclusion, the high value of the essential oil compositions in studied S. bachtiarica populations could provide useful information for conservation and selection of cross parents in breeding programs to develop improved cultivars.  相似文献

Bioaerosol concentrations inside one naturally ventilated and one mechanically ventilated swine finishing barn were assessed by sampling air using membrane filtration and impaction (six-stage Andersen sampler), and assayed by culture method. The barns, located on the same commercial farm in northeast Kansas, did not show any significant difference (p > 0.05) in concentrations of total and respirable airborne microorganisms. The overall mean total concentrations inside the two barns were 6.6 × 104 colony forming units (CFU)/m3 (SD = 3.8 × 104 CFU/m3) as measured by filtration and 8.6 × 104 CFU/m3 (SD = 5.1 × 104 CFU/m3) by impaction. The overall mean respirable concentrations were 9.0 × 103 CFU/m3 (SD = 4.1 × 103 CFU/m3) measured by filtration and 2.8 × 104 CFU/m3 (SD = 2.2 × 104 CFU/m3) by impaction. Total and respirable CFU concentrations measured by impaction were significantly (p < 0.05) higher than that by filtration. The persistent strains of microorganisms were various species of the following genera: Staphylococcus, Pseudomonas, Bacillus, Listeria, Enterococcus, Nocardia, Lactobacillus, and Penicillium. It appears that filtration sampling can be used for a qualitative survey of bioaerosols in swine barns while the Andersen sampler is suitable for both quantitative and qualitative assessments. Microorganisms are central to both the beneficial (organic degradation, nutrient removal, biogas production) and detrimental (odor production, pathogen contamination) effects of swine waste storage systems. In this study, both quantitative (real-time polymerase chain reaction) and qualitative (denaturing gradient gel electrophoresis, cloning, sequence analysis) molecular analyses were used to track spatial and temporal changes in the microbial community of swine slurry from a 0.4 ha anaerobic lagoon. The lagoon, located in a region of western Kentucky which has a humid, subtropical environment, was sampled on a monthly basis (n = 10) over a period of one year at four different depths (top, 51 cm from the top, 152 cm from the top, and bottom >198 cm). The concentration and diversity of Bacteroides sp. was seasonal (up to 90% decrease between March and June). Hespellia sp. and other clostridial species, on the other hand, were endemic in the slurry (concentrations up to 1.0 × 107 cells mL?1 slurry) regardless of time of the year or lagoon depth. Results suggest that there were seasonal effects on the microbial community in the swine lagoon, while the effect of depth was not as pronounced. Seasonal changes in the microbial community in stored wastes may be (directly or indirectly) correlated with changes in malodor emissions from lagoons.  相似文献

Concentrations of hypophosphite and phosphite oxidizing bacteria were found to be high, relative to bacterial concentrations growing on phosphate, in sediment and soil during winter and summer seasons from 12 common terrestrial and aquatic sites using a most probable number method. The percent of total culturable bacterial concentrations that could use these reduced phosphorus compounds as a sole source of phosphorus were as follows: hypophosphite, 7–100%; phosphite, 10–67%; aminoethylphosphonate, 34–270%. The average MPN/g (±SEM) was as follows: phosphate, 6.19 × 106 (±2.40 × 106); hypophosphite, 2.61 × 106 (±1.35 × 106) phosphite, 1.91 × 106 (±1.02 × 106); aminoethylphosphonate, 3.90 × 106 (± 1.95 × 106). Relatively high concentrations of reduced phosphorus oxidizing bacteria were found in both pristine sites and sites with urban and agricultural disturbance. Concentrations of reduced phosphorus oxidizing bacteria in anoxic sediments and soil were equivalent. Our data indicate that reduced phosphorus oxidizing bacteria are abundant in the environment and provide strong evidence for the importance of bacterial P oxidation in nature.  相似文献

