The role of the cuticle and sheath in the water dynamics of the infective larvae of Trichostrongylus colubriformis under osmotic stress and the effect of osmotic stress on behaviour have been investigated. In hypotonic solutions larvae lose the activity response to mechanical stimulation and there was also some increase in coiling. Larvae can regulate their water content in hypotonic solutions but lose water slowly in hypertonic solutions. Removal of the sheath by exposure to sodium hypochloritc or to a ‘natural’ stimulus had little effect on the water dynamics of infective larvae. The sheath thus appears to be freely permeable to water when hydrated whereas the cuticle has a very restricted permeability.  相似文献

The infection rate of the first stage larval nematodes, Elaphostrongylus rangiferi, was studied experimentally, using the juvenile snail Arianta arbustorum as intermediate host. The nematode showed a linear, fivefold increase in infection rate within the temperature range of 4 to 28 C. The snails were exposed to the larval nematodes on three different substrates. The highest infection rate was recorded when snails were exposed in tap water and significantly slower infection rates were obtained when either lettuce or soil was used as the substrate. First stage larvae of E. rangiferi were infective for at least 2 months when stored at 12 C. Throughout this period, the infection rate showed a significant decline, while the motility of the larvae remained unchanged.  相似文献

Nematospiroides dubius, unlike the closely related nematode Nippostrongylus brasiliensis, lacks a sterol requirement for the completion of its development to the L3 stage in an aqueous suspension of formalin-killed Escherichia coli. These larvae were justascapable of completing their development to adults in mice as those grown on charcoal-feces cultures. In addition, sterols had a toxic effect on the larvae of N. dubius which was removed by substituting a Krebs-Ringer solution for distilled water.In order to interpret the absence of a sterol requirement for the development of N. dubius larvae, an analysis of the egg lipids for both species was made. N. brasiliensis had about twice the sterol concentration of N. dubius eggs. A comparison of the development for both species showed, however, that N. brasiliensis grows more than twice as much as N. dubius to reach the L3 larval stage. Therefore, the amount of endogenous sterol is insufficient for N. brasiliensis to develop from the egg to the L3 stage.N. dubius and N. brasiliensis also differ in their porphyrin requirements. Unlike N. brasiliensis, coproporphyrin did not increase the size of N. dubius larvae so that they were comparable to those grown on charcoal-feces cultures. Coproporphyrin did result in larvae that were more robust in appearance than those grown without a supplement.  相似文献

Spores of an unidentified bacterium were discovered adhering to cuticles of third-stage infective juvenile (IJ) Steinernema diaprepesi endemic in a central Florida citrus orchard. The spores were cup-shaped, 5 to 6 mm in length, and contained a central endospore. Based on 16S rDNA gene sequencing, the bacterium is closely related to the insect pathogens Paenibacillus popilliae and P. lentimorbus. However, unlike the latter bacteria, the Paenibacillus sp. is non-fastidious and grew readily on several standard media. The bacterium did not attach to cuticles of several entomopathogenic or plant-parasitic nematodes tested, suggesting host specificity to S. diaprepesi. Attachment of Paenibacillus sp. to the third-stage cuticle of S. diaprepesi differed from Paenibacillus spp. associated with heterorhabditid entomopathogenic nematodes, which attach to the IJ sheath (second-stage cuticle). The inability to detect endospores within the body of S. diaprepesi indicates that the bacterial association with the nematode is phoretic. The Paenibacillus sp. showed limited virulence to Diaprepes abbreviatus, requiring inoculation of larvae with 108 spores to achieve death of the insect and reproduction of the bacterium. The effect of the bacterium on the nematode population biology was studied in 25-cm-long vertical sand columns. A single D. abbreviatus larva was confined below 15-cm depth, and the soil surface was inoculated with either spore-free or spore-encumbered IJ nematodes. After 7 days, the proportion of IJ below 5-cm depth was seven-fold greater for spore-free IJ than for spore-encumbered nematodes. Mortality of D. abbreviatus larvae was 72% greater (P <= 0.01) for spore-free compared to spore-encumbered S. diaprepesi. More than 5 times as many progeny IJs (P <= 0.01) were produced by spore-free compared to spore-encumbered nematodes. These data suggest that the bacterium is a component of the D. abbreviatus food web with some potential to regulate a natural enemy of the insect.  相似文献

