Rapid developments in scientific and technological aspects in stem cell biology and tissue engineering have led to the increased use of human cells and tissues for the treatment of various diseases and injuries. The regulatory environment for CTT products is rapidly evolving and drug regulatory agencies are working towards establishment of a risk-based system with some common features. Various drug regulatory agencies in many countries/regions have implemented regulatory controls in the last few years. This article will highlight some of works done till date to regulate CTT products in Australia, Canada, Europe, Japan, Korea, Singapore and United States of America.  相似文献

microRNAs (miRNAs) are identified as a class of non-protein regulators and a new source for broad control of gene expression in eukaryotes. The past years have witnessed substantial progress in understanding miRNA functions and mechanisms, although a few controversies remain. Various hypotheses and models have been suggested for the mechanisms of miRNA repression, including translational inhibition at the level of initiation or elongation, rapid degradation of the nascent peptide, mRNA degradation, and mRNA sequestration into P bodies (processing bodies) and SGs (stress granules) for degradation or/and storage. Recently, some noncanonical miRNA regulation, such as miRNA activation and de-repression of miRNA inhibition, have been uncovered. This review discusses some recent advances about how miRNAs regulate their targets and various modes of miRNA function.  相似文献

Most ground nesters lay pigmented eggs, and egg pigmentation generally matches the environment. Pigmentation of eggs has evolved as a protective device against predation, but dark-pigmented eggs can be susceptible to overheating when exposed to solar radiation. The Ostrich (Struthio camelus) lays white eggs that are unattended for the first few weeks before incubation, and are quite visible to predators. To evaluate the effect of colour on the surface and core temperatures, we painted some Ostrich eggs dark brown or white, and left some unpainted (control), and exposed all of them directly to the sun during the day. The surface and core temperatures of brown eggs were significantly higher than those of the white-painted and control eggs. In addition, the core temperature of brown eggs exceeded 37.5°C, which is the temperature at which embryo mortality starts to increase. In a second experiment, we placed eggs (brown-painted and control) in various types of vegetation to study their visibility to an observer walking towards them. The white eggs were discovered from a significantly longer distance than the brown eggs, indicating that the predation risk may be much higher for white eggs. The results thus suggest that white eggs minimise overheating and allow the Ostrich to leave its eggs unattended before incubation starts, but they are more susceptible to predation.  相似文献

There is a high prevalence of Fasciolopsis buski in Sawasdee Village in the Nam Som District, Udonthani Province in northeastern Thailand, an endemic area of liver fluke infection. From stool examination of 183 villagers participating in the study, the authors detected a fluke parasitic infection rate of 14.8% (27 cases). Of interest, the authors found a similar high prevalence of both Opisthorchis viverrini (16 cases, 8.7%) and F buski (13 cases, 7.1%). This region is the endemic area of liver fluke, not of F buski. The surrounding environment is the hill area of Nayoong Namsom mountain, which is not a suitable area for F buski larvae. However, the village has been inhabited by a number of immigrants from various regions of Thailand over the past few years. The F buski can be carried by these people, possibly explaining the migration of the fluke parasite to this setting. The nonendemic parasite infection was also detected at high prevalence in the community. Wide spectrum control for various parasitic infections in villages like Sawasdee is necessary.  相似文献

We analyze an increasingly popular NMR method analogous to the black lipid membrane (BLM) isotopic tracer experiment for the study of mediated cation transport but involving the preparation of vesicles with an environment asymmetric in that paramagnetic metal ions are present only outside the vesicles. This asymmetry is manifest in the NMR spectrum as two distinct resonances for magnetic nuclei in outside and inside lipid headgroups. As mediated transport begins and for the paramagnetic metal ions enter the vesicles, the inner headgroup resonance line shifts and changes shape with a time course containing much information on the actual ion transport mechanism. Processes by which the ions enter the vesicles one or a few at a time (such as via a diffusive carrier) are easily distinguishable from those by which the ions enter in large bursts (such as by pore activation). The limiting case where intervesicular mediator exchange is slow relative to cation transport (the situation for integral membrane proteins) is treated analytically. Computer simulated curves indicate conditions necessary for certain changes in the line shape which are analogous to the "current jumps" observed in BLM conductance studies. The theory derived allows estimates of the average number of ions entering the first few bursts, how often the bursts occur, and how they depend on the concentration of the mediating species in the vesicular membrane. Preliminary experimental spectra illustrating some of the various possible line shape behaviors are presented.  相似文献

