华北地区棉铃虫对转Bt基因抗虫棉抗性适应的模拟模型   总被引:2,自引:3,他引:2  
通过对华北地区耕作制度和生态系统的了解,在充分考虑种群遗传学、生物学和人为操纵因子等三大因素的基础上,建立了一个预测棉铃虫对转Bt基因抗虫棉抗性适应的模拟模型。在华北地区典型的耕作制度下,如果所有棉田均为Bt棉,则Bt棉的预期寿命为7年;如果只有春播棉为Bt棉(约占棉田总面积的70%),则其寿命为10年。模型的灵敏度分析表明, Bt棉的使用寿命随抗性基因的显性度、初始抗性频率、Bt棉所占比例等因素的增长而迅速缩短。当Bt棉表达的杀虫蛋白量恰好全部杀死敏感基因型(GSGS)个体时,Bt棉的预期寿命最短。由于国外采用的“高剂量/庇护所”抗性治理策略不适用于棉铃虫及华北棉区的耕作制度,我国需要加强对其它抗性治理措施(如转双基因抗虫棉)的研究与应用。  相似文献

棉花耐害补偿反应可归纳为三种动态类型:1)不足补偿动态反应型;2)完全——不足补偿动态反应型;3)超越——完全——不足补偿动态反应型。其临界指标的建立及其应用可优化棉花病虫害综防决策.以研究害虫防治决策为例,剖析了利用害虫自然种群,人为改变害虫自然种群、人为地接放一定虫量与人工损害模拟等不同测定棉花耐害补偿能力方法的利弊。并探讨改进措施.分析论述了不同量化棉花耐害补偿能力的方法,并就棉花耐害补偿临界指标的建立及其意义作了探讨.棉花耐害补偿临界描标在棉田生态系统有害生物综合治理中可用于指导防治决策或直接用于防治决策,有着十分广阔的应用前景.最后就棉花耐害补偿临界指标及其应用的研究方向及有关问题作了讨论。  相似文献

Dexmedetomidine is a highly selective adrenergic receptor agonist, which has a dose-dependent sedative hypnotic effect. Furthermore, it also has pharmacological properties, and the ability to inhibit sympathetic activity and improve cardiovascular stability during an operation. However, its protective effect on patients with severe craniocerebral injury in the perioperative period remains unclear. Eighty adult male SD rats were used and divided into two groups (n = 40, each group): dexmedetomidine injury group (experimental group), and sodium chloride injury group (control group). Models of severe craniocerebral injury were established in these two groups using the modified Feeney’s free-fall method. As soon as the establishment of models was succeed, rat in the experimental group received 1 μg of dexmedetomidine (0.1 ml), while each rat in the control group was given 0.1 ml of 0.9% sodium chloride. Blood was sampled from an incision at the femoral vein to detect TNF-α and IL-2 levels at 1, 12, 24,36,48 and 72 h after establishing the model in the two groups. After severe craniocerebral injury, TNF-α levels of rats were lower in every stage and at different degrees in the experimental group than in the control group (P < 0.05), while IL-2 levels were lower in the experimental group to different extents (P < 0.05). Dexmedetomidine protects the brain of rats with severe craniocerebral injury by reducing the release of inflammatory mediators.  相似文献

目的:探讨血清IL-6及S-100B水平变化对颅脑损伤患者病情程度及预后评估的临床价值。方法:选择我院2013年7月~2015年7月收治的颅脑损伤患者100例为研究对象,依据格拉斯哥昏迷评分(GCS)将患者分为特重型损伤组、重型损伤组、中型损伤组和轻型损伤组,每组25例。另选择同期在我院接受体检的25位健康志愿者作为对照组。采用酶联免疫吸附法检测各组研究对象不同时间点血清IL-6及S-100B水平,并分析IL-6及S-100B水平变化与颅脑损伤程度及预后的关系。结果:颅脑损伤后第1、3、5、7、14天患者血清IL-6及S-100B水平均显著高于对照组,差异具有统计学意义(P0.05)。在相同检测时间,损伤程度越重的患者其血清IL-6及S-100B水平越高,差异具有统计学意义(P0.05);相同损伤程度颅脑损伤患者随着损伤时间延长,其血清IL-6水平越高,差异具有统计学意义(P0.05);相同损伤程度颅脑损伤患者血清S-100B水平在损伤后第1天开始上升,第3天达到高峰,第5天开始下降,差异具有统计学意义(P0.05)。颅脑损伤患者血清IL-6和S-100B水平与格拉斯哥预后评分(GOS)和格拉斯哥昏迷评分(GCS)呈负相关(rIL-6=-0.812、-0.770,rS-100B=-0.767、-0.831,P0.05),与颅脑损伤程度呈正相关关系(r=0.776、0.791,P0.05)。结论:血清IL-6和S-100B水平与颅脑损伤患者病情严重程度和预后相关,对颅脑损伤程度的临床诊断及预后评估具有重要参考价值。  相似文献