Nematodirus battus has been shown to be subject to a “self-cure” mechanism when lambs 6 weeks of age are infected with 60,000 larvae. It is proposed that this self-cure phenomenon is an immune response similar to that which occurs in rats infected with Nippostrongylus brasiliensis. Changes in the weight of adult Nematodirus sp. and Nippostrongylus sp. occur over the period of an infection in their hosts. Females of both species from high-dose infections increased in weight up to that time postinfection where most adult worms were present, then decreased in weight with the onset of rejection from the host. Male Nematodirus sp. showed a similar pattern of weight change to that of females but male Nippostrongylus sp. maintained a steady weight throughout the infection. The consequences of these changes in weight are discussed with relevance to expression of enzyme activities of the nematodes on a weight of individual nematode basis.  相似文献

Various concentrations of the nematode Heterorhabditis bacteriophora were added to dishes containing second, third, and fourth larval instars of the mosquito, Culex pipiens, respectively. The infective stage nematodes were ingested by the mosquito larvae, they then penetrated through the alimentary tract in the neck region and entered the hemocoel. A melanization reaction killed many invading nematodes, but heavier concentrations overwhelmed the hosts' defense reaction and 100% mortality of third- and fourth-instar larvae was achieved using between 170 and 200 nematodes per host. Death was either due to the nematode releasing cells of the symbiotic bacterium, Xenorhabdus luminescens, into the hemocoel or to foreign bacteria (mostly Pseudomonas aeruginosa), which were introduced by the penetrating nematodes. The potential use of this nematode as a biological control agent of larval culicine mosquito is discussed.  相似文献

Early stages (neonate to 7- or 8-day-old larvae) of Spodoptera exigua and Pseudaletia unipuncta were exposed to the entomogenous nematode, Steinernema feltiae, at concentrations of 0, 10, 25, 60, 100, or 200 nematodes per larva. Larvae of both species were susceptible to nematode infections. However, neonate larvae of S. exigua were significantly less susceptible to nematode infection than 3- or 8-day-old larvae at or above 50 nematodes per larva. Mortalities of neonate larvae exposed to 50 or more nematodes ranged from 68 to 74% while mortalities of 3- and 8-day-old larvae ranged from 91 to 100%. The results with P. unipuncta showed similar trends as described for S. exigua, albeit at a lower mortality level and usually with no statistical differences. Mortalities of neonate larvae exposed to 50 or more nematodes ranged from 34 to 44% while mortalities of 7-day-old larvae ranged from 32 to 91%.  相似文献

Absence of the phasmid was demonstrated with the transmission electron microscope in immature third-stage (M3) and fourth-stage (M4) males and mature fifth-stage males (M5) of Heterodera schachtii, M3 and M4 of Verutus volvingentis, and M5 of Cactodera eremica. This absence was supported by the lack of phasmid staining with Coomassie blue and cobalt sulfide. All phasmid structures, except the canal and ampulla, were absent in the postpenetration second-stage juvenile (J2) of H. schachtii. The prepenetration V. volvingentis J2 differs from H. schachtii by having only a canal remnant and no ampulla. This and parsimonious evidence suggest that these two types of phasmids probably evolved in parallel, although ampulla and receptor cavity shape are similar. Absence of the male phasmid throughout development might be associated with an amphimictic mode of reproduction. Phasmid function is discussed, and female pheromone reception ruled out. Variations in ampulla shape are evaluated as phylogenetic character states within the Heteroderinae and putative phylogenetic outgroup Hoplolaimidae.  相似文献

