Two rice chlorophyll (Chl) b-less mutants (VG28-1, VG30-5) and the respective wild type (WT) plant (cv. Zhonghua No. 11) were analyzed for the changes in Chl fluorescence parameters, xanthophyll cycle pool, and its de-epoxidation state under exposure to strong irradiance, SI (1 700 μmol m−2 s−1). We also examined alterations in the chloroplast ultrastructure of the mutants induced by methyl viologen (MV) photooxidation. During HI (0–3.5 h), the photoinactivation of photosystem 2 (PS2) appeared earlier and more severely in Chl b-less mutants than in the WT. The decreases in maximal photochemical efficiency of PS2 in the dark (Fv/Fm), quantum efficiency of PS2 electron transport (ΦPS2), photochemical quenching (qP), as well as rate of photochemistry (Prate), and the increases in de-epoxidation state (DES) and rate of thermal dissipation of excitation energy (Drate) were significantly greater in Chl b-mutants compared with the WT plant. A relatively larger xanthophyll pool and 78–83 % conversion of violaxanthin into antheraxanthin and zeaxanthin in the mutants after 3.5 h of HI was accompanied with a high ratio of inactive/total PS2 (0.55–0.73) and high 1–qP (0.57–0.68) which showed that the activities of the xanthophyll cycle were probably insufficient to protect the photosynthetic apparatus against photoinhibition. No apparent difference of chloroplast ultrastructure was found between Chl b-less mutants and WT plants grown under low, LI (180 μmol m−2 s−1) and high, HI (700 μmol m−2 s−1) irradiance. However, swollen chloroplasts and slight dilation of thylakoids occurred in both mutants and the WT grown under LI followed by MV treatment. These typical symptoms of photooxidative damage were aggravated as plants were exposed to HI. Distorted and loose scattered thylakoids were observed in particular in the Chl b-less mutants. A greater extent of photoinhibition and photooxidation in these mutants indicated that the susceptibility to HI and oxidative stresses was enhanced in the photosynthetic apparatus without Chl b most likely as a consequence of a smaller antenna size.  相似文献

Seeds of Suaeda salsa were cultured in dark for 3 d and betacyanin accumulation in seedlings was promoted significantly. Then the seedlings with accumulated betacyanin (C+B) were transferred to 14/10 h light/dark and used for chilling treatment 15 d later. Photosystem 2 (PS2) photochemistry, D1 protein content, and xanthophyll cycle during the chilling-induced photoinhibition (exposed to 5 °C at a moderate photon flux density of 500 μmol m−2 s−1 for 3 h) and the subsequent restoration were compared between the C+B seedlings and the control (C) ones. The maximal efficiency of PS2 photochemistry (Fv/Fm), the efficiency of excitation energy capture by open PS2 centres (Fv′/Fm′), and the yield of PS2 electron transport (ΦPS2) of the C+B and C leaves both decreased during photoinhibition. However, smaller decreases in Fv/Fm, Fv′/Fm′, and ΦPS2 were observed in the C+B leaves than in C ones. At the same time, the deepoxidation state of xanthophyll cycle, indicated by (A+Z)/(V+A+Z) ratio, increased rapidly but the D1 protein content decreased considerably during the photoinhibition. The increase in rate of (A+Z)/(V+A+Z) was higher but the D1 protein turnover was slower in C+B than C leaves. After photoinhibition treatment, the plants were transferred to a dim irradiation (10 μmol m−2 s−1) at 25 °C for restoration. During restoration, the chlorophyll (Chl) fluorescence parameters, D1 protein content, and xanthophyll cycle components relaxed gradually, but the rate and level of restoration in the C+B leaves was greater than those in the C leaves. The addition of betacyanins to the thylakoid solution in vitro resulted in similar changes of Fv/Fm, D1 protein content, and (A+Z)/(V+A+Z) ratio during the chilling process. Therefore, betacyanin accumulation in S. salsa seedlings may result in higher resistance to photoinhibition, larger slowing down of D1 protein turnover, and enhancement of non-radiative energy dissipation associated with xanthophyll cycle, as well as in greater restoration after photoinhibition than in the control when subjected to chilling at moderate irradiance.  相似文献

