Acetazolamide produces a characteristic forelimb reduction deformity when administered to pregnant rodents. Past studies indicated that non-rodent species (rabbit and monkey) are resistant to this effect. The present studies confirmed this fact and demonstrated that transport of acetazolamide into the rabbit embryo was similar to that in sensitive rat embryos. In monkeys, however, the concentrations of acetazolamide within maternal plasma and embryo were much lower than in rats. Carbonic anhydrase activity was also measured since inhibition of this enzyme is the primary pharmacologic effect of acetazolamide. Again the rabbit embryo had carbonic anhydrase specific activity levels similar to that of the rat. Monkey embryos, on the other hand, contained negligible levels of enzyme activity during the presumed sensitive period of development. Thus the resistance of monkey embryos to acetazolamide teratogenesis may be due to low carbonic anhydrase activity and/or the small amount of drug reaching the embryo. No basis for the resistance of rabbit embryos to acetazolamide teratogenesis was uncovered.  相似文献

In this study, 198 donor mares of different breeds, ages, and reproductive category were inseminated with fresh, cooled and frozen or frozen and cooled semen at the embryo transfer station or in private artificial insemination centers during 10 breeding seasons. The results of this activity were retrospectively analyzed by Pearson Chi-square test and logistic regression to evaluate factors affecting multiple ovulations, embryo recovery, embryo quality, and embryo diameter. Out of the 661 cycles, 937 ovulations were recorded (mean ovulations/cycle: 1.42 ± 0.58). Ovulation rate and incidence of multiple ovulations were significantly affected by age, breed, and reproductive category. Uterine flushings for embryo recovery were performed between 7 and 10 days after ovulation and resulted in the recovery of 338 embryos (51.1% embryos/cycle and 36.1% embryos/ovulation, respectively). At least one embryo was recovered in 298 flushings (45.1%). The factors affecting embryo recovery were age, breed, reproductive category, type of semen, number of ovulations, and location of artificial insemination. Flushing protocol and day of flushing had no effect on embryo recovery. Age, type of semen, number of ovulations, and day of flushing had a significant influence on embryo diameter (N = 215). None of the factors included in the model had an effect on embryo quality distribution.  相似文献

Multiple ovulation and embryo transfer (MOET) has the potential to increase the rates of genetic improvement in sheep. However, better realization of this potential requires a higher yield of transferable embryos. Thus we investigated some factors that may contribute to high embryo yield and quality under field conditions, as part of an ongoing MOET program. Comparison of the effects of 2 breeding systems (natural service plus laparoscopic intrauterine AI vs natural service only) on embryo yield and quality indicated that while AI did not affect embryo recovery, it significantly (P < 0.05) improved fertilization rate and embryo quality, and increased (P < 0.05) embryo survival rate after transfer to recipients. Two flushing procedures (the original semi-laparoscopic and a modified version) were compared for effects on embryo recovery. The modifications made to the original collection method increased (P < 0.001) embryo recovery from 69.0 +/- 2.4 to 83.2 +/- 0.6%. The effects of the progestagen priming dosage during superovulatory treatment and ewe age on MOET outcome were also investigated. Donor ewes primed with 30-mg progestagen sponges came into estrus 1.9 h earlier (P < 0.05) than those primed with 45-mg sponges, but there was no difference in ovulation rate or embryo recovery, or in embryo survival after transfer between the 2 regimens. However, Chi-square analysis indicated a significant benefit in favor of the higher progesterone dose on both fertilization (P < 0.01) and embryo quality (P < 0.001). Age of donor ewe did not significantly affect the timing of estrus, fertilization rate or embryo survival after transfer. While adult ewes had higher (P < 0.05) ovulation rates and embryo yields, shearling ewes produced a much higher proportion of Grade 1 embryos (P < 0.05).  相似文献

Granulocytic and erythrocytic colonies developed on the chick embryo chorioallantoic membrane (CAM) following the inoculation of chick embryo spleen cells. Dose response and kinetic experiments showed that the colonies were derived from cell aggregates present in the inoculum. Dissociation and reaggregation studies of the CAM colony-inducing cells (CAM-CIC) indicated that these cells must be present as aggregates in order to form colonies. Results from the morphology and cell marker experiments suggested that the colony-inducing aggregates (CAM-CIA) attract and support the differentiation of primitive host hemopoietic cells. The physical characteristics of the CAM-CIC, which are different from those of the hemopoietic progenitor cells, indicated that they represent a stromal cell population of the chick embryo spleen. Further evidence supporting this notion was provided by the radiation studies which showed that the colony-inducing ability of the CAM-CIC is radioresistant. The above characteristics of the CAM-CIC strongly suggest that they represent the stromal cells of the chick embryo spleen which influence hemopoiesis.  相似文献

