通过PCR定点突变的技术,将蛇毒蛋白Echistatin基因的C端进行了突变(Ala48→Arg48→,Thr49→Val49),模拟纤维蛋白N端的四肽(Gly-Pro-Arg-Val),以期增加Ecs(Echistatin)的活性。突变的基因重组到表达质粒pJC264上,经IPTG诱导,以CheY-Ecs融合蛋白方式进行了表达,表达量占菌体总蛋白的15~20%。SephadexG-75初步纯化该融合蛋白,然后用CNBr裂解,透析,冻干,反相HPLC纯化C端突变体Ecs蛇毒蛋白,N端十个氨基酸分析与天然的相符,在PRP(platelet-richplasma)测活体系中,10μmol/L的ADP诱导,C端突变体Ecs抑制血小板凝聚的活性约为野生型4倍。得到了Ecs的C端突变后使Ecs抑制血小板凝聚的活性提高的结果。  相似文献

Scutellaria barbata (S. barbata), a traditional Chinese herbal medicine native to southern China, is widely used as an anti-inflammatory and a diuretic in China. Several studies have indicated that extracts of S. barbata have growth inhibitory effects on a number of human cancers. Treatment of lung cancer, digestive system cancers, hepatoma, breast cancer, and chorioepithelioma by S. barbata extracts was reported. However, the mechanism underlying the antitumor activity was unclear. In this study, we studied the growth inhibitory effect of S. barbata and determined its mechanism of antitumor activity using human lung cancer cell line A549. Our results showed that ethanol extracts of S. barbata greatly inhibited A549 cell growth, with IC50 of 0.21 mg/ml. The major mechanisms of inhibition included cell apoptosis and cytotoxic effects. cDNA microarray analysis showed that 16 genes, involved in DNA damage, cell cycle control, nucleic acid binding and protein phosphorylation, underwent more than 5-fold change. These data indicated that these processes are involved in S. barbata-mediated killing of cancer cells. A surprising finding is that CD209, related to dendritic cell (DC) function, was dramatically downregulated by 102-fold. Further functional studies are needed to assess the role of the array-identified genes in S. barbata mediated anticancer activity.  相似文献

The individual flavonoid component, scutellarin, scutellarein, luteolin and apigenin, in Scutellaria barbata plant was isolated based on the macro porous adsorbent with high adsorption selectivity. These adsorbents were synthesized based on the copolymerization of methyl acrylate and divinylbenzene (MA-co-DVB). So the polarity and the adsorption affinity of these adsorbents can be adjusted through changing MA content in the adsorbents. And then the ability of the adsorbent with different MA contents for isolation of these four individual flavonoid was also investigated. Adsorbents M2 and M4, with MA content of 25% and 45%, respectively, demonstrated the best separation ability. Complete separation of the four flavone compounds was achieved in a continuous process based on combination of adsorbents with different polarities (M2 and M4). Gradient elution using adsorbent M4 separated the four flavonoids into three fractions, which were determined to contain scutellarin, scutellarein and a mixture of luteolin and apigenin. The latter was separated completely by adsorbent M2 subsequently. All four compounds were obtained at high resolution and high recovery yield (96.7%, 94.1%, 95.8% and 93.8%, respectively), suggesting the efficiency of sequentially combined columns with different segregation patterns.  相似文献

In this article, a DPPH·–luminol chemiluminescence (CL) system was reported and the CL mechanism was discussed according to the CL kinetic properties after sequence injecting DPPH· into the DPPH·–luminol reaction mixture. It was observed that scutellarin could inhibit the CL response of the DPPH·–luminol system. Based on this observation, a simple and rapid flow injection CL method was developed for the determination of scutellarin using the inhibition effect in alkaline medium. The optimized chemical conditions for the CL reaction were 5 × 10?6 mol/L DPPH · and 1.0 × 10?4 mol/L luminol in 0.01 mol/L NaOH. Under optimized conditions, the CL intensity was inversely proportional to the concentration of scutellarin over the ranges 5–2000 and 40–3200 ng/ml in pharmaceutical injection and rat plasma, respectively. The limits of detection (S/N  = 3) were 5 and 40 ng/ml in preparations and rat plasma, respectively. Furthermore, the precision, recovery and stability of the validated method were acceptable for the determination of scutellarin in both pharmaceutical injections and rat plasma. The presented method was successfully applied in the determination of scutellarin in pharmaceutical injections and real rat plasma samples.  相似文献

A validated high-performance liquid chromatography method is described for the determination of scutellarin in rat plasma using a liquid-liquid extraction and ultraviolet (UV) absorbance detection. The separation used a Diamonsil ODS column (250 mm x 4.6mm i.d., 5 microm particle size) with an isocratic mobile phase consisting of methanol-acetonitrile-50mM dihydrogen ammonium phosphate buffer (22:15:63 (v/v/v), adjusted to pH 2.5 with 1M phosphoric acid). The ultraviolet detector operated at 335 nm. Plasma samples were extracted with ethyl acetate after acidification. The extraction recovery of scutellarin ranged from 68.1 to 80.5%. High selectivity and a low quantitation limit (0.050 microg/ml) were achieved. The linear range was 0.050-12.5 microg/ml, correlation coefficient r=0.9981. The method has a good reproducibility, R.S.D. values were below 7.9% for within-day and between-day precision. The method is simple, rapid, and applicable to preliminary pharmacokinetic studies of scutellarin in rats.  相似文献

