Selenium (Se) is an essential element for many organisms but also toxic at higher levels. The objective of this study was to identify accessions from the model species Arabidopsis thaliana that differ in Se tolerance and accumulation. Nineteen Arabidopsis accessions were grown from seed on agar medium with or without selenate (50 microM) or selenite (20 microM), followed by analysis of Se tolerance and accumulation. Tissue sulfur levels were also compared. The Se Tolerance Index (root length+Se/root length control) varied among the accessions from 0.11 to 0.44 for selenite and from 0.05 to 0.24 for selenate. When treated with selenite, the accessions differed by two-fold in shoot Se concentration (up to 250 mgkg(-1)) and three-fold in root Se concentration (up to 1000 mgkg(-1)). Selenium accumulation from selenate varied 1.7-fold in shoot (up to 1000 mgkg(-1)) and two-fold in root (up to 650 mgkg(-1)). Across all accessions, a strong correlation was observed between Se and S concentration in both shoot and root under selenate treatment, and in roots of selenite-treated plants. Shoot Se accumulation from selenate and selenite were also correlated. There was no correlation between Se tolerance and accumulation, either for selenate or selenite. The F(1) offspring from a cross between the extreme selenate-sensitive Dijon G and the extreme selenate-tolerant Estland accessions showed intermediate selenate tolerance. In contrast, the F(1) offspring from a cross between selenite-sensitive and -tolerant accessions (Dijon GxCol-PRL) were selenite tolerant. The results from this study give new insight into the mechanisms of plant selenium (Se) tolerance and accumulation, which may help develop better plants for selenium phytoremediation or as fortified foods.  相似文献

The effects of different concentrations of selenite (2–30 μM) and selenate (2–60 μM) on biomass production, leaf area, and concentrations of photosynthetic pigments in lettuce plants were investigated. On the basis of the obtained results, the threshold of toxicity for the selenite and selenate has been designated. The toxicity thresholds for selenite and selenate were determined at concentrations of 15 and 20 μM, respectively. Next, four selenium (Se) concentrations (2, 4, 6 or 15 μM), below or near the toxicity boundary, have been selected for the lettuce biofortification experiment. In the biofortified plants, the oxidant status (levels of lipid peroxidation and H2O2 concentrations), as well as Se and sulphur (S) accumulation were analysed. In the edible parts of the lettuce, the Se concentration was higher for selenate presence compared to selenite; however, this difference was not as obvious as it was noted in the case of the roots, where selenite application caused the high accumulation of Se. An application of 15 μM Se as selenite caused a decline in the biomass and an intensification of prooxidative processes in the plant’s tissues and as toxic should be excluded from further biofortification experiments. These results indicate that an application of either selenate or selenite to the nutrient solution at concentrations below 15 μM can be used for biofortification of lettuce with Se, evoking better plant growth and not inducing significant changes in the oxidant status, the concentration of assimilation pigments and S accumulation.  相似文献

Selenium (Se) is considered an essential trace element for animals because of its nutritional and clinical value, including its special relevance in cancer prevention, and thus Se is at present used in biofortification programmes. However, possible effects of Se application on S metabolism and plant growth are still not clear. Thus, we analysed the effect that Se application in two different forms (selenate versus selenite) exerts on the S metabolism in lettuce plants grown for 66 days. Our results indicate that the application of selenite as opposed to selenate does not affect the foliar concentration of S. With respect to different enzymes in charge of sulphate (SO42−) assimilation, the ATP-sulphurylase activity varies only with the application of different rates of selenate, while the activity of O-acetylserine(thiol)lyase (OAS-TL) and serine-acetyltransferase (SAT) increase in activity mainly when selenite is applied. Finally, the concentration of cysteine (Cys) and total thiols (SH-total), fundamentally in the selenate treatments, increased with shoot biomass. In conclusion, this study confirms that the form and application rate of Se affects S assimilation, selenate being the more suitable form to improve effectiveness of the biofortification programme with this trace element.  相似文献

