Large doses of bone morphogenetic protein 2 (BMP2) are used clinically to induce bone formation in challenging bone defects. However, complications after treatment include swelling, ectopic bone formation, and adjacent bone resorption. While BMP2 can be effective, it is important to characterize the mechanism of the deleterious effects to optimize its use. The aim of this study was to determine the effect of BMP2 on apoptosis in osteoblast lineage cells and to determine the role of the BMP inhibitor Noggin in this process. Human mesenchymal stem cells (MSCs), immature osteoblast‐like MG63 cells, and mature normal human osteoblasts (NHOst) were treated with BMP2. A model system of increased endogenous BMP signaling was created by silencing Noggin (shNOG‐MG63). Finally, the BMP pathway regulating apoptosis in NHOst was examined using BMP signaling inhibitors (5Z‐7‐oxozeaenol, dorsomorphin, H‐8). Apoptosis was characterized by caspase‐3, BAX/BCL2, p53, and DNA fragmentation. BMP2 induced apoptosis in a cell‐type dependent manner. While the effect was minor in MSCs, MG63 cells had modest increases and NHOst cells had robust increases apoptosis after BMP2 treatment. Apoptosis was significantly higher in shNOG‐MG63 than MG63 cells. 5Z‐7‐oxozeaenol and dorsomorphin eliminated the BMP2‐induced increase in DNA fragmentation in NHOst, suggesting roles for TAB/TAK1 and Smad signaling. These results indicate that the apoptotic effect of BMP2 is dependent on cell maturation state, inducing apoptosis in committed osteoblasts through Smad and TAB/TAK1 signaling, and is regulated by Noggin. 目的探讨香菇C91-3菌丝发酵蛋白(LFP91-3)对H22肿瘤细胞抑瘤作用及相关机制。方法应用不同剂量LFP91-3(50、100和150μg)对H22荷瘤小鼠腹水瘤模型和腋下实体瘤进行治疗,观察其生存期和体内抑瘤作用;并用MTT法(LFP91-3浓度5、10、15μg/mL)和流式细胞仪对经LFP作用的H22肿瘤细胞进行观察和检测。结果香菇C91-3菌丝发酵提取蛋白(LFP91-3)能延长H22荷瘤小鼠的生存期,对体外培养的H22有直接杀伤作用,抑瘤率出现对浓度和时间的依赖性。LFP91-3能将H22肿瘤细胞株细胞周期阻滞到S期并诱导出细胞凋亡。结论 LFP91-3在体内、外对H22肿瘤细胞有较好的抑瘤作用,主要是诱导其调亡。  相似文献

摘要 目的:探讨circ_0001461对骨肉瘤细胞增殖和凋亡的影响及调控机制。方法:采用实时荧光定量聚合酶反应(qRT-PCR)检测检测circ_0001461在骨肉瘤组织和细胞中的表达水平。在U2OS和HOS细胞中转染sh-NC和sh-circ_0001461后,采用CCK8检测细胞增殖情况,流式细胞术检测细胞凋亡情况,qRT-PCR检测增殖相关分子Ki-67 mRNA的表达水平,Western Blot检测凋亡相关分子Cleaved-caspase-3蛋白的表达水平。采用双荧光素酶报告基因检测circ_0001461和miR-30a-5p的结合情况。结果:circ_0001461在骨肉瘤组织中的表达水平明显高于癌旁正常组织(P<0.05),circ_0001461在骨肉瘤细胞U2OS和HOS中的表达水平均明显高于成骨细胞NHOst(P<0.05)。低表达circ_0001461能够抑制骨肉瘤细胞U2OS和HOS的增殖和增殖相关分子Ki-67的表达(P<0.05);促进骨肉瘤细胞U2OS和HOS的凋亡和凋亡相关分子Cleaved-caspase-3蛋白的表达(P<0.05)。双荧光素酶结果显示circ_0001461能够靶向结合miR-30a-5p。低表达circ_0001461能够促进miR-30a-5p的表达(P<0.05),circ_0001461和miR-30a-5p在骨肉瘤组织中的表达呈负相关(P<0.05)。在U2OS细胞中共转染sh-circ_0001461和miR-30a-5p mimics后能够进一步加强单独转染sh-circ_0001461对U2OS细胞增殖和凋亡的影响(P<0.05);在HOS细胞中共转染sh-circ_0001461和miR-30a-5p inhibitors后能够逆转单独转染sh-circ_0001461对U2OS细胞增殖和凋亡的影响(P>0.05)。结论:circ_0001461在骨肉瘤组织和细胞中明显高表达,低表达circ_0001461能够靶向促进miR-30a-5p的表达进而抑制骨肉瘤细胞增殖和促进细胞凋亡。  相似文献

