Controlled environment experiments were performed to determinethe effects of temperature and water potential on germination,radicle elongation and emergence of mungbean (Vigna radiata(L.) Wilczek cv. IPB-M79-17-79). The effects of a range of constant temperatures (15–45°C) and water potentials (0 to –2.2 MPa) on germinationand radicle elongation rates were studied using an osmoticumtechnique, in which seeds were held against a semi-permeablemembrane sac containing a polyethylene glycol solution. Linearrelationships were established between median germination time(Gt50) and water potential at different temperatures, and betweenreciprocal Gt50 (germination rate) and temperature at differentwater potentials. Germination occurred at potentials as lowas –2.2 MPa at favourable temperatures (30–40 °C),but was fastest at 40 °C when water was not limiting, withan estimated base temperature (Tb) of about 10 °C. Subsequentradicle elongation, however, was restricted to a slightly narrowertemperature range and was fastest at 35 °C. The conceptof thermal time was used to develop an equation to model thecombined effects of water potential and temperature on germination.Predictions made using this model were compared with the actualgermination obtained in a related series of experiments in columnsof soil. Some differences observed suggested the additionalimportance of the seed/soil/water contact zone in influencingseed germination in soil. Seedling emergence appeared to reflectfurther the radicle elongation results by occurring within anarrower range of temperatures and water potentials than germination.Emergence had an estimated Tb of 12.6 °C and was fastestat 35 °C. For a deeper understanding of the germination of chick–pea(Cicer arietinum) seeds, which is dependent upon ethylene synthesis,a crude extract containing authentic ACC oxidase (ACCO) activitywas isolated in soluble form from the embryonic axes of seedsgerminated for 24 h. Under our optimal assay conditions (200mM HEPES at pH 7.0, 4µM FeS04, 6 mM Na–ascorbate,1 mM ACC, 20% 02, 3% CO2 , and 10%glycerol) this enzyme was5–fold more active than under the conditions we used initiallyin the present work. The enzyme has the following Km: 28 µMfor ACC (approximately 4–fold less than in vivo), 1.2%for O2 (in the presence of an optimal CO2 concentration of 3%),and 1% for CO2 in the presence of O2 (20%). The enzyme is inhibitedby phenanthroline (PNT) (specific chelating agent of ferrousion), and competitively inhibited (K1, =0.5 mM) by 2–aminoisobutyricacid (AIB), and the enzymatic activity was not detectable inthe absence of CO2. Under optimal assay conditions, the enzymehas two optimum temperatures (28 C and 35 C) and is inhibitedby divalent metal cations (Zn2+> CO2+>Ni2+>Cu2+>Mn2+>Mg2+) and by salicylic acid, propylgallate, carbonyl cyanidem–chlorophenyl hydrazone (CCCP), dinitrophenol (DNP),and Na–benzoate. The in vitro ACCO activity which we recoveredin soluble form is equivalent to approximately 80–85%of the apparent activity evaluated in vivo. In "air-grown" Chroomonas sp. cells, low concentrations of DCMU(less than 0.1 µM) could prevent the inhibition of 14CO2fixation by anaerobiosis under light-saturating conditions (morethan 40 W.m–2), with phenazine methosulfate showing asimilar effect. Antimycin A, carbonyl cyanide m-chlorophenylhydrazone(CCCP), and N,N'-dicyclohexylcarbodiimide strongly inhibitedanaerobic photosynthesis at concentrations which did not significantlyinhibit the rate under 2% O2 at high light intensity (200 W.m–2),although 0.2 µM CCCP stimulated the rate under 2% O2 tosome extent. On the other hand, KCN inhibited the rate muchmore strongly under 2% O2 than N2, although it inhibited therate very strongly at concentrations above 5 µM both underN2 and 2% O2. These results suggest that the inhibition of photosynthetic14CO2 fixation by anaerobiosis in this alga result from ATPdeficiency caused by over-reduction of electron carriers ofthe cyclic electron flow and that oxygen can prevent the over-reduction.Cyclic electron flow seems to