The objectives of this study were to examine the relationships between periovulatory endocrine events, ovarian activity, and embryo survival after artificial insemination (AI) in cattle (Bos taurus). Eighty-four reproductively normal beef heifers were estrus synchronized using a prostaglandin-based regimen. Artificial insemination was performed between 5 and 21 h after heat onset. Ultrasonic examination of ovarian structures began 12 h after the onset of heat and continued every 6 h until confirmed ovulation. Blood samples were collected for measurement of estradiol, progesterone, and insulin-like growth factor-1 (IGF-1). Pregnancy diagnosis was conducted on Days 30 and 100 after AI. Embryo survival was defined as the presence of an embryo with a detectable heartbeat in a clear amniotic sac at Day 30 postinsemination. There was no effect of the intervals from the onset of heat to AI or ovulation or from AI to ovulation on embryo survival (P > 0.10). There was a tendency (P = 0.09) of an inverse relationship between preovulatory follicle size and embryo survival that was unrelated to concentrations of estradiol or IGF-1 during the periovulatory period (P > 0.05). There was evidence (P = 0.08) of a positive association between embryo survival and concentrations of progesterone on Day 7; however, this relationship was independent (P < 0.05) of hormonal and follicular measurements during the periovulatory period. This study shows that heifers could be inseminated up to 31.5 h before ovulation without compromising the probability of embryo survival. This study suggests that there is an optimum range of follicle size within which high embryo survival rates can be achieved.  相似文献

Isocupressic acid (ICA) [15-hydroxylabda-8 (17), 13E-dien-19-oic acid], a labdane diterpene acid, isolated from ponderosa pine (Pinus ponderosa), Lodgepole pine (Pinus contorta), common juniper (Juniperus communis) and Monterey cypress (Cupressus macrocarpa), induces abortion in pregnant cows when ingested primarily during the last trimester. The objective of this study was to investigate the effects of isocupressic acid on bovine oocyte maturation (in vitro maturation (IVM)-Experiment I) and preimplantation embryo development (in vitro culture (IVC)-Experiment II) using in vitro embryo production techniques and to subsequently evaluate viability and developmental competence of ICA-cultured embryos via embryo transfer to recipient heifers (Experiment III). A complete randomized block experimental design was used. In Experiment I and II, isocupressic acid was added to IVM or IVC media at 0 (TRT1, control), 1.3 (TRT2), and 2.6 microg/ml (TRT3) Results from Experiment I and II indicated that ICA did not inhibit oocyte maturation and did not adversely affect preinpiantation embryo development. Furthermore, results from Experiment II demonstrated that isocupressic acid enhanced bovine preimplantation embryo development in vitro in a dose dependent manner. Subsequently, Day 8 (Day 0 = IVF) blastocysts cultured in vitro in the medium containing 2.6 microg/ml ICA were transferred to recipient heifers and resulted in normal pregnancies as determined by ultrasound imaging. Subsequently, all but two births were normal as evaluated by post natal veterinary examination. In conclusion, ICA showed no adverse effects on oocyte maturation and preimplantation embryo development in vitro or subsequent viability in vivo using the ICA concentrations and in vitro culture parameters of this study.  相似文献

Histochemical analyses of the ovule of Quercus gambelii show that the major food reserves (starch grains and lipids) are located almost exclusively within the outer integument. Vascular traces are present only within this integument which contains numerous, well-developed plasmodesmata. The inner integument is virtually devoid of any food reserves and has very few plasmodesmata. The ovule has a persistent chalazal extension of residual nucellar cells (called the postament) which projects into the embryo sac. Due to the above information and the fact that the synergids rarely contain starch and no plasmodesmata are present in the walls of any of the cells of the egg apparatus (Mogensen, 1972), it is concluded that the synergids play little or no role in embryo sac nutrition. Rather, it is proposed that the pathway of available food materials in the young ovule is from the outer integument to the chalaza and through the postament into the embryo sac.  相似文献

The anti-tumor agent cis-platinum(II) diamine dichloride caused dose-dependent toxicity in V79 Chinese hamster cells and in secondary Syrian hamster embryo cells. Chromosome aberrations were induced and positive dose--response relationships were observed for induction of sister-chromatid exchanges and 6-thioguanine-resistant mutations in V79 cells and morphologic transformation of secondary Syrian hamster embryo cells. Selection of transgenic embryos prior to embryo transfer is a means to increase the efficiency of transgenic livestock production. Among transgenic reporters, cytoplasmic expression of green fluorescent protein (GFP) has features that make it ideal for transgenic embryo selection. The primary objective of this study was to assess cytoplasmic expression of a specially designed GFP reporter as a tool for transgenic bovine embryo selection. A second objective was to evaluate this reporter for studying transgenic mosaicism related to timing of integration of pronuclear microinjected DNA. Transgenic embryos produced by pronuclear injection showed a discrete pattern of GFP expression with clusters at 25, 50, and 100% of blastomeres expressing GFP. This pattern of mosaicism is interpreted to indicate that the integration of microinjected DNA occurred, not only at the pronuclear stage, but also in the subsequent cell divisions. Among the GFP-positive transgenic embryos, only in 21% did all the blastomeres show the green fluorescence. Using the fraction of positive blastomeres within an embryo, the timing of integration of microinjected DNA was estimated. The frequency of nonmosaic embryos expressing GFP is consistent with published germline transmission success rates of transgenic cattle derived from pronuclear microinjected embryos. These results indicate the possible application of GFP as a marker of transgenic embryos and graphically illustrate underlying complexities in DNA integration in embryos subjected to pronuclear microinjection.  相似文献