In an attempt to address the role of biological behavior on Se uptake by soybean crop and the genotype effects, experiments with time and concentration sequences of Se uptake by seedlings in Hoagland solution are conducted using selenite and selenate respectively. Two soybean cultivars Tong-ai 405 (TA) and Qidong Green-skin (QG) are used as different genotypes. In presence of selenite, Se uptake by both roots and shoots exhibited a linear increase with the growing time at 5 M and with the solution Se concentrations. However, in presence of selenate, the linear response to growing time is only valid before 24 h of growing. While root Se uptake is much slower under selenate than under selenite in the time sequence experiment, shoot Se levels are similar between the two different Se form treatments. Nevertheless, in the experiment of concentration sequence, either root Se or shoot Se responses linearly to solution Se concentration regardless of the Se forms supplied. A big discrepancy of root Se level with a similarity of shoot Se between the two cultivars is observed in the concentration sequence experiment. This supports a much faster passive uptake of selenite but more or less an active uptake of selenate by soybean seedlings. Comparatively, cultivars TA have a consistently higher Se concentration than QG both in roots and shoots under selenate, while no difference of concentration ratio of shoot to root is recognized between them. The higher Se level in seed grains, therefore, may be accounted for not by Se transport form root to shoot but by greater ability of Se uptake and retention under selenate by the former cultivar. Therefore, not only forms of Se supply but also genotype difference affects the Se bioavailability by different soybean cultivars. This should be taken into account for screening the high Se-efficiency plants or cultivars to improve the Se supply of the food chain.  相似文献

Selenium (Se), which has antioxidant, anticancer, and antiviral properties, is an essential micronutrient for humans and animals. This micronutrient is found in high quantity in legumes. Peas have an ever-increasing importance in Spain, and to increase their nutritional value, two foliar Se fertilizers: sodium selenate and sodium selenite, at five different rates: 0, 10, 20, 40, 80 g?ha?1, were studied during the 2010/2011 crop season on semiarid Mediterranean conditions. Sodium selenate was much more effectively taken up by plants compared to sodium selenite. There was a strong linear relationship between the total Se content and Se rate in both sodium selenate and selenite. For each gram of Se fertilization as either sodium selenate or sodium selenite, the increase of total Se concentration in the grain was 148 and 19 μg Se?kg?1 dry weight, respectively. Ingestion of 100 g of peas previously fertilized with 10 g of sodium selenate per hectare would result in an intake of 179 μg of Se. This is almost 90 % of the daily recommended dose needed to reduce the chance of some cancers and about 179 % of the minimum concentration required to prevent Se deficiency diseases in animals. The pea has shown to have a strong ability to uptake and accumulate Se under Mediterranean conditions; therefore, this would make it a very strong candidate for inclusion in biofortification programs aiming to increase Se in the food chain.  相似文献

Selenium (Se) is an essential trace element for humans and animals. A hydroponic experiment was performed to study the effects of sulphur (S) on Se uptake, translocation, and assimilation in wheat (Triticum aestivum L.) seedlings. Sulphur starvation had a positive effect on selenate uptake and the form of Se supplied greatly influenced Se speciation in plants. Compared with the control plants, Se uptake by the S-starved plants was enhanced by 4.81-fold in the selenate treatment, and selenate was readily transported from roots to shoots. By contrast, S starvation had no significant effect on selenite uptake, and selenite taken up by roots was rapidly converted to organic forms and tended to accumulate in roots. X-ray absorption near edge spectroscopy (XANES) analysis showed that organic forms of selenium, including selenocystine, Se-methyl-selenocysteine (MeSeCys), and selenomethionine-Se-oxide, were dominant in the plants exposed to selenite and accounted for approximately 90 % of the total Se. Whereas selenate remained as the dominant species in the roots and shoots exposed to selenate, with little selenate converted to selenite and MeSeCys. Besides, sulphur starvation increased the proportion of inorganic Se species in the selenate-supplied plants, but had no significant effects on Se speciation in plants exposed to selenite. The present study provides important knowledge to understand the associated mechanism of Se uptake and metabolism in plants.  相似文献

