Splenectonuzed rats infected with Angiostrongylus cantonensis had more and longer worms and lower anti-A. cantonensis antibody titres compared to intact and sham operated rats. Splenectomy, however, had no effect on the capacity of rats to mount a cell-mediated immune response, as assessed by the in vitro uptake of tritiated thymidine by peripheral blood lymphocytes following stimulation by PHA and adult worm antigen. Adoptive protection and antibody production against A. cantonensis could be transferred with immune spleen cells. This protection was strongly dose and time dependent. Protection was adoptively conferred with 3 × 109 spleen cells transferred 7 days before infection.  相似文献

Beaver J. P. and Dobson C. 1978. Acetylcholinesterase levels in Angiostrongylus cantonensis in relation to the immune response in rats. International Journal for Parasitology8: 9–13. Angiostrongylus cantonensis larvae and adult nematodes synthesize three acetylcholinesterase (AChE) isozymes. The Km of this isozyme complex changes with the development and migrations of the parasite in the rat host. The levels of parasite AChE changed as the development of A. cantonensis progressed; increasing quantities of AChE were found in young adult A. cantonensis from the brain of rats. After migration to the pulmonary arteries, the quantity of AChE in the parasite was reduced and continued to decline in the aging parasite. Anti-A. cantonensis antibody inhibited parasite AChE activity; this inhibition of the parasite AChE activity changed at stages during development of the parasite which suggested variation in parasite AChE isozyme levels. Haemagglutinating anti-A. cantonensis antibody appeared in the serum of infected rats when the parasites commenced to lay eggs and increased in titre thereafter until 103 days after infection.  相似文献

International Journal for Parasitology9: 97–103. A simple procedure for the transplantation of young adult Angiostrongylus cantonensis into the rat pulmonary vessels was described and its potential usefulness was discussed. Acquired resistance to A. cantonensis was conferred on rats by infection with 30 third-stage larvae or by a similar infection followed by treatment with thiabendazole during 14–19 days postinfection. Nearly comparable levels of resistance to reinfection were also observed in rats that were transplanted with 30 young adult worms, collected from the brain surface of donor animals at 21 and 22 days postinfection. The time-course development of reaginic, hemagglutinating and precipitating antibodies differed in these groups of rats although there appeared to be no positive correlation between the appearance of these antibodies and acquired resistance.  相似文献

Changes in peripheral blood leucocytes were followed in male August rats given one or two infections with the parasitic nematode, Nippostrongylus brasiliensis. During the initial infection, there was a biphasic increase in total numbers of leucocytes, lymphocytes, neutrophils, large mononuclear cells, and eosinophils. All except eosinophils fell rapidly to normal levels as the parasites were expelled, but eosinophils were elevated much longer. All these cell types increased in number to a single peak 5 days after reinfection. Basophils were detected at very low levels in uninfected rats (0.06% or 11600 leucocytes) and increased in number to a peak 13 days after initial infection, at which time they represented about 4.5% of total leucocytes, an 80-fold increase compared with the number in normal rats. In reinfected rats, the basophilia occurred more rapidly than in a primary infection, suggesting that the appearance of these cells in the circulation is probably an immunologically mediated event.  相似文献

Lymph node cells from the posterior cervical and mesenteric lymph nodes of immune rats passively protected syngeneic recipient rats against Angiostrongylus cantonensis better than cells from the spleen, thymic and inguinal lymph nodes either as reduced worms burdens and/or stunted growth. No antibody was detected in the sera of recipient rats after transfer of the cells and before infection which suggested that the protection was cell- rather than antibody-mediated.  相似文献

The nematode Angiostrongylus cantonensis is a rat lungworm, a zoonotic pathogen that causes human eosinophilic meningitis and ocular angiostrongyliasis characteristic of rat lungworm (RLW) disease. Definitive diagnosis is made by finding and identifying A. cantonensis larvae in the cerebral spinal fluid or by using a custom immunological or molecular test. This study was conducted to determine if genomic DNA from A. cantonensis is detectable by qPCR in the blood or tissues of experimentally infected rats. F1 offspring from wild rats were subjected to experimental infection with RLW larvae isolated from slugs, then blood or tissue samples were collected over multiple time points. Blood samples were collected from 21 rats throughout the course of two trials (15 rats in Trial I, and 6 rats in Trial II). In addition to a control group, each trial had two treatment groups: the rats in the low dose (LD) group were infected by approximately 10 larvae and the rats in the high dose (HD) group were infected with approximately 50 larvae. In Trial I, parasite DNA was detected in cardiac bleed samples from five of five LD rats and five of five HD rats at six weeks post-infection (PI), and three of five LD rats and five of five HD rats from tail tissue. In Trial II, parasite DNA was detected in peripheral blood samples from one of two HD rats at 53 minutes PI, one of two LD rats at 1.5 hours PI, one of two HD rats at 18 hours PI, one of two LD rats at five weeks PI and two of two at six weeks PI, and two of two HD rats at weeks five and six PI. These data demonstrate that parasite DNA can be detected in peripheral blood at various time points throughout RLW infection in rats.  相似文献

