The absorption and scattering coefficients and reflectance spectra of ultra-high density Nannochloropsis occulata cultures were investigated in detail to identify the optical properties of the cultures and devise algorithms for remote estimation of dry cell mass in ultra-high cell density cultures. High-spectral resolution measurements of apparent absorption and attenuation as well as reflectance from 400 to 900 nm were carried out in relation to the dry weight, cell count, and pigment concentration in outdoor cultures. Indices calculated as (R(NIR) - R(red))/(R(NIR) + R(red)) and R(NIR)/R(red), in which R(NIR) is reflectance in the range from 750 to 800 nm and R(red) is reflectance in the range 670-680 nm, were used for remote assessment of dry cell mass. Remote estimation in the range 1 to 8 g/L was accomplished with an error of less than 0.66 g/L. A different index, i.e., (R(NIR) - R(red)) was employed for estimation of cell-chlorophyll concentration. This is the first report of in vivo specific absorption coefficient of chlorophyll-a and specific scattering coefficient per dry algal weight of Nannochloropsis sp., providing a basis for remote monitoring of dense phytoplankton masses.  相似文献

Red light and kinetin (10 µm) increased nitrite reductase(NIR) activity by 85 and 47% respectively in excised leavesof etiolated Zea mays. The stimulatory effect of kinetin decayedslower than that of red light. Indoleacetic acid (10 µm)had no effect on NIR activity. In the presence of abscisic acid(10 µm), the kinetin stimulated increase in NIR activitywas totally nullified, however, the red light irradiated plantsretained 20–25% increase in NIR activity over the darkcontrol. If ABA was given 2 h after kinetin treatment or redlight irradiation, it totally blocked kinetin stimulation asnoticed earlier, but red light stimulation was inhibited byonly 11%. Kinetin-treated and the red light irradiated leavesshowed 20–25% increase in nitrate accumulation, whichwas totally nullified by ABA. The Normalized Difference Vegetation Index (NDVI) is widely used for monitoring, analyzing, and mapping temporal and spatial distributions of physiological and biophysical characteristics of vegetation. It is well documented that the NDVI approaches saturation asymptotically under conditions of moderate-to-high aboveground biomass. While reflectance in the red region (rho(red)) exhibits a nearly flat response once the leaf area index (LAI) exceeds 2, the near infrared (NIR) reflectance (PNIR) continue to respond significantly to changes in moderate-to-high vegetation density (LAI from 2 to 6) in crops. However, this higher sensitivity of the rho(NIR) has little effect on NDVI values once the rho(NIR) exceeds 30%. In this paper a simple modification of the NDVI was proposed. The Wide Dynamic Range Vegetation Index, WDRVI = (a * rho(NIR-rho(red))/(a * rho(NIR) + rho(red)), where the weighting coefficient a has a value of 0.1-0.2, increases correlation with vegetation fraction by linearizing the relationship for typical wheat, soybean, and maize canopies. The sensitivity of the WDRVI to moderate-to-high LAI (between 2 and 6) was at least three times greater than that of the NDVI. By enhancing the dynamic range while using the same bands as the NDVI, the WDRVI enables a more robust characterization of crop physiological and phenological characteristics. Although this index needs further evaluation, the linear relationship with vegetation fraction and much higher sensitivity to change in LAI will be especially valuable for precision agriculture and monitoring vegetation status under conditions of moderate-to-high density. It is anticipated that the new index will complement the NDVI and other vegetation indices that are based on the red and NIR spectral bands.  相似文献

Light plays a pivotal role in animal orientation. Aquatic animals face the problem that penetration of light in water is restricted through high attenuation which limits the use of visual cues. In pure water, blue and green light penetrates considerably deeper than red and infrared spectral components. Submicroscopic particles and coloured dissolved organic matter, however, may cause increased scattering and absorption of short-wave components of the solar spectrum, resulting in a relative increase of red and infrared illumination. Here we investigated the potential of near-infrared (NIR) light as a cue for swimming orientation of the African cichlid fish (Cichlidae) Oreochromis mossambicus. A high-throughput semi-automated video tracking assay was used to analyse innate behavioural NIR-sensitivity. Fish revealed a strong preference to swim in the direction of NIR light of a spectral range of 850-950nm at an irradiance similar to values typical of natural surface waters. Our study demonstrates the ability of teleost fish to sense NIR and use it for phototactic swimming orientation.  相似文献