Cotton quality, a major factor determining both cotton profitability and marketability, is affected by not only the overall quantity of but also the type of the foreign matter. Although current commercial instruments can measure the overall amount of the foreign matter, no instrument can differentiate various types of foreign matter. The goal of this study was to develop a hyperspectral imaging system to discriminate major types of foreign matter in cotton lint. A push-broom based hyperspectral imaging system with a custom-built multi-thread software was developed to acquire hyperspectral images of cotton fiber with 15 types of foreign matter commonly found in the U.S. cotton lint. A total of 450 (30 replicates for each foreign matter) foreign matter samples were cut into 1 by 1 cm2 pieces and imaged on the lint surface using reflectance mode in the spectral range from 400-1000 nm. The mean spectra of the foreign matter and lint were extracted from the user-defined region-of-interests in the hyperspectral images. The principal component analysis was performed on the mean spectra to reduce the feature dimension from the original 256 bands to the top 3 principal components. The score plots of the 3 principal components were used to examine clusterization patterns for classifying the foreign matter. These patterns were further validated by statistical tests. The experimental results showed that the mean spectra of all 15 types of cotton foreign matter were different from that of the lint. Nine types of cotton foreign matter formed distinct clusters in the score plots. Additionally, all of them were significantly different from each other at the significance level of 0.05 except brown leaf and bract. The developed hyperspectral imaging system is effective to detect and classify cotton foreign matter on the lint surface and has the potential to be implemented in commercial cotton classing offices.  相似文献

以棉铃虫取食模型、种群动态模型及棉花生长发育模型为基础,通过对棉田生态系统中一些关键环节的分析,进一步发展出田间棉铃虫的管理系统。这个系统较好地反映出了国内外到目前为止在棉铃虫管理方面的最新研究成果。强调了在棉花生长发育过程中超补偿机制的实现,以及人工去早蕾与2代棉铃虫取食对棉花生长和产量形成的影响。本系统可以反映第2、3代棉铃虫不同卵量水平下,及不同人工去早蕾水平下棉花生长发育的动态过程。通过不同卵量水平时各去蕾量对产量的影响,以及不同用药时间的最终产量效果的比较,可以找出最佳的去蕾量以及合理的用药时间。  相似文献

本文对 2种不同栽培措施 (直播棉田与覆盖棉田 ) ,4种不同施药水平 (不施药、施 1次药、施 4次药、施 6次药 )的棉田的昆虫群落的特征指数进行了分析比较。连续多次施药会降低棉田昆虫群落的丰富度、多样性和均匀度。因此 ,在棉田害虫的综合治理中 ,应当不施药或尽量少施药 ,以利于充分利用天敌的自然控制作用 ,维持棉田生态系统的稳定。  相似文献

