丙酮酸高产菌株的选育及中试研究   总被引:9,自引:1,他引:9  
对T.glabrata WSH-IP12进行EMS诱变,挑选以NH4Cl为唯一氮源的平板上透明圈较大的菌株,经初筛和复筛后,发现T.glabrata WSH-IP303生产丙酮酸的能力强且稳定。以NH4Cl为唯一氮源摇瓶培养48h,其丙酮酸产量(35.1g/L)比出发菌株(21.4g/L)提高了64%。采用该菌株在300L罐上进行了4批发酵试验,丙酮酸产量最高可达58.4g/L,对葡萄糖产率0.562g/g。  相似文献

一株高产脯氨酸的嗜醋酸棒杆菌的选育及发酵条件优化   总被引:1,自引:0,他引:1  
以嗜醋酸棒杆菌为出发菌株,经过片段化全基因组体外诱变、重组和连续的磺胺胍抗性筛选,获得一株L-脯氨酸的高产菌株。摇瓶发酵优化结果表明,葡萄糖、生物素和硫胺素的最适用量分别为16%、300μg/L、400μg/L,最适pH为6.8~7.0,装液量为25ml/500ml摇瓶,发酵培养72h后L-脯氨酸产率高达到75.6g/L,与对照相比提高了5%。考察了50L发酵罐中细胞生长对L-脯氨酸产量的影响,补料分批发酵结果表明(比生长速率分别为0.06/h、0.08/h和0.1/h),比生长速率在0.08/h左右时L-脯氨酸的产率最高,L-脯氨酸的比生产速率QP达到0.091 g/(g.h),产率高达82.1 g/L,比优化前提高了14%。  相似文献

研究了酵母摇瓶发酵中pH、装液量、初糖浓度、碳氮磷比和补糖方式对谷胱甘肽(GSH)发酵的影响。结果表明,GSH发酵适宜的初始pH和装液量分别为6 0和500ml锥形瓶内装液量60m1。初糖浓度对GSH发酵有较大的影响,超过12g/l,的初糖浓度将减少胞内GSH含量。应用计算得出的一种以控制比生长速率为目的的摇瓶补糖策略,在总糖浓度为26.2g/L下发酵12h,最终细胞浓度和胞内GSH含量分别达到8.78g/L和13.6mg/g,发酵液内GSH总量达到119.4mg/L,细胞对糖产率达到0.335g/g。  相似文献

以黄色短杆菌(Brevibacterium flavum)NJ-237为出发菌株,通过梯度传代适应性培养及同浓度药物平板富集培养的方式,逐步提高菌体的抗药物性能,获得了1株耐高糖和耐高浓度α-氨基丁酸(-αAB)的菌株NJ-2372。在单因素实验的基础上,利用响应面分析法对影响该菌株L-缬氨酸(L-Val)产量的3个重要因素玉米浆、生物素(VH)、硫胺素(VB1)的添加量进行优化。结果表明:当玉米浆、VH、VB1最佳添加量分别为11 g/L、35μg/L和101μg/L时,摇瓶发酵72 h,L-Val摇瓶发酵产量达到52.9 g/L。  相似文献

产甘油假丝酵母 (Candidaglycerolgenesis)发酵产生的有机酸对丙三醇产品质量和产率均有影响。发现在发酵其它条件恒定 ,装液比和玉米浆浓度增加时 ,发酵液总酸是递增的。在装液比为 0 2和玉米浆浓度为 8g L时 ,丙酮酸和乳酸在细胞生长期可分别积累达 4 1g L和 1 0g L ,比正常发酵时增加 2倍以上 ,丙三醇产率也低 ;然而 ,装液比为 0 0 8和玉米浆浓度为 4g L时 ,丙酮酸和乳酸产生较低 ,丙三醇产率较高 ,但乙酸积累比供氧不足时高 ,可达 2 6g L。发酵过程中有机酸被细胞代谢 ,含量逐渐下降 ,如在初糖浓度为 1 0 0g L时 ,有机酸在细胞生长期积累至高峰后 ,丙三醇和有机酸随之均降低至较低含量 ,并且丙酮酸或乳酸可以转化为乙酸。此外 ,在外加一些添加剂时对其产生有机酸也有影响 ,如添加 1 %油酸和VB1时可以降低乙酸的积累 ,同时增加丙酮酸的含量 ,丙三醇产量也有所增加 ;而丙酮酸结构类似物氟代丙酮酸和亚硫酸盐促进乙酸的产生 ,使酮戊二酸合成减少 ,丙三醇产量约增加 2 0 %。  相似文献

