Recent studies on global climate change report that increase in seawater temperature leads to coastal ecosystem change, including coral bleaching in the tropic. In order to assess the effect of increased seawater temperature on a temperate coastal ecosystem, we studied the inter-annual variation in productivity of Laminaria japonica using long-term oceanographic observations for the Uwa Sea, southern Japan. The annual productivity estimates for L. japonica were 2.7 ± 2.5 (mean ± SD) kg wet wt. m−1 (length of rope) (2003/2004), 1.0 ± 0.6 kg wet wt. m−1 (2004/2005) and 12.1 ± 12.5 kg wet wt. m−1 (2005/2006). Our previous study using the same methodology at the same locality reported that the productivity was estimated for the 2001/2002 (33.3 ± 15.2 kg wet wt. m−1) and 2002/2003 (34.0 ± 8.7 kg wet wt. m−1) seasons. Productivity in 2003/2004 and 2004/2005 was significantly lower than in years 2001/2002, 2002/2003 and 2005/2006. A comparison of oceanographic conditions among the 5 years revealed the presence of threshold seawater temperature effects. When the average seawater temperature during the first 45 days of each experiment exceeded 15.5°C, productivity was reduced to about 10 % of that in cooler years. Moreover the analysis of growth and erosion rates indicates that when the seawater temperature was over 17.5°C, erosion rate exceeded growth rate. Thus, an increase of seawater temperature of just 1°C during winter drastically reduces the productivity of L. japonica in the Uwa Sea.  相似文献

Recently, aquaculture of Laminaria japonica has expanded to the southern coast of Japan and to China along the East China Sea. The southerly distribution of L. religiosa, compared to that of L. japonica, indicated that the aquaculture of L. religiosa along the southern coasts of Japan might be feasible. Thus, we examined the growth, biomass and productivity of L. religiosa cultivated in the Uwa Sea, in southwestern Japan over a period of two years. The seawater temperature ranged from 12.9 to 27.4°C in 2003/2004 and from 12.2 to 28.3°C in 2004/2005. In 2003/2004, the maximum mean density, maximum mean length, and maximum mean wet weight of L. religiosa was 7.8 ± 5.0 ind. m−1 (mean ± SD), 14.8 ± 4.6 cm, and 1.2 ± 0.8 g wet wt., respectively. In 2004/2005, no germination was confirmed through the study period. The maximum biomass and annual production in 2003/2004 were estimated to be 6.9 ± 5.2 g wet wt. m−1 and 8.9 g wet wt. m−1 year−1, respectively. The present study revealed that L. religiosa cultivated in the Uwa Sea were much smaller compared with those of Hokkaido Island, where the alga is naturally found. For the growth of L. religiosa, a relatively long period of seawater temperatures below 13.5°C is required. In the study area, seawater temperatures were below 13.5°C only 11 days in 2003, and 12 days in 2004. As a result, it is thought that expanding the cultivation of L. religiosa to southern areas including the Uwa Sea will be difficult.  相似文献

The specific metabolic rate (SMR) and haemolymph osmolality (HO) of the mud crab Rhithropanopeus harrisii Gould, 1841 from Baltic brackish waters were measured at a habitat salinity of 7 psu (T = 15 °C, full air saturation) and after step-wise acclimation to a salinity range of 3-27 psu. Values of SMR at 7 psu varied between 0.40 and 3.89 J g− 1 WW h− 1 (n = 25, wet weight range 0.051-1.142 g) and were significantly (p < 0.05) related to the specimen's wet weight (WW) according to the power regression SMR = 0.94 WW 0.41 (R2 = 0.68). The SMR of females did not differ significantly (p > 0.05) from those of males. When exposed to higher salinities, the SMR of R. harrisii decreased significantly (p < 0.05) and reached a minimum value at 23 psu (0.55 ± 0.05 J g− 1 WW h− 1, n = 6). Mean haemolymph osmolality at 7 psu amounted to 581 ± 26 mOsm kg− 1 (n = 5) and was 2.9 times higher than that of the external medium. R. harrisii hyperosmoregulated its body fluids up to 24 psu (727 mOsm kg− 1) at which salinity the isosmotic point was reached.  相似文献

