Hydrogen ion titration of an affinity-purified mannose/glucose-specific lectin from Cajanus cajan pulse was carried out at 30°C and ionic strength of 0.15 by a discontinuous method. The titration was reversible in the pH range 2–12.0. The numbers of different ionizable groups per 39,000 g of the lectin were 43 carboxyl groups (pKint = 3.93), 10 imidazole groups, 21 ε-amino groups, 12.8 phenoxyl groups (pKint = 10.0), and 5 guanidyl groups. Only seven tyrosine residues of the lectin were dissociated under native conditions. The remaining six tyrosines became available for titration upon denaturation of the lectin in 9 M urea.  相似文献

Lectins from Canavalia brasiliensis (CnBr), Cratylia floribunda (CFL), Vatairea macrocarpa (VML) and Phaseolus vulgaris (PHA) aggregate Rhizobiumbacteria. The relationship between specific sedimentation rate, (based on bacterial dry biomass) of bacterial aggregates and lectin concentrations was hyperbolic and showed bacterial surface affinity by lectins. R. tropici (Rt), R. leguminosarum bv. phaseoli (Rlp) and R. etli (Re) surfaces showed predominantly receptors of galactosidic nature. The Rt surfaces showed very high affinities (k s = ±8.6 × 10–8 ag lectin protein ml–1) by Gal-specific lectins (PHA and VML), and very low affinities (ks=± 4.9 × 10–6) by Glc-specific lectins (CnBr and CFL). The Rlp surface had intermediate affinities by lectins. The Re surface showed high affinities by PHA (ks= ±1.26 × 10–8) and intermediate affinities by VML, CnBr and CFL. The relationship between sedimentation specific (based on lectin weight) and bacterial density was a sigmoid and showed lectin affinity by Rt surfaces. The bacterial sedimentation showed positive cooperative binding of lectins. The Vmax induced by Glc-specific lectins was ±20 of that produced by Gal-specific lectins. The PHA affinity (ks= 1.19 mg dry biomass ml–1) was larger than VML (ks = 1.23). The Glc-specific lectin affinities were smaller than those of Gal-specific. The apparent binding site number of lectins (napp) was: 2.7-PHA; 2.2-VML; 3.2-CFL and 3.2-CnBr. The dissociation constant, ks, of lectin-binding kinetics decreased with sugar-hapten treatment (10 M). The napp decreased in PHA and CFL, increasing in VML + sugar-hapten treatment. This study showed that there is a difference in Rhizobium surfaces for lectin binding.  相似文献

We have demonstrated thatAmaranthus leucocarpus lectin hemagglutinating activity was powerfully inhibited by the T-antigen, containing Gal(1–3)GalNAc(1–3)Ser/Thr, and the Tn-antigen, which contains GalNAc(1–3)Ser/Thr. This suggests that the acetamido group at C-2 and the axial -OH at C-4 of theN-acetyl-D-galactopyranosylamine ring are important for lectin binding. The hemagglutination assays also established that desialylated and Pronase-treated human typeO erythrocytes with an M phenotype were better recognized than erythrocytes from all other blood groups. The recognition was dependent on pH and ionic strength.  相似文献

Hydrogen ion titration of an affinity-purified mannose/glucose-specific lectin from Cajanus cajan seeds at 30°C and ionic strength of 0.15 was reported earlier by our lab (Salahuddin and Khan, 1998). Further work has been carried out on the systematic study of H+ titration of Cajanus cajan lectin at three different temperatures, i.e., 10°, 20°, and 30°C at ionic strength of 0.15. In all, 88 protons were dissociated reversibly in pH range 2–12. Different ionizable groups present in lectin were also determined with respect to their heat of ionization. Heat of ionization, H, for 43 carboxyl groups was 1.92 Kcal/mol, and for 7 phenoxyl groups, 6.84 Kcal/mol. Spectrophotometric titration was performed in the presence and absence of 0.1 M methyl -d-mannopyranoside to characterize the role of phenoxyl groups present at the active site. One phenoxyl group was found to be involved in carbohydrate binding. The presence of two carboxyl groups per subunit at the active site was confirmed by H+ titration in the presence and absence of 0.1 M methyl -d-mannopyranoside. Hemagglutinating and IgM precipitating activity was checked for the chemically modified carboxyl and phenoxyl group preparation. Loss of activity further confirmed the participation of carboxyl as well as phenoxyl groups at the active site. It is interesting to note the near-UV CD changes in the aromatic environment/conformation of the lectin in the presence of specific sugar, further indicating the possible participation of phenoxyl groups in the carbohydrate-binding pocket.  相似文献

