The authors investigated the response to experimentally elevated water temperature in genotypes of Pocillopora damicornis from three coral reefs in the upwelling Gulf of Panama and four coral reefs in the non-upwelling Gulf of Chiriquí, Panamanian Pacific. Sea-surface temperature in the Gulf of Panama declines below 20 °C during seasonal upwelling, while in the thermally stable Gulf of Chiriquí, the temperature ranges from 27 to 29 °C. Genotypes of P. damicornis from the seven locations were determined by allozyme electrophoresis. The most abundant genotype at each location was selected for a thermal tolerance experiment where corals were exposed to water temperature of 30 °C (1 °C above ambient) for 43 days. Four site coral genotypes can be uniquely differentiated by the GPI locus, two by the LGG-2 locus, and two by a combination of the MDH-1, LGG-2, and LTY-3 loci. A visual assessment of the coral condition after exposure to an elevated temperature showed that corals from localities in the non-upwelling environment retained a normal to slightly pale appearance, while corals from the upwelling environment bleached and their polyps were mostly retracted. A two-way ANOVA confirmed that corals were significantly affected by water temperature and locality. The zooxanthellae were also significantly affected by the interaction of elevated temperature and locality of the corals. Mean zooxanthellae density decreased by 25 and 55%, respectively, in experimentally heated corals from the non-upwelling and upwelling environments. Low concentrations of photosynthetic pigments per live area of the corals were the norm in corals under elevated temperature. The mean concentration of chlorophyll a per live area of the corals was reduced by 17 and 49%, respectively, in heated corals from the non-upwelling and upwelling sites. Coral genotypes from the upwelling Gulf of Panama demonstrated higher vulnerability to thermal stress than coral genotypes from the non-upwelling Gulf of Chiriquí. However, the latter showed greater differences in their responses. Thus, even at small geographic scales, corals can display different levels of tolerance to thermal stress. The difference in thermal tolerance between corals from upwelling and non-upwelling environments is concomitant with greater genetic differences in experimental corals from the thermally stable Gulf of Chiriquí compared with corals from the upwelling Gulf of Panama.Communicated by K.S. High temperature tolerance experiments performed on Pocillopora damicornis, a major reef-building coral in the tropical eastern Pacific, resulted in loss of zooxanthellae, histopathological abnormalities, and mortality similar to that observed during the severe 1982–83 El Niño-Southern Oscillation (ENSO) event. Coral vitality declined significantly at 30–32°C during a 10-week period, but remained high at normal temperatures (26–28°C). Laboratory time courses to coral morbidity and death were similar to those observed in the field. Experimental high temperatures had a greater negative effect on corals from the Gulf of Panama, which experiences seasonally cool upwellings, than on corals from the nonupwelling Gulf of Chiriqui. The condition of obligate symbiotic crustaceans (Trapezia, Alpheus) associated with experimental corals declined with their host's declining condition. All Gulf of Panama corals subjected to 32°C were dead after 5 weeks, and all of their associated crustacean symbionts were dead after 9 weeks. Gulf of Chiriqui corals at 30°C survived for 9 weeks and 42% of their crustacean symbionts were still alive after 10 weeks. Coral mortality in the Gulf of Panama was significantly higher (68.5%) after El Niño warming than after subsequent episodes of unusually intense cool upwellings (10.4%). Low temperature stress (cool currents and upwelling) has been generally suggested as the critical limiting condition that prevents extensive coral reef development in the eastern Pacific. Our results suggest that infrequent but severe ENSO sea warming events also may limit reef development in this region.  相似文献

