Spike activity was analyzed in the course of visual testing for directional sensitivity in 177 neuronal populations in different thalamic nuclei and the striopallidal complex in the brain of nine parkinsonian patients, diagnosed and treated using implanted intracerebral electrodes. Directionally selective neurons were discovered in the centrum medianum, the thalamic zona incerta and reticular nucleus, the caudate nucleus, and the central area of the globus pallidus. In acute experiments on cats anesthetized with thiopental (30–40 mg/kg, intraperitoneally) and immobilized with D-tubocurarine (1 mg/kg) responses of 145 neurons of the reticular and 158 neurons of the ventral anterior nuclei of the thalamus to electrical stimulation of the centrum medianum were investigated. An antidromic action potential appeared after a latent period of 0.3–2.0 msec in 4.1% of cells of the reticular nucleus and 4.4% of neurons of the ventral anterior nucleus tested in response to stimulation. The conduction velocity of antidromic excitation along axons of these neurons was 1.7–7.6 m/sec. Neurons responding with an antidromic action potential to stimulation both of the centrum medianum and of other formations were discovered, electrophysiological evidence of the ramification of such an axon. Altogether 53.8% of neurons of the reticular nucleus and 46.9% of neurons of the ventral anterior nucleus responded to stimulation of the centrum medianum by orthodromic excitation. Among neurons excited orthodromically two groups of cells were distinguished: The first group generated a discharge consisting of 6–12 action potentials with a frequency of 130–640 Hz (the duration of discharge did not exceed 60 msec), whereas the second responded with a single action potential. Inhibitory responses were observed in only 0.7% of neurons of the reticular nucleus and 4.4% of the ventral anterior nucleus tested. Afferent influences from the relay nuclei of the thalamus, lateral posterior nucleus, and motor cortex were shown to converge on neurons responding to stimulation of the centrum medianum.A. A. The morphology and topography of neurons whose axons form the nonspecific thalamic input in the primary somatosensory area were studied in the cat forebrain by the retrograde axonal horseradish peroxidase transport method. Stained cells were found in the dorsolateral part of the nucleus ventralis anterior, and were diffusely distributed in the nucleus centralis, lateralis, the lateral part of the nucleus dorsalis medialis, and the dorsal part of the centrum medianum. In the nucleus paracentralis only solitary, palely stained neurons were detected. Cells stained with horse-radish peroxidase were multipolar, triangular, or fusiform. The results are evidence that besides the ventrobasal complex, the nonspecific nuclei of the diencephalon also project into the somatosensory cortex. After microinjections of horseradish peroxidase into the central tegmental area of the midbrain and centrum medianum thalami in cats, labeled neurons were found in the nucleus of the tractus solitarius, gracile and cuneate nuclei, spinal nuclei of the trigeminal nerve, the external nucleus and nucleus of the brachium of the inferior colliculus, the medial pretectal region, nucleus of the posterior commissure and stratum intermediale of the superior colliculus, and reticular structures of the medulla and pons. Comparison of the location of the sources of ascending afferent projections in the central tegmental area of the midbrain and centrum medianum thalami showed that the reticular formation receives mainly visceral projections through the nucleus of the tractus solitarius, whereas the centrum medianum thalami is innervated mainly by the system of sensory somatic nuclei.I. Responses of caudate neurons to stimulation of the anterior sigmoid and various parts of the suprasylvian gyrus were studied in acute experiments on cats. The experiments consisted of two series: on animals with an intact thalamus and on animals after preliminary destruction of the nonspecific thalamic nuclei. Stimulation of all cortical areas tested in intact animals evoked complex multicomponent responses in caudate neurons with (or without) initial excitation, followed by a phase of inhibition and late activation. The latent periods of the initial responses to stimulation of all parts of the cortex were long and averaged 14.5–25.5 msec. Quantitative and qualitative differences were established in responses of the caudate neurons to stimulation of different parts of the cortex. Considerable convergence of cortical influences on neurons of the caudate nucleus was found. After destruction of the nonspecific thalamic nuclei all components of the complex response of the caudate neurons to cortical stimulation were preserved, and only the time course of late activation was modified.A. Spike response was investigated in 104 neurons of the nucleus reticularis thalami (R) and adjoining thalamic nuclei to acoustic, tactile, and visual stimuli during chronic experiments on cats. Of the test neurons, 29% responded to acoustic stimulation and 11% showed no preference in relation to different acoustic stimuli. Minimum latencies of response to sounds measured 12–37 msec in excitatory and 18–27 msec in inhibitory cells. Duration of excitation produced by acoustic stimuli reached 50–250 msec; inhibition lasted 27–190 msec. Most cells belonging to this nucleus were excited by different stimuli; the