The metabolism of liver nuclear phospholipid acyl components of rats of various age was studied in vitro. It was found that the activity of phospholipases A1 and A2 in the nuclei sharply increased in animals aged 1-3 months, showing a decrease in 12- and 24-month-old animals. The incorporation of labeled arachidonic acid into nuclear phospholipids remained practically unchanged thereby. The age-specific fluctuations in the activity of nuclear phospholipids A1 and A2 may be one of possible reasons of changes in the fatty acid composition of nuclear lipids during ontogenesis.  相似文献

The putative role of the nuclear nucleoside triphosphatase (NTPase) is to provide energy to the nuclear pore complex for poly A(+) mRNA export. Previous work has demonstrated that liver nuclear NTPase activity is greater in 6 month old corpulent (cp/cp) female JCR:LA rats, a hyperlipidemic rat model, compared to lean (+/?) animals. This increase appeared to be related to increases in nuclear membrane cholesterol content. The current study extended these initial data to compare NTPase activity as a function of age and sex in isolated JCR:LA-cp rat liver nuclei, to further test the hypothesis that nuclear membrane cholesterol may modulate NTPase activity. NTPase activity was increased in cp/cp female animals compared to +/? females at all ages studied, with Vmax values increased by 60-176%. Membrane integrity of cp/cp female nuclei was reduced compared to +/? female nuclei. Nuclear membrane cholesterol levels increased linearly with age by 50, 150 and 250% in 3, 6 and 9 month old cp/cp females over leans. In contrast, nuclei from cp/cp males exhibited only minor, isolated changes in NTPase activity. Furthermore, there were no significant changes in nuclear cholesterol content or membrane integrity in the less hyperlipidemic male animals at any age. These data suggest that altered lipid metabolism may lead to changes in nuclear membrane structure, which in turn may alter NTPase activity and functioning of the nuclear pore complex.

The effect of cytosol and ATP-regenerating system on RNA, transport was studied in isolated liver nuclei of adult and old rats. The stimulating effect of cytosol was found not to depend on the age of animals. The release of RNA from old rat liver nuclei activated by the ATP-regenerating system was more expressed compared to adult rats. It is assumed that the age changes of energy-delivering system of the RNA transport through nuclear membrane may be conditioned by the deficit of endogenous energetic substrates in the hepatic cells of old animals.  相似文献

Differences in immunological abnormalities like autoimmunity, abnormal T cell proliferative disorders and accelerated ageing occur between MRL/Mp-lpr/lpr(lpr/lpr) and MRL/Mp-+/+(+/+) mice as a consequence of one gene. The present study was designed to assess the effect of these differences in genotype and diet on the composition and function of the liver nuclear envelope. Mice of both strains were fed nutritionally adequate diets differing only in fatty acid composition for 4 weeks. Phospholipid fatty acid composition of the liver nuclear envelope was determined and the effect of altering the lipid composition of the nuclear membrane on nucleoside-triphosphatase (NTPase) activity, ribonucleic acid (RNA) efflux and binding of L-triiodothyronine (L-T3) was determined. Strain of mouse and level of dietary linoleic acid exhibited significant effects on the phospholipid fatty acid composition of the nuclear envelope. Levels of 18:1(n - 9) and 18:2(n - 6) were lower and 20:4(n - 6) content was higher in nuclear envelope phospholipids of lpr/lpr mice compared with mice of the +/+ strain. Mice fed the high linoleic acid diet exhibited higher levels of 18:0, 18:2(n - 6) and 20:4(n - 6) and lower levels of 16:0 and 18:1(n - 9) in liver nuclear envelope phospholipids, compared with mice fed the low linoleic acid diet. These changes in membrane composition were reflected in alteration of NTPase activity and efflux of RNA from isolated mouse liver nuclei. Nucleoside triphosphatase activity and efflux of ribonucleic acid from isolated nuclei were significantly higher in livers of the lpr/lpr strain. NTPase activity and RNA efflux from isolated nuclei were higher in the high linoleic acid fed group compared with the low linoleic acid group. A single class of binding sites for L-T3 was present in liver nuclear envelopes of these mice and Kd values were not influenced by strain or dietary linoleic acid levels. Nuclear envelopes prepared from +/+ animals exhibited a significantly higher number of binding sites for L-T3 compared with the lpr/lpr group. These observations indicate that the single gene difference characterizing lpr/lpr mice from +/+ mice results in alterations in the composition and function of the nuclear envelope. This genetic difference also alters the response of this membrane to dietary factors known to modulate characteristics and functions of the nuclear envelope.  相似文献