α-酮戊二酸和丙酮酸是重要的酮酸,广泛应用于食品、医药等领域。以本研究室在前期的工作中诱变选育的Yarrowia lipolytica WSH-Z06 C3为出发菌株,高效联产α-酮戊二酸和丙酮酸。在摇瓶水平上初步确定了最佳氮源浓度以及接种密度。在此基础上,重点考察了在15 L发酵罐上硫胺素浓度以及溶氧控制参数对酮酸联产的影响。结果显示,硫胺素最佳浓度为0.2μg/L,α-酮戊二酸和丙酮酸分别高达24.5、53.7 g/L。与未优化的对照相比,酮酸总产量和丙酮酸总转化率均提高了57.9%。溶氧水平控制在50%,发酵96 h酮酸总产量高达53.2 g/L。通过两阶段溶氧调控,酮酸总产量高达64.7 g/L,比未调控前提高了21.2%。通过硫胺素与溶氧水平的优化,显著提高了酮酸的产量,进一步为酮酸的工业化生产提供参考。  相似文献

通过液体发酵的摇瓶实验,考察碳源、氮源、碳氮比和乙醇浓度对云芝糖肽的产率影响,确定葡萄糖和玉米淀粉为适宜碳源,蛋白胨和黄豆饼粉为适宜氮源,合适的碳氮比为20∶1,适宜的乙醇体积分数为1.5%。采用以上的培养基进行70 L发酵罐进行扩大实验和10 t工业生产规模试验。发酵周期32 h,最终生物量达到13.0 g/L,云芝糖肽产率平均为2.21 g/L,与摇瓶发酵水平相当。此外工业实验还确定了云芝液体发酵的pH值、残糖浓度以及菌丝体形态等终点指标。  相似文献

对基因工程菌Pichiapastoris的摇瓶发酵条件进行了试验 ,并根据摇瓶发酵的优化结果进行了补料分批高密度发酵。在摇瓶发酵时 ,甲醇诱导基因工程菌P .pastoris表达重组人血清白蛋白的发酵周期为 96h ;甲醇的最佳诱导浓度为 1 0g L ;发酵pH范围为 5 72~ 6 5 9;在摇瓶培养时 ,随着接种量的增加 ,虽然目的蛋白表达量缓慢增加 ,但单位细胞光密度的蛋白产率却明显下降 ,符合y =1 2 941x- 0 50 59方程 (线性相关系数r=0 9789) ,其限制性因子很可能为溶氧。在分批发酵 ,接种量为 1 0 %且种子细胞光密度 (OD60 0 )为 2 0左右时 ,细胞生长的延迟期为 2 1 1h左右 ,细胞生长光密度与培养时间的关系模型为 :y =0 7841e0 .2 3 19t(线性相关系数r=0 .993 6 ) ;在补料发酵时细胞干重浓度可达到 1 1 5g L— 1 6 0g L ,在 1 2 0h重组人血清白蛋白表达量最大达到 3 6g L。  相似文献

对Torulopsis glabrata WSH-IP303进行NTG诱变,挑选以乙酸为补充碳源的平板上透明圈较大的菌落,经初筛和复筛,发现T.glabrataWSH-LQ307生产丙酮酸能力强且稳定。以乙酸为补充碳源摇瓶培养48h,其丙酮酸产量(46.2g/L)比出发菌株(38.3g/L)提高21%,采用该菌株在5L发酵罐上进行4批发酵实验,丙酮酸产量在64h最高可达68.7g/L,对葡萄糖的转化率为0.651g/g。  相似文献