【目的】转Bt基因棉花和转Bt基因杨树在我国已推广使用。本研究的目的是调查和分析不同转基因杨棉复合系统内地上节肢动物群落多样性变化,为转基因杨树大规模应用提供生态安全方面的数据。【方法】2019年4-10月,在河北任丘采取欧洲黑杨Populus nigra林下种植转基因棉花Gossypium hirsutum的模式,设置转基因杨树 转基因棉花(复合生态系统1)和非转基因杨树-转基因棉花(复合生态系统2)两种杨棉复合系统,调查杨树和棉花地上部植株节肢动物种类和个体数量,比较节肢动物各功能群的物种数量和个体数量以及香农指数、优势集中性指数、均匀度指数等群落多样性指数,并测定了转基因杨树和棉花叶片Bt蛋白含量。【结果】复合生态系统1中转基因杨树和棉花各自植株上鳞翅目种群累计个体数均显著低于复合生态系统2相应植株上的个体数量。转基因杨树上叶甲类累计个体数量显著高于非转基因杨树上的,寄生蜂类累计个体数量则显著低于非转基因杨树上的。在6月5日、6月21日和9月7日的调查中,转基因杨树上的节肢动物群落香农指数显著高于非转基因杨树上的。除鳞翅目害虫外,2个复合系统中棉株上节肢动物各功能群累计个体数量无显著性差异,香农指数、优势集中性指数和均匀度指数随时间变化的趋势基本一致且同一调查时间无显著性差异,仅在8月17调查中复合生态系统1中棉株上节肢动物香农指数显著高于复合生态系统2棉株上的。将杨树和棉花上节肢动物各类群累计数量相加作为系统整体,复合生态系统1和复合生态系统2在香农指数、优势集中性指数和均匀度指数数值上无显著差异。在调查后期,2个复合系统中节肢动物种群均向杨树叶片聚集。转基因棉花叶片Bt蛋白含量在各调查期均显著高于转基因杨树叶片的。【结论】转基因杨树对靶标害虫种群数量有较好的控制作用,对同系统内棉花上鳞翅目害虫数量亦有协同控制作用。转基因杨棉复合系统中地上节肢动物群落结构稳定,对系统内棉花地上部节肢动物群落多样性无显著影响。靶标害虫对转Bt基因杨树的抗性发展需持续监测。  相似文献

Resistance evolution in target insects to Bacillus thurningiensis (Bt) cotton, Gossypium hirsutum L., is a main threat to Bt cotton technology. An increasing trend of population density of Helicoverpa armigera (Hübner) (Lepidoptera: Noctuidae) has been observed since 2001 in Qiuxian County (Hebei, China), where Bt cotton has been planted dominantly since 1998. This region was selected in 2006 and 2007 for estimating frequency of gene alleles conferring resistance to Bt cotton by screening the F1 progeny from single-pair cross between field-collected male and laboratory female of the Bt-resistant strain of H. armigera (F1 screen). F1 offspring from each single-pair line were screened for resistance alleles based on larval growth, development, and survival on Bt cotton leaves for 5 d. Two-year results indicated that approximately equal to 20% of field-collected males carried resistance alleles. The conservative estimate of the resistance allele frequency was 0.094 (95% CI, 0.044-0.145) for 2006 and 0.107 (95% CI, 0.055-0.159) for 2007. This is the first report of resistance allele frequency increase to such a high level in the field in China. Long-term adoption of Bt sprays, dominant planting of single-toxin-producing Bt cotton, and lack of conventional cotton refuge system might accelerate the resistance evolution in the region.  相似文献

Transgenic cotton (Cossypium hirsutum L.) varieties, adapted to China, have been bred that express two genes for resistance to insects, the CrylAc gene from Bacillus thuringiensis (Berliner) (Bt), and a trypsin inhibitor gene from cowpea (CpTI). Effectiveness of the double gene modification in conferring resistance to cotton bollworm, Helicoverpa armigera (Hübner) (Lepidoptera: Noctuidae), was studied in laboratory and field experiments. In each experiment, performance of Bt+CpTI cotton was compared with Bt cotton and to a conventional nontransgenic variety. Larval survival was lower on both types of transgenic variety, compared with the conventional cotton. Survival of first-, second-, and third-stage larvae was lower on Bt+CpTI cotton than on Bt cotton. Plant structures differed in level of resistance, and these differences were similar on Bt and Bt + CpTI cotton. Likewise, seasonal trends in level of resistance in different plant structures were similar in Bt and Bt+CpTI cotton. Both types of transgenic cotton interfered with development of sixth-stage larvae to adults, and no offspring was produced by H. armigera that fed on Bt or Bt+CpTI cotton from the sixth stage onward. First-, second-, and third-stage larvae spent significantly less time feeding on transgenic cotton than on conventional cotton, and the reduction in feeding time was significantly greater on Bt+CpTI cotton than on Bt cotton. Food conversion efficiency was lower on transgenic varieties than on conventional cotton, but there was no significant difference between Bt and Bt+CpTI cotton. In 3-yr field experimentation, bollworm densities were greatly suppressed on transgenic as compared with conventional cotton, but no significant differences between Bt and Bt+CpTI cotton were found. Overall, the results from laboratory work indicate that introduction of the CpTI gene in Bt cotton raises some components of resistance in cotton against H. armigera, but enhanced control of H. armigera under field conditions, due to expression of the CpTI gene, was not demonstrated.  相似文献

