An attempt was done to identify some factors influencing the caecal fermentation pattern in poultry. Experiments were conducted to study effects of carbohydrate substrates (feed components and supplements) and antibiotics on the formation of volatile fatty acids (VFA) and methane in in vitro incubations of the caecal contents of 7-week-old chickens. Stoichiometry of fermentation differed in cultures with different carbohydrates. Fermentation pattern characterized by high propionate and low acetate production was found in cultures with lactose (0.447 and 0.376 mol/1 mol of VFA produced, respectively) and, to a lesser extent, also in cultures with raffinose. Acetate was the predominant metabolite of starch, pectin and xylan (0.727, 0.773 and 0.685 mol/1 mol of VFA produced, respectively). Fermentation of inulin resulted in high proportion of butyrate, 0.221 mol/1 mol of VFA. Other polysaccharides produced only 0.060–0.111 mol of butyrate per 1 mol of VFA. Oligosaccharides (lactose, raffinose) were fermented more rapidly than polysaccharides. Fermentation of inulin yielded more VFA than fermentation of starch, pectin and xylan. No production of VFA from carboxymethylcellulose was observed. On average, 11 mols of VFA were produced per mol of methane. Lasalocid significantly increased molar proportion of propionate, which is potentially beneficial from the point of view of salmonellae control. The magnitude of improvement, however, was small. Other feed antibiotics tested (avoparcin, bacitracin, lincomycin, spiramycin, tylosin, virginiamycin) produced only non-significant or marginal fermentation shifts. Formation of valerate, isoacids and methane was not significantly influenced by the substrate or by antibiotic treatment.  相似文献

A Rusitec semi-continuous flow fermentor was used to study the influence of enzyme pretreatment of food substrates on the fermentation profile over a 2-week period following inoculation with rabbit caecal contents. Three types of substrate were examined: 1) homogenized commercial rabbit feed; 2) the solid remains of this feed after digestion with alpha-amylase for 24 h; and 3) substrate 2 digested for 4 h with pepsin (double enzyme treatment). One of a pair of nylon pouches containing 15 g substrate was replaced each day, thus producing a uniform 48-h fermentation. Fermentation of the untreated feed (1) for 5-6 days produced a fermentation profile quite different from that obtained in vivo in the rabbit caecum: propionic acid accounted for over 35% of total volatile fatty acid (VFA), and butyric acid for about 15%. Amylase digestion (2) gave a stable ferment profile closer to the in vivo profile, except that propionic and butyric acids were similar at 15% of total VFA. Digestion with both amylase and pepsin (3) produced a stable fermentation profile very close to the in vivo profile: C2 > 60%, C3 < 11% and 17% < C4 < 21%. The rate at which membrane constituents (acid detergent fibre, ADF) were lost in 48 h was similar to the digestibility coefficient measured in vivo by others for the same basic feed. Lastly, there was a high percentage (about 5%) of volatile C5 fatty acids; this could be due to the discontinuous fermentor input of one pouch per 24 h. Thus, feed pretreated with both amylase and pepsin simulates, in vitro, rabbit caecal fermentation in a semi-continuous Rusitec type fermentor.  相似文献

Fermentation pattern and yields of microbial protein were investigated in cultures of the rabbit caecal contents supplied with glucose, xylose, starch, pectin and xylan. Rabbits at the age of 4 weeks (before weaning) and 3 months were slaughtered, their caecal contents added at 1.1% to growth media and incubated anaerobically at 39°C for 18 h. Caecal microorganisms of 4‐week‐old rabbits produced no methane and caproate, less butyrate, but more propionate than microorganisms of 3‐month‐old rabbits. In both groups of rabbits, fermentation of xylose produced significantly more propionate and less butyrate than fermentation of glucose. More propionate and less acetate was formed from starch than from pectin. In caecal cultures from 4‐week‐old rabbits with pectin, the molar percentages of acetate was significantly higher and percentages of other short‐chain fatty acids (SCFA) lower than in cultures with starch or xylan. In cultures from 3‐month‐old rabbits, fermentation of pectin and xylan produced similar SCFA profiles, different from SCFA molar composition in cultures with starch. Average production of microbial protein was 129mg per lg of carbohydrate digested (range 110 to 141mg/g). Protein yields were the same on glucose and xylose, but nonsignificantly higher on starch than on pectin and xylan. It can be concluded that the characteristics of substrate affected fermentation pattern in mixed cultures of rabbit caecal microorganisms. Substrate effects on protein yields were not statistically significant, due to high variation.  相似文献