A multi-vitamin auxotroph, Torulopsis glabrata strain WSH-IP303, which can use ammonium chloride as a sole nitrogen source for pyruvate production, was selected. To optimize pyruvate yield and productivity, a simple but useful, orthogonal design method, was used to investigate the relationship between thiamine, nicotinic acid, pyridoxine, biotin, and riboflavin. Thiamine was confirmed to be the most important factor affecting pyruvate production. When the concentration of thiamine was 0.01 mg/l or 0.015 mg/l, glucose consumption was improved by increasing the nicotinic acid concentration. When the concentrations of nicotinic acid, thiamine, pyridoxine, biotin, and riboflavin were 8.0, 0.015, 0.4, 0.04, and 0.1 mg/l, respectively, pyruvate concentration and yield reached 52 g/l and 0.52 g/g, respectively, in a 48-h flask culture. By employing a combination of the optimum vitamin concentrations, a batch culture was conducted in a 2.5-l fermentor with an initial glucose concentration of 112 g/l; and the pyruvate concentration reached 69 g/l after 56 h (yielding 0.62 g/g).  相似文献

The effects of glucose, vitamins, and DO concentrations on efficient pyruvic acid fermentation were investigated using Torulopsis glabrata IFO 0005, and a novel biphasic culture method was developed on the basis of the metabolic flux analysis. T. glabrata requires the four vitamins nicotinic acid (NA), thiamine hydrochloride (B(1)), pyridoxine hydrochloride, and biotin for cell growth. The deficiency of these vitamins plays an essential role in pyruvate fermentation. In the present study, we considered the effects of the first two vitamins on the pyruvate fermentation. On the basis of several batch and fed-batch experiments, it was found that, as a result of glucose inhibition of cell growth, the initial glucose concentration should be around 30-40 g/L, and the glucose concentration during fermentation should be controlled at high level around 30 g/L. On the basis of an analysis of carbon flux distribution, a biphasic fermentation method was developed where the cultivation started with a high DO (at 40-50% of air saturation) for efficient cell growth and then was reduced to 5-10% for efficient pyruvate production. Since a fair amount of ethanol was formed when the DO concentration was decreased, the addition of NA turned out to be effective in reducing the ethanol formation. This may be due to a relaxing of the requirement for NADH oxidation by the alcohol dehydrogenase pathway. Since B(1) affects both the pyruvate dehydrogenase complex and pyruvate decarboxylase, its initial concentration must be carefully determined by considering both the cell growth and pyruvate production phases.  相似文献

Yarrowia lipolytica as an oleaginous yeast is capable of growing in various non-conventional hydrophobic substrate types, especially industrial wastes. In this study, the content of thiamine (vitamin B1), riboflavin (vitamin B2), pyridoxine (vitamin B6), biotin (vitamin B7) and folic acid (vitamin B9) in the wet biomass of Y. lipolytica strains cultivated in biofuel waste (SK medium), compared to the standard laboratory YPD medium, was assessed. Additionally, the biomass of Y. lipolytica A-101 grown in biofuel waste (SK medium) was dried and examined for B vitamins concentration according to the recommended microbial methods by AOAC Official Methods. The mean values of these vitamins per 100 g of dry weight of Y. lipolytica grown in biofuel waste (SK medium) were as follows: thiamine 1.3 mg/100 g, riboflavin 5.3 mg/100 g, pyridoxine 4.9 mg/100 g, biotin 20.0 µg/100 g, and folic acid 249 µg/100 g. We have demonstrated that the dried biomass is a good source of B vitamins which can be used as nutraceuticals to supplement human diet, especially for people at risk of B vitamin deficiencies in developed countries. Moreover, the biodegradation of biofuel waste by Y. lipolytica is desired for environmental protection.  相似文献

A method is described whereby mutants having a directed increased yield of a metabolite may readily be isolated. A culture of bacterial cells is exposed to a gradient concentration of an antimetabolite that is inhibitory to the growth of the organism and mutants are isolated which survive because of an increased yield of the metabolite. This increased yield may be several times that formed by the parent culture. Techniques and results for increasing the yields of pyridoxine, nicotinic acid and thiamine are described.  相似文献