To establish new techniques for automatic classification of rhizosphere components, we investigated the utility of visible (VIS) and near–infrared (NIR) spectral images of the rhizosphere under two soil moisture conditions (mean volumetric water content: 0.39 and 0.16 cm3 cm?3). Spectral reflectance images of the belowground parts of hybrid poplar cuttings (Populus deltoides × P. euramericana, I45/51) grown in a rhizobox were recorded at 120 spectral bands ranging from 480 to 972 nm. We examined which wavelengths were suitable and the number of spectral bands needed to accurately classify live roots of four age classes, dead roots, leaf mold, and soil. VIS reflectance (<700 nm) of live roots first increased and then decreased with age, whereas NIR reflectance (≥700 nm) was stable in mature roots. The reflectance of dead roots was lower than that of mature roots in both the VIS and NIR spectral regions. VIS reflectance did not differ among dead roots, leaf mold, and soil, but the NIR reflectance was clearly lower in soil than in the other materials. The reflectance of leaf mold and soil increased mainly in the NIR spectral region with reducing soil moisture, but this increase did not affect the order of reflectance intensity among the rhizosphere components in general. Although the most suitable spectral bands statistically selected for classifying rhizosphere components differed somewhat between moist and dry conditions, the spectral bands 580–679 nm (VIS) and 848–894 nm (NIR) provided high reliability under both conditions. Classification accuracy was higher when using two to five VIS–NIR images (overall accuracy ≥87.8%) than three VIS images (red, green, and blue; accuracy <67.1%). The high accuracy with VIS–NIR was mainly due to successful separation of leaf mold and soil. Irrespective of soil moisture condition, the overall accuracy tended to be stable at 92–94% with use of four VIS–NIR images. The spectral bands effective in wet soil conditions could also be used for classification in dry conditions, with overall accuracies >86.9%. These results suggest that automatic image analysis using VIS–NIR images at four spectral bands, including red and NIR, allows for accurate classification of the growth stage or live/dead status of roots and distinguishes between leaf mold and soil.  相似文献

Pigments absorbing 350–1,050 nm radiation have had an important role on the Earth for at least 3.5 billion years. The ion pumping rhodopsins absorb blue and green photons using retinal and pump ions across cell membranes. Bacteriochlorophylls (BChl), absorbing in the violet/blue and near infra red (NIR), power anoxygenic photosynthesis, with one photoreaction centre; and chlorophylls (Chl), absorbing in the violet/blue and red (occasionally NIR) power oxygenic photosynthesis, with two photoreaction centres. The accessory (bacterio)chlorophylls add to the spectral range (bandwidth) of photon absorption, e.g., in algae living at depth in clear oceanic water and in algae and photosynthetic (PS) bacteria in microbial mats. Organism size, via the package effect, determines the photon absorption benefit of the costs of synthesis of the pigment–protein complexes. There are unresolved issues as to the evolution of Chls vs. BChls and the role of violet/blue and NIR radiation in PS bacteria.  相似文献

The anthocyanin and chlorophyll contents in leaves provide valuable information about the physiological status of plants. Thus, there is a need for accurate, efficient, and practical methodologies to estimate these biochemical parameters of vegetation. In this study, we tested the performance and accuracy of several nondestructive, reflectance-based techniques for estimating anthocyanin and chlorophyll contents in leaves of four unrelated species, European hazel (Corylus avellana), Siberian dogwood (Cornus alba =Swida alba), Norway maple (Acer platanoides), and Virginia creeper (Parthenocissus quinquefolia), with widely variable pigment content and composition. An anthocyanin reflectance index, which uses reflectances in the green and red edge spectral bands, and a modified anthocyanin reflectance index, employing, in addition, the near-infrared (NIR) band, were able to accurately estimate leaf anthocyanin for all species taken together with no reparameterization of algorithms. Total chlorophyll content was accurately estimated by a red edge chlorophyll index that uses spectral bands in the red edge and the NIR. These approaches can be used to estimate anthocyanin and chlorophyll nondestructively and allow the development of simple handheld field instrumentation.  相似文献

