Human angiogenin was genetically engineered and contained the E. coli Omp A signal sequence for secreting soluble angiogenin to the periplasm under tac promoter control. The angiogenin sequence was encoded in a single gene and expressed as a 14.4 kilodalton soluble protein in E. coli. It was purified by CM-Sepharose ion-exchange chromatography and by a heparin-Sepharose affinity chromatography procedure. The biological activity of angiogenin was established by its ability to inhibit mRNA-dependent rabbit reticulocyte cell-free translation.  相似文献

A method for obtaining authentic human angiogenin utilizing an Escherichia coli recombinant expression system is described. A synthetic gene encoding angiogenin was placed into a vector for direct expression under the control of a modified E. coli trp promoter. The protein was produced by the bacteria in an insoluble form and purified to homogeneity by cation-exchange and reversed-phase HPLC following reduction/solubilization and reoxidation. The protein isolated was identified as Met-(-1) angiogenin by amino acid analysis and tryptic peptide mapping; the latter demonstrated that all three disulfide bonds had formed correctly. Both the enzymatic and angiogenic activities of the Met-(-1) protein were equivalent to those of native angiogenin. A Met-(-1) Leu-30 derivative of angiogenin was also isolated and found to be fully active. Conversion of Met-(-1) angiogenin to the authentic less than Glu-1 protein was achieved by treatment with Aeromonas aminopeptidase under conditions in which the new N-terminal glutamine readily cyclizes nonenzymatically. This aminopeptidase treatment may have more general applicability for removal of undesirable N-terminal methionine residues from foreign proteins expressed in bacteria.  相似文献

A synthetic gene coding for human angiogenin was cloned in pUR290 plasmid in frame with beta-galactosidase, both parts of the resultant fused protein being joined through an Asp-Pro sequence. The fused protein, synthesised in E. coli cells upon IPTG induction and isolated as inclusion bodies, possessed angiogenic activity on the chick chorioallantois membrane and was cleaved upon acid treatment to yield free angiogenin.  相似文献

A synthetic gene coding for the cysteine proteinase inhibitor (desSer1 Ile29 Leu89) chicken cystatin was cloned and expressed in E. coli. The gene was assembled from 12 oligonucleotides and inserted into vector pUC 8. Expression as fusion protein was performed in a temperature-inducible E. coli system. The expression product was synthesized as 20% of total E. coli protein. The fusion protein was purified, the chicken cystatin homologue was split off with CNBr and the N-terminal sequence confirmed up to position 37. The properties of the purified material correspond to those of natural chicken cystatin. The recombinant cystatin variant binds anti-chicken cystatin IgG, is inhibitorily active and displays Ki values with papain and with cathepsin B similar to those determined for natural chicken cystatin.  相似文献

目的:获得大鼠crip2基因片段,并在大肠杆菌中表达、纯化大鼠CRIP2(cysteine-rich intestinal protein 2)蛋白。方法:从大鼠主动脉组织中提取总DNA,RT-PCR扩增出相应大小的crip2 DNA片段,与pGEM-T-easy载体连接后测序;将测序正确的crip2按照BamHⅠ和HindⅢ酶切位点克隆入原核表达载体pRSET A,将连接产物转化大肠杆菌BL21,挑出阳性克隆,IPTG诱导表达重组的6×His融合蛋白,通过镍柱进行纯化。结果:PCR获得的crip2序列与GenBank报道的一致(为707 bp);重组融合蛋白在大肠杆菌BL21中以可溶形式高效表达,经SDS-PAGE和Western印迹分析,在相对分子质量为27×103处有特异的蛋白条带,经镍柱纯化后,得到了高纯度的CRIP2融合蛋白。结论:克隆了大鼠crip2基因片段,并在大肠杆菌BL21中高效表达,亲和层析纯化后获得高纯度的CRIP2融合蛋白。  相似文献

A synthetic gene containing the coding sequence for the human cysteine proteinase inhibitor, cystatin A, was obtained by enzymatic assembly of 20 oligodeoxyribonucleotides which had been chemically synthesized by the solid phase phosphoramidite method. It was cloned into an Escherichia coli plasmid. The expression plasmid for cystatin A was constructed by introducing the synthetic gene downstream of the tac promoter of an E. coli plasmid which is a derivative of pKK223-3 with high copy number. The gene was expressed in E. coli JM109 without IPTG-induction. The expression of cystatin A was detected by SDS-polyacrylamide gel electrophoresis of the E. coli JM109 lysate, followed by immunoblotting using rabbit antiserum raised with human epidermal cystatin A and alkaline phosphatase-conjugated goat anti-rabbit IgG. The result showed that the molecular weight of the expression product is identical with that of the authentic protein and the antigenic properties are also the same. Furthermore, the expression product purified with a CM-papain Sepharose affinity column and FPLC system with a Mono-Q column showed the same inhibitory activity for various cysteine proteinases. Also, purified recombinant cystatin A was found to have identical amino acid composition, NH2-terminal amino acid sequence, and peptide-map on reverse phase HPLC with those of the authentic inhibitor.  相似文献