Three mannose-binding lectins were assayed in artificial diets for their toxic and growth-inhibitory effects on nymphal development of the peach-potato aphid Myzus persicae. The snowdrop (Galanthus nivalis) lectin GNA was the most toxic, with an induced nymphal mortality of 42% at 1500 g ml–1 (30 M) and an IC50 (50% growth inhibition) of 630 g ml–1 (13 M). The daffodil (Narcissus pseudonarcissus) lectin NPA and a garlic (Allium sativum) lectin ASA induced no significant mortality in the range 10–1500 g ml–1, but did result in growth inhibition of 59% (NPA) and 26% (ASA) at 1500 g ml–1 (40 M for NPA, 63 M for ASA). All three lectins were responsible for a slight but significant growth stimulation when ingested at 10 g ml–1, reaching +26%, +18% and +11% over the control values for the garlic lectin, the daffodil lectin and the snowdrop lectin, respectively. GNA, as well as the glucose/mannose binding lectin Concanavalin A, were also provided at sublethal doses throughout the life cycle of the aphids, and effects on adult performance were monitored. Adult survival was not significantly altered, but both lectins adversely affected total fecundity and the dynamics of reproduction, resulting in significant reduction in calculated r ms (population intrinsic rate of natural increase) on lectin-containing diets. These effects are discussed in relation to the use of transgenic plants expressing these toxic lectins for potential control of aphid populations.  相似文献

A sialic acid binding lectin, AchatininH, was purified in single step from the hemolymph of the land snail, Achatina fulica, by the affinity chromatography on sheep submaxillary mucin coupled to Sepharose 4B. The yield of the lectin was found to be 3 mg from 100 ml of hemolymph. The homogeneity of the lectin was established by alkaline gel electrophoresis, immunodiffusion, immunoelectrophoresis and analytical isoelectrophoresis. The molecular weight of the native protein was 242000, having identical subunits of Mr 15000. The lectin agglutinated rabbit erythrocytes in the presence of Ca2–. The inhibition study clearly suggests that the binding site of the lectin recognizes sialic acid as the immunodominant sugar. This was further confirmed by the observation that there was a marked decrease of agglutinating activity of the lectin with neuraminidase treated rabbit erythrocytes and asialofetuin was unable to inhibit the activity of AchatininH. Among the inhibitors used the glycoconjugate containing 2-6 linkages of N-acetylneuraminic acid with subterminal galactopyranose or 2-acetamido-2-deoxy-galactopyranose residue was found to be better inhibitor than that containing 23 linkages of N-acetyl neuraminic acid. Besides that sialoglycoprotein containing both N and O type of glycosidic linkages plays an important role in binding with the lectin. Fetuin was found to be the best inhibitor.  相似文献

Various monosaccharides and oligosaccharides were used to define the specificity of theButea frondosa lectin using the hapten inhibition technique of human erythrocyte agglutination. AlthoughB. frondosa lectin exhibited higher affinity forN-acetylgalactosamine, lactose andN-acetyllactosamine appeared to be relatively good inhibitors of haemagglutination. The behaviour ofN-acetyllactosamine-type oligosaccharides and glycopeptides on a column ofB. frondosa lectin immobilized on Sepharose 4B showed that the sugar-binding specificity of the lectin is directed towards unmaskedN-acetyllactosamine sequences. Substitution of theseN-acetyllactosamine sequences by sialic acid residues completely abolished the affinity of the lectin for the saccharides. The presence of one or several Fuc(1-3)GlcNAc groups completely inhibited the interaction between the glycopeptides and the lectin. Substitution of the core -mannose residue by an additional bisecting (1-4)GlcNAc residue decreases the affinity of the lectin for these structures as compared with the unsubstituted ones.  相似文献

The three-dimensional structure ofDolichos biflorus seed lectin has been constructed using five legume lectins for which high resolution crystal structures were available. The validity of the resulting model has been thoroughly investigated. Final structure optimization was conducted for the lectin complexed with GalNAc, providing thereby the first three-dimensional structure of lectin/GalNAc complex. The role of theN-acetyl group was clearly evidenced by the occurrence of a strong hydrogen bond between the protein and the carbonyl oxygen of the carbohydrate and by hydrophobic interaction between the methyl group and aromatic amino acids. Since the lectin specificity is maximum for the Forssman disaccharide GalNAc(1–3)GalNAc-O-Me and the blood group A trisaccharide GalNAc(1–3)[Fuc(1–2)]Gal-O-Me, the complexes with these oligosaccharides have been also modelled.  相似文献