Light-mediated therapies such as photodynamic therapy (PDT) are considered emerging cancer treatment strategies. However, there are still lots of defect with common photosensitizers (PSs), such as short emission wavelength, weak photostability, poor cell permeability, and low PDT efficiency. Therefore, it is very important to develop high-performance PSs. Recently, luminogens with aggregation-induced emission (AIE) characteristics and red/near-infrared (NIR) emissive have been reported as promising PSs for image-guided cancer therapy, due to them being able to prevent autofluorescence in physiological environments, their enhanced fluorescence in the aggregated state, and generation of reactive oxygen species (ROS). Herein, we developed PSs named TBTCPM and MTBTCPM with donor–acceptor (D–A) structures, strong red/NIR, excellent targeting specificities to good cell permeability, and high photostability. Interestingly, both of them can efficiently generate ROS under white light irradiation and possess excellent killing effect on cancer cells. This study, thus, not only demonstrates applications in cell image-guided PDT cancer therapy performances but also provides strategy for construction of AIEgens with long emission wavelengths.  相似文献

Macrophages play a very important role in the conduction of several regenerative processes mainly due to their plasticity and multiple functions. In the muscle repair process, while M1 macrophages regulate the inflammatory and proliferative phases, M2 (anti‐inflammatory) macrophages direct the differentiation and remodelling phases, leading to tissue regeneration. The aim of this study was to evaluate the effect of red and near infrared (NIR) photobiomodulation (PBM) on macrophage phenotypes and correlate these findings with the repair process following acute muscle injury. Wistar rats were divided into 4 groups: control; muscle injury; muscle injury + red PBM; and muscle injury + NIR PBM. After 2, 4 and 7 days, the tibialis anterior muscle was processed for analysis. Macrophages phenotypic profile was evaluated by immunohistochemistry and correlated with the different stages of the skeletal muscle repair by the qualitative and quantitative morphological analysis as well as by the evaluation of IL‐6, TNF‐α and TGF‐β mRNA expression. Photobiomodulation at both wavelengths was able to decrease the number of CD68+ (M1) macrophages 2 days after muscle injury and increase the number of CD163+ (M2) macrophages 7 days after injury. However, only NIR treatment was able to increase the number of CD206+ M2 macrophages (Day 2) and TGFβ mRNA expression (Day 2, 4 and 7), favouring the repair process more expressivelly. Treatment with PBM was able to modulate the inflammation phase, optimize the transition from the inflammatory to the regeneration phase (mainly with NIR light) and improve the final step of regeneration, enhancing tissue repair.  相似文献

This study was conducted to investigate the efficacy of the simultaneous application of near-infrared (NIR) heating and UV irradiation for reducing populations of food-borne pathogens, including Salmonella enterica serovar Typhimurium and Escherichia coli O157:H7 in red pepper powder and to clarify the mechanisms of the lethal effect of the NIR-UV combined treatment. Also, the effect of the combination treatment on quality was determined by measuring changes in color and pungency constituents. Simultaneous NIR-UV combined treatment for 5 min achieved 3.34- and 2.78-log CFU reductions in S. Typhimurium and E. coli O157:H7, respectively, which involved 1.86- and 1.31-log CFU reductions, respectively, which were attributed to the synergistic effect. Through qualitative and quantitative analyses, damage to the cell envelope was identified as the main factor contributing to the synergistic lethal effect of NIR-UV combined treatment. Color values and capsaicin and dihydrocapsaicin content of NIR-UV simultaneously treated red pepper powder were not significantly (P > 0.05) different from those of untreated samples. These results suggest that simultaneous application of NIR and UV treatment can be effectively used to control food-borne pathogens in powdered red pepper without affecting quality.  相似文献