Absorbance difference spectroscopy and redox titrations have been applied to investigate the properties of photosystem I from the chlorophyll d containing cyanobacterium Acaryochloris marina. At room temperature, the (P740+ − P740) and (FA/B − FA/B) absorbance difference spectra were recorded in the range between 300 and 1000 nm while at cryogenic temperatures, (P740+A1 − P740A1) and (3P740 − P740) absorbance difference spectra have been measured. Spectroscopic and kinetic evidence is presented that the cofactors involved in the electron transfer from the reduced secondary electron acceptor, phylloquinone (A1), to the terminal electron acceptor and their structural arrangement are virtually identical to those of chlorophyll a containing photosystem I. The oxidation potential of the primary electron donor P740 of photosystem I has been reinvestigated. We find a midpoint potential of 450 ± 10 mV in photosystem I-enriched membrane fractions as well as in thylakoids which is very similar to that found for P700 in chlorophyll a dominated organisms. In addition, the extinction difference coefficient for the oxidation of the primary donor has been determined and a value of 45,000 ± 4000 M− 1 cm− 1 at 740 nm was obtained. Based on this value the ratio of P740 to chlorophyll is calculated to be 1:~ 200 chlorophyll d in thylakoid membranes. The consequences of our findings for the energetics in photosystem I of A. marina are discussed as well as the pigment stoichiometry and spectral characteristics of P740.  相似文献

In this study, we have structurally characterized the amidase of a nitrile-degrading bacterium, Rhodococcus sp. N-771 (RhAmidase). RhAmidase belongs to amidase signature (AS) family, a group of amidase families, and is responsible for the degradation of amides produced from nitriles by nitrile hydratase. Recombinant RhAmidase exists as a dimer of about 107 kDa. RhAmidase can hydrolyze acetamide, propionamide, acrylamide and benzamide with kcat/Km values of 1.14 ± 0.23 mM− 1s− 1, 4.54 ± 0.09 mM− 1s− 1, 0.087 ± 0.02 mM− 1s− 1 and 153.5 ± 7.1 mM− 1s− 1, respectively. The crystal structures of RhAmidase and its inactive mutant complex with benzamide (S195A/benzamide) were determined at resolutions of 2.17 Å and 2.32 Å, respectively. RhAmidase has three domains: an N-terminal α-helical domain, a small domain and a large domain. The N-terminal α-helical domain is not found in other AS family enzymes. This domain is involved in the formation of the dimer structure and, together with the small domain, forms a narrow substrate-binding tunnel. The large domain showed high structural similarities to those of other AS family enzymes. The Ser-cis Ser-Lys catalytic triad is located in the large domain. But the substrate-binding pocket of RhAmidase is relatively narrow, due to the presence of the helix α13 in the small domain. The hydrophobic residues from the small domain are involved in recognizing the substrate. The small domain likely participates in substrate recognition and is related to the difference of substrate specificities among the AS family amidases.  相似文献