Larval stages of Oesophagostomum radiatum grown in vitro and adults grown in vivo were incubated in complex media or in a simple salt solution containing radioactive glucose. Glucose disappearance and end product accumulation of third-stage larvae in a simple salt solution indicated that they excreted CO2 and acetic, propionic, and lactic acids. Larvae in third molt, fourth stage, and adults all excreted CO2, acetic, propionic, and lactic acids at twice the rate of third-stage larvae plus an additional product, methylbutyric acid. Carbon dioxide arose primarily from the 3 or 4 carbons of glucose. An anaerobic atmosphere (95% N2:5% CO2) had no apparent effect on metabolism. When incubation was done in complex media, isobutyric and 3-methylbutyric acids were seen as major excretion products (10 and 24%, respectively). However, these acids were quantitatively minor when incubations took place in simple salts-glucose medium (1 and 0–3%, respectively).  相似文献

Studies on 300 worm-free chickens infected with Ascaridia galli indicated that the histotropic phase is a normal part of the life cycle and that it involves both second- and third-stage larvae. The duration of the histotropic phase was dose-dependent. At low dose rates (50 eggs) it occurred from days 3 to 16 and was terminated abruptly by the third ecdysis. At high dose rates (2000 eggs) it was prolonged until at least day 54, because third-stage larvae became arrested in their development and the third ecdysis was delayed. Arrested development of Ascaridia galli was significantly suppressed following treatment of birds with cyclophosphamide, but expulsion of worms was not prevented. This result suggests the involvement of host antibodies in the induction of the arrested state.  相似文献

Chronically elevated circulating plasma glucocorticoid concentrations can have suppressive effects on immune function in mammals. House mice (Mus domesticus) that have been selectively bred for high voluntary wheel running exhibit chronically elevated (two-fold, on average) plasma corticosterone (CORT) levels and hence are an interesting model to study possible glucocorticoid-induced immune suppression. As an initial test of their immunocompetence, we compared the four replicate high runner (HR) lines with their four non-selected control (C) lines by subjecting them to infection by a parasitic nematode, Nippostrongylus brasiliensis. At generation 36 of the selection experiment, 10 adult males from each of the eight lines were inoculated subcutaneously with approximately 600 third-stage larval N. brasiliensis, and then sacrificed 12 days after injection. Neither spleen mass nor number of adult nematodes in the small intestine differed significantly between HR and C lines. However, the eight lines differed significantly in nematode counts, and the line means for nematode infestation were significantly positively related to baseline circulating CORT concentration measured in males from generations 34 and 39. Therefore, although selective breeding for high locomotor activity may not have resulted in a generally compromised immune response, results of this study are consistent with the hypothesis that glucocorticoids can have immunosuppressive effects.  相似文献

How any complex trait has evolved is a fascinating question, yet the evolution of parasitism among the nematodes is arguably one of the most arresting. How did free-living nematodes cross that seemingly insurmountable evolutionary chasm between soil dwelling and survival inside another organism? Which of the many finely honed responses to the varied and harsh environments of free-living nematodes provided the material upon which natural selection could act? Although several complementary theories explain this phenomenon, I will focus on the dauer hypothesis. The dauer hypothesis posits that the arrested third-stage dauer larvae of free-living nematodes such as Caenorhabditis elegans are, due to their many physiological similarities with infective third-stage larvae of parasitic nematodes, a pre-adaptation to parasitism. If so, then a logical extension of this hypothesis is that the molecular pathways which control entry into and recovery from dauer formation by free-living nematodes in response to environmental cues have been co-opted to control the processes of infective larval arrest and activation in parasitic nematodes. The molecular machinery that controls dauer entry and exit is present in a wide range of parasitic nematodes. However, the developmental outputs of the different pathways are both conserved and divergent, not only between populations of C. elegans or between C. elegans and parasitic nematodes but also between different species of parasitic nematodes. Thus the picture that emerges is more nuanced than originally predicted and may provide insights into the evolution of such an interesting and complex trait.  相似文献