The effects of temperature, irradiance, and daylength on Sargassum horneri growth were examined at the germling and adult stages to discern their physiological differences. Temperature–irradiance (10, 15, 20, 25, 30°C × 20, 40, 80 μmol photons m−2s−1) and daylength (8, 12, 16, 24 h) experiments were carried out. The germlings and blades of S. horneri grew over a wide range of temperatures (10–25°C), irradiances (20–80 μmol photons m−2s−1), and daylengths (8–24 h). At the optimal growth conditions, the relative growth rates (RGR) of the germlings were 21% day−1 (25°C, 20 μmol photons m−2s−1) and 13% day−1 (8 h daylength). In contrast, the RGRs of the blade weights were 4% day−1 (15°C, 20 μmol photons m−2s−1) and 5% day−1 (12 h daylength). Negative growth rates were found at 20 μmol photons m−2s−1 of 20°C and 25°C treatments after 12 days. This phenomenon coincides with the necrosis of S. horneri blades in field populations. In conclusion, we found physiological differences between S. horneri germlings and adults with respect to daylength and temperature optima. The growth of S. horneri germlings could be enhanced at 25°C, 20 μmol photons m−2s−1, and 8 h daylength for construction of Sargassum beds and restoration of barren areas.  相似文献

The ability of spring barley (Hordeum vulgare cv. Akcent) to adjust the composition and function of the photosynthetic apparatus to growth irradiances of 25–1200 μmol m−2 s−1 was studied by gas exchange and chlorophyll a fluorescence measurements and high-performance liquid chromatography. The increased growth irradiance stimulated light- and CO2-saturated rates of CO2 assimilation expressed on a leaf area basis up to 730 μmol m−2 s−1 (HL730), whereas at an irradiance of 1200 μmol m−2 s−1 (EHL1200) both rates decreased significantly. Further, the acclimation to EHL1200 was associated with an extremely high chlorophyll a/b ratio (3.97), a more than doubled xanthophyll cycle pool (VAZ) and a six-fold higher de-epoxidation state of the xanthophyll cycle pigments as compared to barley grown under 25 μmol m−2 s−1 (LL25). EHL1200 plants also exhibited a long-term inhibition of Photosystem II (PS II) photochemical efficiency (F v/F m). Photosynthetic capacity, chlorophyll a/b and VAZ revealed a linear trend of dependence on PS II excitation pressure in a certain range of growth irradiances (100–730 μmol m−2 s−1). The deviation from linearity of these relationships for EHL1200 barley is discussed. In addition, the role of increased VAZ and/or accumulation of zeaxanthin and antheraxanthin in acclimation of barley to high irradiance is studied with respect to regulation of non-radiative dissipation and/or photochemical efficiency within PS II. Using 77 K chlorophyll a (Chl a) fluorescence spectra in vivo, the development was studied of Photosystems II (PS II) and I (PS I) during greening of barley under intermittent light followed by continuous light at low (LI, 50 μmol m−2 s−1) and high (HI, 1000 μmol m−2 s−1) irradiances. The greening at HI intermittent light was accompanied with significantly reduced fluorescence intensity from Chl b excitation for both PS II (F685) and PS I (F743), in comparison with LI plants, indicating that assembly of light-harvesting complexes (LHC) of both photosystems was affected to a similar degree. During greening at continuous HI, a slower increase of emission from Chl b excitation in PS II as compared with PS I was observed, indicating a preferred reduction in the accumulation of LHC II. The following characteristics of 77 K Chl a fluorescence spectra documented the photoprotective function of an elevated content of carotenoids in HI leaves: (1) a pronounced suppression of Soret region of excitation spectra (410–450 nm) in comparison with the red region (670–690 nm) during the early stage of greening indicated a strongly reduced excitation energy transfer from carotenoids to the Chl a fluorescing forms within PS I and PS II; (2) changes in the shape of the excitation band of Chl b and carotenoids (460–490 nm) during greening under continuous light confirmed that the energy transfer from carotenoids to Chl a within PS II remained lower as compared with the LI plants. The extent of mercury (Hg) toxicity in the heterocystous cyanobacterium Nostoc muscorum grown for 72 h in three different light intensities was tested for various physiological parameters viz. growth, pigment contents, photosynthesis, respiration, reactive oxygen species (ROS), malondialdehyde formation and antioxidants. A general reduction in growth and pigments, whole cell O2-evolution, photosynthetic electron transport activities and 14CO2-fixation was observed in a metal concentration–dependent manner, and this effect was more pronounced in high light (130 μmol photon m−2 s−1)–exposed cells as compared to low (10 μmol photon m−2 s−1) and normal (70 μmol photon m−2 s−1) light intensity–exposed cells; however, carotenoids and respiration showed reverse trend. Among photosynthetic electron transport activities, whole chain activity was found to be most sensitive in comparison with photosystem II (PS II) and photosystem I (PS I). Comparing the different photosynthetic processes, 14CO2-fixation was most affected in cyanobacterial cells when exposed to Hg and different light intensities. After application of various exogenous electron donors, diphenyl carbazide was found to be more effective to restore PS II activity, suggesting that site of damage lies in between oxygen evolving complex and PS II. Level of oxidative stress (superoxide radical and lipid peroxidation) was maximum at 3.0 μM of Hg when coupled with high light intensity (except hydrogen peroxide). A dose-dependent increase in enzymatic antioxidants such as superoxide dismutase, peroxidase and catalase as well as non-enzymatic antioxidants such as proline, ascorbate, cysteine (except under high light intensity) and non-protein thiols [NP-SH] was observed, which further increased with the increase in light intensity. It was noticed that Hg intoxicates N. muscorum through ROS production, which is aggravated along with the increase in light intensity. Overall results suggest that the severity of the metal stress does increase with Hg concentrations but when coupled with light, it was the light intensity that determines the extent of Hg toxicity.  相似文献