The product of lipid peroxidation, 4-hydroxy-2-nonenal (HNE) is known to cause cell death at high concentrations, while at lower concentrations it can influence cell proliferation and differentiation. In our experiments we used human osteosarcoma cells (HOS), to test the influence of HNE on cell proliferation, differentiation and induction of apoptosis. Apoptosis induction was estimated by TiterTACS TUNEL test. The cells were in parallel counted and the DAPI staining method was used to distinguish between apoptotic and necrotic cells as well as to define the proportion of cells in mitosis. To test the influence of HNE on HOS cell differentiation, cells were treated every second day with HNE. After 10 days, the cells were stained for alkaline phosphatase, a marker for osteoblast differentiation. Cell growth inhibition was caused by supraphysiological concentrations of 10 or 100 microM HNE, while apoptosis was induced with supraphysiological as well as by the physiological amount of the aldehyde (1 microM). Necrosis appeared when cells were treated with 10 or 100 microM, but not with 1 microM HNE. The proportion of cells in mitosis gradually declined with increased HNE concentration. Multiple exposures of HOS cells to 10 microM HNE prevented HOS cell differentiation. These results indicated that HNE inhibits proliferation and differentiation of HOS cells in the same concentration dependent manner as it causes apoptosis. We thus assume that HNE might be one of the important signaling molecules regulating the growth of the human osteosarcoma cells.  相似文献

目的:通过康莱特联合顺铂对宫颈癌SiHa细胞增殖和凋亡的影响,探讨其作用机制。方法:体外培养宫颈癌Siha细胞,分别将康莱特(浓度为1,2,4,6,8 mg/mL),顺铂(浓度梯度为1.5,3,6,9,12μg/mL),单独作用于宫颈癌SiHa细胞,加药24h、48h用噻唑蓝(MTT法)检测细胞增殖情况。用流式细胞术检测康莱特组和顺铂组细胞24h凋亡率,选取合适的药物浓度(康莱特6 mg/mL,顺铂3μg/mL),进行联合用药,加药24h、48h用MTT法检测细胞增殖情况,用流式细胞术检测24h细胞凋亡率。结果:①MTT法显示加药后两组的24h、48h,宫颈癌SiHa细胞的抑制率均高于对照组(P0.05),并且在一定程度上呈浓度和时间依赖性。②联合用药时,细胞的抑制率和凋亡率要显著高于单独用药(P0.01)。结论:康莱特、顺铂单独或联合作用均能抑制SiHa细胞的增殖,促进其凋亡,且康莱特联合顺铂的作用要显著高于单独用药,康莱特与化疗药物联合使用可提高肿瘤细胞对化疗药物的敏感性。  相似文献

