The potential of the MDK4‐20 promoter of Arabidopsis thaliana to direct effective transgenic expression of a secreted nematode‐repellent peptide was investigated. Its expression pattern was studied in both transgenic Arabidopsis and Solanum tuberosum (potato) plants. It directed root‐specific β‐glucuronidase expression in both species that was chiefly localized to cells of the root cap. Use of the fluorescent timer protein dsRED‐E5 established that the MDK4‐20 promoter remains active for longer than the commonly used constitutive promoter CaMV35S in separated potato root border cells. Transgenic Arabidopsis lines that expressed the nematode‐repellent peptide under the control of either AtMDK4‐20 or CaMV35S reduced the establishment of the beet cyst nematode Heterodera schachtii. The best line using the AtMDK4‐20 promoter displayed a level of resistance >80%, comparable to that of lines using the CaMV35S promoter. In transgenic potato plants, 94.9 ± 0.8% resistance to the potato cyst nematode Globodera pallida was achieved using the AtMDK4‐20 promoter, compared with 34.4 ± 8.4% resistance displayed by a line expressing the repellent peptide from the CaMV35S promoter. These results establish the potential of the AtMDK4‐20 promoter to limit expression of a repellent peptide whilst maintaining or even improving the efficacy of the cyst‐nematode defence.  相似文献

Savannas are highly diverse and dynamic environments that can shift to forest formations due to protection policies. Long‐distance dispersal may shape the genetic structure of these new closed forest formations. We analyzed eight microsatellite loci using a single‐time approach to understand contemporary pollen and effective seed dispersal of the tropical tree, Copaifera langsdorffii Desf. (Fabaceae), occurring in a Brazilian fire‐ and livestock‐protected savanna. We sampled all adult trees found within a 10.24 ha permanent plot, young trees within a subplot of 1.44 ha and open‐pollinated seeds. We detected a very high level of genetic diversity among the three generations in the studied plot. Parentage analysis revealed high pollen immigration rate (0.64) and a mean contemporary pollen dispersal distance of 74 m. In addition, half‐sib production was 1.8 times higher than full‐sibs in significant higher distances, indicating foraging activity preference for different trees at long distances. There was a significant and negative correlation between diameter at breast height (DBH) of the pollen donor with the number of seeds (r = ?0.640, P‐value = 0.032), suggesting that pollen donor trees with a higher DBH produce less seeds. The mean distance of realized seed dispersal (recruitment kernel) was 135 m due to the large home range dispersers (birds and mammals) in the area. The small magnitude of spatial genetic structure found in young trees may be a consequence of overlapping seed shadows and increased tree density. Our results show the positive side of closed canopy expansion, where animal activities regarding pollination and seed dispersal are extremely high.  相似文献

A common paradigm in studies of human cooperation is that individuals mediate their investment in a public good according to the investments of others. However, most research on this topic has been conducted in artificial laboratory settings and has assumed that cooperation imposes relatively high monetary costs on players. As such, we do not yet know how humans behave when faced with non‐monetary costs of cooperating in more realistic settings. Here, we present results from a real‐world public good experiment where the monetary costs of cooperation were relaxed. We manipulated the cleanliness of a sink in a staff tea room (a public good) by adding or removing dirty kitchenware and then recorded the number of items subsequently added to the sink by staff members. We found that when the sink had no dirty items, people were less likely to dump their own used kitchenware and the public good was maintained. In contrast, people were more likely to dump items in the sink when it already contained unwashed kitchenware and increasing the number of dirty items in the sink led to an increase in the number of items that were subsequently dumped. This study shows that defection begets defection in real‐life situations, even when cooperation is relatively cheap.  相似文献

The combination of high‐capacity anodes and high‐voltage cathodes has garnered a great deal of attention in the pursuit of high‐energy‐density lithium‐ion batteries. As a facile and scalable electrode‐architecture strategy to achieve this goal, a direct one‐pot decoration of high‐capacity silicon (Si) anode materials and of high‐voltage LiCoO2 (LCO) cathode materials is demonstrated with colloidal nanoparticles composed of electroconductive antimony‐doped tin oxide (ATO). The unusual ATO nanoparticle shells enhance electronic conduction in the LCO and Si electrode materials and mitigate unwanted interfacial side reactions between the electrode materials and liquid electrolytes. The ATO‐coated LCO materials (ATO‐LCO) enable the construction of a high‐mass‐loading cathode and suppress the dissolution of cobalt and the generation of by‐products during high‐voltage cycling. In addition, the ATO‐coated Si (ATO‐Si) anodes exhibit highly stable capacity retention upon cycling. Integration of the high‐voltage ATO‐LCO cathode and high‐capacity ATO‐Si anode into a full cell configuration brings unprecedented improvements in the volumetric energy density and in the cycling performance compared to a commercialized cell system composed of LCO/graphite.  相似文献