将北非蝎昆虫毒素 (AndroctonusaustralisHectorinsecttoxin ,AaHIT1)基因克隆入温度诱导表达载体pBV2 2 0 ,并在宿主菌E .coliDH5α中进行诱导表达 .表达定位分析发现 ,AaHIT1大部分存在于包涵体中 .蛋白质N 端测序证实了体外表达的AaHIT1蛋白的正确性 .将AaHIT1基因转化入烟草中并获得了转基因植株 ,基因组PCR、RT PCR及Northern印迹分别证实AaHIT1基因已正确地插入到烟草基因组中并已得到表达 .抗虫实验表明 ,该转基因烟草对白粉虱有明显的抗性 .该研究为蝎毒AaHIT1的生物学毒性实验以及获得抗虫转基因作物奠定了基础  相似文献

Recombinant bovine angiogenin (rbAng) was expressed in E. coli at up to 30% of total cell proteins but was produced as inclusion bodies. By investigating the effect of various factors on the refolding yield, we obtained about 60% refolding. After chromatographic purification, about 60 mg purified angiogenin was obtained from 1 l culture. The purified recombinant bovine angiogenin was identical to native bovine angiogenin (nbAng) obtained from cow's milk. Our approach is highly efficient and can be generally used for the production of various types of angiogenin for functional and structural studies as well as therapeutic purposes.  相似文献

The nucleotide sequence coding for human angiogenin has been deduced from the published amino acid sequence with the use of codons preferentially utilized in highly expressed E. coli genes. It was divided into forty-three oligonucleotides, which were synthesized by automatic gene assembler and then joined by DNA ligase into three double-stranded blocks, the blocks were consequently cloned and ligated in M13mp8 phage, and the resultant 389-bp DNA sequence coding for human angiogenin was analysed by chain-terminator sequencing technique.  相似文献

通过RT PCR的方法从人外周血白细胞扩增血管生成素 (Ang)cDNA .在计算机分子结构模建的基础上 ,通过柔性连接臂构建了Ang与Gfp融合基因 ,并在大肠杆菌DH5α中实现了高效表达 .重组蛋白占菌体总蛋白的 32 %.融合蛋白经初步纯化后 ,在紫外线激发下可见明显的绿色荧光 ,同时能够显著地促进鸡胚尿囊膜毛细血管的新生 ,而且所获融合蛋白在体外具有促进人脐静脉血管内皮细胞增殖的作用 .这种双功能融合蛋白的表达为阐明Ang的核转位过程奠定了基础 ,同时为阐明血管新生的分子机制创造了条件  相似文献

大肠埃希菌Mn-SOD基因的克隆、表达及多克隆抗体制备   总被引:1,自引:0,他引:1  
目的实现Mn-SOD基因在大肠埃希菌中的高可溶性表达,制备Mn-SOD的多克隆抗体。方法用PCR方法从一株野生型大肠埃希菌(E.coli)基因组中扩增Mn-SOD基因编码区.将它克隆到原核表达载体上进行大量表达和纯化,再用纯化的蛋白对新西兰大白兔进行背部多点注射,40d后取其血清,用Western-blot印迹实验测定抗体效果。结果SDS-PAGE分析表明SOD的表达量约为细菌总蛋白的50%;黄嘌呤氧化酶法测定表达蛋白活性,结果表明每毫克菌体可溶性总蛋白中表达产物酶比活为3921.77U/mg,是对照BL21的276.77倍;并制备了高效价的多克隆抗体。结论该研究成功地构建了大肠埃希菌Mn-SOD基因高效原核表达系统,所表达的Mn-SOD具有良好的免疫原性和免疫反应性。  相似文献

目的:原核表达、纯化DNA损伤检查点蛋白调节子1(MDC1)片段,并制备其多克隆抗体。方法:设计特异引物,通过RT-PCR扩增编码MDC1 N端194个氨基酸残基的基因片段,测序正确后插入含GST基因的原核表达载体pGEX-KG中,以IPTG诱导表达,并经谷胱甘肽琼脂糖珠纯化融合蛋白;用纯化的蛋白免疫小鼠制备多克隆抗体,用ELISA测定抗体的效价,Western印迹鉴定抗体的特异性。结果:原核表达并纯化了MDC1 N端片段,并获得了抗MDC1的多克隆抗体,抗体效价达到1∶12800,Western印迹显示该抗血清能特异识别原核及真核细胞表达的MDC1。结论:MDC1 N端片段能够诱导小鼠产生具有较高效价和特异性的多克隆抗体,为进一步研究MDC1在Fhit特异信号通路中的作用奠定了基础。  相似文献

