The effect of dietary fibres on constituents of complex carbohydrates in various tissues of streptozotocin induced diabetic rats is presented by analysing different constituents of complex carbohydrates in presence and absence of dietary fibre. Wheat bran was effective in preventing the decrease (14%) in total sugars in spleen and an increase in total sugars in stomach (33%) during diabetes. Decrease in uronic acid content during diabetes in spleen was prevented to the extent of 25% by the presence of wheat bran in the diet. The other parameters which were affected by the presence of wheat bran in the diet during diabetes are amino sugar (brain and stomach), sulphates (liver) and protein (lungs and stomach). Guar gum was effective in preventing the decrease in total sugar content in spleen by 28% and sulphate content in liver by 14% during diabetes. Variation in protein content in lungs was observed in diabetes. The results indicated beneficial role of dietary fibres like wheat bran and guar gum on complex carbohydrates to varying extents in different tissues.  相似文献

The adsorption of mutagens by some dietary fibres has been suggested as one mechanism by which dietary fibres protect against colorectal cancer. It is thought that these dietary fibres carry the mutagen out of the digestive tract, decreasing the effective mutagen concentration to which epithelial cells are exposed. The ability of gastrointestinal mucin to alter the extent to which the hydrophobic mutagen 1,8-dinitropyrene (DNP) adsorbs in vitro onto the insoluble dietary fibre alpha-cellulose, was investigated. It was found that crude and purified human ileal mucins themselves adsorbed DNP and decreased the adsorption of DNP onto alpha-cellulose. Purified mucin which had been treated with trypsin also adsorbed DNP. These studies suggest that in the digestive tract there would be competition for the adsorption of DNP between mucin and insoluble dietary fibres, such as alpha-cellulose. This factor must be considered in predictions about the distribution of hydrophobic, mutagenic carcinogens in the digestive tract and their role in the etiology of colorectal cancer.  相似文献

As in many other countries, the New Zealand Cancer Society produces guidelines for cancer prevention. These recommend avoiding asbestos, smoking, sunlight, alcohol, fatty food and obesity. Women are advised to have a regular cervical smear test. Additional 'probably helpful' suggestions include eating plenty of fresh fruit and vegetables and dietary fibre. However, considerable data from animal studies and more slowly accumulating data from human intervention studies suggest additional and more specific advice may be appropriate. Fruit and vegetable servings should total a minimum of five each day. Some specific fruits and vegetables (e.g., tomato, broccoli, onions) may have particular benefits against individual cancer types. Positive human evidence on potential benefits of increasing dietary fibre comes from studies where wheat bran was added to the diet. This is not a dietary fibre per se, but merely a good fibre source. Indeed, our own studies suggest that it could be various phytochemicals in the bran, rather than dietary fibre, which is beneficial. An increase either in whole wheat or wheat bran, rather than fibre, would be a sounder recommendation. Although there is some evidence that multivitamin supplementation can protect against cancer, this may be only in the special situation where the population is already significantly vitamin-deficient. For example, a combination of beta-carotene, vitamin E and selenium significantly reduced cancer mortality in a Chinese population, whereas lung cancer risks (in already high risk groups) were increased in Finnish and American trials with high dose beta-carotene. Various other chemopreventive drugs are being actively developed and at various stages in clinical trials. The enhanced cancer incidence in the beta-carotene trial illustrates the potential benefit of utilising surrogate endpoints of malignant disease rather than incident cancer as a trial endpoint.  相似文献

The effect of three types of fibre, guar, carboxymethyl cellulose (CMC) and wheat bran (WB) upon carbohydrate tolerance in rats has been investigated. Rats fed guar and CMC gained less weight over the eight week test period; however, both of these groups exhibited significantly lower postprandial plasma glucose and insulin. A residual fibre effect was seen postprandially after these animals were maintained on control diet for two weeks. WB had no effect upon carbohydrate tolerance. In a second study, augmenting the test meal with guar resulted in lower plasma glucose in the first 30 min postprandially. Since both acute and chronic fibre effects have been shown, this animal model is suitable to study the mechanism by which different dietary fibres improve carbohydrate tolerance.  相似文献

