直接用戊二醛固定水样中的浮游病毒,通过超速离心使已固定的浮游病毒沉淀到覆有Formvar膜和碳支持膜的铜网上,经醋酸双氧铀染色后,利用透射电镜对湖水中的浮游病毒进行观察.结果可观察到球形、杆状和蝌蚪状等形态各异的浮游病毒颗粒及球形病毒的囊膜子粒、杆状病毒的核衣壳、具有不同尾部的蝌蚪状病毒等的精细超微结构.从而建立了一种简便、快捷和高效研究浮游病毒的电镜方法.  相似文献

We examined mitochondrial-DNA (mtDNA) sequence heterogeneity on four adjacent trapping grids in an island population of meadow voles (Microtus pennsylvanicus) at two different population densities. Four restriction endonucleases revealed 20 different mtDNA composite phenotypes in samples totaling 198 meadow voles. There were significant heterogeneities in the distribution of four common mtDNA composite phenotypes among the four trapping grids, suggesting that there is population subdivision on a fine scale. Genetic distances between grids, mtDNA diversity within grids, and GST also varied during the study period. We found a decrease in genetic distance and an increase in diversity when the population density was high and vice versa when the population density was low. When population density was high, the coefficient of gene differentiation was smaller than the same coefficient observed when the population density was low. These changes in population subdivision and diversity are consistent with theoretical expectations of population structure in which effective female population size and dispersal are the critical variables. The data also support the hypothesis of maintenance of mtDNA diversity by population subdivision, rapid population growth rate, and dispersal.  相似文献

Summary Helix pomatia (Snail) lectin complexed with colloidal gold (HPL-gold) recognized binding sites on elastic fibers in plastic embedded sections of lung tissue from mice of several ages. Deposition of the lectin-gold particles was examined by electron microscopy. Structures such as the elastic laminae of pulmonary vessels and elastic fibers throughout the lung was specifically and intensely decorated by the HPL-gold complex and easily visualized. The binding of the HPL-gold particles was primarily to sites on the amorphous component of elastin, to the virtual exclusion of the microfibrillar elastin elements, collagen fibers and other components of the extracellular matrix. In addition, moderate age differences in the binding of HPL-gold to elastin were apparent. These observations appear to be the first demonstration of the presence, in the amorphous component of elastin, of glycoconjugates that are specifically recognized by HPL and suggest a method by which the involvement of glycoconjugates in lung elastogenesis could be explored.Supported in part by USPHS Grant HL-32870  相似文献

Summary. Vitamin C is accumulated in mammalian cells by two types of proteins: sodium-ascorbate co-transporters (SVCTs) and hexose transporters (GLUTs); in particular, SVCTs actively import ascorbate, the reduced form of this vitamin. SVCTs are surface glycoproteins encoded by two different genes, very similar in structure. They show distinct tissue distribution and functional characteristics, which indicate different physiological roles. SVCT1 is involved in whole-body homeostasis of vitamin C, while SVCT2 protects metabolically active cells against oxidative stress. Regulation at mRNA or protein level may serve for preferential accumulation of ascorbic acid at sites where it is needed. This review will summarize the present knowledge on structure, function and regulation of the SVCT transporters. Understanding the physiological role of SVCT1 and SVCT2 may lead to develop new therapeutic strategies to control intracellular vitamin C content or to promote tissue-specific delivery of vitamin C-drug conjugates. As part of a study sponsored by the Ministry of Health of Italy, a research program on pathogenic actinomycetes, keratinophilic and pathogenic fungi in soil was carried out. Two hundred soil samples, collected from different areas of the city of Rome, Calabria, Emilia Romagna, Latium, Apulia, Sardinia, Sicily, Tuscany and Umbria, were examined by several techniques to detect the widest possible variety of pathogenic actinomycetes and fungi. Seven isolates ofNocardia asteroides, four ofActinomadura madurae and one ofNocardiopsis dassonvillei were isolated for the first time from soil in Italy. In addition, numerous isolates ofPetriellidium boydii, Aspergillus fumigatus, A. flavus, A. niger and keratinophilic fungi of the generaMicrosporum, Trichophyton andChrysosporium were also recovered.Presented at the VII Congress of the International Society for Human and Animal Mycology, Jerusalem, Israel, 11–16 March, 1979.  相似文献