Immunity to the coccidial parasite, Eimeria nieschulzi, in CD-F rats was assessed by the numbers of oocysts shed in relation to the time after inoculation. Intravenous injections of syngeneic thoracic duct lymphocytes (TDL) from immunized rats elicited various degrees of adoptive immunity against primary infections of E. nieschulzi. Of the 16 rats injected with 109 sensitized TDL, 7 were totally immune to a subsequent challenge by the parasite. This number of injected TDL also raised the serum antibody level to that of immune rats. Contact with immune TDL was deleterious to sporozoites of E. nieschulzi in vitro and produced immunocytoadherence of parasite to cell.  相似文献

A method of affinity chromatography developed for the purification of species-specific antigens from Toxocara canis adult worms is described. Immunochemical analyses by polyacrylamide gel electrophoresis, immunoelectrophoresis and immunodiffusion showed that ‘pure’ antigen contained fewer but more specific proteins than ‘crude’ antigen. Purified antigens and parasite sections from four parasite species (Toxocara canis, Dirofilaria immitis, Angiostrongylus cantonensis and Ascaris lumbricoides) were used in immunofluorescence tests to measure serum antibody levels in animals with natural or experimental T. canis infections and people with zoonotic toxocariasis. ‘Pure’ antigen showed higher specificity and sensitivity than ‘crude’ antigen in serological testing.  相似文献

To study immunological responses in chronic filarial infections, a model utilizing inbred Lewis rats infected with Brugia pahangi was developed. Microfilaria were found in the bloodstream of over 90% of the rats by 16 weeks of infection. Using in vitro lymphocyte blastogenesis, cell-mediated immune responses of blood, splenic, and mesenteric node lymphocytes were followed during 1.5 years of infection. Lymphocyte responses to antigen prepared from infective stage filarial larvae were detectable in the early weeks of infection, whereas responses to microfilarial antigen only developed late as microfilaremia waned. Lymphocyte responses to antigen from adult filaria vacillated during the infection. With the mitogens, phytohemagglutinin, pokeweed mitogen, and bacterial lipopolysaccharide, periods of B and T-cell hyporesponsiveness were demonstrable. Between 16 and 36 weeks of infection node lymphocytes from many rats were unresponsive to all mitogens and antigens. The model of B. pahangi in inbred rats offers advantages for immunological studies of filarial infections.  相似文献

Background

Angiostrongylus cantonensis is a lungworm of rats (Muridae) that is the causative agent of human cerebral angiostrongyliasis. The life cycle of A. cantonensis involves rats and mollusks as the definitive and intermediate hosts, respectively. This study was designed to increase the knowledge about the occurrence and distribution of A. cantonensis in its definitive host in the Canary Islands, using parasitological and serological analysis in different areas and age groups.

Methodology/Principal Findings

Between 2009 and 2010, 54 black rats (Rattus rattus) from Tenerife were captured from six human-inhabited areas and sera samples were obtained. The lung nematodes were identified by morphological and molecular tools as A. cantonensis. The 31-kDa glycoprotein antigen was purified from A. cantonensis adult worms by electrophoresis and electroelution. Of the 54 tested rodents, 30 showed IgG antibodies against A. cantonensis 31-kDa antigen by ELISA. Therefore, the overall seroprevalence was 55.6% (95% CI: 42.4–68). Seroprevalent rodents were found in all the 6 areas. This 31-kDa antigen was not recognized by some sera of rats infected by other helminth species (but not A. cantonensis). Seroprevalence of IgG antibodies against A. cantonensis and prevalence based on the presence of adult worms showed significant correlation (R2 = 0.954, p<0.05).

Conclusions/Significance

The present results could indicate a high prevalence of A. cantonensis in Tenerife and suggest the inclusion of two new zones in the distribution area of the parasite. The commonness and wide distribution of A. cantonensis in rats implies the presence of intermediate hosts, indicating that humans may be at risk of getting infected.  相似文献