proportion of inhibitory neurons did not exceed 4–10%.A. A. Structural and ultrastructural changes in the frontal areas of the cortex and in the region of the globus pallidus were investigated after local and extensive destruction of the caudate nucleus. It was shown by the Fink-Heimer method that after local injury to the caudate nucleus by means of electrodes implanted 2–16 months before electrolytic destruction, only a few degenerating fibers of medium and thin caliber were present. Extensive destruction of the caudate nucleus (without preimplantation of electrodes) was followed by massive degeneration of fibers of different caliber in the frontal area of the cortex. After local injury to the caudate nucleus numerous thin degenerating axons 0.5–0.6 µ in diameter and degenerating terminals were found in the region of the globus pallidus. Degenerative changes in the axo-dendritic and axo-somatic terminals followed the "dark" type of course. It is concluded that no considerable direct projections of neurons of the caudate nucleus are present in the cortex. Degenerating fibers of average caliber in frontal areas of the cortex after destruction of the caudate nucleus are evidently axons of thalamic neurons and not from cells of the damaged nucleus.A. A. Responses of 137 neurons of the rostral pole of the reticular and anterior ventral thalamic nuclei to electrical stimulation of the ventrolateral nucleus and motor cortex were studied in 17 cats immobilized with D-tubocurarine. The number of neurons responding antidromically to stimulation of the ventrolateral nucleus was 10.5% of all cells tested (latent period of response 0.7–3.0 msec), whereas to stimulation of the motor cortex it was 11.0% (latent period of response 0.4–4.0 msec). Neurons with a dividing axon, one branch of which terminated in the thalamic ventrolateral nuclei, the other in the motor cortex, were found. Orthodromic excitation was observed in 78.9% of neurons tested during stimulation of the ventrolateral nucleus and in 52.5% of neurons during stimulation of the motor cortex. Altogether 55.6% of cells responded to stimulation of the ventrolateral nucleus with a discharge of 3 to 20 action potentials with a frequency of 130–350 Hz. Similar discharges in response to stimulation of the motor cortex were observed in 30.5% of neurons tested. An inhibitory response was recorded in only 6.8% of cells. Convergence of influences from the thalamic ventrolateral nucleus and motor cortex was observed in 55.7% of neurons. The corticofugal influence of the motor cortex on responses arising in these cells to testing stimulation of the ventrolateral nucleus could be either inhibitory or facilitatory.A. A. Neuronal activity in the ventrolateral thalamus during execution of instrumental reaction before and after parenteral administration of N-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) was investigated in samples of 81 and 70 cells, respectively. After a 5-day course of one 5 mg/kg MPTP injection daily, firing rate of neurons in which activity correlated with forelimb movement rose significantly; this activation increased in length during the initial, flexor, and extensor stages of motor response. Bradykinesia set in together with intensified neuronal activation in the animals. Microinjection of exogenous dopamine into the caudate nucleus brought about correction of motor disturbance and a reduced neuronal firing rate in the ventrolateral (thalamic) nucleus. It was deduced that the nigrostriatal system exercises inhibitory control over the activity of thalamic neurons associated with forelimb movement in thalamic neurons in intact animals.A. A. Functional connections between different parts of the caudate nucleus and particular thalamic nucleic were investigated during the onset and development of the recruiting response in chronic experiments on cats. The method of condioning and testing stimulation of subcortical structures was used. The highest degree of functional connection was observed between the caudate nucleus and ventral anterior thalamic nucleus in the ipsilateral hemisphere. It is suggested that caudatethalamic functional connections in the development of synchronized activity are due to the activity of two mechanisms: general and facultative thalamic pacemakers.A. A. Responses of caudate neurons to electrical stimulation of the afferent input from thepulvinar thalamic nucleus and to visual stimuli of various orientations were studied extracellularly in awake chronic cats. Activation responses dominated among reactions of these neurons. The response latencies have ranged from 4 to 85 msec for units with primary activation and from 20 to 150 msec for inhibited ones. The values are indicative of both rapidly and slowly conducting afferent pathways. A possibility of monosynaptic transmission in thepulvinarcaudate projections is also revealed.Pulvinar stimulation is found to be efficient for a significant (more than 50 percent) number of caudate neurons responding to visual stimuli, including orientation-selective cells. The mode of influences from other structures of the visual system (optic tract, area 17, the Clare-Bishop area) on caudate neurons responding topulvinar stimulation is described. The data are discussed with respect to the possible role of cortical and subcortical projections of the visual system in the creation of sensory specific responses of the caudate nucleus.A. A. Synchronized activity (spindles, augmentation response) evoked by stimulation of thalamic nonspecific, association, and specific nuclei was investigated in chronic experiments on 11 cats before and after successive destruction of the caudate nuclei. After destruction of the caudate nuclei the duration of spindle activity in the frontal cortex and subcortical formations (thalamic nuclei, globus pallidus, putamen) was reduced to only three or four oscillations. In the subcortical nuclei its amplitude fell significantly (by 50±10%); in the cortex the decrease in amplitude was smaller and in some cases was not significant. Different changes were observed in the amplitude of the augmentation response, depending on where it was recorded. In the subcortical formations it was considerably and persistently reduced (by 50±10%); in the cortex these changes were unstable in character. Unilateral destruction of the caudate nucleus inhibited synchronized activity evoked by stimulation of the thalamic nuclei on the side of the operation only. Destruction of the basal ganglia (caudate nucleus, globus pallidus, entopeduncular nucleus, and putamen) did not prevent the appearance of synchronized activity; just as after isolated destruction of the caudate nucleus, after this operation synchronized activity was simply reduced in duration and amplitude. It is suggested that the caudate nucleus exerts an ipsilateral facilitatory influence on the nonspecific system of the thalamus during the development of evoked synchronized activity.A. A. Zusammenfassung Die räumliche Gestalt des Centrum medianum thalami und des Nucleus parafascicularis des Menschen wird beschrieben. Die Darstellung stützt sich auf die Untersuchung von 800–1000 dicken, mit Aldehydfuchsin gefärbten Schnitten unter dem Stereomikroskop. Mit dieser Methode werden Lipofuscinkörnchen und Lysosomen in den Nervenzellen elektiv dargestellt. Während die Nervenzellen im Nucleus parafascicularis und im parvocellulären Teil des Centrum medianum annähernd gleichmäßig verteilt sind, zeigt die Pars magnocellularis einen verwickelten Aufbau aus zelldichten Lamellen und locker gebauten Partien. Der Nucleus parafascicularis ist durch mehrere hörnerartige Vorsprünge mit dem Centrum medianum verzahnt. Seine Grenzlinie zu diesem Kern ist scharf.

Summary The three dimensional shape of the centrum medianum thalami and the nucleus parafascicularis of the human is described on the basis of 800–1000 thick sections, which are stained with aldehydefuchsin and studied under the binoculars. By this method lipofuscin granules and lysosomes in the nerve cells are stained selectively. Whereas the nerve cells of the nucleus parafascicularis and the parvocellular part of the centrum medianum are evenly distributed the magnocellular part exhibits a more complicated composition consisting of dense cell layers and loosely arranged parts. The nucleus parafascicularis is tightly connected with the centrum medianum by mutual indentations, both nuclei, however, being sharply outlined by clear delimitations.

During chronic experiments on unanesthetized cats neuronal response in the caudate nucleus to the presentation of local photic stimuli and electrical stimulation of the specific (field 17) and the association (Clare-Bishop) areas were compared. Stimulation of the Clare-Bishop area proved more effective than stimulating field 17 for neurons of the caudate nucleus; a response was produced in 47% of test neurons in comparison with 8% of units only in the specific area. Lower average values were observed for latency of neuronal response to stimulation of the Clare-Bishop area. An insignificant number of caudate nucleus neurons were activated as a result of stimulation of both cortical areas. A comparison between the response of one set of neurons to electrical cortical and visual stimulation showed that cells responding to visual stimulation were more highly activated by stimulating the Clare-Bishop area than by stimulation of field 17. This type of neuron predominated in the caudate nucleus. A discussion follows of the possible involvement of the Clare-Bishop area in shaping neuronal response to visual stimulation.A. A. Location within the brain of retrogradely labeled neurons putting out projections from the dorsal magnocellularis area of the red nucleus was investigated by means of microiontophoretic injection of horseradish peroxidase into the dorsal magnocellularis area of the cat red nucleus. Projections were found from a number of hypothalamic nuclei, the centrum medianum, parafascicular and subthalamic nuclei, zone incerta, Forel's field, nucleus medialis habenulae, pontine and bulbar reticular formation, and the following midbrain structures: the central gray matter, superior colliculus, Cajal's interstitial nucleus, reticular formation, and the contralateral red nucleus. Projections were also identified proceeding from more caudally located structures: the cerebellar fastigial nucleus, facial nucleus, medial vestibular and dorsal lateral vestibular nuclei, and ventral horns of the spinal cord cervical segments. Connections between the substantia nigra and the red nucleus were clarified. Projections to the red nucleus from the cerebral cortex, interstitial and dentate (lateral) cerebellar nuclei, the nucleus gracilis and cuneate nucleus were found, confirming data presented in the literature. Unitary responses of the caudate nucleus to stimulation of various parts of it were investigated by extracellular