Cultured rodent osteoblastic cells reiterate the phenotypic differentiation and maturation of osteoblasts seen in vivo. As previously shown, the human osteosarcoma cell line HOS 58 represents a differentiated stage of osteoblast development. The potential of HOS 58 for still further in vitro differentiation suggests the line can serve as a model of osteoblast maturation. Using this cell line, we have investigated the influence of 1,25-(OH)2-D3 (D3), TGF-beta and Dexamethasone (Dex) on proliferation and on the protein and mRNA levels of alkaline phosphatase (AP), procollagen 1 (Col 1), and osteocalcin (Oc), as well as mineralization during 28 days in culture. AP mRNA and protein were highly expressed throughout the culture period with further increase of protein AP activity at constant gene expression levels. A differentiation inhibiting effect of either TGF-beta or Dex was seen. Col 1 was investigated without the use of ascorbic acid and showed only minor changes during culture time or stimulation. The gene expression for Oc increased continually whereas protein synthesis peaked at confluence and decreased thereafter. TGF-beta and Dex treatments decreased Oc mRNA and protein levels. Stimulation by D3 was maximal at day 7 with a decrease thereafter. HOS 58 cells showed no mineralization capacity when stimulated with different agents, as measured by energy-dispersive X-ray microanalysis. This was not due to absence of Cbfa1 expression. In conclusion, the HOS 58 osteosarcoma cell line represents a differentiated cell line with highly expressed and physiologically regulated AP expression during further differentiation in culture. We observed a dissociation between osteocalcin gene expression and protein secretion which may contribute to the lack of mineralization in this cell line.  相似文献

考察鹅不食草提取物对人高分化鼻咽癌细胞CNE-1增殖的抑制作用,初步探讨其抗鼻咽癌分子机制。95%乙醇提取鹅不食草全草,噻唑蓝(MTT)法观察不同浓度提取物在不同时间对人高分化鼻咽癌细胞CNE-1增殖的抑制作用,Hoechst 33258荧光染色观察CNE-1凋亡情况及形态学变化,Western Blot检测CNE-1细胞内抗凋亡蛋白Bcl-2和促凋亡蛋白Bax的表达。结果表明:鹅不食草醇提物能显著抑制CNE-1增殖(P<0.01),并呈现明显的时间-剂量依赖性,其中诱导48和72 h的IC50分别为30.0和25.0μg/mL。阳性对照药顺铂的IC50为0.79μg/mL。Hoechst 33258荧光染色观察发现给药组CNE-1细胞出现不同程度细胞核变圆变亮,核染色质浓缩,产生核小体等明显的凋亡特征。Bcl-2蛋白相对表达量随醇提物浓度的增大而下降,Bax蛋白相对表达量随醇提物浓度的增大而上升,二者与对照组相比均有显著性差异(P<0.01)。鹅不食草醇提物对体外人高分化鼻咽癌细胞CNE-1具有明显的增殖抑制和凋亡诱导作用,其分子作用机制可能与Bcl-2蛋白表达下调、Bax蛋白表达上调有关。  相似文献

为达到鼻咽癌(nasopharyngeal carcinoma,NeC)的靶向化疗,该研究通过酰胺化反应和配位偶联技术制备叶酸(folicacid,FA)分子靶向载川页铂(cisplatin,CDDP)羧甲基-β-环糊精(carboxymethyl-β-cyclodextrin,CM—β—CD)纳米复合物(FA-CM—β—CD—CDDP),采用邻苯二胺(o-phenylenediamine,OPDA)比色法检测复合物中CDDP含量,紫外分光光谱检测FA含量,透射电镜观察复合物形态,激光粒度仪测定复合物粒径大小。荧光显微镜观察NPC叶酸受体(folatereceptor,FR)阳性HNE-1细胞及FR阴性CNE-2细胞对偶联FITC的复合物的吞噬及OPDA比色法检测细胞内CDDP的浓度。通过MTT法、集落形成实验和流式细胞术检测复合物对HNE-1细胞增殖能力和凋亡的影响。研究结果显示,复合物中偶联的FA和CDDP浓度分别为340gg/mL和2mg/mL,CDDP包封率达20.00%,复合物粒径均匀且大小为157.8nm。HNE-1细胞内见较多FITC,细胞内CDDP浓度为6.24ng/mL,而CNE-2细胞内FITC较少,细胞内CDDP浓度仅约2.01ng/mL。HNE-1生长抑制率在24h明显高于对照组(CM—β—CD—CDDP),其IC50(4.80μg/mL)明显低于对照组(6.97μg/mL),但当所载的CDDP终浓度达到16.00μg/mL时,两组抑制率均达到80%以上;作用48h两组抑制率无明显差异。在24h,当复合物的CDDP终浓度为1.00μg/mL时,HNE—1的集落形成率为33.21%,明显低于对照组(52.27%)。当复合物的CDDP终浓度为0.25μg/mL和1.00μg/mL时,HNE-1的凋亡率分别达12.65%和22.35%,明显高于对照组(6.91%和14.21%)。研究结果表明,成功构建的FA—CM—β—CD.cDDP纳米复合物能够靶向抑制FR阳性的NPC细胞增殖并促进其凋亡。  相似文献

