A natural feature of freshly harvested Avena fatua L. caryopses is primary dormancy which, however, was relieved partially by putrescine (Put) (10?2 M) and completely by karrikinolide (KAR1) (3 × 10?9 M) or gibberellic acid (GA3) (10?5 M). The sensitivity of A. fatua caryopses to these stimulators was adversely affected by supraoptimal temperature (SOT) (30 °C). A reduced germinability of caryopses due to high temperature even after transferring them to lower temperatures (10 or 20 °C) indicated the induction of thermodormancy. The maintenance of relatively constant levels of abscisic acid (ABA) in embryos but not surrounding tissues during SOT treatment was observed. The application of Put either during the SOT treatment or afterwards counteracted the effects of high temperature but had no significant impact on ABA content. The action of exogenous Put in alleviating the loss of responsiveness to KAR1 and GA3 imposed by SOT treatment in A. fatua PD caryopses is discussed in reference to the interconnection between ABA and GA metabolism and signaling pathways.  相似文献

Backgound

Evolution of cancer cells is characterized by large scale and rapid changes in the chromosomal  landscape. The fluorescence in situ hybridization (FISH) technique provides a way to measure the copy numbers of preselected genes in a group of cells and has been found to be a reliable source of data to model the evolution of tumor cells. Chowdhury et al. (Bioinformatics 29(13):189–98, 23; PLoS Comput Biol 10(7):1003740, 24) recently develop a computational model for tumor progression driven by gains and losses in cell count patterns obtained by FISH probes. Their model aims to find the rectilinear Steiner minimum tree (RSMT) (Chowdhury et al. in Bioinformatics 29(13):189–98, 23) and the duplication Steiner minimum tree (DSMT) (Chowdhury et al. in PLoS Comput Biol 10(7):1003740, 24) that describe the progression of FISH cell count patterns over its branches in a parsimonious manner. Both the RSMT and DSMT problems are NP-hard and heuristics are required to solve the problems efficiently.

Methods

In this paper we propose two approaches to solve the RSMT problem, one inspired by iterative methods to address the “small phylogeny” problem (Sankoff et al. in J Mol Evol 7(2):133–49, 27; Blanchette et al. in Genome Inform 8:25–34, 28), and the other based on maximum parsimony phylogeny inference. We further show how to extend these heuristics to obtain solutions to the DSMT problem, that models large scale duplication events.

Results

Experimental results from both simulated and real tumor data show that our methods outperform previous heuristics (Chowdhury et al. in Bioinformatics 29(13):189–98, 23; Chowdhury et al. in PLoS Comput Biol 10(7):1003740, 24) in obtaining solutions to both RSMT and DSMT problems.

Conclusion

The methods introduced here are able to provide more parsimony phylogenies compared to earlier ones which are consider better choices.

Paster (2014) has argued that grammar does not distinguish between weak and strong suppletion. This claim is countered in this paper, in an analysis of allomorphic effects in several Semitic languages. What has been traditionally called “root” is broken down into three levels: the formless index (Harley 2014), the phonological index (Borer 2005, 2009, 2013) and its underlying representation. The distinction between the first two leads to a principled structural difference between weak and strong suppletion. Several test cases are presented that require a distinction between the latter two. Two general conclusions are drawn: (1) allomorphy at the different levels has different functions, and (2) in the Semitic root-and-pattern system, weakly suppletive allomorphy never targets the root as a whole; there is no root allomorphy in Semitic.  相似文献