Food intervention is a financially sensible way for prevention and treatment of diabetes. Extruded snack foods are considered high glycaemic products. Our previous research illustrated that postprandial glycaemic responses to snacks are manipulated by altering dietary fibre and starch contents. The current research assessed the effect of psyllium and oat bran on postprandial glycaemia and in?vitro digestibility. Addition of psyllium fibre to extruded snack products significantly reduced both the in?vitro and in?vivo glycaemic responses of products compared to a control snack product recipe. Oat bran inclusion reduced in?vitro starch digestibility but not in?vivo glycaemic response. The inclusion of oat bran into the snack products appeared to extend the glycaemic response of individuals compared to the control snack, suggesting a possibility of prolonging glucose release and potentially affecting satiety responses. The positive effect in attenuating glucose response means that psyllium fibre could be a target for inclusion by the snack food industry to effectively manipulate postprandial glucose response of individuals.  相似文献

Despite the physiochemical complexity of dietary fibers (plant cell walls) and their individual components, there is substantial epidemiologic, clinical, and experimental evidence that these dietary components may have a role in modifying certain risk factors in coronary heart disease. Particulate fibers, such as wheat bran, do not appear to significantly alter plasma lipids or lipoprotein distributions in humans, or the atherogenicity of diets in experimental animals. Dietary fibers found in fruits, legumes, and vegetables, in contrast, show more definitive responses. Among the fiber isolates, the gelling and mucilaginous fibers, such as pectins and guar gum, predictably decrease circulating lipids in humans and animals and increase excretion of fecal metabolites of cholesterol, the bile acids. These fibers and fiber components can be shown to influence luminal solubility of lipids and the extent of lymphatic absorption of both cholesterol and triglyceride. In addition, these same fibers are effective in reducing postprandial levels of glucose, insulin, and other hormones. These direct effects on lipid absorption, and secondary effects of glucose and insulin on hepatic and peripheral lipoprotein metabolism, can account for many of the hypolipidemic responses to specific dietary fibers or their components, and may be of long-term consequence in coronary heart disease.  相似文献

Fibrous materials, exemplified by asbestos, that release respirable fibres are in common use and there is considerable knowledge regarding the toxicology of these common fibres. Newer materials or those that are under development, such as synthetic organic fibres and carbon nanotubes may have a different toxicology paradigms. The existing paradigm for silicate fibres suggests that respirable fibre types vary in their ability to cause lung disease and that this can be understood on the basis of the length of the fibres and their biopersistence in the lungs. Because fibres are regulated on a fibre number basis and the hazard is understood on the basis of the number of long fibres, in fibre testing the dose should always be expressed as fibre number, not mass and the length and diameter distribution need to be known. Short-term biological tests are likely to produce false positives in the case of long non-biopersistent fibres, because whilst they may have effects in vitro, they do not persist long enough in the lungs for sufficient dose to build up and produce effects in vivo. The biopersistence of fibres is therefore a key factor that needs to be known in order to interpret short-term tests that may claim to predict fibre pathogenicity.  相似文献

A better understanding of the functionality of probiotics and dietary fibres with prebiotic activity is required for the development of improved synbiotic preparations. In this study, utilization of β(2-1) fructans, galactooligosaccharides, and plant polysaccharides as prebiotics by lactobacilli, bifidobacteria, and pediococci was investigated. Our results demonstrate that prebiotics with linear chains consisting of galactose units are better utilized by probiotics than are those consisting of glucose and fructose units, and the ability of probiotic bacteria to utilize prebiotics is strain-specific. In addition, rye fructooligosaccharides represent a prebiotic fibre that supports the growth of a wide range of probiotic cultures and as such has a potential to improve the successfulness of probiotic treatments. This study also demonstrates dietary fibre utilization by pediococci and provides data supporting the possible use of pediococci as a probiotic in synbiotic combinations.  相似文献

