Solutions of oestrone, oestrone-phosphate, oestrone-sulphate,oestradiol and oestradiol-sulphate in the concentration range103 mol m3 to 107 mol m3 had noobservable morphological or anatomical effects on adventitiousroot formation in Phaseolus vulgaris hypocotyl, epicotyl andprimary leaf cuttings. Oestradiol-sulphate and oestrone-sulphatetreatments at 0.1 mol m3 significantly inhibited rootingin hypocotyls, and the inhibition was almost complete in epicotylsand primary leaves. In the latter, anomalous development ofvascular tissues was noted. However, neither oestrone-phosphateat 0.1 mol m3 nor direct application of up to 100 µgof the oestrogens to apices or primary leaves of explants modifiedthe pattern of root formation. High frequency bud regeneration was observed inpetiole and thin layer explants of 10 different breeding linesof sugar beet tested. Organogenesis resulted when petiole explantsexcised from 8-d-old seedlings grown on half-strength Murashigeand Skoog medium (MS) containing 3.0 mg dm3 naphthaleneacetic acid (NAA), 3.0 mg dm3 6-benzylaminopurine (BAP)and 1.0 mg dm3 2, 3, 5, triiodobenzoic acid (TIBA) werecultured on MS with 3.0 mg dm3 NAA and 3.0 mg dm3BAP. Thin cell layer strips isolated from shoot apices culturedon MS medium supplemented with 09 mg dm3 BAP or1.0 mg dm3 indolebutyric acid (IBA) formed adventitiousbuds on MS medium containing 05 mg dm3 NAA + 5.0mg dm3 BAP. Effects of a Range of O2 Concentrations on Porosity of Barley Roots and on their Sugar and Protein Concentrations 总被引:1,自引:0,他引:1
Barley was grown at a range of oxygen concentrations (0.59mg l1), in nutrient solutions. Growth of both shootsand seminal roots was restricted by O2 concentrations lowerthan 23 mg l1) but nodal root growth was not. Root porosities were increased even at those O2 concentrationswhich did not restrict growth, and were inversely proportionalto the protein levels of the roots. Putrescine, spermidine, and spermine content were analysed inzygotic embryos of barley (Hordeum vulgare L.). Changes in polyaminecontent were observed during zygotic embryo growth. In two cultivars,Bomi and Golden Promise, the totalpolyamine content in the embryos was 2.62.9 nmol mg1fresh weight 10 d after anthesis, the highest content observed.It dropped to 1.3 nmol mg1 fresh weight 14 d after anthesis.This drop was caused by decreases in all three polyamine concentrations.From 14 to 35 d after anthesis the putrescine content continuedto decrease while the spermidine and spermine content increased,thus the total polyamine content remained constant until 35d after anthesis. The mutant Ris? 1508 showeda constant polyamine content around 1.3 nmol mg1 freshweight from 14 to 35 d after anthesis. The polyamine patternwas conserved in all three lines throughout the period of investigationshowing a spermidine content higher than putrescine contentwhich was, in turn, higher or equal to the spermine content.The polyamine content measured as nmol µg1 proteindecreased from 14 to 21 d post anthesis in all three lines,because the protein content (µg mg1 fresh weight)increased during the period. In dedifferentiating zygotic embryoscultured in vitro the putrescine content (nmol mg1 freshweight) rose by a factor of nine and the spermidine contentdoubled within the first week of cultivation, whereas sperminecontent did not change. Histological Observations on Initiation and Morphogenesis in Immature and Mature Embryo Derived Callus of Barley (Hordeum vulgare L.) 总被引:1,自引:0,他引:1
Callus was induced from immature and mature embryos of barley(cv. Haruna Nijo) on Murashige and Skoog medium containing 2mg l-1 2,4-D and 5 mg l-1 picloram, respectively. Paraffin sections(10 µm thick) were prepared for histology during callusinitiation and plant regeneration. Meristems were regeneratedfrom nodular compact callus (NC) derived from scutellar epidermisin immature embryos, whereas they were regenerated from NC derivedfrom epidermal cells of leaf or coleoptile bases in mature embryos.Regardless of the explant source, regeneration was predominantlythrough organogenesis, although regeneration through somaticembryogenesis infrequently occurred. Clonally propagated plants of Dactylis glomerata derived froma well-drained, heavily grazed cliff habitat (clone L) and froman undergrazed poorly-drained soil (clone A) were tested forwaterlogging tolerance in soil-culture. Water-logging did