Antigen-independent selection of intracellular stable antibody frameworks   总被引:1,自引:0,他引:1  
The intracellular expression of highly specific antibody fragments ("intrabodies") in eukaryotes has a great potential in functional genomics and therapeutics. However, since the intracellular reducing environment prevents formation of the conserved intrachain disulfide bonds, most antibodies do not fold properly and are therefore inactive inside cells. The few antibodies that have been found to function in an intracellular environment and that have been characterized for their biophysical properties have generally shown a high degree of stability and solubility. Thus, for intracellular expression and application, very stable antibody frameworks are needed that can correctly fold even in the absence of disulfide bonds and that do not aggregate. Here, we present and discuss a novel method, named "Quality Control," which allows selection of stable and soluble antibody frameworks in vivo without the requirement or knowledge of antigens. This system is based on the expression of single-chain antibody fragments (scFvs) fused to a selectable marker that can control gene expression and cell growth. The activity of such a selectable marker fused to various scFvs that have been biophysically characterized correlated with the solubility and stability of the scFv moieties. This antigen-independent intrabody selection system was applied to screen scFv libraries for identifying stable and soluble frameworks, which subsequently served as acceptor backbones to construct intrabody libraries by randomization of hypervariable loops.  相似文献

AIMS: Mutagenic pollution of natural environment is currently one of the most serious ecological problems. Therefore, rapid detection of the presence of mutagens is a very important issue. Although many mutagenicity assays have already been described, only a few are suitable for testing samples from natural environment. One of such assays is a microbiological mutagenicity test based on genetically modified Vibrio harveyi strains. The aim of this work was to modify and improve the V. harveyi assay. METHODS AND RESULTS: A series of V. harveyi dark and dim mutants were tested for reversion of their phenotype towards efficient light emission in response to incubation with known mutagens. Luminescence of the A16 strain (luxE mutant) increased significantly after a few hours of such a treatment with various mutagenic agents, revealing a dose-response correlation. Sensitivity of the assay has been determined for different mutagens. CONCLUSIONS: The luminescence-based V. harveyi mutagenicity assay is rapid, sensitive and reveals a dose-response correlation for various mutagens. SIGNIFICANCE AND IMPACT OF THE STUDY: The assay developed in this study is a potentially useful tool in studies on mutagenic pollution of environment, especially marine water.  相似文献

It also talks about the history of biofiltration, and also proposes few ideas for the future developments in the biofiltration research pertaining to VOC control. Moreover, the paper also discusses about various important physical, chemical and biological factors which affect the performance of a biofilter both directly and indirectly. Controlled environment sunlit plant growth chambers have been built because of a great interest in plant responses to environmental variables under light intensities approaching those of natural sunlight conditions. Individual research projects have designed sunlit chambers that differ in size, structure, material, and environmental control systems dependent on the goals of the projects. Most literature describes plant organism responses to environmental variables, whereas reports of system design and performance are few. The objective of this article is to present a review of the engineering aspects of the design, environmental control, and performance of sunlit growth chambers that have been described in the literature. Most controlled environment sunlit growth chambers have been constructed with experimental plants grown in either pots or soil bins. Although a few sunlit growth chambers were designed for field grown plants, precise environmental control was not available. Further progress in the development of precise controlled environment sunlit growth chambers should include portability (or movability) so these chambers can be used in multiple field sites for greater cost-effectiveness. Modifications for improvements in guidelines for the design and operation of controlled environment growth chamber studies are also proposed.  相似文献

Bisphenol A (BPA), is an industrially important compound and is widely used for the production of polycarbonates and other plastics. Over the past few years, there have been many issues raised all over the world on the use of BPA. BPA is known to possess estrogenic activities; hence, it mimics the role of estrogen once it enters living systems. Thus, it has been placed in the category of compounds called endocrine disruptors. It can cause damage to reproductive organs, thyroid gland, and brain tissues at developmental stages, and most recently it has also been linked to cancer development in humans. Here, in this review, we aim to summarize the various effects of BPA on humans and animals, and at the same time we wish to throw some light on the emerging field of biodegradation of BPA in the natural environment. A few studies conducted recently have tried to isolate BPA-degrading microorganisms from various sites, like water bodies receiving wastes from industries, landfills, etc. In the present scenario, with huge controversies on the use of BPA, we emphasize on bridging the gap between studies, aiming at finding the damage caused by BPA, and the studies which aim at the safe removal of BPA from the environment, with the help of naturally occurring microbes. Once this gap is filled, we will be able to find a way which will allow the use of BPA in manufacturing plastics, without its accumulation in the environment.  相似文献

