Several studies suggest that the concentration of immunoreactive (I) FSH measured in peripheral plasma by radioimmunoassay does not always reflect the level of bioactive (B) hormone capable of eliciting a biological response (e.g. oestradiol synthesis by Sertoli cells in vitro). The aim of this study was to measure both B-FSH and I-FSH concentrations in male and female sheep during the first year of life, and to relate this to pubertal development. The hypothesis being tested was that B-FSH is present in both male and female sheep during the prepubertal period and that discrete changes in B-FSH are associated with the onset of puberty. Eight ewe lambs and eight ram lambs were blood sampled fortnightly from 2 to 52 weeks of age. All samples were assayed for B-FSH and I-FSH content. Pubertal development was monitored in ewe lambs from behavioural oestrus and from plasma progesterone concentrations, and in ram lambs from penile and testicular development and from plasma testosterone concentrations. Mean I-FSH concentrations varied significantly with time after birth, in both females and males ( P<0.01). In contrast, B-FSH was found to vary with time in females only ( P<0.01). Around the expected time of puberty in ram lambs (i.e. at 30–40 weeks of age), and thereafter, I-FSH concentrations were undetectable (<0.2 ng ml −1), whereas the B-FSH concentrations were measurable at concentrations up to twice the assay detection limit (0.8 ng ml −1) until 38 weeks of age. In ewe lambs, but not ram lambs, there was a significant linear relationship between B-FSH and I-FSH values ( R=0.595; P<0.005). When standardised about the time of puberty, B-FSH ( P<0.05) but not I-FSH was significantly higher in ewe lambs that failed to reach puberty. No differences for either B-FSH or I-FSH between pubertal and non-pubertal ram lambs were noted. In summary, B-FSH was often measurable in plasma throughout prepubertal development in sheep and the concentrations often differed from those of I-FSH, especially in ram lambs. However, there appeared to be no discrete changes in B-FSH that could be directly related to specific pubertal events. It is concluded that although FSH may be a prerequisite for prepubertal testicular development and/or ovarian follicular growth, it is not a critical factor in determining whether puberty is attained during the first year of life in this seasonally breeding species. 相似文献

Summary Male rainbow trout were treated with salmon gonadotropic hormone (GTH) at different stages of the circannual reproductive cycle; spawning fish were also treated with an antiserum against salmon GTH. Injection of GTH led to a several-fold increase of plasma sex steroid levels during spermatogenesis and in the spawning season but was without effect at early stages of testicular development. GTH neutralization during the spawning season was followed by a several-fold decrease of plasma sex steroid levels. During spermatogenesis and in the spawning season, both treatment regimes resulted in an increased sensitivity of testicular explants in response to a subsequent stimulation of steroid secretion in vitro. This up-regulatory response may facilitate and maintain the high sex steroid plasma levels observed during the spawning season. It may also be necessary to allow for concomitant peak values of plasma GTH and sex steroids in the spawning season, a situation difficult to understand within the negative feedback concept. The adaptive capacities of the testicular steroidogenic system indicate that it is not only an effector site for GTH but also an active part of the endocrine system controling reproduction.Abbreviations BSA

Neonates and infants with congenital heart disease with increased pulmonary blood flow suffer morbidity from poor oxygenation and decreased lung compliance. In a previous experiment involving 4-wk-old lambs with pulmonary hypertension secondary to increased pulmonary blood flow following an in utero placement of an aortopulmonary vascular graft, we found a decrease in surfactant protein (SP)-A gene expression as well as a decrease in SP-A and SP-B protein contents. To determine the timing of these changes, the objective of the present study was to characterize the effect of increased pulmonary blood flow and pulmonary hypertension on SP-A, -B, and -C gene expressions and protein contents within the first week of life. Of eight fetal lambs that underwent the in utero placement of the shunt, there was no difference in the expression of SP-A, -B, and -C mRNA levels or SP-A and -B protein contents compared with age-matched controls. The results showed that, in this model of congenital heart disease with pulmonary hypertension and increased pulmonary blood flow, the effect of the shunt on SP gene expression and protein content was not apparent within the first week of life. 相似文献

Male rats of the Wistar/Han strain are treated with ovine FSH, hCG or PMS in various doses for twelve days, beginning on the 1st, 12th, 24th or 36th day of life each. The determination of FSH in plasma by radioimmunoassay at the end of the treatment shows that all exogenous gonadotropins depress the FSH levels to immeasurable values. Plasma testosterone increases after application of hCG and PMS, not ovine FSH. To explain these findings a hypothalamic inhibition of the FSH release by a "short feedback" mechanism is discussed. 相似文献