Angiostrongylus cantonensis and Angiostrongylus mackerrasae are metastrongyloid nematodes that infect various rat species. Terrestrial and aquatic molluscs are intermediate hosts of these worms while humans and dogs are accidental hosts. Angiostrongylus cantonensis is the major cause of angiostrongyliasis, a disease characterised by eosinophilic meningitis. Although both A. cantonensis and A. mackerrasae are found in Australia, A. cantonensis appears to account for most infections in humans and animals. Due to the occurrence of several severe clinical cases in Sydney and Brisbane, the need for epidemiological studies on angiostrongyliasis in this region has become apparent. In the present study, a conventional PCR and a TaqMan assay were compared for their ability to amplify Angiostrongylus DNA from DNA extracted from molluscs. The TaqMan assay was more sensitive, capable of detecting the DNA equivalent to one hundredth of a nematode larva. Therefore, the TaqMan assay was used to screen molluscs (n=500) of 14 species collected from the Sydney region. Angiostrongylus DNA was detected in 2 of the 14 mollusc species; Cornu aspersum [14/312 (4.5%)], and Bradybaenia similaris [1/10 (10%)], which are non-native terrestrial snails commonly found in urban habitats. The prevalence of Angiostrongylus spp. was 3.0% ± 0.8% (CI 95%). Additionally, experimentally infected Austropeplea lessoni snails shed A. cantonensis larvae in their mucus, implicating mucus as a source of infection. This is the first Australian study to survey molluscs using real-time PCR and confirms that the garden snail, C. aspersum, is a common intermediate host for Angiostrongylus spp. in Sydney.  相似文献

There is limited knowledge about parasites of the endangered Ryukyu long-furred rat, Diplothrix legata (Murinae, Rodentia) endemic to Okinawa, Tokunoshima, and Amami-Oshima Islands in Japan. In the present study, postmortem histopathological examination of an individual found on Amami-Oshima Island revealed a mixed helminth infection of Calodium hepaticum, Hydatigera taeniaeformis, and Angiostrongylus cantonensis. These helminths are considered non-native to Amami-Oshima Island and are maintained by invasive mammals, such as non-native rats and outdoor cats. This observation presents a new host record for C. hepaticum and H. taeniaeformis and the first record of A. cantonensis in Ryukyu long-furred rat on Amami-Oshima Island.  相似文献

Adams D. B. and Davies H. I. 1982. Enhanced resistance to infection with Haemonchus contortus in sheep treated with a corticosteroid. International Journal for Parasitology12: 523–529. Fewer worms established from experimental infections with Haemonchus contortus in either immune or naive sheep or in sheep of undefined immune status given the corticosteroid, dexamethasone, around the time of challenge. In four experiments, the number of adult worms present in sheep treated with dexamethasone ranged from 32 to 36% of that in untreated animals. That the phenomenon did not stem from direct action of dexamethasone on the worms themselves was demonstrated by comparing continuous treatment with the drug from infection until patency with treatment given at infection and four days later. Sheep continuously treated with dexamethasone harboured similar numbers of worms as the infection controls whereas fewer parasites were found in sheep given dexamethasone around the time of infection. The results imply that active regulation of immunological unresponsiveness operates in sheep during infection with H. contortus and that disruption of immunoregulation by dexamethasone released protective responses thereby decreasing worm burdens. Because suppressor lymphocytes are implicated, cellular perturbations following treatment with dexamethasone were investigated. Dexamethasone did not cause marked lymphopenia. It, however, reduced blastogenic responses by lymphocytes to con A but not PHA. Comparison of responses to these mitogens in cells from blood and lymph demonstrates that con A and PHA-reactivity resides in identifiably different cell populations in sheep.  相似文献

Three patients with eosinophilic meningitis (EoM) were investigated in two hospitals in Porto Alegre, Southern Brazil. These patients had a common exposure after the ingestion of raw mollusks in a religious ritual. Two of them had an uncommon presentation with intense lower distal extremities pain and small fiber neuropathy as defined by an electroneuromyography (ENMG) study. All three patients were positive for Angiostrongylus cantonensis serology and recovered after antihelminthic and anti-inflammatory treatment. Increased awareness of A. cantonensis infection is important to avoid new infections and to improved recognition and handling of cerebral angiostrongyliasis.  相似文献

The most common route of infection of Biomphalaria glabrata experimentally exposed to first-stage larvae of Angiostrongylus cantonensis is via penetration of the gastric and prointestinal walls after ingestion. Subsequently, most parasites migrate through the kidney and rectal ridge to the mantle collar and head-foot of the gastropod at which sites they become lodged. Nematodes are encapsulated in B. glabrata beginning by 24/2-48 h post-infection. Encapsulation occurs as a two-phase process involving (1) initial infiltration and aggregation of basophilic hemolymph cells around the parasite and (2) subsequent transformation of such cellular aggregates into more fibrous-appearing nodules. The small number of parasites which localize in such tissues as the rectal ridge, kidney, or vascular connective tissue near the gonad are similarly encapsulated.  相似文献