Photoinhibition under irradiance of 2 000 μmol m−2 s−1 (HI) was studied in detached control (C) and water deficit (WD) leaves of grapevine (Vitis vinifera L.) plants. The degree of photoinhibition was determined by means of the ratio of variable to maximum chlorophyll (Chl) fluorescence (Fv/Fm) and electron transport measurements. The potential efficiency of photosystem (PS) 2, Fv/Fm, marginally declined under HI in WD-leaves without significant increase of F0. In contrast, Fv/Fm ratio declined markedly with significant increase of F0 in C-leaves. In isolated thylakoids, the rate of whole chain and PS2 activity under HI were more decreased in C-than WD-leaves. The artificial exogenous electron donors diphenyl carbazide, NH2OH, and Mn2+ failed to restore the HI-induced loss of PS2 activity in both C-and WD-leaves. Thus HI operates at the acceptor side of PS2 in both leaf types. Quantification of the PS2 reaction centre protein D1 following HI exposure of leaves showed pronounced differences between C-and WD-leaves. The marked loss of PS2 activity under HI of C-leaves was due to the marked loss of D1 protein of the PS2 reaction centre.  相似文献

Interferon-alpha (IFN-α) is an immunomodulatory cytokine that is used clinically for the treatment of melanoma in the adjuvant setting. The cellular actions of IFN-α are regulated by the suppressors of cytokine signaling (SOCS) family of proteins. We hypothesized that the anti-tumor activity of exogenous IFN-α would be enhanced in SOCS1-deficient mice. SOCS1-deficient (SOCS1−/−) or control (SOCS1+/+) mice on an IFN-γ−/− C57BL/6 background bearing intraperitoneal (i.p.) JB/MS murine melanoma cells were treated for 30 days with i.p. injections of IFN-A/D or PBS (vehicle). Log-rank Kaplan-Meier survival curves were used to evaluate survival. Tumor-bearing control SOCS1+/+ mice receiving IFN-A/D had significantly enhanced survival versus PBS–treated mice (P = 0.0048). The anti-tumor effects of IFN-A/D therapy were significantly enhanced in tumor-bearing SOCS1−/− mice; 75% of these mice survived tumor challenge, whereas PBS-treated SOCS1−/− mice all died at 13-16 days (P = 0.00038). Antibody (Ab) depletion of CD8+ T cells abrogated the anti-tumor effects of IFN-A/D in SOCS1−/− mice as compared with mice receiving a control antibody (P = 0.0021). CD4+ T-cell depletion from SOCS1−/− mice also inhibited the effects of IFN-A/D (P = 0.0003). IFN-A/D did not alter expression of CD80 or CD86 on splenocytes of SOCS1+/+ or SOCS1−/− mice, or the proportion of T regulatory cells or myeloid-derived suppressor cells in SOCS1+/+ or SOCS1−/− mice. An analysis of T-cell function did reveal increased proliferation of SOCS1-deficient splenocytes at baseline and in response to mitogenic stimuli. These data suggest that modulation of SOCS1 function in T-cell subsets could enhance the anti-tumor effects of IFN-α in the setting of melanoma.  相似文献