The design of a sodium‐ion rechargeable battery with an antimony anode, a Na3V2(PO4)3 cathode, and a low‐cost composite gel‐polymer electrolyte based on cross‐linked poly(methyl methacrylate) is reported. The application of an antimony anode, on replacement of the sodium metal that is commonly used in sodium‐ion half‐cells, reduces significantly the interfacial resistance and charge transfer resistance of a sodium‐ion battery, which enables a smaller polarization for a sodium‐ion full‐cell Sb/Na3V2(PO4)3 running at relatively high charge and discharge rates. The incorporation of the gel‐polymer electrolyte is beneficial to maintain stable interfaces between the electrolyte and the electrodes of the sodium‐ion battery at elevated temperature. When running at 60 °C, the sodium‐ion full‐cell Sb/Na3V2(PO4)3 with the gel‐polymer electrolyte exhibits superior cycling stability compared to a battery with the conventional liquid electrolyte.  相似文献

An investigation of intraspecific habitat‐related patterns of variation in oculoscapular lateral‐line superficial neuromasts (SN) identified a decrease in the ratio of total SNs to pores, and a trend towards decreased asymmetry in SNs in the habitat‐generalist common bully Gobiomorphus cotidianus from fluvial habitats compared to lacustrine habitats, suggesting habitat‐related phenotypic variability. A greater ratio of pores to SNs, as well as less variation in the total number and asymmetry of SNs observed in the fluvial habitat‐specialist redfin bully Gobiomorphus huttoni may provide further evidence of variations in the oculoscapular lateral‐line morphology of fluvial habitat G. cotidianus individuals serving as adaptations to more turbulent environments.  相似文献

The polyproteins of coronaviruses are cleaved by viral proteases into at least 15 nonstructural proteins (Nsps). Consisting of five domains, Nsp3 is the largest of these (180–210 kDa). Among these domains, the so‐called X‐domain is believed to act as ADP‐ribose‐1″‐phosphate phosphatase or to bind poly(ADP‐ribose). However, here we show that the X‐domain of Infectious Bronchitis Virus (strain Beaudette), a Group‐3 coronavirus, fails to bind ADP‐ribose. This is explained on the basis of the crystal structure of the protein, determined at two different pH values. For comparison, we also describe the crystal structure of the homologous X‐domain from Human Coronavirus 229E, a Group‐1 coronavirus, which does bind ADP‐ribose.  相似文献

Washing is a standard step for enzyme‐linked immunosorbent assays (ELISA) performed on a paper‐based chip, in which nonspecific‐binding antibodies and antigens should be removed completely from the paper surface. In this study, a novel three‐dimensional (3D) washing strategy using a heating ring‐oven was carried out on a paper‐based chip. Compared with a plane washing mode by a ring‐oven, this 3D washing strategy obtained a lower background, as gravity played an important role in the washing step. The paper‐based chip was placed on a 3D plastic holder and the waste area was connected to a heating ring. Use of a heating waste area meant that the nonspecific‐binding protein was continuously carried to the waste area through gravity and capillary action. The angle between the plastic holder and the ring plane was carefully selected. The effect of washing on different parts of the detection area was investigated by upconversion fluorescence and chemiluminescence (CL). This novel 3D washing strategy was performed for carcinoembryonic antigen detection through CL and a lower detection limit of 2 pg ml?1 was obtained. This approach provides an effective washing strategy to remove nonspecific‐binding antibody from a paper‐based immunodevice.  相似文献

