Desulfovibrio vulgaris lipopolysaccharide and outer membrane proteins (OMPs) were incorporated into vesicles ofD. vulgaris phospholipid and studied for [55Fe]binding activity. Both lipopolysaccharide and an extract of two major OMPs caused large increases in55Fe uptake over control (phospholipid only) vesicles. CommercialSalmonella typhimurium lipopolysaccharide gave a similar result, but the effect was inhibited by calcium ions; this was not the case forDesulfovibrio. The lipid A portion ofS. typhimurium lipopolysaccharide had a high iron-binding ability, whereasDesulfovibrio lipid A iron binding was little different from control values;D. vulgaris lipopolysaccharide thus has a specific iron-binding site within its polysaccharide side chain.  相似文献

Chloroplast DNA (cpDNA) and isozyme variation were analyzed over a range of populations of two infraspecific taxa of the tetraploidSenecio vulgaris. The isozyme data were supportive of the hypothesis that the weedy and cosmopolitanS. vulgaris var.vulgaris is an evolutionary derivative ofS. vulgaris subsp.denticulatus from the coasts of W Europe and montane altitudes in S Spain and Sicily. The two taxa exhibited a very high genetic identity with subsp.denticulatus containing slightly more isozyme diversity than was found in var.vulgaris. — Three cpDNA haplotypes (A, B, C) already known from other Mediterranean diploid species ofSenecio were resolved in var.vulgaris, and an additional fourth haplotype (E) was found in subsp.denticulatus. Two alternative hypotheses were chosen to account for the origin and maintenance of the observed cpDNA composition ofS. vulgaris. It either reflects (1) the retention of an ancestral polymorphism which stems from the recurrent and polytopic formation of ancestral tetraploid lineages; or (2)S. vulgaris originally was characterized by haplotype E, and haplotypes A, B and C were acquired through repeated introgressive hybridization with related diploid species. The finding that very low levels of nuclear (isozyme) diversity were present in both taxa ofS. vulgaris examined supports the second of these two hypotheses; however, more detailed analysis of nuclear genetic diversity is required before a firm conclusion can be reached on this matter.Dedicated to emer. Using the method for the identification of promoters recognized by the sporulation specific σ factor (σF), we identified a positive 950 pbSau3Al DNA fragment inStreptomyces cœlicolor A3(2). High-resolution S1-nuclease mapping identified a potential promoter, PF35, in theE. coli two-plasmid system similar to the consensus sequence ofBacillus subtilis promoters recognized by the general stress-response σ factor (σB). However, the putativesigF-dependent promoter, PF35, was inactive inS. cœlicolor in the course of diffenentiation and it was located divergently in the promoter region directing expression of thechiC gene encoding chitinase. Sequence analysis of the region potentially governed by PF35 revealed two translationally coupled genes encoding proteins similar to bacterial two-component regulatory systems, and with the highest similarity to the two-component systemchiS, chiR, regulating chitinase activity inStreptomyces thermoviolaceus. However, the genes had a divergent orientation with respect to the PF35 promoter. Disruption of theS. cœlicolor chiR gene appeared to have no obvious effect on growth, morphology, differentiation, and production of pigmented antibiotic actinorhodin and undecylprodigiosin. We studied the effects of 15-months of elevated (700 μmol mol−1) CO2 concentration (EC) on the CO2 assimilation rate, saccharide content, and the activity of key enzymes in the regulation of saccharide metabolism (glycolysis and gluconeogenesis) of four C3 perennial temperate grassland species, the dicots Filipendula vulgaris and Salvia nemorosa and the monocots Festuca rupicola and Dactylis glomerata. The acclimation of photosynthesis to EC was downward in F. rupicola and D. glomerata whereas it was upward in F. vulgaris and S. nemorosa. At EC, F. rupicola and F. vulgaris leaves accumulated starch while soluble sugar contents were higher in F. vulgaris and D. glomerata. EC decreased pyrophosphate-D-fructose-6-phosphate l-phosphotransferase (PFP, EC 2.7.1.90) activity assayed with Fru-2,6-P2 in F. vulgaris and D. glomerata and increased it in F. rupicola and S. nemorosa. Growth in EC decreased phosphofructokinase (PFK, EC 2.7.1.11) activity in all four species, the decrease being smallest in S. nemorosa and greatest in F. rupicola. With Fru-2,6-P2 in the assay medium, EC increased the PFP/PFK ratio, except in F. vulgaris. Cytosolic fructose-1,6-bisphosphatase (Fru-1,6-P2ase, EC 3.1.3.11) was inhibited by EC, the effect being greatest in F. vulgaris and smallest in F. rupicola. Glucose-6-phosphate dehydrogenase (G6PDH EC 1.1.1.49) activity was decreased by growth EC in the four species. Activity ratios of Fru-1,6-P2ase to PFP and PFK suggest that EC may shift sugar metabolism towards glycolysis in the dicots.  相似文献