Trypanosoma musculi infections in CBA mice consist of a phase of increasing parasitemia during which dividing forms of the parasite are present in the blood, followed by a period when only nondividing trypomastigotes are seen. A second crisis terminates the blood infection and leaves the host immune, but small numbers of trypanosomes, including multiplicative forms, persist in the kidneys for many months. Studies were made involving infections in T-lymphocyte deprived mice, the effects of passive transfer of serum and cells, measurement of DNA synthesis by the parasite, serological responses, and in vitro effects of serum on the trypanosomes. These indicated that the initial check on the increase in blood parasitemia is due in part to two humoral factors. One of these has a trypanocidal effect (this is thought to be an IgM antibody) while the other, which may be an IgG antibody, is the ablastin that inhibits further reproduction by the parasite. Both trypanocidal and ablastic effects were demonstrable in the serum of immune mice yet the parasite was still able to survive and multiply in the kidneys.  相似文献

Experiments were conducted defining the possible basis for increased susceptibility of alloxan-treated and genetically diabetic C57Bl/KsJ mice to infections with Candida albicans. Alloxan monohydrate (175 mg/kg) produced a prolonged state of hyperglycemia, which persisted through 31 days. Parameters of immune responses varied depending upon the interval between alloxan administration and testing. In the period immediately following alloxan treatment (1–14 days), the numbers of lymphocytes in the thymus and spleen were reduced, the numbers of recoverable peritoneal macrophages were decreased, and the mice showed an increased susceptibility to intravenous infection with C. albicans. In contrast, splenic lymphocytes responded normally to stimulation with Con A, and in vitro phagocytosis of yeast cells by peritoneal macrophages was normal. Also, in vivo production of such lymphokines as migration inhibitory factor (MIF) and macrophage activating factor (MAF), as well as delayed hypersensitivity footpad responses, was generally within the normal range. In the later phase of alloxan diabetes (21–28 days) after administration of alloxan, lymphoid cellularity recovered progressively and the numbers of recoverable peritoneal macrophages were normal. However, these mice still showed an increased susceptibility to C. albicans infection. Genetically diabetic mice (C57Bl/KsJ, db/db) were abnormal in virtually all the assays involving cell-mediated immunity. The numbers of lymphocytes and peritoneal macrophages were markedly decreased, lymphoid cells responded poorly to Con A, and the phagocytosis of yeast cells by macrophages was depressed. The in vivo production of lymphokines and footpad responses of the delayed-type hypersensitivity were depressed. In addition, these mice were highly susceptible to intravenous infection with C. albicans.  相似文献

The influence of Mesocestoides corti on subsequent Angiostrongylus cantonensis infection in mice (C57BL/6 and BALB/c) was assessed. Both strains of mice infected with M. corti demonstrated a peak blood eosinophilia at around 3 weeks post-infection (p.i.). C57BL/6 and BALB/c mice primarily infected with M. corti were given A. cantonensis infection 18 days later, but pre-existing M. corti infection did not affect the recovery of intracranial worms of A. cantonensis at day 21 p.i. BALB/c mice with mixed parasite infections showed low morbidity and mortality as compared with mice singly infected with A. cantonensis and some mice demonstrated a pulmonary migration of intracranial worms. In C57BL/6 mice, intracranial worms were killed and thus all mice survived. C57BL/6 mice with mixed parasite infections failed to resist A. cantonensis reinfection. The blastogenic responses of spleen cells against A. cantonensis antigen were lower in BALB/c than in C57BL/6 mice and mixed parasite infections also resulted in less blastogenic responses against both concanavalin A and A. cantonensis antigen than monoinfection. The recovery of M. corti biomass was significantly higher in mice with mixed parasite infections than mice with monoinfection with M. corti. These data suggest a distinct difference in response to A. cantonensis infection between C57BL/6 and BALB/c mice, and the induction of immunosuppression in both mouse strains following M. corti infection. Blood eosinophilia provoked by M. corti infection is not directly associated with the killing of worms in subsequent A. cantonensis infection.  相似文献

We investigated cellular immune responses of mice infected with the apicomplexan parasite Eimeria falciformis in order to characterise protective immune mechanisms and effector functions. Adoptive transfer experiments with mesenterial lymph node cells (MLNC) from immune donor mice were performed, and the oocyst output monitored after challenge infection. Phenotypical analysis by fluorescence cytometry and T cell proliferation assay showed that already from day four post infection E. falciformis-specific lymphocytes were present in the MLN. The frequency of parasite-specific, IFN-γ producing CD4+ and CD8+ cells increased in this period by 9.8% and 16.4%, respectively. Infection experiments with IFN-γ deficient mice revealed that IFN-γ is involved in resistance to primary and secondary infection. Transfer of total MLNC from immune donors reduced the oocyst output by 65–74%, as compared to the oocyst output of animals transferred with cells from naïve donors. Transfer of CD8+ cells inhibited parasite development resulting in a reduction of oocyst numbers by 42–64%, whereas CD4+ cells showed no influence on resistance to reinfection.  相似文献
