The present study investigated the effect of prostaglandins (PG) on the in vitro production of polyclonal IgG and IgM by pokeweed mitogen- stimulated normal human peripheral mononuclear cells. Concentrations of PGE1 and PGA1 in excess of 10−6M were suppressive. PGE2 and PGs of the F series were less effective and significant suppression was seen in concentrations greater than 10−3M. Indomethacin added to cell cultures did not enhance Ig production. This discrepancy between physiologic PG concentrations and the very large pharmacologic concentration necessary to suppress Ig synthesis in vitro makes the physiologic role of PG in the modulation of Ig synthesis questionable.  相似文献

In vitro synthesis of progesterone and estradiol-17β from endogenous precursors was studied in the placenta from women in early stage of gestation (< 7 weeks). Radioimmunoassay techniques were used to measure progesterone and estradiol-17β.It was shown that placental tissue from as early as six weeks of gestation can synthesize both progesterone and estradiol-17β in vitro. Prostaglandins F and E2 in concentration of 100 μg/ml of the incubation media did not have any significant effect on the in vitro synthesis of progesterone and estradiol-17β in the placental tissue.It seems unlikely that the abortifacient effect of natural prostaglandins PGE2 and PGF is due to their direct action on the synthesis of progesterone and estradiol-17β in the placenta.  相似文献

We have demonstrated the capacity of isolated follicles from estrous rabbits to synthesize prostaglandins in vitro and to respond to gonadotropins added to the incubation medium with an increased accumulation of these lipids. The increase in both prostaglandins became apparent only after 5 hours of incubation. The effect was specific for hormones with LH activity and the threshold dose of LH appeared to be 0.005 μg/ml. In addition, we have shown that cyclic AMP (0.02 M) added to the incubation medium also increased prostaglandin in this in vitro system and appears to be a likely mediator of this action of LH.  相似文献

Synthesis of COP (prostaglandins; PG and thromboxanes; Tx) from exogenous and endogenous arachidonic acid (AA) was studied in isolated perfused lungs from rats treated in vivo with a single dose of α-naphthylthiourea (ANTU; 10mg/kg;). Lung dry: wet weight ratios showed changes characteristic of oedema between 6 and 16h after ANTU. Bioassay of COP showed that COP synthesis from exogenous AA was raised above control values in lungs from rats treated with ANTU, reaching a maximum at 16h after treatment. By radioimmunoassay, the major increase was in 6-oxo-PGF, with lesser effects on PGE2 and PGF levels. Synthesis of bioassayable COP from endogenous AA induced by the calcium ionophore A23187 was increased as early as 2h after ANTU treatment and remained elevated up to 70h. In lungs 28h after ANTU, 6-oxo-PGF release was greater than in normal lungs. These results show that in this model of pulmonary oedema, the potential for COP synthesis was increased. From the time course of this effect, increased COP synthesis was probably a response to the initial damage rather than a cause of the oedema.  相似文献

A series of straight phase gel chromatography systems have been developed for the separation of prostaglandin methyl esters. Using the methyl esters of prostaglandins B2, E2, F2α and F2β, the basic relationships between elution volume and the polarities of the gel, the solvent system (heptane-chloroform mixtures), and the prostaglandin have been determined. The separation of prostaglandin methyl esters with slight differences in structure has been demonstrated. Examples include oxo and hydroxy prostaglandins, hydroxy epimers, double bond isomers, prostaglandins of varying α- and ω-chain length, and 1- and 2- (5,6 cis double bond) series prostaglandins. In view of the general advantages of liquid-gel chromatography, it is suggested that these systems may be useful for isolation and purification in a number of areas in the prostaglandin field.  相似文献

Antibodies against 15 keto PGF and 13,14 dihydro 15 keto PGF were produced in goats and rabbits using the appropriate prostaglandin protein conjugate. Tritium labeled 15-keto, and 13,14 dihydro 15-keto PGF were prepared from 3H-PGF. These antibodies and 3H-labeled compounds were used to develop radioimmunoassays for the respective F metabolites. The antibodies had relatively little cross-reactivity (≤0.1%) with the parent F molecule. Infusion of PGF in monkeys increased 15-keto-h2 levels 10–20 fold higher than PGF in peripheral plasma. The levels of this metabolite were not altered detectably during clotting, indicating relatively slow rates of PGF metabolism in vitro. These assays should be useful to follow release rates of exogenous prostaglandins from various formulations and delivery systems, and in vivo tissue synthesis of PGF, where low levels preclude measuring the parent compound.  相似文献