We compared the effect of diets containing different nondigestible carbohydrates: cellulose (C), inulin (IN) and carboxymethylcellulose (CMC) as single supplements or in dietary combination on caecal physiology of rats. Sixty male Wistar rats (Rattus norvegicus) were divided into five groups and for 4 weeks were fed a casein diet with the compared carbohydrates (4% of diet) or a combination of IN+C or IN+CMC (both 4+4%). Diet intake and FCR index remained unaffected by the treatments, whereas IN improved the body weight gain of rats compared to CMC. Compared to C group, all diets containing IN and CMC decreased the caecal pH as well as enlarged the caecum, thus increasing the weights of contents and tissue, especially upon CMC treatment. Rats given carboxymethylcellulose (CMC and IN+CMC groups) had watery caecal digesta, and some of them suffered from diarrhoea. In the case of CMC, the caecal enlargement was due to tissue hypertrophy and digesta accumulation mostly in response to an increased bulk of contents. Unlike C+IN, the dietary combination of CMC- and inulin-enhanced fermentation in the caecum of rats, however the proportion of acetate, propionate and butyrate was less beneficial. Compared to CMC, inulin gave a higher concentration of SCFA, especially of butyrate and propionate. The action of inulin in the caecum of rats could be pronounced by dietary treatment combined with CMC.  相似文献

A model experiment was performed on rats to evaluate the effect of partial or total substitution of saccharose (S) and cellulose (C) by preparations of lactulose and inulin on the development and metabolism of the caecum. In the experimental diets given to rats for 4 weeks, the examined preparations were administered either with an equivalent amount of cellulose (each at 4% of the diet) or as sole source of dietary fibre at 8% of the diet. Compared to the saccharose group cellulose had no effect, and low doses of lactulose and inulin in the diet increased to a medium extent the weight of the caecum wall and caecal digesta. The addition of lactulose and inulin at 8% increased significantly the content of caecal digesta (4.62 and 4.11 g/100g BW, respectively) and the weight of the caecal wall (1.10 and 0.86 g/100g BW, respectively), compared to the groups with saccharose and cellulose (0.73, 0.90 and 0.24, 0.28 g/100g BW, respectively). Cellulose and cellulose partially-substituted with lactulose and inulin caused an increase in the dry matter content of caecal digesta (26.5-27.5%), compared to other groups (21.8-22.8%). The administration of lactulose and inulin preparations was accompanied by a significant drop in pH (5.47-5.81), compared to the groups with cellulose or saccharose (6.83-6.91), and a decrease in the ammonia concentration in the caecal digesta, compared to the cellulose control (0.27-0.40 and 0.62 mg/g, respectively). The group with 8% lactulose was characterized by the highest activities of microbiological alpha- and beta-galactosidase and beta-glucosidase in the caecal digesta. Cellulose and both preparations significantly decreased the activity of beta-glucuronidase, compared to the saccharose group (0.39-0.89 and 1.52 U/g, respectively). The highest concentration of VFA in the caecal digesta was observed in the saccharose group (89.2 micromol/g), and the lowest concentration in the group where cellulose was totally substituted by lactulose and inulin (55.1 and 57.5 micromol/g, respectively). The total production of VFA in the caecum was fourfold higher with 8 % lactulose and inulin (254.7 and 236.4 micromol/100g BW, respectively) than in both controls groups (65.1 and 67.8 micromol/100g BW, respectively). The high dose of inulin and lactulose increased the share of propionic acid in the VFA profile (C2:C3:C4) compared to both control groups. When 4% inulin was added to the diet a significant increase of butyrate concentration in the caecum was observed.  相似文献