The goal of the present study was to prospectively assess the plasma selenium (Se) concentrations of term and preterm infants during the first year of life in relation to gestational age and nutrition. Blood specimens were collected from orally formula-fed preterm infants (gestational age < 32 weeks, birth weight < 1500 g): 1.) in hospital and 2.) corrected for gestational age parallel to healthy term breast and formula-fed infants at the ages of 1, 4 and 12 months. All infants were fed according to a standardized nutritional concept, solids and follow-up formula were introduced at the age of 4 months. Plasma selenium in preterm infants in hospital was 11.7 (6.5–20.8) g/l and 11.6 (8.8–16.7) g/l at 4 weeks corrected for gestational age. At the age of 4 months plasma selenium was still significantly lower than in the other groups: Preterm infants: 17.1 (10.4–30.5) g/l; formula-fed term infants: 31.3 (24.3–47.5) g/l; breast-fed term infants: 45.6 (27.1–65.1) g/l). The levels of breast-fed infants were significantly higher than those of both formula-fed groups up until the introduction of solids. Preterm infants had significantly low plasma selenium levels up until a postnatal age of at least 6 months. The levels were lower than those of term infants fed an identical unsupplemented infant formula during the first 4 months of life. These data support routine monitoring in hospital and selenium supplementation of preterm infants, preferably in hospital before discharge. 相似文献

The data on the efficacy of antibacterial drugs and their combinations in treatment of 150 three-month old infants with generalized infection due to Pseudomonas aeruginosa are presented. The clinical isolates of P. aeruginosa were found to be carriers of multiple drug resistance which markedly complicated the chemotherapy. Only combined antibacterial therapy of such infants proved to be rational. High activity of aminoglycosides, azlocillin and cefotaxime against P. aeruginosa and the synergistic action of their combinations observed in the patients permitted to recommend the use of combinations of the above drugs in the empirical chemotherapy. 相似文献

Two experiments were carried out to analyse FSH secretion in the ewe. The first was a long-term study during which four ewes under controlled photoperiods were checked for plasma concentrations of FSH twice daily for a period of 16 months. They were successively anestrous, cycling, gestating and lactating. The results suggested that an endogenous secretion rhythm of FSH persisted throughout each of the physiological states of the ewes. The periodic cycles of FSH production lasted about 5 days during anestrus and gestation but extended to about 6 days during estrus. One of the three waves of secretion we noted during one cycle was represented by the two periovulatory surges, the first coincident with the LH peak, the second occuring 30-40 h later. Plasma levels of FSH were similar during estrous cycles and anestrus, whereas the FSH secretion decreased gradually throughoug gestation. During lactation, large differences were observed among animals before the recovery of cyclic ovarian activity. The second experiment consisted of frequent blood sampling (every ten minutes) of eight ewes for 6 hours during anestrus. FSH was secreted differently compared to LH. No pulsatile production of FSH was demonstrated and no increase in FSH levels was seen at the time of the episodic LH surge. 相似文献

When rats were infused with a solution containing triiodothyronine, amino acids, glucagon and heparin (solution A) the hepatocytes increased DNA synthesis and decreased plasma membrane sialic acid. In order to study whether the reduced levels of sialic acid in the plasma membrane were associated with hepatocyte proliferation, different mixtures of three components of solution A were infused into rats and the DNA synthetic activity as well as the sialic acid content measured. Results reported here show a correlation between DNA synthetic activity and sialic acid reduction suggesting that the decrease in the plasma membrane sialic acid can be a pre-replicative step associated to cell proliferation. 相似文献

To investigate fetal regulation of the endocrine testis during the third trimester of gestation, pregnenolone, progesterone, 17-hydroxyprogesterone, androstenedione, testosterone, FSH and prolactin concentrations were measured in the umbilical circulation of 31–35 preterm (27–37 weeks) and 18–19 fullterm (39–42 weeks) male infants, and postnatally until 5 days of age in 27–39 fullterm male infants. 17-hydroxyprogesterone and prolactin concentrations increased significantly ( P < 0.001) between 27–37 weeks of gestation; the other hormones measured were unchanged. The levels of progesterone in preterm infants, and prenenolone, progesterone and 17-hydroxyprogesterone in the cord vein of fullterm infants were significantly ( P < 0.001–0.05) higher than those in the cord artery. Androstenedione concentrations were similar in the cord artery and vein, and decreased less than pregnenolone, progesterone and 17-hydroxyprogesterone after birth, reflecting major androstenedione production in the fetus. Testosterone concentrations were higher ( P < 0.01–0.05) in the cord artery than in the vein, both in preterm and fullterm infants, showing the main site of testosterone production to be the fetal compartment. Postnatally, testosterone increased clearly from concentrations of 0.25 ± 0.05 (SE) mg/ml in the cord artery and 0.10 ± 0.01 in the cord vein to 0.94 ± 0.14 ng/ml in the peripheral vein on the first postnatal day, and decreased thereafter clearly between 3–5 days. FSH did not change during the first 5 postnatal days. Concentrations of all the other hormones measured decreased significnatly after birth.It is concluded from the cord blood hormone levels of infants born between 27–42 weeks of gestation that: (1) The third trimester of gestation represents a stable phase of endocrine development with relatively small changes in circulating hormone levels; (2) Both the placenta and the fetus seem to be able to produce androstenedione in the perinatal period; and (3) The initial increase in testosterone after birth is indicative of the inhibitory effect of placental steriods on testicular endocrine function during the last trimester of gestation. 相似文献