α-d-Mannopyranosyl units were attached to an aromatic scaffold through disulfide linkages to obtain mono- to trivalent glycosylated ligands for lectin binding studies. Isothermal titration calorimetric (ITC) measurements indicated that binding affinities of these derivatives to Concanavalin A (Con A) were comparable to or slightly higher than that of methyl α-d-mannopyranoside (Ka values in the range of 104 M−1). The stoichiometries of the lectin-ligand complexes were in agreement with the formal valencies (1–3) of the respective ligands indicating cross-linking in interactions with the di- and trivalent derivatives. Multivalency effects could not, however, be observed with the latter. These ligands were shown to bind to the carbohydrate binding site of Con A using saturation transfer difference (STD) NMR competition experiments.  相似文献

The direct double-antibody enzymelinked immunosorbent assay system was used in the detection and measurement of seed lectins from peanut (Arachis hypogaea L.) and soybean (Glycine max L.) plants (PSL and SBL, respectively) that had been inoculated with their respective rhizobia. Concentrations of PSL dropped to undetectable levels in peanut roots at 9 d and stems and leaves at 27 d after planting; SBL could no longer be detected in soybean roots at 9 d and in stems and leaves at 12 d. A lectin antigenically similar to PSL was first detected in root nodules of peanuts at 21 d reaching a maximum of 8 g/g at 29 d then decreasing to 2.5 g/g at 60 d. There was no evidence of a corresponding lectin in soybean nodules.Sugar haemagglutination inhibition tests with neuraminidase-treated human blood cells established that PSL and the peanut nodule lectin were both galactose/lactose-specific. Further tests with rabbit blood cells demonstrated a second mannosespecific lectin in peanut nodule extracts that was not detected in root extracts of four-week-old inoculated plants or six-week-old uninoculated plants, although six-week-old root extracts from inoculated plants showed weak lectin activity. The root extracts from both nodulated and uninoculated plants contained another peanut lectin that agglutinated rabbit but not human blood cells. Summary The effect of pH buffers and related compounds on the conductance of an outwardly rectifying anion channel has been studies using the patch-clamp technique. Single-channel current-voltage relationships were determined in solutions buffered by trace amounts of bicarbonate and in solutions containing N-substituted taurines (HEPES, MES, BES, TES) and glycines (glycylglycine, bicine and tricine), Tris andbis-Tris at millimolar concentrations. HEPES (pKa=7.55) reduced the conductance of the channel when present on either side of the membrane. Significant inhibition was observed with 0.6mm HEPES on the cytoplasmic side (HEPES i ) and this effect increased with [HEPES i ] so that conductance at the reversal potential was diminished 25% with 10mm HEPES i )and 70% at very high [HEPES i ]. HEPES i block was relieved by applying positive voltage but positive currents were not consistent with a Woodhulltype blocking scheme in that calculated dissociation constants and electrical distances depended on HEPES concentration. Results obtained by varying total HEPES i concentration at constant [HEPES] and vice versa suggest both the anionic and zwitterionic (protonated) forms of HEPES inhibit. Structure-activity studies with related compounds indicate the sulfonate group and heterocyclic aliphatic groups are both required for inhibition from the cytoplasmic side. TES (pKa=7.54), substituted glycine buffers (pKa=8.1–8.4) andbis-Tris (pKa=6.46) had no measurable effect on conductance and appear suitable for use with this channel.  相似文献

A Forssman antigen (GalNAc1-3GalNAc1-3Gal1-4Gal1-4Glc1-1Cer)-binding lectin has been purified from velvet bean (Mucuna derringiana) seeds by a combination of affinity chromatography and reversed phase HPLC. This lectin agglutinates both native and trypsin-treated sheep erythrocytes as well as trypsinized rabbit erythrocytes, but neither native rabbit nor human erythrocytes, irrespective of blood group type. SDS-PAGE and gel filtration chromatography reveal the lectin to be a homodimer consisting of two 54 kDa subunits linked by non-covalent bonds. The results obtained by quantitative precipitation, haemagglutination inhibition and TLC overlay assays indicate that theMucuna lectin specifically recognizes Forssman antigen and Forssman disaccharide (GalNAc1-3GalNAc)-related structures. High molecular weight lectins (> 100 kDa) from seeds of the legumes Canavalia brasiliensis (CnBr), Cratylia floribunda (CFL), Phaseolus vulgaris (PHA) and Vatairea macrocarpa (VML), temporarily stimulate the respiration of Rhizobium tropici-CIAT899 and R. etli-CFN42. These stimulants were significant (P < 0.05) in bacterial suspensions (> 2.85 mg dry biomass ml–1), having at least 6200 molecules of lectins per bacteria. The VML (20 g ml–1), induced specific O2 demand of 2.3–2.5 M O2 min–1 mg dry biomass–1, in CFN42 and CIAT899, respectively. However, CnBr, CFL and PHA induced smaller demands of O2 (5×), in both strains. The order of affinities of the lectins was approximately VML > PHA > CFL > CnBr, with regard to respiratory stimuli in CIAT899 strain. The co-administration of 10 g VML ml–1 and 9.8 M galactose, in CIAT899 suspensions, reduced the respiratory stimuli significantly in relation to the treatment with VML alone. These respiratory stimuli, induced by the lectins, increase the significance of the interaction lectin × Rhizobium in terms of bacterial physiology. Its understanding could be important in relation to bacterial symbiotic behaviour.  相似文献