Membrane receptors and ion channels respond to various stimuli and relay that information across the plasma membrane by triggering specific and timed processes. These include activation of second messengers, allowing ion permeation, and changing cellular excitability, to name a few. Gaining control over equivalent processes is essential to understand neuronal physiology and pathophysiology. Recently, new optical techniques have emerged proffering new remote means to control various functions of defined neuronal populations by light, dubbed optogenetics. Still, optogenetic tools do not typically address the activity of receptors and channels native to neurons (or of neuronal origin), nor gain access to their signaling mechanisms. A related method—synthetic optogenetics—bridges this gap by endowing light sensitivity to endogenous neuronal receptors and channels by the appending of synthetic, light‐receptive molecules, or photoswitches. This provides the means to photoregulate neuronal receptors and channels and tap into their native signaling mechanisms in select regions of the neurons, such as the synapse. This review discusses the development of synthetic optogenetics as a means to study neuronal receptors and channels remotely, in their natural environment, with unprecedented spatial and temporal precision, and provides an overview of tool design, mode of action, potential clinical applications and insights and achievements gained.  相似文献

This study compares laying performances between two environments of dwarf laying hen lines segregating for the naked neck mutation (NA locus), a selected dwarf line of brown-egg layers and its control line. Layers with one of the three genotypes at the NA locus were produced from 11 sires from the control line and 12 sires from the selected line. Two hatches produced 216 adult hens in Taiwan and 297 hens in France. Genetic parameters for laying traits were estimated in each environment and the ranking of sire breeding values was compared between environments. Laying performance was lower, and mortality was higher in Taiwan than in France. The line by environment interaction was highly significant for body weight at 16 weeks, clutch length and egg number, with or without Box-Cox transformation. The selected line was more sensitive to environmental change but in Taiwan it could maintain a higher egg number than the control line. Estimated heritability values in the selected line were higher in France than in Taiwan, but not for all the traits in the control line. The rank correlations between sire breeding values were low within the selected line and slightly higher in the control line. A few sire families showed a good ranking in both environments, suggesting that some families may adapt better to environmental change.  相似文献

The present study was undertaken to determine the effect of simulated microgravity on the testis of the rats and to evaluate the possibility of spermatogenesis failure in space environment. Fifty-four adult male albino rats were used in this study. They were divided equally into intact control, stress control and experimental groups. The rats of the intact control group (Group Ia) were kept without intervention. The rats of both the stress control and experimental groups were subjected to inguinal canal ligation and tail-suspension. In the stress control group (Group Ib) the hindlimbs were not elevated above the floor of the housing units whereas in the experimental groups the hindlimbs were elevated for one week (Group II) and six weeks (Group III), respectively. In a third experimental group (Group IV) the rats were hindlimb-suspended for six weeks followed by another six weeks without suspension to allow recovery. Prior to sacrifice, the animals were weighed and anesthetized, and the testes were excised and weighed. Testicular specimens were processed for histological, histochemical and morphometric studies. The results of the present study revealed that only after six weeks of hindlimb-suspension, the rats showed a significant decline in testicular weight compared with the control groups. Histologically, few abnormalities were observed in some seminiferous tubules in one-week hindlimb-suspended group. Spermatogenesis was significantly reduced by six-week of hindlimb-suspension marked by atrophy of the testes and loss of all germ cells, except a few spermatogonia. Spermatogenesis was partially restored in the recovery group. In all groups the appearance of Sertoli cells remained the same. Proliferation of Leydig cells was observed in the experimental groups. It is concluded that spermatogenesis is severely inhibited by six weeks of hindlimb-suspension and that it is partially restored following six weeks of recovery. This study provides further insights regarding the serious effects of long-term exposure to microgravity on the testes of mammals, including human beings.  相似文献

A wheeled mobile mechanism with a passive and/or active linkage mechanism for rough terrain environment is developed and evaluated. The wheeled mobile mechanism which has high mobility in rough terrain needs sophisticated system to adapt various environments.We focus on the development of a switching controller system for wheeled mobile robots in rough terrain. This system consists of two sub-systems: an environment recognition system using link angles and an adaptive control system. In the environment recognition system, we introduce a Self-Organizing Map (SOM) for clustering link angles. In the adaptive controllers, we introduce neural networks to calculate the inverse model of the wheeled mobile robot.The environment recognition system can recognize the environment in which the robot travels, and the adjustable controllers are tuned by experimental results for each environment. The dual sub-system switching controller system is experimentally evaluated. The system recognizes its environment and adapts by switching the adjustable controllers. This system demonstrates superior performance to a well-tuned single PID controller.  相似文献