be necessary to provide additionalATP for CO2 reduction under anaerobic conditions, although itseems to be less necessary under aerobic conditions. Effects of ethylene, CO2 and O2 on the germination of after-ripenedupper cocklebur (Xanthium pennsylvanicum Wallr.) seeds wereexamined in relation to pre-irradiation by red (R) or far-red(FR) light In order to remove the pre-existing Pfr, seeds weresoaked in the dark for various periods prior to light irradiationand gas treatments. Regardless of light, 0.3 Pa C2H4 promotedgermination at 23 ?C, but it strongly inhibited germinationwhen applied at 33 ?C, the optimal temperature for the germinationof this seed. However, delayed application of C2H4 during 33?C incubation stimulated germination independently of lightin a similar manner to that seen at 23 ?C. It is, therefore,suggested that the germination-regulating action of C2H4 iscompletely independent of phytochrome. In contrast, the germination-promoting effect of 3–0 kPaCO2 was pronounced only when the seeds were previously irradiatedby R, regardless of temperature, suggesting that CO2 actionto promote germination depends upon Pfr. A synergism betweenCO2 and C2H4 at 23 ?C was observed only in the germination ofseeds pre-irradiated by R, while at 33 ?C an antagonism occurredindependently of light. The stimulation of C2H4 production byCO2 was most striking in the cotyledonary tissue pre-irradiatedby R. However, the R-dependent enhancement of CO2-stimulatedC2H4 production was negated by the subsequent FR and it wasnot found in the presence of 1-aminocyclopropane-1-carboxylicacid (ACC). Moreover, the R dependency of the germination-promotingCO2 effect disappeared in the presence of C2H4. The R-dependentC2H4 production enhanced by CO2 may thus be involved, at leastpartially, in some step of conversion from methionine to ACC. The germination-promoting effect of C2H4, but not CO2, was enhancedby O2 enrichment regardless of light. The C3 species wheat and rice and the C4 species maize weregrown for 2–3 weeks in controlled environment growth chambersat 20 or 30 °C day and 15 °C night temperatures. CO2assimilation rates (at 20 and 30 °C) and several leaf parametersincluding total nitrogen, soluble protein, and RuBP carboxylaseprotein were determined. When the assimilation rates under atmosphericCO2 and O2 levels were expressed on a total nitrogen basis (=nitrogen use efficiency), the C4 species maize had a greaternitrogen use efficiency than either of the two C3 species examined,regardless of the combination of temperatures used for growthor measurement of CO2 assimilation. Maize is also shown to makemore efficient use of its soluble protein and RuBP carboxylaseprotein than either wheat or rice when measurements are madeat 320 parts 10–6 CO2 and 21% O2. Atmospheric CO2 enrichmentduring CO2 assimilation measurements increased the nitrogenuse efficiency in the C3 species. In one treatment (wheat grownand measured at 20 °C), CO2 assimilation under saturatingCO2 showed nitrogen, soluble protein, and RuBP carboxylase proteinuse efficiencies equal to or greater than that of the C4 species. These data indicate that C4 species may make more efficientuse of their nitrogen, soluble protein, and RuBP carboxylaseprotein than C3 species under atmospheric CO2 conditions. The response of photosynthesis in the flag leaf of rice (Oryzasativa) to elevated CO2 or reduced O2 was investigated relativeto other environmental factors using steady-state gas exchangetechniques. We found under moderate conditions of temperatureand photosynthetic flux density (PFD) (26°C and 700µmolquanta m–2s–1 similar to growth conditions) photosynthesisin the flag leaf of rice during heading and grain filling saturatedat near ambient levels of CO2, with a concomitant loss of O2sensitivity, when a high stomatal conductance was maintainedby high humidity (low vapor pressure deficit). Under 18°Cthere was near complete loss of O2 sensitivity of photosynthesisat normal ambient levels of CO2. This is in contrast to thelarge enhancement of photosynthesis