Studies have been made on the onset and regulation of the activity of sucrase and phosphofructokinase from different parts of the small intestine of chicks after hatching up to the 30th day of postnatal life. It was shown that within this period the activity of both enzymes undergoes synchronous changes. Carbohydrate loading increases the activity of sucrase only in 30-day chicks, its effect being similar to that of a stimulator from the duodenum. Changes in the activity of phosphofructokinase exhibit a different pattern being significantly dependent on the region of the small intestine.  相似文献

Protein deficiency in female rats diet during pregnancy and lactation resulted in deceleration of induction of sucrase both forms in the jejunum and ileum; in acceleration of induction of the maltase membrane from in the jejunum; and in suppression of the lactase membrane form in the ileum; in earlier forming of the adult-type distribution of activity of the membrane form of intestinal alkaline phosphatase and in a decrease in activity of the enzyme soluble form. The findings are corroborated by a suppression of activities of the membrane and soluble forms of the small intestine digestive enzymes in 30-day old rat pups fed with a control (adequate) ration starting 21 days after the birth.  相似文献

Activities of membranous and soluble forms of disaccharidases, alkaline phosphatase, and aminopeptidase M in jejunum and ileum was studied in the 10-, 20-, and 30-day old rat pups, whose mothers were kept at the period of pregnancy or lactation on a low-protein diet. The protein deficiency in mother's nutrition is shown to be accompanied by significant changes in the ratio of two forms of these membrane-bound enzymes. The greatest changes of this parameter are found for maltase, alkaline phosphatase, and aminopeptidase M in the 10-day old rat pups, while for lactase, in rat pups of all age groups. In some cases the maximal changes of the enzyme activity are revealed in rat pups, whose mothers were on the low-protein nutrition at the period of pregnancy (for example, saccharase), in other cases, in rat pups, whose mothers obtained such feeding during lactation (alkaline phosphatase), in the third ones, the changes were practically identical in the both groups of animals (maltase, while in some age groups, aminopeptidase M). The absence of changes in action of some modificators on activities of both forms of the studied enzymes allows suggesting that structure and properties of the studied enzyme proteins in the offspring's small intestine seem to remain unchanged under conditions of protein deficiency in mother's nutrition.  相似文献

We studied the responses in the omnivorous rodent A. azarae submitted to a low quality diet at morphological, physiological and biochemical levels. At short term, a decrease in body mass occurred. A later increase in food consumption constituted a strategy that allowed a temporal recovery of physical condition. However, hyperphagia appeared not to be enough to maintain physical condition after 30 days of low quality diet consumption. At the morphological level, an increase in length (9%) of the anterior portion of the gut occurred, the part of the gut where digestion and absorption take place. A decrease in small intestine weight could be related with the long-term impairment of body condition. Inhibition of sucrase specific activity in small intestine would indicate a down-regulation of sucrase-isomaltase complex. Total maltase specific activity in small intestine was not affected suggesting an up-regulation of sucrase-independent maltase specific activity. A down-regulation of protease specific activity in small intestine occurred in response to low quality diet. The specific activity of disaccharidases in caecum and large intestine was down-regulated. The strategies and constraints at different levels of A. azarae upon low quality diet are discussed.  相似文献

Myelin was isolated from the sciatic nerves of chicks of ages 18-day embryonic, 1-day, 4-day, 7-day post-hatch, and adult to study developmental changes in lipid composition of this structure. The yield of myelin increased throughout the early stages of development and the preparations were of high purity. Although the lipid content of the myelin did not change, significant changes took place in lipid composition during development. The most significant changes were a relative increase in cerebrosides, phosphatidalethanolamine and long-chain fatty acids of cerebrosides, and a relative decrease in the content of phosphatidylserine and phosphatidylethanolamine. A second fraction ("lower band") was obtained during the isolation procedure. This "lower band" was present at all developmental stages and layered consistently at the interface of 1.2 and 0.8 M sucrose on a discontinuous gradient. The quantity of this fraction did not change during development and it differed from myelin in electron microscopic appearance. Its lipid composition, which did not change, resembled that of 18-day embryonic myelin in its high phospholipid:cholesterol ratio and low galactolipid content. The enzyme, 2':3'-cyclic-nucleosidemonophosphate phosphodiesterase was found to be present in both the myelin and "lower band" fractions; however there was no enrichment of this enzyme in purified myelin.  相似文献