A nuclear pore complex-associated nucleoside triphosphatase (NTPase) activity is believed to provide energy for nuclear export of poly(A)+ mRNA. This study was initiated to determine if nuclear membrane lipid composition is altered during chronic hyperlipidemia, and what effect this has on NTPase activity. The JCR:LA-cp corpulent rat model is characterized by severe hypertriglyceridemia and moderate hypercholesterolemia, and thus represents an ideal animal model in which to study nuclear cholesterol and NTPase activity. NTPase activity was markedly increased in purified hepatic nuclei from corpulent female JCR:LA-cp rats in comparison to lean control rats as a function of assay time, [GTP], [ATP], and [Mg2+]. Nuclear membrane cholesterol and phospholipid content were significantly elevated in the corpulent animals. Nuclei of corpulent animals were less resistant to salt-induced lysis than nuclei of lean animals, suggesting a change in relative membrane integrity. Together, these results indicate that altered lipid metabolism in a genetic corpulent animal model can lead to changes in nuclear membrane lipid composition, which in turn may alter nuclear membrane NTPase activity and integrity. © 1996 Wiley-Liss, Inc.  相似文献

In rats injected intraperitoneally with actinomycin D (2 mg/kg body weight) 12 h earlier, the yield of the 30S ribonucleoprotein particles isolated from liver nuclei by extraction with 0.1 M NaCl at pH 8.0 decreased by 60 per cent. The protein-to-RNA ratio of these particles increased to 32:1 from the ratio 4.4:1 found in the same particles isolated from the nuclei of liver of control rats. The particles isolated from the liver nuclei of rats injected with actinomycin D were depleted of all charge isomers of the two most prominent polypeptides (33,000 and 39,000 daltons) present in the particles of liver of control animals. The most abundant protein in these particles was a 43,000 dalton polypeptide. This polypeptide is the least prominent of the 3 major polypeptides present in the control particles. The same charge isomers of the 43,000 dalton polypeptide were present in the nuclear ribonucleoprotein particles isolated from the liver of control animals and from the liver of animals treated with actinomycin D 12 h earlier. In control animals the nuclear ribonucleoprotein monoparticles isolated from kidney contained 3 major polypeptides of the same molecular weight with the same distribution of their charge isomers as were present in the particles isolated from liver nuclei. The injection of actinomycin D 12 h earlier was without any effect on the protein composition of the 30S nuclear ribonucleoprotein particles of rat kidney.  相似文献

In humans and animals, food composition, especially dietary fat, affects androgen secretion and metabolism. On the other hand, disturbances of sex steroid metabolism play an important role in the etiology of hormone-related cancers. In this report the roles of dietary fat, its quantity, fatty acid composition and feeding period on androgens metabolism was described. In conclusion, it should be stated that the amount of dietary fat, and its composition, (i.e. the content of individual fatty acids and/or their groups), as well as the period during which the nutrient is fed to animals affect significantly the secretion and metabolism of androgens.  相似文献

The possible influence of the type of dietary fat on quantitative changes of different lipids in microsomes, nuclei and chromatin in hepatic cells of albino rats in ontogenesis was studied. While the type of diet had no significant influence on the levels of different phospholipids and fatty acids of nuclei, the type of dietary fat exerted definite effect on the levels of whole phospholipids, cholesterol of microsomes, nuclei and fatty acids of chromatin. The effects of in vitro and in vivo modifications of nuclear envelope lipid on DNA leakage and on ATP-stimulated RNA release from isolated rat liver nuclei were investigated. The modifications included corn-oil feeding of the animals to alter the fatty acid composition of the lipids, phospholipase treatment of the isolated nuclei, and extraction of the total lipid with Triton X-100. Significant changes in lipid composition and approximate order parameter values of the spin-label 5-doxylstearate resulted, but there was no significant effect on RNA transport rate. It was concluded that the nuclear envelope lipid does not play any important part in nucleocytoplasmic RNA transport in mammalian liver.  相似文献