Batch fermentation studies were performed to evaluate the potentials of a complex nitrogen source, soybean, as an alternative to yeast extract for the economical production of lactic acid by Lactobacillus rhamnosus. An enzyme-hydrolysate of soybean meal, Soytone, with an adequate supplementation of vitamins was found to be highly effective in supporting lactic acid production from glucose and lactose. The effects of seven selected vitamins: d-biotin, pyridoxine, p-aminobenzoic acid, nicotinic acid, thiamine, pantothenic acid, and riboflavin, on cell growth and lactic acid production were investigated to provide the basis for the optimization of vitamin supplementation to minimize the cost. Pantothenic acid was the most required compound while the other six vitamins were also essential for high lactic acid productivity. As a result of the optimization, 15 g/l yeast extract could be successfully replaced with 19.3 g/l Soytone supplemented with the vitamins, resulting in a production of 125 g/l lactic acid from 150 g/l glucose. The volumetric productivity and lactate yield were 2.27 g/l/h and 92%, respectively, which were higher than those with 15 g/l yeast extract. The raw material cost was estimated to be 21.4 cent/kg lactic acid, which was only approximately 41% of that with yeast extract.  相似文献

The levels of seven water-soluble vitamins in Methanobacterium thermoautotrophicum, Methanococcus voltae, Escherichia coli, Bacillus subtilis, Pseudomonas fluorescens, and Bacteroides thetaiotaomicron were compared by using a vitamin-requiring Leuconostoc strain. Both methanogens contained levels of folic acid and pantothenic acid which were approximately two orders of magnitude lower than levels in the nonmethanogens. Methanobacterium thermoautotrophicum contained levels of thiamine, biotin, nicotinic acid, and pyridoxine which were approximately one order of magnitude lower than levels in the nonmethanogens. The thiamine level in Methanococcus voltae was approximately one order of magnitude lower than levels in the nonmethanogens. Only the levels of riboflavin (and nicotinic acid and pyridoxine in Methanococcus voltae) were approximately equal in the methanogens and nonmethanogens. Folic acid may have been present in extracts of methanogens merely as a precursor, by-product, or hydrolysis product of methanopterin.  相似文献

构建了共表达烟酸转磷酸核糖激酶(NAPRTase)和丙酮酸羧化酶(PYC)的重组质粒pTrc99a-pncB-pyc,并考察了重组菌E.coli NZN111/pTrc99a-pncB-pyc生产丁二酸的能力。结果表明:重组菌NZN111/pTrc99a-pncB-pyc的NAPRTase和PYC的比酶活达到最高,分别为20.75和1.04 U/mg,同时,辅酶NADH、NAD+及NAD(H)总量达到最高。厌氧摇瓶发酵结果:48 h能够消耗17.5 g/L的葡萄糖生成14.08 g/L的丁二酸,而丙酮酸的产量大幅度降低,仅为0.11 g/L。本研究为基因工程菌大肠杆菌厌氧条件下发酵生产丁二酸提供了一定的基础。  相似文献

Inhibition of rabbit skeletal muscle glycogen phosphorylase b by biotin, pyridoxine, lipoic acid, as well as by thiamine and cobalamine vitamins and coenzymes has been found. The values of "half-saturation" concentration and Hill coefficients are determined for biotin (27 mM, 1.3), pyridoxine (19 mM, 1.7), 5'-deoxyadenosyl-cobalamine (2.5 mM, 1.5), lipoic acid (3.4 mM, 1.1), thiamine (11 mM, 1.3), thiamine diphosphate (11 mM, 1.0). Effectiveness of the enzyme inhibition by vitamins and coenzymes containing different heterocyclic groups is analysed; riboflavin and its coenzymic forms are suggested to be the most effective inhibitors.  相似文献

研究了温度、培养基初始pH、碳源、氮源对被孢霉(Mortierella sp.)产生花生四烯酸的影响。正交试验结果表明,Mortierella sp.M10最佳培养基组成为(g/L):葡萄糖100,酵母膏10,KNO_3 4.0,KH_2PO_4 2.0,CaCl_2·2H_2O 0.1,MgSO_4·7H_2O 0.5,FeCl_3·6H_2O0.015,ZnSO_4·7H_2O 0.0075,CuSO_4·5H_2O 0.0005。采用最佳培养基及发酵条件,细胞干重和花生四烯酸产量分别为33.51g/L和0.827g/L。同时对摇瓶发酵过程进行了分析。  相似文献