No interactions were noted between the contents of antibiotic and MOS in the diet in any of the parameters examined. Depending on dietary dose, MOS had a different influence on caecal digesta parameters. The medium level of dietary MOS (0.25%) resulted in the highest caecal pH, dry matter and protein concentrations as well as the bacterial glycolytic activity (including beta-glucuronidase). Compared to other dietary treatments, the highest amount of MOS (0.5%) reduced ammonia concentration and enhanced volatile fatty acids concentration, especially of acetate and butyrate, in the caecal digesta. The medium level of dietary MOS caused a significant enhancement of propionate, iso-butyrate and iso-valerate concentrations in the digesta. Volatile Fatty Acids (VFA) production by anaerobic fermentation of organic solid wastes was studied at laboratory scale. The influence of initial substrate concentration was evaluated on VFA production. Completely mixed reactors (0.9?l) were used at mesophilic temperature (35?°C). Food wastes had 43.8% Total Solids content. Three dilutions of substrate (1/25, 1/10 and 1/5) corresponding to 1.75%, 4.38% and 8.76% of Total Solids and five values of Organic Loading Rates: 2, 5, 10, 12.5 and 25?kg COD/m3?d were studied. It was found that substrate 1/10 led to 14?g VFA/l at a loading rate of 12.5?kg COD/m3?d and an hydraulic retention time of 3.7 d. The main VFA produced were especially acetate and butyrate. Substrate diluted 1/5 led to 26.1?g VFA/l at a loading of 5?kg COD/m3?d and an hydraulic retention time of 15.1 d, but biomass production was not optimal. In a second study, a cascade of three reactors was used. An effluent with 42?g VFA/l was obtained at steady-state conditions at a loading of 12.5?kg of COD/m3?d and an hydraulic retention time of 12.5?d. The distribution of VFA was the following: 36% of propionate, 34% of acetate and 22.5% of butyrate.  相似文献

Two Rusitec fermentors were operated under identical conditions. One was seeded with an inoculum of rabbit caecal contents, and the other with bovine rumen contents. The fermentation substrate was rabbit feed that had been digested with amylase and pepsin. The substrate constituents (organic matter, OM and NDF) were lost in 48 h at a significantly higher rate in the presence of rumen inoculum (OM: +10%, NDF: +15%). The pHs of the 2 fermentors were similar at pH 6.6. The fermentors produced similar amounts of protein nitrogen per 24 h, after 6 d of adaptation. Volatile fatty acid production was slightly higher in the presence of rumen inoculum. The fermentor inoculated with rumen contents produced a higher percentage of propionic acid (25%) than of butyric acid (7%), while fermentation with rabbit caecal contents gave the opposite ratio (C3/C4 = 0.81). Consequently, only the rabbit caecal inoculum provided the fermentation profile characteristic of the species.  相似文献

The brown hare and the domestic rabbit are mid-sized herbivorous mammals and hindgut fermenters, though their digestive physiologies differ in some traits. The objective of this study was to estimate and compare the caecal microbial activity in hares and rabbits via an analysis of the following end-products of in vitro caecal fermentation: methane, total gas production, short chain fatty acids and ammonia concentration. Hare caecal methanogenesis occurred at a much lower level (0.25 mmol/kg for samples incubated without substrate and 0.22 mmol/kg for samples incubated with substrate) than that of the rabbit (15.49 and 11.73 mmol/kg, respectively) (P<0.001). The impact of the substrate’s presence on caecal methanogenesis was not significant, though its presence increased the total gas production during fermentation (P<0.001). Hare caecal microflora produced a lower short chain fatty acids concentration than did rabbit microorganisms (P<0.05). In unincubated hare samples, the short chain fatty acids concentration was 28.4 mmol/kg, whereas in unincubated rabbit samples, the short chain fatty acids concentration was 51.8 mmol/kg. The caecal fermentation pattern of the hare was characterised by higher propionate and isobutyrate molar proportions compared with those observed in rabbit caecum (P<0.01). No significant changes in the ammonia concentration in either rabbit or hare caecum were found. The results obtained indicate some differences in the activity of the microbial populations colonising the hare and rabbit caecum, particularly in regards to methanogenic Archaea.  相似文献

The caecal fermentation pattern was studied in four litters of rabbits. Rabbits were sequentially slaughtered at the age of 4 (before weaning), 6, 8 and 11 weeks. Their caecal contents were analyzed and incubated in vitro at 39°C for 6 and 12h. Net productions of short‐chain fatty acids (SCFA), hydrogen and methane were determined. The average caecal weight increased three‐times within two weeks after weaning, from 31.4 to 93.7g. At the end of the experiment, the caecal weight was on average 134.4g. A large variability of the SCFA concentration observed before weaning decreased after weaning. Measurements of caecal metabolite profiles and results of in vitro experiments indicated a certain decrease of propionate in favour of butyrate, associated with the weaning. The establishment of methanogens in rabbits was slow. Methanogenesis started in one out of four rabbits at the age of 6 weeks. Five weeks later, one of four rabbits still did not produce methane. The hydrogen recovery decreased between the 4th and the 6th week of age, due to the increase of the butyrate/propionate ratio. After the 6th week, the hydrogen recovery increased with age, apparently because of the increase in methane production. Hydrogen recovery tended to increase during incubation, suggesting a decrease of reductive acetogenesis. This increase was observed both in methanogenic and non‐methanogenic rabbit caecal cultures. In former cultures, the ratio CH4/SCFA rose with time of incubation.  相似文献

