The cotyledons of Copaifera langsdorfii Desf, have been shown to contain a water-soluble xyloglucan (amyloid), which represents about 40% of the seed's dry weight. On acid hydrolysis its composition (Glc:Xyl:Gal = 4.0:2.8–2.9:1.5–1.7) was similar to that of the well-characterized xyloglucan of Tamarindus indica L. (Glc:Xyl:Gal = 4.0:3.0–3.1:1.4). On hydrolysis with pure Trichoderma viride cellulase, both C. langsdorfii and T. indica xyloglucan gave the same xyloglucan oligosaccharides: but in significantly different proportions A:B1:B2:C = 1:0.4–0.5:2.1–2.2:3.1–3.4 in T. indica , and 1:1.1:1.8:7.4 and 1:1.3:2.6:12 for C. langsdorfii , savanna and forest populations respectively. This demonstrated a difference in fine molecular structure, notably in the distribution of the terminal non-reducing galactose substituents, between the xyloglucans of the two species and indicated differences in the specificities of their biosynthetic mechanisms. The xyloglucans obtained from C. langsdorfii seeds harvested from savanna and forest environments were slightly different, one from the other, in their sugar-residue composition (Glc:Xyl:Gal = 4.0:2.9:1.5 and 4.0:2.8:1.7, respectively), and were significantly different in the relative proportions of the xyloglucan oligosaccharides released on cellulase hydrolysis (above). Using light microscopy and biochemical methods, no difference in the pattern or rate of postgerminative xyloglucan metabolism was detected in seeds of savanna and forest origin. This is the first clear experimental evidence for differences in a storage xyloglucan structure between populations of the same species. It may indicate environmental influences on xyloglucan biosynthesis.  相似文献

Three seeds of Turkish origin, flax, poppy and safflower were analyzed for their proximate, fatty acids, tocols (tocopherols and tocotrienols) and total phenolic composition, and oxidative stability of their oil. The major fatty acid in the flax oil was alpha-linolenic acid, comprising 58.3% of total fatty acids, whereas poppy and safflower oils were rich in linoleic acid at 74.5% and 70.5% level, respectively. The amount of total tocols was 14.6 mg/100g flax, 11.0mg/100g poppy and 12.1mg/100g safflower seed. Flax and poppy oil were rich in gamma-tocopherol as 79.4 mg/100g oil and 30.9 mg/100g oil, respectively, while alpha-tocopherol (44.1g/100g oil) was dominant in safflower oil. Only alpha- and gamma-tocotrienol were found in the oils. Oxidative stability of oils was measured at 110 degrees C at the rate of 20 L/h air flow rate, and poppy oil (5.56 h) was most stabile oil followed by safflower oil (2.87 h) and flax oil (1.57). There were no correlation between oxidative stability and unsaturation degree of fatty acids and tocol levels of the oils. All of the seeds investigated provide a healthy oil profile and may have potential as a source of specialty oils on a commercial scale.  相似文献

Mature seeds of Tropaeolum majus L. contain the cell wall polysaccharide xyloglucan (amyloid), protein and lipid as storage substances. The transitory occurrence of starch during the process of seed development could be substantiated.[U-14C]-labelled xylose, glucose and glucuronic acid were fed to ripening seeds and the incorporation of radioactivity into xyloglucan, starch and the sugar nucleotide fraction of the cotyledons was determined. The results indicate that exogenous supplied xylose is not incorporated directly into xyloglucan, but is transformed to glucose before incorporation into xyloglucan and starch. Radioactivity from glucuronic acid was predominantly found in the xylose moiety of xyloglucan. Composition of tocopherols, tocotrienols, carotenoids, fatty acids, as well as hydrophilic and lipophilic antioxidant activities, were determined in seeds of two Salvia species and oils obtained from them. Both seeds contained a large amount of oil (around 20%) rich in polyunsaturated fatty acids. While Salvia officinalis seed oil can be classified as oleic‐linoleic oil, the predominant fatty acid in Salvia sclarea was α‐linolenic acid (around 54%). Among tocols, the main isomers in both seeds and oils were γ‐tocopherol, followed by α‐tocopherol. Concerning carotenoids, their concentration was around 0.75 mg/100 g of seeds and 0.16 mg/100 g of oils, with a predominance of lutein. Oil and seeds of S. officinalis exhibited higher antioxidant potential compared to S. sclarea investigated samples which could be attributed to higher content of total vitamin E and carotenoids. This study provides results that enables use of two Salvia species as new alternative sources of vegetable oils.  相似文献

