The short-wave fluorescence of bidistilled water treated in the cathode and anode chambers of two types of electrolysers made from different materials was investigated by fluorescence spectroscopy. It was shown that the electrochemical treatment of water did not induce intrinsic short-wave flyorescence of catholyte and anolyte in the case of quartz glass electrolyser. The increase observed in the intensity of intrinsic short-wavelength fluorescence of bidistilled water in the catholyte and anolyte of the electrolyser made from technical organic glass was caused by microcontaminations released from electrolyser material in the samples.  相似文献

The biological activity of the catholyte and anolyte of double distilled water was studied in experiments on the germination of wheat grains in the period from March to May. The activity of the solutions, which was characterized by a growth index, was high early in this period, then decreased almost to zero in the middle of the period, and then increased to about the initial value by the end of the period. Throughout, the efficiency of the anolyte of double distilled water generally exceeded the efficiency of the catholyte. Early and late in the period, the stimulatory effect of the anolyte exceeded that of the catholyte by a factor of 5–5.5. The changes in the biological activity of the catholyte and anolyte of double distilled water were also compared with the changes in the biological activity of the catholyte of nutrient medium M9. The stimulatory effect of the catholyte of the nutrient medium was evaluated from the change in the growth of E. coli cells. Early in the period at a cultivation temperature of 20°C, the stimulatory effect determined from the increase in the optical density of the cell suspension in the experiment with respect to a reference value was 55–60%. Next, the stimulatory effect decreased almost to zero in the middle of the period and increased to approximately initial value by the end of the period. It was assumed that the physicochemical mechanisms of action of the catholyte and anolyte of double distilled water on the wheat seed germination and of the catholyte of the nutrient medium on E. coli cell growth are of different nature.  相似文献

Chlorophyll fluorescence measurements were used to test whether externally applied abscisic acid (ABA) has an effect on the desiccation tolerance of lichens and their sensitivity to constant water saturation. A surplus of ABA did not decrease the time required to regain full photosynthetic capacity after prolonged dry periods. However, an effect of ABA could be observed when lichens were permanently hydrated at moderately high temperatures. Lichens suffered less from constant saturation if ABA was added to the incubation medium. These results suggest a positive effect of ABA on membrane function.  相似文献

Using a (8, 8) carbon nanotube (CNT) as a model for nanochannel, we studied the effects of carbonyl (–CO) groups and external electric field on the properties of confined water molecules by molecular dynamics simulation. We analysed the density profiles, the distribution of orientation and the hydrogen bonds of water molecules in the axial and radial directions of the CNT. The results show that –CO groups are more powerful than electric fields on controlling water properties, and property changes induced by –CO groups are less affected by electric fields. Meanwhile, the particular behaviour of water molecules induced by the –CO groups is not limited near the –CO groups, but also expands to the whole CNT.  相似文献

In this study, the fluorescence spectra of sarafloxacin (SAR) under different pH conditions were investigated to determine the structural changes due to protonation that result from change in pH. At pH < 1.02, SAR exists in the H3L2+ form for which the maximum fluorescence emission wavelength was about 455 nm. At pH 1.87–4.94, SAR exists in the H2L+ form in which H3L2+ loses one proton in the nitrogen molecule at the 1‐position in the quinoline ring. Fluorescence intensity was strong and steady and the maximum emission wavelength was 458 nm. At pH 7.14–9.30, the maximum emission wavelengths were gradually blue shifted to 430 nm with increase in pH, here SAR exists in the form of a bipolar ion HL in which H2L+ loses a carboxyl group proton. At pH > 11.6, HL transforms into anionic L? in which HL loses one proton from the piperazine ring, leading to a decrease in fluorescence intensity, and the maximum emission wavelength was red shifted to approximately 466 nm. The two‐step dissociation constant pKa for SAR was calculated, pK a1 was 6.06 ± 0.37 and pK a2 for SAR was 10.53 ± 0.19. In a pH 3.62 buffer solution with quinine sulfate as the reference, the fluorescence quantum yield of SAR at the maximum excitation wavelength of 276 nm was 0.09.  相似文献