The goal of this research was to study the effect of the substitution of wheat starch by potato starch (PS) on the performance, health and digestion of growing rabbits. Three experimental diets were formulated with 0%, 7% and 14% PS (PS0, PS7 and PS14, respectively) and similar starch contents (22% dry matter basis), proteins and fibre. The three diets were administered to three groups of 48 rabbits from weaning (28 days) to slaughter (70 days), and growth and health measurements were made. Another 10 rabbits per diet (30 rabbits at each age), reared under similar conditions, were slaughtered at 6 to 10 weeks of age, and the digesta were collected to analyse the caecal microbial activity (pH, volatile fatty acids (VFA) levels, fibrolytic activity) and the starch concentration in the ileal digesta. At the same ages, the whole tract digestibility coefficients were measured in 10 other rabbits for each treatment (30 rabbits). The feed intake between 28 and 42 days of age (days) increased by 11% (P < 0.05) in PS0 v. PS14. Over the whole growth period (28 to 70 days), weight gain was similar among diets (40.5 g/day), whereas the feed intake and feed conversion increased (8.5% and 5.2%, respectively; P < 0.05) with the PS14 diet. Mortality and morbidity were not affected by the diets. The starch concentration of the ileal contents increased (P < 0.01) with the addition of PS to the diet (0.39%, 0.77% and 1.08% for diets PS0, PS7 and PS14, respectively). Starch digestibility was 0.8 percentage units higher (99.8% v. 99.0%) with the PS0 diet than the PS14 diet (P = 0.04). The bacterial cellulolytic activity in the caecum tended to be higher with the PS14 diet (P = 0.07). The total VFA caecal concentration increased (P < 0.01) only in 6-week-old rabbits with PS7 compared with PS0 (54.7 v. 74.5 mmol/l). Protein digestibility and ileal starch concentration decreased (P < 0.05) with age (6 v. 10 weeks), and hemicelluloses digestibility increased (P < 0.05). At 10 weeks of age, rabbits showed a higher VFA pool (6.25 mol) and proportion of butyrate (15.9%) and a lower proportion of acetate (79.3%), ammonia level (7.5 mmol/l) and C3/C4 ratio (0.31) than at 6 weeks of age. The intake of potato starch had no effect on the performance, caecal microbial activity or digestive health of growing rabbits.  相似文献

The products of the microbial activity in the large intestine are an important source of energy for herbivores. A previous study showed differences in caecal methanogenesis and short-chain fatty acid (SCFA) profiles between brown hares and domestic rabbits. The present study was performed on animals which were offered the same diet to eliminate the impact of diet on the comparative analyses of microbial metabolites. Caecal samples of hares and rabbits were incubated in triplicate, i.e. without any supplementary substrate (control) or with the addition of wheat bran or oat bran. Calculated as percentage of body weight, the stomachs of the rabbits were heavier than those of the hares, but caeca of the hares weighed more than those of the rabbits. The total SCFA concentration in caecal samples was higher in rabbits than in hares, and it increased in both species when the supplementary fermentation substrates were added. In hares, the molar proportion of propionate was higher and that of butyrate was lower compared to rabbits. The addition of substrate decreased acetate and propionate but increased the molar proportion of butyrate. Microbial fermentation resulted in greater gas release in rabbit caecal samples compared to those of hares. Methanogenesis tended to be lower in hares than in rabbits, but high individual variability was observed, especially in hares. Our study stated lower microbial activity in the caeca of brown hares compared to domestic rabbits. The presented results might lead to assumption that differences between fermentation patterns were not caused by diet but resulted from the peculiarities of both species.  相似文献