Five forms of xyloglucan endotransglycosylase/hydrolase (XTH) differing in their isoelectric points (pI) were detected in crude extracts from germinating nasturtium seeds. Without further fractionation, all five forms behaved as typical endotransglycosylases since they exhibited only transglycosylating (XET) activity and no xyloglucan-hydrolysing (XEH) activity. They all were glycoproteins with identical molecular mass, and deglycosylation led to a decrease in molecular mass from approximately 29 to 26.5 kDa. The major enzyme form having pI 6.3, temporarily designated as TmXET(6.3), was isolated and characterized. Molecular and biochemical properties of TmXET(6.3) confirmed its distinction from the XTHs described previously from nasturtium. The enzyme exhibited broad substrate specificity by transferring xyloglucan or hydroxyethylcellulose fragments not only to oligoxyloglucosides and cello-oligosaccharides but also to oligosaccharides derived from β-(1,4)-d-glucuronoxylan, β-(1,6)-d-glucan, mixed-linkage β-(1,3; 1,4)-d-glucan and at a relatively low rate also to β-(1,3)-gluco-oligosaccharides. The transglycosylating activity with xyloglucan as donor and cello-oligosaccharides as acceptors represented 4.6%, with laminarioligosaccharides 0.23%, with mixed-linkage β-(1,3; 1,4)-d-gluco-oligosaccharides 2.06%, with β-(1,4)-d-glucuronoxylo-oligosaccharides 0.31% and with β-(1,6)-d-gluco-oligosaccharides 0.69% of that determined with xyloglucan oligosaccharides as acceptors. Based on the sequence homology of tryptic fragments with the sequences of known XTHs, the TmXET(6.3) was classified into group II of the XTH phylogeny of glycoside hydrolase family GH16.  相似文献

A water soluble galactomannan isolated from Leucaena leucocephala seeds gave an intrinsic viscosity of 3.5dl/g and viscosity average molecular mass, M(v), of 6.98×10(5)g/mol. This was in reasonably good agreement with the value of the weight average molecular mass, M(w), of 5.44±0.20×10(5)g/mol determined by GPC-MALLS coupled to RI. The onset of polymer coil overlap occurred at c*[η] of 2.1, with slope of 3.0 above and 1.3 below the point of polymer coil overlap. The shear viscosity of the polysaccharide was temperature dependent and decreased with increasing temperature. The activation energy for viscous flow of 3.0% polysaccharide concentration obtained by Arrhenius plot of zero shear viscosity as a function of temperature was 26.4kJ/mol. Both the storage modulus (G') and loss modulus (G″) showed strong dependence on frequency indicating the presence of entangled coils. The Cox-Merz plot gave close superimposition of the complex and shear viscosities.  相似文献

The jojoba plant (Simmondsia chinensis L.) produces seeds that contain around 50 to 60 wt.% of inedible long-chain wax esters that are suitable as a potential feedstock for biodiesel (BD) production. Jojoba oil methyl esters (JME) were prepared from acid-catalyzed pretreated jojoba oil in order to evaluate important fuel properties of jojoba-based BD, including kinematic viscosity, cloud point (CP), pour point (PP), cold filter plugging point (CFPP), acid value (AV), oxidative stability, and lubricity. A comparison was made with soybean oil methyl esters (SME) and relevant BD fuel standards such as ASTM D6751 and EN 14214. JME was characterized using Fourier transform infrared spectroscopy and 1H and 13C nuclear magnetic resonance. The CP, PP, and CFPP of JME were ?13°C, ?16°C, and ?14°C, respectively, which were superior to SME. The kinematic viscosity (40°C) of JME was 6.67 mm2/s, which was higher than observed for SME. Blends (B5 and B20) of JME in ultra-low sulfur diesel fuel (ULSD) were also evaluated for the aforementioned fuel properties and compared to an analogous set of blends of SME in ULSD and relevant petro diesel fuel standards such as ASTM D975 and D7467. JME blends in ULSD displayed improved low-temperature properties in comparison to neat ULSD and blends of SME in ULSD. In summary, jojoba oil has potential as an alternative, nonfood feedstock for BD production.  相似文献