The archaeobotanical study of the charred macro-remains recovered from the burnt settlement of La Fontanaccia, Allumiere, 50 km northwest of Rome, a small hut from the time of the end of the late Roman Empire, provided results on the use of food of its inhabitants, their living conditions, and the natural environment. The fire which destroyed the small settlement was archaeologically dated to the middle of the 5th century a.d., few years before the end of the Roman Empire. This was a period in which the state structure, undermined by the barbarian invasions which provoked famine and destruction, was in deep economic and political crisis, and the population in Rome and in the countryside lived in precarious conditions. No archaeo-botanical data have been available until now for this period in the region of Rome. The presence of grass peas, acorns, two-rowed barley caryopses, and small horse bean seeds demonstrate the general state of regression in the late Roman Empire, when misery and famine were widespread. The finds of charcoal from chestnut, deciduous oak, maple and elm suggest the presence of thermophilous deciduous woods and environmental conditions similar to today’s. It deserves mention that this is the first site in which macro-remains (charcoal) of Castanea have been found in central Italy.  相似文献

Summary The present investigation was undertaken in an effort to differentiate histochemically between sialo-mucins and acid mucopolysaccharides in connective tissues.On the basis of previous studies regarding the histochemical identification of sialic acid in sections of animal salivary glands the author applied different technical procedures on cartilage and tooth germs of newborn rats.The results obtained indicated that in the tissues examined sialic acid is a normal component of the polysaccharide fraction of the connective tissue ground substance.On leave of absence at the University of Rome, Medical School, Viale Regina Elena 287-A, Rome, Italy.  相似文献

The fine structure of resting and activated platelets was compared using two approaches novel to this dense cytoplasm. First, rapid lysis of platelets on carbon-coated grids was following by negative staining of the "cytoskeleton." Second, a brief, minimal fixation of platelets in plasma was coupled with partial lysis and examination of the unstained whole mounts at 200 kV. The results showed that the dense ground cytoplasm of discoid, fully resting platelets appeared granular or amorphous, and microfilaments were not observed. A coiled microtubule terminated in one, free, straight end. When any slight degree of activation occurred, microfilaments could be detected in the platelets. In fully spread specimens, the amorphous character of the resting cytoplasm was strikingly altered into an interconnected network of microfilaments. Stereo views of the whole mounts showed that dense granules, 100-250 nm in diameter, appeared as if suspended in the filament nets. The results support the view that platelet activation involves a major assembly of microfilaments from amorphous precursors. The change can only be seen convincingly when stringent precautions are taken during preparation because the platelets are very easily activated by thermal or mechanical stimuli.  相似文献

We have examined the shape of apolipoprotein B (apoB) from low density lipoproteins (LDL) using a new method to prepare the electron microscope grids. After adsorption of the lipoproteins to a carbon-coated copper grid, lipids were extracted with ethanol-ether 4:1; an aqueous negative stain was then applied. When the LDL residue was examined after this treatment, apoB, together with residual lipid, appeared as an elongated flexible structure about 600-700 A in length consisting of multiple domains of variable width from 20-70 A. Occasionally, the elongated apoB formed an irregular ring-shaped structure, but most of the rings were open. When LDL were pretreated with glutaraldehyde, then adsorbed, extracted, and stained, most of the images were closed rings with an average contour length of 700 A, again consisting of multiple domains of variable sizes. These results are consistent with apoB being composed of multiple domains arranged in an elongated structure on the surface of the LDL, and with distant domains possessing a mutual affinity that favors their cross-linking.  相似文献