Within the fossil collection from the Sandelzhausen Lagerstätte in the Upper Freshwater Molasse near Mainburg, Germany, are remains of the schizotheriine chalicothere Metaschizotherium bavaricum, von Koenigswald, 1932. This new material includes elements from a large part of the body, and allows the dentition and postcranial skeleton of Metaschizotherium to be described in detail for the first time. At approximately 16 Ma (MN5), M. bavaricum is now the best-known Early and Middle Miocene European schizotheriine and is important for comparative studies. It differs to some degree from earlier Miocene (MN2–MN4) European material attributed to Moropus sp. or Metaschizotherium wetzleri (Kowalewsky, 1873) and to a larger degree from the Late Miocene species Ancylotherium pentelicum (Gaudry and Lartet, 1856). At Sandelzhausen, M. bavaricum apparently lived in a moist forested environment, where it probably fed on leaves, fruit, and seeds. Members of the Chalicotheriinae, such as Anisodon and Chalicotherium, are not found at Sandelzhausen and may not have been present in Europe at this time. M. bavaricum, like other Schizotheriinae, did not have the bizarre gorilla-like proportions of the Chalicotheriinae. Instead, its general body proportions resemble those of contemporary schizotheriine chalicotheres on other continents, for example, Moropus from North America. M. bavaricum is slightly smaller than the type species of Metaschizotherium, M. fraasi von Koenigswald, 1932 (MN6–MN7) and differs from it in small ways that are still being explored as variation within and differences between these species are clarified. The schizotheriine chalicothere from La Grive St.-Alban (France) referred to M. fraasi by von Koenigswald (Palaeontographica, Beitrage zur Naturgeschichte der Vorzeit 8:1–24, 1932) and Viret (Nouvelles Archives Musée d’Histoire Naturelle de Lyon 6:53–81, 1961) should be restudied and referred to a different taxon.  相似文献

Theoretical chemistry calculations using the Density Functional Theory (DFT) were carried out to understand the interaction between oxygen (O2) and MnN4 type manganese-based complexes during the formation of MnN4-O2 adducts. In order to understand how this interaction is affected by different macrocyclic ligands, O2 was bonded to manganese-porphyrin (MnP), manganese-octamethylporphyrin (MnOMP), manganese-tetraaza[14]annulene (MnTAA), manganese-dibenzo [b,i] [1, 4, 8, 11]-tetraaza [14] annulene (MnDBTAA), manganese-2,3,9,10-tetramethyl-1,4,8,11-tetraazacyclotetradeca-1,3,8,10-tetraene ([(tim)Mn]2+), and manganese-2,3,9,10-tetraphenyl-1,4,8,11-tetraazacyclotetradeca-1,3,8,10-tetraene ([(ph-tim)Mn]2+). The binding and activation of the oxygen molecule was facilitated by an increasing trend in the O-O bond lengths and a decreasing one in the O-O vibrational frequency, with preference for the O2 side-on interaction among MnN4 macrocycles. The catalytic activities of the MnN4 complexes toward the O2 binding process increased in the following order: [(ph-tim)Mn]2+?<?MnP?<?MnOMP?<?MnDBTAA?<?MnTAA?<?[(tim)Mn]2+. Therefore, it was concluded that the [(tim)Mn]2+complex was the most active for the binding and activation of molecular oxygen.  相似文献

Purpose

The purpose of this study was to test the chain-organization environmental footprint (chain-OEF) approach by applying it to part of a pork production chain in Belgium. The approach is supposed to provide insight into the environmental impact of a specific production chain in an efficient manner by applying pragmatic data collection throughout the chain. This is achieved by allocating the environmental impact of each of the production sites to the product of interest using straightforward allocation rules.

Methods

The cradle-to-gate (up to retail) environmental impact of pork was determined by life cycle assessment (LCA), in line with the product and organisation environmental footprint guidelines (PEF and OEF; European Commission 2013b). Foreground data was gathered at a feed production site, two farmers, a slaughterhouse and a meat processing site. All foreground operations are part of the same pork production chain in Belgium. The chain was completed using background data from Ecoinvent v3.01 (Wernet et al. 2016), Agri-Footprint v1.0 (Blonk 2014), European Life Cycle Database v3.0, LCA Food Database (Nielsen et al. 2003) and OEF Sector Rules Retail (Humbert et al. 2015b). The impact was quantified using the international reference life cycle data system (ILCD) midpoint method for 14 impact categories, but focussing on climate change.

Results and discussion

The total carbon footprint of the cradle-to-gate pork production system equals 0.46 kg CO2-eq. (100 g pork)?1. This result is quite similar to that of earlier studies analysing the pork production chain: 0.58 and 0.57 kg CO2-eq. (100 g pork)?1 (Bracquené et al. 2011, Agri-Footprint 2014). Most of the carbon footprint was caused by feed production and more specifically, by the feed ingredients and their transport. Grains, soy and palm oil have the largest impact contributions. The farms are responsible for most of the remaining impact. N2O and CH4 emissions are the largest cause of greenhouse gas emissions at the farms. Also, in the other 13 considered impact categories, feed production and farming are responsible for more than half of the total impact, mostly followed by meat processing.