Because apelin may play an important regulatory role in human cardiac dysfunction, we investigated alterations in cardiovascular content of apelin and its receptor, APJ, during hypertension and the effect of exercise training on the cardiovascular apelin/APJ system in hypertensive animals. Spontaneously hypertensive rats (SHRs) underwent swimming training consisting of 54 swimming sessions of 60 min each (6 days/week for 9 weeks). Systolic blood pressure (SBP) was verified weekly by tail-cuff plethysmography. Apelin levels in plasma and cardiovascular tissues were determined by radioimmunoassay. The level of apelin/APJ mRNA was determined by RT-PCR. SHRs showed severe hypertension and pathological cardiomegaly. The level of apelin immunoreactivity (apelin-ir) in plasma and ventricular and aortic tissues was lower, by 40%, 40% and 42% (all P<0.01), respectively, in SHRs than in control Wistar-Kyoto rats, and the mRNA level of apelin and APJ in myocardium and aorta was markedly decreased. Compared with sedentary SHRs, swimming-trained SHRs showed decreased SBP and elevated mRNA expression of apelin and APJ in cardiovascular tissues and elevated apelin-ir level in plasma, myocardium and aorta (all P<0.01). SBP and level of apelin-ir in plasma and cardiovascular tissues were negatively correlated. Long-term swimming training relieved the pathogenesis of hypertension and reversed the downregulation of the cardiovascular apelin/APJ system induced by hypertension, which suggests that the improving effect of exercise training on hypertension could be mediated by upregulating the cardiovascular apelin/APJ system.  相似文献

摘要 目的:分析血清乳酸脱氢酶在评估颅脑手术患者重症肺炎的中的应用价值。方法:选取2020年7月~2022年6月在本院神经外科进行收治的颅脑择期手术患者268例,根据是否合并术后肺炎,分为非感染组(n=185)和肺炎组(n=83),肺炎组根据肺炎严重程度分为重症肺炎31例及普通肺炎52例。采用酶联免疫吸附法测定血清PCT、CRP水平;采用酶比色法检测血清 LDH 水平,采用Pearson相关性检验分析合并肺炎患者血清LDH、PCT、CRP水平与病情严重程度的相关性。采用ROC曲线分析血清LDH、PCT、CRP水平对颅脑手术患者合并重症肺炎的临床诊断价值,血清LDH、PCT、CRP水平预估颅脑手术合并重症肺炎患者预后的价值。结果:术后1 d、3 d、7 d与未感染组对比,普通肺炎组患者血清LDH、PCT、CRP水平均明显升高(P<0.05);与普通肺炎组对比,重症肺炎组患者血清LDH、PCT、CRP水平均明显升高(P<0.05)。经Pearson相关性检验分析,颅脑手术合并肺炎患者血清LDH、PCT、CRP水平与PSI评分呈明显正相关(P<0.05)。以术后1 d血清LDH、PCT、CRP水平建立ROC曲线,结果发现,血清LDH、PCT、CRP的曲线下面积分别为0.840、0.825、0.746。以术后7 d血清LDH、PCT、CRP水平建立ROC曲线,结果发现,血清LDH、PCT、CRP的曲线下面积分别为0.819、0.725、0.684。结论:LDH在颅脑手术合并肺炎患者中高表达,与肺炎严重程度正相关。LDH在区分非重症肺炎患者和重症肺炎患者以及预测重症肺炎患者的28天死亡率方面具有良好价值。  相似文献