The direct effect of 1alpha,25(OH)(2)D(3) on osteoblasts remains unclear. In this study, we evaluated the in vitro effects of 1alpha,25(OH)(2)D(3) and its analogue, 2-methylene-19-nor-(20S)-1,25-dihydroxyvitamin D(3) (2MD), on osteoblasts from three different species, i.e. bone marrow stromal cells from the Sprague-Dawley (SD) rat, from the C57BL/6 mouse, as well as human osteoblast NHOst cells and human osteosarcoma derived MG-63 cells. We found that in rat cells, both compounds increased cell proliferation, inhibited cell apoptosis and increased alkaline phosphatase (ALP) activity. In mouse cells, however, both compounds initiated cell apoptosis and inhibited ALP activity. In human cells, although cell proliferation was inhibited by both compounds, cell apoptosis was inhibited and ALP activity was enhanced. In each species, 2MD was much more potent than 1alpha,25(OH)(2)D(3). To summarize, species differences should be taken into account in studies of vitamin D effects. However, in all tested species - rat, mouse and human - 2MD is considerably more potent in its effects on osteoblastic cells in vitro than 1alpha,25(OH)(2)D(3).  相似文献

【背景】虫生真菌是非常重要的自然资源,但被发现和利用的种类相对较少。【目的】鉴定从野外采集的4株虫生真菌,并探讨4株菌的醇提物对宫颈癌Hela细胞的抑制活性。【方法】结合形态学特征与rDNA ITS和β-Tubulin序列分析对4株虫生真菌进行种类鉴定,采用四甲基偶氮唑盐[3-(4,5-Dimethyl-2-Thiazolyl)-2,5-Diphenyl-2-H-Tetrazolium Bromide,MTT]比色法分析它们的菌丝醇提物(M-1、M-2、M-3、M-4)和发酵液醇提物(F-1、F-2、F-3、F-4)对Hela细胞增殖的抑制活性,并进一步利用细胞形态学观察和流式细胞术检测所筛选抑制作用最强的醇提取物的抑制活性。【结果】经鉴定,菌株IS和LS均为环链棒束孢(Isaria cateniannulata),菌株HLS和HLS2均为粉棒束孢(Isaria farinosa);MTT实验结果显示8种醇提物对Hela细胞增殖均有一定的抑制作用,而且菌丝醇提物和发酵液醇提物的抑制作用大小排序均为:粉棒束孢HLS>环链棒束孢LS>粉棒束孢HLS2>环链棒束孢IS,其中,粉棒束孢HLS的发酵液醇提物F-3的抑制活性最强(P<0.001),处理24h后其IC50值为0.6678 mg/mL,而其他7种醇提物的IC50值均大于1 mg/mL;细胞形态学观察显示F-3在处理时间24h时,对Hela细胞生长的抑制作用随着浓度的增加而加大,而对正常细胞BHK-21仅有轻微的毒性作用;流式细胞仪检测发现F-3可诱导Hela细胞凋亡,当处理浓度为1.0 mg/mL时,细胞总凋亡率可达到32.45%。【结论】4株菌对Hela细胞增殖的抑制作用存在着显著差异,这可能与4株菌的地理来源、寄主等因素有关;来源于粉棒束孢HLS株系的发酵液醇提物F-3对正常细胞BHK-21的增殖和生长无显著抑制作用,但对Hela癌细胞的增殖和生长抑制活性最强,而且具有诱导肿瘤细胞凋亡的作用,暗示其具有研究利用价值。  相似文献