notaffect the two clones differentially, a result, which contrastedstrongly with that of a previous experiment in which simulatedgrazing (clipping to 20 cm) unexpectedly caused clone A to beless tolerant of waterlogging than clone L. Maximum leaf andleaf sheath length was reduced more by water-logging in cloneL than in clone A (P < 0.05). In solution-culture when providedwith factorial combinations of 0.5, 5 and 50 mg dm2 ofFe2+ and Mn2+ the shoot dry weight yield of the dry-soil clonewas reduced more than that of the wet-soil clone by 50 mg Fedm3 irrespective of Mn2+ concentration (P < 0.01)but the reduction of growth was less at higher Mn2+ concentrations.Fifty milligrams of Mn2+ dm3 reduced the growth of thedry soil clone but increased the growth of the wet soil clonewith Fe2+ at 5 mg dm2 (P < 0.05). Iron at 0.5 mg dm2was suboptimal for shoot growth of both clones at any levelof Mn2+ and caused more severe leaf chlorosis in the wet soilclone. Leaf tissue of clone L contained more iron than thatof clone A after waterlogging (P < 0.01) but in solutionculture, though increasing iron from 0.5 to 50 mg dm3almost doubled leaf iron content (P < 0.001), the interactionClones x Mn x Fe just failed to reach significance at P <0.05. The manganese content of leaf tissue from the two clonesvaried differently in response to solution manganese (Clonesx Mn P < 0.01), clone A showing a slightly greater increaseof manganese content at high solution concentration. Iron at50 mg dm3 suppressed Mn uptake (Mn x Fe, P < 0.001)in both clones. The two clones thus show marked environmentaladaptation to the chemistry of wet and dry soils. Inhibition of Light-Stimulated Leaf Expansion by Abscisic Acid 总被引:9,自引:2,他引:7
Abscisic acid (ABA) applied to intact bean (Phaseolus vulgaris)leaves or to isolated leaf discs inhibits light-stimulated cellenlargement This effect may be obtained with 104 molm3 ABA, but is more significant at higher concentrations.The inhibition of disc expansion by ABA is greater for discsprovided with an external supply of sucrose than for discs providedwith KC1, and may be completely overcome by increasing the KC1concentration externally to 50 mol m3. Decreased growthrate of ABA-treated tissue is not correlated with loss of solutesfrom growing cells, but is correlated with a decrease in cellwall extensibility. ABA does not prevent light-stimulated acidificationof the leaf surface, and stimulates the acidification of theexternal solution by leaf pieces. However, the capacity of thecell walls to undergo acid-induced wall loosening is diminishedby ABA-treatment. The possibility that ABA acts directly byinhibiting growth processes at the cellular level, or indirectlyby causing stomatal closure, is discussed. During the ripening stage of barley and rice, the sink activitywas defined as the dry matter increase per units sink size,leaf area and time, as follows: NAR = A.SinkW+NAR', where NAR is the net assimilation rate (g d.wt dm2d1);A is the sink activity [g d.wt g1d.wt (ear) dm2d1]; Sink W is ear wt per plant at heading (g d.wt);and NAR' is net assimilation rate excluding the assimilate ofsink organ (g d.wt dm2 d1). Plant material with 16 combinations of mutually different sink(ear) and source (leaf) size were produced at heading for eachcrop: parts of each leaf and ear were removed to produce fourgrades in barley, and also a part of each leaf was removed producingfour grades for four rice varieties showing different ear size.NAR and NAR' were determined during 26 and 21 d in barley andrice after heading, respectively. Sink activity (A), representedas the assimilation rate induced by the sink organ, was estimatedfrom the relationship between SinkW and NAR using the previousequation. The sink activity was significantly higher in ricewith a value of 00.028 g d.wt g1 d.wt (ear) dm2d1 vs. 00.0017 in barley, suggesting that therelative role of leaves for grain filling is considerably higherin rice than in barley. The sink activity obtained in the studymight be introduced into a model to predict the yields of barleyand rice. The specific respiration rates of nodulated root systems, ofnodules and of roots were determined during active nitrogenfixation in soya bean, navy bean, pea, lucerne, red clover andwhite clover, by measurements on whole plants before and afterthe removal of nodule populations. Similar measurements weremade on comparable