The methanolic extract of pasuchaca (Geranium dielsiaum) (PsEx) was found to suppress blood glucose elevation after oral administration of sucrose, maltose, and starch, but not after oral administration of glucose, in the mouse. In vitro examination of the inhibitory effect of PsEx on maltase activity revealed that PsEx strongly inhibited mouse small intestine maltase activity. Taken together, these results suggest that the inhibitory effect of PsEx on alpha-glucosidase activity might contribute to delay in carbohydrate digestion and subsequent lowering of the blood glucose level, thereby leading to prevention and cure of diabetes.  相似文献

The aim of the present work was to study the changes in the activity of disaccharidase enzymes (lactase. maltase, saccharase) in the small intestine of gnotobiotic pigs aged 0–35 days and inoculated with Enterococcus faecium. The continual decrease of lactase activity was observed from the 14th day of age up to the end of the experiment. The most significant decrease of specific lactase activity in the duodenum (2.1 μmol/mg protein/hour) was noted from the 21st to the 28th day of age. On the other hand, the specific saccharase activity increased moderately during the post weaning period and maltase activity maintained a constant level. We studied the occurrence of digestive strategies at different levels in females of the subterranean herbivorous rodent Ctenomys talarum living in their natural habitat. We determined the dimensions of different parts of the gastrointestinal tract and organs along as the activity of key digestive enzymes(sucrase, maltase and N-aminopeptidase) in small intestine in females seasonally caught. Females of C. talarum did not show seasonal variations in the mass of the different parts of the gastrointestinal tract. In nonreproductive females large intestine was longer in autumn, whereas reproductive females did not show seasonal variations in the length of the different parts of the gut. Females of C. talarum exhibited a high sucrose, maltase and N-aminopeptidase activity in small intestine, although these activities were higher in small intestine of females caught in autumn (nonreproductive) than in females caught in winter (reproductive). The results show that C. talarum females exhibit characteristics in the gut at the morphological and biochemical level, which could represent digestive strategies to face the constraints imposed by their costly particular way of life.  相似文献

The development of maltase, sucrase, and lactase activity has been examined in the small intestine of the mouse. After increasing during 2 days before birth, maltase remains unchanged for 14 days, after which activity surges up throughout the intestine. Sucrase is absent during the first 14 days, but then rises in a pattern similar to, but distinct from, that for maltase. Both enzymes rise faster in the proximal third of the intestine than in the terminal third. Lactase, which is high in the infant intestine, falls after 12 days in the proximal segment, but only after 16 days in the more posterior segments.Cortisone administered at 8 days causes a rise of maltase activity that continues for at least 72 hours. At 4 days the same treatment causes an increase that ceases after 48 hours. Sucrase activity is elicited by cortisone at 8 days but not at 4 days. Between 10 and 13 days both actinomycin D and cycloheximide evoke significant increases of both maltase and sucrase activity in all regions of the intestine. When administered in concert with cortisone, actinomycin D inhibits, but does not prevent, the stimulatory influence of the hormone on sucrase; with maltase activity, significant inhibition occurs only in the middle third of the intestine. Cycloheximide does not interfere with the effects of cortisone. No additive effects between hormone and antibiotics were obtained.These results are discussed in relation to results of similar studies on intestinal alkaline phosphatase and leucylnaphthylamidase.  相似文献