A case study on Centaurea gymnocarpa Moris & De Not., a narrow endemic species, was carried out by analyzing its morphological, anatomical, and physiological traits in response to natural habitat stress factors under Mediterranean climate conditions. The results underline that the species is particularly adapted to the environment where it naturally grows. At the plant level, the above-ground/below-ground dry mass (1.73 ± 0.60) shows its investment predominately in the above-ground structure with a resulting total leaf area per plant of 1399 ± 94 cm2. The senescent attached leaves at the base of the plant contribute to limit leaf transpiration by shading soil around the plant. Moreover, the dense C. gymnocarpa leaf pubescence, leaf rolling, the relatively high leaf mass area (LMA = 12.3 ± 1.3 mg cm−2) and leaf tissue density (LTD = 427 ± 44 mg cm−3) contribute to limit leaf transpiration, also postponing leaf death under dry conditions. At the physiological level, a relatively low respiration/photosynthesis ratio (R/PN) in spring results from high R [2.26 ± 0.59 μmol (CO2) m−2 s−1] and PN [12.3 ± 1.5 μmol (CO2) m−2 s−1]. The high photosynthetic nitrogen use efficiency [PNUE = 15.5 ± 0.4 μmol (CO2) g−1 (N) s−1] shows the large amount of nitrogen (N) invested in the photosynthetic machinery of new leaves, associated to a high chlorophyll content (Chl = 35 ± 5 SPAD units). On the contrary, the highest R/PN ratio (1.75 ± 0.19) in summer is due to a significant PN decrease and increase of R in response to drought. The low PNUE [1.5 ± 0.2 μmol (CO2) g−1 (N) s−1] in this season is indicative of a greater N investment in leaf cell walls which may contribute to limit transpiration. On the contrary, the low R/PN ratio (0.05 ± 0.02) in winter is resulting from the limited enzyme activity of the respiratory apparatus [R = 0.23 ± 0.08 μmol (CO2) m−2 s−1] while the low PNUE [3.5 ± 0.2 μmol (CO2) g−1 (N) s−1] suggests that low temperatures additionally limit plant production. The experiment of the imposed water stress confirms that the C. gymnocarpa growth capability is in conformity with the severe conditions of its natural habitat, likewise as it may be the case with others narrow endemic species that have occupied niches with similar extreme conditions.  相似文献

Our study aimed to test the ability of aquatic plants to use bicarbonate when acclimated to three different bicarbonate concentrations. To this end, we performed experiments with the three species Ceratophyllum demersum, Egeria densa, Lagarosiphon major to determine photosynthetic rates under varying bicarbonate concentrations. We measured bicarbonate use efficiency, photosynthetic performance and respiration. For all species, our results revealed that photosynthetic rates were highest in replicates grown at low alkalinity. Thus, E. densa had approx. five times higher rates at low (264 ± 15 μmol O2 g−1 DW h−1) than at high alkalinity (50 ± 27 μmol O2 g−1 DW h−1), C. demersum had three times higher rates (336 ± 95 and 120 ± 31 μmol O2 g−1 DW h−1), and L. major doubled its rates at low alkalinity (634 ± 114 and 322 ± 119 μmol O2 g−1 DW h−1). Similar results were obtained for bicarbonate use efficiency by E. densa (136 ± 44 and 43 ± 10 μmol O2 mequiv. L−1 g−1 DW h−1) and L. major (244 ± 29 and 82 ± 24 μmol O2 mequiv. L−1 g−1 DW h−1). As to C. demersum, efficiency was high but unaffected by alkalinity, indicating high adaptation ability to varied alkalinities. A pH drift experiment supported these results. Overall, our results suggest that the three globally widespread worldwide species of our study adapt to low inorganic carbon availability by increasing their efficiency of bicarbonate use.  相似文献

The ontogenetic changes of MAAs in the soft coral Heteroxenia fuscescens was studied in relation to their symbiotic state (azooxanthellate vs. zooxanthellate) under different temperature conditions in the Gulf of Eilat, northern Red Sea. The HPLC chromatograms for extracts of the planulae, azoo- and zooxanthellate primary polyps of H. fuscescens from all dates of collection yielded a single peak at 320 nm that has been identified as the compound palythine. Concentration of palythine in planulae at 23 °C was 7.57 ± 1 nmol mg− 1 protein and at 28 °C reached 17.29 ± 1 nmol × mg− 1 protein. Concentration of palythine in azooxanthellate primary polyps was 16.4 ± 3 nmol × mg− 1 protein and 28.37 ± 2.8 nmol × mg− 1 protein at 23 °C and 28 °C respectively. The palythine concentration for zooxanthellate primary polyps at 23 °C was 13 ± 3 nmol × mg− 1 protein and at 28 °C 32.7 ± 2 nmol mg− 1 protein. Palythine concentrations were significantly higher at 28 °C in the different animal groups and correlated linearly with the ambient collection temperature. This study shows for the first time that UVR and temperature act synergistically and affect the MAA levels of early life-history stages of soft corals.  相似文献