On aqueous extraction, Hymenaea courbaril var. stilbocarpa, known in Brazil as jatobá, furnishes a high yield of viscous xyloglucan (45%) from its seeds. The crude polysaccharide (B1) was hydrolysed and the products, analysed as alditol acetates, were glucose, xylose, galactose and arabinose in the ratio 50:35:13:2. After further fractionation on a DEAE-cellulose column (chloride form), the main fraction (70% yield, B2) was obtained. The basic structure of the xyloglucan was determined as a cellulose-type (1 → 4)-linked β-d-glucan backbone partially substituted with side chains at 06 of -d-xylopyranose, some of which were themselves substituted at 02 by the units of β-d-galactopyranose. Treatment of the xyloglucan (B2) with commercial cellulase from Trichoderma sp. yielded six oligosaccharides. These oligosaccharides were isolated by preparative paper chromatography, and their structures were determined by gas-liquid chromatography-mass spectroscopy of the derived partially O-methylated alditol acetates. There are several reports indicating that hypergravity and microgravity influence the mechanical properties of cell walls in shoots, resulting in changes in the growth rate. The mechanical properties of cell walls in dicots are mainly determined by the physicochemical properties of xyloglucan, a matrix polysaccharide. An increase in the molecular mass of xyloglucan correlated with a decrease in cell wall extensibility. Hypergravity is known to increase the molecular mass of xyloglucan. The cell wall enzyme, xyloglucan endotransglucosylase/hydrolase (XTH) is involved in xyloglucan metabolism. Using Arabidopsis, it was examined whether or not the expression of XTH genes in the floral stem and rosette leaf is influenced by hypergravity. RT-PCR analysis revealed that the expression of XTH genes changes in response to hypergravity of 300 g.  相似文献

Despite widespread and its local available as a naturalized hedge and shade plant, the potential of Trichilia emetica was not utilized in soap making by the majority of local community in various parts of Dodoma, Tanzania. This study aimed to assess the quantity (yields) and quality (Acid Values (AVs), %Free Fatty Acids (%FFAs) and Saponification Values (SVs) of seed oil from water plant (T. emetica), suitable for soap making application. Solvent extraction method was used during oil extraction, where by 50gm of preheated and powdered seed materials were immersed in 250 ml of n-hexane in 1:5 (w/v) to dissolve the oil contained in the seed cake. The oil was collected by vaporizing solvent out through Rotary evaporation at 60 °C. Also standard titration methods were used to obtain SVs, AVs and %FFAs of the extracted oil. Results showed that T. emetica seeds contained higher quantity of oil (48.4%−50.2%) than many reported commercial plant seed oils. Also, the study found higher AV (7.4 mgKOH/g−7.8 mgKOH/g), %FFA (3.7% to 3.9%) and SVs (189.5 mgKOH/g 191.4 mgKOH/g) than the maximum acceptable limits of 0.50 mg KOH/g, 0.020% and 175 mgKOH/g − 187 mgKOH/g prescribed by ASTM standards (2002). The obtained results showed that, T. emetica seeds yielded high oil quantity with low qualities due to higher levels of acidity. But high SVs guarantees the possibility of using T. emetica seed oil in soap making. However, the oil requires purification in order to bring levels of acidity to acceptable standards and guarantee its normal use in soap making.  相似文献