Nitrite reductase (NIR; EC 1.7.7.1) is a central enzyme in nitrate assimilation and is localized in plastids. The present study concerns the regulation of the appearance of NIR in cotyledons of the mustard (Sinapis alba L.) seedling. It was shown that light exerts its positive control over the nitrate-mediated induction of NIR via the farred-absorbing form of phytochrome. Without nitrate the light effect cannot express itself; even though the light signal is accumulated in the cotyledons it remains totally cryptic in the absence of nitrate. Moreover, it was recognised that intact plastids are important in the control of the appearance of NIR. If the plastids are damaged by photooxidation the action of nitrate and phytochrome on NIR appearance is abolished. The appearance of nitrate reductase (NR; EC 1.6.6.1) responds similarly to photooxidative damage even though this enzyme is cytosolic. Application of nitrate leads to an induction of nitrate reductase (NR; EC 1.6.6.1) and nitrite reductase (NIR; EC 1.7.7.1) in the cotyledons of dark-grown mustard (Sinapis alba L.) seedlings, and this induction can strongly be promoted by a far-red-light pretreatment — operating through phytochrome — prior to nitrate application. This light treatment is almost ineffective — as far as enzyme appearance is concerned — if no nitrate is given. When nitrate is applied, the stored light signal potentiates the appearance of NR and NIR in darkness, even in the absence of active phytochrome, to the same extent as continuous far-red light. This action of previously stored light signal lasts for approx. 12 h.Storage of the light signal was measured for NR and NIR. The process shows enzyme-specific differences. Storage occurs in the absence as well as in the presence of nitrate, i.e. irrespective of whether or not enzyme synthesis takes place. The kinetics of signal transduction and signal storage indicate that the formation and action of the stored signal are a bypass to the process of direct signal transduction. Non-destructive methods have been widely recognized for evaluating fruit quality traits of many horticultural crops and food processing industry. Destructive (analytical) test, and non-destructive evaluation of the quality traits were investigated and compared for ‘Red Rose’ tomato (Solanum lycopersicum L.) fruit grown under protected environment. Fresh tomato fruit at five distinctive maturity stages namely; breaker (BK), turning (TG), pink (PK), light-red (LR), and red (RD) were labeled and scanned using the handheld near infra-red (NIR) enhanced spectrometer at a wavelength range of 285–1200 nm. The labeled tomato samples were then measured analytically for flesh firmness, lycopene, β-carotene, total phenolic content (TPC) and total flavonoids content (TFC). The results revealed that quality traits could be estimated using NIR spectroscopy with a relatively high coefficient of determination (R2): 0.834 for total phenolic content, 0.864 for lycopene, 0.790 for total flavonoid content, 0.708 for β-carotene; and 0.679 for flesh firmness. The accumulation of Lyco and β-Car rapidly increased in tomatoes harvested between the TG and the LR maturity stages. Harvesting tomatoes at BK maturity stage resulted in significantly higher flesh firmness than harvesting at the later maturity stages. Tomato fruits had the lowest TPC and TFC contents at the earliest maturity stage (BK), while they had intermediate TPC and TFC levels at LR and RD maturity stages. NIR spectroscopic measurements of fruit firmness and lipophilic antioxidants in tomato fruit at various maturity stages were partially in accordance with those estimated by destructive (analytical) methods. Based on these findings, we recommend using non-destructive NIR spectroscopy as an effective tool for predicting tomato fruit quality during harvest stage and postharvest processing.  相似文献

The purpose of this research was to analyze and compare the responses of three Process Analytical Technology (PAT) techniques applied simultaneously to monitor a pilot-scale fluidized bed granulation process. Real-time measurements using focused beam reflectance measurement (Lasentec FBRM) and near-infra red spectroscopy (Bruker NIR) were taken by inserting in-line probes into the fluidized bed. Non-intrusive acoustic emission measurements (Physical Acoustic AE) were performed by attaching piezoelectric sensors on the external wall of the fluidized bed. Powder samples were collected at regular intervals during the granulation process and characterized offline using laser diffraction, scanning electron microscopy, stereo-optical microscopy and loss on drying method. PAT data comprising chord length distribution and chord count (from FBRM), absorption spectra (from NIR) and average signal levels and counts (from AE) were compared with the particle properties measured using offline samples. All three PAT techniques were able to detect the three granulation regimes or rate processes (wetting and nucleation, consolidation and growth, breakage) to varying degrees of sensitivity. Being dependent on optical signals, the sensitivities of the FBRM and NIR techniques were susceptible to fouling on probe windows. The AE technique was sensitive to background fluidizing air flows and external interferences. The sensitivity, strengths and weaknesses of the PAT techniques examined may facilitate the selection of suitable PAT tools for process development and scale-up studies.  相似文献