Bicyclic dioxetanes 2a–c bearing a 2′‐alkoxy‐2‐hydroxy‐1,1′‐binaphthyl‐7‐yl moiety were effectively synthesized and their base‐induced chemiluminescent decomposition was investigated by the use of alkaline metal (Na+ and K+) or Mg2+ alkoxide in MeOH. When 2a–c were treated with tetrabutylammonium fluoride (TBAF) in dimethyl sulfoxide (DMSO) as a reference system, they showed chemiluminescence as a flash of orange light (maximum wavelength λmaxCL = 573–577 nm) with efficiency ΦCL = 6–8 × 10–2. On the other hand, for an alkaline metal (Na+ or K+) alkoxide/MeOH system, 2a–c decomposed slowly to emit a glow of chemiluminescence, the spectra of which were shifted slightly toward red from the TBAF/DMSO system, and ΦCL (= 1.4–2.3 × 10–3) was considerably decreased. In addition, Mg(OMe)2 was found to play a characteristic role as a base for the chemiluminescent decomposition of 2a–c through coordination to the intermediary oxidoaryl‐substituted dioxetanes 13. Thus, Mg2+ increased ΦCL to more than twice those with Na+ or K+, while it shifted λmaxCL considerably toward blue (λmaxCL = 550–566 nm). Copyright © 2012 John Wiley & Sons, Ltd.  相似文献

Abstract. Vegetation changes in a semi‐natural grassland of wooded meadow type that had been grazed for centuries are described following the introduction of various management regimes: mowing each year, mowing every third year, burning, mechanical removal of woody plants, chemical treatment of woody plants, continuous grazing and abandonment. The experiment was established in southern Sweden in 1972 and has been in progress for 15 years. In 1972, 1980 and 1986 the botanical composition in these plots was investigated in permanent subplots. The study clearly demonstrates that mowing or grazing is necessary to preserve community structure and that mowing is to be preferred in cases where maintaining species richness is of primary concern. Mowing every third year delayed vegetation change and prevented woody species from spreading. Therefore, periodic mowing might be an alternative way to preserve the flora. In contrast, yearly burning does not seem to be a viable management in this type of semi‐natural grassland. To preserve the open landscape regeneration of woody plants has to be prevented. However, in plots where woody plants were removed the typical grassland flora declined. Abandonment resulted in closed forest.  相似文献

Short, alpha‐helical coiled coils provide a simple, modular method to direct the assembly of proteins into higher order structures. We previously demonstrated that by genetically fusing de novo–designed coiled coils of the appropriate oligomerization state to a natural trimeric protein, we could direct the assembly of this protein into various geometrical cages. Here, we have extended this approach by appending a coiled coil designed to trimerize in response to binding divalent transition metal ions and thereby achieve metal ion‐dependent assembly of a tetrahedral protein cage. Ni2+, Co2+, Cu2+, and Zn2+ ions were evaluated, with Ni2+ proving the most effective at mediating protein assembly. Characterization of the assembled protein indicated that the metal ion–protein complex formed discrete globular structures of the diameter expected for a complex containing 12 copies of the protein monomer. Protein assembly could be reversed by removing metal ions with ethylenediaminetetraacetic acid or under mildly acidic conditions.  相似文献

This study is aimed at elucidating the structure of a novel T‐cell adhesion inhibitor, cyclo(1,8)‐CPRGGSVC using one‐ and two‐dimensional (2D) 1H NMR and molecular dynamics (MD) simulation. The peptide is derived from the sequence of its parent peptide cIBR (cyclo(1,12)‐PenPRGGSVLVTGC), which is a fragment of intercellular adhesion molecule‐1 (ICAM‐1). Our previous results show that the cyclo(1,8)‐CPRGGSVC peptide binds to the LFA‐1 I‐domain and inhibits heterotypic T‐cell adhesion, presumably by blocking the LFA‐1/ICAM‐1 interactions. The structure of the peptide was determined using NMR and MD simulation in aqueous solution. Our results indicate that the peptide adopts type‐I β‐turn conformation at the Pro2‐Arg3‐Gly4‐Gly5 (PRGG) sequence. The β‐turn structure at the PRGG motif is well conserved in cIBR peptide and ICAM‐1 receptor, which suggests the importance of the PRGG motif for the biological activity of cyclo(1,8)‐CPRGGSVC peptide. Meanwhile, the Gly5‐Ser6‐Val7‐Cys8‐Cys1 (GSVCC) sequence forms a “turn‐like” random coil structure that does not belong to any structured motif. Therefore, cyclo(1,8)‐CPRGGSVC peptide has only one structured region at the PRGG sequence, which may play an important role in the binding of the peptide to the LFA‐1 I‐domain. The conserved β‐turn conformation of the PRGG motif in ICAM‐1, cIBR, and cyclo(1,8)‐CPRGGSVC peptides can potentially be used to design peptidomimetics. © 2009 Wiley Periodicals, Inc.