The biomass production of three common aquatic macrophytes,viz. Azolla pinnata, Eichhornia crassipes andHydrilla verticillata, was high at the prevailing environmental conditions and by the enriched water of River Ganga. The biomass production ofAzolla andEichhornia was positively correlated with the orthophosphate phosphorus and nitrate-nitrogen concentrations of the enriched water. The biomass ofAzolla andHydrilla was positively correlated with the electrical conductivity of the water. The average yield of crude protein was highest in Azolla (8,520 kg.ha–1.yr–1), and somewhat lower inEichhornia (6,520 kg.ha–1.yr–1). The annual biogas production was highest inEichhornia (44,381 litres), and somewhat lower inAzolla (17,186 litres).  相似文献

Conclusions GHT1 was isolated as suppressor ofd-glucose uptake deficiency ofS. pombe mutant YGS-5. The correspondingS. pombe DNA encodes a putative protein with significant amino acid sequence identity to theS. cerevisiae HXT transporters. Heterologous expression ofGHT1 inS. cerevisiae hxt mutant RE700A (strain HLY709) enabled the mutant to grow ond-glucose as the sole carbon source. HLY709 cells take up hexoses with similar specificity toS. pombe wild strain and accumulate the non-metabolizable analogues of glucose (2DG and 6DG) intracellularly, thus matchingS. pombe wild strain. Southern blot analysis revealed the existence of other putative glucose transporters inS. pombe and the search for related transporter genes inS. pombe genome is in progress.  相似文献

Minimal inhibitory concentrations for 10 new condensed isoindole derivatives were established by the agar diffusion method. Benzo[b]-6H-pyrimido[2,1-a]isoindole-4-one exhibited a broad antifungal effect; the observed MIC (mg/L) were 7 (K. lactis), 8 (C. pseudotropicalis), 17 (W. fluorescens), 22 (E. magnusii), 31 (P. membranaefaciens), 32 (S. alluvius) and 32 (S. cerevisiae). The compound also inhibited the growth ofB. subtilis (MIC 9 mg/L). No effect on the growth ofE. coli was found.  相似文献

2C nuclear DNA amounts were determined in 30 collections belonging to 10 species ofEleusine. About a 2.5-fold variation in genome size is evident in the genus. The 2C DNA amount in the diploid species ranged from 2.50 pg inE. verticillata to 3.35 pg inE. intermedia. In contrast, the tetraploid species showed a range from 4.95 pg inE. africana to 6.13 pg inE. floccifolia. At intraspecific level 10 collections ofE. coracana, 6 ofE. indica, 4 ofE. africana, 2 ofE. tristachya, and 2 ofE. kigeziensis did not show any significant variation. However, 2 collections ofE. floccifolia, connected with polyploidy, displayed about 90% variation. Polyploid species showed approximately double the genome size of that of their corresponding diploids. An evolutionary increase in DNA amount is evident inE. coracana during the course of its origin and domestication fromE. africana.  相似文献

It has been shown by means of double wavelength cytophotometry of DNA (Feulgen reaction) and histone (fast green, pH 8.2) inTriturus vulgaris spermatocytes that the doubling of DNA content in nuclei terminates at the end of preleptotene to beginning of leptotene whereas the doubling of histone content begun at premeiotic interphase is delayed and proceeds till the end of leptotene to beginning of zygotene. As a result preleptotene spermatocytes contain approximately 4C DNA and only 3C histone. Histone content in leptotene amounts to 93% of 4C, and in zygotene, pachytene and metaphase I both DNA and histone contents equal 4C. Thus, the temporal pattern of nucleo-histone doubling in meiotic chromosomes ofT. vulgaris differs from the synchronous DNA and histone doubling in mitotic chromosomes of all previously studied species. The delay of histone doubling inT. vulgaris meiocytes is less pronounced than in the previously studied insectsAcheta domestica andPyrrhocoris apterus where the histone content amounts to 3C in leptotene—zygotene and the equal histone/DNA ratio is restored only in pachytene.—Responsibilities for this phenomenon and its biolgoical sinnificance are discussed in connection with recent hypotheses concerning mechanisms of homologous chromosome pairing.  相似文献