populations of the six legumes, lacking nodulesbut receiving abundant nitrate-nitrogen, to determine the specificrespiration of their roots. All plants were grown in a controlled-environmentclimate which fostered rapid growth. The specific respiration rates of nodulated root systems ofthe three grain and three forage legumes during a 714-dayperiod of vegetative growth varied between 10 and 17 mg CO2g1 (dry weight) h1. This mean value consistedof two components: a specific root respiration rate of 69mg CO2 g1 h1 and a specific nodule respirationrate of 2246 mg CO2 g1 h1. Nodule respirationaccounted for 4270 per cent of nodulated root respiration;nodule weight accounted for 1240 per cent of nodulatedroot weight. The specific respiration rates of roots lackingnodules and utilizing nitrate nitrogen were generally 2030per cent greater than the equivalent rates of roots from nodulatedplants. The measured respiratory effluxes are discussed in thecontext of nitrogen nitrogen fixation, nitrate assimilation. Plants of Phaseolus vulgaris grown at 7 and 28 W m2 showedno differences in rate of development of leaves or flowers.At 7 W m-Z plants had longer internodes, more succulent stemsand leaves, higher ratios of shoot:root and greater leaf areasthat those at 28 W m2. These differences were establishedprior to detectable differences in photosynthesis and couldpartly be attributed to an increased proportion of far-red light. Although the final d. wt, carbon content, and fruit yield werehigher at 28 W m2, plants at 7 W m2 apparentlyhad similar relative growth rates and greater photosyntheticefficiency. Ten-day old kidney bean plants (Phaseolus vulgaris L. cv. Shin-edogawa)were exposed to 2.0 and 40 parts 106 NO2, and0.1, 0.2, and 0.4 parts 106 O3 alone or in combinationfor 2, 4, and 7 d. The effects of these air pollutants wereexamined with respect to the growth, partitioning of assimilates,nitrogen uptake, soluble sugar content, and root respiration. Decreased dry matter production was significant with all treatmentsexcept 2.0 parts 106 NO2 and 0.1 parts 106 O3.Exposure to mixtures of the gases produced more severe suppressionof growth than exposure to the single gases. Root/shoot ratiowas significantly lowered at 7 d by the gas treatments otherthan 2.0 parts 106 NO2 and 0.1 parts 106 O3. Thetotal nitrogen content of plants was increased by all treatments;the higher percent of nitrogen found with O3 exposure will resultfrom the growth retardation which increases the concentrationof nitrogen in the plants because the absorption of nitrogenby roots was unaffected. The combination of O3 with NO2 significantlydecreased the assimilation of NO2 by the plants. The concentration of soluble sugars in roots was decreased bythe gas treatments. There was a strong positive correlationbetween soluble sugar content and dry weight of the roots harvestedat 7 d. Root respiration was relatively unchanged until 5 dand then decreased significantly at 7 d by 2.0 parts 106NO2 and 02 parts 106 O3. Retarded growth of theroots and the decreased root respiration may be due to diminishedtranslocation of sugars from leaves to roots caused by exposureto air pollutants. Explants obtained from the basal portion of leaves of Hordeumvulgare (cv. Karan 92) gave rise to callus when cultured onMurashige and Skoog (MS) basal medium supplemented with 2, 4-dichlorophenoxyaceticacid (2, 4-D). Initially, the callus was friable, shiny-whiteand watery but subsequently some compact, nodular callus appeared.The latter were cultured on MS medium containing 0.05 mg l12, 4-D and 0.1 mg l1 N6-furfurylaminopurine (kinetin),when plantlets were generated. Protoplasts were successfully isolated from internodal callustissues of both Oxalis glaucifolia and O. rhombeo-ovata whenthey were digested in a solution containing 0.1% (w/v) MacerozymeR-10, 0.5% (w/v) cellulase Onozuka R-10 and 0.3 mmol m3sucrose. Protoplasts proliferated to give cell colonies on Gamborget al.'s B5 medium supplemented with 0.3 mmol m3 mannitol,0.5 mg dm32, 4-D, and 2.0 mg dm3 kinetin. Calluswas produced upon transfer of cell colonies to Murashige andSkoog medium containing 2.0 mg dm3 l-naphthaleneaceticacid (NAA) and 0.1 mg dm3 kinetin for O. glaucifolia,or with 5.0 mg dm3 NAA and 0.5 mg dm3 6-benzylaminopurine,for O. rhombeo-ovata. Plants were regenerated from O. glaucifoliaprotoplasts on a medium containing 0.1 mg dm3 NAA, 1.0mg