Numbers of Steinernema sp. (CB2B) and S. carpocapsae (Agriotos) exponentially declined after application into a clay loam soil. Over a 35-day sampling period, Steinernema sp. (CB2B) was more persistent than S. carpocapsae (Agriotos). The presence or absence of the second-stage cuticle on the third-stage juveniles (J3) at the time of application did not alter the rate of population decline of Steinernema sp. (CB2B). Nearly all J3 of Steinernema sp. (CB2B) and S. carpocapsae (Agriotos) lost their cuticle within 24 hours of being in soil. Centrifugal flotation recovered the greatest number of nematodes, with a lower variance than either the live bait or Baermann funnel techniques. A strong positive linear relationship was evident between numbers of nematodes present in the soil and the numbers that established in a bait insect. Approximately 40% of Steinernema sp. (CB2B) and 30% of the S. carpocapsae (Agriotos) present in the soil established in Galleria mellonella larvae. The extraction techniques had different efficiencies and gave different relative estimates of persistence for the two species. Persistence and infectivity was best measured using a combination of live bait and flotation techniques.  相似文献

The impact of the nematode-parasitic fungus Hirsutella rhossiliensis on the effectiveness of Steinernema carpocapsae, S. glaseri, and Heterorhabditis bacteriophora against Galleria mellonella larvae was assessed in the laboratory. The presence of Hirsutella conidia on the third-stage (J3) cuticle of S. carpocapsae and H. bacteriophora interfered with infection of insect larvae. Conidia on the J3 cuticle of S. glaseri and on the ensheathing second-stage cuticle of H. bacteriophora did not reduce the nematodes'' ability to infect larvae. The LD₅₀ values for S. carpocapsae, S. glaseri, and H. bacteriophora in sand containing H. rhossiliensis were not different from those in sterilized sand when Galleria larvae were added at the same time as the nematodes. However, when Galleria larvae were added 3 days after the nematodes, the LD₅₀ of S. glaseri was higher in Hirsutella-infested sand than in sterilized sand, whereas the LD₅₀ of H. bacteriophora was the same in infested and sterilized sand. Although the LD₅₀ of S. carpocapsae was much higher in Hirsutella-infested sand than in sterilized sand, the data were too variable to detect a significant difference. These data suggest that H. bacteriophora may be more effective than Steinernema species at reducing insect pests in habitats with abundant nematode-parasitic fungi.  相似文献

全氟辛烷磺酸(Perfluorooctane sulfonate, PFOS)是一种广泛存在于水生生态系统的新型持久性有机污染物(Persistent Organic Pollutants, POPs),其对鱼类健康的影响以及水生生态系统安全的潜在威胁是当前人们高度关注的水环境问题。为探究PFOS对斑马鱼(Danio rerio)胚胎及仔鱼的生态毒理效应,本文研究了不同浓度(0,0.1,1,10 mg/L)PFOS暴露对斑马鱼胚胎孵化率、仔鱼畸形率与死亡率、仔鱼心率、仔鱼运动行为以及生长的影响。结果表明:PFOS暴露对斑马鱼胚胎孵化率、孵出仔鱼死亡率与畸形率的影响显著(P﹤0.05),10 mg/L PFOS暴露导致胚胎孵化率下降,孵化延迟,仔鱼死亡率与畸形率升高;PFOS暴露4 dpf(day post-fertilization,dpf)或8 dpf 对斑马鱼仔鱼心率影响显著(P﹤0.05),心率随PFOS暴露浓度升高而增加;PFOS 暴露6 dpf 或9 dpf 对斑马鱼仔鱼的运动行为影响显著(P﹤0.05),10 mg/L PFOS暴露6 dpf 导致运动斑马鱼仔鱼比例和仔鱼最大持续运动距离增加(P﹤0.05),PFOS暴露9 dpf,单位时间内仔鱼的运动距离、停顿频率、平均每次运动距离随PFOS暴露浓度升高而减少(P﹤0.05),最大持续运动距离随PFOS暴露浓度升高而增加(P﹤0.05),呈剂量依赖的毒理学效应;PFOS暴露导致斑马鱼仔鱼体长和吻宽下降(P﹤0.05)或有下降的趋势,并对吻宽/体长、吻宽/头长影响显著(P﹤0.05)。以上研究结果提示:PFOS对斑马鱼胚胎及仔鱼具有显著的发育与行为毒性,仔鱼心率、运动行为、吻宽/体长以及吻宽/头长等是评估水体PFOS污染敏感而有效的生物标志物。  相似文献