The responses of the early development of Laminaria japonica collected from Kiaochow Bay in China to enhanced ultraviolet-B radiation (UV-B, 280–320 nm) were studied in the laboratory. The low UV-B radiations (11.7–23.4 J·m−2·d−1) had no significant effects on zoospores attachment, but when the UV-B dose > 35.1 J·m−2·d−1 the attachment decreased significantly compared with the control. Germination of embryospores was >93% under the low (11.7–35.1 J·m−2·d−1) doses, and in the range of 78.5%–88.5% under the high (46.8–70.2 J·m−2·d−1) UV-B doses, indicating a significant radiation effect. Under the higher UV-B exposure (35.1–70.2 J·m−2·d−1), all of the few gametophytes formed from embryospores died 120 h post-release. After exposure to the low UV-B radiation (11.7–23.4 J·m−2·d−1), the formation of sporophytes decreased and the female gametophyte clones increased compared with the control. However, the sex ratio and the relative growth of female gametophytes/sporophytes had not significantly changed. According to the results, enhanced UV-B radiation has a significant effect on the early development of L. japonica under laboratory conditions, suggesting that the UV-B radiation could not be overlooked as one of the important environmental factors influencing the ontogeny of macroalgae living in marine ecosystems. Rates of net photosynthesis (P N) and transpiration (E), and leaf temperature (TL) of maintenance leaves of tea under plucking were affected by photosynthetic photon flux densities (PPFD) of 200–2 200 μmol m−2 s−1. P N gradually increased with the increase of PPFD from 200 to 1 200 μmol m−2 s−1 and thereafter sharply declined. Maximum P N was 13.95 μmol m−2 s−1 at 1 200 μmol m−2 s−1 PPFD. There was no significant variation of P N among PPFD at 1 400–1 800 μmol m−2 s−1. Significant drop of P N occurred at 2 000 μmol m−2 s−1. PPFD at 2 200 μmol m−2 s−1 reduced photosynthesis to 6.92 μmol m−2 s−1. PPFD had a strong correlation with TL and E. Both TL and E linearly increased from 200 to 2 200 μmol m−2 s−1 PPFD. TL and E were highly correlated. The optimum TL for maximum P N was 26.0 °C after which P N declined significantly. E had a positive correlation with P N.  相似文献

Photosynthetic properties of carnivorous plants have not been well characterized and the extent to which photosynthesis contributes to carbon gain in most carnivorous plants is also largely unknown. We investigated the photosynthetic light response in three carnivorous plant species, Drosera rotundifolia L. (sundew; circumpolar and native to northern British Columbia, Canada), Sarracenia leucophylla Rafin. (‘pitcher-plant’; S.E. United States), and D. capensis L. (sundew; Cape Peninsula, South Africa), using portable gas-exchange systems to explore the capacity for photosynthetic carbon gain in carnivorous plant species. Maximal photosynthetic rates (1.32–2.22 μmol m−2 s−1 on a leaf area basis) and saturating light intensities (100 to 200 μmol PAR m−2 s−1) were both low in all species and comparable to shade plants. Field or greenhouse-grown D. rotundifolia had the highest rates of photosynthesis among the three species examined. Dark respiration, ranging from −1.44 (S. leucophylla) to −3.32 (D. rotundifolia) μmol m−2 s−1 was high in comparison to photosynthesis in the species examined. Across greenhouse-grown plants, photosynthetic light compensation points scaled with light-saturated photosynthetic rates. An analysis of gas-exchange and growth data for greenhouse-grown D. capensis plants suggests that photosynthesis can account for all plant carbon gain in this species.  相似文献