探讨了桦褐孔菌提取物对胃癌MGC - 80 3细胞株的抗增殖、诱导凋亡作用及对凋亡相关基因表达的影响。MTT比色法结果显示 ,桦褐孔菌提取物在 0 .5~ 16 0 μg/mL范围内 (其IC50 为 4 μg/mL)对胃癌MGC- 80 3细胞株均有抑制作用 ,并表现出浓度依赖性关系 ;凋亡形态学观察结果 ,药物浓度 2 μg/mL ,作用时间 12~ 2 4h后 ,细胞核染色质固缩并凝结成块 ,聚集在核膜周边 ,凋亡小体形成 ;TUNEL法检测结果 ,不同浓度药物均诱导胃癌MGC - 80 3细胞株凋亡 ,细胞凋亡率随药物浓度增加而上升 ,显示明显的量效关系。Ki- 6 7抗原检测结果 ,不同浓度药物均抑制胃癌MGC - 80 3细胞株增殖 ,表现出浓度、时间依赖性关系 ;2μg/mL桦褐孔菌提取物作用胃癌MGC - 80 3细胞株 4 8h之后 ,明显下调Bcl - 2基因蛋白表达。因此 ,通过此项研究可得出 ,桦褐孔菌提取物对胃癌MGC - 80 3细胞株有抗增殖作用和诱导凋亡作用 ,其凋亡的分子生物学机制可能与下调凋亡抑制基因Bcl 2表达有一定关系。  相似文献

为研究橙盖鹅膏多糖的体外免疫调节活性、细胞毒性和抗肿瘤活性,本研究运用细胞学技术探究AC-1对T细胞、B细胞和RAW264.7三种免疫细胞的体外免疫调节活性;研究AC-1对小鼠成纤维细胞(L929)的细胞毒性以及对小鼠胃癌细胞(MFC)、小鼠肉瘤细胞(S180)的体外抗肿瘤活性。结果表明,AC-1能在体外显著刺激三种免疫细胞的增殖及RAW264.7细胞的吞噬,表明AC-1在增强机体免疫方面具有重要作用,进一步研究发现AC-1主要通过显著促进IgE和IgG的分泌来增强体液免疫。在浓度为5~20μg/mL时AC-1对L929细胞无显著的促进及抑制作用,说明AC-1对正常细胞无毒性;在浓度为10~20μg/mL时AC-1能显著抑制小鼠胃癌细胞(MFC)的生长,但对小鼠肉瘤细胞(S180)的抑制效果较弱,说明AC-1在体外能够直接抑制肿瘤细胞的生长,但对不同的肿瘤细胞具有不同的抑制效果。综上所述,橙盖鹅膏多糖(AC-1)在体外具有良好的免疫调节活性、抗肿瘤活性,但对不同的肿瘤细胞具有不同的抑制效果并且对正常细胞无毒性。  相似文献

目的考察胡桃楸提取液对肿瘤细胞Hela、K562的抑制作用和相关机制。方法用MTT方法分析胡桃楸提取液对Hela、K562细胞增殖的影响。采用端粒酶PCR ELISA试剂盒分析胡桃楸提取液对Hela、K562细胞端粒酶的影响。结果 Hela细胞24、48和72 h的LD50分别为406.18μg/mL、319.48μg/mL和112.84μg/mL。K562细胞24 h LD50为154.50μg/mL。HLF细胞LD50为918.69μg/mL。胡桃楸提取液可抑制Hela细胞和K562细胞的端粒酶活性,而对HLF细胞端粒酶活性影响不大。结论胡桃楸提取液对Hela细胞、K562细胞有抑制作用,在低浓度下对HLF细胞杀伤不大。对肿瘤细胞抑制作用可能与抑制端粒酶活性相关。  相似文献