Conclusions

Applying the chain-OEF approach in this study has shown that a chain LCA can be performed successfully and pragmatic data collection allows obtaining LCA results relatively fast, especially for small or medium-sized enterprises (SMEs). Whereas data availability was not such an issue, the main bottlenecks identified are data management and the link of LCA to other disciplines such as engineering, policy, etc. which could increase the added value of LCA studies.

Support and management staff in elite sport experience work-related stress and emotional disturbance to a similar extent as athletes (Fletcher and Wagstaff 2009). The resonant frequency breathing technique (Lehrer et al. 2000) can inhibit autonomic changes associated with stressful situations or events and as such provides a potential emotional regulation tool. The present study utilised five practitioner-led heart rate variability (HRV) biofeedback sessions and home practice via mobile applications to train support and management staff (n = 9) in resonant frequency breathing techniques. Although baseline HRV did not change from pre to post training, participants increased total HRV (i.e., SDNN; p = .006), parasympathetic HRV (i.e., RMSSD; p = .028) and HRV reflective of baroreflex function (i.e., low frequency power; p = .018) while accurately performing resonant frequency breathing without a breath pacer. Post-intervention questionnaire data revealed an increase (p = .032) in habitual use of somatic strategies for emotional regulation, and social validation data suggested that the technique enhanced emotional regulation at home, work and during international competition. HRV biofeedback and the resonant frequency technique provided an on-demand emotional regulation technique for elite sport support and management staff.  相似文献

This paper analyzes two patterns of number marking in the DP in Nez Perce (Sahaptian) within the framework of Distributed Morphology. The first involves under-realization of plural on nouns. Number has classically been understood as a feature inherent to nouns, rather than to adjectives that modify them. In Nez Perce, however, only a small set of nouns show number morphology, whereas number morphology is highly productive on adjectival modifiers. Adjectives in fact may realize the plural more than once per word—an instance of multiple exponence. I show that the puzzle of under-realization for nouns can be solved through conditioned allomorphy, providing new evidence for the presence of gender features on n (Lowenstamm 2007; Kramer 2014). The puzzle of multiple exponence (over-realization) for adjectives can be solved through Local Dislocation (Embick and Noyer 2001) combined with very late insertion of reduplicative content (Haugen 2008, 2011), demonstrating one way the DM architecture may produce multiple exponence without recourse to dedicated mechanisms.  相似文献

In this study, aflatoxin B1 (AFB1) toxicity toward the earthworm Eisenia fetida (Savigny 1826) was evaluated in contact paper test systems containing distilled water and ethanol or 20 to 400 μg/ml of AFB1 over 72 h of exposure. The results indicated that AFB1 could induce significant damage to earthworms (coiling, curling, excessive mucus secretion, clitellum swelling) at greater than 75 μg/ml. Moreover, AFB1 had harmful effects on E. fetida (degenerative changes such as bulging of the clitella regions) at levels higher than 150 μg/ml. The calculated LD50 was 168.5 μg/ml. These findings confirm that E. fetida and standardized methods based on this organism (OECD 207 1984) are applicable and useful in mycotoxin related toxicity studies.  相似文献