The p-nitrophenacyl esters of a number of closely related and isomeric prostaglandins were resolved by HPLC on a microparticulate silica gel column (Zorbax-Sil ®, DuPont). Ten F-series prostaglandin analogs, eight E-series prostaglandin analogs, the isomeric 15(R)- and 15(S)-methyl prostaglandins of the E- and F-series and, lastly, PGA2 and PGB2 were chromatographed under conditions generating 2,000 to 7,000 theoretical plates. Conditions are described for quantitative conversion of prostaglandins to p-nitrophenacyl esters in less than 6 minutes at room temperature. Linear peak height and peak area plots were obtained for in-situ esterified PGE2 p-nitrophenacyl ester over the range of 0.4 – 3.1 μg. The lower limit of detection of this ester is about 1 ng. A linear relationship is observed between silica gel TLC 1/Rf values and HPLC retention times as predicted by theory.  相似文献

In view of recent findings which suggest that renal prostaglandins mediate the effect of hypoxia on erythropoietin production, we have studied whether hypoxia is a stimulus for in vitro prostaglandin synthesis. Studies were carried out in rat renal mesangial cell cultures which produce erythropoietin in an oxygen-dependent manner. Production rates of PGE2 and in specified samples also of 6-keto-PGF, as a measure of PGI2, and PGF were determined by radioimmunoassay after incubation at either 20% O2 (normoxic) or 2% O2 (hypoxic) in gas permeable dishes for 24 hrs. Considerable variation in PGE2 production was noted among independent cell lines. PGE2 production appeared to be inversely correlated to the cellular density of the cultures. In addition, PGE2 production was enhanced in hypoxic cell cultures. The mean increase was 50 to 60%. PGF and 6-keto-PGF increased by about the same rate. These results indicate that hypoxia is a stimulus for in vitro prostaglandin production.  相似文献

Because of its highly unstable nature, TXA2, produced by platelet metabolism of arachidonic acid, does not lend itself to use as a receptor probe for its own receptor. As such, the stable TXA2/PGH2 antagonist, trans-13-azaprostanoic acid (trans-13-APA, 12b), was prepared as the [17,18 3H] derivative ([3H] trans-13-APA, 12c) to study this receptor and to better evaluate the mechanism of action of these azaprostanoids. Tritiated trans-13-APA, 12c, was prepared in nearly theoretical specific activity (57 Ci/mmole) from (17z)-trans-13-azaprost-17-enoic acid (11b) by catalytic tritiation. The unsaturated 11b was prepared by condensation of cis-7-amino-3-heptene (8) with 2-(6-carboxyhexyl) cyclopentanone (9), NaBH4 reduction, chromatography, and hydrolysis of the trans isomer so isolated. The olefins 11a and b were also of biochemical interest because of the unsaturation in the lower side chain. The presence of similar unsaturation in PGH3 (4) and TXA3 (3) renders these prostaglandins inactive as proaggregatory agents. Evaluation of the antiaggregatory activity of 11a and b indicated it to be about the same potency in inhibiting human platelet aggregation as the parent cis and trans-13-APAs, suggesting that introduction of a double bond at the 17 position in platelet prostaglandin antagonists is unlikely to result in enhanced antiplatelet activity.  相似文献

The effect of extra- and intracellularly injected prostaglandins (PG) E2 and F2 on electrical activity and responses to acetylcholine and serotonin were studied in experiments on identified neurons ofHelix pomatia. As a rule prostaglandins modified the typical electrical activity of the identified neurons: PG E2 enhanced and PG F2 depressed it. These substances mainly weakened responses of the nerve cells to mediators: PG E2 caused a greater change in the response to serotonin and PG F2 in the response to acetylcholine. Effects of the prostaglandins when injected extracellularly and intracellularly differed. The possible molecular-cellular mechanisms of the central action of prostaglandins are discussed in the light of their functional connections with other universal regulators of cellular metabolism and with proteins specific for nerve tissues.P. Cysts of Entamoeba invadens obtained under axenic culture conditions have been reported to be similar to cysts of the human intestinal parasite E histolytica both in morphology and chitin presence in their wails. Mature E. invadens cyst forms, isolated from cultures following discontinuous Percoll gradient sedimentation were resistant (>80%) to detergent treatment. Addition of chitin synthesis inhibitors such as Polyoxin D and Nikkomycin (50 μg/ml) to cultures in encystation media markedly inhibited (>85%) the formation of detergent resistant cysts and prevented the incorporation of radiolabeled chitin precursor N-acetyl[3H]glucosamine. These findings suggest that chitin synthesis inhibitors may serve as drugs which specifically block the life cycle of the Entamoeba parasite.  相似文献

