1. The effects of arachidonate (AA) on myocardial and coronary function, and the ability to metabolize AA have been explored for the first time in the heart of rainbow trout, Oncorhynchus mykiss.2. On the isolated and perfused working heart exogenous AA (10−7 and 10−5 M) elicits a significant reduction of cardiac output and power output while heart rate is unaffected.3. The negative inotropic effect is abolished in presence of 10−5 M indomethacin. At the same AA concentration a pronounced increase in coronary resistance (175% change from baseline values) is apparent which is reduced but not abolished in the presence of 10−5 M indomethacin.4. 14C-arachidonate is metabolized by trout ventricle homogenate into PGs E2, F, D2, and in lesser amount into TXB and 6-keto-PGF. Ca-ionophore A23187 enhances the production of both PGE2 and PGF. The lipoxygenase products assayed as the hydroxy acids (HETEs) appear to be less actively synthesized than prostanoids.  相似文献

Myometrial low speed supernatant prepared from non-pregnant rhesus uteri was incubated with 3H-Prostaglandin (PG)E1 with or without addition of unlabelled prostaglandins. The uptake of 3H-PGE1 was inhibited in a dose dependent fashion by PGE2>PGE1>PGA1>PGF=PGA1>PGB1=PGB2≥PGD2. PGE1 metabolites inhibited 3H-PGE1 binding in the following order: 13,14-dihydro-PGE1>13,14-dihydro-15-keto-PGE1=15-keto-PGE1. The specific binding of 3H-PGE1 and 3H-PGF was similarly affected by the temperature and time of incubation. Equilibrium binding constants determined using rhesus uteri obtained during the luteal phase of the menstrual cycle indicate the presence of high affinity PGE1 binding sites with an average (n=3) apparent dissociation constant of 2.2 × 10−9M and a lower affinity PGE1 binding site with a Kd ≅ 1 × 10−8M. No high affinity — low capacity 3H-PGF sites could be demonstrated.Relative uterine stimulating potencies of some natural prostaglandins and prostaglandin analogs tested after acute intravenous administration in mid-pregnant rhesus monkeys corresponded with the PGE1 binding inhibition of the respective compound. The uterine stimulating potencies of the prostaglandin analogs tested were: (15S)-15-methyl-PGE2=16,16-dimethyl-PGE2>17-phenyl-18,19,20-trinor-PGE2>16 phenoxy-17,18,19,20-tetranor-PGF=PGE2=PGE1=(15S)-15-methyl-PGF>PGF.  相似文献

The effects of prostaglandin F (PGF) on propulsive activity in segments of isolated colon and on isolated strips of guinea-pig colon were investigated.Using experimental conditions under which spontaneous propulsive activity was negligible, PGF (5×10−8×1×10−6M), added to the bathing medium, increased propulsive activity in a concentration dependent manner. This increase of propulsive activity was abolished in the presence of atropine or tetrodotoxin (1×10−7g/ml).The contractions produced by PGF(5×10−7 − 1×10−5M) in isolated longitudinal and circular smooth muscle strips of guinea-pig colon were unaffected in the presence of atropine or tetrodotoxin (1×10−7g/ml).From these results it is concluded that under the conditions employed in this study propulsive activity stimulated by PGF may depend on the contractions of both muscle layers and stimulation of the peristalic reflex.  相似文献

Transformed cells from human lung carcinoma (Line A549), resembling type II pneumocytes, were cultured in monolayer at 37°C and incubated for five hours with 3H-choline and 14C-palmitate in the presence of various concentrations of prostaglandins (PGs) E2 and F. In the control (no PG) the level of % palmitate incorporation was 13.5 × as high as that of choline, after taking isotope dilution into account. Between the concentrations studied, 0.1 and 10 μM, both prostaglandins stimulated markedly the incorporation of both precursors, though choline up to 3 × better than palmitate. This was indicated by a change in the palmitate/choline incorporation ratio from 13.5 to as low as 4.2. At the lowest PG concentration, 0.1 μM, PGE2 was much more effective than PGF in stimulating the incorporation of both precursors.  相似文献