Romney ewes were infused with ovine FSH (NIADDK-oFSH-16) for 48 h from the initiation of luteolysis with cloprostenol. Doses of 2.5 or 5 micrograms/h which partly or completely prevented the normal preovulatory decline in plasma FSH concentrations caused a significant increase in mean ovulation rates. Ovulation rates were not increased significantly if the FSH (5 micrograms/h) was infused for only 20 h starting from the initiation of luteolysis or 24 h later. Infusion of a less potent and relatively impure preparation of FSH (i.e. FSH-P) at 0.5 mg/h for 48 h after cloprostenol treatment also increased the mean ovulation rate significantly. However, if the FSH-P was given for only the first 24 h, or if the start of the infusion was delayed for more than 12 h, mean ovulation rates were not increased significantly. Infusion of LH (NIADDK-oLH-25, 5 micrograms/h) for 48 h from the initiation of luteolysis decreased the mean ovulation rate significantly. Administration of bovine follicular fluid to suppress plasma FSH concentrations below normal during the first 24 h after cloprostenol injection did not delay oestrus. However, oestrus was delayed by approximately 2 days if plasma FSH concentrations were reduced by bovine follicular fluid 24 h after the initiation of luteolysis. As ovulation rate increased, the mean weight of individual corpora lutea of each ewe decreased. In ewes with a single ovulation, most corpora lutea weighed greater than 600 mg, but as the ovulation rate increased the proportion of corpora lutea present weighing less than 400 mg rose steadily.(ABSTRACT TRUNCATED AT 250 WORDS) 相似文献

A positive correlation between male social status and testosterone levels is expected and often found in social species with high rates of agonistic interactions or when social relationships among males are unstable. In contrast, in species with low rates of agonistic interactions or when social relationships are stable, testosterone levels should not correlate with social status. The "challenge hypothesis" predicts that androgen levels should rise during periods of courtship or mate guarding. We addressed these questions in free-ranging spotted hyenas, a species with low rates and low intensities of aggression among males but where males spend extensive effort to court females. In males, we measured testosterone, its precursor androstenedione, and its metabolite 5alpha-dihydrotestosterone. As predicted, testosterone levels were significantly higher and androstenedione levels tended to be higher in males that, at the time of sampling, defended a female, compared with males that did not defend a female. Also, as predicted, there was no correlation between social status and androgen levels in male spotted hyenas. 相似文献

Gonadotropins, prolactin (PRL), testosterone (T), delta 4-androstenedione, dehydroepiandrosterone sulfate and cortisol (F) levels were determined from 14 days before birth to term in 3 female and 3 male ovine fetuses with a chronically implanted venous catheter, and in the same animals from birth to 72 h of age. In both sexes, plasma gonadotropins and androgens were low throughout the period of study while plasma F increased with gestational age. After birth, plasma gonadotropins and PRL tended to increase progressively with time while PRL concentrations were significantly higher in female than in male lambs. F and T concentrations decreased significantly within the first 12 and 6 h of postnatal life. Higher T values were again observed at 36 h in male lambs. These data indicate that the fetal hypothalamic-pituitary-gonadal axis is relatively quiescent in the last 14 days of gestation but is activated within the first 72 h after birth. 相似文献

PRL plasma levels and FSH and LH pituitary reserve were tested in ten apparently healthy male subjects. A good correlation was found between PRL on one hand and FSH plasma levels (p less than 0,05), LH plasma levels (p less than 0,01) and FSH pituitary reserve (p less than 0,01) on the other hand. This seems to support the current hypothesis that prolactin may cause a progressive clinically latent impairment in the spermatogenetic function of the testis. Further evidence is needed. 相似文献