为明确晚成型小鼠胎后发育肠道消化酶活力的建立过程和发育模式,探讨其与适应性调节假说的关系,测定了从出生后至27日龄小鼠小肠前、中、后段的乳糖酶、蔗糖酶、麦芽糖酶和氨基肽酶的酶活力。结果发现单位组织酶活力方面,乳糖酶活力先增后降,小肠前段在9日龄而中后段在12日龄达到最高,至27日龄时仅中段有微弱的酶活力;蔗糖酶活力12日龄始出现,前段和后段自15日龄迅速升高,至18日龄达最高,但随后显著降低,而中段在15日龄后持续升高至21日龄达到最高,此后维持在较高水平;麦芽糖酶出生时已具有活力,但在15日龄前维持较低水平,此后迅速升高,前后段在18日龄,中段在21日龄达到峰值,此后下降;小肠前段的氨基肽酶活力出生后至27日龄持续下降,而后段和中段从出生到断乳前则持续升高,断乳后略有下降。除乳糖酶总酶活力先增后降,在15日龄达峰值外,其余3种酶的总酶活力均持续增加。在小肠不同位置4种酶活力的分布具有显著差异,且日龄对不同位置酶活力的影响趋势不同。总之,小鼠小肠4种消化酶的酶活力随时间的变化能够与其食物转变的消化需求相匹配,部分地支持适应性调节假说。  相似文献

Actinomycin D affects a number of functions of the epithelial cells of the small intestine. Maltase, saccharase and lactase levels in the small intestine of hamsters treated with various dosages of actinomycin D over various periods of time, differed from those observed in control animals: administration of 0.25 micrograms/g body weight, gave rise to a statistically significant increase in the maltase and saccharase levels measured after 4 h and a statistically significant reduction in the lactase levels measured after 8 h; administration of 1.5 micrograms/g body weight reduced the activity of all three enzymes at all times post-administration, the decrease being statistically significant for maltase after 2 and 8 h.  相似文献

The study on remote consequences of hypomagnetic conditions during roach Rutilus rutilus embryogenesis (at the stages of embryo and prelarva) revealed multidirectional changes in body length/weight, activities of glycosidases (maltase, sucrase, and amylolytic activity), and kinetic characteristics of carbohydrates hydrolysis in the intestine of the yearlings. The exposure of embryos to hypomagnetic conditions leads to the increase in body length and weight in the yearlings; the exposure of prelarvae leads to the decrease in these parameters. The impact of hypomagnetic conditions at the stage of prelarva results in most pronounced changes in the physiological and biochemical parameters in the yearlings. Decline in the Michaelis constant values, reflecting affinity of an enzyme to substrate, may be considered as an adaptive response of the fish digestive system to the lack of magnetic field during embryogenesis.  相似文献

It is revealed that administration of exogenous dexamethasone and thyroxin to the 7-8-day old rat pups produces different effect on activity and ratio of membrane and soluble forms of digestive enzymes (lactase, saccharase, maltase, alkaline phosphatase, aminopeptidase M, and glycyl-L-leucine dipeptidase) in jejunum and ileum of animals, whose mothers received different rations during lactation: the standard (control) and low-protein (experiment) ones. Disturbances of the hormonal regulation of the activity and the ratio of membrane and soluble forms of the digestive enzymes in small intestine in early ontogenesis consisted in that the effect of hormones was much less pronounced in the rat pups of the experimental group. An increase of the portion of the soluble form of the enzymes was also revealed. These results indicate a possibility of retardation of maturation of the intestinal digestive enzymes. The most significant difference of these effects in animals of the experimental and control groups was observed for saccharase, maltase, and peptidases.  相似文献

1. To characterize the development of peptide hydrolysis the activities of pancreatic carboxypeptidase A (CPA) and intestinal glycylleucine dipeptidase (GLDP) were registered in 1-45 days old chicks, as well as GLDP activities in newborn and adult guinea pigs. 2. The highest values of CPA and GLDP relative activities were found immediately after hatching, maximal decrease of activities took place during the first week. 3. GLDP activities gradient on the surface of the small intestine of chicks has two maximums: one in the upper jejunum, the other one--in the lower ileum. The development of proximo-distal gradient began at the age of 7 days and finished at the end of the first month. 4. Total CPA and GLDP activities decreased during the first week; up to the 15-20 day they reached the initial level and later exceeded it. 5. Relative GLDP activity in guinea pigs declined with aging, while the total activity increased, as it was demonstrated for chicks. 6. GLDP activity was distributed equally along the surface of the small intestine in newborn guinea pigs as well as in mature animals.  相似文献