The relation between oxygen evolution rate (OER) and quantum yield of photochemical reactions in photosystem 2 (ΦPS2) was examined in lichen symbiotic alga Trebouxia erici Ahmadjian (strain UTEX 911) exposed to different irradiances and osmotic stress (2 M sucrose for 60 h). Linear relationship was found between OER and ΦPS2 in control cell suspension within irradiance range of 0 – 500 μmol m−2 s−1. Under osmotic stress, OER and ΦPS2 were significantly reduced. Relation between OER and ΦPS2 was curvilinear due to strong osmotically-induced inhibition of OER at high irradiance. The highest used irradiance (500 μmol m−2 s−1) was photoinhibitory for osmotically-stressed T. erici because non-photochemical quenching (NPQ) increased substantially. Energy-dependent quenching represented major part of NPQ increase. Osmotic stress led also to the reduction of capacity of photochemical processes in PS 2 (FV/FM) and increase in F0/FM. These changes indicated negative effects of osmoticum on structure and function of photosynthetic apparatus.  相似文献

Citrus volkameriana (L.) plants were grown for 43 d in nutrient solutions containing 0, 2, 14, 98, or 686 μM Mn (Mn0, Mn2, Mn14, Mn98, and Mn686, respectively). To adequately investigate the combined effects of Mn nutrition and irradiance on photosystem 2 (PS2) activity, irradiance response curves for electron transport rate (ETR), nonphotochemical quenching (qN), photochemical quenching (qP), and real photochemical efficiency of PS2 (ΦPS2) were recorded under 10 different irradiances (66, 96, 136, 226, 336, 536, 811, 1 211, 1 911, and 3 111 μmol m−2 s−1, I66 to I3111, respectively) generated with the PAM-2000 fluorometer. Leaf chlorophyll content was significantly lower under Mn excess (Mn686) compared to Mn0; its highest values were recorded in the treatments Mn2-Mn98. However, ETR and ΦPS2 values were significantly lower under Mn0 compared to the other Mn treatments, when plants were exposed to irradiances ≥96 μmol m−2 s−1. Furthermore, Mn0 plants had significantly higher values of qN and lower values of qP at irradiances ≤226 and ≥336 μmol m−2 s−1, respectively, than those grown under Mn2-Mn686. Irrespective of Mn treatment, the values of ΦPS2 and qN decreased, while those of qP increased progressively by increasing irradiance from I136 to I3111. Finally, Mn2-Mn98 plants were less sensitive to photoinhibition of photosynthesis (≥811 μmol m−2 s−1) than the Mn686 (≥536 μmol m−2 s−1) and Mn0 (≥336 μmol m−2 s−1) ones.  相似文献

Photosynthetic parameters, growth, and pigment contents were determined during expansion of the fourth leaf of in vitro photoautotrophically cultured Nicotiana tabacum L. plants at three irradiances [photosynthetically active radiation (400–700 nm): low, LI 60 μmol m−2 s−1; middle, MI 180 μmol m−2 s−1; and high, HI 270 μmol m−2 s−1]. During leaf expansion, several symptoms usually accompanying leaf senescence appeared very early in HI and then in MI plants. Symptoms of senescence in developing leaves were: decreasing chlorophyll (Chl) a+b content and Chl a/b ratio, decreasing both maximum (FV/FM) and actual (ΦPS2) photochemical efficiency of photosystem 2, and increasing non-photochemical quenching. Nevertheless, net photosynthetic oxygen evolution rate (P N) did not decrease consistently with decrease in Chl content, but exhibited a typical ontogenetic course with gradual increase. P N reached its maximum before full leaf expansion and then tended to decline. Thus excess irradiance during in vitro cultivation did not cause early start of leaf senescence, but impaired photosynthetic performance and Chl content in leaves and changed their typical ontogenetic course.  相似文献