The main objective of this work was to determine whether nitrogen-use efficiency (NUE) was affected by the application of different forms and dosages of selenium (Se), to ascertain the influence of this trace element in a biofortification program in lettuce plants. The parameters analyzed were biomass, NO3 concentration, and total reduced N as well as those defining NUE in plants: total nitrogen accumulation (TNA), nitrogen efficient ratio (NER), nitrogen-utilization efficiency (NUtE), and nitrogen-uptake efficiency (NUpE). According to our results, application of Se affected NUE in lettuce plants. With the application of selenite as well as selenate NO3 uptake was reduced, thus diminishing the NUpE and the foliar concentration of this anion. In addition, selenate application at a rate of 20 μM and selenite at 5 μM induced N utilization, reflected by an increase in NER and NUtE; this result coincides with augmented biomass production. Notably, our results indicate that when Se is applied at high rates, selenite is far more phytotoxic, this being associated with a higher reduction of NUE in these plants.  相似文献

There has been an interest in using hyperaccumulating plants for the removal of heavy metals and metalloids. High selenium (Se) concentrations in the environment are detrimental to animals, humans, and sustainable agriculture, yet selenium is also an essential nutrient for humans. This experiment was conducted to screen fern plants for their potential to accumulate selenium. Eleven fern species, Pteris vittata, P. quadriaurita, P. dentata, P. ensiformis, P. cretica, Dryopteris erythrosora, Didymochlaena truncatula, Adiantum hispidulum, Actiniopteris radiata, Davallia griffithiana, and Cyrtomium fulcatum, were grown under hydroponic conditions for one week at 20 mg L(-1) selenate or selenite. Root Se concentrations reached 245-731 and 516-1082 mg kg(-1) when treated with selenate and selenite, respectively. The corresponding numbers in the fronds were 153-745 and 74-1,028 mg kg(-1) with no visible toxicity symptoms. Only three fern species were able to accumulate more Se in the fronds than the roots, which were D. griffithiana when treated with selenate, P. vittata when treated with selenite, and A. radiata regardless of the forms of Se. A. radiata was the best species overall for Se accumulation. More research is needed to further determine the potential of the fern species identified in this study for phytoremediation of the Se contaminated soils and water.  相似文献

Indian mustard (Brassica juncea L.) accumulates high tissue Se concentrations and volatilizes Se in relatively nontoxic forms, such as dimethylselenide. This study showed that the presence of bacteria in the rhizosphere of Indian mustard was necessary to achieve the best rates of plant Se accumulation and volatilization of selenate. Experiments with the antibiotic ampicillin showed that bacteria facilitated 35% of plant Se volatilization and 70% of plant tissue accumulation. These results were confirmed by inoculating axenic plants with rhizosphere bacteria. Compared with axenic controls, plants inoculated with rhizosphere bacteria had 5-fold higher Se concentrations in roots (the site of volatilization) and 4-fold higher rates of Se volatilization. Plants with bacteria contained a heat-labile compound in their root exudate; when this compound was added to the rhizosphere of axenic plants, Se accumulation in plant tissues increased. Plants with bacteria had an increased root surface area compared with axenic plants; the increased area was unlikely to have caused their increased tissue Se accumulation because they did not accumulate more Se when supplied with selenite or selenomethionine. Rhizosphere bacteria also possibly increased plant Se volatilization because they enabled plants to overcome a rate-limiting step in the Se volatilization pathway, i.e. Se accumulation in plant tissues.  相似文献