1. The carbohydrase activities of homogenates of mucosa from the abomasum, small intestine, caecum and colon, and of the pancreas of cattle were studied. 2. The disaccharidase activities were located mainly in the small intestine and showed a non-uniform pattern of distribution along the small intestine; trehalase activity was highest in the proximal part, lactase and cellobiase activities were highest in the proximal and middle parts and maltase activity was highest in the distal part. 3. The intestinal lactase and cellobiase activities were highest in the young calf and decreased with age, whereas the intestinal maltase and trehalase activities, which were very low compared with the lactase activity, did not change with age. 4. No intestinal sucrase or palatinase activity was detected in the calf or in the adult cow. 5. Homogenates of intestinal mucosa also exhibited amylase and dextranase activity. 6. Homogenates of the pancreas possessed a strong amylase activity and a weak maltase activity. The maltase activity did not change with age, whereas the amylase activity increased with age. 7. No marked differences were observed between the carbohydrase activities of calves fed solely on milk and those of calves given a concentrate-hay diet from 6 weeks of age.  相似文献

Hypoxia in the neonate is known to alter the activity of hepatic and pancreatic enzymes involved in lipid and carbohydrate metabolism. The purpose of this study was to evaluate the effect of neonatal hypoxia on the activity of intestinal enzymes, and to determine whether the administration of glucocorticoids to neonates can mimic the effects of hypoxia. Hypoxia in neonatal rats (0-7 days) increased protein content, and lactase and maltase activity in the duodenal and the jejunal segments of the small intestine compared with normoxic controls. Hypoxia in juvenile rats (28-35 days) did not change these enzymes. Two weeks after returning hypoxic (0-7 days) pups to normoxia, their body weight remained lower than the age-matched controls. In the group recovering from hypoxia, sucrase, maltase, and leucine aminopeptidase activities were lower in the duodenal and the jejunal segment. Compared with controls, LDH activity was lower only in the jejunal intestine in the group recovering from hypoxia. All enzyme activities returned to control levels 3 weeks after recovery. Neonatal rats treated with dexamethasone had a decrease in body weight, but increases in sucrase and maltase activity in both the duodenal and the jejunal segment. Hypoxia in newborn rats caused a delayed maturation of small intestinal enzymes. Increases in serum glucocorticoids after hypoxic exposure probably do not play a major role in the delayed maturation of the disaccharidase activity in the small intestine.  相似文献

1. The maltase, sucrase, isomaltase and palatinase activities of the chick small intestine are localized in particles that sediment when centrifuged at 100000g for 90min. 2. Solubilization of the particle-bound disaccharidases without loss of activity was achieved by digestion with papain. Trypsin was less effective and caused a preferential solubilization of the sucrase, isomaltase and palatinase activities. 3. On Sephadex G-200 columns, the solubilized preparations yielded two disaccharidase peaks. The first peak was eluted close to the void volume of the column and contained all the sucrase, isomaltase and palatinase activities and some of the maltase activity. The remainder of the maltase activity was eluted beyond the total volume of the column. 4. Precipitation with ethanol did not affect the behaviour of the disaccharidases of gel filtration. 5. The maltase activity of the second peak on rechromatography in a buffer containing 0.01m-maltose was eluted close to the void volume. 6. Similar pH optima but different K(m) values were obtained for the maltase activities of the two peaks. 7. Heat-inactivation studies showed that the first peak contained two disaccharidase enzymes; one hydrolysed sucrose and maltose and the other hydrolysed isomaltose, palatinose and maltose. The second peak contained three disaccharidase enzymes all specific for the hydrolysis of maltose. 8. It is proposed that the intestinal disaccharidases of the chick exist in the form of two complexes: a sucrase-isomaltase complex and a maltase complex.  相似文献