by supra-atmospheric levelsof CO2 and sub-atmospheric levels of O2 by suppression of photorespirationwhen there is no limitation on utilizing the initial productof CO2 assimilation (triose-P) as predicted from Ribulose-l,5-bisphosphatecarboxylase/oxygenase (Rubisco) kinetic properties. Thus, lossof sensitivity to CO2 and O2 has been previously explained asa limitation on utilization of triose-P to synthesize carbohydrates.Under high PFD at 25°C, the rate of photosynthesis in ricedeclined over a period of hours around midday, while the intercellularlevels of CO2 remained constant suggesting a limitation on utilizationof photosynthate. Short-term fluctuations in climatic factorsincluding temperature, light and humidity could result in afeedback limitation on photosynthesis in rice which may be exacerbatedby rising CO2. Nodulated white clover plants (Trifolium repens L. cv. Huia)were grown for 71 d in flowing nutrient solutions containingN as 10 mmol m–3 NH4NO3, under artificial illumination,with shoots at 20/15°C day/night temperatures and root temperaturereduced decrementally from 20 to 5°C. Root temperatureswere then changed to 3, 7, 9, 11, 13, 17 or 25°C, and theacquisition of N by N2 fixation, NH4+ and NO3 uptakewas measured over 14 d. Shoot specific growth rates (d. wt)doubled with increasing temperature between 7 and 17°C,whilst root specific growth rates showed little response; shoot:root ratios increased with root temperature, and over time at11°C. Net uptake of total N per plant (N2 fixation + NH4++ NO3) over 14 d increased three-fold between 3 and 17°C.The proportion contributed by N2 fixation decreased with increasingtemperature from 51% at 5°C to 18% at 25°C. Uptake ofNH4+ as a proportion of NH4+ + NO3 uptake over 14 d variedlittle (55–62%) with root temperature between 3 and 25°C,although it increased with time at most temperatures. Mean ratesof total N uptake per unit shoot f. wt over 14 d changed littlebetween 9 and 25°C, but decreased progressively with temperaturebelow 9°C, due to the decline in the rates of NH4+ and NO3uptake, even though N2 fixation increased. The results suggestthat N2 fixation in the presence of sustained low concentrationsof NH4+ and NO4 is less sensitive to low root temperaturethan are either NH4+ or NO3 uptake systems. The rates of CO2 assimilation by potted spray carnation plants(cv. Cerise Royalette) were determined over a wide range oflight intensities (45–450 W m–2 PAR), CO2 concentrations(200–3100 vpm), and leaf temperatures (5–35 °C).Assimilation rates varied with these factors in a way similarto the response of single leaves of other temperate crops, althoughthe absolute values were lower. The optimal temperature forCO2 assimilation was between 5 and 10 °C at 45 W m–2PAR but it increased progressively with increasing light intensityand CO2 concentration up to 27 °C at 450 W m–2 PARand 3100 vpm CO2 as expressed by the equation TOpt = –6.47-h 2.336 In G + 0.031951 where C is CO2 concentration in vpmand I is photo-synthetically active radiation in W m–2.CO2 enrichment also increased stomatal resistance, especiallyat high light intensities. Wheat plants were grown in a controlled environment with daytemperatures of 18 ?C and with 500 µ Einsteins m–28–1 of photosynthetically active radiation for 16 h. Beforeanthesis and 2 to 3 weeks after, rates of net photosynthesiswere measured for leaves in 2 or 21% O2 containing 350 vpm CO2at 13, 18, 23, and 28 ?C and with 500 µEinsteins m–2s–1 of photosynthetically active radiation. Also, underthe same conditions of light intensity and temperature, therates of efflux of CO2 into CO2-free air were measured and,for mature flag leaves 3 to 4 weeks after anthesis, gross andnet photosynthesis from air containing 320 vpm 14CO2 of specificactivity 39?7 nCi µmol–1. When the O2 concentration was decreased from 21 to 2% (v/v)the rate of net photosynthesis increased by 32 per cent at thelowest temperature and 54 per cent at the highest temperature.Efflux of CO2 into CO2-free air ranged from 38 per cent of netphotosynthesis