The relative plant availability of selenate versus selenite depends on the concentrations of competing ions, specifically sulfate and phosphate, respectively. In solution culture, the concentration of phosphate is typically 100- to 1000-fold greater than in soil solution, an artifact that could lead to underestimation of the phytoavailability of selenite. A nutrient solution study was conducted to compare the availability of selenite and selenate to perennial ryegrass (Lolium perenne L. cv. Evening Shade) and strawberry clover (Trifolium fragiferrum L. cv. O'Conner) at basal concentrations of SO4 (0.5 mM) and PO4 (2 μM) similar to those found in soil solution. Concentrations up to 5 mM SO4 and 200 μM PO4 allowed quantitative comparison of the efficacy of the competing ions. In both species, selenite was more phytotoxic than selenate, especially for shoot growth. Selenate was less toxic, and tended to preferentially inhibit root growth. Translocation percentages were much higher with selenate (≥84%) than with selenite (≤47%). A 10-fold increase in sulfate decreased uptake from selenate by >90% in both species. In ryegrass, 10-fold increases in phosphate caused 30% to 50% decreases in Se accumulation from selenite, but in clover such decreases only occurred in the roots. Sulfate-selenate antagonisms were thus stronger than phosphate-selenite antagonisms. Nonetheless, conventional nutrient solutions with millimolar phosphate will significantly underestimate Se availability from selenite, and moderate levels of sulfate salinity can inhibit selenate uptake sufficiently to reverse the apparent relative availability of the two forms of Se. Abstract

Selenate and selenite are the most prevalent bioavailable selenium (Se) forms and most easily taken up by plants. Some studies indicate that they are differently absorbed and accumulated in plants and that selenium is toxic if accumulated at high concentrations. Toxicity is due to substitution of sulphur by selenium in cysteine and methionine aminoacids with alteration of the tertiary structure and catalytic activity of proteins and with inhibition of enzymes involved in chlorophyll biosynthesis. Moreover, the interaction between Se and thiol groups induces loss of efficiency of plant defence systems and increases the reactive oxygen species (ROS) production thus enhancing the oxidative stress. To further elucidate the role of Se in higher plants, in this study the antioxidative response to the phytotoxicity of selenite and selenate in Senecio scandens L. was evaluated. The data indicate that while selenite induces oxidative stress enhancing ROS production, lipid peroxidation and the oxidised forms of ascorbate and glutathione, selenate does not significantly affect the analysed pathways. This article outlines that the synergistic action of different antioxidant components is necessary to overcome the phytotoxicity of selenium in Senecio.  相似文献

The aquatic plant Lemna minor L. was treated with sodium selenite or sodium selenate to test the toxicity of these salts in relation to high or low levels of sulfate in the culture medium. Several morphophysiological aspects, such as multiplication rate (MR), ratio of the number of fronds to number of colonies (Nfr/Ncol), frond size, cell ultrastructure, pigment content and guaiacol peroxidase (EC 1.11.1.7) activity were evaluated. Their variations might be an indirect means of evaluating the degree of susceptibility or tolerance of this plant to selenium (Se). Sodium selenite or sodium selenate treatments at concentrations ranging from 1 to 256 μ M generally decreased the investigated parameters. Moreover, the sulfate concentration influenced the toxicity of both Se salts. In general, with treatments in a medium containing a high sulfate (HS) content, sodium selenite appeared more toxic than sodium selenate, whereas in a low sulfate (LS) medium, sodium selenate seemed more toxic. MR was significantly increased at 1–4 μ M selenite and LS or 8 μ M selenate and HS levels, suggesting that Se may be an essential nutrient for this plant.  相似文献

Treatment of fathead minnows (Pimephales promelas) with either [75Se]selenate, -selenite or -l-selenomethionine by gavage at 20 ng Se/g resulted in organ uptake and early distribution patterns which differed significantly between compounds. The greatest differences in uptake between compounds was observed in liver tissue which accumulated much less [75Se]selenate than either selenite or l-selenomethionine. The 75Se burdens and relative distribution among the various organs were nearly identical during the elimination phase for [75Se]selenate and -selenite. This suggests that selenium derived from these compounds converge to a common metabolic pool. The whole body T1/2, rate of 75Se uptake and magnitude of 75Se accumulation were generally greater for [75Se]selenomethionine than the inorganic forms. Selenium-75 was present in the bile following the oral administration of each compound. The partitioning of selenate and selenite into the plasma and cellular fraction of blood differs with both the compound and time following exposure.  相似文献