This study investigated the absorption of arsenic (As), sulfur (S), and phosphorus (P) in the desert plant Chilopsis linearis (Desert willow). A comparison between an inbred line (red flowered) and wild type (white flowered) plants was performed to look for differential responses to As treatment. One month old seedlings were treated for 7 days with arsenate (As2O5, AsV) at 0, 20, and 40 mg AsV L−1. Results from the ICP-OES analysis showed that at 20 mg AsV L−1, red flowered plants had 280 ± 11 and 98 ± 7 mg As kg−1 dry wt in roots and stems, respectively, while white flowered plants had 196 ± 30 and 103 ± 13 mg As kg−1 dry wt for roots and stems. At this treatment level, the concentration of As in leaves was below detection limits for both plants. In red flowered plants treated with 40 mg AsV L−1, As was at 290 ± 77 and 151 ± 60 mg As kg−1 in roots and stems, respectively, and not detected in leaves, whereas white flowered plants had 406 ± 36, 213 ± 12, and 177 ± 40 mg As kg−1 in roots, stems, and leaves. The concentration of S increased in all As treated plants, while the concentration of P decreased in roots and stems of both types of plants and in leaves of red flowered plants. X-ray absorption spectroscopy analyses demonstrated partial reduction of arsenate to arsenite in the form of As-(SX)3 species in both types of plants.  相似文献

The Iberian Peninsula encompasses more than 80% of the species richness of European aquatic ranunculi. The floristic diversity of the phytocoenosis characterised by aquatic Ranunculus and the main physical–chemical factors of the water were studied in 43 localities of the central Iberian Peninsula. Four aquatic Ranunculus communities are found in most of the aquatic environments. These are species-poor and have an uneven distribution: three species of Batrachium are heterophyllous and their communities are distributed in different aquatic ecosystems on silicated substrates; one species is homophyllous and its community occurs in various aquatic ecosystems with carbonated waters. In the Mediterranean climate, Ranunculus species are present in different habitats, as shown by the results of all the statistical analyses. Ranunculus trichophyllus communities occur in base-rich waters with a high buffering capacity (2273.44 ± 794.57 mg CaCO3 L−1) and a high concentration of cations (Ca2+, 121 ± 33.12 mg L−1; Mg2+, 71.64 ± 82.77 mg L−1), nitrates (2.89 ± 4.80 mg L−1), ammonium (2.19 ± 1.36 mg L−1) and sulphates (216.25 ± 218.54 mg L−1). Ranunculus penicillatus communities are found in flowing waters with a high concentration of phosphates (0.48 ± 0.6 mg L−1) and intermediate buffering capacity (683.66 ± 446.76 mg CaCO3 L−1). Both Ranunculus pseudofluitans and Ranunculus peltatus communities grow in waters with low buffering capacity (R. pseudofluitans, 385.91 ± 209.2 mg CaCO3 L−1; R. peltatus, 263.3 ± 180.36 mg CaCO3 L−1), and a low concentration of cations (R. pseudofluitans: Ca2+, 12.57 ± 9.42 mg L−1; Mg2+, 3.42 ± 1.67 mg L−1; R. peltatus: Ca2+, 15 ± 18.26 mg L−1; Mg2+, 6.26 ± 8.89 mg L−1) and nutrients (R. pseudofluitans: nitrates, 0.23 ± 0.2 mg L−1; phosphates, 0.09 ± 0.1 mg L−1; R. peltatus: nitrates, 0.19 ± 0.21 mg L−1; phosphates, 0.09 ± 0.12 mg L−1); the first in flowing waters, the latter in still waters.  相似文献

