The instrumentation of a precise yet flexible system for the formation and fractionation of linear density gradients is described. The use of a special peristaltic pump and gradient-forming chambers permits the rapid formation of both dilute and concentrated solutions into ca. 5 to 300 ml of gradient. This gradient can be delivered to a single centrifuge tube or equally apportioned to as many as 24 tubes.The special peristaltic pump, which is described in the paper that follows, delivers the concentrated solution into the gradient-forming chamber as well as conveying the gradient from the chamber to the special centrifuge tube. A capillary in the wall of the tube conveys the gradient to the bottom, where it is dispersed into 18 equidistant orifices. This dispersion permits the rapid formation and fractionation of the gradient without disruption. A presettable counter coupled to the peristaltic pump permits the gradients to be apportioned into equal volume fractions.Results obtained with both dilute and concentrated gradients are presented.  相似文献

A temperature gradient chamber (TGC) is described which enables elevated CO2 concentrations and a dynamic temperature gradient to be imposed on field crops throughout their life cycle under standard husbandry. Air is circulated through two double-walled polyethylene-covered tunnels connected to a split heat pump system to give a near-linear temperature gradient along each tunnel. Solar energy gain along each tunnel and exchange with outer tunnel air flow contribute to the temperature gradient and also produce diurnal and seasonal temperature fluctuations corresponding to ambient conditions. Mean temperature gradients of between 3 and 5°C have been recorded throughout the growing seasons of crops of lettuce, carrot, cauliflower and winter wheat. Elevated or present CO2 concentrations are maintained in each of two pairs of tunnels throughout the cropping season using pure CO2 injected through motorized needle valves. This system can realistically simulate aspects of the effects of projected future environmental change on crop growth, development and yield, and in particular tin-possible interaction of the effects of increased CO2 and temperature.  相似文献

A method for the fractionation of centrifuged density gradients is described. A piston forced into the centrifuge tube from above displaces the gradient, while its tip collects liquid from the small volume immediately below it without disturbing the unfractionated part of the gradient. The centrifuge tube holder permits sensitive visual detection of bands of light scattering particles which may then be rapidly and precisely extracted. Continuous fractionation of the entire gradient is also possible.  相似文献

The origin of antigen-binding (rosette-forming) and antibody-forming (plaque-forming) cells in mouse spleen is determined with the use of anti-thymus (anti-theta) and anti-bone marrow (anti-beta) sera. Evidence is presented that anti-theta serum is specific for thymus and thymus-derived cells. Anti-beta serum acts only on bone marrow and bone marrow-derived cells and does not affect the viability or functional activity of thymus cells. The cytotoxic activity of anti-theta and anti-beta serum does not overlap.  相似文献

A theoretical model of the gramicidin A channel is presented and the kinetic behavior of the model is derived and compared with previous experimental results. The major assumption of the model is that the only interaction between ions in a multiply-occupied channel is electrostatic. The electrostatic calculations indicate in a multiply-occupied channel is electrostatic. The electrostatic calculations indicate that there will be potential wells at each end of the channel and, at high concentrations, that both wells can be occupied. The kinetics are based on two reaction steps: movement of the ion from the bulk solution to the well and movement between the two wells. The kinetics for this reaction rate approach are identical to those based on the Nernst-Planck equation in the limit where the movement between the two wells is rate limiting. The experimental results for sodium and potassium are consistent with a maximum of two ions per channel. To explain the thallium results it is necessary to allow three ions per channel. It is shown that this case is compatible with the electrostatic calculations if the presence of an anion is included. The theoretical kinetics are in reasonable quantitative agreement with the following experimental measurements: single channel conductance of sodium, potassium, and thallium; bi-ionic potential and permeability ratio between sodium-potassium and potassium-thallium; the limiting conductance of potassium and thallium at high applied voltages; current-voltage curves for sodium and potassium at low (but not high) concentrations; and the inhibition of sodium conductance by thallium. The results suggest that the potential well is located close to the channel mouth and that the conductance is partially limited by the rate going from the bulk solution to the well. For thallium, this entrance rate is probably diffusion limited.  相似文献

Summary CsCl density gradient analysis showed that the DNA of plaque forming particles ofSalmonella phageP22 is lighter than the host DNA. The DNA of transducing phages exhibits an intermediate density, but close to host DNA. BU labelling of DNA synthesized in the cells after phage infection resulted in a density increase of transducing DNA of about 0.004 gxcm-3, whereas infectious DNA increased by about 0.045 gxcm-3. Shearing of isolated DNA molecules from unlabelledP22 lysates demonstrated that transducing DNA consists of two pieces of DNA of different density: 90% stem from the bacterial host whereas 10% are phage DNA and therefore responsible for the BU lable in transducing phages.  相似文献

Summary The current-voltage curve of theChara membrane was obtained by applying a slow ramp de- and hyperpolarization by use of voltage clamp. By inhibiting the electrogenic pump with 50m DCCD (dicyclohexylcarbodiimide), theI–V curve approached a steady state within 100 min, which gave thei d -V curve of the passive diffusion channel. Thei p -V curve of the electrogenic pump channel was obtained by subtracting the latter from the former. With the increase of external pH, thei d -V curve showed only a slight change, while thei p -V curve of the pump channel showed almost a parallel shift, in the hyperpolarizing direction, along the voltage axis in the pH range between 6.5 and 7.5. The sigmoidali p -V curve in this pH range could be simulated satisfactorily with the five-state model reported previously (U. Kishimoto, N. Kami-ike, Y. Takeuchi & T. Ohkawa,J. Membrane Biol. 80:175–183, 1984) as well as with a lumped two-state model presented in this report. The analysis based on these models suggests that the electrogenic pump of theChara membrane is mainly a 2H+/1ATP pump. The forward rate constant in the voltage-dependent step increased with the increase of external pH, while the backward one decreased. On the other hand, the forward rate constant in the voltage-independent step remained almost unchanged with the increase of external pH, while the backward one increased markedly. The pump conductance at the resting membrane potential showed either a slight increase or a decrease with the increase of external pH, depending on the sample. Nevertheless, the pump current showed generally a slight increase with the increase of external pH.  相似文献