A barley (Hordeum vulgare L.) mutant (cool) with leaf transpiration unaffected by the application of 1 mM abscisic acid (ABA) was isolated from the population of M2 seedlings using thermography (electronic visualization, and quantitation of the temperature profiles on the surface of the leaves). Stomata of the mutant plants were insensitive to exogenously applied ABA, darkness, and such desiccation treatments as leaf excision and drought stress. The evaporative cooling of the leaves of the cool barley was always higher than that of the wild-type barley, even without ABA application, indicating that the diffusive resistance of the mutant leaves to water loss was always lower. Guard-cell morphology and stomatal density as well as ABA level and metabolism were seemingly unaltered in the mutant plants. Most psycholinguistic models of lexical processing assume that the comprehension and production of inflected forms is mediated by morphemic constituents. Several more recent studies, however, have challenged this assumption by providing empirical evidence that information about individual inflected forms and their paradigmatic relations is available in long-term memory (Baayen et al. 1997; Milin et al. 2009a, 2009b). Here, we investigate how whole-word frequency, inflectional paradigm size and morphological family size affect production latencies and articulation durations when subjects are asked to read aloud isolated Estonian case-inflected nouns. In Experiment 1, we observed that words with a larger morphological family elicited shorter speech onset latencies, and that forms with higher whole-word frequency had shorter acoustic durations. Experiment 2, for which we increased statistical power by using 2,800 words, revealed that higher whole-word frequency, inflectional paradigm size, and morphological family size reduced both speech onset times and acoustic durations. These results extend our knowledge of morphological processing in three ways. First, whole-word frequency effects of inflected forms in morphologically rich languages are not restricted to a small number of very high-frequency forms, contrary to previous claims (Niemi et al. 1994; Hankamer 1989; Yang 2016). Second, we replicated the morphological family size effect in a new domain, the acoustic durations of inflected forms. Third, we showed that a novel paradigmatic measure, inflectional paradigm size, predicts word naming latencies and acoustic durations. These results fit well with Word-and-Paradigm morphology (Blevins 2016) and argue against strictly (de)compositional models of lexical processing.  相似文献

The discovery of the TROSY effect (Pervushin et al. in Proc Natl Acad Sci USA 94:12366–12371, 1997) for reducing transverse relaxation and line sharpening through selecting pathways in which dipole–dipole and CSA Hamiltonians partially cancel each other had a tremendous impact on solution NMR studies of macromolecules. Together with the methyl TROSY (Tugarinov and Kay in J Biomol NMR 28:165–172, 2004) it enabled structural and functional studies of significantly larger systems. The optimal field strengths for TROSY have been estimated to be on spectrometers operating around 900 MHz (21.14 T) for the 1HN TROSY (Pervushin et al. in Proc Natl Acad Sci USA 94:12366–12371, 1997) while the aromatic 13C (13Caro) TROSY is posited to be optimal at around 600 MHz (14.09 T) (Pervushin et al. in J Am Chem Soc 120:6394–6400, 1998b; Pervushin in Q Rev Biophys 33:161–197, 2000). The initial rational was based on the consideration of where the quadratic B0 field dependences of the TROSY relaxation rates reach a minimum. For sensitivity consideration, however, it is interesting to estimate which field strengths yield the tallest peaks. Recent studies of 15N-detected TROSYs suggested that maximal peak heights are expected at 1.15 GHz (27.01 T) although the slowest relaxation rates or longest transverse relaxation times T2 are indeed expected around 900 MHz (21.14 T) (Takeuchi in J Biomol NMR 63:323–331, 2015; Takeuchi et al. in J Biomol NMR 64:143–151, 2016). This was based on the fact that the heights of Lorentzian lines are proportional to B o 3/2 * T2 (Bo). Thus, multiplying the parabolic T2(Bo) dependence with the increasing function of B o 3/2 shifts the maxima of peak-height field dependence from the T2 maximum at 900 MHz to higher fields. Moreover, besides shifting the peak height maximum for 15N TROSY, this analysis yields estimates for optimal peak heights for 1HN detected TROSY to 1.5 GHz, and to 900 MHz for 13C-detected 13CaroTROSY as is detailed below. To our knowledge, this aspect of field dependence of TROSY sensitivity has not been in the attention of the NMR community but may affect perspectives of NMR at ultra-high fields.  相似文献

Two western Pacific triplefins, Enneapterygius fuscoventer Fricke 1997 and E. howensis Fricke 1997 (Perciformes: Tripterygiidae), are similar to each other in sharing 15–19 (usually 17) notched lateral-line scales and the mandibular pore formula 3–5 + 1 + 3–5 (usually 4 + 1 + 4), in addition to similar coloration, viz. body with four vertical bands, the first and second forked ventrally, dorsal-fin membrane semi-transparent, anal fin entirely blackish, and caudal fin blackish with a semi-transparent margin. These species have previously been known only from preserved specimens. Examination of additional specimens plus color photographs of males and females of both species when fresh, and comparisons with type specimens resulted in several features, including coloration and counts of second dorsal-fin spines, anal-fin soft rays, pored lateral-line scales and longitudinal scale rows, being regarded as new diagnostic characters. Enneapterygius fuscoventer and E. howensis have been newly recorded from southern Japan and coastal eastern Australia, respectively.  相似文献