Changes in the volumes of second-stage juveniles of Meloidogyne incognita were monitored in aqueous solutions of polyethylene glycol supplemented with dilute balanced salts. At key points within a 48-hour cycle of fluctuating water potential, nematodes were placed under hypoxic conditions or exposed to the respiratory inhibitor, sodium cyanide, to detect any respiration-dependent process that regulates volume. Aerobic respiratory arrest at -500 kPa induced pronounced water loss, lateral and dorsoventral collapse of the body wall, and abnormal failure to shorten longitudinally. Durations of hypoxia that were innocuous in dilute solutions were lethal during 500 kPa increases and decreases in water potential; the same water potential changes under aerobic conditions had no effect on viability. Data are consistent with the hypothesis that respiration is essential to survive water potential changes.  相似文献

Infection of cat flea, Ctenocephalides felis, larvae by the entomophilic nematode Neoaplectana carpocapsae was accomplished in the laboratory. The Breton strain of N. carpocapsae provided higher larval mortality at lower dosages than did the DD-136 strain. Adult nematodes were evident in the insect hemocoel after 48 h; however, no infective third-stage larvae were produced. Larval flea infection increased with an increase in the moisture content of sand from 2% to 7% and of sandy clay from 7% to 12%. Larval flea infection was also obtained on turf containing dauer larvae. Nematode penetration of cocoons with invasion of prepupal and pupal fleas was apparent.  相似文献

Mice infected orally with third-stage larvae of Heligmosomoides polygyrus were killed at various times after infection. Their small intestines were removed, tied at each end and incubated at 37 C in dilute culture medium. When intestines were taken from mice infected for a period of between 1 and 7 days, a number of developing larvae comprising up to 20% of the infective dose emerged within 60 min through the intestinal wall into the medium. The recovery of emergent larvae was highest using intestines from mice infected 36 to 120 hr previously. The proportion of parasites emerging from the intestines of 48-hr-infected mice was similar for doses of 100 to 2400 larvae. Significantly fewer larvae emerged from the intestines of mice resistant to reinfection and challenged with third-stage larvae 36–72 hr before necropsy.  相似文献

During a study on the breeding rate of the earthworms Lumbricus rubellus and Eisenia foetida, nematodes identified as Rhabditis terricola were found consistently in large numbers in earthworm cocoons that failed to hatch after an adequate incubation period. This nematode species, which was previously known as a saprophyte, was found to invade earthworm cocoons and reproduce within, causing extensive productivity losses in earthworm cultures. In this study, R. terricola was effectively eradicated from earthworm cultures by rinsing the earthworms in tap water and transferring them repeatedly to sterile bedding.  相似文献

The embryological and postembryological development of Thelastoma bulhoesi was determined. Initial cleavage was into unequal cells and occurred within 1-2 hours at 25 C. Cell division was holoblastic but no true morula is formed. Gastrulation occurred at approximately 48 hours by epibolic synectic mechanisms. First-stage larvae were fully developed at 96 hours. The molt to second-stage larvae was initiated in the egg and was completed at hatching. Second-stage larvae were first observed in the host at 11 hours postinfection, third-stage larvae at 18 hours, and fourth-stage larvae at 192 hours. Adult female worms were observed at 32 days. Thiabendazole, in even the lowest concentrations, inhibited the developmem of unembryonated ova.  相似文献