Patch dynamics of the Mediterranean slow-growing seagrass Posidonia oceanica was studied in two shallow sites (3–10 m) of the Balearic Archipelago (Spain) through repeated censuses (1–2 year−1). In the sheltered site of Es Port Bay (Cabrera Island), initial patch density (October 2001) was low: 0.05 patches m−2, and the patch size (number of shoots) distribution was bimodal: most of the patches had less than 6 shoots or between 20 and 50 shoots. Mean patch recruitment in Es Port Bay (0.006 ± 0.002 patches m−2 year−1) exceeded mean patch loss (0.001 ± 0.001 patches m−2 year−1), yielding positive net patch recruitment (0.004 ± 0.003 patches m−2 year−1) and a slightly increased patch density 3 years later (July 2004, 0.06 patches m−2). In the exposed site of S’Estanyol, the initial patch density was higher (1.38 patches m−2, August 2003), and patch size frequency decreased exponentially with size. Patch recruitment (0.26 patches m−2 year−1) and loss (0.24 patches m−2 year−1) were high, yielding a slightly increased patch density in the area 1 year later (October 2004, 1.40 patches m−2). Most recruited patches consisted of rooting vegetative fragments of 1–2 shoots. Seedling recruitment was observed in Summer 2004 at both sites. Episodic, seedling recruitment comprised 30% and 25% of total patch recruitment in Es Port Bay and S’Estanyol, respectively. Patch survival increased with patch size and no direct removal was observed among patches of 5 shoots or more. Most patches grew along the study, shifting patch distribution towards larger sizes. Within the size range studied (1–150 shoots), absolute shoot recruitment (shoots year−1) increased linearly with patch size (R2 = 0.64, p < 4 × 10−5, N = 125), while specific shoot recruitment was constant (about 0.25 ± 0.05 year−1), although its variance was large for small patches. Given the slow growth rate and the high survival of patches with 5 or more shoots, even the low patch recruitment rates reported here could play a significant role in the colonisation process of P. oceanica.  相似文献

We studied the decolorization of malachite green (MG) by the fungus Cunninghamella elegans. The mitochondrial activity for MG reduction was increased with a simultaneous increase of a 9-kDa protein, called CeCyt. The presence of cytochrome c in CeCyt protein was determined by optical absorbance spectroscopy with an extinction coefficient (E550-535) of 19.7 ± 6.3 mM−1 cm−1 and reduction potential of + 261 mV. When purified CeCyt was added into the mitochondria, the specific activity of CeCyt reached 440 ± 122 μmol min−1 mg−1 protein. The inhibition of MG reduction by stigmatellin, but not by antimycin A, indicated a possible linkage of CeCyt activity to the Qo site of the bc1 complex. The RT-PCR results showed tight regulation of the cecyt gene expression by reactive oxygen species. We suggest that CeCyt acts as a protein reductant for MG under oxidative stress in a stationary or secondary growth stage of this fungus.  相似文献

Calcification and primary production responses to irradiance in the temperate coralline alga Lithothamnion corallioides were measured in summer 2004 and winter 2005 in the Bay of Brest. Coralline algae were incubated in dark and clear bottles exposed to different irradiances. Net primary production reached 1.5 μmol C g−1 dry wt h−1 in August and was twice as high as in January–February. Dark respiration showed significant seasonal variations, being three-fold higher in summer. Maximum calcification varied from 0.6 μmol g−1 dry wt h−1 in summer 2004 to 0.4 μmol g−1 dry wt h−1 in winter 2005. According to PE curves and the daily course of irradiance, estimated daily net production and calcification reached 131 μg C g−1 dry wt and 970 μg CaCO3 g−1 dry wt in summer 2004, and 36 μg C g−1 dry wt and 336 μg CaCO3 g−1 dry wt in winter 2005. The net primary production of natural L. corallioides populations in shallow waters was estimated at 10–600 g C m−2 y−1, depending on depth and algal biomass. The mean annual calcification of L. corallioides populations varied from 300 to 3000 g CaCO3 m−2. These results are similar to those reported for tropical coralline algae in terms of carbon and carbonate productivity. Therefore, L. corallioides can be considered as a key element of carbon and carbonate cycles in the shallow coastal waters where they live.  相似文献