The aim of this study was to observe the cytotoxicity and apoptotic effects of nickel oxide nanoparticles on human cervix epithelioid carcinoma cell line (HeLa). Nickel oxide precursors were synthesized by an nickel sulphate-excess urea reaction in boiling aqueous solution. The synthesized NiO nanoparticles (<200 nm) were investigated by X-ray diffraction analysis and transmission electron microscopy techniques. For cytotoxicity experiments, HeLa cells were incubated in 50-500 μg/mL NiO for 2, 6, 12 and 16 hours. The viable cells were counted with a haemacytometer using light microscopy. The cytotoxicity was observed low in 50-200 μg/mL concentration for 16 h, but high in 400-500 μg/mL concentration for 2-6 h. HeLa cells' cytoplasm membrane was lysed and detached from the well surface in 400 μg/mL concentration NiO nanoparticles. Double staining and M30 immunostaining were performed to quantify the number of apoptotic cells in culture on the basis of apoptotic cell nuclei scores. The apoptotic effect was observed 20% for 16 h incubation.  相似文献

目的 探讨牡蛎多糖体外抗乙型肝炎病毒(HBV)的作用.方法 采用HepG2.2.15细胞为体外细胞模型,给予不同浓度牡蛎多糖进行混合培养,作用9d后收集上清液,用ELISA法测定上清液中HBsAg、HBeAg的水平,观察药物对HepG2.2.15细胞分泌病毒杭原的影响,同时以MTT法检测药物在体外对HepG2.2.15细胞生长的抑制作用.结果 牡蛎多糖浓度在0.1~1 000 μg/mL时,对细胞无明显的毒性作用;牡蛎多糖可明显抑制HBsAg、HBeAg的分泌,其半数有效浓度( IC50)分别为362.2、558.6 μg/mL,治疗指数(TI)为>27.6和>17.9.结论 牡蛎多糖体外具有一定的抗HBV作用.  相似文献

The induction of apoptosis by azadirachtin, a well‐known botanical tetranortriterpenoid isolated from the neem tree (Azadirachta indica A. Juss) and other members of the Meliaceae, was investigated in Spodoptera frugiperda cultured cell line (Sf9). Morphological changes in Sf9 cells treated by various concentrations of azadirachtin were observed at different times under light microscopy. Morphological and biochemical analysis indicated that Sf9 cells treated by 1.5 μg/mL azadirachtin showed typical morphological changes, which were indicative of apoptosis and a clear DNA ladder. The flow cytometry analysis showed the apoptosis rate reached a maximum value of 32.66% at 24 h with 1.5 μg/mL azadirachtin in Sf9 cells. The inhibition of Sf9 cell proliferation suggested that the effect of azadirachtin was dose dependent and the EC50 at 48 and 72 h was 2.727 × 10−6 and 6.348 × 10−9 μg/mL, respectively. The treatment of azadirachtin in Sf9 cells could significantly increase the activity of Sf caspase‐1, but showed no effect on the activity of Topo I, suggesting that the apoptosis induced by azadirachtinin Sf9 cells is through caspase‐dependent pathway. These results provided not only a series of morphological, biochemical, and toxicological comprehensive evidences for induction of apoptosis by azadirachtin, but also a reference model for screening insect cell apoptosis inducers from natural compounds.  相似文献