The inotropic and chronotropic actions of prostaglandin (PG) types PGE1, PGA1, and PGF were studied in isolated guinea pig right and left atria, and papillary muscles; rabbit atria; and toad ventricular strips in order to more completely define the cardiac contractile properties of PG. All three prostaglandins, in muscle bath concentrations of 10μg/ml, exerted positive inotropic and chronotropic actions on guinea pig atrium. These contractile effects were persistent after removal of PG from the muscle bath and appeared to limit the relative response to a subsequent dose of PG. The inotropic action of PGE1 was present over a wide range of bath calcium concentrations (1.1 to 4.4 mM/L). Beta adrenergic receptor blockade, histamine blockade, and pretreatment with reserpine failed to significantly affect the inotropic actions of PG. Norepinephrine and histamine produced more potent inotropic and chronotropic effects on guinea pig atria than did PG and these contractile effects did not exhibit persistence or tachyphylaxis. The prostaglandins did not significantly affect dose response curves for norepinephrine inotropic and chronotropic actions. The prostaglandins had no effect on the force or frequency of contraction in rabbit atria. PGE1 exerted a positive inotropic effect on toad ventricular myocardium whereas PGA1 had no effect and PGF had a negative inotropic action.  相似文献

Binding sites on human lymphocytes for prostaglandins were examined by incubating cells with [3H]prostaglandin (PG) A1, E1, E2, F, and F. Specific reversible binding for [3H]PGE1 and E2 was found with a Kd of ~2 × 10?9M and a B max of ~200 binding sites per cell, assuming uniform distribution. We detected no specific binding of [3H]PGA1, F, or F to lymphocytes. Also, the addition of 10- to 1000-fold greater amounts of unlabeled PGA, F, or F did not inhibit the binding of [3H]PGE. The time course of [3H]PGE binding appeared to be bimodal with one component complete within 5 min at 37 °C and another component of binding increasing over a 40-min incubation. We feel that the rapid component of binding may represent cell surface receptors for PGE while the slower component may represent a specific uptake mechanism for PGE into the cell. Glass adherent cells had fewer binding sites than nonadherent cells. Preincubation of the cells overnight resulted in a loss of binding sites.  相似文献

The purpose of this study was to determine the concentrations of prostaglandins E2 and F (PGE2 and PGF) in the blood, testis and seminal plasma of mature male rainbow trout and in the ovarian fluid to assess the effects of these prostaglandins on sperm motility parameters when present in activation media. Also prolonged incubation with prostaglandins on sperm motility and calcium influx were studied. The profile of PGE2 and PGF differed in concentration between blood, testicular supernatant and seminal plasma. PGE2 was predominant in the blood sample (0.29 ng ml?1) and testicular supernatant (3.1 ng ml?1) whereas their level in seminal plasma was lower than PGF (0.23 ng ml?1). The concentrations of PGF in blood, testis and seminal plasma were 0.04, 0.99, 1.3 ng ml?1, respectively. In the ovarian fluid the concentrations of both prostaglandins were higher than in the male reproductive tract. Adding both prostaglandins to activation buffer (at concentrations 15 and 70 ng ml?1) had no effect on any CASA parameters. Calcium influx related to rainbow trout sperm incubations with PGE2, and PGF was not detected. After 24 h incubation of sperm in artificial seminal plasma solution without and with prostaglandins all sperm samples increased their motility potential and intracellular calcium concentration. Therefore, this effect was not related to the presence of prostaglandins. In summary PGE2, and PGF were present in the rainbow trout male reproductive tract, and their profile varies from that of blood, testis and seminal plasma. The specific role of both prostaglandins in salmonid sperm biology remains unclear.  相似文献

Effects of prostaglandins (PG) E1, E2, F, A1, A2 nad B2 were studied on guinea-pig isolated tracheal chains. PGF, B2 and A2 produced contraction, PGE1 and E2 relaxation of the chain, but A1 produced no response. 1) From the cumulative dose response curves, PGF was more active in producing concentration than B2 or A2, though its effect was less than that of acetylcholine (ACh). PGE1-induced relaxation was less than the response to isoproterenol. 2) PGE1 and E2 1 μg/ml caused a 26.1 ± 3.83% (n=5) or a 9.5 ± 3.36 (n=6) decrease of ACh (1 μg/ml)-induced contraction respectively. The degree of relaxation produced by E1 was greater than that by E2 (P<0.01). 3) After five minutes preincubation with each of PGA1, A2, B2 and F in concentrations which did not produce any effect, ACh-induced contraction was augmented only after PGA2 (P<0.05).  相似文献