dm3 kinetin and 1.0 mg dm3 gibberellic acid,but only vascular nodules were differentiated by O. rhombeo-ovataprotoplast-derived calli. The pigment changes that occur during transformation of etioplaststo fully developed chloroplasts have been studied in seedlingsof barley (Hordeum vulgare L.) by greening with white lightof low (1525 µmol m2 s1) and medium(150 µmol m2 s1) intensity. At least 24h longer was required in the low light regime for the same concentrationof pigment to be accumulated in the seedlings. The increasein pigment content was mainly due to the synthesis of chlorophyllsa and b. Of the carotenoids present, the increases in the levelsof neoxanthin and, especially, ß-carotene were muchgreater than those observed for the other carotenoids. Levelsof lutein also increased but this change was small by comparisonto those observed for neoxanthin and ß-carotene. Inthe long-term the concentration of violaxanthin remained unalteredalthough significant transient changes were recorded. The levelsof antheraxanthin and zeaxanthin were markedly reduced duringgreening. The rate of pigment synthesis decreased with increasingcell age, i.e. from the base to the tip of the primary leaf.Overall, carotenoid levels increased by approximately 100% atthe base of the seedling but hardly at all at the tip. Use of Rotary Variable Displacement Transducers attached togrowing primary leaves of Phaseolus vulgaris has shown thatwhen the root systems were rapidly cooled from 23°C to 10°Cleaf extension rate fell to very low values within a few minutes.When the root systems were returned to 23°C leaf extensionincreased almost immediately to overshoot the control rateswithin 510 min, before declining to control values overthe next 5060 min. When lights went off at the end ofthe day cycle there was an immediate and very large increasein leaf extension rate in both root-cooled and control plants;the rate then slowly declined over the next 60 min. This effectwas seen when the photoperiod was artificially shortened orlengthened and was reduced in magnitude when the photoperiodwas ended gradually by dimming the lights. The effect was notattributable to effects on leaf temperature but appears to bethe result of an endogenous rhythm interacting with the endof the photoperiod. At the beginning of the photoperiod therewas a gradual reduction in leaf extension rate occurring over3045 min. The Response of Bean Plants to UV-B Radiation Under Different Irradiances of Background Visible Light 总被引:14,自引:1,他引:13
Plants of Phaseolus vulgaris L. (cv. Stella) were grown in controlledconditions under three different irradiances of visible lightwith or without UV-B (280320nm) radiation. The biologicallyeffective UV-B radiation (UV-BBE) was 6.17 kJ m2 d1,and simulated a c. 5% decrease in stratospheric ozone at 55.7?N,13.4?E. The photon flux densities of the photosyntheticallyactive radiation (PAR, 400700 nm) were either 700 µmolm21 (HL), 500, µmol m2 s1(ML) or 230 µmol m2 s1 PAR (LL). Under highlight (HL) conditions plus UV-B radiation, bean plants appearedmost resistant to the enhanced levels of UV-B radiation, andresponded only by increasing leaf thickness by c. 18%. A smallincrease in UV screening pigments was also observed. Both thelower irradiances (ML and LL) increased the sensitivity of theplants to UV-B radiation. Changes in leaf structure were alsoobserved. Photosystem II was inhibited under ML and LL togetherwith UV-B radiation, as determined by Chi fluorescence inductionand calculation of the fluorescence half-rise times. Leaf reflectivitymeasurements showed that the amount of PAR able to penetrateleaves of UV-B treated plants was reduced, and that a possiblecorrelation may exist between the reduced PAR levels, loss ofChi and lowered photosynthetic activity, especially for LL +UV-Bgrown plants, where surface reflection from leaves was highest.Changes in leaf chlorophyll content were mostly confined toplants grown under LL + UV-B, where a decrease of c. 20% wasfound. With regard to protective pigments (the carotenoids andUV screening pigments) plants subjected to different visiblelight conditions responded differently. Among the growth parametersmeasured, there was a substantial decrease in leaf area, particularlyunder LL + UV-B (c. 47% relative to controls), where leaf dryweight was also reduced by c. 25%. Isolated embryos of three contrasting barley genotypes