In exponentially growing cells of Saccharomyces cerevisiae, cycloheximide stimulated intracellular protein degradation to the same extent as did starvation for required amino acids. By using inhibitors of macromolecular synthesis and temperature-sensitive mutants defective in different steps of RNA and protein synthesis it could be demonstrated, that this stimulation of protein degradation was directly related to the inhibition of protein synthesis per se, but not connected to the cessation of ribosomal RNA synthesis or to the inhibition of cell growth.  相似文献

The hemodynamic effects of PGF, PGE2, and norepinephrine injected into the umbilical arterial circulation were compared in nine fetal lambs in utero. Umbilical blood flow was measured with radioactive microspheres and an electromagnetic flow transducer implanted on the distal aorta of the fetus after ligation of external iliac arteries and other accessible distal aortic branches.PGF and norepinephrine increased fetal arterial pressure and umbilical blood flow while umbilical vascular resistance increased slightly (PGF) or not at all (norepinephrine). PGE2 increased fetal arterial pressure, decreased umbilical blood flow, and exerted a profound active vasoconstrictor effect on the fetal placental bed. Our data taken together with the observations of others suggest that prostaglandins may play a role in the circulatory adaptations of the fetus at birth and that PGE2 in high concentrations is likely to have deleterious hemodynamic consequences in the fetus in utero.  相似文献

The synthesis of PGC2 methyl ester is described. Comparison of the synthetic material with the methyl ester prepared from natural PGC2 showed the two to be identical, thus confirming the structure assignment. The physical and biological data of PGC2 methyl ester are presented.Horton and Jones have recently shown that PGA1 (Ic) and PGA2 (Ia) are deactivated on incubation with cats blood (1,2). Jones has proposed that this deactivation involves the enzymic conversion of PGA (I) to an 11,12-double bond isomer (PGC, II) which is subsequently isomerized by base to the inactive PGB (III) (3). Structure assignment of the PGC's in the earlier study were based on chromatographic mobilities and uv and mass spectrometric properties. We report here a total synthesis of PGC2 methyl ester (IIb). Comparison of the biological and physical properties of this material with those of diazomethane-esterified natural material confirms the earlier structure assignment for PGC2 (IIa).  相似文献

The ability of various prostaglandins (PGs) to affect the in vitro anamnestic immune response of keyhole limpet hemocyanin (KLH)-primed rabbit popliteal lymph node cells was investigated. Of the four PGs studied (PGA1, PGE2 and PGF), PGE1 was found to have a stimulatory effect, whereas PGA1, PGE2 and PGF were ineffective in stimulating or inhibiting the in vitro anamnestic response. Under the conditions studied, a 3.5-fold increase in antibody production was obtained in PGE1-treated, KLH-stimulated cultures. Maximum enhancement was obtained when 0.2 μg of PGE1 were added at the time of culture initiation and were allowed to remain in contact with the lymph node cells for 24 hours.  相似文献

Summary Enzyme synthesis, foaming behaviour and its effects were studied using two common cellulolytic fungi;Trichoderma reesei andSporotrichum pulverulentum in a medium containing sugar beet cosette as a cellulosic substrate. Summary Grape must and concentrated rectified grape must were used for the gluconic acid synthesis using Aspergillus niger batch cultures. The latter substrate was the better, with a production, at 72 h, of 67.43 g/l and a yield (calculated on converted glucose) of 0.96. Citric acid was also observed as a by-product. In order to decrease the residual fructose content, at the end of the gluconate production cycle, an experimental model of sequential fermentation A. nigerRhizopus arrhizus was proposed for the synthesis of gluconic and fumaric acid. The use of Glucose-isomerase (EC 5.3.1.5) to convert fructose to glucose was also tested.  相似文献

The four deoxynucleoside triphosphate pools in unfertilized eggs of L. pictus and S. purpuratus were measured and found to be very large, ranging from 10?3 to 10?2 pmoles per egg. The high levels of the individual dNTP pools are sufficient for one to eight rounds of DNA synthesis. During the first division cycle these pools fluctuate with the highest levels being attained prior to DNA synthesis. The pools then decrease just preceding or during the S period. There is a large reduction in the total cellular dNTP in later stages of development when DNA synthesis is reduced relative to the cleavage stages.  相似文献