We used various approaches to establish a comprehensive budget of methane (CH4) emissions from the Seine basin, including direct emissions from livestock and soils as well as emissions from the drainage network. For the direct emissions from livestock, we used official livestock census numbers and emission factors (CH4 emitted by each animal species per head per year) available in the literature. For the emissions from soils, we based our estimates on experimental measurements in closed chambers installed on different agricultural plots, forest, and grasslands in 2008 and 2009. The results were extrapolated to the whole Seine basin, including grassland, cropland, and forest soil distributions in the Seine basin. The CH4 emissions from the Seine drainage network were also based on measurements of sampled waters in various rivers and streams (from headwaters to estuary) during different seasons in 2007, 2008, and 2010. After chemical analysis of CH4 concentrations in the water samples using a gas chromatographic technique and calculation of the CH4 supersaturation by stream order in rivers of the Seine basin (from 1 to 8) and by season we could estimate the CH4 emissions for the whole water surface area of the Seine drainage network. The livestock of the Seine basin produce CH4 emissions amounting to 166 × 106 kg C year?1, among which cattle are responsible for 85 %. The total CH4 emission from the Seine drainage network was estimated at 0.3 × 106 kg C year?1, large rivers being responsible for the largest proportion. Ebullition could account for an additional 0.2 × 106 kg C year?1. Soils of the Seine basin are a net sink for CH4 (9.4 × 106 kg C year?1). The water and soils fluxes are low with regard to emissions by livestock, but domestic waste, through landfills, could contribute an additional 40 × 106 kg C year?1.  相似文献

Odor control and disposal of swine waste have inhibited expansion of swine production facilities throughout the United States. Swine waste odor is associated primarily with high concentrations of volatile fatty acids (VFAs). Here, we demonstrate that stimulated Fe(III) reduction in hog manure can rapidly remove the malodorous compounds and enhance methane production by 200%. As part of these studies, we enumerated the indigenous Fe(III)-reducing population in swine waste and identified members of the family Geobacteraceae as the dominant species. These organisms were present at concentrations as high as 2 × 105 cells g−1. Several pure cultures of Fe(III) reducers, including Geobacter metallireducens, Geobacter humireducens, Geobacter sulfurreducens, Geobacter grbiciae, Geothrix fermentans, and Geovibrio ferrireducens, readily degraded some or all of the malodorous VFAs found in swine manure. In contrast, Shewanella algae did not degrade any of these compounds. We isolated an Fe(III) reducer, Geobacter strain NU, from materials collected from primary swine waste lagoons. This organism degraded all of the malodorous VFAs tested and readily grew in swine waste amended with Fe(III). When raw waste amended with Fe(III) was inoculated with strain NU, the VFA content rapidly decreased, corresponding with an almost complete removal of the odor. In contrast, the raw waste without Fe(III) or strain NU showed a marked increase in VFA content and a rapid pH drop. This study showed that Fe(III) supplementation combined with appropriate bioaugmentation provides a simple, cost-effective approach to deodorize and treat swine waste, removing a significant impediment to the expansion of pork production facilities.  相似文献

Management practices from large-scale swine production facilities have resulted in the increased collection and storage of manure for off-season fertilization use. Odor and emissions produced during storage have increased the tension among rural neighbors and among urban and rural residents. Production of these compounds from stored manure is the result of microbial activity of the anaerobic bacteria populations during storage. In the current study, the inhibitory effects of condensed quebracho tannins on in vitro swine manure for reduction of microbial activity and reduced production of gaseous emissions, including the toxic odorant hydrogen sulfide produced by sulfate-reducing bacteria (SRB), was examined. Swine manure was collected from a local swine facility, diluted in anaerobic buffer, and mixed with 1 %?w/v fresh feces. This slurry was combined with quebracho tannins, and total gas and hydrogen sulfide production was monitored over time. Aliquots were removed periodically for isolation of DNA to measure the SRB populations using quantitative PCR. Addition of tannins reduced overall gas, hydrogen sulfide, and methane production by greater than 90 % after 7 days of treatment and continued to at least 28 days. SRB population was also significantly decreased by tannin addition. qRT-PCR of 16S rDNA bacteria genes showed that the total bacterial population was also decreased in these incubations. These results indicate that the tannins elicited a collective effect on the bacterial population and also suggest a reduction in the population of methanogenic microorganisms as demonstrated by reduced methane production in these experiments. Such a generalized effect could be extrapolated to a reduction in other odor-associated emissions during manure storage.  相似文献

Gamma-ray-induced DNA single-strand scissions and the ability to repair the scissions in spermatogonia from young mice and in spermatozoa from adult mice were studied quantitatively by an alkaline sucrose density-gradient centrifugation method. The average size of DNAs in non-irradiated spermatogonia was 2.6–3.0 × 108 daltons, similar to those of a spermatid-rich population, and the size of DNA in non-irradiated spermatozoa was 1.2 × 108 daltons.In spermatogonia, the radiosensitivity of DNA was 0.42 single-strand breaks/ 1012 daltons of DNA/rad in oxic conditions and only 0.24 under anoxic conditions. In spermatozoa the break efficiency of DNA was 0.22 single-strand breaks/1012 daltons of DNA/rad under oxic conditions and altered little under anoxic irradiation. The DNA scissions were efficiently repaired in spermatogonia within 10 min, whereas the breaks in spermatozoa were not rejoined at all even after two days of post-irradiation time.The radiosensitivities of DNA, repair capability and non- and/or slow-reparable DNA scissions were compared in spermatogonium-rich, spermatid-rich and spermatozoan-rich populations.  相似文献