Aqueous extraction of sunflower seeds and rapeseeds with mixtures of hydrolytic enzymes were carried out in 2-L reactor and uronic acid and reducing sugar concentrations were measured during digestion. The goal of the study was to determine if a correlation between those concentrations and free oil exists that would allow predicting free oil release from aqueous solution measurements. Similar concentrations of 4.5 and 4.7 g/L of reducing sugars and 43 and 55 g/L of uronic acid were found for sunflower and rapeseeds, respectively. The corresponding yields of free oil from the two seeds were 83% and 16% of total oil, and this difference was due to the formation of an emulsion in the rapeseed dispersion. Therefore, measurement of reducing sugars or uronic acid concentrations are not sufficient to predict a release of free oil from all oil seeds.  相似文献

The objective of this study was to evaluate the impact of diets enriched with plant oils or seeds, high in polyunsaturated fatty acids (PUFA), on the fatty acid profile of sheep intramuscular and subcutaneous adipose tissue (SAT). Sixty-six lambs were blocked according to initial body weight and randomly assigned to six concentrate-based rations containing 60 g fat/kg dry matter from different sources: (1) Megalac (MG; ruminally protected saturated fat), (2) camelina oil (CO), (3) linseed oil (LO), (4) NaOH-treated camelina seed (CS), (5) NaOH-treated linseed (LS) or (6) CO protected from ruminal saturation by reaction with ethanolamine; camelina oil amides (CA). The animals were offered the experimental diets for 100 days, after which samples of m. longissimus dorsi and SAT were collected and the fatty acid profile determined by GLC. The data were analyzed using ANOVA with 'a priori' contrasts including camelina v. linseed, oil v. NaOH-treated seeds and CS v. CA. Average daily gain and total fatty acids in intramuscular adipose tissue were similar across treatments. The NaOH-treatment of seeds was more effective in enhancing cis-9, trans-11 conjugated linoleic acid (CLA) incorporation than the corresponding oil, but the latter resulted in a higher content of trans-11 18:1 in both muscle neutral and polar lipids (P < 0.01, P < 0.001, respectively). Inclusion of LS resulted in the highest PUFA:saturated fatty acid (SFA) ratio in total intramuscular fat (0.22). The NaOH-treatment of seeds resulted in a higher PUFA/SFA ratio (0.21 v. 0.18, P < 0.001) than oils and on average, linseed resulted in a higher PUFA/SFA ratio than camelina (P < 0.01). Lambs offered LS had the highest concentration of n-3 PUFA in the muscle, while those offered MG had the lowest (P < 0.001). This was reflected in the lowest (P < 0.001) n-6: n-3 PUFA ratio for LS-fed lambs (1.15) than any other treatment, which ranged from 2.14 to 1.72, and the control (5.28). The trends found in intramuscular fat were confirmed by the data for SAT. This study demonstrated the potential advantage from a human nutrition perspective of feeding NaOH-treated seeds rich in PUFA when compared to the corresponding oil. The use of camelina amides achieved a greater degree of protection of dietary PUFA, but decreased the incorporation of biohydrogenation intermediates such as cis-9, trans-11 CLA and trans-11 18:1 compared to NaOH-treated seeds.  相似文献

A xyloglucan (XG) was extracted from the seeds of Hymenaea courbaril (Jatobá) harvested from trees growing at different locations in Brazil. They were from Foz do Chopin Forest Reserve, State of Paraná (HC-I), Cuiaba (HC-II) and Sinope/State of Mato Grosso (HC-III), Natal/State of Rio Grande do Norte (HC-IV), Fortaleza/State of Ceará (HC-V) and Tamarindus indica (TM) from Conceição de Almeida/State of Bahia, Brazil. It was submitted to SEC-MALLS analysis and its intrinsic viscosity determined. When a solution of XG was heated to 85 °C for 2 h, there was a reduction of aggregation and the XG from the seeds of H. courbaril had a higher value of C when compared with that of from T. indica, probably due to the presence of an unique oligossacharide series present in the H. courbaril samples (XXXXG). This gives rise to a more rigid molecule when compared with the XG from T. indica.  相似文献