Introduction

Despite much work over past decades, whether antigen-specific immune reactions occur in rheumatoid arthritis (RA) and to what extent such reactions are directed towards joint-specific autoantigens is still questionable. One strong indicator for antigenic involvement in RA is the fact that certain major histocompatibility complex (MHC) class II genotypes [human leucocyte antigen (HLA)-DR4 and HLA-DR1] predispose for the development of the disease [1]. In the present report, collagen type II (CII) was studied as a putative autoantigen on the basis of both clinical and experimental data that show an increased frequency of antibodies to CII in RA patients [2,3,4] and that show that CII can induce experimental arthritis [5].It is evident from the literature that RA peripheral blood mononuclear cells (PBMCs) respond poorly to antigenic stimulation [6,7,8], and in particular evidence for a partial tolerization to CII has been presented [9]. The strategy of the present work has accordingly been to reinvestigate T-cell reactivity to CII in RA patients, to relate it to the response to commonly used recall antigens and to analyze IFN-γ responses as an alternative to proliferative responses.  相似文献

Introduction

Mass spectrometry imaging (MSI) is a technology that enables the visualization of the spatial distribution of hundreds to thousands of metabolites in the same tissue section simultaneously. Roots are below-ground plant organs that anchor plants to the soil, take up water and nutrients, and sense and respond to external stresses. Physiological responses to salinity are multifaceted and have predominantly been studied using whole plant tissues that cannot resolve plant salinity responses spatially.

Objectives

This study aimed to use a comprehensive approach to study the spatial distribution and profiles of metabolites, and to quantify the changes in the elemental content in young developing barley seminal roots before and after salinity stress.

Methods

Here, we used a combination of liquid chromatography–mass spectrometry (LC–MS), inductively coupled plasma mass spectrometry (ICP–MS), and matrix-assisted laser desorption/ionization (MALDI–MSI) platforms to profile and analyze the spatial distribution of ions, metabolites and lipids across three anatomically different barley root zones before and after a short-term salinity stress (150 mM NaCl).

Results

We localized, visualized and discriminated compounds in fine detail along longitudinal root sections and compared ion, metabolite, and lipid composition before and after salt stress. Large changes in the phosphatidylcholine (PC) profiles were observed as a response to salt stress with PC 34:n showing an overall reduction in salt treated roots. ICP–MS analysis quantified changes in the elemental content of roots with increases of Na+ and decreases of K+ content.

Conclusion

Our results established the suitability of combining three mass spectrometry platforms to analyze and map ionic and metabolic responses to salinity stress in plant roots and to elucidate tolerance mechanisms in response to abiotic stress, such as salinity stress.

Background & Aims

The effects of an alfalfa plant (Medicago sativa L.) hydrolysate-based biostimulant (EM) containing triacontanol (TRIA) and indole-3-acetic acid (IAA) were tested in salt-stressed maize plants.

Methods

Plants were grown for 2 weeks in the absence of NaCl or in the presence (25, 75 and 150 mM). On the 12th day, plants were supplied for 48 h with 1.0 mg L?1 EM or 11.2 μM TRIA.

Results

EM and TRIA stimulated the growth and nitrogen assimilation of control plants to a similar degree, while NaCl reduced plant growth, SPAD index and protein content. EM or TRIA increased plant biomass under salinity conditions. Furthermore, EM induced the activity of enzymes functioning in nitrogen metabolism. The activity of antioxidant enzymes and the synthesis of phenolics were induced by salinity, but decreased after EM treatment. The enhancement of phenylalanine ammonia-lyase (PAL) activity and gene expression by EM was consistent with the increase of flavonoids.