Changes in photosynthetic pigment ratios showed that the Chlorophyll d-dominated oxyphotobacterium Acaryochloris marina was able to photoacclimate to different light regimes. Chl d per cell were higher in cultures grown under low irradiance and red or green light compared to those found when grown under high white light, but phycocyanin/Chl d and carotenoid/Chl d indices under the corresponding conditions were lower. Chl a, considered an accessory pigment in this organism, decreased respective to Chl d in low irradiance and low intensity non-white light sources. Blue diode PAM (Pulse Amplitude Modulation) fluorometry was able to be used to measure photosynthesis in Acaryochloris. Light response curves for Acaryochloris were created using both PAM and O2 electrode. A linear relationship was found between electron transport rate (ETR), measured using a PAM fluorometer, and oxygen evolution (net and gross photosynthesis). Gross photosynthesis and ETR were directly proportional to one another. The optimum light for white light (quartz halogen) was about 206 ± 51 μmol m− 2 s− 1 (PAR) (Photosynthetically Active Radiation), whereas for red light (red diodes) the optimum light was lower (109 ± 27 μmol m− 2 s− 1 (PAR)). The maximum mean gross photosynthetic rate of Acaryochloris was 73 ± 7 μmol mg Chl d− 1 h− 1. The gross photosynthesis/respiration ratio (Pg/R) of Acaryochloris under optimum conditions was about 4.02 ± 1.69. The implications of our findings will be discussed in relation to how photosynthesis is regulated in Acaryochloris.  相似文献

Colurella dicentra clones isolated from bay water in the Mississippi Gulf Coast were cultured with artificial seawater. Experiments were conducted to determine the effects of six algae species (Nannochloropsis oculata, Tetraselmis chuii, Chaetoceros gracilis, Rhodomonas salina, Isochrysis galbana, and Prorocentrum micans), six C. gracilis densities, and six N. oculata densities (25,000, 50,000, 100,000, 250,000, 500,000, and 1,000,000 cells ml− 1) on C. dicentra population growth. Algae type influenced rotifer production (p < 0.0001). C. gracilis treatment (9120 ± 3351SD) produced the highest number of rotifers followed by N. oculata (5760 ±2232SD). P. micans had the lowest number of rotifers, although not significantly different from numbers in T. chuii, R. salina, and I. galbana treatments (p > 0.05).The population growth rate (r) varied with algae species treatment. The highest values were recorded for C. gracilis treatment (0.22 to 0.26 d− 1), followed by N. oculata (0.21 to 0.24 d− 1), and the lowest for P. micans (− 0.19 to 0.14 d− 1). C. gracilis and N. oculata densities had significant effects (p < 0.0001) on C. dicentra population growth. The highest rotifer production was recorded at a C. gracilis density of 100,000 cells ml− 1, followed by 250,000 cells ml− 1 and 50,000 cells ml− 1. Algae densities of 500,000 cells ml− 1 and above produced the lowest rotifer numbers. Population growth rate (r) varied with C. gracilis densities. The highest values were observed for C. gracilis concentrations of 100,000 cells ml− 1 (0.17 to 0.19 d− 1), and the lowest for concentrations of 500,000 cells ml− 1 and above (− 0.19 to 0.09 d− 1). The 100,000 cells ml− 1N. oculata density gave the highest rotifer production followed by 50,000, 250,000, 25,000, and 500,000 cells ml− 1. Algae densities of 1,000,000 cells ml− 1 produced the lowest rotifer numbers. Population growth rate (r) varied with N. oculata densities, with the highest values obtained for algae densities of 100,000 cells ml− 1 (0.35 to 0.40 d− 1), and the lowest for concentrations of 1,000,000 cells ml− 1 (0.05 to 0.012 d− 1). This is the first report of C. dicentra in Mississippi Coastal waters, and perhaps the smallest marine rotifer species (93 by 49 μm) ever cultured successfully.  相似文献

Sublevel structure of the 8S7/2 electronic ground state of anionic bis(phthalocyaninato)gadolinium(III) has been determined by simulation analysis of an ESR spectrum in frozen solution. The simplex multidimensional minimization algorithm was employed to find the zero-field-splitting parameter set giving the minimum RMS error from the observed spectrum. The parameter set {B20,B40,B60} for the potential of D4d symmetry has been determined to be ±{(1.54 ± 0.01)× 10−2 cm−1, (0.9 ± 0.1)× 10−4 cm−1, (−0.6 ± 0.9)× 10−6 cm−1}. The energy difference between the lowest and highest sublevels has been found to be about 0.5 cm−1.  相似文献