A highly heat-stable amylase gene from an obligately anaerobic and extremely thermophilic bacterium, Dictyoglomus thermophilum, was cloned and expressed in Escherichia coli. The nucleotide sequence of the amylase gene predicts a 686-amino-acid protein of relative molecular mass 81,200, which is consistent with that determined by sodium dodecyl sulfate/polyacrylamide gel electrophoresis of the purified enzyme. The NH2-terminal sequence determined using the enzyme purified from E. coli cells corresponds precisely to that predicted from the nucleotide sequence, except for the absence of the NH2-terminal methionine in the mature protein. When the amylase gene was expressed in E. coli cells, the enzyme was localized in the cytoplasmic fraction; this is probably explained by the absence of the signal sequence for secretion. By using the amylase purified from the E. coli transformant, some enzymatic properties, such as optimum pH, optimum temperature, pH-stability and heat-stability, were examined. Two improved plasmid vectors, containing the gene coding for staphylococcal protein A and adapted for gene fusions, have been constructed. These vectors allow fusion of any gene to the protein A moiety, giving fusion proteins which can be purified, in a one-step procedure by IgG affinity chromatography. One vector, pRIT2, is designed for temperature-inducible expression of intracellular fusion proteins in Escherichia coli and the other pRIT5, is a shuttle vector designed for secretion. The latter gives a periplasmatic fusion protein in E. coli and an extracellular protein in Gram-positive hosts such as Staphylococcus aureus. The usefulness of these vectors is exemplified by fusion of the protein A gene and the E. coli genes encoding the enzymes beta-galactosidase and alkaline phosphatase. High amounts of intact fusion protein are produced which can be immobilized on IgG-Sepharose in high yield (95-100%) without loss of enzymatic activity. Efficient secretion in both E. coli and S. aureus, was obtained for the alkaline phosphatase hybrid, in contrast to beta-galactosidase which was only expressed efficiently using the intracellular system. More than 80% of the protein A alkaline-phosphatase hybrid protein can be eluted from IgG affinity columns without loss of enzymatic activity.  相似文献

目的:原核表达人Pin2/TRF1结合蛋白X1(PinX1),确定该蛋白与人端粒酶逆转录酶(hTERT)催化亚基的相互作用。方法:用PCR方法从乳腺文库中扩增PinX1基因的编码序列,克隆至pET-32a载体构建重组质粒pHis-PinX1,经双酶切鉴定后转化大肠杆菌BL21并进行诱导表达,SDS-PAGE和Western印迹检测His-PinX1的表达;用His磁珠纯化His-PinX1,在人肾胚细胞293T内检测His-PinX1与hTERT的相互作用。结果:扩增得到980bp的PinX1基因;Western印迹检测表明,相对分子质量约57x10。的His-PinX1获得表达,且纯化的His-PinX1与hTERT具有相互作用。结论:表达并纯化得到了与hTERT相互作用的His-PinX1,为深入研究PinX1的功能奠定了基础。  相似文献

A cDNA coding mutated cecropin CMIV from Bombyx mori was synthesized according to its amino acid sequense using E .coli biased codons .The gene was cloned into the fusion expression vector pEZZ318 and was expressed in E .coli HB101.The fusion protein produced was purified by affinity chromatography to yield 26 mg/L fusion product .The anti-bacterial activities of recombinant cecropin CMIV were recovered after cleavage by chemical method.  相似文献

Aerolysin is a toxin (protein in nature) secreted by the strains of Aeromonas spp. and plays an important role in the virulence of Aeromonas strains. It has also found several applications such as for detection of glycosylphosphatidylinositol (GPI)-anchored proteins etc. A. hydrophila is a ubiquitous Gram-negative bacterium which causes frequent harm to the aquaculture. To obtain a significant amount of recombinant aerolysin in the active form, in this study, we expressed the aerolysin in E. coli under the control of T7 RNase promoter. The coding region (AerA-W) of the aerA gene of A. hydrophila XS91-4-1, excluding partial coding region of the signal peptide was cloned into the vector pET32a and then transformed into E. coli b121. After optimizing the expression conditions, the recombinant protein AerA-W was expressed in a soluble form and purified using His.Bind resin affinity chromatography. Recombinant aerolysin showed hemolytic activity in the agar diffusive hemolysis test. Western blot analysis demonstrated good antigenicity of the recombinant protein.  相似文献