分别对具4~5张完全叶桶栽甘蔗品种ROC22#和园林6#叶面喷施黄腐酸,分别为正常供水0(CK1)、0(CK2)、200、400、800、1200mg/L黄腐酸,研究在不同胁迫程度下不同浓度黄腐酸对甘蔗叶片叶绿素荧光参数、丙二醛(MDA)含量变化的影响。结果表明,经过黄腐酸处理后甘蔗叶片的PSⅡ原初光能转换效率(Fv/Fm)、光合量子产额(Yield)、光化学淬灭系数(qP)数值在中度和重度水分胁迫下都高于干旱对照CK2,品种和处理间参数的变化有差异;非光化学淬灭系数(qN)值则与正常供水对照比较没有大的差异。黄腐酸处理后甘蔗叶片MDA含量随处理浓度增大而降低,且明显低于干旱对照CK2。从2个参试品种6个处理浓度综合分析,水分胁迫下喷施黄腐酸可提高ROC22#和园林6#甘蔗苗期的抗旱性,以200、400mg/L处理效果较好。  相似文献

研究了水分胁迫对牛心朴子叶片光合色素及叶绿素荧光动力学参数的影响。结果表明,在长期的水分胁迫中,牛心朴子叶片的叶绿素a(Chl a)、叶绿素b(Chl b)和类胡萝卜素(Car)含量没有下降或下降不明显。直到处理末期才显著下降;叶片叶绿素荧光动力学参数Fo、Fm、Fv、Fv/Fm变化不大,在处理末期各处理Fo降低,轻度、重度水分胁迫的Fm、Fv、Fv/Fm升高。说明K期水分胁迫后牛心朴子的光合功能受到影响,但牛心朴子仍表现出较强的适应干旱的能力。  相似文献

Aims:  To expose bacteria to anolyte and subsequently investigate the effect of anolyte on the protein profiles of treated bacteria.
Methods and Results:  Proteins were extracted from bacteria treated with different concentrations of anolyte and analysed using SDS-PAGE. Fewer and more faint protein bands were observed for concentrated halide anolyte treated bacteria when compared to untreated bacteria while extra protein bands were observed for bacteria exposed to dilute concentrations.
Conclusions:  The undiluted and the 10−1 dilution of halide derived anolyte was effective in killing the test bacteria. Anolyte caused bacterial death by complete destruction of proteins or by causing oxidative stress which resulted in protein fragmentation.
Significance and Impact of the Study:  The results of this study provide information on the antimicrobial mechanism of anolyte on other bacteria for which the information is currently unavailable.  相似文献

In presence of water, the naked eye color of DFPBEN changes from red to yellow, while it turns to green from red under UV light. Molecular dynamics simulations of liquid water were performed at 258K and a density of 1.0?g/cm3 under various applied external electric field, ranging 0~1010?V/m. The influence of external field on structural and dynamical properties of water was investigated. The simple point charge (SPC) model is used for water molecules. An enhancement of the water hydrogen bond structure with increasing strength of the electric field has been deduced from the radial distribution functions and the analysis of hydrogen bonds structure. With increasing field strength, water system has a more perfect structure, which is similar to ice structure. However, the electrofreezing phenomenon of liquid water has not been detected since the self-diffusion coefficient was very large. The self-diffusion coefficient decreases remarkably with increasing strength of electric field and the self-diffusion coefficient is anisotropic.  相似文献

夏蜡梅是浙江省特有的濒危单种属物种.本文研究了不同程度土壤水分胁迫和不同温度处理对2年生盆栽夏蜡梅光合作用的影响.结果表明:轻度和中度水分胁迫下夏蜡梅净光合速率分别下降至对照的92.3%和74.3%,净光合速率的降低主要由气孔限制引起;重度水分胁迫下,净光合速率仅为对照的44.4%,主要由非气孔限制引起.夏季夏蜡梅的光合适宜温度范围在20 ℃~28 ℃,39 ℃下其净光合速率、水分利用效率和最大光化学效率显著降低,暗呼吸速率和蒸腾速率显著升高.随着水分胁迫的加重及处理温度的升高,夏蜡梅光补偿点上升,光饱和点、表观量子效率和最大净光合速率下降.重度水分胁迫及高温是制约夏蜡梅生存的重要环境因子.  相似文献