The expression changes of baculovirus inhibitor of apoptosis repeat-containing protein5 in brain glioma after administration of Scutellarin was detected. To explore the effort of scutellarin on anti-glioma by downregulating BIRC5.The effect of scutellarin on tumour growth and animal survival was detected by administering scutellarin to nude mice subcutaneous tumour formation and SD rats in situ tumour formation models. A significantly different gene BIRC5 was found by using the combination of TCGA databases and network pharmacology. And then qPCR was performed to detect the expression of BIRC5 in glioma tissues, cells and normal brain tissues and glial cells. CCK-8 was used to detect the IC50 of scutellarin on glioma cells. The wound healing assay, flow cytometry and MTT test were used to detect the effect of scutellarin on the apoptosis and proliferation of glioma cells. The expression of BIRC5 in glioma tissues was significantly higher than that in normal brain tissues. Scutellarin can significantly reduce tumour growth and improve animal's survival. After scutellarin was administered, the expression of BIRC5 in U251 cells was significantly reduced. And after same time, apoptosis increased and cell proliferation was inhibited. This original research showed that scutellarin can promote the apoptosis of glioma cells and inhibit the proliferation by downregulating the expression of BIRC5.  相似文献

在相同栽培管理条件下,分别以黑色石灰土、红色石灰土、红壤、黄红壤、紫色土和腐殖土为生长介质对短葶飞蓬(Erigeron breviscapus)进行了人工栽培试验。结果显示,生长在不同土壤类型的短葶飞蓬植株体内总咖啡酸酯和灯盏乙素含量有显著的差异。红色石灰土、紫色土及黄红壤生长的植株总咖啡酸酯和灯盏乙素含量最高,黑色石灰土和腐殖土的最低。相同土类的不同亚类土壤生长植株间2种次生代谢产物含量的差异程度,会超过不同土类间的。植株总咖啡酸酯和灯盏乙素含量与土壤P和K含量没有显著相关关系,但与土壤速效N含量及植株含N量则都有极显著负相关关系。不同土壤生长植株叶片的最大光能转换效率(Fv/Fm)值与灯盏乙素和总咖啡酸酯含量都表现出极显著负相关关系。土壤类型对短葶飞蓬次生代谢影响规律与碳素/营养平衡假说(CNB)及资源获得假说(HA)的预测一致。  相似文献

The binding of scutellarin with human serum albumin (HSA) was investigated at four temperatures, 296, 303, 310, and 318 K, by fluorescence, circular dichroism (CD), Fourier transform infrared spectroscopy (FT-IR), and molecular modeling study at pH 7.40. The binding parameters were determined by Scatchard's procedure, which are approximately consistent with the results of Stern-Volmer equation. The thermodynamic parameters were calculated according to the dependence of enthalpy change on the temperature as follows: DeltaH degrees is a small negative value (-8.55 kJ/mol), whereas DeltaS degrees is a positive value (65.15 J/mol K). Quenching of the fluorescence HSA in the presence of scutellarin was observed. Data obtained by fluorescence spectroscopy and CD experiment, FT-IR experiment, and molecular modeling method suggested that scutellarin can strongly bind to the HSA and the primary binding site of scutellarin is located in site I of HSA. It is considered that scutellarin binds to site I (subdomain II) mainly by a hydrophobic interaction and there are hydrogen bond interactions between the scutellarin and the residues Arg222 and Arg257.  相似文献

Selenium has both nutritional function and toxicity according to its concentration and species. To counteract the toxicity of selenium, scutellarin was investigated. Wistar rats were supplemented with 40 μg Se/kg/d as sodium selenite, 40 μg Se/kg/d with 20 mg/kg/d scutellarin, and 20 mg/kg/d scutellarin, respectively, for 15 d. The mRNA levels and activities of glutathione peroxidase (GSH-Px) and thioredoxin reductase (TR), and the malondialdehyde (MDA) contents were measured. Reactive oxygen species (ROS) were detected by chemiluminescence assay, and tissue conformation was investigated by histological study. The results showed significant decreases of mRNA levels and activities of GSH-Px and TR and a significant increase of MDA content in livers of the Se-treated rats (p<0.05, compared with the control). Supplementation of scutellarin to the Se-treated group significantly inhibited the decreases of mRNA levels and activities, and the increase of MDA content (p<0.05, compared with the Setreated group). Meanwhile, scutellarin-scavenged ROS generated in the mixture of sodium selenite, reduced glutathione, and oxygen. Liver injury was displayed in slices exposed to selenium at the present dose. The groups treated with both selenium and scutellarin or only scutellarin did not show significant tissue damage. Thus, scutellarin had an antagonistic effect against the toxicity of selenium.  相似文献