Photoacoustic imaging is a noninvasive imaging technique having the advantages of high‐optical contrast and good acoustic resolution at improved imaging depths. Light transport in biological tissues is mainly characterized by strong optical scattering and absorption. Photoacoustic microscopy is capable of achieving high‐resolution images at greater depth compared to conventional optical microscopy methods. In this work, we have developed a high‐resolution, acoustic resolution photoacoustic microscopy (AR‐PAM) system in the near infra‐red (NIR) window II (NIR‐II, eg, 1064 nm) for deep tissue imaging. Higher imaging depth is achieved as the tissue scattering at 1064 nm is lesser compared to visible or near infrared window‐I (NIR‐I). Our developed system can provide a lateral resolution of 130 μm, axial resolution of 57 μm, and image up to 11 mm deep in biological tissues. This 1064‐AR‐PAM system was used for imaging sentinel lymph node and the lymph vessel in rat. Urinary bladder of rat filled with black ink was also imaged to validate the feasibility of the developed system to study deeply seated organs.   相似文献

通过大田试验,测量了水稻不同生育期的冠层光谱,并通过在TM红光波段和近红外波段范围内不断扩展波段宽度,计算了各个波段宽度对应的归一化植被指数(NDVI)及其与叶面积指数(LAI)的最大相关系数,再根据NDVI与LAI最佳拟合方程的最大R2值随波段宽度的变化特征确定了最佳波段宽度.结果表明:NDVI与LAI的相关系数和最佳拟合方程的最大R2值具有相同的变化趋势,在小于红光波段宽度60nm范围内,均随波段宽度的增大而减小,即窄波段NDVI比宽波段NDVI对LAI的估算效果好;在超过60nm后,由于受近红外波段宽度的影响而略有波动.对R2规律的分析表明,使用NDVI估算水稻LAI的最佳波段宽度为15nm.  相似文献

The modern wine industry needs tools for process control and quality assessment in order to better manage fermentation or bottling processes. During wine fermentation it is important to measure both substrate and product concentrations (e.g. sugars, phenolic compounds), however, the analysis of these compounds by traditional means requires sample preparation and in some cases several steps of purification are needed. The combination of visible/near-infrared (Vis/NIR) spectroscopy and chemometrics potentially provides an ideal solution to accurately and rapidly monitor physical or chemical changes in wine during processing without the need for chemical analysis. The aim of this study was to assess the possibility of combining spectral and multivariate techniques, such as principal component analysis (PCA), discriminant partial least squares (DPLS), or linear discriminant analysis (LDA), to monitor time-related changes that occur during red wine fermentation. Samples (n = 652) were collected at various times from several pilot scale fermentations with grapes from either Cabernet Sauvignon or Shiraz varieties, over three vintages (2001-2003) and scanned using a monochromator instrument (Foss-NIRSystems 6500, Silver Spring, MD) in transmission mode (400-2,500 nm). PCA was used to demonstrate consistent progressive spectral changes that occur through the time course of the fermentation. LDA using PCA scores showed that regardless of variety or vintage, samples belonging to a particular time point in fermentation could be correctly classified. This study demonstrates the potential of Vis/NIR spectroscopy combined with chemometrics, as a tool for the rapid monitoring of red wine fermentation.  相似文献

Cyanobacteriochromes (CBCRs) are linear tetrapyrrole bilin-binding photoreceptors of cyanobacteria that exhibit high spectral diversity, gaining attention in optogenetics and bioimaging applications. Several engineering studies on CBCRs were attempted, especially for designing near-infrared (NIR) fluorescent proteins with longer fluorescence wavelengths. However, despite continuous efforts, a key component regulating fluorescence emission property in CBCRs is still poorly understood. As a model system, we focused on red/green CBCR Slr1393g3, from the unicellular cyanobacterium Synechocystis sp. PCC 6803 to engineer Pr to get far-red light-emitting property. Energy profiling and pairwise structural comparison of Slr1393g3 variants effectively reveal the mutations that are critical to the fluorescence changes. H497 seems to play a key role in stabilizing the chromophore environment, especially the α3 helix, while H495, T499, and Q502 are potential key residues determining fluorescence emission peak wavelength. We also found that mutations of α2 and α4 helical regions are closely related to the chromophore binding stability and likely affect fluorescence properties. Taken together, our computational analysis suggests that the fluorescence of Slr1393g3 is mainly controlled by the stabilization of the chromophore binding pocket. The predicted key residues potentially regulating the fluorescence emission property of a red/green CBCR will be advantageous for designing improved NIR fluorescent protein when combined with in vitro molecular evolution approaches.  相似文献