Weaning is a critical phase in intensive piglet production marked by the frequent occurrence of digestive disorders posing health and economic burden. To stave off such weaning-related problems, antibiotics and supplements containing high levels of ZnO are often used. In this study, we investigated whether natural fibres obtained from two plants known for their dietary fibre profile and antioxidant properties: flax (Linum usitatissimum L.) and hemp (Cannabis sativa L.), can serve as a health-promoting diet supplement. The study was conducted on a total of 191 Polish Large White piglets from 18 L, from their birth to 56 days of age. Piglets were divided by litters into six groups: W1.5, F1.5, H1.5, W2.0, F2.0, and H2.0. Groups W1.5, F1.5, and H1.5 received feed supplemented with 1.5% fibre from wood cellulose (W), flax (F), and hemp (H), respectively, while groups W2.0, F2.0, and H2.0 received feed with a higher 2.0% content of the same fibres. Flax and hemp fibres were characterised by a complex composition, antioxidant properties due to the presence of phenolic acids, and low risk of mycotoxin contamination. Flax fibre resulted in best weight gains and feed conversion ratio (P ≤ 0.05) of piglets, while hemp fibre had higher positive effect on antioxidant status (P ≤ 0.05) compared to the other two fibre additives. Neither flax nor hemp fibres had any adverse effect on the haematological and biochemical blood parameters. Piglets receiving a diet with 1.5% added fibre showed better growth performance, while diet supplementation with 2% fibre had a beneficial effect on the content of butyric acid in the small intestinal chyme (P ≤ 0.05). The results suggested that both flax and hemp fibres can be innovative feed additives for weaned piglets. However, further studies should be conducted in commercial farms, as the effects of dietary fibre could vary in more challenging environmental conditions.  相似文献

The influence of wheat bran content in biodegradable composites based on cassava starch and containing glycerol and potassium sorbate were studied. Films were produced by casting and three different fractions of wheat bran fibre were used: 1.5 mg, 13.5 mg and 27.1 mg/g of matrix.It was observed that the addition of wheat bran, which contains 40 g of water insoluble fibre per 100 g of bran, shifted the glycerol-rich phase glass transition temperature toward higher temperatures, broadening and diminishing in intensity the peak associated with this relaxation. This effect suggests that the presence of fibre led to an enhancement in the glycerol dispersion.At room temperature, an increase in fibre content did not affect density of the matrix but caused the increase of the storage modulus and the decrease of loss tangent, moisture content and water vapor permeability. Besides, the addition of fibres led to the increase of the yellow index.The improvement in water vapor barrier properties jointly with the enhancement of mechanical properties when fibre was present, lead to the idea that the composite developed can be used to protect food and extend its shelf life.  相似文献

This paper describes the structure of the flagella, basal bodies, and some of the associated fibre systems in three genera of complex flagellates, Trichonympha, Pseudotrichonympha, and Holomastigotoides. Three groups of longitudinal fibres occur in a flagellum: two central and nine outer fibres such as have been repeatedly described in other material, and an additional set of nine smaller secondary fibres not previously identified as such. Each central fibre shows a helical substructure; the pair of them are enveloped in a common sheath. Each outer fibre is a doublet with one subfibre bearing projections—called arms—that extend toward the adjacent outer fibre. The basal body is formed by a cylinder of nine triplet outer fibres. Two subfibres of each triplet continue into the flagellum and constitute the doublets. The third subfibre terminates at the transition of basal body to flagellum, possibly giving rise to the nine radial transitional fibres that seem to attach the end of the basal body to the surface of the organism. The central and secondary flagellar fibres are not present in the lumen of the basal body, but other complex structures occur there. The form of these intraluminal structures differs from genus to genus. The flagellar unit is highly asymmetrical. All the flagella examined have possessed the same one of the two possible enantiomorphic forms. At least two systems of fibres are associated with the basal bodies of all three genera.  相似文献