The response of endothelial adenylate cyclase (AC) to prostaglandins (PGE1, PGE2, PGF, PGF, PGD2 and PGI2) and the relationship of PGE2 to adrenergic systems were investigated in cerebrovascular endothelial cultures. E-type prostaglandins and PGI2 were more effective in stimulating endothelial AC (EC50 = 3 × 10?7M, and 3 × 10?7M, respectively) than prostaglandins of the F-series and PGD2 which activated AC at high doses only. A modulation of endothelial AC response to either PGE2 or norepinephrine (NE) was observed in the presence of both agents in the system. It was manifested by a dose-dependent NE inhibition of the PGE2-stimulated formation of cAMP, which was partially restored by phentolamine. Alpha and β-adrenergic agonists (α, clonidine and 6-fluoronorepinephrine; β, isoproterenol) also partly blocked while forskolin and PGE2 synergistically stimulated the production of cAMP in the endothelial cultures. These findings strongly suggest that the interaction of prostaglandins and α- and β-adrenergic agonists with the AC system in cerebrovascular endothelium may play a role in the regulation of the cerebral microcirculation and/or blood pressure.  相似文献

Isolated bovine, canine, and human coronary arteries exhibited dose dependent contractions to prostaglandin (PG) E2 and F (50 ng/ml to 10 μg/ml). The ED50 value for both PGE2 and PGF was 500 ng/ml in the bovine and human coronary arteries. Paradoxically, although PGE2 and PGF are vasoconstrictors, administration of their precursor, arachidonate (100 ng/ml to 10 μg/ml) caused relaxation of the bovine, canine and human coronary arteries. This observation suggests that arachidonate is not being converted by the coronary PG synthetase to PGE2 or PGF. However, the arachidonate induced coronary relaxation was inhibited by pretreatment with PG synthetase inhibitors, indomethacin, meclofenemate and aspirin. Indomethacin addition to the strips previously relaxed by arachidonate caused contraction. In contrast to other PGs (E2 and F), PGE1 (10 ng/ml to 10 μg/ml) caused dose dependent relaxation of the bovine coronary arteries (ED50 = 100 ng/ml). Indomethacin induced further relaxation of the blood vessels previously relaxed by PGE1. Since PGE1 cannot arise from arachidonate, the arachidonate coronary dilation and reversal by indomethacin must be independent of PGE1 formation. Linolenate (100 ng/ml to 10 μg/ml) and oleate (100 ng/ml to 10 μg/ml) also caused relaxation of the bovine coronary blood vessels both before and after indomethacin, thereby eliminating a direct non-specific fatty acid effect as the cause of the arachidonate relaxation. These results suggest that in isolated coronaries, arachidonate undergoes a novel conversion, possibly by PG synthetase, to a dilating substance which exerts different contractile effects than exogenously administered PGE2, PGF and PGE1.This work was supported by (USPHS) training grants NS 05221, RCDA (P.N.) The effects of several prostaglandins (PGs) injected through the subclavian artery toward the cardiac sympathetic ganglia of spinal dogs were studied by utilizing changes of the heart rate as indicator of ganglionic function. PGF (10–270 μg) administered intra-arterially in the presence or absence of preganglionic stimulation produced weak positive chronotropic effects, which were increased by physostigmine. This positive chronotropic effect of F after physostigmine was inhibited by hexamethonium plus atropine, and depressed after hemicholinium-3 except for the response elicited by the first dose of F. PGE1 and E2 injected during preganglionic stimulation did not affect the heart rate. Intra-arterially administered epinephrine and dopamine depressed dose-dependently transmission in the cardiac ganglia, the effect being inhibited by E1 and E2 but not by F. These results suggest that F facilitates the release of acetylcholine from preganglionic nerve ending, whereas E1 and E2 antagonize the inhibitory actions of catecholamine in the cardiac ganglia.  相似文献

A possible mechanism to explain the suppression of mitogen-induced lymphocyte proliferation in vitro by histamine-stimulated mononuclear cells was investigated. In initial experiments, the inhibitory action of histamine-induced suppressor factor (HSF) on lymphocyte proliferation was documented to be reduced by the addition of indomethacin (1 μg/ml). Moreover, the addition of exogeneous PGE2 (10?7-10?8 M) to mononuclear cell cultures reconstituted HSF activity in the presence of indomethacin. In order to ascertain the nature of the target cell responding to HSF, control and suppressor supernatants were incubated with human lymphocytes or monocytes (5 × 106 cells/ml) for 24 hr. Following incubation, the supernatants were assayed for their content of prostaglandin E2, F, and thromboxane B2. Monocytes (but not lymphocytes) incubated with supernatants containing HSF increased their production of prostaglandin E2, F, and thromboxane B2 by 169, 53, and 49%, respectively. Suppressor supernatants were generated with histamine or an H-2 agonist (dimaprit) and chromatographed by gel filtration on Sephadex G-100. The elution profiles for the factor(s) inducing suppression of lymphocyte proliferation (25–40,000 daltons) and augmenting PGE2 production (25,000 daltons) overlapped but were not identical. Collectively, these data suggest that HSF-mediated inhibition of lymphocyte proliferation may occur in part through the augmented production of prostaglandins and/or thromboxane B2 by human monocytes.  相似文献