Elastin, a core protein of the elastic fibers, exhibits the coacervation (temperature‐dependent reversible association/dissociation) under physiological conditions. Because of this characteristic, elastin and elastin‐derived peptides have been considered to be useful as base materials for developing various biomedical products, skin substitutes, synthetic vascular grafts, and drug delivery systems. Although elastin‐derived polypeptide (Val‐Pro‐Gly‐Val‐Gly)n also has been known to demonstrate coacervation property, a sufficiently high (VPGVG)n repetition number (n > 40) is required for coacervation. In the present study, a series of elastin‐derived peptide (Phe‐Pro‐Gly‐Val‐Gly)5 dimers possessing high coacervation potential were newly developed. These novel dimeric peptides exhibited coacervation at significantly lower concentrations and temperatures than the commonly used elastin‐derived peptide analogs; this result suggests that the coacervation ability of the peptides is enhanced by dimerization. Circular dichroism (CD) measurements indicate that the dimers undergo similar temperature‐dependent and reversible conformational changes when coacervation occurs. The molecular dynamics calculation results reveal that the sheet‐turn‐sheet motif involving a type II β‐turn‐like structure commonly observed among the dimers and caused formation of globular conformation of them. These synthesized peptide dimers may be useful not only as model peptides for structural analysis of elastin and elastin‐derived peptides, but also as base materials for developing various temperature‐sensitive biomedical and industrial products. A scaffold-like structure is observed under the electron microscope when mouse chromosomes are digested with the restriction endonuclease Hae III. This structure, located in the inner part of chromatids, may correspond to those fragments of chromatin loops anchored to the chromosome scaffold and is obtained when chromosomes are treated either in suspension or attached to grids. The width of the structure is correlated with the extent of digestion in chromosomes treated in suspension. Those treated on grids show this structure whenever chromatids do not collapse. These results agree with the model of chromosome organization based on a non-histone protein scaffold.  相似文献

BackgroundThe nature of fibrillar deposits from aqueous solutions of human serum and recombinant human transferrin on mica and carbon-coated formvar surfaces has been investigated.Methods and ResultsAtomic force microscopy showed that the deposition of recombinant transferrin onto the hydrophilic surface of mica resulted in the formation of a monolayer-thick film composed of conformationally-strained flattened protein molecules. Elongated fibres developed on top of this layer and appeared to be composed of single proteins or small clusters thereof. Monomeric and dimeric transferrins were separated by gel permeation chromatography and their states of aggregation confirmed by mass spectrometry and dynamic light scattering. Transmission electron-microscopy showed that dimeric transferrin, but not monomeric transferrin, deposited on carbon-coated formvar grids forms rounded (circular) structures ca. 250 nm in diameter. Small transferrin fibrils ca. 250 nm long appeared to be composed of smaller rounded sub-units. Synchrotron radiation-circular dichroism and, Congo red and thioflavin-T dye-binding experiments suggested that transferrin aggregation in solution does not involve major structural changes to the protein or formation of classical β-sheet amyloid structures. Collisional cross sections determined via ion mobility–mass spectrometry showed little difference between the overall protein shapes of apo- and holo-transferrin in the gas phase.General significanceThe possibility that transferrin deformation and aggregation are involved in neurological disorders such as Parkinson's and Alzheimer's disease is discussed. The fine structure of the spermatozoa and spermatophores of the zuwai crab, Chionoecetes opilio, was examined electron microscopically. The spermatophores embedded in the secretory droplets within the vasa deferentia showed a spherical structure with an extremely wrinkled envelope and contained numerous spermatozoa. The mature spermatozoa of this crab, similar to those of other brachyurans, were stellate in shape and had a globular acrosome surrounded by a cup-like nucleus with several radiating processes. The acrosome was ultrastructurally complex and its apical part was characterized by an electron-dense discoid structure, whereas its innermost part was occupied by an electron-lucent cylindrical structure containing assemblies of thin tubules and a reticular formation of electron-dense material. The cytoplasm interposed between the nucleus and acrosome was remarkably reduced in volume and displayed a membranous lamellar complex, few mitochondria, and a centriole. The nuclear chromatin was not condensed but represented by finely flocculent material. The morphological aspects of the zuwai crab spermatozoa are discussed in comparison with those of other decapod crustaceans.  相似文献