The timing of skeletal band formation and concomitant changes in calcification rates and linear skeletal extension were investigated in Pavona corals growing under two distinct thermal regimes along the Pacific coast of Panama: fluctuating, marked by seasonal upwelling (Gulf of Panama) and stable, nonupwelling (Gulf of Chiriqui). The purpose of this study was to test the hypothesis that banding in corals is largely mediated by seasonal variations in temperature (Highsmith 1979). Our results indicate that the timing of band formation is synchronous at these two environmentally distinct locations. The low density (LD) portion of the annual band is accreted over a five month period (January–June) and represents an increase in linear skeletal extension (mm/mo.) as well as a marked increase in calcification rate (g CaCO3 · cm-2 · mo-1) relative to the high density portion which forms over the remaining seven month period (July through December). In contrast to the predictions of the Highsmith model these findings indicate that variations in light levels rather than fluctuation in temperature is a better correlate to changes in skeletal density. Qualitatively, banding patterns were similar at the two sites; however, higher growth rates (particularly with respect to the LD band) for Pavona clavus in the Gulf of Panama indicate that lower water temperatures and higher productivity, or both, may be responsible for quantitative differences in banding between sites. We found that formation of the HD band corresponds to lower light levels and the production of gametes. We propose that banding in corals is a complex phenomenon governed by endogenous processes (e.g. reallocation of energy from growth to reproduction) which may be mediated by exogenous factors (e.g light and productivity).  相似文献

Mediterranean junipers are of special ecological importance as key components of resource islands in semi-arid mountain ecosystems of the Mediterranean basin. The fragmentation of their habitat, which was primarily natural and driven by climatic drought conditions, is currently being aggravated by anthropogenic pressure. In the framework of this concern, the present work aims to contribute establishing a genomic profile of Juniperus in its western Mediterranean range, with a special emphasis placed on J. thurifera. DNA contents were assessed by flow cytometry in 43 populations of nine taxa within their Mediterranean range (first reports for J. navicularis, J. thurifera subsp. africana and J. thurifera subsp. thurifera). Chromosome numbers were determined by orcein staining in eight taxa (first counts for J. oxycedrus subsp. badia, J. phoenicea subsp. phoenicea, J. phoenicea subsp. turbinata, of 2n?=?2x?=?22, and for J. thurifera subsp. thurifera, of 2n?=?4x?=?44). Tetraploid cytotypes have been the only ones found in the 19 populations of J. thurifera studied, this being the first report of a Juniperus species exclusively polyploid. In J. thurifera, C-value does not respond to habitat fragmentation, in the same way that genetic diversity within populations was previously shown to be unaltered, suggesting that this factor has not had, at least to date, a significant impact on populations at genomic and genetic levels. Habitat fragmentation leads to deeply age-biased populations with a male-biased imbalanced sex ratio (lack of females), indicating an urgent need to improve regeneration within the populations of this species.  相似文献

Anadenanthera colubrina var. cebil is a native South American tree species inhabiting seasonally dry tropical forests (SDTFs). Its current disjunct distribution presumably represents fragments of a historical much larger area of this forest type, which has also been highly impacted by human activities. In this way the hypothesis of this study is that the natural populations of A. colubrina var. cebil from Northern Argentina represent vestiges of ancient fragmentation, but they are additionally influenced by a certain degree of gene flow among them. We aimed to analyze the genetic structure of both nuclear and chloroplast DNA to evaluate the relative role of ancient and recent fragmentation on intraspecific diversity patterns. Sixty-nine individuals of four natural populations were analyzed using eight nuclear microsatellites (ncSSR) and four chloroplast microsatellite loci (cpSSR). The level and distribution of genetic variation were estimated by standard population genetic parameters and Neighbor Joining as well as Bayesian analyses. The eight ncSSR loci were highly polymorphic, while genetic diversity of cpSSRs was low. Nuclear SSRs displayed lower genetic differentiation among populations than cpSSR haplotypes (F ST 0.11 and 0.95, respectively). However, high differentiation between phytogeographic provinces was observed in both genomes. The high genetic differentiation detected emphasizes the role of ancient fragmentation. However, the Paranaense province also shows the effects of recent fragmentation on genetic structure, whereas gene flow by pollen preserves the effects of genetic drift in the Yungas province.  相似文献

Patterns of reproductive periodicity in the regular echinoid species Diadema mexicanum A. Agassiz and Echinometra vanbrunti A. Agassiz from the Bay of Panama, and Diadema antillarum Phillipi, Echinometra lucunter (Linnaeus) and E. viridis A. Agassiz from the Caribbean coast of the Isthmus of Panama exhibit dissimilarities that reflect the differences of the environments they inhabit. Populations of the two species from the seasonal Bay of Panama display synchronous, well-defined, reproductive cycles. Spawning appears to be timed so that newly metamorphosed sea urchins, rather than larvae, can benefit from the increased food production concomitant with dry season upwelling. On the less seasonal Caribbean shore reproductive periodicity is less defined. Populations of Diadema antillarum and Echinometra lucunter from the vicinity of the Panama Canal show indications of periodicity, while those from a locality 20 km to the east display little tendency for synchrony between individuals. The much rarer E. viridis, on the other hand, maintains well-defined, population-wide cycles in both localities. It is suggested that in a constant environment the intensity of selection for synchrony between individual gametogenic cycles may be inversely proportional to population density.  相似文献