Testicular levels of prostaglandin E2 and F were measured in decapsulated adult rat testis following hCG stimulation. Basal levels were, respectively, 342 ± 74 and 502 ± 89 pg/testis. Following hCG administration these basal values are not significantly modified up to 2 hours. From 2 to 24 hours the concentrations are clearly increased above the basal level: at 12 hrs they are 1925 ± 165 for E2 and 3200 ± 190 for F. Levels are back to normal at 48 hrs and remain so until 144 hrs. An identical pattern of prostaglandin release is observed in vitro in Leydig cell preparations isolated at different times following in vivo hCG injection. This suggests that prostaglandins are secreted by Leydig cells. In hypophysectomized animals the release of both prostaglandins E2 and F is similar to controls indicating that prostaglandin secretion is not directly linked to testosterone production. alternatively testosterone injections (10 mg) does not modify prostaglandin levels. Binding sites for prostaglandins E1, E2 and F are present on the Leydig cells and consequently Leydig cell function may be modulated by endogenous or exogenous prostaglandins. Their level is slightly increased at 24 hrs following hCG stimulation. Since the acute changes in prostaglandin E2 and F secretion occur during the period of “desensitization” and of acute “down regulation” of the LH-hCG receptor in the Leydig cells it is suggested that prostaglandins are involved in both phenomena.  相似文献

Early effects of various prostaglandins on the production of hexosamine-containing substances by cultured fibroblasts, which were derived from a rat carrageenin granuloma, were studied. At the stationary phase, the cells were exposed for 6 h to one of the prostaglandin A1 (PGA1), A2, B1, B2, D2, F, F, E1, E2 or arachidonic acid in various concentrations ranging from 0.01 to 10 μg/ml for all the stimuli and from 10 pg to 10 μg/ml for PGF. The activity of the cells in incorporating 3H-glucosamine into hexosamine-containing substances (acidic) glycosaminoglycans and glycoproteins) during this period was compared with that of control cells. All the stimuli tested showed more or less stimulative effect on the synthesis of hexosamine-containing substances at their specific concentrations. PGF was found to be the most potent stimulant and its stimulative effect was found significant even at the low concentration of 100 pg/ml. PGD2, F and E2 were the next potent stimuli. Their optimum dose were around 1 μg/ml but they still had significant stimulation at the concentration of 0.01 μg/ml. Effect of PGE2 was rather mild. Stimulation by PGA1, A2, B1 and B2 or arachidonic acid was seen at high dose, and its seemed to be non-specific. The results suggested that these prostaglandins such as PGF, D2, F and E2 play some important role on regulating the production of intercellular ground substances.  相似文献

Administration of exogenous prostaglandins at the time of mating may improve fertility via their effects on uterine contractility. The present study was undertaken to compare the effects of three prostaglandins that affect either the male or female reproductive uterine contractility. Contractions in the uterine body of anesthetized ewes during estrus were studied before, during and after a 5 min interval of systemic infusion of prostaglandin F-THAM salt (PGF; 5 mg), prostaglandin E1 (PGE1; 5 mg), prostaglandin E2 (PGE2; 5 mg) or vehicle. Pressure changes were detected by the use of an open-ended intrauterine catheter and a transducer. Each of the three prostaglandins initially caused a single prolonged contraction that lasted about 10 minutes and had a maximum pressure of 50 mm Hg. Prior to the prolonged contraction, PGE1 and E2 caused a relaxation for about 1 minute. In addition, PGE1 and E2 caused more secondary contractions (15–20) during the prolonged contraction than did PGF (7–9). The effects of prostaglandin (PG) treatment lasted for 20–30 minutes. The authors conclude that with the dose used the three prostaglandins studied do not have greatly different effects on uterine contractility in estrous ewes.  相似文献