Conclusion

The present study proves that the EM increases plant biomass even when plants are grown under salinity conditions. This was likely because EM stimulated plant nitrogen metabolism and antioxidant systems. Therefore, EM may be proposed as bioactive product in agriculture to help plants overcome stress situations.  相似文献

Multiple investigators have reported thepresence of defects in the immune response of the elderly [Castle In: Clin Infect Dis 31:578, 2000; Ortqvist et al. In: Eur Respir J 30:414–422, 2007; Saurwein-Teissl et al. In: J Immunol 168:5893, 2002; Haynes et al. In: Proc Natl Acad Sci USA 100:15053–15058, 2003]. These defects reduce the magnitude of the immune response to infection and to vaccination. In individuals greater than 55 years of age, the probability of developing a fully protective neutralizing antibody response to the yearly multivalent particle inactivated influenza vaccine is less than 20% [Jefferson et al. In: Lancet 264:1165–1174, 2005; Goodwin et al. In: Vaccine 24:1159–1169, 2006; Jackson et al. In: Lancet 372:398–405, 2008; Simonsen and Taylor In: Lancet 7:658–666, 2007]. The defects in the aged immune system that are responsible for this limited response to vaccination in the older age groups include functional defects of the antigen presenting cells, functional defects in CD4 helper CD4 T cells and monocytes, and an altered microenvironment [Eaton et al. In: J Exp Med 200:1613–1622, 2004; Dong et al. In: J Gen Virol 84:1623–1628, 2003; Deng et al. In: Immunology 172:3437–3446, 2004; Cella et al. In: J Exp Med 184:747–752, 1996]. Starting at puberty, the involution of the thymus and the consequent reduction of the export of naïve T cells specific to neo-antigens leads to the reduction of the ratio of antigen naïve to memory cells as chronological age advances [Prelog In: AutoimmunRev 5:136–139, 2006; McElhaney et al. In: J Immunology 176:6333–6339, 2006]. Changes in glycosylation of T cells and target antigens acquired during the aging process and the antibodies to these new glycopeptides and glycoproteins may also contribute to a reduction in the functioning of the adaptive immune response [Ishii et al. In: J Clin Neurosci 14:110–115, 2007; Shirai et al. In: Clin Exp Immunol 12:455–464, 1972; Adkins and Riley In: Mech AgeingDev 103:147–164, 1998; Ben-Yehuda and Weksler In: Cancer Investigation 10:525–531, 1992]. One of the more interesting examples of the functional defects in the cells of the adaptive immune response is a reduced level of expression in the surface cytoadhesion and activation receptor molecules on CD4 helper T cells undergoing activation during vaccination. Upon infection or vaccination, CD40L is typically increased on the surface of CD4 helper T cells during activation, and this increased expression is absolutely essential to the CD40L promotion of expansion of antigen-specific B cells and CD 8 effector T cells in response to infection or vaccination [Singh et al. In: Protein Sci 7:1124–1135, 1998; Grewal and Flavell In: Immunol Res 16: 59–70, 1997; Kornbluth In: J Hematother Stem Cell Res 11:787–801, 2002; Garcia de Vinuesa et al. In: Eur J Immunol 29:3216–3224, 1999]. In aged human beings and mice, the reduced levels of expression of CD40 ligand (CD40L) in activated CD4 helper T cells is dramatically reduced [Eaton et al. In: J Exp Med 200:1613–1622, 2004; Dong et al. In: J Gen Virol 84:1623–1628, 2003]. To circumvent the reduction in CD40L expression and the subsequent reduction in immune response in the elderly, we have developed a chimeric vaccine comprised of the CD40L linked to the target antigen, in a replication incompetent adenoviral vector and in booster protein. The Coburg Sandstone member of the Hassberge Formation, Middle Keuper, Carnian, Upper Triassic, in the Germanic Basin of Central Europe contains spinicaudatan branchiopods which considerably broaden the known spectrum and provide important data on the phylogenetic history and intercontinental correlation of this systematic group. The spinicaudatan fauna for this lithostratigraphic unit is described in detail and includes Euestheria kozuri sp. nov., E.? sp. nov. Additional discussions deal with Euestheria multicostata (Geyer, 1987), E. winterpockensis (Bock, 1953a), Laxitextella laxitexta (Sandberger in Jones, 1890), Laxitextella multireticulata (Reible, 1962), and “Laxitextella? hausmanni (Schmidt, 1938),” as well as problems with (sub)global zonations that are based on spinicaudatans and suggested to be of Carnian and Norian age.  相似文献