Apart from being an energy source, dietary fibre is also discussed to act as anti-nutritional factor reducing apparent precaecal protein and amino acid (AA) digestibility due to reduced absorption or increased endogenous secretion or both. However, the amounts of protein and AA of endogenous origin determined at the terminal ileum in cannulated animals do not represent the total losses associated with endogenous secretion. A high proportion of secreted protein is reabsorbed and does not reach the terminal ileum, and losses occur during synthesis of endogenous protein. Therefore, the present study used an alternative indirect approach, taking the reduction of nitrogen (N) retention in a threonine (Thr) limited diet as a sensitive indicator for fibre-associated Thr losses. Two experiments were conducted with 12 castrated male pigs each between 37 and 75 kg body weight to measure the effect of the intake of Thr and 150 and 300 g/d fibre from wheat bran (Exp. 1), or 150 g/d fibre from rape seed, cassava leaves, and cassava root peels, respectively (Exp. 2), on N retention. During two (Exp. 1) and three (Exp. 2) balance periods the animals were subjected to the dietary treatments according to a cross-over design. All animals received 1350 g/d of a wheat-soybean-based diet supplemented with free AA to ensure Thr being the first-limiting AA. To determine the effect of Thr on N retention, intake of the basal diet was reduced to 1150 g/d and supplemented with corn starch to reach equal energy intake and an unchanged AA pattern. With increasing BW additional starch was added to all diets to ensure a constant energy intake of 1.25 MJ ME/kg BW(0.75). Since the fibre sources contained small amounts of Thr, N retentions were corrected for precaecal digestible Thr intake originating from the fibre sources according to the Thr effect on N retention as determined in experiment 1. Corrected N retentions were affected by fibre level (p = 0.007) and source (p < 0.001). Fibre-associated Thr losses amounted to 3.3, 3.2, 1.2, and 1.1 g/kg fibre from wheat bran, rapeseed, cassava leaf, and cassava root peel, respectively. It is concluded that Thr losses per gram of dietary fibre depend on the fibre source and that fibre concentration and source in pig diets should be considered as a factor affecting Thr requirement.  相似文献

One of the theories to explain the protective action of some dietary fibres against colon cancer is that certain mutagens and/or cancer promoters are adsorbed to these dietary fibres making the mutagens and/or cancer promoters less available to gut mucosal cells. The abilities of 2 contrasting cell wall preparations (dietary fibre preparations) from potato tubers to adsorb in vitro the hydrophobic mutagen, 1,8-dinitropyrene (DNP), were studied using an incubation mixture containing DNP in phosphate-buffered saline (PBS). Walls from potato skins strongly adsorbed DNP and, at the highest wall concentration tested, only a small porportion of the DNP remained in solution. In marked contrast to the skin walls, potato flesh walls adsorbed only a small proportion of the DNP. Unexpectedly, the flesh walls also caused a large increase in the proportion of DNP found in solution. When flesh walls were pre-extracted with PBS, the ability of the extracted walls to bind DNP increased. The material extracted from the flesh walls was able to maintain DNP in solution, when added to the incubation medium in the absence of cell walls. Pectic polysaccharides appear to be the soluble component responsible for maintaining the DNP in solution. Competition between soluble and insoluble fibre components may have major implications for the availability and distribution of hydrophobic mutagens in the alimentary tract.  相似文献

AIMS: To optimize the solid-state cultivation conditions for xylanase production using agriresidues and testing the biobleaching efficiency of xylanase on nonwoody plant fibre materials. METHODS AND RESULTS: An extracellular cellulase free xylanase was produced from Bacillus subtilis C 01 using various inexpensive substrates under solid-state cultivation. High level of xylanase production (135 IU gds(-1)) was observed when grown on wheat bran followed by maize powder (50 IU gds(-1)). The maximum xylanase (136 IU gds(-1)) production was occurred in wheat bran-to-moisture ratio of 1 : 1 at 72 h. The xylanase pretreated pulp samples of banana, silk cotton and cotton showed an increased brightness of 19.6, 11.6 and 7.9%, respectively. CONCLUSIONS: The enzyme-aided biobleaching results indicate that the xylanase has potential application in enhancing the brightness of nonwoody plant fibre pulp. SIGNIFICANCE AND IMPACT OF THE STUDY: This is the first report on biobleaching of banana fibres, silk cotton and cotton pulps using xylanase. The biobleaching results of secondary fibres are promising and can be transferred to paper mills, which utilize nonwoody plant fibres as a raw material for paper production.  相似文献