The most extensively studied ficins have been isolated from the latex of Ficus glabrata and Ficus carica. However the proteases (ficins) from other species are less known. The purification and characterization of a protease from the latex of Ficus racemosa is reported. The enzyme purified to homogeneity is a single polypeptide chain of molecular weight of 44,500 ± 500 Da as determined by MALDI-TOF. The enzyme exhibited a broad spectrum of pH optima between pH 4.5-6.5 and showed maximum activity at 60 ± 0.5 °C. The enzyme activity was completely inhibited by pepstatin-A indicating that the purified enzyme is an aspartic protease. Far-UV circular dichroic spectra revealed that the purified enzyme contains predominantly β-structures. The purified protease is thermostable. The apparent Tm, (mid point of thermal inactivation) was found to be 70 ± 0.5 °C. Thermal inactivation was found to follow first order kinetics at pH 5.5. Activation energy (Ea) was found to be 44.0 ± 0.3 kcal mol−1. The activation enthalpy (ΔH), free energy change (ΔG) and entropy (ΔS) were estimated to be 43 ± 4 kcal mol−1, −26 ± 3 kcal mol−1 and 204 ± 10 cal mol−1 K−1, respectively. Its enzymatic specificity studied using oxidized B chain of insulin indicates that the protease preferably hydrolyzed peptide bonds C-terminal to glutamate, leucine and phenylalanine (at P1 position). The broad specificity, pH optima and elevated thermal stability indicate the protease is distinct from other known ficins and would find applications in many sectors for its unique properties.  相似文献

Vermicomposting potential of Allolobophora parva is well proven in recent experiments but little is known about its growth and reproduction performance. Efforts were made in this study to assess the biological productivity of A. parva in cattle waste solids under laboratory conditions. The growth and reproduction performance of A. parva was monitored up to its termination state in experimental beddings. A. parva was weighed weekly and cocoons produced during the interval were also counted. The maximum individual biomass and maximum growth rate (wt. mg worm−1 week−1) was 190.9 ± 0.07 mg (after 13 weeks) and 2.66 (after 12th week), respectively. A. parva showed the maximum values of cocoon number (within a week) and reproduction rate: 26 ± 1.12 and 0.74 ± 0.05 cocoon worm−1 week−1, respectively, during the 8th week of vermiculture. Cocoon production in earthworms was terminated after the 17th week of culture. Data suggested that A. parva may acts as a potential candidate to convert negligible organic waste resources into worm biomass for sustainable environmental management.  相似文献

“Reduced minus oxidized” difference extinction coefficients Δ? in the α-bands of Cyt b559 and Cyt c550 were determined by using functionally and structurally well-characterized PS II core complexes from the thermophilic cyanobacterium Thermosynechococcus elongatus. Values of 25.1 ± 1.0 mM−1 cm−1 and 27.0 ± 1.0 mM−1 cm−1 were obtained for Cyt b559 and Cyt c550, respectively. Anaerobic redox titrations covering the wide range from −250 up to +450 mV revealed that the heme groups of both Cyt b559 and Cyt c550 exhibit homogenous redox properties in the sample preparation used, with Em values at pH 6.5 of 244 ± 11 mV and −94 ± 21 mV, respectively. No HP form of Cyt b559 could be detected. Experiments performed on PS II membrane fragments of higher plants where the content of the high potential form of Cyt b559 was varied by special treatments (pH, heat) have shown that the α-band extinction of Cyt b559 does not depend on the redox form of the heme group. Based on the results of this study the Cyt b559/PSII stoichiometry is inferred to be 1:1 not only in thermophilic cyanobacteria as known from the crystal structure but also in PSII of plants. Possible interrelationships between the structure of the QB site and the microenvironment of the heme group of Cyt b559 are discussed.  相似文献