High dose glucocorticoid (GC) treatment induces osteoporosis partly via increasing osteoblast apoptosis. However, the mechanism of GC-induced apoptosis has not been fully elucidated. Osteoblast-derived tissue inhibitor of metalloproteinase-1 (TIMP-1) was recently reported to be involved in bone metabolism. Our previous study demonstrated that TIMP-1 suppressed apoptosis of the mouse bone marrow stromal cell line MBA-1 (pre-osteoblast) induced by serum deprivation. Therefore, we tested the effect of the GC dexamethasone (Dex) on TIMP-1 production in murine osteoblastic MC3T3-E1 cells and further determined whether this action is associated with Dex-induced osteoblast apoptosis. Dex decreased TIMP-1 production in MC3T3-E1 cells, and this effect was blocked by the glucocorticoid receptor (GR) antagonists, RU486 and RU40555. Recombinant TIMP-1 protein reduced caspase-3 activation and apoptosis induced by Dex in MC3T3-E1 cells. In addition, the pro-apoptotic effect of the Dex was augmented by suppression of TIMP-1 with siRNA. Furthermore, mutant TIMP-1, which has no inhibitory effects on MMPs, yet protects MC3T3-E1 cells against Dex-induced apoptosis. Our study demonstrates that Dex suppresses TIMP-1 production in osteoblasts through GR, and this effect is associated with its induction of osteoblast apoptosis. The anti-apoptotic action of TIMP-1 is independent of its inhibitory effects on MMPs activities. The decrease in TIMP-1 production caused by Dex may contribute to the mechanisms of Dex-induced bone loss.  相似文献

紫芝酸性三萜类化合物体外抑癌和抑菌作用的研究   总被引:2,自引:0,他引:2  
采用噻唑蓝比色(MTT)法研究紫芝酸性三萜对几种癌细胞体外增殖的影响,并用管碟法检测了紫芝胞内酸性三萜对几种细菌和霉菌的体外抑菌作用。结果表明,紫芝胞内酸性三萜和胞外酸性三萜在250μg/mL时,对人肝癌细胞BEL7402和人乳腺癌细胞MCF-7均有显著抑制作用(P<0.05),但对人胃癌细胞SGC-7901没有显著抑制作用(P>0.05)。BEL7402细胞的生长曲线试验表明,胞内酸性三萜组的细胞受到显著抑制,未出现指数增长期,且BEL7402细胞培养3d后,对照组细胞数目多、均匀,而胞内酸性三萜组的细胞数目明显减少,且细胞变小。抑菌试验结果表明,胞内酸性三萜在40mg/mL时,对大肠杆菌Escherichia coli和金黄色葡萄球菌Staphylococcus aureus的生长均具有显著抑制作用(P<0.01),对枯草芽孢杆菌Bacillus subtitis和青霉的Penicillium chrysogenum抑制作用较弱;而在此浓度下对黑曲霉Aspergillus niger没有抑制作用。该样品对大肠杆菌、金黄色葡萄球菌、枯草芽孢杆菌、黑曲霉和青霉的MIC分别为20mg/mL、20mg/mL、40mg/mL、80mg/mL和40mg/mL。此外,该酸性三萜的抑菌成分在60℃下(处理2h)较稳定,但在80℃以上,热稳定性较差,活性降低。  相似文献

目的:研究淫羊藿苷体外对致宫颈癌TC-1细胞的增殖抑制及促凋亡作用。方法:利用细胞培养,用不同浓度的淫羊藿苷在一定的时间处理致宫颈癌TC-1细胞,光学显微镜直接观察药物对细胞的作用;MTT法检测淫羊藿苷对TC-1细胞的增殖抑制作用;Dapi核染色、Annexinv-FITC/PI流式细胞学检测细胞凋亡。结果:淫羊藿苷对TC-1细胞有显著的抑制作用,且呈时间、剂量依赖,20μg/ml作用72小时后,细胞抑制率达99%;DAPI核染色和流式细胞术检测可发现典型细胞凋亡特征。结论:淫羊藿苷对TC-1细胞增殖有抑制和促凋亡作用,并呈时间浓度依赖性。  相似文献