at 13 ?C to 86 per cent at 28 ?C. Gross photosynthesis,measured by the 14C assimilated during 40 s, was greater thannet photosynthesis by some 10 per cent at 13 ?C and 17 per centat 28 ?C. Stands of groundnut (Arachis hypogaea L.), a C3 legume, weregrown in controlled-environment glasshouses at 28 °C (±5°C)under two levels of atmospheric CO2 (350 ppmv or 700 ppmv) andtwo levels of soil moisture (irrigated weekly or no water from35 d after sowing). Elevated CO2 increased the maximum rate of net photosynthesisby up to 40%, with an increase in conversion coefficient forintercepted radiation of 30% (from 1–66 to 2–16g MJ–1) in well-irrigated conditions, and 94% (from 0–64to 1·24 g MJ–1) on a drying soil profile. In plantswell supplied with water, elevated CO2 increased dry matteraccumulation by 16% (from 13·79 to 16·03 t –1) and pod yield by 25% (from 2·7 to 3·4t ha–1).However, the harvest index (total poddry weight/above-grounddry weight) was unaffected by CO2 treatment. The beneficial effects of elevated CO2 were enhanced under severewater stress, dry matter production increased by 112% (from4·13 to 8·87 t ha–1) and a pod yield of1·34t ha–1 was obtained in elevated CO2, whereascomparable plotsat 350 ppmv CO2 only yielded 0·22 t ha-1.There was a corresponding decrease in harvest index from 0·15to 0·05. Following the withholding of irrigation, plants growing on astored soil water profile in elevated CO2 could maintain significantlyless negative leaf water potentials (P<0·01) for theremainder of the season than comparable plants grown in ambientCO2, allowing prolonged plant activity during drought. In plants which were well supplied with water, allocation ofdry matter between leaves, stems, roots, and pods was similarin both CO2 treatments. On a drying soil profile, allocationin plants grown in 350 ppmv CO2 changed in favour of root developmentfar earlier in the season than plants grown at 700 ppmv CO2,indicating that severe waterstress was reached earlier at 350ppmv CO2. The primary effects of elevated CO2 on growth and yield of groundnutstands weremediated by an increase in the conversion coefficientfor intercepted radiation and the prolonged maintenance of higherleaf water potentials during increasing drought stress. Ribulose bisphosphate carboxylase (E.C.4.1.1.39) was purifiedfrom leaves of Triticum aestivum, Hordeum vulgare, Spinaceaoleracea, Petroselinum crispum, salad mustard-most likely Brassicanapus, Helianthus annuus, Solanum tuberosum, Beta vulgaris,Lolium perenne, Equisetum arvense, Zea mays, Ginkgo biloba,Pteris aquilina, Salix babylonica, Chamaecyparis lawsonianaand Atrichum undulatum by density gradient centrifugation andgel filtration or by ammonium sulphate fractionation, densitygradient centrifugation, ion-exchange chromatography and gelfiltration. Purified enzymes were freeze-dried and then storedat 0 °C to 4 °C. Portions of each enzyme preparationwere reactivated at 25 °C for 5 h in the presence of 10mM HCO2 and 20 mM MgCl2-RuBP carboxylase activities were measuredat four different concentrations of CO2 at 25 °C and pH8.2 in solutions equilibrated with pure nitrogen or air (21%O2, 79% N2). Km(CO2), Vmax and K1(O2) values were computed fromthe results. Significant differences were found in the Km(CO2)values for enzymes isolated from different species. The effect of sucrose, H2BO3, KNO3, Ca(NO2)2.4H2O and MgSO4.7H2O on pollen germination of Phytolacca dodecandra L. (endod)in a liquid medium was investigated. Sucrose and H3BO3 werecritical to pollen germination. A concentration of 10% sucroseand 161.8 µm H2BO3 gave over 70% germination. The germinationof pollen was not enhanced by Ca(NO3)2.4H2O, KNO3 and MgSO4.7H2O.Endod pollen was dehydrated over CaCl2 and stored in gelatincapsules in cryogenic vials at –175 °C, 1±1°C and 24±2 °C. The pollen moisture content atcollection was approx. 