Selenium (Se) is an essential micronutrient for animal and human nutrition, but whether it is essential to plants remains controversial. However, there are increasing experimental evidences that indicate a protective role of Se against the oxidative stress in higher plants through Se-dependent glutathione peroxidase (GSH-Px) activity. The effects of the Se chemical forms, selenite and selenate, the rate of their application on shoot Se concentration and their influence on the antioxidative system of ryegrass (Lolium perenne cv. Aries), through the measurement of GSH-Px activity and lipid peroxidation, were evaluated in an Andisol of Southern Chile. Moreover, a soil–plant relationship for Se was determined and a simple method to extract available Se from acid soils is proposed. In a 55-day experiment ryegrass seeds were sown in pots and soil was treated with sodium selenite or sodium selenate (0–10 mg Se kg−1). The results showed that the Se concentration in shoots increased with the application of both selenite and selenate. However, the highest shoot Se concentrations were obtained in selenate-treated plants. For both sources of Se, there was a significant positive correlation between the shoot Se concentration and the GSH-Px activity; and the Se-dependence of this enzymatic activity was related especially with the chemical form of applied Se rather than the Se concentration in plant tissues. Furthermore, the lipid peroxidation, as measured by Thiobarbituric Acid Reactive Substances (TBARS), decreased at low levels of shoot Se concentration, reaching the lowest level at approximately 20 mg Se kg−1 in plants and then increased steadily above this level. In addition, the acid extraction method used to evaluate available Se in soil showed a positive good correlation between soil Se and shoot Se concentrations irrespective of chemical form of Se applied.  相似文献

Selenium (Se)-fortified broccoli (Brassica oleracea var. italica) has been proposed as a functional food for cancer prevention, based on its high glucosinolate (GSL) content and capacity for Se accumulation. However, as selenate and sulphate share the initial assimilation route, Se fertilization could interfere with sulphur metabolism and plant growth. Consequently, GSL accumulation could be compromised. To evaluate these potentially adverse effects of Se fertilization, we performed a comprehensive study on sand-grown young broccoli plants (weekly selenate applications of 0.8 μmol plant(-1) via the root) and field-grown adult broccoli plants during head formation (single foliar selenate application: 25.3 or 253 μmol plant(-1) ). The results show that under these conditions, Se application does not affect plant growth, contents of cysteine, glutathione, total GSL, glucoraphanin (major aliphatic GSL) or the expression of BoMYB28 (encoding a functionally confirmed master regulator for aliphatic GSL biosynthesis). Conversely, due to the changed expression of sulphate transporters (BoSULTR1;1, 1;2, 2;1, and 2;2), sulphate and total S contents increased in the shoot of young plants while decreasing in the root. We conclude that broccoli can be fertilized with Se without reduction in GSL content, even with Se accumulation exceeding the level recommended for human consumption.  相似文献

Pot culture experiments containing sand and soil, at two levels of mercury (2 and 5 g mL-1) added through irrigation with increasing supplementation of selenium (selenite and selenate) led to a decrease in the uptake of mercury by tomato (Lycopersicum esculentum) plant. Both the forms of selenium (selenite and selenate) were found to be equally effective in reducing the mercury accumulation by plants. The observed reduction pattern of mercury accumulation in plant tissues has been discussed on the basis of the formation of insoluble HgSe complex in soil-root environment.  相似文献

After 3 h, selenate uptake by roots of Phaseolus vulgaris L.cv. Contender resulted in more than 50% of the Se absorbed beingconveyed to the aerial organs. This distribution was sensitiveto respiratory inhibitors and when roots were soaked in a solutionsupplied with hydroxylamine, the level of Se decreased by about80% in the whole plant, suggesting that selenate uptake requiresenergy. Addition of glucose to the nutrient medium resultedin slightly decreased uptake and distribution. Under the same growth conditions and 3 h incubation with selenite,a major part of the Se had accumulated in the roots, while asmall fraction was conveyed towards the aerial organs. Thispercentage was decreased by about 20% when plants were transferredto a solution supplied with hydroxylamine, suggesting that partof the selenite entered the roots passively. Addition of glucoseto the nutrient solution, resulted in enhanced levels of Sein the whole plant. Application of plant growth substances affected Se transport.When roots were incubated in abscisic acid (ABA), selenate uptakewas affected, while foliar spraying of gibberellin A3 (GA3)enhanced selenite uptake and translocation.