The specificity ofMoluccella laevis lectin was investigated by analysing its binding to glycosphingolipids separated on thin-layer chromatograms or adsorbed on microtitre wells. The binding activity of the lectin was highest for glycosphingolipids with terminal -linkedN-acetylgalactosamine, both in linear structures, as the Forssman glycosphingolipid, GalNAc3GalNAc3Gal4Gal4Glc1Cer, and in branched structures, as glycosphingolipids with the blood group A determinant, GalNAc3(Fuc2)Gal. In addition, the lectin bound, though considerably more weakly, to linear glycosphingolipids with terminal -linked galactose. When considering the use of theM. laevis lectin for biochemical and medical purposes this cross-reactivity may be of importance. Nomenclature: The glycosphingolipid nomenclature follows the recommendations by the IUPAC-IUB Commission on Biochemical Nomenclature (CBN for Lipids:Eur J Biochem (1977)79:11–21,J Biol Chem (1982)257:3347–51, andJ Biol Chem (1987)262:13–18). It is assumed that Gal, Glc, GlcNAc, GalNAc, and NeuAc are of thed-configuration, Fuc of thel-configuration, and all sugars present in the pyranose form.  相似文献

The dietary lectins, edible mushroom (ABL) and Jacalin (JAC) inhibit the proliferation of colonic cancer cells, whereas Amaranth (ACL) and peanut (PNA) stimulate their proliferation. All these lectins share as their preferred ligand the Thomsen-Friedenreich (TF) antigen galactosyl 1,3 N-Acetylgalactosamine (Gal1,3GalNAc), but differ in their finer specificities for modifications of this determinant and in their specificities for cancerous epithelia. We have investigated, using a resonant mirror biosensor, the kinetics of binding of these lectins, and Maclura pomifera lectin (MPL), which is similar to JAC, to two different Gal-GalNac bearing glycoproteins, antarctic fish antifreeze glycoprotein (AFG) and asialofetuin. JAC had the highest affinity for AFG [K d 0.027 M] due to a fast association rate constant [k ass 610,000 (Ms)–1]. The other lectins had considerably lower affinities, with K d ranging from 0.16 M (ABL) to 5.7 M (PNA), largely due to slower k ass [ABL 74,000 (Ms)–1 to PNA 2700 (Ms)–1]. Similarly, JAC had a much higher affinity for asialofetuin [K d 0.083 M] than the other lectins [K d 1.0 M–4.5 M]. Affinities were also calculated from the extent of binding at equlibrium and were generally similar to those calculated from the kinetic parameters indicating the true nature of these values.  相似文献

Legume lectins are considered to be antinutritional factors (ANF) in the animal feeding industry. Inactivation of ANF is an important element in processing of food. In our study on the stability ofPisum sativum L. lectin (PSL), a conserved hydrophobic amino acid (Val103) in a surface loop was replaced with alanine. The mutant lectin, PSL V103A, showed a decrease in unfolding temperature (T m ) by some 10 °C in comparison with wild-type (wt) PSL, and the denaturation energy (H) is only about 55% of that of wt PSL. Replacement of an adjacent amino acid (Phe104) with alanine did not result in a significant difference in stability in comparison with wt PSL. Both mutations did not change the sugarbinding properties of the lectin, as compared with wt PSL and with PSL from pea seeds, at ambient temperatures. The double mutant, PSL V103A/F104A, was produced inEscherichia coli, but could not be isolated in an active (i.e. sugar-binding) form. Interestingly, the mutation in PSL V103A reversibly affected sugar-binding at 37 °C, as judged from haemagglutination assays. These results open the possibility of production of lectins that are activein planta at ambient temperatures, but are inactive and possibly non-toxic at 37 °C in the intestines of mammals.  相似文献