7.8% (f. wt basis) and dehydration overCaCl2 reduced it to about 1.4%. Pollen stored at 1±1°C and –175 °C maintained viability for over 6months. Pollen stored at room temperature lost viability within4 weeks of storage. Pollination with cryopreserved pollen resultedin normal fruit set. Several models have been proposed to describe germination rates,but most are limited in statistical analysis and biologicalmeaning of indices. Therefore, a mathematical model is proposedto utilize the logistic function. The function was defined asan overall response including time, temperature, and the interactionbetween time and temperature. Cumulative germination percentagesover time were used to develop the model. Germination tests were conducted on indiangrass (Sorghastrumnutans (L.) Nash) strain ‘IG-2C-F1’, at constanttemperatures of 9, 12, 15, 20, 25, and 30 °C. The functionfitted the observed data over six temperatures at r2 = 0.99.Time to reach 10% of final germination (Gt10) increased from2.5 d at 30 °C to 44.0 d at 9 °C, and Gt50 (time toreach 50% of final germination) increased from 3.6 d at 30 °Cto 53.8 d at 9 °C. True germination rate (% d–1) foreach temperature was maximum at Gt50. A linear model of 1/Gt50versus temperature was used to estimate the base temperatureof 8.3 °C for germination. An Arrhenius plot indicated achange occurred between 20 °C and 25 °C for temperatureresponse of germination. Published data on hypocotyl growthof Cucumis melo L. were recalculated using the model. Absolutegrowth rates showed a temperature response similar to the publishedweighted-mean elongation rates. Base temperature for hypocotylgrowth of C. melo was estimated as 8.8 °C. The respiratory effluxes of nodules and of roots of FiskebyV soyabean (Glycine max (L.) Merr.), grown in a controlled environment,were measured at intervals in air and 3% O2 from shortly afterthe onset of N2 fixation until plant senescence. The respiratoryburdens linked with nitrogenase plus ammonia metabolism, andnodule growth and maintenance, were calculated from gas exchangedata and related to the concurrent rates of N2 fixation. The specific respiration rates of nodules increased to a maximumof 21 mg CO2 g–1 h–1 at the time pods began development:the equivalent maximum for roots was c. 4.5 mg CO2 g–1h–1. Maximum nodule and root respiration rates per plantwere attained about 25 d later at the time N2 fixation peakedat 15 mg N d–1 plant–1. The relationship between nodule respiration and N2 fixationindicated an average respiratory cost of 13.2 mg CO2 mg–1N until the last few days of plant development Separation ofnodule respiration into the two components: nitrogenase (+ NH3metabolism) respiration and nodule growth and maintenance respiration,indicated that the latter efflux accounted for c. 20% of nodulerespiration while N2 fixation was increasing and new noduletissue was being formed. When nodule growth ceased and N2 fixationdeclined, this component of respiration also declined. The respiratorycost of nitrogenase activity plus the associated metabolismof NH3 varied between 11 mg CO2 mg–1 N during vegetativeand early reproductive growth, to 12.5 mg CO2 mg–1 N duringthe later stages of pod development. The leaves of willow cuttings were allowed to assimilate 14CO2,and the rate of increase of activity in the sieve-tube sap,collected as aphid honeydew, was compared with the output ofrespiratory 14CO2 from the stem at 25 °C and at 0 °C. In the case of 3–5-week-old young shoots, the relativerate of breakdown of the 14C-labelled sugars was reduced at0 °C as compared with 25 °C. With 2–4-year-oldmature stems, however, the rate of breakdown of labelled sugarsin transit through the phloem was increased at 0 °C relativeto that at 25 °C. When the labelled sugars were being respiredin isolated stem segments where no transport was taking place,then low temperatures markedly reduced the rate of breakdownin both young shoots and mature stems. These results are discussed in relation to the results of otherworkers who have studied temperature effects on sieve-tube transport.  相似文献