Relationships were analyzed between sea surface temperature (SST) and annual growth characteristics (density, extension rate and calcification rate) of the Caribbean reef-building coral Montastraea annularis. Colonies were collected from 12 localities in the Gulf of Mexico and the Caribbean Sea. Two well-separated relationships were found, one for the Gulf of Mexico and the other for the Caribbean Sea. Calcification rate and skeletal density increased with increasing SST in both regions, while extension rate tended to decrease. Calcification rate increased ∼0.57 g cm−2 year−1 for each 1 °C increase in SST. Zero calcification was projected to occur at 23.7 °C in corals from the Gulf of Mexico and at 25.5 °C in corals from the Caribbean Sea. The 24 °C annual average SST isotherm marks the northern limit of distribution of M. annularis. Montastraea annularis populations of the Gulf of Mexico are isolated from those of the Caribbean Sea, and results indicate that corals from the Gulf of Mexico are adapted to growth at lower minimum and average annual SST. Corals from both the Gulf of Mexico and the Caribbean Sea, growing at lower SSTs and having lower calcification rates, extend their skeletons the same or more than those growing at higher SSTs. They achieve this by putting more of their calcification resources into extension and less into thickening, i.e., by sacrificing density.  相似文献

Jacaranda decurrens (Bignoniaceae) is an endemic species of the Cerrado with validated antitumoral activity. The genetic diversity of six populations of J. decurrens located in the State of São Paulo was determined in this study by using molecular markers for randomly amplified polymorphic DNA (RAPD) and amplified fragment length polymorphism (AFLP). Following optimization of the amplification reaction, 10 selected primers generated 78 reproducible RAPD fragments that were mostly (69.2%) polymorphic. Two hundred and five reproducible AFLP fragments were generated by using four selected primer combinations; 46.3% of these fragments were polymorphic, indicating a considerable level of genetic diversity. Analysis of molecular variance (AMOVA) using these two groups of markers indicated that variability was strongly structured amongst populations. The unweighted pair group method with arithmatic mean (UPGMA) and Pearson''s correlation coefficient (RAPD -0.16, p = 0.2082; AFLP 0.37, p = 0.1006) between genetic matrices and geographic distances suggested that the population structure followed an island model in which a single population of infinite size gave rise to the current populations of J. decurrens, independently of their spatial position. The results of this study indicate that RAPD and AFLP markers were similarly efficient in measuring the genetic variability amongst natural populations of J. decurrens. These data may be useful for developing strategies for the preservation of this medicinal species in the Cerrado.  相似文献

Mangrove forests in the Gulf of California, Mexico represent the northernmost populations along the Pacific coast and thus they are likely to be source populations for colonization at higher latitudes as climate becomes more favorable. Today, these populations are relatively small and fragmented and prior research has indicated that they are poor in genetic diversity. Here we set out to investigate whether the low diversity in this region was a result of recent colonization, or fragmentation and genetic drift of once more extensive mangroves due to climatic changes in the recent past. By sampling the two major mangrove species, Rhizophora mangle and Avicennia germinans, along the Pacific and Atlantic coasts of Mexico, we set out to test whether concordant genetic signals could elucidate recent evolution of the ecosystem. Genetic diversity of both mangrove species showed a decreasing trend toward northern latitudes along the Pacific coast. The lowest levels of genetic diversity were found at the range limits around the Gulf of California and the outer Baja California peninsula. Lack of a strong spatial genetic structure in this area and recent northern gene flow in A. germinans suggest recent colonization by this species. On the other hand, lack of a signal of recent northern dispersal in R. mangle, despite the higher dispersal capability of this species, indicates a longer presence of populations, at least in the southern Gulf of California. We suggest that the longer history, together with higher genetic diversity of R. mangle at the range limits, likely provides a gene pool better able to colonize northwards under climate change than A. germinans.  相似文献