Since peroxidase and manganese have been implicated in both auxin destruction and ethylene production, the effect of auxins and high tissue levels of manganese on the peroxidative indoleacetic acid oxidase system and the internal level of ethylene was determined in cotton (Gossypium hirsutum L. cv. Watson GL-7). The highest level of manganese tested produced manganese toxicity symptoms, including necrotic lesions, accompanied by an increase in internal ethylene levels at about 15 days after treatment initiation. Statistically significant increases in indoleacetic acid oxidase and peroxidase activity were first observed 2 days later and were paralleled by tissue manganese levels above 7.4 milligrams per gram dry weight and internal ethylene levels of 0.77 microliters per liter air. Eight hours after application of 2,4-dichlorophenoxyacetic acid or indoleacetic acid, the internal levels of ethylene were increased to above 6.6 microliters per liter air in cotton plants, and levels of this magnitude were maintained for a 72-hour period of observation. Modification of peroxidase and indoleacetic acid oxidase activity in auxintreated plants definitely occurred well after the elevation of internal ethylene levels. While ethylene levels and indoleacetic acid oxidase activity were increased by both experimental approaches, the earlier appearance of increased ethylene indicates that the peroxidative indoleacetic acid oxidase system in cotton is not involved in ethylene synthesis or that this enzyme is not the rate-limiting factor when ethylene synthesis is increased. Ethylene, as well as auxin destruction, may be involved in some of the long term plant responses to toxic levels of manganese. The findings also suggest that auxin-induced ethylene may play a role in the elevation of peroxidase and indoleacetic acid oxidase activity eventually seen in extracts of plants treated with auxins. The data support the assumption that the enzymatic portion of the indoleacetic acid oxidase system in cotton is a peroxidase.  相似文献

In tick-borne encephalitis certain immunopathological reactions develop in the tissues of the central nervous system; alpha 2-macroglobulin may serve as the marker of the activity of these reactions. The dynamic study of liquor taken from 16 patients with the meningeal and focal forms of tick-borne encephalitis (TBE), 8 patients with severe craniocerebral traumas accompanied by meningitis and 10 patients with osteochondrosis was made. As revealed in this study, in TBE patients the level of alpha 2-macroglobulin increases 3.5-fold and remains stable during the acute period of the disease.  相似文献

Cotton is an important cash crop worldwide, and is a significant source of fiber, feed, foodstuff, oil and biofuel products. Considerable effort has been expended to increase sustainable yield and quality through molecular breeding and genetic engineering of new cotton cultivars. Given the recent availability of the whole-genome sequence of cotton, it is necessary to develop molecular tools and resources for large-scale analysis of gene functions at the genome-wide level. We have successfully developed an Agrobacterium-mediated virus-induced gene silencing (VIGS) assay in several cotton cultivars with various genetic backgrounds. The genes of interest were potently and readily silenced within 2 weeks after inoculation at the seedling stage. Importantly, we showed that silencing GhNDR1 and GhMKK2 compromised cotton resistance to the infection by Verticillium dahliae, a fungal pathogen causing Verticillium wilt. Furthermore, we developed a cotton protoplast system for transient gene expression to study gene functions by a gain-of-function approach. The viable protoplasts were isolated from green cotyledons, etiolated cotyledons and true leaves, and responded to a wide range of pathogen elicitors and phytohormones. Remarkably, cotton plants possess conserved, but also distinct, MAP kinase activation with Arabidopsis upon bacterial elicitor flagellin perception. Thus, using gene silencing assays, we have shown that GhNDR1 and GhMKK2 are required for Verticillium resistance in cotton, and have developed high throughput loss-of-function and gain-of-function assays for functional genomic studies in cotton.  相似文献

A range of virulence levels was found in four populations of Meloidogyne incognita collected from cotton fields of the Punjab region of Pakistan. The most virulent population was associated with development of larger gall size, larger giant cell formation and improved success of juveniles transitioning into reproducing adults. The most virulent nematode population, MI-78, emanated from cotton cultivar NIAB-78. This cotton cultivar also possessed the greatest level of resistance to the three other nematode populations evaluated in this study. The source of plant resistance was not evident during root penetration by second-stage juveniles (J2), but became apparent as nematode feeding was attempted. Although one other cotton cultivar, CIM-506, could also be designated as showing a level of resistance, none of the other cultivars reduced any nematode stage by more than 75% of that achieved on the best host. These data provide an example of a single cotton cultivar that could have short-term utility in field settings. The data also provide insight for future cotton breeding programs.  相似文献