Spartina alterniflora (S. alterniflora) is a dominant invasive alien species that occurs in Yancheng Wetland National Nature Reserve, the largest coastal wetland in China. It expands rapidly and exerts great threats to local ecosystem. The main native species there are Phragmites australis (P. australis) and Suaeda salsa (S. salsa), respectively. In order to monitor their dynamics, it is of great significance to analyze their spectral discrimination. Canopy spectra of these typical species were measured in July and October in order to compare species differences, as well as the seasonal variation. The Normalized Difference Vegetation Index (NDVI) was calculated based on canopy spectra. The significances of differences in spectral characteristic among species were tested using the one-way analysis of variance (ANOVA) (P < 0.05). The results showed that the visible (VI) bands were the optimum wavelengths for species discrimination in both seasons. S. alterniflora always had the greatest green peak height and red absorption depth. S. salsa had no obvious green peak, but an obvious red reflection peak. P. australis generally had intermediate values. Spectra in the near-infrared (NIR) bands were not appropriate for delineating S. alterniflora and P. australis in summer, as they showed similar values in these wavelengths. But NIR bands could be used to distinguish S. salsa in summer, as it had significantly lower reflectance in NIR bands than the others. Meanwhile, the reflectance in short-wavelength infrared (SWIR) bands became another suitable approach for distinguishing species in autumn. S. salsa had significantly higher values than the others in SWIR regions. Significant NDVI differences between different species proved that it could improve the species discrimination. S. alterniflora always had the highest NDVI values, while S. salsa had the lowest values. The seasonal trend of canopy spectra was also revealed. With plant maturity, the reflectance values in green bands and NIR bands decreased significantly, but increased significantly around the yellow-red wavelengths. The green peak heights, red absorption depths, and NDVI values of the species decreased remarkably. Furthermore, S. alterniflora and P. australis illustrated obvious ‘blue shift’ of the red edge with senescence. These results pointed out the the potential bands and appropriate spectral parameters appropriate for species discrimination, and highlighted the influence of seasonality on spectral information. Spectral changes with leaf aging in Amazon caatinga   总被引:1,自引:1,他引:0  
 Significant gaps exist in the knowledge of tropical leaf spectra and the manner in which spectra change as leaves age in their natural environment. Leaf aging effects may be particularly important in tropical vegetation growing on nutrient poor soils, such as Amazon caatinga, a white sand community common in the Amazon Basin. Spectral changes observed in six caatinga dominants include decreased reflectance and transmittance and increased absorptance for epiphyll-coated older leaves. Near-infrared (NIR) changes were most significant. More detailed spectral and physical changes were studied in one dominant, Aldina heterophylla. Over 16 months, Aldina study plants produced one or two leaf flushes. During leaf expansion, leaf water content and Specific Leaf Area decreased rapidly. Over the first 6 months spectral changes occurred across the spectrum, resulting in decreased transmittance and increased absorptance in the visible and NIR and decreased visible and increased NIR reflectance. In contrast, significant spectral changes were restricted to the NIR over the last 9 months, which showed a 10% absorptance increase associated primarily with increasing epiphylls and necrosis. At the canopy scale, increased NIR absorptance provides a mechanism for producing seasonally varying forest albedo and changing NIR to red ratios, independent of changes in other canopy attributes. In the Amazon caatinga studied, all canopy dominants were subject to epiphyllic growth providing a mechanism for distinguishing these forest types spectrally from more diverse terra-firme forest or forest types with more rapid leaf turnover, such as second growth. These changes are observable using remote sensing and could be used to map caatinga and monitor interannual or seasonal variability in phenology. If these results can be extended to other communities with long-lived foliage, they may offer a means for mapping vegetation on the basis of leaf longevity.