It was discovered that there is sphingomyelinase activity in the rat liver nuclei. The maximum of enzyme activity is at pH 7.1. The data obtained demonstrated that the main part of sphingomyelinase is located in the nuclear membrane. Comparison of sphingomyelinase activity in cell nuclei, liver and brain homogenates shows high level of enzyme activity in the nuclei. The authors discuss possible participation of sphingomyelinases in changes of phospholipids composition in nuclear structure under different functional activity of cell nuclei.  相似文献

The effects of dietary fat on fatty acid compositions of phospholipids in liver nuclei were investigated. By feeding a diet containing 5% corn oil or fish oil to rats, the proportions of n -6 or n -3 polyunsaturated fatty acids (PUFA), respectively, were increased in phospholipids of the liver nuclei. By feeding a fat-free diet, endogenous PUFA were increased. Even after feeding the fat-free diet for 40 weeks, the n - 6 still remained at about 10% in both phosphatidylcholine (PC) and phosphatidylethanolamine (PE), while the n -3 PUFA remained at about 5 and 16% in PC and PE, respectively. The fatty acid compositions of phospholipids in the liver nuclei were influenced by dietary fat and were roughly similar to those in the microsomes. The proportion of n - 3 PUFA was high in PE of both the nuclear membrane and matrix. The proportion of n - 6 was higher in both PC and PE of the nuclear matrix than in those of the membrane.  相似文献

Sphingomyelin metabolism in liver cell nuclei of rats of various age has been studied. It was found that the level of sphingomyelin hydrolysis in cell nuclei is the highest in young animals, showing a decrease in 24-month-old animals. The age-specific fluctuations in the activity of phospholipase C may be one of possible reasons for sphingomyelinase activity changes in liver nuclei during ontogenesis. Changes of antioxidative activity (AOA), lipid composition and microviscosity of different membrane regions in tumor cell nuclei and in the liver of tumor-host with Ehrlich ascite carcinoma (EAC) after irradiation were studied. On the basis of the obtained data the analysis of the control system of lipid oxidation in the membrane was carried out. This control system involves a relationship between AOA changes, lipid composition, their oxidative ability and the nuclear membrane structure. It was shown that after irradiation the control system in the nuclei of tumor cells had the same state as before irradiation and was different from the normal one. The control system in the nuclei of tumor-host liver after irradiation starts to work in a regime which is characteristic of irradiated cells. It was shown that the principle difference in the control system functioning in tumor and tumor-host nuclei disappeared after irradiation.  相似文献

Diets supplemented with relatively high levels of either saturated fatty acids derived from sheep kidney fat (sheep kidney fat diet) or unsaturated fatty acids derived from sunflower seed oil (sunflower seed oil diet) were fed to rats for a period of 16 weeks and changes in the thermotropic behaviour of liver and heart mitochondrial lipids were determined by differential scanning calorimetry (DSC). The diets induced similar changes in the fatty acid composition in both liver and heart mitochondrial lipids, the major change being the omega 6 to omega 3 unsaturated fatty acid ratio, which was elevated in mitochondria from animals on the sunflower seed oil diet and lowered with the mitochondria from the sheep kidney fat dietary animals. When examined by DSC, aqueous buffer dispersions of liver and heart mitochondrial lipids exhibited two independent, reversible phase transitions and in some instances a third highly unstable transition. The dietary lipid treatments had their major effect of the temperature at which the lower phase transition occurred, there being an inverse relationship between the transition temperature and the omega 6 to omega 3 unsaturated fatty acid ratio. No significant effect was observed for the temperature of the higher phase transition. These results indicate that certain domains of mitochondrial lipids, probably containing some relatively higher melting-point lipids, independently undergo formation of the solidus or gel phase and this phenomenon is not greatly influenced by the lipid composition of the mitochondrial membranes. Conversely, other domains, representing the bulk of the membrane lipids and which probably contain the relatively lower melting point lipids, undergo solidus phase formation at temperatures which reflect changes in the membrane lipid composition which are in turn, a reflection of the nature of the dietary lipid intake. These lipid phase transitions do not appear to correlate directly with those events considered responsible for the altered Arrhenius kinetics of various mitochondrial membrane-associated enzymes.  相似文献