BACKGROUND AND AIMS: Hymenaea courbaril (Leguminosae-Caesalpinioideae) is a tree species with wide distribution through all of the Neotropics. It has large seeds (approx. 5 g) with non-photosynthetic storage cotyledons rich (40 %) in a cell wall polysaccharide (xyloglucan) as a carbon reserve. Because it is found in the understorey of tropical forests, it has been considered as a shade-tolerant, late-secondary species. However, the physiological mechanisms involved in seedling establishment, especially regarding the interplay between storage and light intensity, are not understood. In this work, the ecophysiological role of this carbon cotyledon reserve (xyloglucan) is characterized, emphasizing its effects on seedling growth and development during the transition from heterotrophy to autotrophy under different light conditions. METHODS: Seedlings of H. courbaril were grown in environments with different light intensities, and with or without cotyledons detached before xyloglucan mobilization. Development, growth, photosynthesis and carbon partitioning (dry mass and [14C]sucrose) were analysed in each treatment. KEY RESULTS: The detachment of cotyledons was not important for seedling survival, but resulted in a strong restriction (50 % less) of shoot growth, which was the main sink for the cotyledon carbon reserves. Carbon restriction promoted an early maturation of the photosynthetic apparatus without changes in the net CO2 fixation per unit area. The reduced surface area of the first leaves in seedlings without cotyledons was evidence of limited growth and development of seedlings in low light conditions (22 micromol m(-2) s(-1) photon flux). CONCLUSIONS: There is an increase in the importance of storage xyloglucan in cotyledons for H. courbaril seedling development as light intensity decreases, confirming that this polymer plays a key role in the adaptation of this species to establish successfully in the shadowed understorey of the forest.  相似文献

Siberian apricot (Prunus sibirica L.), an excellent woody oil plant unique to Asia, is well known for its ability to produce high‐oil seeds for use as a promising feedstock of biodiesel. Based on the investigation of natural Siberian apricot resources in China in the early stage, seeds of Siberian apricot from 74 geographic provenances which can fully reflect the overall information were collected. In this research, seeds oil content, fatty acid composition and biodiesel properties were evaluated, and the key environmental factors that caused the variation of these in different geographic provenance were analyzed. The oil content of Siberian apricot seeds is 45.48%–61.07%, and the average was 50.95% for all provenances. The characteristics of oil can identify and quantify eight fatty acids. The most abundant fatty acids were oleic acid (C18:1; 54.02%–76.54%), followed by linoleic acid (C18:2; 16.78%–38.49%) and erucic acid (C16:0; from 3.27% to 6.12%). Monounsaturated fatty acids are the most abundant in 54.75%–77.03% compared with saturated fatty acids and polyunsaturated fatty acids. The biodiesel properties of most provenance seeds meet the standards of the ASTM D6751 and GB/T 20828, and a few meet the standards of the EN14214. Through the clustering of oil content and fatty acid composition and the analysis of biodiesel properties indexes, it is concluded that KSK provenance is the most suitable for biodiesel production. The XBZ, HHE, AES, ZLQ and LD provenances may be preserved as potential biodiesel. RDA and VPA showed that the effects of environmental factors on the oil properties of Siberian apricot were ranked as terrain factor > climate factor > soil factor, among which longitude, latitude and altitude are the main terrain indicators. These evaluations can provide reference for the effective utilization and further development of Siberian apricot as a bioenergy feedstock.  相似文献

Arabidopsis thaliana is frequently used as a model for the study of oilseed biology and metabolism. However, the very small seeds of Arabidopsis can complicate analysis of their oil content and influence the application of results to larger-seeded plants. Here, we describe how seed anatomy, light, and plant-to-plant variation influence the content and measurement of oil in Arabidopsis seeds. The anatomy of Arabidopsis and Brassica napus seeds were compared and the distribution of mass, oil and the fatty acid composition of different seed parts were determined. In Brassica, 90% of the seed oil resides in the cotyledons that contribute 74% of seed mass. By contrast, the values for Arabidopsis are 60% and 45%, respectively, with a higher fraction of the oil deposited in the radicle, hypocotyl, endosperm and seed coat. Growth of Arabidopsis plants with 600 micromol m(-2) s(-1) light resulted in a two-fold higher seed yield, a 40% increase in mass per seed and a 60% increase in oil per seed compared to growth at 100 micromol m(-2) s(-1). Factors that influence the analysis of oil content were evaluated. Intact-seed transmethylation followed by gas chromatography (GC) analysis provided reproducible analysis of Arabidopsis seed oil. However, plant-to-plant variation in oil content is large and we analyzed how this influences the ability to detect statistically valid changes in oil between different genotypes. These observations establish a reference data set on the fatty acid composition and distribution of mass and oil between tissues of Arabidopsis seeds that should help to predict the applicability of results obtained with Arabidopsis to other oilseeds.  相似文献