recording. Latent periods of response discharges varied from 3.5 to 40 msec. Most neurons were excited by stimulation of the most rostral part of the head of the caudate nucleus. Irrespective of the site of stimulation, in most cases responses consisted of initial excitation in the form of one or, less frequently, two discharges followed by a period of depression of spontaneous activity. Recovery of activity took place gradually, without postinhibitory facilitation. No afterdischarges or periodic repetitions of spikes were observed after the initial response. Repetitive stimulation of the caudate nucleus showed that the neurons of this nucleus reproduce frequencies of stimulation badly above 30/sec, and under these circumstances in many cases they continued to discharge on average at a frequency of 5–15/sec. The results are examined from the standpoint of participation of the caudate nucleus in the formation of spindle activity.A. A. Stimulation of the head of the caudate nucleus in cats anesthetized with chloralose and pentobarbital evoked spike responses of the Purkinje cells and other cerebellar cortical neurons in the paramedian lobes, lobulus simplex, and the tuber of the vermis. Phasic responses in the form of simple discharges (on account of activation of the neurons through mossy fibers) appeared mainly after a latent period of 5–12 and 14–20 msec; the latent period of responses consisting of complex discharges (on account of activation of Purkinje cells through climbing fibers) was 5–6, 9–22 msec, or more. Depending on the latent period, the spike responses differed in their rhythm of generation. In response to stimulation of the caudate nucleus with a frequency of 4–6/sec recruiting responses were found. An inhibitory pause was an invariable component of the tonic responses. During stimulation of the globus pallidus responses of the same types (phasic and tonic) appeared as during stimulation of the caudate nucleus, but they differed in the distribution of the neurons by latent period of spike responses. The minimal latent period was 4 msec. Recruiting also was observed during repetitive stimulation of the globus pallidus. During stimulation of the substantia nigra Pukinje cells activated by climbing fibers responded. Evoked complex discharges appeared after a stable latent period of 8.5±0.3 msec. Arguments are put forward regarding the role of the substantia nigra, the globus pallidus, nuclei of the inferior olive, and also the thalamic nuclei in the mechanism of caudato-cerebellar oligosynaptic and polysynaptic connections.N. Responses of 141 neurons of the caudate nucleus to acoustic stimuli — tones (500 and 2000 Hz) and clicks of different frequency (0.2 and 0.8/sec) and intensity (75, 80, 95 dB) — were recorded extracellularly in chronic experiments on cats. The responses recorded showed great variability with respect to character (phasic, tonic), structure (one or two phases of excitation), latent periods (from 7.5 to 300.0 msec), and burst discharge frequency (from 90 to 800 spikes/sec). Analysis of averaged poststimulus histograms and graphs of the dynamics of the responses showed that responses of 74% of neurons were much better expressed if less frequent stimuli were used: The regularity of the responses and the number of spikes in each response increased. Responses of neurons also increased and acquired a more distinct temporal structure if the intensity of the clicks increased. The character of responses to clicks and tones differed qualitatively in 17% of neurons studied: Phasic excitation arose in response to clicks, tonic changes in spike activity to tones. Extracellular investigations on the activity of 269 caudate neurons during electrical stimulation of the midbrain reticular formation were carried out during chronic experiments on cats. Stimuli of different sensory modalities were used: auditory, mechanical, and visual. A response was observed to both reticular and peripheral stimulation in single neurons. The former produced an orthodromic response in 53% of caudate neurons, notable for its high probability of occurrence. A total of 23% of caudal neurons responded to this type of stimulation and application of stimuli of a single modality, while 14% responded polymodally. An excitatory response pattern prevailed during all types of stimulation. By applying twin stimuli to 100 caudate neurons, a capacity for interaction between reticular and acoustic inputs was discovered. Interaction was similarly observed in neurons which had reacted neither to separate application of both stimuli nor to either of the stimuli in isolation.A. A. Spontaneous and evoked unit activity in response to repeated application of clicks at a frequency of 0.3–2.0 Hz in the caudate nucleus was studied by an extracellular recording technique in chronic experiments on cats. Four types of spontaneous unit activity in the caudate nucleus were distinguished. Altogether 44% of neurons tested responded by changes in spontaneous activity to clicks. Five types of responses of caudate neurons to clicks were discovered: phasic excitation, phasic inhibition, tonic activation, tonic inhibition, and mixed tonic responses; the commonest type was tonic activation. During prolonged stimulation by clicks extinction of the phasic responses was not observed. Complete or partial extinction of tonic responses in the course of frequent repetition of stimulation was observed in 33% of responding neurons.