Abstract

Arsenic (As) is found naturally in the geological strata within the Ba Men Region of Inner Mongolia, China. A study was conducted to compare the total As measurements from two analytical techniques: instrumental neutron activation analysis (INAA) and atomic fluorescence spectrometry (AFS), and to verify nails as an exposure biomarker in this population. In 1999, nail and water samples were collected in a pilot study. Fingernails and toenails were pooled from 32 participants and analysed for total As by both INAA and AFS. Mean nail As values were 14.8±2.4 and 19.4±2.8 µg?g?1 (±SEM) for INAA and AFS, respectively. Results from these two methods were significantly correlated (r=0.93, p<0.0001). In 2000, a second study was conducted and INAA was used to measure total As in toenails from 314 Ba Men residents. Well water samples were collected from 121 households and analysed by AFS. A significant correlation was observed between toenail and well water As (r=0.84, p<0.0001). Based on the results, INAA was significantly correlated with AFS and proved to be a reliable measure of nail As levels. In this population, toenail samples are a useful internal As exposure biomarker from drinking water sources.  相似文献

The function of photosystem II (PSII) during desiccation was investigated via analysis of Chl a fluorescence emission in thalli from Parmelia quercina (Willd.) Vainio, Parmelia acetabulum (Necker) Duby, Ramalina farinacea (L.) Ach., Pseudevernia furfuracea (L.) Zopf., and Evernia prunastri (L.) Ach. Water loss followed the same exponential pattern in all these species, the half time being dependent on species. Desiccation affected the fluorescence parameters. Dark-adapted maximum fluorescence (Fm), instantaneous fluorescence (Fo) and the ratio of variable (Fm–Fo) to Fm were dependent on water content and decreased in two distinct phases: a slow and apparently linear phase, followed by a more steep decline at low water content. Actual PSII photochemical yield (φPSII), non-photochemical quenching (NPQ), efficiency of photon capture (φexc), and photochemical quenching (qp) remained nearly constant until 30% relative water content (RWC), decreasing rapidly thereafter. In contrast, increased NPQ appeared to occur only at water content values lower than 20%. Treatment of thalli with dithiothreitol (DTT) effectively reduced NPQ during desiccation and increased susceptibility to photoinhibition caused by exposure to high light as measured by dark recovery of the FvFm ratio. HPLC analysis showed that the level of the de-epoxidized xanthophyll cycle pigments antheraxanthin (Anth) and zeaxanthin (Zea) increased during lichen desiccation. The results point towards the existence of a photoprotective mechanism with the involvement of Zea and Anth in non-radiative dissipation of the desiccation-induced excess of energy.  相似文献

为研究依据不同土层的土壤质量含水量进行测墒补灌对小麦(Triticum aestivum)拔节期与开花期旗叶荧光特性和水分利用效率的影响, 2011-2012和2012-2013年度两个小麦生长季, 设置0-20 (D1)、0-40 (D2)、0-60 (D3)和0-140 cm (D4) 4个土层进行处理, 测定土壤质量含水量, 以各土层平均土壤相对含水量在拔节期为65%和在开花期为70%为目标相对含水量进行补灌, 全生育期不灌溉为对照(D0)。结果表明: (1) D2处理拔节至开花期40-100 cm土层和开花至成熟期40-140 cm土层的土壤贮水消耗量高于其他处理, 开花至成熟期是小麦贮水消耗的最大时期。(2)开花后旗叶水分利用效率、PSII潜在活性(Fv/Fo)、PSII电子传输活性(Fm/Fo)、相对电子传递速率(ETR)和光化学猝灭系数(qP) D2处理最高, D3次之, D0最低。(3)两个小麦生长季, 各处理的籽粒产量为D2 > D3 > D1 > D4 > D0, D2的水分利用效率分别为20.19 kg·hm-2·mm-1和21.92 kg·hm-2·mm-1, 高于D0、D3和D4处理, 与D1处理间无显著差异。综合分析, 小麦拔节期和开花期依据0-40 cm土层的土壤质量含水量进行测墒补灌可兼顾高产和高水分利用效率。  相似文献