A yellow leaf colouration mutant (named ycm) generated from rice T-DNA insertion lines was identified with less grana lamellae and low thylakoid membrane protein contents. At weak irradiance [50 μmol(photon) m−2 s−1], chlorophyll (Chl) contents of ycm were ≈20 % of those of WT and Chl a/b ratios were 3-fold that of wild type (WT). The leaf of ycm showed lower values in the actual photosystem 2 (PS2) efficiency (ΦPS2), photochemical quenching (qP), and the efficiency of excitation capture by open PS2 centres 1 (Fv′/Fm′) than those of WT, except no difference in the maximal efficiency of PS2 photochemistry (Fv/Fm). With progress in irradiance [100 and 200 μmol(photon) m−2 s−1], there was a change in the photosynthetic pigment stoichiometry. In ycm, the increase of total Chl contents and the decrease in Chl a/b ratio were observed. ΦPS2, qP, and Fv′/Fm′ of ycm increased gradually along with the increase of irradiance but still much less than in WT. The increase of xanthophyll ratio [(Z+A)/(V+A+Z)] associated with non-photochemical quenching (qN) was found in ycm which suggested that ycm dissipated excess energy through the turnover of xanthophylls. No significant differences in pigment composition were observed in WT under various irradiances, except Chl a/b ratio that gradually decreased. Hence the ycm mutant developed much more tardily than WT, which was caused by low photon energy utilization independent of irradiance.  相似文献

Fast cyclic electron transport (CET) around photosystem I (PS I) was observed in sunflower (Helianthus annuus L.) leaves under intense far-red light (FRL) of up to 200 μmol quanta m−2 s−1. The electron transport rate (ETR) through PS I was found from the FRL-dark transmittance change at 810 and 950 nm, which was deconvoluted into redox states and pool sizes of P700, plastocyanin (PC) and cytochrome f (Cyt f). PC and P700 were in redox equilibrium with K e = 35 (ΔE m = 90 mV). PS II ETR was based on O2 evolution. CET [(PS I ETR) − (PS II ETR)] increased to 50–70 μmol e m−2 s−1 when linear electron transport (LET) under FRL was limited to 5 μmol e m−2 s−1 in a gas phase containing 20–40 μmol CO2 mol−1 and 20 μmol O2 mol−1. Under these conditions, pulse-saturated fluorescence yield F m was non-photochemically quenched; however, F m was similarly quenched when LET was driven by low green or white light, which energetically precluded the possibility for active CET. We suggest that under FRL, CET is rather not coupled to transmembrane proton translocation than the CET-coupled protons are short-circuited via proton channels regulated to open at high ΔpH. A kinetic analysis of CET electron donors and acceptors suggests the CET pathway is that of the reversed Q-cycle: Fd → (FNR) → Cyt cn → Cyt bh → Cyt bl → Rieske FeS → Cyt f → PC → P700 →→ Fd. CET is activated when PQH2 oxidation is opposed by high ΔpH, and ferredoxin (Fd) is reduced due to low availability of e acceptors. The physiological significance of CET may be photoprotective, as CET may be regarded as a mechanism of energy dissipation under stress conditions.  相似文献