A total of 200 weaned (35 days) hybrid Hyla rabbits were randomly divided among five groups housed in bicellular cages (20 cages per group). Between 35 and 60 days of age, the groups were submitted to the following treatments: group ANT (positive control) fed a basal diet supplemented with antibiotics (colistin sulphate, 144 mg/kg; tylosin, 100 mg/kg; and oxytetracyclin, 1000 mg/kg); groups MOS_0.5, MOS_1.0 and MOS_1.5 fed the basal diet supplemented with 0.5, 1.0 and 1.5 g/kg mannanoligosaccharides (MOS), respectively; another group fed the basal diet without antibiotics or mannanoligosaccarides supplementation (negative control). Along the trial, an episode of epizootyc rabbit enteropathy occurs so that in the control group mortality rate was very high (78%) and survivor rabbits showed severe symptoms of disease (diarrhoea). Thus, the control group was discarded from the trial. At 60 days of age, samples of caecal content were collected from 10 rabbits per group and used as inocula for an in vitro gas production trial. At the end of fermentation (120 h of incubation), organic matter digestibility (OMd), cumulative gas production, fermentation kinetics, pH, volatile fatty acid (VFA) and NH3 productions were measured. Inoculum from MOS_1.0 rabbits showed the significant higher values of OMd (64.21%, P < 0.05), gas production (262.32 ml/g, P < 0.05), acetate (96.99 mmol/g OM, P < 0.05) and butyrate (26.21 mmol/g OM, P < 0.05) than the other groups. Slight differences were recorded among the groups ANT, MOS_0.5 and MOS_1.5. In addition, branched chain acids, in proportion to total VFAs, were significantly higher in MOS_1.0 inoculum (0.04, P < 0.05). Korean food wastes were anaerobically digested to produce volatile fatty acids (VFA) that can be used as a carbon source in biological nutrient removal in a sequential batch reactor (SBR). Acetate, propionate and butyrate were produced at a yield of 379-400 g VFA/kg VS0 (initial volatile solids). The ratio of SCOD (Soluble Chemical Oxygen Demand) of VFA to ammonia nitrogen (N) was in the range of 36.2-36.5 and the ratio of SCOD to phosphorus was between 151 and 162. The removal rate of nitrogen and phosphorus improved from 44% and 37% to 92% and 73%, respectively when the VFA were added to the influent of the Taejon municipal wastewater plant. The concentration of nitrogen and phosphorus were maintained below 3 mg/l and 1 mg/l, respectively. The N- and P-content of the food waste was low enough not to influence the final N- and P-concentrations of the wastewater.  相似文献

Twenty-six Alpine and Saanen goats, 12 fitted with a rumen cannula and a T-type cannula in the duodenum, were used in a 6-week experiment on the effect of a monoterpene blend on rumen fermentation, duodenal terpene and nutrient flows, milk yield and composition. The monoterpene blend consisted of 45.2, 36.7, 16.0 and 2.2 mol/100 mol terpene for linalool, p-cymene, α-pinene and β-pinene, respectively. The four compounds were considered as good models of northwest Mediterranean sward terpenes and were supplied in the same proportions as in the spring diet of dairy goats in the Basilicata (Southern Apennine range, Italy). The goats were assigned to three experimental groups fed a total mixed ration with no terpene or with the monoterpene blend at two levels, 0.043 and 0.43 g/kg dry matter intake. Ruminal fermentation characteristics remained unchanged among groups. In average, the total volatile fatty acids (VFA) concentration was 89.0 mM and the proportions of acetate, propionate and butyrate were 66.4, 21.1 and 10.1 mol/100 mol VFA, with an acetate:propionate ratio of 3.19. In average, 0.0, 2.7, 3.5 and 23.5 mol/100 mol of the ingested amounts of linalool, α-pinene, p-cymene and β-pinene were recovered in the duodenum. The mean terpene composition of duodenal digesta was 11.7, 9.2 and 79.1 mol/100 mol terpene for p-cymene, α-pinene and β-pinene, differing markedly from the dietary blend in both supplemented groups. Apparent rumen digestibility of dry matter (0.40), of neutral detergent fibre (0.54), and of acid detergent fibre (0.45) was not affected by either level of terpene supplementation. Dry matter intake and milk production, milk fat content or milk fatty acid profile were not affected by terpenes. It is concluded that, because of extensive ruminal degradation of each terpene, the monoterpene blend had no effect on ruminal digestion of dietary constituents or on milk performance whatever the supplementation level.  相似文献

Wistar rats fed diets containing different contents of cellulose or 5% transgalactosylated oligosaccharides (TOS) for 7 weeks were examined for faecal microflora and caecal short-chain fatty acids. High (15%) cellulose diet showed remarkable increases in concentrations of faecal bifidobacteria and caecal butyrate with a reduced concentration of propionate as compared with the other diets. TOS-fed rats showed strikingly increased concentrations of bifidobacteria and acetate. A comparison of pH values between the caecal contents and faeces suggested more fermentation of cellulose in the distal than proximal part of the large bowel of rats.  相似文献