Noncovalent aggregation as a side reaction competing with the reconstitution of oligomeric enzymes is enhanced by slow conformational changes within the partially unfolded subunits. This has been shown for lactic dehydrogenase from pig muscle after acid dissociation [G., Zettlmeissl R. Rudolph, and R. Jaenicke (1981)Eur. J. Biochem.121, 169–175]. The present experiments confirm previous spectroscopic evidence (from circular dichroism) applying pepsin digestion and subsequent analysis of the fragments on sodium dodecyl sulfate-polyacrylamide gradient gels. The susceptibility of certain fragmentation sites toward pepsin digestion changes with increasing incubation at acid pH, in accordance with a slow M1 → M2 transition of the acid-dissociated monomers. Constant pulses of pepsin at varying times after transferring native enzyme to pH 2.3 yield distinct changes in the fragmentation pattern consisting of undigested monomers (Mr = 35,000) plus 12 fragments ranging from 31,000 to 5000. Short digestion of the M2 species at low concentrations of pepsin preferentially yields 25,000 and 10,500 fragments (molar ratio pepsin:lactic dehydrogenase = 1:24). The time-dependent decrease of monomers upon incubation in 0.1 m sodium phosphate, pH 2.3, at 20 °C strictly parallels the formation of the two fragments. The quantitative kinetic analysis of the changes in peptide pattern yields a first-order rate constant K1 = 8 ± 2 × 10?4 s?1. The observed increase in proteolytic susceptibility is in the time range of the above mentioned decrease in the far-ultraviolet circular dichroism, and the parallel decrease in the yield of reactivation. The results suggest that during the M1 → M2 transition at acid pH a specific interdomain cleavage site is becoming exposed. As taken from the molecular weight of the two main fragments the trp 225-lys 226 peptide bond is the most probable candidate for this cleavage site.  相似文献

Among the loss of genetic diversity due to population declines, population fragmentation and habitat loss, hybridization also stands as a threat to Morelet’s crocodile (Crocodylus moreletii) populations. Genetic surveys in Belize and the Yucatan Peninsula have detected evidence of hybridization with the American crocodile (C. acutus). Admixture between these two species is most likely driven by human-mediated translocations. Along the central gulf coast of Mexico, C. moreletii populations are presumed to be purebred. To test this, we use nine microsatellite loci and sequence data from the mitochondrial control region to detect if C. acutus alleles have introgressed into populations of C. moreletii from central Veracruz. In 2010, C. moreletii was transferred from Appendix I to II of CITES based on a whole species demographic analysis, which indicated that populations had recovered across its range. Our study shows that populations in central Veracruz are purebred, although they exhibit low levels of genetic diversity most likely caused by inbreeding. Our data also suggest there is fragmentation among populations of C. moreletii, which may lead to further loss of genetic variation. Due the purity and low genetic diversity of C. moreletii populations from central Veracruz, we recommend increased protection and active management practices that take genetic data into account.  相似文献

Habitat fragmentation can have severe effects on plant pollinator interactions, for example changing the foraging behaviour of pollinators. To date, the impact of plant population size on pollen collection by pollinators has not yet been investigated. From 2008 to 2010, we monitored nine bumble bee species (Bombus campestris, Bombus hortorum s.l., Bombus hypnorum, Bombus lapidarius, Bombus pascuorum, Bombus pratorum, Bombus soroensis, Bombus terrestris s.l., Bombus vestalis s.l.) on Vaccinium uliginosum (Ericaceae) in up to nine populations in Belgium ranging in size from 80 m2 to over 3.1 ha. Bumble bee abundance declined with decreasing plant population size, and especially the proportion of individuals of large bumble bee species diminished in smaller populations. The most remarkable and novel observation was that bumble bees seemed to switch foraging behaviour according to population size: while they collected both pollen and nectar in large populations, they largely neglected pollen collection in small populations. This pattern was due to large bumble bee species, which seem thus to be more likely to suffer from pollen shortages in smaller habitat fragments. Comparing pollen loads of bumble bees we found that fidelity to V. uliginosum pollen did not depend on plant population size but rather on the extent shrub cover and/or openness of the site. Bumble bees collected pollen only from three plant species (V. uliginosum, Sorbus aucuparia and Cytisus scoparius). We also did not discover any pollination limitation of V. uliginosum in small populations. We conclude that habitat fragmentation might not immediately threaten the pollination of V. uliginosum, nevertheless, it provides important nectar and pollen resources for bumble bees and declining populations of this plant could have negative effects for its pollinators. The finding that large bumble bee species abandon pollen collection when plant populations become small is of interest when considering plant and bumble bee conservation.  相似文献