The effect of light intensity (50–300 μmol photons m−2 s−1) and temperature (15–50°C) on chlorophyll a, carotenoid and phycobiliprotein content in Arthronema africanum biomass was studied. Maximum growth rate was measured at 300 μmol photons m−2 s−1 and 36°C after 96 h of cultivation. The chlorophyll a content increased along with the increase in light intensity and temperature and reached 2.4% of dry weight at 150 μmol photons m−2 s−1 and 36°C, but it decreased at higher temperatures. The level of carotenoids did not change significantly under temperature changes at illumination of 50 and 100 μmol photons m−2 s−1. Carotenoids were about 1% of the dry weight at higher light intensities: 150 and 300 μmol photons m−2 s−1. Arthronema africanum contained C-phycocyanin and allophycocyanin but no phycoerythrin. The total phycobiliprotein content was extremely high, more than 30% of the dry algal biomass, thus the cyanobacterium could be deemed an alternative producer of C-phycocyanin. A highest total of phycobiliproteins was reached at light intensity of 150 μmol photons m−2 s−1 and temperature of 36°C, C-phycocyanin and allophycocyanin amounting, respectively, to 23% and 12% of the dry algal biomass. Two 60-day experiments were conducted to study the influence of photon flux density (PFD) and temperature on the attachment and development of Gloiopeltis tenax and Gloiopeltis furcata tetraspores. In the first experiment, tetraspores of the two Gloiopeltis species were incubated at five temperature ranges (8°C, 12°C, 16°C, 20°C, 24°C) under a constant PFD of 80 μmol photons m−2 s−1 with a photoperiod of 12:12. In a second experiment, tetraspores were incubated under five PFD gradients (30, 55, 80, 105, 130 μmol photons m−2 s−1) at a constant temperature of 16°C with a photoperiod of 12:12. Maximum density of attached tetraspores was observed at 16°C for both species. Maximum per cent of spore germinating into disc was recorded at 12–16°C for G. tenax and 8–12°C for G. furcata. Maximum per cent of discs producing erect axes for G. tenax and G. furcata were recorded at 24°C and 20°C, respectively. Light had no significant effect on tetraspore attachment and developing into disc, but it affected the growth, sprouting and survival of its discs. Under 30–55 μmol photons m−2 s−1, the discs of the two species of Gloiopeltis did not form thallus until the end of the experiment. Optimum PFD range for G. tenax discs was 80–105 μmol photons m−2 s−1, whilst it was 80–130 μmol photons m−2 s−1 for G. furcata. Results presented in this study are expected to assist the progress of artificial seeding of Gloiopeltis.  相似文献

Symbiotic dinoflagellates of the species Amphidinium are expected to be pharmaceutically useful microalgae because they produce antitumor macrolides. A microalgae production system with a large number of cells at a high density has been developed for the efficient production of macrolide compounds. In the present study, the effects of culture conditions on the cellular growth rate of dinoflagellates were investigated to determine the optimum culture conditions for obtaining high yields of microalgae. Amphidinium species was cultured under conditions with six temperature levels (21–35°C), six levels of photosynthetic photon flux density (15–70 μmol photons m−2 s−1), three levels of CO2 concentration (0.02–0.1%), and three levels of O2 concentration (0.2–21%). The number of cells cultured in a certain volume of solution was monitored microscopically and the cellular growth rate was expressed as the specific growth rate. The maximum specific growth rate was 0.022 h−1 at a temperature of 26°C and O2 concentration of 5%, and the specific growth rate was saturated at a CO2 concentration of 0.05%, a photosynthetic photon flux density of 35 μmol photons m−2 s−1 and a photoperiod of 12 h day−1 upon increasing each environmental parameter. The results demonstrate that Amphidinium species can multiply efficiently under conditions of relatively low light intensity and low O2 concentration.  相似文献

Abies faxoniana is a key species in reforestation processes in the southeast of the Qinghai-Tibetan Plateau of China. The changes in growth, photosynthesis and nutrient status of A. faxoniana seedlings exposed to enhanced ultraviolet-B (UV-B), nitrogen supply and their combination were investigated. The experimental design included two levels of UV-B treatments (ambient UV-B, 11.02 KJ m−2 day−1; enhanced UV-B, 14.33 KJ m−2 day−1) and two nitrogen levels (0; 20 g N m−2). The results indicated that: (1) enhanced UV-B significantly caused a marked decline in growth parameters, net photosynthetic rate (Pn), photosynthetic pigments and F v/F m, (2) supplemental nitrogen supply increased the accumulation of total biomass, Pn, photosynthetic pigments and F v/F m under ambient UV-B, whereas supplemental nitrogen supply reduced Pn, and not affect biomass under enhanced UV-B, (3) enhanced UV-B or nitrogen supply changed the concentration of nutrient elements of various organs.  相似文献