The changes in the excitability of the reorganized axonal membrane in myelinated and demyelinated nerve fibres as well as the causes conditioning such changes have been investigated by paired stimulation during the first 30 ms of the recovery cycle. The variations of the action potential parameters (amplitude and velocity) are traced also. The simulation of the conduction along the normal fiber is based on the Frankenhaeuser and Huxley (1964) and Goldman and Albus (1968) equations, while the demyelination is considered to be an elongation of the nodes of Ranvier. The axonal membrane reorganization is achieved by means of potassium channel blocking and increase of the sodium-channel permeability. It is shown that potassium channels block decreases membrane excitability for the myelinated and demyelinated fibres in the cases of initial and paired stimulation. With increasing sodium-channel permeability on the background of the blocked potassium channels, the membrane excitability is increased. For the fibres with a reorganized membrane, a supernormality of the membrane excitability is obtained, the latter remaining unrecovered during the 30 ms cycle under investigation. The supernormality of the excitability grows from the demyelinated fibre without reorganized membrane to the demyelinated fibre with reorganized one. For short interstimulus intervals, the second action potential propagates along the fibres with a reduced velocity and a decreased amplitude. No supernormality of the potential parameters (amplitude, velocity) is observed during the cycle up to 30 ms. The membrane properties of the myelinated and demyelinated fibres with blocked potassium channels recover in the interval from 15 to 20 ms depending on whether the sodium channels' increase of the permeability is added on the background of the blocked potassium channel or not. In the recovery cycle, the axonal membrane reorganization leads to an improvement of the conduction along most severely demyelinated fibres.  相似文献

We analysed the influence of several enzymatic treatment processes using an alkaline cellulase enzyme from Bacillus spp. on the sorption properties of cotton fabrics. Although cellulases are commonly applied in detergent formulations due to their anti-redeposition and depilling benefits, determining the mechanism of action of alkaline cellulases on cotton fibres requires a deeper understanding of the morphology and structure of cotton fibres in terms of fibre cleaning. The accessibility of cellulose fibres was studied by evaluating the iodine sorption value and by fluorescent-labelled enzyme microscopy; the surface morphology of fabrics was analysed by scanning microscopy. The action of enzyme hydrolysis over short time periods can produce fibrillation on cotton fibre surface without any release of cellulosic material. The results indicate that several short consecutive treatments were more effective in increasing the fibre accessibility than one long treatment. In addition, no detectable hydrolytic activity, in terms of reducing sugar production, was found.  相似文献

Diets containing wheat bran (WB) protect against cancers of the colon or breast in rats, and may be beneficial in humans. In a previous study of rats treated with the carcinogen 2-amino-3-methylimidazo[4,5-f]quinoline (IQ), inclusion of 10% wheat bran in the diet led to an apparent reduction in IQ metabolites but not of intact IQ in plasma. In the present study, male Wistar rats were fed diets containing 0, 10 or 20% wheat bran, and effects on xenobiotic metabolising enzymes compared. Wheat bran-supplementation showed differential effects on phase I enzymes, significantly increasing the activity of hepatic cytochrome P450 isozyme CYP3A2, but slightly reducing the activity of CYP1A1/2. The activities of both hepatic phase II detoxification enzymes glutathione-S-transferase and glucuronosyl transferase were also reduced. Western blotting revealed similar effects on expression of the proteins. Interestingly, the expression of xenobiotic metabolising enzymes (XME) in the colon appeared to be modulated independently of hepatic XME. Although the wheat bran-supplemented diet still led to an increased expression of CYP3A, it now slightly increased CYP1A in the colon. However, 20% wheat bran significantly increased the expression of both glutathione transferase isozymes, GST A1 & A2, in the colon. Natures Gold (NG) is a commercial wheat bran derivative which is lower than wheat bran in dietary fibre, but enriched in vitamins, minerals and various phytochemicals. Dietary supplementation with 20% Natures Gold led to similar trends as seen in wheat bran-fed rats, but more potent effects in both hepatic and colonic enzymes. The significance of these changes for activation of carcinogens to mutagenic metabolites was investigated using the Salmonella/mammalian microsome mutagenicity test. The activation of IQ and benzo[a]pyrene, but not cyclophosphamide, to a mutagen by hepatic S9 from wheat bran-fed or Natures Gold-fed rats was significantly reduced compared with S9 from animals on a diet lacking wheat bran. We suggest that modulation of xenobiotic metabolising enzymes may be an important component of cancer protection by wheat bran, and this effect may relate to micronutrients or cancer-protective non-nutrient phytochemicals rather more than to dietary fibre.  相似文献