  • Due to ongoing human impacts, plant species increasingly occur in landscapes that are highly fragmented, with remaining natural habitats occupying small areas, resulting in populations that are smaller and more isolated than in previous time periods. This changed metapopulation structure is expected to have negative impacts on seed production. For example, the proportion of female plants within gynodioecious populations may be more volatile due to genetic drift in small populations associated with small habitat fragments, with concomitant impacts on seed production. My aims were to determine: (i) if variation in proportion of females is larger in smaller fragments; and (ii) if such changes in female frequency in small fragments result in reduced seed production.
  • Thirty‐two populations of Lobelia spicata Lam., a gynodioecious species, were surveyed in 2000, 2001 and 2009 in the tallgrass prairie region of Midwestern North America (Illinois and Indiana, USA). Data were collected for: proportion of female plants, total number of flowering plants (measure of population size), seed set per plant and prairie fragment size (another measure of population size).
  • The proportion of females is more variable in smaller prairie fragments. Seed number per fruit decreases as the proportion of females increases in a population, but only significantly for female plants. The number of flowering plants is positively associated with fruit production for both genders. Populations within larger prairie fragments have higher seed production.
  • The reproductive consequences of habitat fragmentation depend on the plant breeding system. While both sexes were negatively impacted, females were more adversely affected.

The introduction and spread of non-indigenous species (NIS) in marine ecosystems accelerated during the twentieth century owing to human activities, notably international shipping. Genetic analysis has proven useful in understanding the invasion history and dynamics of colonizing NIS and identifying their source population(s). Here we investigated sequence variation in the nuclear ribosomal Internal Transcribed Spacer region of the ctenophore Mnemiopsis leidyi, a species considered one of the most invasive globally. We surveyed four populations from the native distribution range along the Atlantic coasts of the United States and South America, as well as six populations in the introduced range from the Black, Azov, Caspian and Baltic seas. Allelic and nucleotide diversity of introduced populations were comparable to those of native populations from which they were likely drawn. Introduced populations typically exhibited lower genetic differentiation (F ST = ?0.014?C0.421) than native populations (F ST = 0.324?C0.688). Population genetic analyses supported the invasion of Eurasia from at least two different pathways, the first from the Gulf of Mexico (e.g., Tampa Bay) to the Black Sea and thence to the Caspian Sea, the second from the northern part of the native distribution range (e.g., Narragansett Bay) to the Baltic Sea. The relatively high genetic diversity observed in introduced populations is consistent with large inocula and/or multiple invasions, both of which are possible given ballast water transport and the extensive native distribution of the ctenophore in the Atlantic Ocean.  相似文献