Summary The incidence of virus infections in three lepidopterous insect species was studied from 1965 to 1968 in alfalfa fields in California. The insects were the alfalfa caterpillar,Colias eurytheme; the beet armyworm,Spodoptera exigua; and the alfalfa looper,Autographa californica. InC. eurytheme, the major virus was a nuclear polyhedrosis virus (NPV); inS. exigua, a granulosis virus (GV) and an NPV; inA. californica, a GV. Virus epizootics did not develop in very high densities ofC. eurytheme. Virus epizootics occurred in low host densities of the three insect species, especially in populations ofA. californica. The virus acted as a density-dependent factor in the regulation of the populations ofS. exigua andA. californica. Temperature, humidity and rainfall had no marked effect on the incidence of virus infections.  相似文献

An electrophoretic survey of isozyme variation was conducted to test the hypothesis thatSenecio vulgaris L. (2n = 40) is of autotetraploid origin fromS. vernalis Waldst. & Kit. (2n = 20). It was established thatS. vulgaris exhibited fixed heterozygosity at three loci examined, showed disomic inheritance at all polymorphic loci, and contained a gene (Est-1) and an allele (Aat-3b) which were not present in the single population ofS. vernalis surveyed. From this it is concluded thatS. vulgaris is not of autotetraploid origin. Instead, the genetic evidence is in keeping with an allopolyploid origin ofS. vulgaris with the possibility thatS. vernalis acted as one of its two parents.  相似文献

In the semi-desert environment of the Mu Us Sandland, the vegetation is composed chiefly of shrubs and semi-shrubs, coverage normally amounting to 30-40%. However, an exception can be found in the community of Sabina vulgaris Antoine, an evergreen shrub, which tends to grow so densely that it covers the sand dunes completely. Previous research has indicated that the high density of Sabina is parallelled by the very low transpiration rate. Based on anatomical and scanning electronic microscopic observations, this paper also points out that two structural features should play an important role in the potential mechanism of water conservation in this plant. First, a low stomatal density in S. vulgaris can lead to a high stomatal resistance and low transpiration. Second, the morphological feature of S. vulgaris leaves being tightly pressed to the stems might be a structural feature of water conservation, because in this way the leaf side bearing more (and mostly larger) stomatal pores could be protected from direct high radiation, while the side with fewer (and mostly smaller) pores (the outer side) is exposed to the dry desert air.  相似文献

A DNA fragment coding for a carboxymethylcellulase (CMCase) ofFibrobacter succinogenes S85 was isolated from a pUC18 gene library inEscherichia coli JM109. The CMCase gene was present as a single copy in theF. succinogenes S85 genome and was found in all the otherF. succinogenes strains tested. The gene was expressed from an endogenous promoter inE. coli and was not subject to glucose repression. Most of the CMCase activity was located in the membrane ofE. coli. Zymogram analysis and35S labeling of the proteins encoded by the CMCase gene-containing plasmid indicated that the enzyme has a molecular mass of 58,000. The optimal pH and temperature of activity on CMC were respectively 6.4 and 30°C. The enzyme was active on CMC, barley -glucan, and lichenan but would not hydrolyze laminarin and exhibited no exoglucanase-type activity, suggesting that it is an endo-(1,4)--d-glucanase.  相似文献

The 16 species of theSedum acre-group were investigated for the presence of alkaloids. They areS. acre ofS. ser.Acria, S. alpestre, S. annuum, S. apoleipon, S. borissovae, S. euxinum, S. grisebachii, S. laconicum, S. multiceps, S. sexangulare, S. tuberiferum, S. tuberosum, S. ursi, andS. urvillei ofS. ser.Alpestria, S. samium ofS. ser.Samia, andS. litoreum ofS. ser.Litorea. S. acre differs significantly from the other species. It contains sedamine, hydroxy sedamine, and a number of 2,6-disubstituted piperidine alkaloids. The leafy parts of the species ofS. ser.Alpestria, S. ser.Samia, andS. ser.Litorea contain 4 piperidine alkaloids which also occur inS. acre, and in addition 4 pyrrolidine alkaloids not present inS. acre. The composition of the alkaloid fraction agrees with the infrageneric classification (series) based on the hybridization patterns of the species (comparia).  相似文献

采用中华蜜蜂Apis cerana cerana F.临床自然发病的欧洲幼虫腐臭病(European Foulbrood, EFB)幼虫,对其病原体进行了分离鉴定。结果表明:中华蜜蜂EFB的病原系蜂房蜜蜂球菌Melissococcus pluton。该菌为革兰氏阳性的兼性厌氧菌,形态学及染色特性、致病性试验、血清学试验和细菌DNA G+C mol%试验均证明中华蜜蜂和西方蜜蜂的EFB病原属于同属的蜂房蜜蜂球菌。生理生化特性结果与国外的早期研究结果相近似。该试验为不同地理位置、不同种寄主间蜂房蜜蜂球菌的遗传差异的研究,以及中华蜜蜂EFB病的防治学研究打下基础。  相似文献