The CO2 compensation point at 25 °C and 250 µEinsteinsm–2 s–1 wasmeasured for 27 bryo-phyte species, andwas found to be in the range of 45–160 µl CO2 I–1air. Under the same conditions Zea mays gave a value of 11 µlI–1 and Horde um vulgare 76 µI–1. The rate of loss of photosyntheticallyfixed 14CO2 in the light and dark in six bryophytes (three mosses,two leafy liverworts, one thalloid liverwort) was determinedin CO2-free air and 100% O2. The rate of 14CO2 evolution inthe light was less than that in the dark in CL2-free air, butin 100% O2 the rate in the light increased, so that in all butthe leafy liverworts it was greater than that in the dark. Raisingthe temperature tended to increase the rate of 14CO2 evolutioninto CO2-free air both in the light and dark, so that the light/dark(L/D) ratio did not greatly vary. The lower rate of loss of14CO2 in the light compared tothe dark could be due to partialinhibition of ‘dark respiration’ reactions in thelight, a low rate of glycolate synthesis and oxidation, or partialreassimilation of the 14CO2 produced, or a combination of someor all of these factors.  相似文献

Opuntia ficus-indica, an extremely productive CAM plant cultivatedin many countries, was exposed to 36, 52, and 72–73 PaCO2 in field plots and open-top chambers. Initiation of newcladodes (stem segments) was monitored until the canopy closed,after which bimonthly harvests maintained the plants for oneyear at a cladode area per unit ground area that is optimalfor biomass production. Doubling the CO2 partial pressure slightlyincreased the number of first-order daughter cladodes growingon the basal (planted) cladodes after 3 months and nearly doubledthe number and area of second-order cladodes. When the C02 levelwas doubled, cladodes were 5% thicker after a few months and11 to 16% thicker after one year. Although the productivityenhancement by elevated C02 tended to decrease during the year,the annual above-ground dry-mass gain was 37 to 40% higher whenthe C02 level was doubled, reaching 65 tons hectare–1year–1 in a field plot. Well-watered cladodes at day/nightair temperatures of 25°C/15°C and a total daily photosyntheticphoton flux (PPF) of 15 mol m–2 d–1 in controlledenvironment chambers had 74% more net CO2 uptake over 24 h at73 Pa than at 37 Pa CO2. With doubled CO2, the percentage enhancementof net CO2 uptake increased as the PPF was lowered, as the temperaturewas raised, and during drought. Using an environmental productivityindex based on such factors, net CO2 uptake and hence productivityof O. ficus-indica can be predicted for elevated CO2 levelsand other variations accompanying global climate change. Soybeans were grown for three seasons in open-top field chambersto determine (1) whether elevated CO2 (360 versus 700 µmolmol–1) alleviates some of the yield loss due to pollutantO3, (2) whether the partial stomatal closure resulting fromchronic O3 exposure (charcoal-filtered air versus 1.5 ambientconcentrations) is a cause or result of decreased photosynthesis,and (3) possible implications of CO2/O3 interactions to climatechange studies using elevated CO2. Leaf conductance was reducedby elevated CO2, regardless of O3 level, or by exposure to O3alone. As.a result of these effects on conductance, high CO2reduced estimated midday O3 flux into the leaf by an averageof 50% in charcoal-filtered air and 35% in the high O3 treatment.However, while exposure to O3 reduced seed yields by 41% atambient CO2 levels, the yield reduction was completely amelioratedby elevated CO2. The threshold midday O3 flux for yield lossappears to be 20–30 nmol m–2 s–1 in this study.Although elevated CO2 increased total biomass production, itdid not increase seed yields. A/Ci curves show a large reductionin the stomatal limitation to photosynthesis due to elevatedCO2 but no effect of O3. These data demonstrate that (1) reducedconductance due to O3 is the result, and not the cause, of reducedphotosynthesis, (2) 700 µmol mol–1 CO2 can completelyameliorate yield losses due to O3 within the limits of theseexperiments, and (3) some reports of increased yields underelevated CO2 treatments may, at least in part, reflect the ameliorationof unrecognized suppression of yield by O3 or other stresses.