1. Acetylcholine reduced atrial contractions by 82.5% in guinea pig, 50.8% in rat, and 41.5% in rabbit.2. The EC50 values for the negative inotropic effect of acetylcholine were 3.3 × 10−7 M in rat and guinea pig atria and 4.1 × 10−6 M in rabbit atria.3. There was no correlation between the species differences in the negative inotropic effect of acetylcholine in atria and the density or affinity of acetylcholinesterase or muscarinic receptors.4. Inhibition of atrial acetylcholinesterase with soman reduced the ec50of acetylcholine three-fold in all species, but did not change the maximal inotropic effect of acetylcholine.5. Species differences in the negative inotropic effect of acetylcholine may be caused by differences in the coupling between myocardial muscarinic receptors and the ion channels that mediate negative inotropy.  相似文献

THIS report describes the biosynthesis of the naturally occurring renal prostaglandins E2 (PGE2) and F (PGF)1,2 by homogenates and slices of rabbit renal medulla, from endogenous precursors. I have confirmed that rabbit renal cortex contains little prostaglandin and cannot synthesize them from endogenous lipids3. Hamberg has reported that arachidonic acid, which is converted to PGE2 and PGF by enzymes present in ram seminal vesicles4, can be efficiently converted to PGE2 and PGF by homogenates of rabbit renal medulla3. I have now confirmed that arachidonic acid, added to such medullary homogenates, can increase the quantities of prostaglandins synthesized. There was no evidence that the major prostaglandin biosynthesized, PGE2, was further metabolized to inactive products.  相似文献

The effects of morphine on the constancy of spontaneous contractions (isometric developed TENSION = IDT and contractile FREQUENCY = CF), in uterine strips isolated from ovariectomized rats and the influence of naloxone, were explored. The inotropic responses to added prostaglandins (PGs) E2 and F and the influences of morphine and of morphine in the presence of naloxone on PG actions, were also determined. Moreover, the synthesis and outputs of PGs E and F from uteri and the effects of morphine alone and of morphine plus naloxone, were studied. Morphine (10−6 M) significantly depressed uterine constancy of IDT during the first hours following delivery, but its action on CF did not differ from controls. Naloxone, neither at 10−8 M nor at 10−6 M, altered the negative inotropoic influence of morphine on IDT. Exogenous PGs E2 and F, stimulated uterine inotropism in a concentration-dependent fashion. Morphine altered dose-response curves for exogenous PGE2, evoking a parallel surmountable shift to the right, but did not affect the inotropic action of added PGF. This antagonistic effect of the opioid was not altered by preincubation with naloxone. Basal synthesis and outputs of PGs E and F in uteri from ovariectomized rats were significantly depressed by morphine (10−6 M) but not altered by incubating tissues with morphine in presence of naloxone. Results are discussed in terms of a presumptive dual action of morphine on uterine motility, i.e., antagonizing PGE2 receptors and inhibiting the synthesis of some PGs by the uterus. These influences of morphine do not appear to be subserved by the activation of μ opioid receptors. Moreover, the possibility that endogenous opioids could play a relevant role modulating uterine PG influences, is also discussed.  相似文献

Radioimmunoassays of platelet prostaglandins E1 and F in platelet rich plasma or platelet suspension, demonstrate that both PGE1 and PGF are present at higher concentrations than prostaglandins E2 and F. Gas chromatography — mass spectrometry determinations of prostaglandins E1 and E2 in resting washed platelets confirm this difference. Lastly, there is a greater incorporation of [1-14C] acetate into prostaglandins E1 and F compared to that into prostaglandins E2 and F.  相似文献

The behaviour of the pulmonary metabolites of prostaglandins E1, E2 and F were examined in several thin-layer chromatography (TLC) systems commonly used to differentiate parent prostaglandins. Although the systems chosen readily distinguished between a prostaglandin and its own metabolites, they often did not differentiate between a parent prostaglandin and the metabolites of another. In particular, 13, 14-dihydro-PGF was virtually indistinguishable from PGE2, and 13, 14-dihydro-PGE2 was similarly indistinguishable from PGE1, in all systems investigated. These pairs of prostaglandins could not be distinguished by bioassay on the rat stomach strip alone. Although distinction could be made by parallel assay on the rat stomach strip, chick rectum and rat colon, the differential assay obtained would not be enough to allow identification of these prostaglandins and metabolites in samples containing their mixtures. The 13, 14-dihydro-prostaglandin metabolites were also active in contracting the isolated rat uterus. The findings indicate that TLC and bioassay together may not permit identification of prostaglandins in biological fluids.  相似文献