Neither insulin nor epinephrine influenced the incorporation of glucose into the acid-soluble or acid-insoluble glycogen pool of mouse embryos at the morula-early blastocyst stage during 5 h culture in the presence of radiolabelled glucose. During a 5 h chase culture of pulse-labelled embryos at this stage of development, acid-soluble glycogen labelled during the pulse was not utilized by the embryo but acid-insoluble glycogen was reduced. Addition of glucagon, insulin, epinephrine, cAMP, theophylline or caffeine during chase culture had no effect on the turnover of label in the glycogen pools of the embryo. These results indicate that the turnover of embryonic glycogen observed in vivo is not due to the direct effect of the hormones that regulate glycogen metabolism in the mother. Insulin was found to stimulate incorporation of glucose into non-glycogen macromolecules during both pulse and chase culture. Thus, whilst an effect of insulin on glycogen metabolism was absent, the anabolic effects of this hormone appear to have been expressed in the embryo at this stage of development.  相似文献

The herbaceous vine Aristolochia contorta (Aristolochiaceae) is a rare plant with a fragmented area in East Asia. Confocal laser-scanning microscopy and stereomicroscopy were employed to examine the seed and embryo structure. This is the first research on embryo anatomy in Aristolochia. Shape and structure of this embryo may be phylogenetically significant. The seed structure of A. contorta is similar to that of other Aristolochia species. Considerable variation is found for productivity traits; the average seed set is rather low (26.7%). The studied seed and embryo traits of A. contorta are found to be variable, but may be potentially informative at the genus and species level. The restricted productivity and reduced population size threaten the continued survival of A. contorta and, perhaps, the butterfly that depends on it.  相似文献

The aim of this work was to study morphological and biochemical aspects during zygotic embryogenesis in O. catharinensis, by measuring changes in the endogenous concentrations of proteins, amino acids, polyamines (PAs), indole-3-acetic acid (IAA) and abscisic acid (ABA). Buffer-soluble and insoluble protein contents were determined by spectrometry, and amino acids, PAs, IAA and ABA concentrations were determined by high performance liquid chromatography. Total amino acid accumulation, predominantly asparagine, occurred when the embryo showed completely developed cotyledons, with posterior reduction in the mature embryo. This decrease in total amino acid concentration in the mature embryo may result from their use in storage␣as well as for LEA protein synthesis. Free putrescine (Put) concentration decreased, while free spermine (Spm) increased during embryo development. This suggest a role for Put in the initial phases of embryogenesis when high rates of cell division occur, while elevated concentration of Spm are essential from the middle to the end of embryo development, when growth is mainly due to cell elongation. An IAA peak in zygotic embryos occurred during initial development, suggesting a link between growth and cellular division as well as with the establishment of bilateral symmetry. ABA concentration declined during initial stages of development then increased at the mature embryo stage, suggesting a possible relationship with dormancy and recalcitrance characteristics. Our results show that changes in the phytohormones (IAA, ABA and PAs) concentrations in combination with amino acids are likely important factors determining the developmental stages of O.␣catharinensis zygotic embryos.  相似文献

A recessive EMS-induced mutation inherited in Mendelian fashion caused monocotyledonous embryo formation and seed germination on high salt medium inCatharanthus roseus. Availability during embryo development of exogenously supplied cytokinin kinetin suppressed the mutant phenotype. These observations suggest that, inC. roseus, (i) insufficiency in endogenous kinetin may lead to monocotyledonous embryo patterning and (ii) dicotyledonous embryo formation requires a critical amount of kinetin in certain cells of early embryos.  相似文献

UDP-Glucuronyltransferase (GT) activity increases in chick embryo liver during culture from zero to a steady-state level at or above adult values. The GT activity (o-aminophenol as acceptor) is located entirely in the membranes of the endoplasmic reticulum (ER) and the question arises whether ER increases along with GT. Earlier work showed that the synthesis and degradation rates of GT can be varied in culture over wide ranges by choosing embryo livers of different ages and both phenobarbital. In the present study we measured the GT activities and the concentrations of ER (using stereologic methods) in 5- and 11-day embryo liver during culture with and without phenobarbital. We found that GT and ER always increased in a constant ratio of 2.2 X 10(-9) U of GR activity per square micrometer of membrane, suggesting that the synthesis and degradation of GT are coupled to the synthesis and degradation of ER. A general structure for ER is proposed to explain this finding.  相似文献