Cytochrome c (cyt c) was reduced by a tyrosine-containing peptide, tyrosyltyrosylphenylalanine (TyrTyrPhe), at pH 6.0–8.0, while tyrosinol or tyrosyltyrosine (TyrTyr) could not reduce cyt c effectively under the same condition. Cyt c was reduced at high peptide concentration, whereas the reaction did not occur effectively at low concentration. The reaction rate varied with time owing to a decrease in the TyrTyrPhe concentration and the production of tyrosine derivatives during the reaction. The initial rate constants were 2.4×10–4 and 8.1×10–4 s–1 at pH 7.0 and 8.0, respectively, for the reaction with 1.0 mM TyrTyrPhe in 10 mM phosphate buffer at 15°C. The reciprocal initial rate constant (1/kint) increased linearly against the reciprocal peptide concentration and against the linear proton concentration, whereas logkint decreased linearly against the root of the ionic strength. These results show that deprotonated (TyrTyrPhe), presumably deprotonated at a tyrosine site, reduces cyt c by formation of an electrostatic complex. No significant difference in the reaction rate was observed between the reaction under nitrogen and oxygen atmospheres. From the matrix-assisted laser desorption ionization time-of-flight mass spectra of the reaction products, formation of a quinone and other tyrosine derivatives of the peptide was supported. These products should have been produced from a tyrosyl radical. We interpret the results that a cyt cox/(TyrTyrPhe)cyt cred/(TyrTyrPhe) equilibrium is formed, which is usually shifted to the left. This equilibrium may shift to the right by reaction of the produced tyrosyl radical with the tyrosine sites of unreacted TyrTyrPhe peptides.  相似文献

The X-ray crystal structure of lentil lectin in complex with -d-glucopyranose has been determined by molecular replacement and refined to anR-value of 0.20 at 3.0 Å resolution. The glucose interacts with the protein in a manner similar to that found in the mannose complexes of concanavalin A, pea lectin and isolectin I fromLathyrus ochrus. The complex is stabilized by a network of hydrogen bonds involving the carbohydrate oxygens O6, O4, O3 and O5. In addition, the -d-glucopyranose residue makes van der Waals contacts with the protein, involving the phenyl ring of Phe123. The overall structure of lentil lectin, at this resolution, does not differ significantly from the highly refined structures of the uncomplexed lectin.Molecular docking studies were performed with mannose and its 2-O and 3-O-m-nitro-benzyl derivatives to explain their high affinity binding. The interactions of the modelled mannose with lentil lectin agree well with those observed experimentally for the protein-carbohydrate complex. The highly flexible Me-2-O-(m-nitro-benzyl)--d-mannopyranoside and Me-3-O-(m-nitro-benzyl)--d-mannopyranoside become conformationally restricted upon binding to lentil lectin. For best orientations of the two substrates in the combining site, the loss of entropy is accompanied by the formation of a strong hydrogen bond between the nitro group and one amino acid, Gly97 and Asn125, respectively, along with the establishment of van der Waals interactions between the benzyl group and the aromatic amino acids Tyr100 and Trp128.RL and FC are joint first authors.  相似文献

Hemagglutinating proteins were isolated by affinity chromatography from seeds of each of five cultivars of soybeans (Clycine max (L.) Merr.) previously reported to lack detectable lectin (S.P. Pull et al., 1978; Science 200, 1277). Quantities were between 1,000 and 10,000 times less than that found in the seeds of the reference cultivar, Chippewa. The sensitivity of the hemagglutinating assay was 0.05 g ml-1. Hemagglutinating activity was demonstrated in affinity-purified fractions from bulk seeds and seeds from individual plants in two cultivars, 30–70% ammonium-sulfate-precipitable fractions of seeds from individual plants of all five cultivars, and in whole crude extracts of individual seeds from each cultivar. In all instances, hemagglutinating activity was inhibited by galactose, anti-soybean agglutinin (SBA), and lectin-binding polysaccharide produced by Rhizobium japonicum. Affinity-purified lectin from seeds of a single Columbia plant was labeled with fluorescein isothiocyanate (FITC) and observed by fluorescence microscopy to bind to R. japonicum cells with specificity, intensity and localization indistinguishable from FITC-SBA. Lectins from distinguishable from FITC-SBA. Morniga M is a jacalin-related and mannose-specific lectin isolated from the bark of the mulberry (Morus nigra). In order to understand the function and application of this novel lectin, the binding property of Morniga M was studied in detail using an enzyme-linked lectinosorbent assay and lectin-glycan inhibition assay with extended glycan/ligand collection. From the results, it was found that the di-, tri-, and oligomannosyl structural units of N-glycans such as those of the bovine 1-acid glycoprotein (gp) and lactoferrin were the most active gps, but not the O-glycans or polysaccharides including mannan from yeast.