Insulin binding to the plasma membrane is known to be altered by modifying the membrane composition through dietary treatment. As insulin binding receptors are also present on nuclear membrane, this study was undertaken to investigate if specific binding of insulin to the liver nuclei is altered by diet. 8-wk-old female C57 B 6J lean and ob/ob mice were fed semipurified diets containing 20% (w/w) fat of either high or low polyunsaturated-to-saturated (P/S) fatty acid ratio for 4 wk. Liver nuclei were prepared, insulin binding was measured and nuclear phospholipids were isolated for lipid analysis. Insulin binding was highest in nuclei prepared from lean mice fed a high P/S diet. Specific binding of insulin to nuclei prepared from obese mice was also increased by the high P/S diet, but to a lesser extent compared to lean mice. Feeding a high P/S diet increased polyunsaturated fatty acid content of membrane phospholipids from both lean and ob/ob mice. Obese mice were characterized by higher levels of arachidonic acid and lower levels of linoleic acid in phosphatidylcholine. The present study establishes that insulin binding to liver nuclei is increased by feeding a high P/S diet, and that insulin binding to liver nuclei from obese mice is lower than from lean mice.  相似文献

Injections of 6-hydroxydopamine in mouse neonates caused extensive and long lasting damage to the sympathetic nervous system and impaired brown fat development. Brown adipose tissue (BAT) thermogenic capacity of sympathectomized mice (up to 120 days old) was reduced because of marked reductions in the tissue mitochondrial protein content and the mitochondrial concentration of uncoupling protein, as assessed by [3H]GDP binding and immunoassay. Neonatal sympathectomy did not affect BAT DNA content. Sympathectomized mice also had reduced epinephrine-stimulated rates of oxygen consumption. BAT of sympathectomized mice failed to respond by increases in [3H]GDP binding to isolated mitochondria and uncoupling protein concentration when animals were offered a palatable high-fat dietary supplement that increased calorie intake of both normal and sympathectomized mice. The high-fat diet caused increases in body weight, carcass fat, and gonadal white fat pad weights in sympathectomized animals that were similar to those of control mice. These results show that inactivation of BAT metabolism did not accentuate the development of obesity caused by a dietary supplement rich in fat and suggest that stimulation of BAT metabolism was not very effective in counteracting the obesity-inducing effect of this diet.  相似文献

Three experiments were designed to investigate the metabolism of dietary nervonic acid (24:1n-9, NA) during reproduction in the rat. The first experiment determined the effect of early development on the sphingomyelin (SM) composition of rat heart and liver tissues. Rats were fed a standard chow diet and the SM fatty acid composition of the hearts and livers were analyzed of 18-20 day old fetuses, 14 day old sucklings and adult rats. The 18:0 content of SM decreases with age, while 23:0 and iso 24:0 increase with age. In the second experiment pregnant rats were fed diets supplemented with either canola, corn or peanut oil to determine the effect of diets high in 24:1n-9 and 24:0 on liver and heart SM at birth and after 14 days of suckling. Pups from the dams fed the corn oil diet had elevated 24:2n-6 in SM from heart and liver at birth, but the content of NA was not altered by dietary fat type. In the third experiment oil mixtures were designed to provide elevated levels of 22:1 and 24:1 (canola-N25), 22:0 and 24:0 (peanut-flax) or <0.01% of these fatty acids (olive-flax) and were supplemented to the diets of lactating rats. Canola-N25 oil supplemented to lactating rats resulted in increased 24:1n-9 and 24:1/24:0 with decreased 22:0 and 24:0 in milk SM relative to the other groups. The SM composition of livers of the suckling rats showed significant changes reflecting the changes in milk SM composition after 6 days of milk consumption. These experiments suggest that dietary NA and is not readily transferred across the placental barrier but does readily cross the mammary epithelium and is incorporated into milk SM. In addition, NA in milk appears to cross the intestinal epithelium where it is incorporated into the SM of heart and liver of suckling rats.  相似文献