werecultured in vitro on a range of media comprising 16 combinationsof sucrose (312 per cent) and 2, 4-D (08 mg 11)concentration. Cultures were incubated at a range of temperaturesfrom 5 to 25°C and were examined after 21 days when shootlengths as well as fresh and dry weights were recorded. Therelative influence on growth of increasing concentrations ofsucrose and 2, 4-D was investigated, as was their interactionwith the incubation temperature. The genotypes were found todiffer markedly in their response to these two media components,with each parameter of growth differentially affected. After growing barley (Hordeum vulgare L.) in nutrient solutionscontaining EDTA, uptake of the nutrient metals was determinedat three harvests and concentrations of the various chemicalspecies of each metal in the growth solutions was modelled bycomputer simulation. Complexation with EDTA had different effectson the uptake of the ions Fe3+, Mn2+, Cu2+, and Zn2+. At thehighest EDTA level (EDTA/Fe=2/l) the plants were chlorotic andgrowth was inhibited. This is attributed to a deficiency inZn rather than in Fe. The critical level of free Zn2+ requiredin nutrient solutions for healthy growth was found to be approximately10101010 mol dm3, which is consistent withthat found by earlier workers for other plant species. Barleytolerated much lower levels of the free ions of copper and ironwithout exhibiting any obvious adverse effects. The anatomical structure of the second leaf blade of barley{Hordeum vulgare L. cv. Koral) was studied in plants exposedto a photosynthetic photon flux density (PPFD) of 200 µmolm2 s1 compared with those grown under 25µmolm211. Design-based stereological methods wereused for the estimation of various leaf anatomical characteristicssuch as mesophyll volume, proportion of intercellular spaces,number of mesophyll cells, mean mesophyll cell volume, and internalleaf surface area. The structure of the mesophyll was more affectedby different levels of PPFD than were the stomatal characteristics.Increased PPFD produced thicker leaves with a larger mesophyllvolume having a higher number of less elongated mesophyll cellsand a larger internal leaf surface area. Differences in the Interacting Effects of Light and Temperature on Growth of Four Species in the Vegetative Phase 总被引:3,自引:0,他引:3
A comparative study, employing the concepts of growth analysis,has been made of the varying responses in the early vegetativephase of Gossypium hirsutum, Helianthus annuus, Phaseolus vulgaris,and Zea mays to combinations of light intensity (1.08, 2.16,3.24, 4.32, and 5.4 x 104 lxphotoperiod 14 h) and constantdiurnal air temperatures (10, 15, 20, 25, 30, and 35 °C).Depending on the combination of treatments, the temperatureof the internal tissues departed from air temperature by 6.9to 1.4 °C: so only the internal temperatures are cited here. For each species there are complex interactions between theeffects of light and temperature on the net assimilation rate,the leaf-area ratio, and the relative growth-rates of plantweight and leaf area. The magnitude of the changes induced bythe two factors vary both with the growth component and thespecies. The temperature responses are maximal up to 205°C while at the highest temperatures they may be negative.The temperature coefficients for leaf-area ratio are consistentlyless than those of the other three components: here betweenspecies the coefficients over 1020 °C vary by a factorof 9.6, 5.4, and 5.1 for the rates of gain in plant weight andleaf area and the net assimilation rate, while the orderingwithin each growth component is species dependent. Under conditions of optimal temperature the relative growth-rateand net assimilation rate progressively increase, accordingto the species, up to either 4.32 or 5.4x 104 lx. The leaf-arearatio is always largest at the lowest intensity. The level oflight at which the rate of gain in leaf area reaches a maximumranges from 2.16x 104 lx for Phaseolus to between 4.32 and 5.40x104 lx for Gossypium. The highest relative growth-rate and net assimilation rate ofHelianthus exceed those of Zea substantially. Indeed the maximalassimilation rate for Helianthus of 2.10 g dm2 week1is the highest ever recorded under field or controlled conditions.Possible reasons for this reversal of the photosynthetic potentialsof the two species observed by previous workers are discussed. 相似文献