Early pregnancy in the mare is a poorly understood, high risk period during which the embryo communicates its presence to the maternal endometrium. Remarkably, the maternal recognition of pregnancy signal is unknown in the horse. This study aimed to profile the proteins secreted by equine blastocysts into their immediate environment, along with proteins contained in the blastocoel and within the acellular embryo capsule. Embryos were recovered on day 8 after ovulation and cultured for 48 hours. Secretomes of day 9 and day 10 embryos were analyzed by LC‐MS/MS and supported by analysis of blastocoel fluid and embryo capsule. Analyses revealed 72 (24 h) and 97 (48 h) unique protein IDs in the embryo secretome, 732 protein IDs in blastocoel fluid, and 11 proteins IDs in the embryo capsule. Novel findings of interest include secretion of a pregnancy specific proteinase (PAG) by the equine embryo at day 10, along with detection of a prostaglandin receptor inhibiting protein (PTGFRN) and a progesterone potentiating factor (FKBP4) in blastocoel fluid. This is the first comprehensive proteomic analysis of the equine embryo secretome, and provides new insights into the unique physiology of early pregnancy in this species.  相似文献

Immature fruits of Aesculus yield powerful stimuli to growthand cell division. Therefore, the developing fruit of Aesculuswoerlitzensis Koehne has been investigated from pollinationto maturity. The fluid, or liquid endosperm, which containsthe growth-promoting substances is produced in a large vesiclewhich forms at the chalazal tip of the embryo sac. As the vesiclegrows, it encroaches upon the nucellus and, when the embryodevelops, one of its cotyledons penetrates into the vesicleof the embryo sac where it grows and absorbs the contents. Theembryo, which has only a vestigial suspensor, reaches the vesicleby growing along the neck of the long curved embryo sac. Thecotyledon which first penetrates the vesicle grows into a massivestructure; the other remains small. The tip of the cotyledonseems to function as an absorbing surface, for the endospermwith which it comes into contact disorganizes. Fertilizationand the presence of a viable embryo at the micropylar end ofthe embryo sac therefore sets in train a number of other events.These are the extensive development of the nucellus at the chalazalend of the embryo sac, the swelling of the vesicle and the formationof a free nuclear and some cellular endosperm, and the disorganizationof the nucellus as it is encroached upon by the vesiculate embryosac. Attention is directed to the organization of the nucellusin the vicinity of the embryo sac. Files or richly protoplasmicnucellar cells(hypostase) which converge upon the chalazal tipof the embryo sac suggest a principal route by which the vesiclemay be nourished. Special attention is drawn to the very differentsizes of cells, their nuclei and nucleoli, in the differentparts of the nucellus. The growth and development of the embryohas also been traced from the zygote to the mature seed. Thenutritive role of the veaiculate embryo sac, and the supplyof growthstimulating substances, through the function of a cotyledonas an absorbing organ, are now seen as important features ofthe development of the Aesulus embryo in the ovule. Many outstandingproblems still remain. The sequential events that follow fertilizationin the different interdependent regions (nucellus, embryo sac,cotyledon, &c.) are here described, but not casually explained.  相似文献

There is little information on the fate of embryo sacs in plantovules if pollination is prevented. In this study embryo sacsfrom watermelon were observed over a 13 day period followingflowering with (a) normal pollination, (b) non-pollination and(c) induction of parthenocarpic fruit development with naphthaleneacetic acid. Following pollination, and prior to fertilizationapproximately 2 days later, the embryo sacs completed developmentand consisted of two synergids with prominent filiform apparatus,an egg cell, a central cell with two polar nuclei and threeantipodal cells. Sperm nuclei were observed within the embryosac at 2 days and by 4 days the endosperm was proliferating.In the non-pollination treatment the embryo sac was still intactafter 4 days although the antipodal nuclei were becoming hardto distinguish. By 7 days only the two synergids and the eggcell were still well defined, the polar nuclei appeared in somepreparations to be fused, and the antipodals had degenerated.By 10 days the embryo sac was a structure-less watery mass.In parthenocarpic fruit the fate of the embryo sac was similarto that in non-pollinated fruit except that final breakdownwas delayed past 10 days. Maturity of the majority of embryo sacs in an ovary appearedto be contemporaneous with penetration of the pollen tube, andon the basis of the anatomical results it seems possible thatembryo sacs could be fertilized up to 2 days beyond the normaltime.