Diurnal heliotropic leaf movements, photosynthetic gas exchange, and the ratio of variable fluorescence to maximum fluorescence (Fv/Fm) of unrestrained and horizontally restrained leaves from soybean (Glycine max cv. Cumberland) plants grown in two different water and two different nitrogen treatments were measured. Leaves of plants grown in low water or low nitrogen availability treatments displayed more pronounced diaheliotropism (solar tracking) in the afternoon and a longer period of paraheliotropism (light avoiding) at midday relative to those of well-watered, high-nitrogen-grown plants. Photosaturated photosynthetic rates and the photon flux required to saturate photosynthesis were reduced by water stress and nitrogen deficiency. Compared to horizontal leaves, irradiance on orienting leaves was nearer to the breakpoint of the photosynthetic light response curve, where photosynthesis is co-limited by ribulose biphosphate regeneration and carboxylation. This would increase the carbon return on investments of nitrogen into photosynthesis. A positive linear relationship between Fv/Fm and quantum yield of photosynthesis was measured. Leaves of low-nitrogen-grown plants had earlier and more prolonged reductions in Fv/Fm at midday compared to leaves of high nitrogen grown plants of the same water treatment. Within the same water and nitrogen treatment, horizontally restrained leaves had lower midday Fv/Fm in relation to orienting leaves. Nitrogen deficiency and water stress enhanced this difference such that horizontally restrained leaves of low water and low nitrogen grown plants had earlier and longer midday depressions in Fv/Fm.  相似文献

The interactions of caffeine (CF) with chlorogenic acid (CGA) and caffeic acid (CFA) were investigated by fluorescence quenching, UV/vis and Fourier transform infrared (FTIR) spectroscopic techniques. The results of the study indicated that the fluorescence quenching between caffeine and hydroxycinnamic acids could be rationalized in terms of static quenching or the formation of non‐fluorescent CF–CFA and CF–CGA complexes. From fluorescence quenching spectral analysis, the quenching constant (KSV), quenching rate constant (kq), number of binding sites (n), thermodynamic properties and conformational changes of the interaction were determined. The quenching constants (KSV) between CF and CGA, CFA are 1.84 × 104 and 1.04 × 104 L/mol at 298 K and their binding site n is ~ 1. Thermodynamic parameters determined using the Van't Hoff equation indicated that hydrogen bonds and van der Waal's forces have a major role in the reaction of caffeine with caffeic acid and chlorogenic acid. The 3D fluorescence, UV/vis and FTIR spectra also showed that the binding of CF with CFA and CGA induces conformational changes in CFA and CGA. Copyright © 2015 John Wiley & Sons, Ltd.  相似文献

Water transport across plant cell membranes is difficult to measure. We present here a model assay, based on chlorophyll (Chl) a fluorometry, with which net water transport across the cell membrane of freshwater cyanobacterium Synechococcus sp. PCC7942 (S7942) can be followed kinetically with millisecond-time resolution. In cyanobacteria, the phycobilisome (PBS)-sensitized Chl a fluorescence increases when cells expand (e.g., in hypo-osmotic suspension) and decreases when cells contract (e.g., in hyper-osmotic suspension). The osmotically-induced Chl a fluorescence changes are proportional to the reciprocal of the suspension osmolality (ΔF ∝ Osm−1; Papageorgiou GC and Alygizaki-Zorba A (1997) Biochim Biophys Acta 1335: 1–4). In our model assay, S7942 cells were loaded with NaCl (passively penetrating solute) and shrunk in hyper-osmotic glycine betaine (nonpenetrating solute). Upon injecting these cells into hypo-osmotic medium, the PBS-sensitized Chl a fluorescence rose to a maximum due to the osmotically-driven water uptake. The rise of Chl a fluorescence (water uptake) was partially inhibited by HgCl2, at micromolar concentrations. Arrhenius plots of the water uptake rates gave activation energies of EA=4.9 kcal mol−1, in the absence of HgCl2, and EA=11.9 kcal mol−1 in its presence. These results satisfy the usual criteria for facilitated water transport through protein water pores of plasma membranes (aquaporins), namely sensitivity to Hg2+ ions and low activation energy.  相似文献