Animal waste odors arising from products of anaerobic microbial metabolism create community relations problems for livestock producers. We investigated a novel approach to swine waste odor reduction: the addition of FeCl3, a commonly used coagulant in municipal wastewater treatment, to stimulate degradation of odorous compounds by dissimilatory iron-reducing bacteria (DIRB). Two hypotheses were tested: (i) FeCl3 is an effective source of redox-active ferric iron (Fe3+) for dissimilatory reduction by bacteria indigenous to swine manure, and (ii) dissimilatory iron reduction results in significant degradation of odorous compounds within 7 days. Our results demonstrated that Fe3+ from FeCl3 was reduced biologically as well as chemically in laboratory microcosms prepared with prefiltered swine manure slurry and limestone gravel, which provided pH buffering and a substrate for microbial biofilm development. Addition of a 1-g liter−1 equivalent concentration of Fe3+ from FeCl3, but not from presynthesized ferrihydrite, caused initial, rapid solids flocculation, chemical Fe3+ reduction, and Eh increase, followed by a 2-day lag period. Between 2 and 6 days of incubation, increases in Fe2+ concentrations were accompanied by significant reductions in concentrations of volatile fatty acids used as odor indicators. Increases in Fe2+ concentrations between 2 and 6 days did not occur in FeCl3-treated microcosms that were sterilized by gamma irradiation or amended with NaN3, a respiratory inhibitor. DNA sequences obtained from rRNA gene amplicons of bacterial communities in FeCl3-treated microcosms were closely related to Desulfitobacterium spp., which are known representatives of DIRB. Use of iron respiration to abate wastewater odors warrants further investigation.  相似文献

Carvacrol has been recognized as an efficient growth inhibitor of food pathogens. However, carvacrol oil is poorly water-soluble and can be oxidized, decomposed or evaporated when exposed to the air, light, or heat. To overcome these limitations, a carvacrol nanoemulsion was developed and its antimicrobial activity against food pathogens evaluated in this study. The nanoemulsion containing 3% carvacrol oil, 9% surfactants (HLB 11) and 88% water, presented good stability over a period of 90 days. In general, the carvacrol nanoemulsion (MIC: 256 µg ml−1 for E. coli and Salmonella spp., 128 µg ml−1 for Staphylococcus aureus and Pseudomonas aeruginosa) exhibited improved antimicrobial activity compared to the free oil. The carvacrol nanoemulsion additionally displayed bactericidal activity against Escherichia coli, P. aeruginosa and Salmonella spp. Therefore, the results of this study indicated that carvacrol oil nanoemulsions can potentially be incorporated into food formulations, wherein their efficacy for the prevention and control of microbial growth could be evaluated.  相似文献

In order to clarify possible cytological mechanisms that underlie the beneficial effects of carvacrol-bearing essential oils on health and mental abilities, we studied one of them (oregano essential oil) in experiments on transformed cultured Chinese hamster cells. Possible cytotoxic or mitogenic effects of the preparation at various concentrations were preliminarily estimated by analyzing the cell culture density after 4 days of cultivation. The preparation concentration in the growth medium (on carvacrol basis) varied from 1 × 10?15 up to 5 × 10?4 M (on carvacrol basis). As a result, two concentrations were selected for further experiments, including 2.5 × 10?5 M as the maximal absolutely non-toxic concentration and 2.5 × 10?4 M as the concentration at which the oregano essential oil decreased approximately 2-fold the final cell density of the grown culture. It was found that the preparation at 2.5 × 10?5 M had no effect on either the colony-forming ability of the cells or the saturation density of the culture (which is a marker of its ??biological age??) or kinetics of its ??stationary phase aging?? (degradation of cultured cells in the stationary phase of growth, similar to age-related changes of the cells in aging organism). On the contrary, the oregano essential oil at 2.5 × 10?4 M abruptly diminished colony-forming ability of the cells and influenced as a ??pro-aging?? factor on the saturation density of the cell culture and kinetics of the cell death induced by ??stationary phase aging.?? Based on our own concept of aging and the data obtained, we assumed that detected increase in the life span of mice under the influence of the oregano essential oil could be determined by certain functional changes at the organismal level only, but is not associated with any geroprotective (anti-aging) activity of the preparation, which is manifested at the cellular level and improves the cell viability.  相似文献