The two essential thiol groups of myosin, SH-1 and SH-2, have been localized in an ~ 20K segment of the heavy chain by analysis of the distribution of radioactivity after tryptic digestion of tryptic heavy meromyosin (HMM) or papain-HMM subfragment-1, both labeled at SH-1 and SH-2 with [14C]iodoacetamide and [14C]N-ethyl maleimide, respectively. The results are discussed in the framework of earlier work (Bálint, M., Sréter, F. A., Wolf, I., Nagy, B., and Gergely, J. (1975) J. Biol. Chem. 250, 6168–6177) on the tryptic fragmentation of myosin heavy chain and in the light of more recent work on the location of a fragment that reacts with a photoaffinity analog of ATP (Szilágyi, L., Bálint, M., Sréter, F. A., and Gergely, J. (1978) Fed. Proc. 37, 1695) and of suggestions concerning the binding of ATP in the region containing the SH-1 and SH-2 (Elzinga, M., and Collins, J. H. (1977) Proc. Nat. Acad. Tropical late‐successional tree species are at high risk of local extinction due to habitat loss and fragmentation. Population‐growth rates in fragmented populations are predicted to decline as a result of reduced fecundity, survival and growth. We examined the demographic effects of habitat fragmentation by comparing the population dynamics of the late‐successional tree Poulsenia armata (Moraceae) in southern Mexico between a continuous forest and several forest fragments using integral projection models (IPMs) during 2010–2012. Forest fragmentation did not lead to differences in population density and even resulted in a higher population‐growth rate (λ) in fragments compared to continuous forests. Habitat fragmentation had drastic effects on the dynamics of P. armata, causing the population structure to shift toward smaller sizes. Fragmented populations experienced a significant decrease in juvenile survival and growth compared to unaltered populations. Adult survival and growth made the greatest relative contributions to λ in both habitat types during 2011–2012. However, the relative importance of juvenile survival and growth to λ was highest in the fragmented forest in 2010–2011. Our Life Table Response Experiment analysis revealed that positive contributions of adult fecundity explained most of the variation of λ between both habitats and annual periods. Finally, P. armata has a relatively slow speed of recovery after disturbances, compromising persistence of fragmented populations. Developing a mechanistic understanding of how forest fragmentation affects plant population dynamics, as done here, will prove essential for the preservation of natural areas.  相似文献

The Rhinella margaritifera species group consists of 17 species of toads distributed in tropical and subtropical South America and eastern Central America. The identity of some of its species is poorly understood and there are numerous undescribed cryptic species. Among them, the status of Rhinella margaritifera is one of the most problematic. Its range includes lowland rainforests separated by the Andes, the Chocoan rainforest to the west and the Amazonian rainforest to the east. This distribution is puzzling because the Andes are an old and formidable barrier to gene flow and therefore should generate vicariant speciation between disjunct lowland populations. Herein we clarify the taxonomy of populations of the Rhinella margaritifera complex from Central America and the Chocó region of South America. The morphological and genetic variation of Rhinella margaritifera was examined from 39 populations from Chocó, 24 from the upper Amazon region of Ecuador, and 37 from Panama, including the holotype of the Panamanian Rhinella alata. Phylogenetic analyses were performed based on mitochondrial genes 12S rRNA, 16S rRNA, and cytochrome c oxidase I (COI) and the nuclear gene Tyrosinase (Tyr). The genetic and morphological data show that Panamanian and Chocoan populations are conspecific. In the phylogeny, populations from Chocó and Panama form a well-supported clade. The morphology of the holotype of Rhinella alata falls within the variation range of Panamanian and Chocoan populations. Based on all this evidence, we assign the populations from western Ecuador and Panama to Rhinella alata and demonstrate that the unusual distribution pattern of “Rhinella margaritifera” on both sides of the Andes was an artifact of incorrectly defined species boundaries.  相似文献

Asexual reproduction via fragmentation is an integral component of the life history of many marine organisms. In south-eastern Australia the green macroalga Caulerpa filiformis (Suhr) Hering is abundant on many rocky shorelines and fragments are very abundant in the water column. We examined some of the factors that may influence the growth and successful reattachment of fragments of C. filiformis that potentially contribute to its success. Field surveys revealed that C. filiformis fragments were of variable morphologies (from small fronds to entire thalli) and sizes (0.1-60 cm in length). All fragments of C. filiformis were viable propagules that survived and grew well in the laboratory. However, some fragments grew more than others, in particular larger sizes (7.5 cm class) of certain morphologies (fragments that consisted only of a single frond). Rhizoids were produced by all fragments, but again, larger fragments produced more rhizoids than smaller fragments. The force required to detach fragments was proportional to the number (but not size) of rhizoids, demonstrating the importance of rhizoids for attachment. The rate of attachment of fragments was also fast, usually within 12 h, but the longer these fragments were attached to a substratum the greater the force required to dislodge them. Interestingly, fragments only produced rhizoids if they were resting on a substratum which may explain the small proportion of fragments with rhizoids in the field (< 5%). Ultimately, the long term success of C. filiformis fragments was low as only 1.6% of fragments attached within 24 h and none of these persisted for longer than 2 days. Nonetheless, the abundance and viability of fragments of C. filiformis available to attach suggest that asexual fragmentation is a successful reproductive strategy in this seaweed.  相似文献