The stomatal pores of plant leaves, situated in the epidermis and surrounded by a pair of guard cells, allow CO2 uptake for photosynthesis and water loss through transpiration. Blue light is one of the dominant environmental signals that control stomatal movements in leaves of plants in a natural environment. This blue light response is mediated by blue/UV A light-absorbing phototropins (phots) and cryptochromes (crys). Red/far-red light-absorbing phytochromes (phys) also play a role in the control of stomatal aperture. The signaling components that link the perception of light signals to the stomatal opening response are largely unknown. This review discusses a few newly discovered nuclear genes, their function with respect to the phot-, cry-, and phy-mediated signal transduction cascades, and possible involvement of circadian clock.  相似文献

From a phylogenetic point of view, few organs of vertebrateshave shown such a plasticity of form and presumed function ashas the epiphyseal complex. From paired, probably sensory, evaginationsthrough the skulls of primitive vertebrates, the single remnantremaining in the higher vertebrates has evolved into a denselycellular, vascular organ with presumably secretory functions.Too few studies are available from throughout the vertebrateseries to allow conclusions as to function. Demonstrated effectson reproductive mechanisms, on phototaxis and pigment migration,on behavior, activity, and a variety of other mechanisms aretoo inconsistent among and between the various groups to permitgeneralizations other than the likelihood that the epiphysealcomplex directly or indirectly is responsive to some manifestationof light or to its absence. Working backward from this assumption,it seems likely that underlying the activity of these complexstructures will be a modulation of some basic, homeokineticmechanism having much to do with the seasonal adjustment ofthe organism to its environment.  相似文献

Roots are important to plants for a wide variety of processes, including nutrient and water uptake, anchoring and mechanical support, storage functions, and as the major interface between the plant and various biotic and abiotic factors in the soil environment. Understanding the development and architecture of roots holds potential for the exploitation and manipulation of root characteristics to both increase food plant yield and optimize agricultural land use. This theme issue highlights the importance of investigating specific aspects of root architecture in both the model plant Arabidopsis thaliana and (cereal) crops, presents novel insights into elements that are currently hardly addressed and provides new tools and technologies to study various aspects of root system architecture. This introduction gives a broad overview of the importance of the root system and provides a snapshot of the molecular control mechanisms associated with root branching and responses to the environment in A. thaliana and cereal crops.  相似文献

Lipid-rich, unilamellar membranes appear to be relatively common structures lining the apical or 'exposed' surface of epithelial cells. They have now been described in the intestinal tract from the esophagus to the rectum and have been isolated from tissues, such as the stomach, the small bowel, the colon, and the bladder. The presence of a lining layer in the lungs has been known for some time, and its functions, structure, and metabolism have been extensively studied, as can be gleaned from the multitude of reports presented at this symposium. The 'other' surfactants, however, have attracted far less attention and have been investigated in detail in only a few reports. This paucity of information, when compared to the pulmonary system, is most likely due to the fact that a generalized function (sufficiency state) or disease (deficiency state) has not yet been recognized for either the intestinal or urinary forms of surfactant. It seems reasonable to assume that the role of the SLP will vary, at least in part, with the organ or tissue with which it is associated, although the widespread nature of the membrane assumes that some functions (e.g. protective) will be shared. Thus, pulmonary surfactant's primary function in the lung may be to reduce surface tension and prevent lung collapse; but it also plays a significant part in the lung's defenses against bacterial and/or chemical invasion. It is hoped that future studies will shed some light on the function of the various SLPs and lead to a better appreciation for their role in both maintaining a healthy environment and contributing to the proper functioning of their host tissues.  相似文献

Arsenic is a known toxin and carcinogen that is present in industrial settings and in the environment. The mechanisms of disease initiation and progression are not fully understood. In the last a few years, there has been increasing evidence of the correlation between the generation of reactive oxygen species (ROS), DNA damage, tumor promotion, and arsenic exposure. This article summarizes the current literature on the arsenic mediated generation of ROS and reactive nitrogen species (RNS) in various biological systems. This article also discusses the role of ROS and RNS in arsenic-induced DNA damage and activation of oxidative sensitive gene expression.  相似文献