We report the results of three mark–release–recapture experiments carried out in an urban area in Rome, Italy, to study the active dispersal of Aedes albopictus (Diptera: Culicidae). The 4.3% recapture rate obtained supports the use of sticky traps in MRR experiments to study the dispersal of Ae. albopictus females. Most fluorescent dust‐marked females were recaptured at the gravid stage at 50–200 m from the release sites during the first 9 days after release. The average of daily‐MDTs (Mean Distance Traveled) was 119 m and the maximum observed distance travelled ranged from 199 m to 290 m in the three replicates. These data provide the first information about the dispersal of Ae. albopictus in a temperate European area and appear to be consistent with the few data available on this subject from other urban areas, where dispersal was constrained by physical barriers. Although caution should be taken in generalizing these results, they should be considered when planning control activities in urban areas in Italy, as well as in other European countries. This is particularly relevant if control is intended to interrupt pathogen transmission in cases of possible arbovirus epidemics, such as the Chikungunya outbreak that occurred in Ravenna, Italy in 2007.  相似文献

Summary The kinetics of protein accumulation, the variation in RNA, the soluble amino nitrogen content of developing endosperm of two varieties of Triticum aestivum, with high and low protein content in the mature seed, suggest a possible relation between maintenance of the RNA content and the ability to synthesize protein. A sudden halt in protein accumulation is observed as the RNA starts to decrease. The elastin digestion assay was examined to determine if it would facilitate the identification of Coccidioides immitis when non-pathogenic fungi resembling C. immitis are encountered. Fungal isolants tested have anamorphs that closely resemble the macroscopic or microscopic morphology of C. immitis. Elastin hydrolysis was measured by elastin-agar plate assays. Approximately 80% of the isolants hydrolyzed elastin; thus, the elastin digestion assay as a differential test appears to have little value.  相似文献

Cryoelectron microscopy makes it possible to record high-resolution detail from large and complex structures. However, its application to understanding cellular structure is limited by the requirement that samples should be no thicker than approximately 0.5-1 microm. Therefore it is important to develop the ability to section biological material so that it can be imaged in its native frozen state. Here we have adapted standard methods of preparing cryosections so that they can be imaged by cryoelectron microscopy. As used for immunolabeling, cryosections of chemically fixed, cryoprotected frozen rat cardiac muscle were thawed, applied to carbon-coated grids, and rinsed on a drop of buffer. The special step here is that the cryosections were then refrozen by being plunged into liquid ethane and imaged at approximately -180 degrees C in a 200-kV field-emission gun electron microscope. The unstained cryosections have good contrast, allowing the identification of optimum regions of the sample. Considerable fine detail is observed within the substructure of the sarcomere A-band and I-band. Fourier transform analysis of the micrographs shows that this method preserves high structural order, hence these sections are well-suited to 3D reconstruction. We conclude that this approach has considerable potential for obtaining intermediate- and high-resolution structural detail from bulk tissue.  相似文献

The nuclear magnetic spin-lattice and transverse relaxation processes for the 1H and 2D nuclei in purified elastin (ligamentum nuchae), exchanged and hydrated with excess D2O, have been studied in the temperature range 276°–340°K. The 2D relaxation results clearly show the presence of D2O (1) external to the bulk elastin sample, (2) in spaces within the bulk elastin, and (3) as an integral part of the protein on a molecular level. It is shown from these measurements that the protein on a molecular level. It is shown from these measurements that the water content of the protein itself changes from ~0.8 g D2O/g dry elastin at ~280°K to ~0.2 g D2O/g dry elastin at ~335°K, a decrease of 400%. The D2O content of the interfiber spaces decreases by less than 20% over the same temperature range. This fact throws considerable doubt on the validity of the values of β, the thermal expansion coefficient of elastin, used by other workers in discussion of the elastic mechanism in elastin. The elastin proton transverse relaxation shows the presence of three regions in elastin having different degrees of molecular mobility. These are assigned to protons associated with the crosslinks, a fairly mobile, hydrophobic, and low-water-content region, and a more mobile higher water-content region. The temperature variation of the relative proportions of these three regions is explained in terms of a hypothetical temperature-composition phase diagram in which the two mobile regions are represented as two partially miscible phases with different negative temperature coefficients of ‘solubility’ in water. The implications of these observations for current views of the nature of elastin are assessed. It is concluded that the spin-relaxation results are consistent with a multiphase structural model for elastin. An approximate sorption isotherm for the water/elastin system is reported and shows the relatively weak nature of the water/elastin interaction.  相似文献