This study focuses on the variability of chromosomal location and number of ribosomal DNA (rDNA) sites in some diploid and autotetraploidFestuca pratensis andLolium perenne cultivars, as well as on identification of rDNA-bearing chromosomes in their triploid and tetraploidF. pratensis ×L. perenne hybrids. The rDNA loci were mapped using fluorescence in situ hybridization (FISH) with 5S and 25S rDNA probes, and the origin of parental genomes was verified by genomic in situ hybridization (GISH) withL. perenne genomicDNAas a probe, andF. pratensis genomic DNA as a block. FISH detected variation in the number and chromosomal location of both 5S and 45S rDNA sites. InF. pratensis mostly additional signals of 5S rDNA loci occurred, as compared with standardF. pratensis karyotypes. Losses of 45S rDNA loci were more frequent inL. perenne cultivars and intergeneric hybrids. Comparison of theF. pratensis andL. perenne genomes approved a higher number of rDNA sites as well as variation in chromosomal rDNA location inL. perenne. A greater instability ofF. pratensis-genome-like andL. perenne-genome-like chromosomes in tetraploid hybrids was revealed, indicating gains and losses of rDNA loci, respectively. Our data indicate that the rDNA loci physically mapped on chromosomes 2 and 3 inF. pratensis and on chromosome 3 inL. perenne are useful markers for these chromosomes in intergenericFestuca ×Lolium hybrids.  相似文献

Here we report the seed morphology and anatomy of some Nigerian pulses, an investigation that revealed a wide diversity of characteristics in a small collection of seeds. Although these genera share common features, their anatomical differences make it possible to generate a key for identification and classification. Sizes ranged from 5 to 7 x 5 x 3 mm forCajanus cajan up to 25 to 30 x 18 x 19 mm forCanavalia ensiformis. Seed colors were monochromatic black/brown to dichromatic eye/mottled (streaked), and surfaces were either smooth-glossy or puckered. Forms varied from ellipsoid or oblong-ellipsoid to spheroid or reniform, and halos (hilar rims) were either complete or incomplete. Lens shapes were spot, linear, broadly linear, or deltoid. The hila were elliptic to linear (or in-between), with sizes ranging from 0.5 to 1.0 mm (Mucuna pruriens) up to 1.5 to 3.0 mm (Phaseolus vulgaris). Their positions could be completely covered, as inP. vulgaris, partially covered and raised above the seed surface (Vigna subterranea), or naked (without any funicular residue) arid level with the surface, as in Gensiformis (Tce1 ). The aril caps were collar-like inLablab purpureus or cushion-like, as inM. pruriens. Palisade cell sizes ranged from 80.7pm inGlycine max to 1733 μm inC. ensiformis (Tce1), and their shapes varied from a uniformly wall-thickened type (T1), to a bulbous-end type (T2), to one with a corrugated structure on the inner wall (T3). Although these anatomical variations exist, they may not warrant demarcation into sub-familiar or generic classifications. However, we do propose that specific and sub-specific alterations are necessary.  相似文献

The carnivorous bladderworts (Utricularia) possess complicated suction traps. Remarkably, information on the prey trapped is relatively sparse. We have conducted a detailed survey on the prey spectra found in traps of selected aquatic bladderworts (U. australis, U. vulgaris) occurring in ponds in northeastern Germany. A close examination of more than 200 traps revealed cladocerans, copepods, rotiferas, ciliates and insect larvae as being common prey.Of particular interest was the considerable amount of phytoplankton (i.e. algae, cyanobacteria) found in the traps. In total, more than 160 algae species (among others, Kirchneriella lunaris, Scenedesmus quadricauda and S. acuminatus) belonging to more than 50 genera were present, with Chlorophyceae being dominant. The role of the vegetarian diet for nutrient supply of bladderworts is discussed.  相似文献

Summary We demonstrated that a washed suspension ofE. coli does not subsist in Vittel mineral water more than one to four days depending on the size of the inoculum. On the other hand,P. vulgaris and especiallyK. pneumoniae can survive much longer in this water. Thereafter we showed that the longevity ofE. coli andS. faecalis introduced into mineral water by fecal matters may be 1.5 months if the amount of feces is too small to allow multiplication of the bacteria, and 3–4 months if the amount of feces is sufficient to allow multiplication.Our results also show that various organic products introduced into mineral water have the same protective effect as feces uponE.coli andS.faecalis, notably if they allow these bacteria to proliferate.The importance of the time interval between bottling the water and sampling for bacteriological analysis is discussed with the aim of understanding the significance of the presence or absence of test-bacteria as indicator of fecal pollution of bottled mineral water.  相似文献