Density gradient centrifugation was used to monitor DNA replication during sporulation of a 5-bromo-2'-deoxyuridine-tolerant, thymidine-requiring strain of Bacillus subtilis. DNA of heavy, intermediate, and light density was found in cells induced to sporulate in the presence of bromodeoxyuridine, but only intermediate DNA was detected in mature spores. Cells grown with bromodeoxyuridine until DNA was in the heavy form formed spores containing intermediate and light DNA when sporulated with thymidine alone.  相似文献

A general stochastic theory is presented for analysis of current records of a patch containing an arbitrary number (N) of independent homologous channels in the steady-state. We give the "basic theorem" that at the instant of any open (or shut) transition of a channel, the other N-1 channels are located in each state with a probability equal to those in the steady-state, if enough transitions are observed. Using the "basic theorem", we derived: (a) the time-dependent open and shut frequencies after a definite type of transition, and (b) the probability density functions (pdf) of the duration of any period between two successive transitions. Briefly, the main results obtained were: (1) The time-dependent open (or shut) transition frequency after every shut (or open) transition at t = 0 in an N-channel patch, fJSh,Op(t)(N) (or fJOp,Sh(t)(N)), is the same as that of a one-channel patch except for the value of the constant. (2) In the all-shut (or all-open) period of a patch, the average duration of the period is 1/N, and the slowest exponential decay constant contained in the pdf is N times those of a single channel patch, respectively. (3) An example calculation for small N showed that the stochastic properties of a single channel can be obtained even when N is uncertain, if the channel open probability is small and exponential decay constants are separated. (4) When the channels are in equilibrium, the pdf of duration of every type of period in the patch is described by a sum of exponential terms with positive coefficients. This paper describes an inexpensive, space-saving, and labor-efficient method for housing and making repeated observations of individual ticks held under specific photoperiodic, temperature, and humidity conditions. Using 96-well assay plates to house ticks, various developmental parameters, sex, survival, etc., of several thousand individual ticks can be recorded routinely in less than 1 hr. This design also permits more than 3,500 individual ticks to be maintained in 1 small (18.9-L) aquarium that serves as a humidity chamber. A small, unilluminated constant temperature incubator was converted inexpensively into a light and temperature programmable unit to contain the aquarium by adding a timing system, light fixture, and a second thermostat.  相似文献

Voltage-dependent gating behavior of Shaker potassium channels without N-type inactivation (ShB delta 6-46) expressed in Xenopus oocytes was studied. The voltage dependence of the steady-state open probability indicated that the activation process involves the movement of the equivalent of 12-16 electronic charges across the membrane. The sigmoidal kinetics of the activation process, which is maintained at depolarized voltages up to at least +100 mV indicate the presence of at least five sequential conformational changes before opening. The voltage dependence of the gating charge movement suggested that each elementary transition involves 3.5 electronic charges. The voltage dependence of the forward opening rate, as estimated by the single- channel first latency distribution, the final phase of the macroscopic ionic current activation, the ionic current reactivation and the ON gating current time course, showed movement of the equivalent of 0.3 to 0.5 electronic charges were associated with a large number of the activation transitions. The equivalent charge movement of 1.1 electronic charges was associated with the closing conformational change. The results were generally consistent with models involving a number of independent and identical transitions with a major exception that the first closing transition is slower than expected as indicated by tail current and OFF gating charge measurements.  相似文献

Chromatography of serum lipids on a silicic acid column by gradient elution and with continuous detection is described. The gradient is formed in a new organic gradient device based on the principle of E. Peterson and H. A. Sober ((1959) Anal. Chem.31, 857), starting from six initial organic solvents. The detection is realized by flame ionization on a LCM2 liquid chromatography detector (Pye-Unicam). The technique is less solvent- and time-consuming and is of interest in preparative separation and analysis of lipophilic substances.  相似文献

Components of seed yield of cv. Ultra (Lupinus albus L.) andcv. Unicrop (L. angustifolius L.) were measured when grown atthree densities. The low density (10 plants m–2) Unicropyield (34 g seed per plant) was 1.8 times that of Ultra as ithad more branches, pods and seeds per pod. Ultra seeds (310mg per seed) were heavier than Unicrop seeds (180 mg). The branchingpattern of Ultra was less dependent on plant density, henceat 93 plants m–2 it gave a higher per plant yield (7.4vs 6.4 g) than Unicrop at lower densities (83 plants m–2).Density had most influence on pod formation and only small effectson seeds per pod and seed weight. Yield components on the main-steminflorescence were influenced less by density than componentson branch inflorescences. Later formed, higher order generationsof inflorescences were most affected by increased inter- andintra-plant competition. Pod numbers on the main-stem were similarfor both species. Pods formed at higher flower nodes in Unicrop,but the lower flower nodes were less fertile than those in Ultra.Node position of flowers had no influence on seed set in main-stemUnicrop pods, but pods from higher nodes in Ultra formed fewerseeds.