African green monkeys were fed diets containing either 11% (by weight) fish oil or lard for 2.5 yr. To test the hypothesis that fish oil decreases hepatic secretion of triglyceride (TG) and apoB, livers from these animals were perfused with a fatty acid mixture [85% (w/w) oleate containing [14C]oleate and 15% n-3 containing [3H]eicosapentaenoic acid (EPA)] at a rate of 0.1 mumol fatty acid/min per g liver. Liver perfusate was sampled every 30 min during 4 h of recirculating perfusion. The concentration of triglyceride was similar for livers of animals of both groups and there was no difference between groups in the extent of incorporation of [3H]EPA or [14C]oleate into hepatic TG. While the secretion rate for the mass of TG was less in the fish oil-fed group (8.3 +/- 2.5 vs 18.3 +/- 4.4 mg/h per 100 g liver, P less than 0.05), the apoB secretion rate was similar (0.92 +/- 0.15 vs 1.01 +/- 0.13 mg/h per 100 g liver). Significantly less [3H]EPA was incorporated into secreted TG in the fish oil group (0.4 +/- 0.1 vs 1.0 +/- 0.1% infused dose/h; P less than 0.01). The rate of secretion of [14C]TG was similar for both groups (1.3 +/- 0.3 vs 1.4 +/- 0.1% infused dose/h for fish oil and lard groups, respectively). No significant diet-related differences in [3H]TG or [14C]TG fatty acid specific activity were observed for perfusate TG or hepatic TG. After perfusion, livers from fish oil-fed monkeys contained significantly more [3H]EPA in hepatic phospholipid than livers from lard-fed monkeys (19.5 +/- 1.8 vs 11.4 +/- 1.7% infused dose; P less than 0.01) although hepatic phospholipid mass concentrations were similar. The liver phospholipids of the fish oil group were enriched in n-3 fatty acid mass and were relatively depleted of oleate and linoleate. We conclude that although apoB secretion was unaffected, dietary fish oil significantly decreased hepatic TG secretion through relatively poor utilization of EPA for the synthesis of TG destined for secretion in VLDL; at the same time, increased incorporation of [3H]EPA into hepatic phospholipid accompanied the decreased incorporation into secreted TG and these events may be coupled.(ABSTRACT TRUNCATED AT 400 WORDS)  相似文献

Experiments carried out to study changes induced by hard-to-cook (HTC) phenomenon in the non-starch polysaccharides of beans stored at 30 °C and 75% RH for 8 months showed that the development of HTC did not affect the amounts of soluble and insoluble fibre in cooked seeds but changed their carbohydrates physical properties. Aged beans non-starch polysaccharides presented lower water-solubility and underwent lower degradation of galacturonans and arabinose-rich polysaccharides when submitted to cooking. The decrease in non-starch polysaccharides water-solubility produced a shift in the polymers fractionation profile which resulted in an increase of weak and middle-alkali soluble polymers bulk as well as in their arabinose and uronic acid contents. Uronic acid contents were higher in polymers released by 1 M NaOH and in the cellulose-rich residues while the arabinose contents were higher in the mild-alkali soluble polymers of aged seeds. Methylation analysis showed no evident alterations in the xyloglucans and arabinans branching degree with beans ageing. However, both, the molecular mass of water-soluble pectins and CDTA-soluble pectins, increased. Even though changes in the non-starch polysaccharide solubility were not related to the decrease in the arabinan and xyloglucan degree of branching they may be related to the formation of new chemical interactions other than hydrogen bond. There was a correlation between acidic and neutral polysaccharides insolubilisation in beans ageing and probably in beans hardening. After processing, aged seeds present higher amounts of insoluble fibre when compared to normal beans.  相似文献
